Full textThe objective of this study is to develop a low environmental impact solution for the consolidation of partially submerged coastal areas. This solution, the formation of a limestone concretion based on seawater electolysis, relies on two main aspects: firstly, the efficient use of local resources through the valorization of inert construction waste (recycled aggregates (RA)); and secondly, the biomineralization process involving the hydration of CO2 by the enzyme carbonic anhydrase (CA) found in marine bacteria sampled from the Port of La Rochelle. Three major axes were studied to optimize the precipitation of a binder within the limeston concretion composed of CaCO3 and Mg(OH)2 (the calcareous deposit): the effect of RA dissolution in seawater, the application of cathodic polarization via seawater flow, and the study of CaCO3 bio-precipitation by CO2 capture (the role of CA) by marine strains. Seawater flow allowed the formation of a 200 cm3 agglomerate in 60 days at -500µA/cm², resulting in a growth rate of 3 cm3/day. A 10% increase in compactness was observed when the grid was buried (within the RA) either submerged or emerged. Seawater flow and the presence of RA favored the precipitation of CaCO3, particularly in the form of calcite, leading to an Mg(OH)2/CaCO3 ratio less than or equal to 1, whether under continuous or cyclic polarization. The excessive release of calcium and sulfate ions into solution due to the dissolution of the cementitious matrix within the RA could explain the increase in CaCO3. All strains bio-precipitated CaCO3 in their optimal medium and in the presence of natural seawater. Their production drastically decreased at 3% CO2 (atmospheric CO2 = 0.4%) and in the presence of leachate from recycled aggregates. At 3% CO2, the pH of the medium increased in the presence of the strains, which could indicate the activity of CA

Full textThe flow cytometry techniques allow the analysis and sorting of living biologic cells at rates above five for then thousand events per second. After a short review, we present in this report the design and development of a "high-tech" apparatus intended for research laboratories and the experimental results. The first part deals with the physical principles allowing morphologic and functional analysis of cells or cellular components. The measured parameters are as follows: electrical resistance pulse sizing, light scattering and fluorescence. Hydrodynamic centering is used, and in the same way, the division of a water-stream into droplets leading to electrostatic sorting of particules. The second part deals with the apparatus design at the "Commissariat à l'Energie Atomique" (C. E. A. ) and industrialised by "ODAM" (ATC 3000). The last part of this thesis work is the performance evaluations of this cytometer. The difference between the two size measurement methods are analyzed: electrical resistance pulse sizing versus small-angle light scattering. By an original optics design, high sensitivity has been reached in the fluorescence measurement: the equivalent noise corresponds to six hundred fluorescein isothiocyanate (FITC) molecules. Full textThis study is about the development of a new ring shear apparatus which is able to shear an undisturbed sensitive clay sample at large strains in test conditions equivalent to an undrained test, by the mean of a constant volume test. A series of tests were realised on samples of undisturbed sensitive clay cored with a large diameter sampler on the site of the Saint-Jude spread that occurred on May 10th 2010. The results of those tests show that the apparatus is working properly. The results are compared with results of triaxial compression test and direct simple shear test realised on samples from the same soil. The outcome of the comparison is that the ring shear test is the better of the three types of test to characterise properly the shear strength at large strains of the clays. It is also the only one of the three types of test that is able to reach the residual shear strength.

Full textMechanical ventilation induces cyclic changes in left ventricular stroke volume. These changes are related to the expiratory decrease in left ventricular preload following the inspiratory decrease in right ventricular filling and ejection. The magnitude of respiratory changes in left ventricular stroke volume reflects the sensitivity of the heart to the mechanical insufflation-induced changes in ventricular preload. In clinical practice, respiratory changes in left ventricular stroke volume can be assessed by analysis of the arterial pressure wave form (arterial catheter) or of aortic blood velocity (Doppler). In ventilated patients with acute lung injury, the respiratory changes in arterial pulse pressure predict the hemodynamic effects of positive end-expiratory pressure (Study # 1). In septic patients with acute circulatory failure, the respiratory changes in arterial pulse pressure (Study # 2) and in aortic blood velocity (Study # 3) predict the hemodynamic effects of volume expansion. These dynamic parameters are better predictors of fluid responsiveness than static indicators of right and left ventricular preload (Study # 4)

Full textThe incidence of cardiovascular disease differs between men and women. Symptoms, disease’s evolution, response to treatment and even care are different depending on the sex. Cardiac hypertrophy (HC) is the heart's response to sustained and significant hemodynamic overload. HC can be concentric (thickening of the walls) or eccentric (dilation of the chamber). Mechanisms involved in the sexual dimorphism of HC are still poorly understood. Our team is interested in a model of severe aortic valve regurgitation (RA) in rats. We have already shown that women’s heart has a more concentric remodeling, a better capillary density and a gene profile of mitochondrial function and energy metabolism closer to normal, compared to males. We were first interested in the involvement of sex hormones in this sexual dimorphism. Echocardiographic data and tissue analyze show that androgen deficiency in RA males reduces hypertrophic response, improves cardiac function and tends to improve survival. Estrogen loss is not associated with significant hypertrophic changes in RA females. We then studied the effect of valsartan, a treatment targeting the renin angiotensin system (RAAS), a highly activated hormonal pathway in HC. Inhibition of RAAS activity prevents concentric hypertrophy in female’s heart in the early weeks of RA, whereas it has little effect in RA males. Finally, we were interested in the morphological and functional evolution of the heart in RA rats. Echocardiographic analysis shows that RA females’ left ventricle rapidly loses contractile function, circumferential deformation capacity, and changes its normal elliptical shape, compared to males. The results presented here improve our understanding of sexual differences in HC.

Full textThe trained athlete is a physiological model of extreme cardiac adaptation for whom the distinction between adaptive cardiac remodeling induced by chronic exercise and certain early cardiomyopathies can be difficult to assess. Echocardiography is the first-choice imaging modality to evaluate the athlete’s heart at rest and during exercise. Semi-recent developments in speckle tracking and 3D ultrasound imaging have shown clinical interest in the echocardiographic description of the athlete’s heart. However, some technical aspects require further investigation. Moreover, current post-treatment tools provide only a partial analysis of cardiac hemodynamics and regional myocardial function. Using an experimental model mimicking stress echocardiography, we first demonstrated the validity of speckle tracking in comparison to sonomicrometry to measure regional deformation in a large range of deformation rates. Secondly, we studied in volunteers without heart disease (athletes and non- athletes) the reliability of a method to assess non-invasively the left ventricular (LV) systolic and diastolic intraventricular pressure gradients (IVPGs) based on post-processing of intracardiac flow velocity data acquired using color Doppler M-mode. This hemodynamic index was highly feasible and well correlated with LV suction. Analysis of IVPG measurements revealed inter-vendor variability which was mainly related to differences in color Doppler image resolution. Finally, using a multiparametric echocardiographic approach (speckle tracking, IVPGs, and 3D volumes), we studied the physiological relationship between the type of exercise training and the left and right cardiac remodeling among a Canadian football team followed longitudinally. The regional analysis of right ventricular (RV) morphological and functional changes induced by chronic exercise was performed using a new computational method based on 3D echocardiography that volumetrically parcellated the RV into three segments (apex, outlet, and inlet). In conclusion, our workhas shown that the echocardiographic post-processing tools studied for the global and regional analysis of cardiac function and morphology apply to the athlete’s heart and could be useful in the characterization of the exercise-induced cardiac remodeling
