Abstract:
Heinz bodies (HBs) are known to interfere with automated hematology in cats, particularly with the white blood cell (WBC) count. We evaluated the influence of feline HBs on the complete blood count (CBC) results obtained using a flow cytometry–based analyzer. We retrospectively selected cats with circulating HBs and reviewed the results of their CBCs, including red blood cell (RBC) indices, basophil/lobularity (Baso) WBC count (WBCB), peroxidase (Perox) WBC count (WBCP), and cytograms. Based on the presence or absence of HB-related artifacts in their Baso cytogram, cats were grouped into Baso-HBs and HBs groups, respectively, for comparison. The WBCB and WBCP were compared to manual counts of WBCs carried out on blood smears at 400× (MC-WBC). We included 32 cats in our study: 9 of 32 were in the Baso-HBs group, and 23 of 32 were in the HBs group. Baso-HBs cats had a significantly increased HB percentage ( p < 0.001), WBCB ( p < 0.001), difference between WBCB and WBCP ( p < 0.001), lymphocyte count ( p < 0.001), mean corpuscular hemoglobin concentration ( p < 0.001), and difference between calculated and measured erythrocyte hemoglobin concentrations ( p < 0.001) compared to HBs cats. In Baso-HBs cats, the WBCB was significantly higher than the WBCP ( p = 0.02); no significant difference was detected between the WBCP and the MC-WBC ( p = 0.88). Evaluation of automated CBC results raised the suspicion of HB-related interference when using a hematology analyzer in cats; hence, blood smear examination remains essential in routine practice.

Abstract:
Introduction. The estimation of malaria parasite density using a microscope heavily relies on White Blood Cells (WBCs) counts. An assumed WBCs count of 8000/µL has been accepted as reasonably accurate in estimating malaria parasite densities due to the challenge to accurately determine WBCs count. Method. The study used 4944 pieces of laboratory data of consented participants of age group less than 5 years. The study compared parasite densities of absolute WBCs, assumed WBCs, and the WBCs reference values in Central Ghana. Ethical approvals were given by three ethics committees. Results. The mean (±SD) WBCs and geometric mean parasite density (GMPD) were 10500/µL (±4.1) and 10644/µL (95% CI 9986/µL to 11346/µL), respectively. The difference in the GMPD compared using absolute WBCs and densities of assumed WBCs was significantly lower. The difference in GMPD obtained with an assumed WBCs count and that of the WBCs reference values for the study area, 10400/µL and 9200/µL for children in different age groups, were not significant. Discussion. Significant errors could result when assumed WBCs count is used to estimate malaria parasite density in children. GMPD generated with WBCs reference values statistically agreed with density from the absolute WBCs. When obtaining absolute WBC is not possible, the reference value can be used to estimate parasite density.

Abstract:
The scintigraphy with radiolabelled autologous leukocytes (WBCs) is considered the gold-standard technique for imaging infections. Leukokit® is a commercially available, disposable, sterile kit for labelling WBCs ex vivo. In this kit, WBCs isolation from red blood cells (RBCs) was performed using poly(O-2-hydroxyethyl)starch (HES) as the RBCs sedimentation agent. Due to its poor availability, HES has been recently replaced by Gelofusine as the RBC sedimentation agent. The aim of this study was to compare the labelling efficiency and the diagnostic accuracy of WBCs labelled with Leukokit® with HES vs Leukokit® with Gelofusine. WBCs were isolated using HES or Gelofusine for 45 minutes and then purified from platelets (PLTs) and labelled with 1.1 ± 0.3 GBq of freshly prepared 99mTc-HMPAO. The following parameters were evaluated: the number and type of recovered WBCs, RBCs contamination, PLTs contamination, vitality of neutrophils, and chemotactic properties of neutrophils. Clinical comparison was performed between 80 patients (33 males; age 67.5 ± 14.2) injected with 99mTc-HMPAO-WBCs, using HES as the sedimentation agent, and 92 patients (38 males; age 68.2 ± 12.8) injected with 99mTc-HMPAO-WBCs using Gelofusine as the sedimentation agent. Patients were affected by prosthetic joint infections, peripheral bone osteomyelitis, or vascular graft infection. We compared radiolabelling efficiency (LE), final recovery yield (RY), and diagnostic outcome based on microbiology or 2-year follow-up. Results showed that HES provides the lowest RBCs and PLTs contamination, but Gelofusine provides the highest WBC recovery. Both agents did not influence the chemotactic properties of WBCs, and no differences were found in terms of LE and RY. Sensitivity, specificity, and accuracy were also not significantly different for WBCs labelled with both agents (diagnostic accuracy 90.9%, CI = 74.9–96.1 vs 98.3%, CI = 90.8–100, for HES and Gelofusine, respectively). In conclusion, Gelofusine can be considered a suitable alternative of HES for WBCs separation and labelling.

