Abstract:
A doença de Aujeszky (DA) é uma infecção causada por um herpesvírus, o vírus da DA (VDA), primariamente em suínos. Esta doença está presente no Estado de Santa Catarina (SC) desde 1984. Devido ao impacto da DA no mercado exportador de carne suína, no comércio de reprodutores e nas perdas de produtividade, um programa de erradicação, financiado por uma parceria entre a indústria e a associação de produtores tem tido sucesso em eliminar gradualmente a DA de rebanhos suínos de SC. O último caso de DA no estado foi identificado em julho de 2004. Durante o processo de despovoamento/repovoamento, foi detectado um rebanho suíno positivo para o VDA localizado no oeste de SC. Estudos de rastreabilidade da origem daqueles animais indicaram que a fonte era uma granja que distribuía reprodutores ilegalmente sem certificação sanitária. Este suinocultor mantinha um sistema de integração que incluía 40 diferentes produtores para quem eram comercializados reprodutores e/ou suínos para terminação. Testes de soroprevalência detectaram anticorpos anti-VDA em 12 daqueles rebanhos. Devido a sua localização dentro do raio de 2,5km do foco inicial, uma outra granja, onde havia uma central de inseminação artificial que distribuía sêmen suíno para outras 5 granjas do mesmo proprietário, foi testada e também resultou positiva. Os objetivos deste artigo são descrever as condições sanitárias frente ao VDA naquelas granjas que receberam suínos ou sêmen suíno daqueles suinocultores, as medidas para controlar e eliminar o VDA dos rebanhos positivos e a situação atual decorrente deste trabalho. Além disso, o artigo busca alertar que medidas de vigilância ativa e normas sanitárias para comércio e distribuição de material genético devem ser seguidas, caso contrário, a DA pode recrudescer e tornar-se fora de controle como ocorria antes do início deste programa.

Abstract:
AbstractHematopoietic stem and progenitor cells (HSPCs) are multipotent cells giving rise to all blood lineages during life. HSPCs emerge from the ventral wall of the dorsal aorta (VDA) during a specific timespan in embryonic development through endothelial hematopoietic transition (EHT). We investigated the ontogeny of HSPCs in mutant zebrafish embryos lacking functional pten, an important tumor suppressor with a central role in cell signaling. Through in vivo live imaging, we discovered that inptenmutant embryos a proportion of the HSPCs died upon emergence from the VDA, an effect rescued by inhibition of phosphatidylinositol-3 kinase (PI3K). Surprisingly, inhibition of PI3K in wild-type embryos also induced HSPC death. Surviving HSPCs colonized the caudal hematopoietic tissue (CHT) normally and committed to all blood lineages. Single-cell RNA sequencing indicated that inhibition of PI3K enhanced survival of multipotent progenitors, whereas the number of HSPCs with more stem-like properties was reduced. At the end of the definitive wave, loss of Pten caused a shift to more restricted progenitors at the expense of HSPCs. We conclude that PI3K signaling tightly controls HSPCs survival and both up- and downregulation of PI3K signaling reduces stemness of HSPCs.

We identified a novel Drosophila protein of ≈ 400kDa, hemolectin (d-Hml), secreted from haemocyte-derived Kc167 cells. Its 11.7kbp cDNA contains an open reading frame of 3843 amino acid residues, with conserved domains in von Willebrand factor (VWF), coagulation factor V/VIII and complement factors. The d-hml gene is located on the third chromosome (position 70C1-5) and consists of 26 exons. The major part of d-Hml consists of well-known motifs with the organization: CP1-EG1-CP2-EG2-CP3-VD1-VD2-VD′-VD3-VC1-VD′′-VD′′′-FC1-FC2-VC2-LA1-VD4-VD5-VC3-VB1-VB2-VC4-VC5-CK1 (CP, complement-control protein domain; EG, epidermal-growth-factor-like domain; VB, VC, VD, VWF type B-, C- and D-like domains; VD′, VD′′, VD′′′, truncated C-terminal VDs; FC, coagulation factor V/VIII type C domain; LA, low-density-lipoprotein-receptor class A domain; CK, cysteine knot domain). The organization of VD1-VD2-VD′-VD3, essential for VWF to be processed by furin, to bind to coagulation factor VIII and to form interchain disulphide linkages, is conserved. The 400kDa form of d-Hml was sensitive to acidic cleavage near the boundary between VD2 and VD′, where the cleavage site of pro-VWF is located. Agarose-gel electrophoresis of metabolically radiolabelled d-Hml suggested that it is secreted from Kc167 cells mainly as dimers. Resembling VWF, 7.9% (305 residues) of cysteine residues on the d-Hml sequence had well-conserved positions in each motif. Coinciding with the development of phagocytic haemocytes, d-hml transcript was detected in late embryos and larvae. Its low-level expression in adult flies was induced by injury at any position on the body.

