Abstract:
Anopheles gambiaeis the major malaria vector in Africa. Examining the molecular basis ofA. gambiaetraits requires knowledge of both genetic variation and genome-wide linkage disequilibrium (LD) map of wildA. gambiaepopulations from malaria-endemic areas. We sequenced the genomes of nine wildA. gambiaemosquitoes individually using next-generation sequencing technologies and detected 2,219,815 common single nucleotide polymorphisms (SNPs), 88% of which are novel. SNPs are not evenly distributed acrossA. gambiaechromosomes. The low SNP-frequency regions overlay heterochromatin and chromosome inversion domains, consistent with the lower recombinant rates at these regions. Nearly one million SNPs that were genotyped correctly in all individual mosquitoes with 99.6% confidence were extracted from these high-throughput sequencing data. Based on these SNP genotypes, we constructed a genome-wide LD map for wildA. gambiaefrom malaria-endemic areas in Kenya and made it available through a public Website. The average size of LD blocks is less than 40 bp, and several large LD blocks were also discovered clustered around theparagene, which is consistent with the effect of insecticide selective sweeps. The SNPs and the LD map will be valuable resources for scientific communities to dissect theA. gambiaegenome.

Abstract:
AbstractPCR-RFLP techniques have been used to characterize wild and laboratory isolates of the trichostrongyle nematodeHeligmosomoides polygyrusfrom the wood mouseApodemus sylvaticusand the laboratory mouseMus musculusrespectively. Both isolates can be distinguished by eight endonuclease digestions of the ITS region of the rDNA repeat namely,AluI,DdeI,HpaII,HaeIII,HinfI,HhaI,PvuII andSalI. In two of the digests,HinfI andRsaI, a minor polymorphism was observed in the wild isolate ofH. polygyruswhich has been cultured in laboratory-bredA. sylvaticusfor several generations when compared withH. p. polygyrusfrom wildA. sylvaticus. A minor polymorphism was also identified in further wild isolates ofH. polygyruscollected fromA. sylvaticusin a field site in Egham, Surrey. However no evidence of polymorphism was observed in the laboratory isolate ofH. polygyrusfrom the CD1 strain ofM. musculusand the laboratory-bredA. sylvaticus. Reasons for this are discussed and further studies on the population genetics ofH. polygyrusare suggested.

Abstract:
BackgroundAnoectochilus roxburghiiis a widespread herbaceous plant with high medicinal value. WildA. roxburghiiresources face extinction due to their slow growth rate and over exploitation. The growing market demand has led to advances in the field of artificial planting ofA. roxburghii. Methods to increase the economic benefits of cultivation and the production of medicinal ingredients are very useful.MethodsA. roxburghiiwas exposed to red light, blue light (BL), yellow light (YL), green light, or white light as supplemental lighting at night (18:00–02:00) in a greenhouse or were left in darkness (control, CK) to investigate the effects of various light qualities on growth indices, photosynthetic pigments, chlorophyll fluorescence, root vitality, stomatal density, soluble proteins, sugars, and the accumulation of secondary metabolites.ResultsSupplementation of BL had a positive effect onA. roxburghiigrowth and secondary metabolite accumulation. Leaf number, stem diameter, fresh weight, dry weight, chlorophyll a content, and secondary metabolite (total flavonoids, total polyphenols) content increased significantly. YL treatment showed significantly higher soluble sugar and polysaccharide contents than the control.DiscussionBL treatment was conducive to promoting the growth and accumulation of secondary metabolites (total flavonoids, total polyphenols); YL treatment significantly increased the content of soluble sugar and polysaccharides more than the control. Polysaccharides and total flavonoids are important medicinal ingredients ofAnoectochilus, so future research will focus on the combination of blue and YL.

One technology that can be developed on swamp land is utilization of new superior variety and planting system. Economic feasibility is one of considerations for farmers to adopt a technology innovation. The study aimed to determine the combination of the most economically viable new superior variety and planting system to be developed in swamp rice farming. The study was conducted from May-August 2016 in Karang Anyar Village Semidang Alas Maras district, Seluma Regency. There are 7 New Superior Varieties of swamp paddy cultivated; Inpara 1, 2, 3, 6, 7, 8, and Dendang and 1 existing variety as benchmark variety, Cigeulis. The planting system applied were legowo planting system (2:1) with insertion and without insertion. The data used to achieve the research objectives is primary data in the form of farming data to calculate economic benefits and feasibility. Farm profit is calculated using partial analysis whereas economic feasibility is calculated by evaluating the break even point and R/C ratio. The results showed that Inpara 6 with legowo planting system with insertion yield the largest profit Rp 15.096.600,-/ha with R/C ratio of 2.92.

Abstract:

Abstract:
In recent years, the determination of the antioxidant and antibacterial activity of essential oils in wild plants, such as Mexican oregano (Lippia graveolens Kunth), has become increasingly important. The objective was to compare the antioxidant and antibacterial activity of Mexican oregano essential oil obtained from plants occurring naturally in semiarid areas (Wild1 and Wild2), and those cultivated in the field (CField) and greenhouse (CGreenhouse) in northern Mexico. The Mexican oregano essential oil extraction was performed using the hydrodistillation method, the antioxidant activity was determined using the ABTS method, and the antibacterial activity was assessed through bioassays under the microwell method at nine different concentrations. The aim was to determine the diameter of the inhibition zone and, consequently, understand the sensitivity level for four bacterial species. The results revealed an antioxidant activity ranging from 90% to 94% at the sampling sites, with Wild1 standing out for having the highest average antioxidant activity values. Likewise, six out of the nine concentrations analyzed showed some degree of sensitivity for all the sampling sites. In this regard, the 25 µL mL−1 concentration showed the highest diameter of inhibition zone values, highlighting the Wild2 site, which showed an average diameter greater than 30 mm for the four bacteria tested. Only in the case of S. typhi did the CGreenhouse site surpass the Wild2, with an average diameter of the inhibition zone of 36.7 mm. These findings contribute to the search for new antioxidant and antibacterial options, addressing the challenges that humanity faces in the quest for opportunities to increase life expectancy.

