Об’єкт дослідження – агреговані анонімізовані повідомлення з чату користувачів Африканського маркетплейсу. Предмет дослідження – методи машинного, глибинного навчання та ембедингу: W2V, NLP, кластеризація, LSTM, RNN. Мета роботи – створити продукт, проаналізувати алгоритми необхідні для його створення. Метод дослідження – розгляд та аналіз методів машинного та глибокого машинного навчання. Проведений аналіз отриманих результатів, виконано порівняльний аналіз розглянутих методів машинного навчання. Шляхи подальшого розвитку предмету дослідження – збільшити кількість вхідних даних.

Abstract:
This document describes study of an active star forming region in our galaxy (the Milky Way) known as W22. Physical conditions in these regions can help us in understanding star formation processes in the universe and hence the structure and evolution of the universe. Zeeman effect measurements in 18 cm OH absorption line were carried out in order to estimate the line of sight magnetic field strength in the molecular cloud associated with this star forming region. Other physical parameters such as hydrogen column density, optical depth, critical magnetic field were also determined from these measurements. The region was mapped at 18 cm and the distribution of molecular gas within this star forming complex was described.

La presente investigación detalla un plan de marketing que busca evaluar el lanzamiento de una nueva marca de accesorios para canes en Lima Metropolitana, cuya propuesta de valor radica en la calidad, moda, y responsabilidad social contra el maltrato y abandono de canes y medio ambiente, a través de la venta por medio de una plataforma de comercio electrónico donde se ofrecerán experiencias convenientes, informativas y personalizadas. La investigación de mercado se basó en la investigación exploratoria y concluyente, para la cual se realizaron entrevistas de profundidad a expertos, así como en una encuesta a compradores potenciales. El objetivo fue estimar la demanda potencial, medios de consumos de publicidad, canal de venta, frecuencia de compra y conocer el grado de aceptación de la propuesta de valor de I Wuv You, para determinar nuestro mercado objetivo. Este está comprendido por personas habitantes en los distritos seis y siete en Lima Metropolitana y pertenecientes a los NSE A y B, pensadores e interesados en la calidad, moda y responsabilidad social. El objetivo de marketing para I Wuv You es introducir una nueva marca en la categoría de accesorios de canes, con una nueva propuesta que logre en el plazo de cuatro años el liderazgo dentro de la categoría. La estrategia genérica elegida para este proyecto es el de enfoque en diferenciación, debido a que nos orientamos a un segmento específico y los productos que I Wuv You ofrecerá serán percibidos como exclusivos, diferenciándose de la competencia por su preocupación social referente a los canes y al medio ambiente. La fijación de precios será con base en el valor percibido; para ello, se ha establecido un programa en la comunicación para incrementar la percepción de valor en la mente de los compradores. El análisis económico determinó que la implementación de este plan de marketing es viable, poco riesgoso y que genera utilidades desde el primer año.

Projeto de Graduação apresentado à Universidade Fernando Pessoa como parte dos requisitos para obtenção do grau de Licenciada em Fisioterapia
Objetivo: Estudar o efeito de três técnicas de alongamento: alongamento estático (AE), alongamento estático mais vibração de corpo inteiro (AEV) e alongamento de cadeia estática (CE) na acuidade propriocetiva do joelho, nomeadamente no senso da posição articular (SPA). Metodologia: foi realizado um estudo experimental, randomizado, e controlado, com 41 adultos saudáveis (25 mulheres) distribuídos aleatoriamente por quatro grupos: grupo de AE (5 repetições de 60 segundos); grupo de AEV (5 repetições de 60 segundos, 30 Hz a baixa amplitude); grupo de CE (5 minutos de alongamento); grupo de controlo (GC) (5 minutos de repouso). Realizou-se uma avaliação de SPA do joelho antes e depois de cada protocolo (45º para a flexão e extensão do joelho). Resultados: não foram encontradas alterações significativas (p>0.05) para o SPA do joelho em nenhum dos grupos estudados. Conclusão: a aplicação das três técnicas de alongamento estudadas não altera significativamente a acuidade propriocetiva do joelho.
Objective: to study the effect of three stretching techniques: static stretching (SS), static stretching plus whole-body vibration (SSWBV) and static chain stretching (SC) in the knee proprioceptive acuity, namely in the joint position sense (JPS). Methods: A randomized controlled trial was conducted with 41 healthy adults (25 women) randomly assigned to four groups: SS group (5 repetitions of 60 seconds); SSWBV group (5 repetitions of 60 seconds, 30 Hz to low amplitude); SC group (5 minutes stretching); and a control group (GC) (5 minutes rest). A knee JPS evaluation was performed before and after each protocol (45º for knee flexion and extension). Results: no significant changes (P> 0.05) were found for knee JPS in any of the studied groups. Conclusion: the application of the three stretching techniques studied does not significantly alter the knee proprioceptive acuity.

