Full textIn eukaryotes, secretory proteins are synthesized on cytoplasmic ribosomes and transported into the endoplasmic reticulum (ER), where they obtain their biologically active structure. Defect proteins that arise from errors in this maturation process are degraded by the “ER-associated protein degradation” (ERAD) pathway. A central component of ERAD is the HRD-ligase, a membrane-bound protein complex that contains the ubiquitin ligase Hrd1. Some published work raised speculations on an involvement of this trans-membrane domain in a novel transport system for the export of misfolded proteins from the ER. In the course of this work new insights for the importance of the transmembrane domain of Hrd1 for the degradation of ERAD substrates in the model organism Saccharomyces cerevisiae were obtained. The substitution of two amino acids in the third trans-membrane helix of Hrd1 (Hrd1E84L, H101L) specifically impairs the turnover of misfolded ER-luminal proteins whereas the processing of membrane-bound Hrd1 substrates remained largely unaffected. The Hrd1 variant therefore displays most likely a defect in the transport of luminal ERAD substrates through the ER membrane. No major changes in the assembly of the HRD-ligase by the Hrd1 variant were detected. Still, in site specific in vivo photo-crosslinking assays substantial changes in the spatial orientation of the ligase subunit Der1 towards Hrd1E84L, H101L were detected. Der1 is exclusively required for the degradation of ER-luminal ERAD substrates. This implies that the proper alignment of Hrd1 and Der1 within the ER membrane is a prerequisite for the transport of ERAD substrates into the cytoplasm. This work shows for the first time a direct involvement of the trans-membrane region of Hrd1 in the degradation of luminal ERAD client proteins.

Full textIn contrast to the monomer leaderpeptidase of the prokaryotic plasmamembrane, the eukaryotic signalpeptidase of the ER-membrane is a heteromer protein complex. In yeast the signalpeptidase consist of the four subunits Sec11p, Spc1p, Spc2p and Spc3p. Additional to Sec11p also Spc3p is essential for cell growth and cell life. The depletion of Spc3p cause lethal accumulation of precursor proteins in vivo and lost of cleavage activity in vitro. Spc1p and Spc2p are not essential for the cell. We show here, that the Spc2p subunit interacts with the ß-subunits of the Sec61- and the Ssh1-complex. These data implicate that Spc2p facilitates the interactions between different components of the translocation site. In yeast, efficient protein transport across the endoplasmic reticulum (ER) membrane may occurco-translationally or post-translationally. The latter process is mediated by a membrane protein complex that consists of the Sec61p complex and the Sec62p-Sec63p subcomplex. In contrast, in mammalian cells protein translocation is almost exclusively co-translational. This transport depends on the Sec61 complex, which is homologous to the yeast Sec61p complex and has been identified in mammals as a ribosome-bound pore-forming membrane protein complex. We report here the existence of ribosome-free mammalian Sec61 complexes that associate with two ubiquitous proteins of the ER membrane. According to primary sequence analysis both proteins display homology to the yeast proteins Sec62p and Sec63p and are therefore named Sec62 and Sec63, respectively. The probable function of the mammalian Sec61-Sec62-Sec63 complex is discussed with respect to its abundance in ER membranes, which, in contrast to yeast ER membranes, apparently lack efficient post-translational translocation activity.

Full textMicrosoft har i SharePoint 2013 förbättrat användarnas personliga sidor (My Site) genom att sammankoppla dem i ett socialt nätverk. I förbättringen har ett personligt nyhetsflöde som visar händelser som användaren följer tillagts. Denna rapport avser att undersöka möjligheten att utöka det personliga nyhetsflödet med att lägga till en oberoende komponent i My Site. Komponenten ska tillåta användarna att prenumerera på företagsnyheter genom att välja bland olika nyhetskategorier. En prototyp av komponenten implementerades och resultatet uppfyllde de flesta kraven som ställdes i början av arbetet. Komponenten har lagts till i användarens My Site som en webb del och hämtar automatiskt de senaste företagsnyheterna som matchar användarens prenumeration. Den utvecklade prototypen kan förbättras både i funktion och design, men har uppfyllt behovet för denna rapport som avser att undersöka möjligheten att utöka det personliga nyhetsflödet i My Site med en sådan komponent. Eftersom projektmedlemmarna saknade förkunskaper i SharePoint och .NET utveckling innebar projektet nya utmaningar. Studenterna lärde sig att arbeta i SharePoint miljö samt i Microsoft Visual Studio för .NET utveckling.

Full textInformation retrieval has achieved great significance in form of search engines for the Internet. Retrieval systems are used in a variety of research scenarios, including corporate support databases, but also for the organization of personal emails. A current challenge is to determine and predict the performance of individual components of these retrieval systems, in particular the complex interactions between them. For the implementation and configuration of retrieval systems and retrieval components professionals are needed. By using the web-based learning application Xtrieval Web Lab students can gain practical knowledge about the information retrieval process by arranging retrieval components in a retrieval system and their evaluation without using a programming language. Game mechanics guide the students in their discovery process, motivate them and prevent information overload by a partition of the learning content
