Abstract:
Vitamin D (vit D) has anti-inflammatory properties and modulates lung growth, but whether vit D can prevent lung injury after exposure to antenatal inflammation is unknown. We hypothesized that early and sustained vit D treatment could improve survival and preserve lung growth in an experimental model of bronchopulmonary dysplasia induced by antenatal exposure to endotoxin (ETX). Fetal rats (E20) were exposed to ETX (10 μg), ETX + Vit D (1 ng/ml), or saline (control) via intra-amniotic (IA) injections and delivered 2 days later. Newborn pups exposed to IA ETX received daily intraperitoneal injections of vit D (1 ng/g) or saline for 14 days. Vit D treatment improved oxygen saturations (78 vs. 87%; P < 0.001) and postnatal survival (84% vs. 57%; P < 0.001) after exposure to IA ETX compared with IA ETX alone. Postnatal vit D treatment improved alveolar and vascular growth at 14 days by 45% and 25%, respectively ( P < 0.05). Vit D increased fetal sheep pulmonary artery endothelial cell (PAEC) growth and tube formation by 64% and 44%, respectively ( P < 0.001), and prevented ETX-induced reductions of PAEC growth and tube formation. Vit D directly increased fetal alveolar type II cell (ATIIC) growth by 26% ( P < 0.001) and enhanced ATIIC growth in the presence of ETX-induced growth suppression by 73% ( P < 0.001). We conclude that antenatal vit D therapy improved oxygenation and survival in newborn rat pups and enhanced late lung structure after exposure to IA ETX in vivo, which may partly be due to direct effects on vascular and alveolar growth.

Abstract:
The general (α,t)-path connectivity index of a molecular graph originates from many practical problems such as three-dimensional quantitative structure-activity (3D QSAR) and molecular chirality. It is defined as Rtα(G)=∑Pt=vi1vi2⋯vit+1⊆G[d(vi1)d(vi2)⋯d(vit+1)]α, where the summation is taken over all possible paths of length t of G and we do not distinguish between the paths vi1vi2⋯vit+1 and vit+1⋯vi2vi1. In this paper, we focus on the structures of Polycyclic Aromatic Hydrocarbons (PAHn), which play a role in organic materials and medical sciences. We try to compute the exact general (α,3)-path connectivity indices of this family of hydrocarbon structures. Furthermore, we exactly derive the monotonicity and the extremal values of R3α(PAHn) for any real number α. These valuable results could produce strong guiding significance to these applied sciences.

Abstract:
Background: High prevalence of both vitamin D (vit. D) deficiency and obesity in world population as reported by many studies drown attention to establish the association and its direction between these two modifiable risk factors and their impact on health status which are still uncertain. Objective: To measure vit. D levels among a sample of obese women, describing some factors that may contribute for vit. D values in this sample and assess the correlation between vit. D levels and body mass index (BMI). Methods: A cross-sectional study was carried out among 100 obese women. Their age ranged between (20-50) years, during the period from 1st February to 1st August 2019. The sample was gathered from laboratory of private medical center at Palestine Street in Baghdad. vit. D was measured by (Ichroma vit D), which is a fluorescence immunoassay. Results: The mean±SD of vit. D values of the participants were found to be (16.05±6.9) ng/ml. Only two women from the sample had sufficient vit. D level of more than 30 ng/ml, (72%) of the participants found to have insufficient vit. D level 10-29.9 ng/ml and the remaining had deficiency in vit. D level <10 ng/ml. A significant association was found between vit. D levels and the age participants and beverages drinking (P≤0.01). Indirect correlation was found between vit. D values of the participants, their BMI values, and waist/ hip ratios and was significant with the last P≤0.012. Conclusion: No association was found between vit. D and BMI, but there is indirect correlation between vit. D and waist/hip ratio. Highest deficiency of vit. D among age group (30-39) years old and among those who were drinking carbonated beverage. Keywords: Obese, women, vitamin D, body mass index Citation: Abdulkader HD, Al-Saffar AJ. Abstract:
The purpose of this study was to investigate the effects of vitamin B6 (Vit B6) on oxidant and antioxidant status in streptozotocin-induced diabetic rats. Thirty-two Wistar rats were divided into four groups: control (C), control + Vit B6 group (C + Vit B6), diabetes (D), and diabetes + Vit B6 group (D + Vit B6). Vit B6 (4 mg/kg body weight) was administered in drinking water for 4 weeks after the induction of diabetes. Vitamin B6 reduced serum total cholesterol level in the C + Vit B6 (P< 0.01) and D + Vit B6 (P< 0.05) groups. Plasma and tissue malondialdehyde levels were reduced in the C + Vit B6 and D + Vit B6 groups. Whole blood glutathione peroxidase (GSH-Px) and erythrocyte superoxide dismutase (SOD) activities were higher in the D group (P< 0.05). GSH-Px and SOD activities were increased in C + Vit B6 group while these parameters decreased in the D + Vit B6 group. Paraoxonase and arylesterase activities were decreased in the D group while they were increased in C + Vit B6 and D + Vit B6 groups. The results of present study suggest that vitamin B6 supplementation might be a promising adjunctive agent for improving oxidative stress and metabolic disturbances and for preventing diabetic complications including atherogenesis.