Purpose The elevated white blood cells (WBCs) count has been reported to be a predictor of cardiovascular diseases, diabetes, hypertension and metabolic syndrome. This study aims to determine the associations between WBCs count and obesity in apparently healthy young adults. Design/methodology/approach In this cross-sectional study, the authors evaluated the body mass index (BMI) in 392 apparently healthy young adults of both sexes. The WBCs count was measured using standard counter techniques. The inclusion criteria were the agreement to participate in the study, between 18 and 25 years of age, lack of self-reported diseases such as cardiovascular diseases, hypertension, kidney and infectious diseases. Findings According to the BMI classification, underweight and overweight or obesity were observed in 14.58 and 11.48 per cent of young adults, respectively. The mean WBC was 6.5 ± 1.5 (×10³ cells/µL). Higher values of WBCs were found in women than in men (p = 0.02). The young adults with higher BMI had a higher WBCs count. There was a positive correlation between WBCs count and weight and BMI (r = 0.19 and r = 0.22, p < 0.001, respectively). Research limitations/implications This research was a cross-sectional study. Future studies are suggested using longitudinal studies to examine more relationships between obesity and WBCs count in apparently healthy young adults. Practical implications The results of this study provide evidence for weight management in this age group to reduce diseases associated with increased WBCs count. Originality/value The WBCs count was related to increasing levels of BMI per cent 2 C even in the normal range.

Abstract:
During this short term study a total of 110 samples were collected from the selected individuals of study area. This study was aimed to count the White blood cells in the blood samples of individuals, who were mostly exposed to exhaust fumes (air pollution), like traffic constables, cooks and shopkeepers. For this purpose blood samples were taken into an evacuee container having anticoagulant Ethylene diamine tetra-acetic acid (EDTA) so to prevent coagulation of blood, for the count of WBCs, heamocyto-meter was used. During the study the body mass index and blood pressure was also measured by digital blood pressure measuring apparatus. Samples were observed under the microscope. It was found that 15% (13,133±2544.081) sampled individuals had increased WBCs and 85% (7821±1482.76) normal WBCs. In female samples 50% (11900±2151.27) were observed with above range of WBCs and other 50% (8540±844.39) with normal range of WBCs count, non-had WBCs count lower than the normal WBCs. Mostly had normal and few had above the range. One way ANOVA has been used to analyze connection of exposure to air pollution with counts of circulating white blood cells.

Abstract:
Introduction White blood cells (WBCs) are immunity cells which fight against viruses and bacteria in the human body. Microscope images of captured WBCs for processing and analysis are important to interpret the body condition. At present, there is no robust automated method to segment and classify WBCs images with high accuracy. This paper aims to improve on WBCs image segmentation and classification method. Methods A triple thresholding method was proposed to segment the WBCs; meanwhile, a convolutional neural network (CNN)-based binary classification model that adopts transfer learning technique was proposed to detect and classify WBCs as a healthy or a malignant. The input dataset of this research work is the Acute Lymphoblastic Leukemia Image Database (ALL-IDB). The process first converts the captured microscope images into HSV format for obtaining the H component. Otsu thresholding is applied to segment the WBC area. A 13 × 13 kernel with two iterations was used to apply morphological opening on image to ameliorate output results. Collected cell masks were used to detect the contour of each cell on the original image. To classify WBCs into a healthy or a malignant category, characteristics and conditions of WBCs are to be examined. A transfer learning technique and pre-trained InceptionV3 model were employed to extract the features from the images for classification. Results The proposed WBCs segmentation method yields 90.45% accuracy, 83.81% of the structural similarity index, 76.25% of the dice similarity coefficient, and is computationally efficient. The accuracy of fine-tuned classifier model for training, validation and test sets are 93.27%, 92.31% and 96.15% respectively. The obtained results are high in accuracy and precision are over 96% and with lower loss value. Discussion Triple thresholding outperforms K-means clustering in segmenting smaller dataset. Pre-trained InceptionV3 model and transfer learning improve the flexibility and ability of classifier.