As ferramentas de bioinformática tem sido amplamente utilizada para o melhor entendimento de diversos microorganismos. Neste trabalho foram realizados três estudos utilizando estas ferramentas para avaliar diferentes questões biológicas. No primeiro estudo realizou-se uma caracterização molecular de 57 sequências do gene pol, provenientes de pacientes infectados pelo HIV-1 de Salvador, Bahia, Brasil. Para identificar os subtipos e formas recombinantes do HIV-1 circulante na cidade de Salvador foi realizado análises filogenéticas, e através do algoritmo do banco de dados Stanford HIV resistance as mutações associadas à resistência aos ARVs foram detectadas. Entre as 57 sequências analisadas foram identificados neste estudo 45 (77,2%) pertencem ao subtipo B, 11 (21,0%) recombinantes BF e uma (1,8%) do subtipo F1. Além disto, uma alta frequência de eventos de recombinação entre os subtipos B e F foram detectados com 5 padrões de recombinação, duas intergênicas e três intragênicas, mostrando uma alta diversidade. As mutações encontradas com uma maior prevalência foram: I54V (PI) em 7,0%; M184V (NRTI) em 14,0% e K103N (NNRTI) em 10,5% das sequências analisadas. Estes resultados contribuem para traçar o perfil da epidemiologia molecular e diversidade do HIV-1 em Salvador. O segundo estudo avaliou a filodinâmica do HIV-1 em pares de mãe e filho infectados, e em diferentes fases da infecção, três pares na fase aguda e um na fase crônica, e que apresentavam sequências de diferentes tempos. Para este fim foi realizado inferências filogenéticas bayesianas, onde a hipótese do relógio molecular e de diferentes crescimentos populacional foram testadas. Não foi possível observar uma diferença entre a dinâmica da população viral da mãe e a encontrada no filho. Porém, quando observamos o crescimento populacional e o tamanho da população efetiva, ao longo do tempo, sequências provenientes de pares em fase crônica da infecção tem um crescimento mais constante, enquanto as sequências dos pares na fase aguda da infecção se observa uma dinâmica das populações virais, provavelmente devido à pressão do sistema imune e a não adaptação destes vírus. No terceiro estudo, 104 sequências do genoma completo do WNV, disponíveis no Genebank, foram estudadas para identificar a região genômica que apresenta máximo poder interpretativo para inferir relações temporais e geográficas entre as cepas do vírus. Alinhamentos de cada gene foram submetidos à avaliação do sinal filogenético através do programa TREEPUZZEL. As regiões NS3 e NS4 apresentaram um sinal filogenético acima de 70%, sendo as regiões mais indicadas para construção filogenética. Além disto, árvores bayesianas foram inferidas utilizando as regiões NS3, NS5 e E, onde os clados das árvores NS3 e NS5 apresentaram um maior suporte e estrutural temporal geográfica, diferente da região E. Estes achados mostram que os genes NS3 e NS5 são os mais indicados para análises filogenéticas. Neste trabalho foi demonstrando o uso de ferramentas de bioinformática para a melhor caracterização da diversidade, epidemiologia molecular, dinâmica populacional e determinação das relações temporal e geográfica dos vírus.
The bioinformatics tools have been widely used for better understanding of several microorganisms. Here three studies were performed using these tools to answer different biological questions. In the first study, it was conducted the molecular characterization of 57 HIV-1 pol gene sequences from infected patients from Salvador, Bahia, Brazil. To identify the HIV-1 subtypes and recombinants forms, phylogenetics analyses were performed and the Stanford HIV resistance Database were used to analyze the antiretroviral susceptibility. Among all analyzed sequences, 45 of them were (77.2%) subtype B, 11 (21.0%) were BF recombinant and one sequence was (1.8%) subtype F1. Furthermore, a high frequency of recombination events between subtypes B and F was detected with five different patterns: two intergenic and three intragenic. The mutations found with higher prevalence were: I54V (PI) in 7.0%; M184V (NRTI) in 14.0% and K103N (NNRTI) in 10.5% of the analyzed sequences. These results contribute for the knowledge of the molecular epidemiology and diversity of HIV-1 in Salvador. The second study have evaluated the HIV-1 phylodynamics in mother and child infected pairs in different stages of infections: three pairs acutely infected and one chronically infected. Phylogenetic inference was performed using the Bayesian framework were the molecular clock and different population growth models hypothesis were tested. We did not find any difference of the population dynamics between mother and child. However, when observing the population growth and the effective population size through time, the chronically infected pair sequences showed a constant growth, while the acutely infected pair sequences showed a more dynamic population growth, probably due to the immune system selective pressure. In the third study, 104 WNV full genome sequences were selected from Genbank, to identify the best genomic region, which could provide the maximal interpretative power to infer temporal and geographic relationships among the virus strains. The phylogenetic signal was evaluated using the TREEPUZZEL program. The results showed that the NS3 and NS5 regions are the best ones to infer phylogeny since their phylogenetic signal was higher than 70%. Furthermore, Bayesian trees were constructed using the NS3, NS5 and E regions, and the NS3 and NS5 tree clades showed a higher support and a temporal geographic structure, different from the E region. These findings show that the NS3 and NS5 genes are the most informative genes for phylogenetic analyses. These studies demonstrated the use of bioinformatics tools for the better characterization of the virus diversity, molecular epidemiology, and population dynamics.

Deux agents phytopathogenes , un geminivirus, celui du nanisme du ble (vnb=wdv), et un organisme de type mycoplasme (otm=mlo), a l'origine de la phyllodie de la feve (fbp) ont ete soumis a des etudes au laboratoire et au champ. Les etudes de laboratoire consistent en la transmission des phytopathogenes par insectes en utilisant diverses plantes hotes et la detection des agents dans les insectes et dans les plantes inoculees. Les etudes ecologiques et biologiques comprennent: cycle biologique, longevite, preference pour les plantes-hotes, comportement et oviposition, en examinant six especes de cicadellides, euscelidius variegatus kbm, psammotettix alienus (dahlbom), neolimnus aegyptiacus (mats. ), rhytidodus decimusquartus (schrank), recilia sp. (edwards) et anaceratagallia sp. Les resultats ont conduit a prouver, pour la premiere fois, l'existence du vnb en france et sa transmission par p. Alienus. Divers aspects de la transmission de cette virose ont ete etudies. . .

Abstract:
Les aînés sont ceux qui manifestent le plus la peur du crime même s'ils font partie du groupe le moins à risque d'être victimisé. Ce paradoxe tend à disparaître lorsqu'on introduit le concept de l'inquiétude face à la victimisation en prenant en compte les dimensions émotionnelle, cognitive et behaviorale. À notre connaissance, il existe un seul outil validé mesurant ce concept: le Worry about victimization (WAV, de Williams et al., 2000). Le but de cette étude est de traduire en français et valider cet outil permettant de répertorier les insécurités des aînés en lien avec la victimisation criminelle. Le WAV a été traduit de l'anglais vers le français, selon une approche de traduction inversée parallèle, par une équipe de traducteurs et chercheurs, et cette version fut critiquée et révisée, lors de focus groups d'aînés, afin d'en améliorer sa compréhension. Par la suite, le questionnaire fut administré à 350 participants de trois villes québécoises. Les qualités métrologiques de la version française sont comparables à la version américaine. Nous avons produit une version abrégée et validée qui se prête bien à une utilisation auprès d'une population âgée francophone québécoise.

The aim of this thesis is to examine the theoretical commitments informing W.V. Quine’s rejection of alethic modality and to advance a Quinean response to Saul Kripke’s arguments in support of modal metaphysics. The novelty of this thesis consists in it being the first detailed explanation of how Quine’s arguments against modality are situated within his system and informed by his epistemological and ontological views and the first adequate study of the epistemological and metaphysical criticisms Quine would advance against Kripke’s defence of modality. The Quinean response to Kripke presented here is guided by four tenets that Quine takes to be central to the current scientific worldview and which he consequently adopts as the guiding methodological constraints of his own project: empiricism, regimentation, physicalism and simplicity. I explain how Quine’s referential opacity and mathematician-cyclist arguments against modality hang together with the rest of his philosophical project; and I show that while these arguments may seem unpersuasive and easily refuted by Kripke when taken in isolation, they are quite powerful when understood within the context of Quine’s entire system and seen in light of his guiding methodological constraints. By the end of this thesis, it will be clear why Quine remains unconvinced by Kripke’s arguments in support of modal metaphysics and how his response to Kripke is grounded in his deepest methodological constraints. He ultimately rejects Kripke’s arguments because they conflict with the tenets he takes to be the deepest commitments of the scientific worldview. Quine’s arguments against modality must be understood within the context of his philosophical system as a whole and are best seen not as arguments to be met by Kripke on Kripke’s terms but as illustrations of why, from the standpoint of Quine’s project and the standards it adheres to (the standards of science, as Quine understands it), modality is a flawed and unnecessary addition to reconstructed scientific theory. While there may well be reasons for rejecting Quine’s views about modality – and even reasons that would compel Quine, on his own terms, to reject his views – I argue that Kripke does not provide them.