Abstract:
Vitamin D (vit D) is widely known for its role in calcium metabolism and its importance for the bone system. However, various studies have revealed a myriad of extra-skeletal functions, including cell differentiation and proliferation, antibacterial, antioxidant, immunomodulatory, and anti-inflammatory properties in various cells and tissues. Vit D mediates its function via regulation of gene expression by binding to its receptor (VDR) which is expressed in almost all cells within the body. This review summarizes the pleiotropic effects of vit D, emphasizing its anti-inflammatory effect on different organ systems. It also provides a comprehensive overview of the genetic and epigenetic effects of vit D and VDR on the expression of genes pertaining to immunity and anti-inflammation. We speculate that in the context of inflammation, vit D and its receptor VDR might fulfill their roles as gene regulators through not only direct gene regulation but also through epigenetic mechanisms.

Abstract:
Objectives: Vitamin D (Vit D) deficiency has been identified as a global epidemic, sparking numerous studies into its more detailed effects on the body. In orthopedics, Vit D deficiency has been found to correlate with an increase in a variety of musculoskeletal injuries. Although current evidence suggests a connection between the number of musculoskeletal injuries sustained and an athlete’s vitamin D level, this correlation has not yet been thoroughly investigated in the collegiate athlete population. We hypothesized that lower levels of serum vitamin D would be associated with an increased number of musculoskeletal injuries and increased recovery time. Methods: A retrospective study was performed on 285 student athletes at the authors’ institution, a Division I university (Table 1). Serum 25-hydroxyvitamin D (25(OH)D) levels obtained on each athlete via chart review in conjunction with the athletic department and categorized into normal (≥32 ng/mL), insufficient (20 to 31 ng/mL) and deficient (≤19 ng/mL). Additional data collected included demographics, athletic performance, injury history and bone density. Results: 139/285 (48.8%) of athletes were Vit D insufficient, with an additional 50/285 (17.5%) being deficient (Table 2). For football players specifically (our largest cohort of athletes), 71/91 (78%) had a Vit D level below normal, with the average value falling at 24.5 ng/mL. For injury susceptibility, there was a 7% increase in injuries for those with a below normal Vit D. (40% vs 33%). For recovery length, athletes with a below normal Vit D took twice as long to recover (23.7 days vs 10.2 days). Conclusions: A large percentage collegiate athletes have insufficient or deficient levels of Vit D. Our results also suggest that a lower Vit. D level correlates to an increase in the number of musculoskeletal injuries sustained and an increase in recovery length. [Table: see text][Table: see text]

Abstract:
Hyperparathyroidism (HPT) can occur after gastric bypass because of the alteration in vitamin D and calcium absorption. Adequate serum vitamin D concentrations have not been clearly defined in this patient population. Vitamin D (Vit D) and parathyroid hormone (PTH) were assessed 1 year after laparoscopic gastric bypass (LGB). The prevalence of HPT and Vit D deficiency were determined and their association was evaluated using Fisher's exact test. Ninety-three patients (aged 44 ± 1.1 years, 49.6 ± 0.67 kg/m2 body mass index, 79.6% female, 69.6% white) were evaluated. The prevalence of Vit D deficiency (less than 20 ng/mL) and HPT (greater than 65 pg/mL) was 23.6 per cent (n = 22) and 25.7 per cent (n = 28), respectively. Among patients with HPT, only eight of 28 (28.6%) had Vit D deficiency, and of those with Vit D deficiency, only eight of 22 (36.4%) had HPT. There was a weak inverse correlation (r = –0.37) between PTH and Vit D. Blacks are at higher risk for Vit D deficiency. There was no significant association between Vit D deficiency and HPT, Vit D deficiency and Roux limb length, or HPT and Roux limb length. After LGB, Vit D deficiency and hyperparathyroidism occur commonly. Body mass index and Roux limb length are not associated with these two conditions, but racial differences do exist. There is a weak inverse correlation between Vit D and PTH. Further research is needed to elucidate the causes, treatments, and significance of HPT after LGB.