Abstract:
The effects of endotoxemia on cardiopulmonary parameters, before and after cyclooxygenase blockade, were determined in anesthetized ponies spontaneously breathing a mixture of halothane and 100% O2. Escherichia coli endotoxin was infused intravenously at 20 micrograms/kg for 1 h followed by 10 micrograms X kg-1 X h-1 the subsequent 4 h. By 15 min endotoxin increased mean pulmonary arterial pressure (Ppa), pulmonary vascular resistance (PVR), and alveolar dead space ventilation (VDA/VT), and these were followed by a return to base-line values by 30 min. A second increase in PVR occurred by 5 h of endotoxemia. The early increases in Ppa, PVR, and VDA/VT were blocked by flunixin meglumine (FM), a cyclooxygenase inhibitor. Endotoxin decreased central plasma volume by 1 h and cardiac index by 3 h; hematocrit and plasma protein concentration were increased by 0.5 and 1.5 h, respectively, indicating a loss of plasma volume. These changes were also blocked or attenuated by FM. Moreover, in ponies treated with endotoxin + FM, cardiac index increased, indicating the presence of a cardiac-stimulating factor. We conclude that endotoxemia in ponies causes cardiopulmonary dysfunction that is mediated by cyclooxygenase-dependent and possibly cyclooxygenase-independent metabolites.

Immune therapy improves cancer outcomes, yet many patients do not respond. This pre-clinical study investigated whether vascular disrupting agents (VDAs) could convert an immune unresponsive tumor into a responder. CDF1 mice, with 200 mm3 C3H mammary carcinomas in the right rear foot, were intraperitoneally injected with combretastatin A-4 phosphate (CA4P), its A-1 analogue OXi4503, and/or checkpoint inhibitors (anti-PD-1, PD-L1, or CTLA-4 antibodies), administered twice weekly for two weeks. Using the endpoint of tumor growth time (TGT5; time to reach five times the starting volume), we found that none of the checkpoint inhibitors (10 mg/kg) had any effect on TGT5 compared to untreated controls. However, CA4P (100 mg/kg) or OXi4503 (5–50 mg/kg) did significantly increase TGT5. This further significantly increased by combining the VDAs with checkpoint inhibitors, but was dependent on the VDA, drug dose, and inhibitor. For CA4P, a significant increase was found when CA4P (100 mg/kg) was combined with anti-PD-L1, but not with the other two checkpoint inhibitors. With OXi4503 (50 mg/kg), a significant enhancement occurred when combined with anti-PD-L1 or anti-CTLA-4, but not anti-PD-1. We observed no significant improvement with lower OXi4503 doses (5–25 mg/kg) and anti-CTLA-4, although 30% of tumors were controlled at the 25 mg/kg dose. Histological assessment of CD4/CD8 expression actually showed decreased levels up to 10 days after treatment with OXi4503 (50 mg/kg). Thus, the non-immunogenic C3H mammary carcinoma was unresponsive to checkpoint inhibitors, but became responsive in mice treated with VDAs, although the mechanism remains unclear.

Abstract. Ozone Monitoring Instrument (OMI), launched in July 2004, is dedicated to the monitoring of the Earth's ozone, air quality and climate. OMI is the successor of the Total Ozone Mapping Spectrometer (TOMS) instruments and provides among other atmospheric and radiometric quantities the total column of ozone (TOC), the surface ultraviolet (UV) irradiance at several wavelengths, the erythemal dose rates and the erythemal daily doses. The main objective of this work is to compare OMI data with data from ground-based instruments in order to use OMI products (collection 2) for scientific studies. The Laboratoire d'Optique Atmosphérique (LOA) located in Villeneuve d'Ascq (VdA) in the north of France performs solar UV measurements using a spectroradiometer. The site of Briançon in the French Southern Alps is also equipped with a spectroradiometer operated by Interaction Rayonnement Solaire Atmosphère (IRSA). The OMI total ozone column data is obtained from the OMI-TOMS and OMI-DOAS algorithms. The comparison between the TOC retrieved with ground-based measurements and OMI-TOMS data shows good agreement at both sites for all sky conditions with a relative difference for most of points better than 5%. For OMI-DOAS data, the agreement is generally better than 7% and these data show a significant dependence on solar zenith angle. Comparisons of spectral UV on clear sky conditions are also satisfying with relative differences smaller than 10% except at solar zenith angles larger than 65°. On the contrary, results of comparisons of the erythemal dose rates and erythemal daily doses for clear sky show that OMI overestimates surface UV doses at VdA by about 15% and that on cloudy skies, the bias increases. At Briançon, such a bias is observed if data corresponding to snow-covered surface are excluded.