Abstract:
The effect of pH on fiber and paper properties, during beating and sheet formation, was investigated for three different pulps. The pulps were pH adjusted to four different pH levels between 3 and 9. Isotropic laboratory sheet were made of both unbeaten and beaten pulps. The beaten neutral sulfite semi-chemical pulp and bleached softwood kraft pulp were affected by changes in pH; bleached softwood kraft pulp in a minor extent due to less fiber surface charges. Compared to the other pH levels, pH 3 showed a lower fiber surface charge, water retention value, tensile index, tensile stiffness index, compressive index and edge crush resistance index. SEM pictures showed a denser network at pH 9 than for pH 3. This was seen for both neutral sulphite semi-chemical and softwood kraft pulp. The unbeaten pulps and beaten bleached hardwood kraft pulp were not affected by changes in pH. SEM pictures showed no difference in the fiber network for bleached hardwood kraft pulp. A mill trial, with neutral sulphite semi-chemical pulp, at pH levels between pH 4.8 and pH 5.6 was completed. No significant difference was seen for any mechanical property.

Abstract:
The diploma thesis aims on the matter of data hiding (steganography) in audio records. It describes general steganographic principles and aims further on the specifics of data hiding in audio records and also aims on the LSB steganographic technique and suitability of different types of audio records to be used with this technique. The thesis also describes a laboratory exercise focused on steganography in audio records and influence of related aspects on the final audio record which contains secret data. Further, the thesis describes a program that was created especially for the laboratory exercise, from the design, functional as well as implementation-related view. Within the exercise, students will be able to examine how the amount of data to be hidden in the audio record and different ways of data hiding will affect quality of the resulting audio record, and therefore they can become more familiar with the matter of steganography based on the LSB method used in audio records.

Les geminivirus sont cause de beaucoup de maladies sur les plantes cultivees. L'etude du virus du jaunissement et de la courbure des feuilles de la tomate (tylcv) a revele que ce dernier possede une seule molecule d'adn genomique, au contraire des geminivirus transmis par bemisia tabaci et caracterises par deux adn genomiques. La sequence nucleotidique complete de l'isolat de sardaigne (tylcv-s) comporte 2770 nucleotides. Des phases de lecture ouvertes sur les deux brins d'adn codent pour des proteines potentielles dont le poids moleculaire varie de 11 a 40 kda. L'adn clone et sequence est infectieux sur plantes de tomate et de tabac via agroinoculation. Sur des arns prepares a partir de tomates infectees au champ, l'etude des transcrits du tylcv-s, par northern blot, cartographie avec la nuclease s1 et cartographie des extremites 5' et 3', a revele la presence de quatre transcrits: deux d'entre eux sont colineaires au brin complementaire et permettent l'expression des orfs c1 a c4 et les deux autres transcrits, plus abondants, sont colineaires au brin viral, et permettent l'expression des orfs v1 et v2. Dans le but de lutter contre le tylcv, une combinaison des strategies arn antisens et ribozyme ont ete utilisees. Des arn antisens ou des arn antisens-ribozymes ont ete diriges contre la partie 5' terminale du transcrit colineaire au gene c1 essentiel pour la replication. In vitro, les ribozymes utilises, possedent une bonne activite catalytique sur le transcrit c1. Des plantes nicotiana benthamiana transgeniques exprimant ces constructions, presentent une bonne resistance a la maladie. Cette resistance est maintenue dans la descendance des plantes transgeniques, avec une absence de symptomes et d'accumulation d'adn viral lors d'une infection par le tylcv-s. En parallele, nous avons identifie et caracterise un nouvel isolat du virus du nanisme du ble en france (wdv-f) en utilisant une nouvelle methode (methode guanidinium/alcalin) pour purifier specifiquement l'adn des geminivirus sous forme simple et double brins a partir de plantes infectees. L'adn clone est infectieux sur plantes de ble (triticum aestivum) via agroinoculation. La sequence nucleotidique du clone infectieux du wdv-f a ete determinee et comporte 2750 nucleotides

Objetivo: verificar a eficácia de 3 técnicas de alongamento no aumento da flexibilidade da musculatura posterior. Metodologia: é um estudo experimental, controlado e randomizado, com 41 estudantes universitários distribuídos de forma aleatória em 4 grupos: Grupo de Controlo, Grupo de CM, Grupo de Alongamento+WBV e o Grupo de Alongamento. O protocolo da avaliação da flexibilidade o S&R e o YMCA S&R são realizados no início de no fim das intervenções. Resultados: na diferença entre o teste S&R (inicial e o final) e a na diferença entre o teste YMCA S&R (inicial e o final) verificaram-se alterações significativas em todos os grupos de intervenção exceto no grupo de controlo. Por fim ao comparar a diferença do teste S&R e a diferença do YMCA S&R em relação aos grupos de intervenção, o teste S&R, apresentou alterações significativas na relação do grupo de controlo e de alongamento+WBV (p = 0.011). Conclusão: neste estudo a técnica Alongamento+WBV foi a que mais eficácia demonstrou no aumento da flexibilidade da musculatura posterior.
Objective: to verify the effectiveness of 3 stretching techniques in increasing the flexibility of the posterior musculature. Methodology: It is an experimental, controlled and randomized study with 41 university students randomly distributed in 4 groups: Control Group, CM Group, Stretching Group + WBV and Stretching Group. The flexibility assessment protocol the S & R and YMCA S & R are carried out at the beginning of the end of the interventions. Results: The difference between the S & R test (initial and final) and the difference between the YMCA S & R test (initial and final) they were verified significant changes in all intervention groups expect in the control group. Finally, when comparing the S & R test difference and the YMCA S & R difference in relation to the intervention groups, the S & R test showed significant changes in the control group and the WBV + stretching ratio (p = 0.011). Conclusion: in this study, the group of Stretching + WBV technique was the one that showed the most effective for increasing the flexibility of the posterior musculature.

This dissertation seeks to address three research questions through the context of spatial associations between land use / land cover (LULC) and vector-borne infectious disease (VBID). These research questions are: (1) Do spatial associations exist between the linear (edge) density of LULC boundaries and VBID occurrence? (2) Do patterns of spatial associations repeat over time? (3) Do patterns of spatial associations repeat across space?