Abstract:
White Blood Cells (WBCs) analysis is an important procedure to detect diseases is that closely related to human immunity system. Manual WBCs analysis is laborious and hence computer aided system (CAD) is a better option to alleviate the shortcoming. Since conventional segmentation-classification approach is tedious to configure, a Convolutional Neural Network (CNN) become recent trend for WBCs classification. Previously, there are many works proposed for WBCs identification. However, the models that can be generalised to works well among various datasets is remain vague. In this paper, an analysis of various CNN models which are simple Alexnet, embedded friendly Mobilenet, inception based Googlenet, systematic architecture VGG-16 and skip connection based model (Resnet & Densenet), are tested with three major WBCs datasets (Kaggle, LISC and IDB-2). From the rigorous experiments, it can be concluded that simple CNN model of Alexnet performs well across all three datasets with 98.08% accuracy on Kaggle, 96.34% accuracy on IDB-2 and 84.52% on LISC. This outcome can be utilise as a basis to improve the CNN classification model that can be generalize to works under various WBCs datasets.

Purpose: 99mTc-HMPAO radiolabeled autologous leukocyte scintigraphy is routinely used clinically for infection imaging. Leukocytes are mostly separated via sedimentation. It is unknown whether leukocytes are clearly separated by sedimentation or selectively labeled. Therefore, in this study, the blood cell numbers were investigated after leukocyte radiolabeling to identify the cells strongly radiolabeled by 99mTc-HMPAO. Methods: This study was performed with leftover blood samples of the patients who underwent 99mTc-HMPAO scintigraphy at Chonbuk National University Hospital (2018-2019). The blood of 22 patients was drawn for 99mTc-HMPAO scintigraphy. WBCs were separated via conventional sedimentation at our clinic and radiolabeled. The concentration of cell components was determined using an automatic hematology analyzer. The cells in the final sample injectate sample were separated using Histopaque and counted with a dose calibrator. Results: The average numbers of RBCs, WBCs, and PLTs in the final injection sample were 79 ± 33, 23.26 ± 11.95, and 229.5 ± 206.57 x 103/μL, respectively. The PLT number was almost 10-fold the number of WBCs. The number of RBCs was nearly 3-fold higher than WBCs [RBC/WBC ratio = 4.67 ± 3.58, and PLT/WBC ratio = 10.65 ± 12.46]. Following Histopaque separation, the activity of each layer showed 99mTc-HMPAO labeling of WBC > RBC > PLT in order. The total activity/cell numbers of WBCs, RBCs and PLTs were 0.016 ± 0.010, 0.005 ± 0.005 and 0.003 ± 0.002, respectively (p > 0.05). Conclusion: Although the numbers of RBCs and PLTs were highly increased after sedimentation, their individual cellular activity was lower than that of WBCs. 99mTc-HMPAO was more selective to WBCs than RBCs or PLTs. In conclusion, a higher number of WBCs were radiolabeled compared with RBCs and PLTs.