48 Background: A functional vascular system is essential for growth of solid malignancies including gastric cancer. For establishment and maintenance of such a vascular system endothelial cells (ECs) and pericytes (e.g. vascular smooth muscle cells, VSMC) are required. We hypothesized that targeting tumor vasculature with the vascular disrupting agent (VDA) ASA404 (Novartis Oncology) reduces tumor growth in a model of gastric cancer. Methods: Gastric cancer (GC) cell lines, ECs and VSMCs were used for experiments. Effects of ASA404 on growth of GC, EC and VSMC were assessed by MTT assays. Impact of ASA404 (20 mg/kg on day 1, 5, 9) in combination with paclitaxel (10 mg/kg on day 1 and 7) on tumor growth was assessed in a subcutaneous tumor model. Treatment was started when tumors reached a size of approximately 200 mm3. Tumors were measured and harvested on day 23 for IHC analyses. Effect of ASA404 on blood perfusion of tumors during therapy was monitored by contrast-enhanced ultrasound (CEUS). Results: In vitro ASA404 impaired growth of ECs and VSMCs upon stimulation with condition media from gastric cancer cells. No direct effect on tumor cells was observed. In vivo, treatment with ASA404 led to marked decrease of tumor perfusion and an increase of necrosis as determined by CEUS. Furthermore, combination of ASA404 with paclitaxel showed significant reduction of tumor growth compared to controls (p < 0.05). In addition, tumor vascularisation and tumor cell proliferation were significantly reduced as determined by CD31-positive vessel area and BrdU-positive cells (p < 0.05). Conclusions: Combination of the VDA ASA404 with paclitaxel impairs tumor growth and perfusion of gastric cancer in an experimental model. Hence, targeting tumor vasculature with ASA404 appears to be a promising strategy for therapy of gastric cancer. No significant financial relationships to disclose.

The diploma thesis deals with the capability assessment of an optical coordinate measuring machine according to VDA 5. The capability assessment is an effective tool for deciding whether the evaluated measuring machine is capable for the intended use and under which conditions its use is possible. The theoretical part of the thesis is devoted to the analysis of coordinate measuring machines, with a closer focus on a subset of optical measuring machines and the analysis of their capability assessment procedures. In the practical part of the thesis, the selected procedure is applied on the specific measuring machine Micro-Vu Sol 311. The main contribution of this thesis is a description of the possibilities of the considered coordinate measuring machine for its further use in the educational process at the Faculty of Mechanical Engineering of Brno University of Technology.

Abstract:
The principal aim of this project was to synthesise a range of functionalised heterocycles using novel transition metal promoted radical cyclisation reactions as an alternative to the well-established tin hydride method. On achieving this we hoped to successfully apply these methods towards the synthesis of natural product templates. In chapter two, a series of copper(l)-amine catalysts were employed to mediate the 5- endo-trig radical cyclisation of trichloroacetamides, which led to the formation of a variety of highly functionalised bicyclic y-Iactams in good to excellent yield. A variety of cyclisation precursors were examined. This work was also extended towards the synthesis of analogues of the heterocyclic ring fragment of non-peptide bradykinin inhibitor, L-755,807. In chapter 3, the reaction of a series of tertiary bromoacetamides with catalytic copper(I) bromide/N,N,N',N',N",N" -hexamethyltriethylenetetramine (Me6-tren) at room 'temperature is described. This reaction furnished regioisomeric mixtures of unsaturated pyrrolidinones via a highly efficient 5-endo-trig radical cyclisation reaction. We also illustrated that a variety of less activated secondary bromoacetamides undergo efficient 5-endo-trig radical cyclisation reactions to give a,l3-unsaturated monoene lactams under atom transfer conditions mediated by copper(l) bromide and tripyridylamine (TP A) in refluxing toluene. This approach was used towards the synthesis of analogues of the sesquiterpenic alkaloid, eremophilene y-Iactam. Chapter 4 describes an alternative route to constrained bicyclic ring systems mediated by ceric ammonium nitrate (CAN). In addition, the first reported 5-endo-trig radical cyclisation of l3-amido esters to afford functionalised y-Iactams was developed. This methodology was exploited in the production of analogues of the heterocyclic ring fragments of a' number of biologically active natural products.