Understanding how LULC change influences disease emergence informs the prevention and mitigation of local disease outbreaks prior to transmission growth into regional epidemics or global pandemics. Close and frequent human contact with infected arthropod vectors near local-level LULC boundaries drives VBID emergence. Increasingly dense and fragmented LULC boundaries result from human activities in the expansion of urban, pastoral, and agricultural areas. Fragmentation increases the likelihood of pathogen spillover at local-level LULC boundaries, the human-physical interface. Unmitigated and uncontrolled local spillover events can grow in spatial scale and result in significant social and economic impacts. Measuring the human-physical interface to identify spillover hotspots prior to VBID emergence and increasing levels of disease transmission is paramount to protecting public health.

Methods that measure the human-physical interface influence our ability to identify areas with elevated risk of VBID emergence. Prior research used remote sensing, field research, or literature reviews to identify substantive associations between LULC and VBID emergence. The research within this dissertation focuses on the spatial association between the linear density of LULC boundaries and VBID occurrence through spatial statistical methods, to include Principal Component Analysis and negative binomial regression. Proportion abundance and patch density are supplemental landscape metrics that add context to linear (edge) density. Case studies involve West Nile Virus in the contiguous United States from 2003 through 2014 and Zika in Colombia, South America during 2016. The goal is a method that can make use of land development plans to identify areas that could experience elevated VBID occurrence.

Les Cucurbitaceae sont une famille de plantes représentée par une grande diversité d’espèces et cultivées dans toutes les régions du monde. Le melon, qui appartient à cette famille et au genre Cucumis, est particulièrement sensible aux Potyvirus, vis-à-vis desquels aucun allèle du facteur d’initiation de la traduction eIF4e n’a permis d’induire de résistance. Chez le concombre, une Cucurbitaceae du genre Cucumis, Amano et al. (2013) ont identifié un allèle du gène codant pour la protéine VPS4 (Vacuolar protein sorting protein 4), impliqué dans la résistance au Potyvirus Zuchini yellow mosaic virus (ZYMV). Dans la cellule, la protéine VPS4 appartient au complexe ESCRT (Endosomal sorting complexes required for transport) et est impliquée dans la déformation des membranes au niveau des endosomes pour former les corps multivésiculaires (MVBs). L’ESCRT est fortement conservée chez tous les organismes eucaryotes et bien décrite chez les levures et les mammifères. L’utilisation des membranes cellulaires par les virus à ARNsb(+) par le biais de l’ESCRT et de VPS4 est largement décrite pour les virus infectant les animaux, notamment pour les virus du papillome humain (VPH) et de l’immunodéficience humaine (VIH) . Ces dernières années, le rôle majeur du transport vésiculaire dans l'infection virale a également été mis en évidence chez les plantes. Les virus à ARNsb+ forment des vésicules et des quasi-organites, en intégrant les complexes de réplication virales (VRCs) et tous les éléments nécessaires à la réplication de l'ARN viral et à la traduction des protéines virales .Cette thèse se concentre sur deux aspects : (1) la caractérisation d’allèles du gène Vps4 impliqués dans la résistance récessive au Potyvirus Watermelon mosaic virus (WMV) chez le melon (2) et l’étude du rôle de l’ESCRT dans la formation des vésicules virales chez les Potyvirus. Dans le premier objectif, j’ai montré qu’une mutation ponctuelle sur le gène codant pour la protéine VPS4 induit la résistance au WMV chez l’accession de melon TGR-1551. Pour cela, j’ai utilisé un virus transformé avec l’allèle sauvage pour infecter l’accession TGR-1551. Cela a permis d’observer la restauration de la sensibilité chez l’accession résistante. De plus, l’ensemble des accessions portant cet allèle sont également résistantes au WMV. Ces résultats représentent la première caractérisation des bases moléculaires d’une résistance au WMV chez le melon. Associés aux résultats de Amano et al., (2013), ces travaux contribuent à l’identification d’un gène de résistance à plusieurs Potyvirus. Dans le second objectif, j’ai montré l’implication de l’ESCRT dans la réplication des Potyvirus. Par une approche de double hybride associé aux protéines transmembranaires, nous avons montré l’interaction de la protéine virale 6K2 avec les protéines CmVPS28, CmVPS2, CmVPS24 et CmVPS4 du melon. De plus, nous avons localisé l’interaction 6K2/VPS4 au niveau des vésicules virales dans la cellule de plante. L’ESCRT ayant été caractérisée uniquement au niveau des MVBs, la présence de VPS4 au niveau des vésicules virales, en particulier au niveau des chloroplastes, implique le détournement de l’ensemble du mécanisme ESCRT par le virus. Pris ensemble, ces travaux contribuent à une meilleure compréhension du mécanisme de formation des vésicules virales au cours d’une infection d’une cellule de plante par un Potyvirus. Ils offrent également l’opportunité d’identifier de nouveaux allèles apportant une résistance aux Potyvirus afin de développer de nouvelles variétés résistantes
Cucurbitaceae are a family of plants represented by a great diversity of species and cultivated crops all around the world. Within this family and in the genus Cucumis, the melon is particularly susceptible to Potyviruses, against which no allele of the translation initiation factor eIF4e gene has been able to induce a resistance. In cucumber, also a Cucurbitaceae in the genus Cucumis, Amano et al. (2013) identified an allele of the gene encoding for the VPS4 protein, which is involved in the resistance to the Potyvirus Zuchini yellow mosaic virus (ZYMV). In the cell, the VPS4 protein belong to the ESCRT complex (Endosomal sorting complexes required for transport) and is involved in the membrane deformation at the endosome level to create multivesicular bodies (MVBs). The ESCRT complex is highly conserved in all eukaryotic organisms and well described in yeasts and mammals. The use of cell membranes by ssRNA(+) viruses through ESCRT and VPS4 has been widely described for viruses infecting animals, including Human Papillomavirus (HPV) and Human Immunodeficiency (HIV) viruses. In recent years, the major role of vesicular transport in viral infection has also been demonstrated in plants. ssRNA(+) viruses form vesicles and quasi-organelles using cell endomembrane, integrating viral replication complexes (VRCs) and all the elements necessary for the replication of viral RNA and for the translation of viral proteins.This thesis focuses on two aspects: (1) the characterization of alleles of the Vps4 gene involved in melon in a recessive resistance to the Potyvirus Watermelon mosic virus (WMV) (2) and the study of the ESCRT involvement in the formation of viral vesicles in Potyvirus.In the first objective, I demonstrated that a point mutation in the gene encoding the VPS4 protein induces resistance to WMV in the melon accession TGR-1551. For this, I used a virus transformed with the wild type allele of Vps4 to infect the TGR-1551 accession. This allowed us to observe the restoration of the susceptibility in the resistant accession. In addition, all accessions from the natural diversity carrying this allele are also resistant to WMV. These results represent the first characterization of the molecular basis of resistance to WMV in melon. Combined with the results of Amano et al. (2013), this work is part of the identification of a gene for resistance to several Potyviruses.In the second objective, I demonstrated the involvement of ESCRT in the replication of Potyviruses. Using a yeast two-hybrid approach adapted to transmembrane proteins, we showed the interaction of the viral protein 6K2 with the CmVPS28, CmVPS2, CmVPS24 and CmVPS4 melon proteins. In addition, we localized the 6K2 / VPS4 interaction at the viral vesicle level in the plant cell. Since ESCRT has been characterized only at the level of MVBs, the presence of VPS4 at the level of viral vesicles, especially at the level of chloroplasts, implies the hijack of the entire ESCRT mechanism by the virus.Taken together, this work provides a better understanding of the viral vesicle formation mechanism during infection of a plant cell by a Potyvirus. It also offers the opportunity to identify new alleles providing resistance to Potyviruses in order to develop new resistant varieties

