Abstract:
Abstract CDK4/6 control a key checkpoint by phosphorylating Rb, leading to Rb degradation and subsequent S-phase entry. CDK4/6 inhibition, together with endocrine therapy, is standard of care for advanced ER+ breast cancer; however, CDK4/6 inhibition typically is cytostatic. This study aims to identify breast cancer patients who may better respond to treatments targeting CDK4/6. NF1/neurofibromin is a key tumor suppressor that we have shown can represses not only RAS as a GAP but also ER as a transcriptional co-repressor. Dual activation of RAS and ER promotes endocrine therapy resistance. We computed kinase activity scores from phosphor-proteomic data in the CPTAC breast cancer cohort and found that both CDK4/6 activity scores and Rb-pS780 levels negatively correlated with NF1 protein levels. Furthermore, NF1 mutations co-occur with mutations in CDK4/6 and CCND1-3 in breast tumors. These results suggest a functional dependency between NF1 loss and activation of the CDK4/6-Rb pathway. To delineate how NF1 and CDK4 interact molecularly, we depleted NF1 in ER+ breast cancer cells and found an increase in Rb-pS780 and in phosphorylation in CDK4’s activation loop (CDK4-pT172). Cyclin-D binding is required for CDK4 activation. CCND1 expression is under the direct control by ER, and upon NF1-depletion, Cyclin D1 expression was increased, partly due to enhanced ER recruitment to an ERE in CCND1. Cyclin-D-bound CDK4 must be phosphorylated at T172 to be fully active; however, what kinase is responsible for this is largely unknown. We have evidence that CDK4-T172 phosphorylation is RAF, but not ERK, dependent. When the NF1-depleted ER+ cells were seeded in the presence of fulvestrant together with either palbociclib or abemaciclib, apoptosis readily occurred. Remarkably, fulvestrant plus palbociclib efficiently caused durable tumor regression in two ER+ PDX models, in which NF1 is undetectable by IHC. In contrast, no tumor regression was seen in a PDX model with detectable NF1. Finally, greater growth inhibition and apoptosis were detected in NF1-low tumors in the neoadjuvant NeoPalAna trial when palbociclib was added after anastrozole. These data support a model whereby ER and RAS signaling converge upon CDK4/6, and CDK4/6 activation is a key survival mechanism when ER signaling is attenuated by treatment in NF1-depleted ER+ breast cancer cells. This apparent addiction for CDK4/6 activity makes NF1-depleted ER+ breast tumors vulnerable to CDK4/6 inhibition, thus creating a potential therapeutic opportunity to match CDK4/6 inhibition with patients who can benefit the most. Abstract:
Abstract Background Tic disorders (TD) is a neuropsychiatric disease with twitch as the main manifestation in childhood. Jiu-Wei-Xi-Feng granules has been marketed in China for treating children with TD. As Long Gu (Os Draconis) in the composition of this Chinese patent medicine is a rare and expensive medicinal material protected by the Chinese government, therefore, we consider replacing it with Mu Li (Concha Ostreae) that has the same effect and is cheaper. This study is designed to evaluate the clinical equivalence between Jiu-Wei-Xi-Feng granules (Os Draconis replaced by Concha Ostreae) (JWXFD) and Jiu-Wei-Xi-Feng granules (original formula) (JWXFO) in children with TD (consumption of renal yin and liver wind stirring up internally syndrome). Methods/design This is a multicenter, randomized, double-blind, equivalence trial comparing the efficacy and safety of JWXFD and JWXFO in treating Children with tic disorders (consumption of renal yin and liver wind stirring up internally syndrome). A total of 288 patients will be recruited and randomly assigned to two groups in a 1:1 ratio. The treatment course is 6 weeks, with a 2 weeks follow-up. The primary outcome is the mean change