Full textThe search for new compounds to prevent or attenuate skin aging is a priority in current research in cosmetics. In this context, Asian Hornet venom (Vespa velutina nigrithorax) has been studied as a particular source of potentially bioactive molecules for dermacosmetic interest.The first study focused on the implementation of a reliable venom extraction and sampling protocol. Then, the peptide - small molecules fraction was selected to evaluate, in comparison with crude venom, the presence of active molecules with respect to antioxidant, anti-microbial (C. acnes) and enzyme inhibition (tyrosinase, elastase, collagenase) activity in-tubo and in-cellulo. These studies led to the identification in crude venom, by UHPLC-ESI-QTOF-HRMS/MS, of one molecule responsible for antioxidant activity on HaCaT keratinocytes.In a second study, a peptidomic approach based on UHPLC-ESI-QTOF-HRMS/MS followed by statistical processing (PCA, PLS-DA) was applied to the differential study of venom, according to the collection period, castes and behavior. The latter aims at evaluating the influence of these different factors on the venom molecular heritage. At the same time, in a third study, a ligand/enzyme interaction screening approach by mass spectrometry on solid-supported elastase enzymes was developed. The aim of this method is to detect the presence of inhibitors or substrates in more or less complex fractions. Two hornet venom peptides presenting in the hornet venom were identified to be capable of interacting with the enzyme elastase. Their peptide sequences were then partially obtained by de novo sequencing

Full textMarine coccolithophores are microalgae known for their blooms visible from space. These photosynthetic microorganisms are presumed to resist to strong light radiation like UV A and B, what make them of great interest for dermo-cosmetic applications. The present study aimed to identify and lift the blocking points for metabolite production in intensified culture. A screening resulted in the selection of two productive species, among them Emiliania huxleyi, on which the effect of nutrient and light limitation on growth, photosynthesis and calcification was investigated in PBR. A kinetic analysis of the phosphate (P) compartmentation in E. huxleyi cultures was examined in relation to calcification and growth under P limitation. A specific culture parameter, mainly extracellular P, was furthermore shown to synchronize the switch from naked to calcifying cells. Such observation has never been reported in the literature for coccolithophores and may actively improve knowledges on their life cycle. These results are discussed on the basis of a model inferring a potential role of calcium binding P rich molecules, the polyphosphates, in the control of calcification and life-cycle of coccolithophores. These physiological data allowed the design of a fed-batch process in a 9 L laboratory pilot photobioreactor, resulting in the highest productivities in metabolites ever reported for these two species

Full textThe aim of this work was to synthesize and characterize the carbohydrates derivatives modified in anomeric position or in primary position regioselectively. In the first time, a regioselective disubstitution of maltotriose (DP3) was carried out and optimized in only one step giving a key intermediary to the accesses of dialkylmaltotriosides derivatives. Then, we carried out the synthesis of S-alkylglycosides and O-alkylglycosides modified in anomeric position, with good yields, in order to undertake a study structure/activity. In the second time, we evaluated the antimicrobial properties of all these compounds. We observed that the S-alkylglycosides derivatives exhibit weak or no activity whereas the O-alkylmaltodextrines compounds alone or in mixture showed interesting antimicrobial activities. In the third time, an evaluation of the anti-inflammatory drug activity of all these compounds was carried out. Once again, the S-alkylglycosides derivatives presented weak or no activity, while the O-alkylmaltodextrines derivatives alone showed significative anti-inflammatory drug activities. Moreover, mixtures of the latter led to a synergy of action on the inhibition of IL8 never yet described in the literature

Full textIn skin care formulation, "bio mimetism" consists in bringing the composition of the formulas closer to that of the human skin. The use of bio-mimetic ingredients, copies of the components of the human organism, is one of the possible means to improve the effectiveness and the tolerance of the formulas. The monoglycerides, molecules naturally present in the human skin, are emulsifier of choice for the stabilization of emulsions. Using various techniques of physicochemical analyses and studies of molecular modelling, it was possible to specify the role of their structure on their interfacial behaviour and to confirm that these molecules are efficient surfactants. Three monoglycerides were found particularly interesting: Glyceryl laurate, Glyceryl oleate and Glyceryl 12-hydroxystearate. A galenical application carried out on a formula optimized by experimental modelling, then proved that it is possible to obtain a better tolerance with "bio mimetic" formulations