A proliferação de dispositivos móveis e de redes sem fio tem encorajado um crescente interesse em sistemas e serviços baseados na localização de dispositivos portáteis, especialmente em ambientes internos, como edifícios e residências. A localização de um dispositivo portátil é um parâmetro crítico em aplicações baseadas no contexto, as quais requerem grande precisão na estimativa de localização. Entretanto, projetar e desenvolver sistemas de localização em interiores, com crescente precisão na estimação e decrescente custo de instalação, é um problema desafiador. Uma abordagem bastante interessante para satisfazer os requisitos de baixo custo consiste em utilizar as infra-estruturas existentes de redes locais sem fio (WLAN) no padrão IEEE 802.11, que já estão instaladas em muitos ambientes. A maioria das abordagens para localização usando WLAN propostas na literatura é baseada em técnicas probabilísticas, que apresentam bom desempenho e estão cada vez mais populares. Estas técnicas usam um mapa com a informação da potência recebida do sinal (RSSI) juntamente com a freqüência de presença de sinal coletada de múltiplos pontos de acesso Wi-Fi, em diferentes localizações físicas no ambiente. Porém, a informação sobre freqüência de presença de sinal pode ser muito ruidosa devido à natureza imprevisível das falhas de transmissão, as quais podem ocorrer decorrentes de diversos fatores. Este trabalho propõe um novo sistema de localização Wi-Fi, o WBLS (Wireless Based Location System), que não considera a informação sobre freqüência de presença de sinal no processo de estimação, visando eliminar os ruídos a ela associados. O WBLS explora o fato da potência do sinal Wi-Fi variar com a localização e usa um HMM descrito em um grafo onde os nós representam as localizações e as arestas, as probabilidades de transição em função da topologia do ambiente e das velocidades esperadas de um pedestre portando um dispositivo móvel. Investiga-se em que situações a eliminação da informação sobre freqüência de presença de sinal devido a seus ruídos associados aumenta a exatidão da estimativa de localização, apesar do descarte da informação em si. Os experimentos realizados demonstram que a característica mais importante do WBLS é uma particular robustez ao lidar com desligamentos de pontos de acesso, os quais podem ocorrer sem nenhum aviso ou previsão em um ambiente onde pouco controle se tem sobre sua infra-estrutura.
The proliferation of mobile computing devices and wireless networks has fostered a growing interest in location-based systems and services for Portable Wireless Devices, specially in indoor environments. The location of a handheld device is a critical parameter in context-aware applications, which require high degree of accuracy of location estimation. However, designing and developing indoor location systems with increasing estimation accuracy and decreasing cost installation is a challenging problem. A very interesting approach to reach low-cost requirements consists in using the pre-existing IEEE 802.11 wireless local area network (WLAN) infrastructure that is already installed in many places. Most of the WLAN indoor location approaches proposed in the literature are based on probabilistic techniques which show good performance and are becoming increasingly popular. Such approaches use a map of received signal strength information and signal presence frequency collected from multiple Wi-Fi access points at different physical locations in the environment. However, the signal presence frequency information can be very noisy due to the unpredictable nature of transmissions failures, which can be caused by several factors. This work proposes a new probabilistic-based Wi-Fi location system, WBLS (Wireless Based Location System), which doesn\'t take the signal presence frequency information into account in the estimation process, in an attempt to eliminate its associated noise. WBLS exploits the fact that Wi-Fi signal strength vary with location, and uses an HMM on a graph of location nodes whose transition probabilities are a function of the building\'s floor plan and expected speeds of a pedestrian holding a portable device. We investigate if eliminating signal presence frequency information due to its associated noise increases the accuracy of the location estimation, despite the amount of information about the signal presence that is discarded. Experiments show that the most important feature of WBLS is a particular robustness while dealing with access points shutdowns that may happen without any warning in an environment where there is little control over the infrastructure.

Advances in Information and Communication Technology (ICT) and the drastic growth of the Internet over the past decade have considerably changed the environment for end users of computer systems. Therefore, it is important to review how user satisfaction is measured with information systems, especially in a web-based environment. User satisfaction has become one of the most important measures of the success or effectiveness of information systems; however, there is a need of better understanding of its structure and dimensionality in the web environment. The objective of this research was to develop and validate an instrument for measuring user satisfaction in a web-based environment. In this research, three commercial websites were taken as representatives of Web-Based Information Systems (WBIS) for e-services. This study involved testing the validity and reliability of the end-user satisfaction over a set of heuristics, based on responses from users of three e-service websites. This thesis explores not only the role of user satisfaction but also the role of aesthetics and other factors, which play a significant role in service oriented websites. In the past, satisfaction was thought to originate from the effectiveness and efficiency of a product and less from its aesthetic qualities. This theory is beginning to change as numerous studies have indicated the importance of visual aesthetics in web design. In service interfaces, aesthetics (the perceived visual appeal and appropriateness of an object) has shown to correlate positively with many aspects of usability and emotional satisfaction.