value from baseline to 6th week by the Yale Global Tic Severity Scale total tic score (YGTSS-TTS). Secondary outcomes include total effective rate of tic, Yale Global Tic Severity Scale (YGTSS) scores and its factor scores (the degree of motor tics, phonic tics and social function damage), Clinical Global Impression-Severity scale, and TCM syndrome efficacy. Discussion The design of this study refers to a large number of similar research design points, and asked for opinions of peer experts, and finally reached a consensus. This trial will provide high-quality evidence on the clinical equivalence between JWXFD and JWXFO and provide a basis for the marketing of JWXFD. This paper proposes the concept of multisensory landscape construction, pointing out that people’s perception and appreciation of a landscape is a process of overall impression and judgment formed with a combination of visual, auditory, olfactory, tactile, and even thermal and humid sense. Examples can be found in many famous traditional Chinese gardens. Around the West Lake area, there are famous soundscape scenic spots, e.g., Liu Lang Wen Ying (Orioles Singing in the Willows), Nan Ping Evening Bell (Evening Bell Ringing at the Nanping Hill), Zhejiang Qiu Tao (Autumn Wave of Zhejiang), and Jiu Li Song Tao (Wave of Pine Trees Lasts Nine Miles) . There are also some famous lightscape scenic spots, e.g., San Tan Ying Yue (Moon and Candlelight Mirrored in the Lake) and Ping Hu Qiu Yue (Moon over the Peaceful Lake in Autumn) . In terms of smellscape, in addition to the famous scenic spot Qu Yuan Feng He (Curved Yard and Lotus Pool in Summer), the West Lake area is also widely planted with osmanthus and other fragrant plants, forming a smellscape in which "late autumn is fragrant with osmanthus flowers and lotus in bloom for miles and miles." At Humble Administrator Garden, there are soundscape scenic spots such as Wu Zhu You Ju (Secluded Residence among Bamboo Bushes) and Liu Ting Ge (Pavilion to Pause and Listen); there are smellscape scenic spots such as the Orchid Field, the Magnolia Courtyard, the Panicum Pavilion, etc.; lightscape scenic spots such as Liu Ying Ge (Hall of Reflecting Shadows) and Ta Ying Ting (Pavilion of Shadow of Tower) can also be found there. In Chengde Summer Resort, there are soundscape scenic spots such as Wan He Song Feng (Wind of Ten Thousand Ravines and Pines) and lightscape scenic spots such as Xi Ling Chen Xia (Morning Sunset on West Ridge), etc.; smellscape scenic spots such as Qu Shui Hua Xiang (Fragrance of Flowers in the Curved Water) and Yuan Xiang Tang (Hall of Fragrance of Far Away) can also be found.The above classic cases eloquently prove that the creation of multisensory landscape and the integration of them are the valuable experience in traditional Chinese gardens, which play an important role in the achievement of famous landscape.Therefore, the design of landscape must pay attention to the creation and integration of soundscape, smellscape and lightscape. Another key point of the theory of multisensory landscape construction is that it is necessary to pay attention to both spatial and temporal dimensions so that the constructed landscape can be enjoyed everywhere and at all time periods. In this regard, the creation of the three-scape (specifically refer to soundscape, smellscape and lightscape) can also highlight their regional and temporal characteristics. By analyzing some classic cases of traditional Chinese gardens, this paper proposes that the construction of multisensory landscape and the integration of soundscape, smellscape and lightscape are the valuable experience in traditional Chinese gardens, which are also important for the achievement of famous landscapes and are excellent traditions that we should vigorously inherit and carry forward.