Abstract:
Background Deficiency of vitamin-D (Vit-D) was associated with poor survival outcome in several studies across different tumour types. The present study aims to assess the prevalence and prognostic value of Vit-D deficiency among breast cancer patients in a single institution in Saudi Arabia. Methods In this retrospective study, we screened patients who presented with non-metastatic breast cancer to King Abdullah Medical City, Saudi Arabia from June 2011 to December 2015. We checked baseline Vit-D level before starting systemic therapy in addition to other clinicopathological factors. Low Vit-D was defined as Vit-D level less than 30 ng /ml. The relations of Vit-D level (taking the median as the cutoff) with clinicopathological factors were assessed using Chi-Square test. Differences in survival outcome were compared using log rank test. Results We screened 340 patients with non-metastatic breast cancer. Baseline Vit-D levels were available for 189 patients. The median age was 50 years (range: 26- 86 years). Noteworthy, 169 (89.4%) of patients had Vit-D level <30 ng/ml with a median of 14.9 ng/ml (range: 4.0 - 45.0). Low Vit-D level (below the median) was significantly more common in premenopausal (p=0.011) and ER-negative patients (p=0.011). However, lymphovascular invasion (p=0.001), clinically (p=0.023) and pathologically positive axillary LNs (p=0.041) were linked with higher Vit- D level. After a median follow up period of 58.2 months, 14 patients died and 40 relapsed. The 5-year disease-free survival (DFS) rates was 74.8%. The 5-year DFS rate in patients with higher Vit-D level above the median was 78.8% compared to 71.1% in patients with lower Vit-D level with no statistically significance difference (p= 0.22). The 5-year overall survival (OS) rate was 90.2%. Meanwhile, no difference in 5-year OS rate in patients with higher and lower Vit-D levels (90.3% and 89.7% respectively, p=0.6). Conclusion Low Vit-D level was prevalent among the studied breast cancer patients. Low Vit-D level was associated with ER-negative phenotype and premenopausal patients. Baseline Vit-D level was not significantly linked with survival outcome.

Abstract:
1α,25-dihydroxyvitamin D3 (1,25(OH)2D3), the active metabolite of Vitamin D (Vit D), increases intestinal absorption of calcium and phosphate, maintaining a correct balance of bone remodeling. Vit D has an anabolic effect on the skeletal system and is key in promoting osteoblastic differentiation of human Mesenchymal Stem Cells (hMSCs) from bone marrow. In order to overcome some negative aspects connected with the use of MSCs from bone marrow, i.e. morbidity, pain and low yield of cells, other sources of MSCs have been examined. MSCs can be also isolated from the immature form of the tooth, the dental bud: Dental Bud Stem Cells (DBSCs) are adult stem cells that can effectively undergo osteoblastic differentiation. The aim of the project is to study the effect of Vit D on DBSCs, which represent a useful model to understand the anabolic activity of the bone cells, starting from very undifferentiated osteoblast precursors. Since MSCs adhesion to Extra Cellular Matrix (ECM) is still poorly characterized, as well as ECM proteins role in regulating osteogenic differentiation, we studied our cell model of DBSCs for the expression of Integrins and Cadherins as well as for ECM protein interaction. We demonstrated a functional osteogenic differentiation of DBSCs towards an osteoblastic phenotype, being incremented by the Vit D treatment. We observed that DBSCs treated with 1,25(OH)2D3, expressed increased levels of the main osteoblastic markers and formed mineralized matrix nodules in vitro. These results reflected the mesenchymal origin of DBSCs and confirmed their osteoblast-like features. Moreover, in order to elucidate the effect of the microenvironment on DBSCs behavior, we also analyzed αVβ3 integrin distribution and Focal Adhesion formation in cells seeded on surfaces with different coatings in the presence of Vit D. We observed that Vit D seems to induce the expression of focal adhesions in our cell model. Our observations suggest that Vit D acts directly on these cells, that can be considered osteoblast precursors, directing them to an increased bone formation.

