Abstract:
The eggs of the parasitic blood fluke, Schistosoma, are the main drivers of the chronic pathologies associated with schistosomiasis, a disease of poverty afflicting approximately 220 million people worldwide. Eggs laid by Schistosoma mansoni in the bloodstream of the host are encapsulated by vascular endothelial cells (VECs), the first step in the migration of the egg from the blood stream into the lumen of the gut and eventual exit from the body. The biomechanics associated with encapsulation and extravasation of the egg are poorly understood. We demonstrate that S. mansoni eggs induce VECs to form two types of membrane extensions during encapsulation; filopodia that probe eggshell surfaces and intercellular nanotubes that presumably facilitate VEC communication. Encapsulation efficiency, the number of filopodia and intercellular nanotubes, and the length of these structures depend on the egg’s vitality and, to a lesser degree, its maturation state. During encapsulation, live eggs induce VEC contractility and membranous structures formation in a Rho/ROCK pathway-dependent manner. Using elastic hydrogels embedded with fluorescent microbeads as substrates to culture VECs, live eggs induce VECs to exert significantly greater contractile forces during encapsulation than dead eggs, which leads to 3D deformations on both the VEC monolayer and the flexible substrate underneath. These significant mechanical deformations cause the VEC monolayer tension to fluctuate with the eventual rupture of VEC junctions, thus facilitating egg transit out of the blood vessel. Overall, our data on the mechanical interplay between host VECs and the schistosome egg improve our understanding of how this parasite manipulates its immediate environment to maintain disease transmission.

Abstract:
Leukocyte transmigration across vessel walls is a critical step in the innate immune response. Upon their activation and firm adhesion to vascular endothelial cells (VECs), leukocytes preferentially extravasate across junctional gaps in the endothelial monolayer (paracellular diapedesis). It has been hypothesized that VECs facilitate paracellular diapedesis by opening their cell–cell junctions in response to the presence of an adhering leukocyte. However, it is unclear how leukocytes interact mechanically with VECs to open the VEC junctions and migrate across the endothelium. In this study, we measured the spatial and temporal evolution of the 3D traction stresses generated by the leukocytes and VECs to elucidate the sequence of mechanical events involved in paracellular diapedesis. Our measurements suggest that the contractile stresses exerted by the leukocytes and the VECs can separately perturb the junctional tensions of VECs to result in the opening of gaps before the initiation of leukocyte transmigration. Decoupling the stresses exerted by the transmigrating leukocytes and the VECs reveals that the leukocytes actively contract the VECs to open a junctional gap and then push themselves across the gap by generating strong stresses that push into the matrix. In addition, we found that diapedesis is facilitated when the tension fluctuations in the VEC monolayer were increased by proinflammatory thrombin treatment. Our findings demonstrate that diapedesis can be mechanically regulated by the transmigrating leukocytes and by proinflammatory signals that increase VEC contractility.

Aim: Talimogene laherparepvec (T-VEC) is an intralesional therapy for unresectable, metastatic melanoma. T-VEC real-world use in the context of anti-PD1-based therapy requires further characterization. Materials & methods: A retrospective review of T-VEC use from 1 January 2017 and 31 March 2018 for melanoma patients was conducted at seven US institutions. Results: Among 83 patients, three categories of T-VEC and anti-PD-1 therapy were identified: T-VEC used without anti-PD-1 (n = 29, 35%), T-VEC after anti-PD-1-based therapy (n = 22, 27%) and concurrent T-VEC and anti-PD-1-based therapy (n = 32, 39%). 25% of patients discontinued T-VEC therapy due to no remaining injectable lesions, 37% discontinued T-VEC due to progressive disease. Discontinuation of T-VEC did not differ by anti-PD-1-based therapy use or timing. Conclusion: In real-world settings, T-VEC may be used concurrently with or after anti-PD-1-based therapy.