Lighter alcohols like ethanol and methanol are easily miscible with gasoline and by blending alcohols with gasoline; a part of conventional fuel can be replaced while contributing to fuel economy. Several researchers tested various ethanol blends on different engine test rigs and identified ethanol as one of the most promising ecofriendly fuels for spark ignition engine. Its properties high octane number, high latent heat of vaporization give better performance characteristics and reduces exhaust emissions compared to gasoline. This paper focuses on studying the effects of blending 50 of ethanol by volume with gasoline as it hardly needs engine modifications. Gasoline (E0) and E50 fuels were investigated experimentally on single-cylinder, four-stroke port fuel injection spark ignition engine by varying engine speed from 1500 rpm to 3500 rpm. Custom AFOs provide better fit, comfort and performance than pre-manufactured ones. The technique of 3D-printing is suitable for making custom AFOs. Fused deposition modelling (FDM) is a 3D-printing method for custom AFO applications with the desired resistance and material deposition rate. Generally, FDM is a thermal process; therefore materials thermal behaviour plays an important role in optimizing the performance of the printed parts. The objective of this study is to evaluate the thermal behaviour of PLA, ABS, nylon and WF-PLA filaments before manufacturing the AFO components using the FDM method. In the study, the sequence of testing materials provides a basic measuring method to investigate AFO device parts thermal stability. Thermal analysis (TG/DTG and DSC) was carried out before 3D printing is to characterize the thermal stability of each material. Moreover, energy levels in these deposits are low and need to be elevated by introducing efficient operative conversion technologies to utilize these residues as fuels. In this context, the utilization of a fluidized bed innovation enables a wide range of non-uniform-sized low-grade fuels to be effectively converted into other forms of energy.This study was undertaken to evaluate the effectiveness of fluidized conversion method for transformation of agricultural by-products such as rice husk, sawdust, and groundnut shells into useful energy. The present investigation was conducted to know the mixing characteristics of sand and fuel have been found by conducting experiments with mixing ratio of rice husk (1:13), saw dust(1:5) and groundnut shells (1:12), the variation of particle movement in the bed and mixing characteristics are analyzed. The impact of sand molecule size on the fluidization speed of two biofuel and sand components is studied and recommended for groundnut shells using a sand molecule of 0.6 mm size and for rice husk, sawdust 0.4 mm sand particle size. Also, establish that the particle size of sand has a significant effect on mingling features in case of sawdust. In the next part of the investigation, the CFD simulations of the fluidized bed are done to investigate the mixing behavior of sand and biomass particles. A set of simulations are conducted by ANSYS FLUENT16; the state of the bed is the same as that of the test. The findings were presented with the volume fraction of sand and biomass particles in the form of contour plots. Tulong (1543-1606), once had another name Changqing, Wei-Zhen , and Chi-Shui. Who was bone in Ningbo Yinxian。He was a famous writer and drmatist. This research which range was all Tulong’s life, his friends about drama and his works " feng- yi-ge-yue-fu". This research used Geographic Information System(GIS)and pai-chang theory, to discuss Tulong’s influence about drama in late ming dynasty. The first chapter discussed all Tulong’s life, and background which about late ming dynasty. The first chapter referenced Xu Shuo Fang’s "Wan Ming Qu Jia Nian Pu" and Wang Chao Hong’s "Ming Qing Zhe Ji Qu Jia Kao" to study period, location, dating events, friends, and works……exc fourteen project. Tulong’s life had tow very important things which is “zhong ju ren guan” and “ba guan gui xiang”. These Thing divided all Tulong’s life into three parts. This chapter will draw painting to discuss.At last it will research the location which about Tulong’s drama and fridnds to discuss Tulong’s influences in drama. The second chapter is about Tulong’s script . Tulong creared three scripts which is “tan hua ji”, “cai hao ji ”and “xiu wen ji”. Three scripts was named “ feng- yi-ge-yue-fu”. This chaper introduced story what time was creared about three scripts, theme, and version. This chapter introduced plot, architecture and character image in three scripts at last. The third chapter is about “tan hua ji” pai chang artistic research. It introduced pai chang theory in the first. The theory was according to luo li rong “Qu Xue Gai Yao”. This theory is on the basis of this research. This chapter arranged character in each parts, and discussed form , characteristic, and pai chang leng re whichis according to pai chang theory. This chapter analysised character ‘s work and reset by character appearances and songs. This chapter is to discuss “tan hua ji” achievements in drama. The fourth chapter is about “cai hao ji ” pai chang art. It discussed pai chang leng re by counting character’s appearance, form and characteristic in each parts. It discussed analysised character ‘s work and reset by character appearances and songs. The fifth chapter is about “cai hao ji ” pai chang art. It discussed pai chang leng re by counting character’s appearance, form and characteristic in each parts. It discussed analysised character ‘s work and reset by character appearances and songs. Conclusion part summarizes the TuLong and location. It discussed all Tulong’s life has been to place, and friends about drama, and influences in drama. It discussed “Qu Xue Gai Yao” pai chang achienement in drama.