Abstract:
If the litterature states reservations of the family caregivers, to ask for help, their psychic springsremain badly known. Nevertheless the sense of guilt was already located, as an obstacle at the request ofhelp, of the caregiver, but this factor was little explored in a research theme. This research bases on thehypothesis that the sense of guilt of the family caregiver, would represent the main brake, susceptible toprevent his request of help and support, in front of the Alzheimer's disease or the related disease, when thepatient lives at home. A sub-hypothesis aims at placing differently the sense of guilt of the familiy caregiver, according to its spouse's position, or more widely of descendant (child, son-in-law, daughter-in-law) in therelation of help. To test this hypothesis, thirty eight semi-directive conversations were realized andcompleted by the signing of the scales of attachment (RSQ), the caregiver (CRA), and depression (Beck). This research aims at clarifying foudations, and mechanisms of the sense of guilt of the family caregivers, byarticulating it in the problem of loss, which is heart of Alzheimer's disease or the related diseases. It thusgives a new perspective on the psychic work of the family caregiver, which deviates from its only accessunder the angle of the burden and the exhaustion, to envisage it in the light of the work of the pre-mourning,which appears at the keystone of the relation of help. Consequently, the research will follow the cycle of thedependence of the patient, to release in each of its stages, the incidences of the loss in the psychic andintersubjective space at the family caregiver, according to the nature of the links of attachment developpedwith the patient, but also with the family group. From there, we shall try to connect the main register ofelaboration of the loss, in a profile of singular caregiver in the relation of help, to light the links between hisdemonstrations of guilt and his demand of help

Drouin, Christian. "Vitamin D analogues via dynamic combinatorial chemistry." Thesis, McGill University, 2011. http://digitool.Library.McGill.CA:80/R/?func=dbin-jump-full&object_id=96890.

Full text
Abstract:
Calcitriol (1alpha,25-(OH)2 D3, 1,25D) is known for its calcium regulatory function, but also for being a powerful inhibitor of cell growth in a variety of normal and neoplastic cells. As such, it is a lead structure in the development of new cancer drugs. The goal of our project is to identify analogues of calcitriol which bind tightly to the vitamin D receptor (VDR) yet do not exhibit unwanted hypercalcemic effects. The development of novel analogues using conventional chemistry and dynamic chemistry was investigated. First, the numerous building blocks required for the exploration of the different libraries were created through the use of conventional techniques. We then developed different synthetic schemes based primarily on thiol alkylation and amide couplings, enabling rapid parallel synthesis of potential 1,25D analogues. Preliminary investigations allowed us to implement with our collaborators a set of biological assays to evaluate our compounds. Although some analogues appeared to bind to the VDR, it was found that none of the 8 analogues tested in the preliminary studies seemed to be vitamin D receptor (VDR) agonists. In a second part, our research focused on the study of dynamic combinatorial chemistry as a tool for the synthesis of 1,25D analogues. Our first dynamically-generated libraries were created via thioester exchange. It was found that the rate of thioester exchange was dependent on the nature of the thiols involved and the acyl portion of the thioesters. Aliphatic acid thioesters exchanged more slowly than aromatic acid thioesters. The use of branched (alpha-substituted) thiols also slowed the rate compared to primary ones. However, the use of large concentration of aliphatic or aromatic thiols could accelerate the thioester exchange. When thioesters were placed in presence of protein targets in a dynamic system, their inherent electrophilic nature rendered them prone to chemical decomposition (hydrolysis and acylation of protein nucleophilic residues) at basic pHs. Nonetheless, we created thioester libraries of up to 40 members from as few as 7 building blocks under near neutral conditions. We showed that these dynamic libraries ere influenced by the presence of proteins. We have not been able to confirm if thioester libraries were influenced exclusively by the VDR via its binding site. Some of our observations challenged the viability of the thioester exchange as a reversible process in the context of VD3 analogue synthesis. Finally, we created dithiol building blocks, amenable to disulfide exchange in dynamic libraries. Chemically stable, these disulfide entities easily generated libraries of at least 30 members from as few as 7 building blocks. Our preliminary disulfide libraries seemed uninfluenced by VDR's presence. These results still allowed us to understand better the characteristics and limitations of dynamic systems applied to the development of novel vitamin D analogues.