Abstract:
Previous analyses of developmentally programmed capillary regression suggested two distinct causes of vascular endothelial cell (VEC) death. The first appeared to be macrophage-dependent (Lang, R. A. and Bishop, M. J. (1993) Cell 74, 453–462) while the second was proposed to result from cessation of blood flow (Lang, R. A., Lustig, M., Francois, F., Sellinger, M. and Plesken, H. (1994). Development 120, 3395–3403). Combined, these analyses suggested a model in which initial, macrophage-mediated endothelial cell apoptosis blocked blood flow within a capillary segment and, as a consequence, caused apoptosis of all remaining cells in the affected segment. In the current study, we have tested this model using a new method that combines vital and histological analyses as a means of determining the fate of whole capillary segments and individual cells in vivo. This technique revealed that one of the first events in regression was the apoptosis of a single VEC in otherwise normal, flowing capillary segments (initiating apoptosis). These isolated, dying VECs projected into and restricted the capillary lumen, imposing either a temporary or permanent block to blood flow. Following cessation of flow, synchronous apoptosis of VECs occurred (secondary apoptosis). In addition, a quantitative analysis revealed a reciprocal relationship between plasma flow and VEC apoptosis. These observations are consistent with a model for capillary regression in which macrophages induce apoptosis in a limited number of VECs and, as a consequence of a block to blood flow, also cause apoptosis in those remaining.

For some groups $G$ of affine transformations of the complex plane, we deduce simple-to-apply formulas for calculating the $G$-deviation between $n$-gons on the complex plane. We apply these formulas for defining new measures of asymmetry of triangles. These new measures can be applied in Electric Power Engineering for evaluating the quality of 3-phase electric power. One of such measures, namely the affine deviation, is espressible via the unbalance degree, which is a standard characteristic of quality of three-phase electric power.

Abstract:
The non-resonant bounded uniserial representations of Vec(R) form a certain class of extensions composed of tensor density modules, all of whose subquotients are indecomposable. The problem of classifying the extensions with a given composition series is reduced via cohomological methods to computing the solution of a certain system of polynomial equations in several variables derived from the cup equations for the extension. Using this method, we classify all non-resonant bounded uniserial extensions of Vec(R) up to length 6. Beyond this length, all such extensions appear to arise as subquotients of extensions of arbitrary length, many of which are explained by the psuedodifferential operator modules. Others are explained by a wedge construction and by the pseudodifferential operator cocycle discovered by Khesin and Kravchenko.

Abstract:
This paper examines the effects of adjustments in the key policy rate, included as the interbank rate, on the Swedish household sector debt from the second quarter of 2000 to the third quarter of 2019. The effect is examined through a Vector Error-Correction (VEC) model to determine if it provides results that are aligned with the given theory. The research question is “Does an increase in the repo rate temporarily decrease the Swedish household sector debt?”. The theory suggests that an increase in the policy rate will have a short-run negative effect on household debt. To short-run effect the economy and the inflation is also the intention of the central bank of Sweden, called Riksbanken. With regards to the short-run dynamics, the previous researchers have constructed quite simple models of estimation and concluded contradictory results. The VECM distinguish from the previous models by a long-run equilibrium. The model concludes a short-run negative effect on debt for an increase in the policy rate, with an insignificant long-run equilibrium. By regard to this, the findings of this paper agree with the given theory. The ongoing increase in the policy rate conducted by Riksbanken can be justified by this paper.

O objetivo deste trabalho é apresentar o Modelo Vetorial Autorregressivo (VAR) e uma das suas variações, o Modelo Vetorial de Correções de Erros (VEC), segundo uma abordagem Bayesiana, considerando componentes regionais, que serão inseridos nos modelos apresentados através de informações a priori que levam em consideração a localização dos dados. Para formar tais informações a priori são utilizados conceitos referentes à econometria espacial, como por exemplo, as relações de contigüidade e as implicações que estas trazem. Como exemplo ilustrativo, o modelo em questão será aplicado a um conjunto de dados regionais, coletados por estados brasileiros. Este conjunto de dados consiste em observações da variável produção industrial para oito estados, no período de janeiro de 1991 a setembro de 2006. Em função da escolha do modelo adequado, a questão central foi descobrir em que medida a incorporação destas informações a priori no modelo VEC Bayesiano é coerente quando estimamos modelos que consideram informações localizacionais.
The main goal of this work is to present the Vector Autoregressive Model (VAR) and one of its variations, the Vector Error Correction Model (VEC), according to a Bayesian variant, considering regional components that will be inserted in the models presented through prior information, which takes in consideration the data localization. To form such prior information, spatial econometrics is used, as for example the contiguity relations and the implications that these bring to the modeling. As illustrative example, the model in question will be applied to a regional data set, collected for Brazilian states. This data set consists of industrial production for eight states, in the period between January 1991 and September 2006. The central question is to uncover whether the incorporation of these prior informations in the Bayesian VEC Model is coherent when we use models that consider contiguity information.