A conservação da biodiversidade é uma das questões mais urgentes e importantes da atualidade. Pesquisadores da biodiversidade usam modelos de distribuição de espécies para fazer inferências sobre ocorrência e localização de espécies. Estes modelos são úteis para políticas de conservação de biodiversidade. Ferramentas para modelagem de distribuição de espécies usam grandes conjuntos de dados de biodiversidade que estão globalmente distribuídos, podendo estar em diferentes plataformas computacionais, o que dificulta seu acesso e manipulação. A comunidade científica precisa de infraestrutura onde pesquisadores de biodiversidade possam colaborar e compartilhar modelos, dados e resultados, e estejam aptos a reproduzir experimentos de outros pesquisadores. Neste contexto uma Arquitetura Orientada a Serviços (SOA) que suporta a colaboração em uma rede de modelagem de distribuição de espécies na Web é apresentada. Este ambiente computacional baseia-se em um catálogo de experimentos de modelagem e em um conjunto de serviços web geoespaciais, o Web Biodiversity Collaborative Modelling Services WBCMS.
Biodiversity conservation has become a most urgent and important theme at present. Biodiversity researchers use species distribution models to make inferences about species occurrences and locations. These models are useful for biodiversity conservation policies. Species distribution modelling tools use large biodiversity datasets which are globally distributed, sometimes in different computational platforms, and are hard to access and manipulate. The scientific community needs infrastructure in which biodiversity researchers can collaborate and share models, data, results, as well as reproduce experiments from others researchers. In this context, we present a Service-Oriented Architecture (SOA) that supports the collaboration in species distribution modelling network on the Web. This computational environment is based on a Catalogue of modelling experiments and on a set of Geoweb services, the Web Biodiversity Collaborative Modelling Services WBCMS.

Abstract:
Patientnära instrument kan i vissa vårdsituationer ge snabba analysresultat på ett kostnadseffektivt sätt. En viktig patientnära analys är differentialräkning av leukocyter, som visar fördelning och utmognad av dessa blodceller. Störningar i bildningen av leukocyter sker vid infektioner och hematologiska sjukdomar. Med manuell mikroskopi räknas leukocyter i ett blodutstryk färgat med May-Grünwald Giemsa. En kvantitativ och kvalitativ bedömning utförs av alla blodceller. Advia 2120 utför differentialräkning av leukocyter genom cytokemiska reaktioner och ljusspridning i olika vinklar vid belysning av celler. Celler identifieras baserat på kärnkonfiguration, granulering och cellstorlek.Hemocue White Blood Cell (WBC) Diff har en charged coupled device (CCD) kamera som scannar celler i en mikrokyvett. Genom beräkning av optisk densitet från pixlars gråskalor och jämförelse med referensceller identifieras celler. Syftet med studien var att jämföra analysresultat gällande differentialräkning av leukocyter i venöst blod mellan Hemocue WBC Diff och Advia 2120 samt i kapillärt blod mellan Hemocue WBC Diff och manuell mikroskopi. Provmaterialet bestod av 62 venösa prover från patienter och 20 kapillära prover från friska vuxna personer. Resultatet vid jämförelse mellan Hemocue WBC Diff och Advia 2120 visade en korrelation på 0,99 vid beräkning av totala antalet leukocyter, neutrofila granulocyter och lymfocyter. En större spridning av resultat sågs vid beräkning av monocyter, eosinofila- och basofila granulocyter. Resultatet vid jämförelse mellan Hemocue WBC Diff och manuell mikroskopi visade en korrelation på 0,95 för neutrofila granulocyter och 0,84 för lymfocyter. För de andra celltyperna var korrelationen dålig.Hemocue WBC Diff fungerar bra vid beräkning av leukocyter på friska personer. Instrumentet är dock under utveckling och förbättringar på vissa parametrar kan behövas.