Full textFlavonoids are natural polyphenolic compounds widely distributed in many plants. Many of them possess biological activities (radical scavengers, anti-inflammatory. . . ). Chromones are compounds structurally parented to flavonoids, far less ubiquitous in nature and whose biological activities were less studied and less known compared to flavones. We were interested on the potential cosmetical and pharmacological activities of some flavones and chromones. In the first part of this work, cosmetical activities of some chromones were studied. Starting from a mono or polyhydroxylated phenyl ring derivatives, simple known and new synthetic methods were used to afford new mono or polyhydroxylated chromone compounds substituted or not at position 2 and (or) 3. These molecules were submitted to biological tests used in cosmetic or dermatology. Some of the synthesised molecules were found to possess highly interesting biological activities concerning melanin synthesis, keratinocytes differentiation, and antioxidant capacities. In a second part, the protein kinases inhibitory activity of some flavones and chromones were tested. After a preliminary study in molecular modeling, a flavone library was synthesised containing 5,7-dihydroxyflavones and 7-ester-5-hydroxyflavones. These compounds were tested as Tie2 and protein kinase B inhibitors. Biological results showed that all synthesised flavones of our library possess an inhibitory activity for these protein kinases. Some chromone derivatives were then synthesised in order to study the variations of activity in accordance with different substitutions at position 2 and 3 of the chromone core. New chromones with acryloyl group at position 3 or at positions 2 and 3, or with aceto-piperazine at position 2 were synthesised and tested

Full textWild plant with many applications, milk thistle (Silybum marianum L. Gaertn) accumulates in its achenes a mixture of flavonolignans (resulting from the oxidative coupling of taxifolin and coniferyl alcohol), called silymarin. This plant used in pharmaceutical (Legalon ™) thanks to the hepatoprotective properties of flavonolignans, remains underutilized in the cosmetic industry despite many biological activities, particularly antioxidant and anti-inflammatory. The development of silymarin extraction and separation methods made it possible to study the natural variability of the contents of this mixture in wild ecotypes. The flavonolignans accumulation during achene maturation has been investigated at transcriptional level with the study of genes expression involved in the pathway of phenylpropanoids and flavonoids. It led to the identification of genes involved in flavonolignans biosynthesis. Biotechnological approaches have highlighted the interest of hairy root and callus culture for the bioproduction of these compounds. Finally, the characterization of the antioxidant and anti-aging activities of natural or semisynthetic flavonolignans shows that some of them can be used as replacement for widely-decried synthetic antioxidants. This work constitutes a scientific basis for future more targeted studies on the bioproduction conditions of flavonolignans for cosmetic application

Full textAmong the species of oleaginous plants of Guadeloupe, two species were selected for this studt, Pouteria multiflora and Calophyllum calaba, of share their local and traditional uses. The study concerning the Pouteria multifora species, made it possible to provide data on the composition in major and minors components of the fruit:glucids, proteins, lipids and rock salt. The description of the principal molecules constitutive of the greasy substances after identification and quantification by associating various techniques, such as the chromatography, supplements the chimical knowledge of the fruit. Study of the glyceridic fractions of the oils extracted from the fruit of this species, we will quote the fatty acids associated with waxes. In addition, the study of the components of the unsaponifiable enabled us to raise the prevalence of the tripernic molecules such as erythrodiol and uvaol. Concerning Calophyllum calaba, our study emphasizes two essentials points: the choice of the criteria and conditions allowing an optimal extraction of an oil of quality by single -scew pressing of almonds, chimiometric study; as well as the analytical characteristics and the qualitative and quantitative constitution of oil : distribution of the fatty acids, tocopherols and tocotrienols contents, presence of triperpenols and sterols. These last results confirm the properties concerned a traditional use relating to the healing and calming actions of this oil, having for origin the quoted molecules and which are well-known for their antioxydant activies. A local company retained this oleaginous matter for some of these formulations; indeed, the oli of Calophyllum calaba is presented as a excellent raw material for the beauty care

Full textAnimal venoms, by their biochemical richness and diversity of molecular targets, are a highly significant source of new molecules, with numerous potential applications. A major thrust of present pharmacological research now concerns drugs to replace certain antibiotics, proven ineffective due to the appearance of resistant strains. And discoveries of animal toxins over the past 40 years or more, have shown that venomous species could be the « pharmacists of the future », with only a few species of ants' venoms having been studied, but each time resulting in the description of original toxins. The aim of this thesis has been to study the venom of tropical ants using a multidisciplinary approach combining biochemical analysis and toxinology, centred on two main areas: (1) Research into their biological activity on insects and bacteria; (2) Fractionation of venoms using separative techniques, plus biochemical analysis of the peptides isolated using mass spectrometry. Two species of ant have been chosen based on their lifestyles: Crematogaster striatula is arboricolous and Tetramorium bicarinatum, terricolous, and the different biological activities of their crude venoms have been investigated using the MTT test. The Dufour liquid of C. striatula, in addition to its ability to scare away competitors, can definitively paralyse termites. For the venom of T. bicarinatum, two peptides have been isolated, and identified using mass spectrometry. Bicarinalin, a short 20 amino acid residues C-terminally amidated peptide, was notable for its wide spectrum antibacterial activity which, when tested on two Staphylococcus strains, proved to be at least if not more potent than mellitin, the antimicrobial peptide from bee venom. Bicarinalin would thus appear to be a good candidate for the development of new antibiotic drugs