Com a introdução do euro a análise do uso e circulação efetiva de notas por cada um dos países da zona euro passou a ser uma tarefa bastante desafiante devido à sua contínua migração entre países. É também cada vez mais importante a estimação e previsão da evolução da circulação de notas estando o DET encarregue de assegurar a emissão e a colocação em circulação da moeda legal necessária à economia nacional. O objetivo deste estudo é auxiliar o DET na sua tarefa e fazer um exercício comparativo da capacidade preditiva de um conjunto de modelos econométricos clássicos para prever a circulação de notas em Portugal, através de uma abordagem econométrica. Numa primeira fase é feita uma análise sobre a relação entre as notas em circulação e um conjunto de variáveis económicas através da abordagem VEC para o período entre 2002 e 2014. Os resultados demonstram evidência estatística de existência de uma relação de cointegração, havendo portanto uma relação de equilíbrio de longo prazo entre as variáveis. Numa segunda fase são testadas várias metodologias de previsão para avaliar qual o modelo com melhor desempenho para prever as notas em circulação. Tanto os três critérios para avaliar os erros de previsão como o teste Diebold Mariano apontaram para o método de Holt como aquele com melhor capacidade preditiva sobre as notas em circulação. Relativamente à previsão para o período de 2015 até 2017, todos os diferentes métodos de previsão utilizados apontam para a permanência da tendência verificada ao longo dos anos.
Since the euro cash changeover in 2002 it has become increasingly difficult to analyze and compare developments in banknote usage in different countries of the euro area, as a result of the migration of banknotes across borders. Given that the “Departamento de Emissão e Tesouraria do Banco de Portugal” is responsible for ensuring the issue and putting into circulation of the national economy’s legal tender currency, it is increasingly important to estimate and forecast the evolution of banknotes. Thus, the main objective of this study is to make a comparative assessment of the predictive ability of a set of classic econometric models to predict the banknotes in Portugal, through an econometric approach. Initially we study the relationship between banknotes in circulation and a set of macroeconomic variables through the VEC approach to the period between 2002 and 2014. The results show statistical evidence of the existence of one cointegration relation, so apparently there is one long-run equilibrium relationship between the variables under study. The second part consists on comparing multiple forecasting methods to evaluate which is the best model to predict the banknotes in circulation. All the three criteria to evaluate forecasting errors (RMSE, MAE and MAPE) and the Diebold Mariano test, point out the Holt method as the one with better predictive ability on banknotes in circulation. Regarding the forecast from 2015 to 2017, all the different forecasting methods point out to the continuation of the same pattern observed in the past few years.

The object of this dissertation is to analyze the influence of net revenue, total shareholders' equity and long-term debt on the profitability of the value of the shares of ARCADIS, by applying methods of time series, such as unit root test, test Johansen cointegration, the vector autoregression model (VAR), vector error correction (VEC), impulse response and variance decomposition of forecast errors function. The database is quarterly from January 2000 to December 2012 According to the Johansen cointegration test, the results indicate that the series that have unit roots are all integrated of order one in level and show a relationship term between them. The coefficients in the estimated model is presented according to the literature;can also observe that the tests demonstrated a strong influence of net revenue on the share value when compared with the other variables. From the evaluations performed in this study, it can be stated that the variables are relevant to explain the variations over time in the dependent variable value of the share on the stock exchange. That paper presents an analysis of the evolution of consulting engineering companies and the increasing degree of internationalization. Highlights the relevance of the study compared with the purpose of understanding the growth of the company ARCADIS in consulting engineering and project management segment. And since expanded its market competitiveness.
O objeto dessa dissertação é analisar a influência da receita líquida, patrimônio líquido total e dívidas de longo prazo sobre a rentabilidade do valor das ações da ARCADIS, através da aplicação de métodos de séries de tempo, tais como, teste de raiz unitária, teste de cointegração de Johansen, o modelo autorregressivo vetorial (VAR), vetor de correção de erros (VEC), função impulso-resposta e decomposição da variância dos erros de previsão. A base de dados é quadrimestral no período de janeiro de 2000 a dezembro de 2012. De acordo com o teste de cointegração de Johansen, os resultados indicam que as séries que apresentam raiz unitária são todas integradas de ordem um em nível e mostram uma relação de longo prazo entre elas. Os coeficientes obtidos no modelo estimado se apresentaram de acordo com a literatura. Pode-se observar também que os testes demonstraram uma forte influência da receita líquida sobre o valor da ação quando comparados com as outras variáveis. A partir das avaliações realizadas neste estudo, pode-se afirmar que as variáveis são relevantes para explicar as variações ocorridas no decorrer do tempo, na variável dependente valor da ação na bolsa de valores. O referido trabalho apresenta uma análise da evolução das empresas de engenharia consultiva e o crescente grau de internacionalização. Abstract:
In 1980 Feigin and Fuchs classified the length 2 bounded representations of Vec(R), the Lie algebra of polynomial vector fields on the line, as a result of their work on the cohomology of Vec(R). This dissertation is concerned mainly with the uniserial (completely indecomposable) representations of Vec(R) with a single Casimir eigenvalue and weights bounded below. Such representations are composed of irreducible representations with semisimple Euler operator action, bounded weight space dimensions, and weights bounded below. These are known to be the tensor density modules with lowest weight λ, for any non-zero complex number λ, and the trivial module C, with Vec(R) actions π_λ and π_C, respectively. Our proofs are cohomology arguments involving the first cohomology groups of Vec(R) with values in the space of homomorphisms between two irreducible representations. These results classify the finite length uniserial extensions, with a single Casimir eigenvalue, of admissible irreducible Vec(R) representations with weights bounded below. In almost every case there is at most one uniserial representation with a given composition series. However, in the case of an odd length extension with composition series {π_1,π_C,π_1,…,π_C,π_1}, there is a one-parameter family of extensions. We also give preliminary results on uniserial representations of the Virasoro Lie algebra.