На основі аналітичних даних щодо діяльності досліджуваного підприємства запропоновано відкриття тваринного комплексу як стратегію його розвитку. У першому розділі представлені теоретичні основи та наукові підходи проекту для розвитку підприємства на основі врахування факторів, які впливають на перспективи розвитку компанії. У другому розділі здійснено аналіз поточного стану агропромислового підприємства на основі кількісних та якісних показників його функціонування. А також вплив внутрішнього та зовнішнього середовища, У третьому розділі на основі досліджених науково-теоретичних підходів до формування проекту розвитку підприємства та здійсненого аналізу поточного стану агропромислового підприємства, його внутрішнього та зовнішнього середовища розроблено бізнес-план проекту компанії, за допомогою якого підприємство знизить рівень залежності сезонної дохідності Спираючись на отримані результати, сформовано комплекс практичних пропозицій щодо розробки та реалізації проекту розвитку досліджуваного підприємства.
В работе рассмотрены теоретические и методические основы разработки проекта предприятия агропромышленной отрасли. Рассмотрены вопросы формирования проекта предприятия как одного из существенных компонентов усовершенствования экономической деятельности предприятия и реализации в современных условиях хозяйствования. На основе аналитических данных о деятельности исследуемого предприятия предложено открытие животного комплекса как стратегию его развития. В первой главе представлены теоретические основы и научные подходы проекта для развития предприятия на основе учета факторов, влияющих на перспективы развития компании. Во второй главе проведен анализ текущего состояния агропромышленного предприятия на основе количественных и качественных показателей его функционирования. А также влияние внутренней и внешней среды, В третьем разделе на основе исследованных научно-теоретических подходов к формированию проекта развития предприятия и анализа текущего состояния агропромышленного предприятия, его внутренней и внешней среды разработан бизнес-план проекта компании, с помощью которого предприятие снизит уровень зависимости сезонной доходности. Опираясь на полученные результаты, сформирован комплекс практических предложений по разработке и реализации проекта развития изучаемого предприятия.
The theoretical and methodical bases concerning development of the project of the enterprise of agro-industrial branch are considered in the work. The issue of forming the project of the enterprise as one of the essential components of improvement of economic activity of the enterprise and realization in modern conditions of managing is considered. On the basis of analytical data on the activities of the studied enterprise, the opening of the animal complex as a strategy for its development is proposed. The first section presents the theoretical foundations and scientific approaches of the project for enterprise development based on factors that affect the prospects of the company. The second section analyzes the current state of the agro-industrial enterprise on the basis of quantitative and qualitative indicators of its operation. As well as the impact of internal and external environment, In the third section, based on the researched scientific and theoretical approaches to the formation of the enterprise development project and the analysis of the current state of the agro-industrial enterprise, its internal and external environment, the company's business plan is developed. Based on the obtained results, a set of practical proposals for the development and implementation of the project of development of the researched enterprise has been formed.

In the frame of the European Water Framework Directive (WFD, 2000/60/EC), a macroalgae based index (CARLIT) was applied along the Sicilian coastal water bodies (WBs) in order to assess for the first time their ecological status and collect accurate information on the distribution and abundance of shallow-water communities, especially of those most sensitive. The ecological quality ratio values, sensu WFD, showed “high”/“good” levels in all WBs with lushy forests of Cystoseira amentacea except two with “moderate” level due to the presence of stress-tolerant species related to local factors.