Abstract:
This study aimed to evaluate the percentage of Vitamin D deficiency in women’s in Erbil city, Kurdistan, Iraq by direct assessment and intervention, carried out in the department of nutrition and dietetics, at Cihan University, Erbil. The methods of work designed for females participants in different ages who visited health care centers and they were ready to take part in this study and filling the questionnaire, after they accepted and agree to participate then they have to do Vitamin D3 blood analysis test and depend on the result our team continue to contact with them and deliver brochures’ about Vit. D and follow up the cases one by one as much as possible especially those who had deficiency, our goal was to increase the awareness and they have to do second blood test to evaluate the progress and impact of the intervention. The main result of this study was, the percentage of deficiency more than (53%) of the participants, meanwhile many correlations was found between the health and nutritional situation and Vit. D deficiency. On the other hand, the intervention which was carried out to increase the awareness helped many females who had deficiency and accept to continue participating in the program, more than (83 %) of them they skipped the vit. D3 deficiency depend on the second blood test which was done after period between (6 - 8 weeks) only. So, we can conclude that, females in Erbil suffering from this nutrient deficiency because of many reasons, as one of the nutritional problems, and they need more and more efforts and programs to help them in their life to be in good health.

Abstract:
Vortex-Induced Motion (VIM) is another way to denominate the Vortex-Induced Vibration (VIV) in floating units. The main characteristics of VIM in such structures are the low aspect ratio (L/D < 4.0) and the unity mass ratio (m* = 1.0, i.e. structural mass equal water displacement). The VIM can occur in MPSO (Monocolumn Production, Storage and Offloading System) and spar platforms. These platforms can experience motion amplitudes of around their characteristic diameters. In such cases, the fatigue life of mooring and riser systems can be greatly reduced. Typically, the VIM model testing campaigns are carried out in the Reynolds range between 200,000 and 400,000. VIV model tests with low aspect ratio cylinders (L/D = 1.0, 1.7 and 2.0) and unity mass ratio (m* = 1.0) have been carried out at the Circulating Water Channel facility available at NDF/EPUSP. The Reynolds number range covered in the experiments was between 10,000 and 50,000. The characteristic motions (in the transverse and in-line direction) were obtained using the Hilbert-Huang Transform method (HHT) and then compared with results obtained in experiments found in the literature. The aim of this investigation is to definitely establish the similarity between the VIM and VIV phenomena, making possible to increase the understanding of both and, at same time, allowing some analytical models developed for VIV to be applied to the VIM scenario on spar and monocolumn platforms, logically under some adaption.

Abstract:
Autonomous marine vehicles (AMVs) are typically deployed for long periods of time in the ocean to monitor different physical, chemical, and biological processes. Given their limited energy budgets, it makes sense to consider motion plans that leverage the dynamics of the surrounding flow field so as to minimize energy usage for these vehicles. In this paper, we present two graph search based methods to compute energy optimal paths for AMVs in two-dimensional (2-D) time-varying flows. The novelty of the proposed algorithms lies in a unique discrete graph representation of the 3-D configuration space spanned by the spatio-temporal coordinates. This enables a more efficient traversal through the search space, as opposed to a full search of the spatio-temporal configuration space. Furthermore, the proposed strategy results in solutions that are closer to the global optimal when compared to greedy searches through the spatial coordinates alone. We demonstrate the proposed algorithms by computing optimal energy paths around the Channel Islands in the Santa Barbara bay using time-varying flow field forecasts generated by the Regional Ocean Model System. We verify the accuracy of the computed paths by comparing them with paths computed via an optimal control formulation.

Abstract:
Altering milk composition via transgenesis depends on three main factors. (1) The availability of an efficient regulatory sequences for targeting transgene(s) to the mammary gland; (2) a reliable in vitro model to test the expression of transgenes prior to their introduction to the animal genome; and (3) better understanding of the major factors which determine the rate of gene expression and protein synthesis. The current studies provide the necessary means and knowledge to alter milk protein composition via transgenesis. The following specific goals were achieved: a: Identifying regulatory regions in the b-lactoglobulin (BLG) gene and the cross-talk between elements which enabled us to construct an efficient vector for the expression of desirable cDNA's in the mammary gland. b: The establishment of a sheep mammary cell line that serves as a model for the analysis of endogenous and exogenous milk protein synthesis in the mammary gland of livestock. c: An accurate comparison of the potency of the 5' regulatory sequences from the BLG and whey acidic protein (WAP) promoters in directing the expression of human serum albumin (HSA) to the mammary gland in vitro and in vivo. In this study we have also shown that sequences within the coding region may determine a specific pattern of expression for the transgene, distinct from that of the native milk protein genes. d: Characterizing the dominant role of ECM in transgene expression in mammary epithelial cells. e: Further characterization of the BCE-1 enhancer element in the promoter of the b-casein gene as a binding site for the c/EBP-b and Stat5. Identifying its interaction with chromatin and its up regulation by inhibitors of histone deacetylation. f: Identifying a mechanism of translational control as a mediator for the synergistic effect of insulin and prolactin on protein synthesis in the mammary gland.