Nas últimas décadas, identificou-se uma crescente preocupação com a sustentabilidade e com a qualidade ambiental, ao mesmo tempo em que se observou uma expansão do comércio internacional e uma procura por um maior grau de liberalização comercial. Nesse contexto, surgiram órgãos destinados a defender cada uma das questões, ambiental e comercial, e questionamentos e conflitos sobre os impactos que cada um deles estava causando ao outro. Diante desses questionamentos identificou-se, então, a importância da inter-relação entre as duas áreas, de forma que passou a buscar-se uma maior interação entre as discussões englobando comércio e meio ambiente. Contudo, muitas pesquisas ainda tendem a avaliar as questões ambientais e comerciais separadamente. Em virtude disso, este trabalho tem como objetivo analisar conjuntamente as duas questões, a fim de ampliar a discussão acerca de sua inter-relação; ao mesmo tempo, propõe-se um modelo para analisar a relação entre a liberalização comercial e a qualidade ambiental (representada aqui pelas emissões de CO2 equivalente) para o Brasil, estimado por meio de um Modelo de Autorregressão Vetorial estrutural com correção de erro (VAR-VEC). A análise abrangeu o período de 2003 a 2015, período que já incorpora a entrada da China no mercado internacional, ocorrida no ano de 2002, fato que usualmente tem bastante impacto sobre os fluxos comerciais e sua modelagem. As variáveis utilizadas no modelo foram o índice de abertura comercial, importações mundiais, como proxy de renda mundial, preços totais de commodities e emissões brasileiras de CO2 Equivalente. O modelo identificou que um aumento da abertura comercial brasileira contribui para a ampliação das emissões de CO2 do país, seguindo o esperado de acordo com a literatura.
In the last decades, a concern about sustainability and environmental quality has grown, simultaneously to the process of the international trade expansion, pursuing also for greater degree of trade liberalization. In this context, international bodies were created to defend each of these issues, environmental and commercial, raising questions and conflicts about the impacts that each one was causing to the other. Thus, the importance of the interrelation between the two themes became evident, in a way to highlight the necessity to promote a greater interaction between debates encompassing trade and environment. However, many surveys still tend to analyze environmental and trade issues separately. As a result, this work aims to examine the two issues together, in order to broaden the discussion about their interrelationship. At the same time, a model has been proposed to quantify the relationship between trade liberalization and environmental quality (here represented by CO2 equivalent emissions) for Brazil, estimated by a VAR- VEC model. The analysis covered the period from 2003 to 2015, comprising the tie of China\'s entry into the international market in 2002, which is important once this event usually has a significant impact on international trade flows and their modelling. The variables used in the model are the index of trade liberalization for Brazil, the world imports as a proxy for the world income, total commodity prices and the Brazilian emissions of equivalent CO2. The model identified that an increase in Brazilian trade liberalization contributes to an increase in the country\'s CO2 emissions, following expectations from the reviewed literature.