The aim of this paper is to perform segmentation of white blood cells (WBCs) using blood smear images with the help of image processing techniques. Traditionally, the process of morphological analysis of cells is performed by a medical expert. This process is quite tedious and time consuming. The equipments used to perform the experiments are very costly and might not be available in all hospitals. Further, the whole process is quite lengthy and prone to error easily because of the lack of standard set of procedure. Hence there is a need for innovative and efficient techniques. An automated image segmentation system can make the blood test process much easier and faster. Segmentation of a nucleus image is one of the most critical tasks in a leukemia diagnosis. In this work, we have investigated and implemented image processing algorithms to segment cells. The proposed model detects WBCs and converts cell images from RGB to HSV color space using Otsu thresholding. The resultant image is then processed with the morphological filter because the segmented image contains noise which affects the system performance. Lastly, the Marker-based watershed algorithm is implemented in which specific marker positions are defined. The proposed model is tested on publically available ALL-IDB2 dataset. The system’s performance was overall examined and resulted in 98.99% overall precision for WBC segmentation.

Abstract:
Adhesion of leukocytes (WBC's) to vascular endothelial cells (EC's) is a component of inflammation, thrombosis and atherosclerosis. The purpose of this study was to assess the effect of WBC adhesion on the EC contribution to these pathologic events. Human WBC's were isolated and co-incubated with cultured human umbilical cord EC's. Supernatants and cell lysates (4 wells × 2) were obtained at 0.5,1,2, and 4 hours of incubation. EC's and WBC's (5 × 105) were-incubated alone or in combination. Supernatants and cell lysates were assayed for leukotriene B4 (LTB4), the thromboxane metabolite thromboxane B2 (TXB2) and the prostacyclin metabolite 6-keto prostaglandin FT alpha (6 keto) by RIA. Cell lysates were analyzed for cell associated procoagulant activity (PCA) by an APTT procedure, for plasminogen activator inhibitor (PAI) by an amidolytic assay and for IL-1 by a T-cell co-stimulator assay. Cellular and supernatant LTB4 was unmeasurable for both WBC's and WBC/EC cultures. WBC TXB2 showed a time dependent elevation which was unaffected by EC's. IL-1 activity was measurable at 2 hours and reach 14 U/ml in WBC's and 6 U/ml in EC/WBC cultures. Co-incubation of WBC's with EC's induced a 200% increase in both supernatant and cell associated 6 keto concentrations compared to EC's incubated alone. EC's and WBC's produced no PCA when incubated alone. PCA activity of the EC/WBC co-cultures was measurable at 2 hours and was 300-1500 U/ml after 4 hours. Coincubated EC's had a 50% decrease in cell PAI, suggesting an increased release of inhibitor from the cells. The prostacyclin and PAI release -along with the delayed expression of PCA activity are responses similar to those expected after EC exposure to cytokines. A source of these cytokines appears to be the WBC's which secreted measurable amounts of IL-1. WBC released IL-1 was sufficient to induce biochemical changes in EC's which can stimulate coagulation (PCA synthesis), inhibit fibrinolysis (PAI release), and enhance inflammation (prostacyclin synthesis). These results suggest that the release of WBC IL-1 can be sufficient to produce pro-thrombotic and inflammatory changes in EC's which are similar to those observed with the addition of exogenous IL-1 to EC cultures.

Insulator-based dielectrophoresis (iDEP) is known as a powerful technique for separation and manipulation of bioparticles, using arrays of insulating posts and external electrical field. In this research, we utilized numerical simulation to study, in detail, the Joule heating which is one the most important phenomena in iDEP technique specially related to bioparticles separation and manipulation in physiological samples. Although Joule heating has been observed in both electrode-based and insulator dielectrophoresis, its effect is more significant in iDEP since higher electric potentials are required in this technology. As a result of the external electrical field, the temperature gradients would create conductivity, permittivity, viscosity and density local gradients in the solution, and consequently cause bulk fluid forces and fluid motion, known as electrothermal flow (ET). These flow circulations can cause unpredicted behavior of the device and even cause problems due to clogging. Moreover, the temperature rise due to the Joule heating could threaten the cell viability. In this study, we are going to develop a robust numerical model for predicting the flow behavior in the existence of external electric field and determining the temperature and velocity profile which can determine the cell viability and clogging problem in iDEP microdevices. The developed numerical tool was used based on the properties of circulating tumor cells (CTCs) and White blood cells (WBCs) and their separation.