During 1984-85 I investigated population ecology and habitat relationships of river cooters (Pseudemys concinna) in the New River Gorge National River (NRGNR), West Virginia. Cooter colonies occurred in 3 pool habitats characterized by slow current velocities (x=0.22m/s), shallow water (0-2m), aquatic macrophyte beds, and basking sites. The estimated adult population during summer 1985 was 64 individuals. Six, 25%, and 35% of the captures at the three study sites respectively, were juveniles. Most hatchling cooters apparently overwintered in the nest and emerged in April. Juvenile growth was rapid and linear until 6 years of age. Adult female cooters were larger (P=0.03) and heavier (P=0.0004) than males. Adult cooters consumed mostly eelgrass (Vallisneria americanaElodea canadensis), although some crayfish remains were found in fecal samples. Juveniles consumed vegetation as well as invertebrates and fish. Cooters did not move out of the pool habitats in which they were marked. Movements within pool habitats were influenced by basking site availability and location, which varied with river flow fluctuations. Two radio-marked cooters wintered in shallow backwater channels. Potential factors limiting the population are collecting and high artificial summer flows which scour weed beds. Addition of basking sites may increase habitat suitability for cooters.

Abstract:
Purpose: To evaluate the effectiveness of the West Virginia Prescription Drug Abuse Quitline (WVPDAQ). Methods: Descriptive data and inferential analyses are provided for the period of operations from 9/11/2008 to 3/1/2010. Chi-square tests for Independence compared differences between callers completing at least one follow-up survey against callers completing only the baseline intake survey were observed. A Wilcoxon signed rank sum test tested differences between the scores of intake callers and follow-up callers at the one-week (n=177) and one-month (n=89) intervals. Results: A total of 1,056 calls were received, including 670 intakes, 177 1st follow-ups, 89 2nd follow-ups, and 36 3rd follow ups, and 84 caller satisfaction surveys. Chi-square analyses determined that callers who only completed the intake survey tended to have initiated drug use at a younger age ([less than or equal to] 39 years of age, [chi square]=7.63, p=.02). Longitudinal findings indicated significant self-reported declines in daily drug use (p<.0001), increased intentions to quit in the next 30 days (p<.0001), and declines in requesting a referral for treatment (p<.0001) at the one-month follow-up. Finally, approximately 19% (n=17) of callers reported obtaining a counseling/ treatment appointment as a result of calling the WVPDAQ at the one-month follow up. Conclusions: Preliminary results suggest the WVPDAQ has established itself as a meaningful resource to combat prescription drug abuse in West Virginia. Sustainable funding and greater integration of the WVPDAQ into existing and developing treatment infrastructure could provide more affordable options for the state of West Virginia and its citizens.

Internet-based self-management interventions have the potential to reduce the current disparity in access to psychological support for people with tinnitus. One example is the Tinnitus E-Programme, which was developed in the United Kingdom to support self-management in people with tinnitus. Although freely available online, there was little understanding of how the intervention is used, its active ingredients, how it works, the circumstances in which it works best, and whom it works best for. This PhD aimed to address these issues by evaluating and optimising the Tinnitus E-Programme. A systematic review of self-help interventions for tinnitus was conducted, which concluded that there was a need for further evaluations of unguided self-help interventions in UK populations. A mixed methods study explored past, current, and new users’ (n=40) views and usage of the Tinnitus E-Programme (1.0), demonstrating that it was acceptable to people with tinnitus. However, its implementation was limited by instances of poor usability, user engagement, and adherence to behavioural goals. Consistent with a person-based approach, the findings from this mixed methods study were used alongside evidence-based (i.e. systematic and literature reviews) and theory-based (i.e. behavioural analysis and logic modelling) approaches to develop the Tinnitus E-Programme 2.0. Think aloud interviews with 19 people with tinnitus evaluated this new version of the intervention and findings revealed that the Tinnitus E-Programme 2.0 was acceptable to its target users. The two primary research studies highlighted how users’ pre-existing beliefs regarding tinnitus and self-management, their perceptions of relevance, and the nature of tinnitus can influence users’ engagement with the Tinnitus E-Programme 1.0 and 2.0. Several cognitive factors (e.g. illness beliefs), behavioural factors (e.g. practicing relaxation), and behavioural determinants (e.g. motivation to practice relaxation) were identified by users to explain how changes in intervention outcomes may occur. Further development and implementation work is needed that introduces and evaluates additional intervention content and design features, and explores how the intervention can fit into current clinical service models for tinnitus. Future evaluation work should test the hypothesised mechanisms of impact and contextual factors proposed in this work, and assess the acceptability and feasibility of procedures for subsequent randomised controlled trials that will assess the efficacy of the intervention.