Abstract:
Abstract Background: Epstein-Barr Virus (EBV)-associated gastric carcinoma (GC) (EBVaGC) is a subtype of GC which represents approximately 10% of this tumor. CDKN2B antisense RNA 1 (CDKN2B-AS1) is a long non-coding RNA associated with chronic diseases like cardiovascular disease and cancer. This study aimed to evaluate if the loss of CDKN2B-AS1 shifted the balance of the competing endogenous RNA (ceRNA) networks favoring an apoptosis resistant phenotype through ebv-miR-BART4-5p/BID in EBVaGC. Material and Methods: Seven gastric cancer cell lines were used to determine the expression of CDKN2B-AS1. Specific cell lines were used for CDKN2B-AS1 inhibition assays, the transfection of ebv-miR-BART4-5p and the binding between CDKN2B-AS1 and ebv-miR-BART4-5p. The detection of BID protein was determined by Western Blot. To validate the in vitro results, we detected CDKN2B-AS1, BID, and ebv-miR-BART4-5p expression levels by RT-qPCR in four EBVaGC positive and five negative cases. Apoptosis was detected by in situ hybridization (ISH). Results and Discussion: CDKN2B-AS1 is expressed at higher levels in NCI-N87 and SNU-1 cell lines and at lower levels in AGS and KATO-III. A significant decrease in BID protein expression was observed after co-transfection with ebv-miR-BART4-5p and CDKN2B-AS1 inhibitor. However, these results were not significant at mRNA level. The luciferase assay validated the predicted binding between ebv-miR-BART4-5p and CDKN2B-AS1. In EBVaGC samples, a significant decrease in BID expression and cleaved caspase-3 in the presence of miR-BART4-5p was observed. Our results show that decreasing CDKN2B-AS1 expression favors viral miR binding on BID mRNAs through ceRNA networks. Also, these results could explain why positive EBVaGCs have lower apoptosis rate than EBVaGC negative cases. Abstract:
Mouse models are critical tools in tuberculosis (TB) research. Recent studies have demonstrated that the wild mouse gut microbiota promotes host fitness and improves disease resistance. Here we examine whether the wild mouse gut microbiota alters the immunopathology of TB in BALB/c mice. Conventional BALB/c mice (LabC) and mice born to germ-free BALB/c mothers reconstituted with the wild mouse gut microbiota (WildR) were used in our studies. WildR mice controlled initial TB infection better than LabC mice. The microbial gut communities of LabC mice and WildR mice had similar richness but significantly different composition prior to infection. TB reduced the gut community richness in both cohorts while differences in community composition remained indicating a general TB-induced dysbiosis. The wild mouse gut microbiota did not alter the typical lung histopathology of TB in the BALB/c model that includes unstructured immune cell infiltrates with infected foamy macrophages invading alveolar spaces. Animals of both cohorts mounted robust T cell responses in lungs and spleen with lower absolute counts of CD4 and CD8 T cells in lungs of WildR mice during acute infection, corresponding with observed differences in pathogen load. In summary, LabC mice and WildR mice showed largely overlapping TB immunopathology and pathogen kinetics, with WildR mice controlling early acute infection better than LabC mice.

Abstract:
The Wild and Scenic Rivers Act (WSRA) marked its fiftieth anniversary in 2018 without much fanfare. The WSRA has been somewhat overshadowed by the Wilderness Act, which preceded it by four years, and by the National Environmental Policy Act and the pollution control statutes which followed in the 1970s. But the WSRA was a significant conservation achievement, has now extended its protections to over 200 rivers, and has the potential to provide watershed protection to many more in the future. This article explains the statute and its implementation over the last half-century as well as a number of challenges to fulfilling its laudable goals of protecting free-flowing rivers, their water quality, and their “outstandingly remarkable values.” We make a number of suggestions to the managing agencies and to Congress if the WSRA’s achievements over the next half-century are to match the last fifty years, including reviving congressional interest in study rivers, updating managing agencies’ river plans to focus on non-federal lands within river corridors, and ensuring that those river plans provide the watershed protection Congress envisioned when it included a significant amount of riparian land within WSRA river corridors. We also call for a new emphasis on rivers that should be studied for their restoration potential and for more states to take advantage of the statute’s unusual pathway for state-designated rivers to gain WSRA protections. A postscript briefly explains a recent congressional proposal to expand the WSRA system, and an appendix catalogues all 226 WSRA river segments designated during the statute’s first fifty years.

Abstract:
Wild plant genetic resources are increasingly becoming valuable for breeding, genomics, and ornamental horticulture programs. Wild relatives of horticultural species may offer desirable traits that are not available in cultivated varieties, but “wilds” often also have traits that are highly undesirable. Advances in comparative genomics and marker-assisted breeding facilitate the inclusion of the valued traits from wild materials in plant breeding programs. As technologies advance, wild plant genetic resources will become even more valuable for future research developments. This serves as an introduction to a series of proceedings articles from the American Society of Horticultural Science meetings in 2010 workshop entitled “Horticultural Value of Wild Genetic Resources.”