Psychological treatment is effective at alleviating tinnitus distress when delivered by a psychologist. In the United Kingdom (UK), best practice guidance for tinnitus advises that audiologists should deliver Cognitive Behavioural Therapy (CBT) and other counselling techniques where psychologists are unavailable. However, there is a lack of evidence for the effectiveness of audiologist-delivered psychological tinnitus treatment, and no consensus among clinicians and researchers on what form, if any, this should take. Furthermore, training in CBT and other counselling techniques is not a part of standard audiologist training in the UK. The aims of this PhD were to define the components and measure the effects of an audiologist-delivered psychological tinnitus treatment for people with tinnitus. Potential treatment components to include were identified in two studies. The first study was a comprehensive scoping review in which components were extracted from 64 records on psychological tinnitus treatment delivered by psychologists. The components were predominantly extracted from studies of CBT. The second study was a Delphi survey of a panel of 39 patients and clinicians who were asked to rate the importance of 160 different treatment components for an audiologist-delivered psychological tinnitus treatment. These data were used to inform the development of a treatment manual, underpinned by a cognitive behavioural model of tinnitus distress. The manual comprised tinnitus education, psychoeducation, relaxation, graded exposure, thought challenging, physical exercise, sleep hygiene, an introduction to acceptance and defusion techniques, and sound enrichment, which could be implemented flexibly according to patient need. To measure the effects of the psychological tinnitus treatment, appropriate outcome domains and instruments were identified by applying template analysis to the treatment manual. Potential domains were selected from a list constructed by grounded theory using existing questionnaires. Outcome instruments were identified if they were responsive to therapeutic change and possessed good content validity and internal consistency. It was predicted that the treatment would affect tinnitus-specific emotional impact and negative thoughts. Based on current evidence, the Tinnitus Functional Index (TFI) and the Tinnitus Cognitions Questionnaire (TCQ) were identified to best measure these domains. A feasibility randomised controlled trial (RCT) was conducted over six months to determine the feasibility of a definitive RCT of audiologist-delivered psychological tinnitus treatment. Nineteen patients were recruited to the trial. Attrition rates were high, though all patient withdrawals occurred before their first appointment. The trial indicted that a definitive RCT of audiologist-delivered psychological tinnitus treatment is not feasible due to poor site compliance with returning questionnaires, and low recruitment rates in smaller trial sites. Patients and audiologists were invited to attend post-treatment semi-structured interviews to explore treatment fidelity and experience. Patients reported that the treatment was acceptable and effective in reassuring them about their tinnitus. Audiologists reported focusing on psychoeducation with limited use of specific techniques such as thought challenging and graded exposure. Patients and audiologists thought that the TFI and TCQ comprehensively measure the important tinnitus domains and are useful for planning treatment and encouraging discussion about patients’ negative thoughts. A lack of confidence due to the brevity of training was given by audiologists as a reason they avoided implementing certain treatment components.

Given an integer matrix A, there is a unique matrix S of a particular form, called the Smith Normal Form, and non-unique unimodular matrices P and Q such that PAQ = S. It is often the case that these matrices P and Q will be used for further calculation, and as such it is desirable to find P and Q with small entries. In this thesis we address the problem of finding such P and Q with small entries, in particular in the case where A is a diagonal matrix, which arises as a final step in many published algorithms. Heuristic algorithms are developed which appear to do well in practice and some theory is developed to explain this behaviour. We also give an account of the implementation of an alternative algorithm which bypasses this intermediary diagonal form. The basic theoretical development of this is work by Storjohan.

Abstract:
Gap-induced pre-pulse inhibition of the acoustic startle (GPIAS) is a behavioural test for tinnitus in animals. It relies on a short gap in a continuous background noise which provides a cue to the loud startling stimulus which follows. As a result, gap conditions demonstrate an inhibition of the response to the startling stimulus compared to no-gap conditions. The disrupting effect of tinnitus on the normal GPIAS has been shown in a number of species, including in humans. Such disruption was originally thought to be caused by tinnitus ‘filling in’ the gap, but recent studies have challenged this explanation. Preliminary work in humans measuring the eye blink reflex showed gap detection deficits in tinnitus subjects, but the underlying mechanisms of this effect are unclear. The eye blink response has a relatively long latency (>40ms) and therefore is not a simple primary reflex, nor is it specifically related to the auditory system. In small rodents the acoustic startle is measured by the whole body response involving axial muscles but in larger animals like the guinea pig this response habituates very quickly. Thus here a variation of the GPIAS method is used in which the acoustic startle is measured in guinea pigs using the simple pinna reflex. This reflex has been used to provide evidence of tinnitus in guinea pigs and postulated that it might be possible to use a similar method to obtain objective evidence of tinnitus in humans. The post-auricular muscle reflex (PAMR) is the human analogue of the pinna reflex and may represent a metric for an objective tinnitus test. The PAMR is a short-latency (15-18ms) response that involves only two or three synapses in the brainstem and provides a much tighter link between auditory input and motor output than the eye blink reflex. However, gap-induced pre-pulse inhibition (PPI) of the PAMR has not previously been demonstrated. This question is one of the main objectives examined in this thesis, using measures taken in guinea pigs and in humans, with and without tinnitus. The work reports two feasibility experiments and two-hypothesis testing studies in which I have sought to optimise the stimulus parameters for maximising the magnitude of the PAMR, and reflects on the challenges of working at the first translational gap developing adequate animal models of human hearing-related problems.

Abstract:
Hearing loss is a prevalent condition that can have negative implications on the day-to-day lives of those with hearing loss, and their communication partners. Several reviews have identified numerous questionnaires that explore the impact of hearing loss, with little consensus among researchers as to their preference. One reason for this is a lack of an agreed framework concerning exactly what is the impact of hearing loss, and what is important to those who live with the condition. To address this gap in understanding, the experiences of those living with hearing loss or with someone that has hearing loss was investigated using various systematic research methods and tools. A comprehensive review of published studies exploring the impact of hearing loss on those with hearing loss and/or their communication partner was conducted. An inductive (data-driven) synthesis of the research evidence led to the development of two frameworks; one for each perspective. Each framework comprised of three higher-level ‘supra-domains’ (i.e. Auditory, Social and Self) which capture the broader implications of hearing loss, and numerous lower-level ‘domains’ and ‘subdomains’ that tap into distinct aspects of life. The frameworks were evaluated for face-validity in focus groups with hearing aid users, communication partners and audiologists. Hearing aid users and communication partners were then asked to rank the domains in order of importance based on their experience of living with hearing loss. The domain ‘communicating’ was ranked most important from both perspectives. A subdomain mutual to both frameworks, ‘raising the volume of the television/radio’ was explored further in a case-control study. The preferred TV volume across 42 couples where one partner was a hearing aid user and the other was a non-hearing aid user was measured. A mean difference in volume preference of 6.3dB was observed between groups across the TV programmes viewed by all participants. The hearing aid prescription of hearing aid users was not verified. Future research should explore how domains in the frameworks are currently being measured and addressed in clinic. Also, data-driven methods should be applied to identify coping mechanisms adopted by people with hearing loss and their partner to accommodate for the diverse range of hearing-related difficulties depicted in the frameworks.

Introduction Adaptive behaviour has become increasingly important in the assessment of intellectual disabilities (ID), but is also useful in terms of developing individual behavioural goals. Although measures of adaptive behaviour are widely used, there is little agreement on how to conceptualise the construct and how this may vary cross-culturally. Researchers have previously noted methodological and validity difficulties with the assessment of ID. This research aimed to clarify the construct of adaptive behaviour and consider how to measure this with d/Deaf adults with suspected ID. Methods A systematic literature review, identification of critique of current measures, and gaps within the literature formed the structure of a new item pool. An expert panel (N = 13) were consulted about the usefulness and relevance of these items through a Delphi consensus methodology. Based on the feedback obtained throughout the research process, the design was modified from pursuing a statistics-driven approach to item refinement, to using mixed-methods to clarify issues of construct validity before the content could be further addressed. The second round of the Delphi comprised a feedback report, concluding with a working definition of adaptive behaviour. Participants were asked to comment upon the findings, and provide additional responses to develop a normal base standard of adaptability of a Deaf adult of average functioning. Results The first round of the Delphi consensus yielded wide variation in item ratings. A thematic analysis of the questionnaire comments identified two overarching themes related to the way in which adaptive behaviour was conceptualised: ‘structure’ and ‘content’. The theme of structure contained sub-themes of ‘assessment’, ‘language’, and ‘repetition’ which are arguably common to all psychometric development. The ‘content’ factors pertained to ‘accessibility’, ‘developmental factors’, and ‘cultural differences’, highlighting differences in the expression of adaptive behaviour based on the unique experiences of d/Deaf people. The second round of the Delphi procedure elicited feedback on a working definition of adaptive behaviour generated from the analysis, showing that the adaptive behaviour of d/Deaf people may be conceptualised differently, particularly in hearing contexts where there are differential communication demands. These research findings have been summarised to form initial guidelines for the assessment of adaptive behaviour in Deaf people. Discussion This research provided some insight into how adaptive behaviour may be measured with Deaf people. Limitations of this research included not being able to generate consensus through the Delphi methodology used and, due to the newly developed understanding of adaptability for this population advanced throughout the research process, it was not possible to subject the items from the proposed scale to further psychometric testing. However, recommendations for further research were made in terms of expanding and validating this preliminary work with a sample of Deaf adults.

Vibrations are often experienced in the workplace and may influence performance and executive function. Research has shown that vibrations may have an affect effect on drowsiness and tests related to inhibitory control. Previous work investigating whole body vibrations (WBV) and their effect was evaluated to inform the decisions for this study. WBV effects on cognitive abilities were examined and the different tests used in these studies were identified and compared. Electroencephalogram (EEG) and event related potentials (ERP) were selected to be used to measure inhibitory and cognitive processes. The N2 ERP, which reflects inhibitory control processes, was examined as well as the dominant frequency of the Fourier fast transform (FFT). A total of 94 participants between the ages of 18-55 (Mage = 20.49 SDage = 1.68) completed this study (51 female, 38 male and 5 with no gender listed). A go/no-go task was used to elicit the N2 ERP after WBV and a simultaneous EEG recording while the participants experienced WBV was used to gather the needed data. Stimulus frequencies used for the N2 ERP included 15 Hz, 20 Hz, and 40 Hz. During the simultaneous recording stimulus frequency varied every 30 seconds by 10 Hz from 20 Hz to 110 Hz. Data were analyzed using both a linear mixed effects model for normally distributed data and a generalized linear mixed effects model for data taken as percentages. It was hypothesized that there would be an effect on performance as measured in the raw go/no-go results, that this change in performance showing improved accuracy would be linked to inhibitory control, and be seen as a decrease in the magnitude of the N2 ERP. It was also hypothesized that the exploratory FFT portion of the study would produce a shift from a higher to a lower frequency in the dominant waveform . The results show that there were no main effects in either the behavioral performance or in the N2 ERP of the participants but that there was a significant interaction at 40 Hz with improved simple go trial activity and decreased no-go inhibition. The results also show that there was a statistically significant shift in neural oscillation activity but that this shift was not real-world relevant within the context of this study.

Abstract:
Water quality models in the Colorado River Basin have been developed for the basin, river, and individual reservoirs. They are used to support water quality programs within the basin. The models are periodically reviewed and updated to improve the accuracy of simulations. Improving the usefulness of the Lake Powell model, one of the key reservoirs in the basin, is the subject of this study. Lake Powell is simulated using a hydrodynamic and water quality model, CE-QUAL-W2. Previously the model has been used at Lake Powell to simulate hydrodynamics, temperature, and total dissolved solids with a reasonable degree of accuracy. An additional parameter, dissolved oxygen, will be added to the simulations and then calibrated with observed data to verify accuracy. Dissolved oxygen distributions in Lake Powell vary seasonally and change under different hydrologic cycles. They are a function of physical, biological, and chemical processes. Few measurements of these processes in Lake Powell exist. To compensate for the lack of data an empirical method of loading oxygen demand to the model is developed and tested. Observed limnological processes in the reservoir guide the development of the empirical methods. The methods are then tested in 16 year model simulations and compared with dissolved oxygen measurements from the 16 year period. By accurately reproducing the dissolved oxygen distributions the Lake Powell model will have improved accuracy and also broaden its usefulness.

Erwin Schulhoff (1894–1942) was a Czechoslovakian musician born in Prague, to a German-Jewish family, and whose life came to a premature end in 1942 at the Wülzburg concentration camp, near Weißenburg, Bavaria. Schulhoff’s life, compositional style, and two of his flute works are addressed in this dissertation: Sonata for flute and pianoforte, WV 86, and Concertino for Flute, Viola, and Double Bass, WV 75. Each work is considered as a discrete entity, and insight provided into the structure of the music; stylistic and compositional influences, form, phrase structure, and other aspects are discussed. The intended audience is the flutist seeking knowledge regarding the historical significance and performance of each piece. The analysis and summary of Schulhoff’s life and primary flute works will contribute to the understanding of performance scholarship of his music and provide a deeper understanding of the composer, from the perspective of musical and compositional style.

West Nile virus (WNV) belongs to the Flaviviridae family, a virus family of which many members are known as human pathogens. WNV has a worldwide distribution and strains that cluster in lineage II is endemic to sub-Saharan Africa. The complete nucleotide sequence of four lineage II West Nile virus strains, isolated in South Africa from patients with mild or severe WNV infections, were determined. Using a murine model, these strains had been shown to produce either highly or less neuroinvasive infections and induced similar genes to corresponding highly or less neuroinvasive lineage I strains. Nucleotide and amino acid sequence comparison between highly and less pathogenic lineage II strains demonstrated that the non-structural genes and in particular the gene coding for the NS5 proteins were the most variable. All the lineage II strains sequenced in this study were found to possess the E-protein glycosylation site previously postulated to be associated with virulence. Comparison of the signalase cleavage sites suggested that lineage II strains may be cleaved slightly more efficiently than lineage I strains in the C-prM junction, but less efficiently between prM and E genes. Relative to the highly neuroinvasive strains sequenced in this study major deletions were found in the 3’ noncoding region of 2 lineage II strains shown in previous studies to be either less- or not at all neuroinvasive. This is the first report of full genome sequences of highly neuroinvasive lineage II WNV strains. Currently available commercial WNV ELISA kits were developed with lineage I WNV strains and are expensive to use. For these reasons the development of a potential ELISA diagnostic assay based on the South African lineage II strain, H442, was envisaged. Such assay, if reliable and efficacious would be a useful tool towards WNV surveillance. The prM and E genes were selected to be expressed as recombinant antigens because of their co-expression nature and because the envelope protein is the principal target for neutralization. After cloning of the respective genes and verification of integrity, a mammalian expression system was utilized. Different mammalian cells and transfection media were tested and BHK 21 cells with SuperFect transfection medium were found to be best. Attempted expression of proteins was tested with immunofluorescent antibody testing as well as SDS-PAGE and Western blot analysis. Expression of recombinant WNV antigens were also tested in indirect and sandwich ELISA’s systems. It was however not possible to perform these two ELISA systems at a satisfactory level or clearly indicated if expression of proteins was successful.

As aves podem carregar um grande número de patógenos. As aves migratórias, por viajarem longas distâncias, são as principais responsáveis pela disseminação de agentes infecciosos. Entre os agentes, destacam-se os vírus, como por exemplo o retrovírus da Reticuloendoteliose aviária (REV), amplamente distribuído; e o vírus da febre do Oeste do Nilo (WNV), uma virose reemergente, com caráter zoonótico. Os principais sintomas da Reticuloendoteliose aviária incluem anemia, doença de Runting e síndrome não neoplásica aguda. Já o agente etiológico da Febre do Nilo Ocidental, é o Flavivirus West Nile (WNV).. As aves são seus hospedeiros definitivos e os humanos são hospedeiros acidentais, podendo manifestar quadro febril, e em menor porcentagem, meningite e encefalite. Mosquitos dos gêneros Culex e Aedes spp são os principais transmissores do vírus. Ao contrário do REV que não dispõe de evidências de sua circulação no Brasil, há evidências do WNV em aves e equinos e mais recentemente, em humanos. O objetivo desse trabalho foi investigar a presença do REV e do WNV em aves silvestres e de cativeiro da cidade de São Paulo e do Norte do estado do Pará. Sangue, soro e swab de cloaca foram coletados, totalizando mais de 1000 amostras. Através de técnicas moleculares foi possível detectar a presença do REV em 74 amostras (16%), todas do estado do Pará. O sequenciamento parcial dessas amostras e sua filogenia sugeriu que a migração de aves EUA-Brasil possa ter sido a rota utilizada. Através de ELISA anti-IgG de WNV, 4 amostras de São Paulo foram positivas. Apresentamos a primeira evidência do REV no país e sugerimos a presença do WNV no estado de São Paulo.
Birds can carry a large number of pathogens. The migratory birds are most responsible for the spread of infectious agents due to long distance travels. Among these pathogens, the most notable are viruses, such as the avian Reticuloendotheliosis retrovirus (REV), widely distributed; and the West Nile virus (WNV), a reemerging zoonotic disease. The main symptoms of avian reticuloendotheliosis include anemia, Runting\'s disease and acute nonneoplastic syndrome. The etiological agent of West Nile fever is Flavivirus West Nile (WNV). Birds are their definitive hosts and humans are accidental hosts, which generaly present febrile symptoms, but at less proportion,, meningitis and encephalitis. Mosquitoes of the genus Culex and Aedes spp are the main vectors of the virus. Differently from the REV that has no evidence of its circulation in Brazil, there is evidence of WNV in birds and horses and more recently in humans. The objective of this work was to investigate the presence of REV and WNV in wild birds and captive birds from the city of São Paulo and Northern from Pará State. Blood, serum and cloacal swab were collected, resulting in more than 1000 samples. Through molecular techniques it was possible to detect the presence of REV in 74 samples (16%), all from the State of Pará. The partial sequencing of these samples and their phylogeny suggested that the migration of US-Brazil may have been the route for the virus entry. Through anti-WNV IgG ELISA, 4 samples from São Paulo were positive. We present the first evidence of REV in the country and suggest the presence of WNV in the state of São Paulo.

One of the most important purposes of surface water resource management is to develop predictive models to assist in identifying and evaluating operational and structural measures for improving water quality. To better understand the effects of external and internal nutrient and organic loading and the effects of reservoir operation, a model is often developed, calibrated, and used for sensitivity and management simulations. The importance of modeling and simulation in the scientific community has drawn interest towards methods for automated calibration. This study addresses using an automatic technique to calibrate the water quality model CE-QUAL-W2 (Cole and Wells, 2013). CE-QUAL-W2 is a two-dimensional (2D) longitudinal/vertical hydrodynamic and water quality model for surface water bodies, modeling eutrophication processes such as temperature-nutrient-algae-dissolved oxygen-organic matter and sediment relationships. The numerical method used for calibration in this study is the particle swarm optimization method developed by Kennedy and Eberhart (1995) and inspired by the paradigm of birds flocking. The objective of this calibration procedure is to choose model parameters and coefficients affecting temperature, chlorophyll a, dissolved oxygen, and nutrients (such as NH4, NO3, and PO4). A case study is presented for the Karkheh Reservoir in Iran with a capacity of more than 5 billion cubic meters that is the largest dam in Iran with both agricultural and drinking water usages. This algorithm is shown to perform very well for determining model parameters for the reservoir water quality and hydrodynamic model. Implications of the use of this procedure for other water quality models are also shown.

Introduction: This work explored the concept of ‘empowerment’ in health care. A concept map of ‘empowerment’ was generated based on theories of linguistic analysis to locate aspects of empowerment in conversational data. The process of empowerment was understood within the theory of transformative learning as ‘perspective change’. A ten phase model of the transformative learning process was used to provide evidence of perspective transformation. The empowerment process was explored through the delivery of the intervention Video Interaction Guidance to 16 hearing families of pre-lingual deaf children. Methods: The parents of the deaf children and the intervention guide engaged in conversations reviewing video clips of the parent and child in interaction. These conversations were processed through a corpus analysis software programme to discern key extracts of the participants’ conversational data. These extracts were to subject to discourse analysis to find evidence of transformative learning. Analysis: Transformative learning was observed in 10 families. Transcript extracts representing the model of transformative learning were variable. Principles of conversation analysis were applied to explore the learning process in interaction. Participant speech was tagged to assess their level of engagement. Outcomes: The corpus analysis-driven tagging process offers an original approach to representing the key content of large sets of interview data but in this work, was limited in showing how the interaction created opportunities for learning. Transformative learning processes were variable and the ten phase model could not be characterised in terms of discourse features alone. The intervention encouraged critical reflection but warrants directed focus to achieve learning. The participants were effective in acting as co-collaborators in the intervention process. Discussion and concluding remarks: Models of empowerment must focus on the process, of the ways in which the guide and the participant create learning opportunities and evidence of this must be multimodal. Patient-centred interventions should be supported by an open, communicative relationship with the health service provider.