This work explored the adsorption of organic molecules to achieve change on surfaces. In this context it was proposed, the adsorption of organic molecules of halogenated derivatives of uracil, 5 Fluorouracil and 5 Chlorouracil, in monocrystalline substrate of HOPG (highly oriented pyrolytic graphite), sizzling was the technique used for construction of self-assembled monolayers (SAM). The atomic resolution of the STM (Scanning Tunneling Microscope) images of HOPG substrate prove its hexagonal structure and that their interatomic distance is 0.246 nm. While in the molecular resolution images of 5 Fluorouracil and of 5 Chlorouracil adsorbed on the HOPG substrate, is presented evidence that after in the adsorption some changes occur in the intermolecular distances in the structures and the positioning of these molecular films formed on the substrate because the molecular films behaved in a horizontal position and were stabilized laterally by hydrogen bonds when they are protonated, but when we performed the same procedure of adsorption for these organic molecules, only varying the pH of the environment, the change caused the deprotonation of the molecules, thus allowing one to simulate electrochemical environment and being able to extract information about the pKa of adsorbates using UV-Vis spectroscopy, and was also observed that these deprotonated molecular films behaved in a vertical position on the substrate.
Neste trabalho foi explorada a adsorção de moléculas orgânicas para realizar modificação em superfícies. Neste contexto foi proposta a adsorção das moléculas orgânicas de derivados halogenados do uracil, o 5 Fluorouracil e o 5 Clorouracil no, substrato monocristalino do HOPG (Grafite Pirolítico Altamente Orientado do inglês, Highly Oriented Pyrolitic Graphite), sendo usada a técnica de sizzling para construção de monocamadas automontadas (SAM Self-Assembled Monolayers). As imagens de STM (Scanning Tunneling Microscopy) de resolução atômica do substrato de HOPG comprovam sua estrutura hexagonal e sua distância interatômica que é de 0,246 nm. Já nas imagens de resolução molecular do 5 Fluorouracil e do 5 Clorouracil adsorvido no substrato do HOPG, são apresentadas evidências de que após a adsorção ocorrem mudanças nas distâncias intermoleculares, nas estruturas e no posicionamento desses filmes moleculares formados no substrato, pois os filmes moleculares se comportaram em um posicionamento horizontal e são estabilizados lateralmente por ligações de hidrogênio quando estão protonadas, mas quando foi realizado o mesmo procedimento de adsorção para essas moléculas orgânicas, só que variando o pH do meio, esta variação provocou a deprotonação das moléculas, assim permitindo simular um ambiente eletroquímico e sendo possível extrair informação sobre o pKa dos adsorbatos usando a espectroscopia UV-Vis, e foi observado também que estes filmes moleculares deprotonados se comportaram em um posicionamento vertical no substrato.

Abstract:
Catalytic degradation of PMMA was successfully performed at temperatures below 300°C. The use of zeolite catalyst could reduce reaction temperature in comparison with an ordinary thermal degradation process. It was found that the product distribution obtained from batch experiment depends on zeolite acid properties whereas the composition of the liquid fraction is directly related to the shape selectivity of the catalyst. A continuous fixed bed process was designed that allowed to obtain MMA monomer as main product. The increase of reaction temperature from 200 to 270°C showed a positive effect on the liquid product yield. However, at higher temperatures, the light product was further cracked into gaseous products. Significant deactivation of ZSM-5 catalyst was observed after 120 hours of operation, resulting in a decrease in liquid product yield. Regeneration of the coked ZSM-5 extrudates was achieved by oxidation with ozone at low temperatures, below 150°C. The effects of temperature, GHSV and inlet concentration of ozone on carbon removal efficiency were studied. Carbon removal with ozone started at 50°C and reached a maximum of 80% at 100°C. Higher temperatures were not beneficial due to the strong limitation of ozone diffusion which confines radical production then the regeneration process to the outer surface. In optimal conditions, ozonation almost fully restored the zeolite activity without damaging the texture and active sites of zeolite, as shown from the results of regenerated catalyst in PMMA cracking

Depression is a common mood disorder. Although major ethical challenges make it nearly impossible to invasively and directly measure serotonin (5-hydroxytryptamine, 5-HT) levels in human brains, neuroimaging technologies have shown macroscopic structural and functional abnormalities in the prefrontal cortex (PFC) of depressed patients. The monoamine hypothesis of depression is based on the neurotransmitter imbalance, such as deceased serotonin brain levels are implicated in the cause of depression. Research has focused on the control mechanisms involved in the dorsal raphé nucleus (DRN) which is the serotonergic control center located in the midbrain. We hypothesized that activation 5-HT 2A receptor in PFC would increase serotonin levels by an AMPA-dependent mechanism in both DRN and PFC. Enhancement of the 5-HT in DRN may inhibit 5-HT level in PFC by 5-HT 1A receptor. This becomes the full feedback loop system. While 5-HT levels in the PFC have been well studied, pathway that modulate this DRN pool through upstream cascade interactions leading to a downstream feedback loop have been difficult to elucidate. Developing a mechanism-based pharmacokinetics (PK) and pharmacodynamics (PD) model to quantitatively describe the effect of 5-HT 2A receptors regulation to serotonin in the DRN and PFC would help us to better understand the complex brain. 5-HT 2A receptor agonist and AMPA receptor agonist and antagonist were used to activate or block the related receptor. Male Wistar rats underwent neurosurgery for implantation of microdialysis (MD) probes. Three to five rats were randomly assigned to experimental arms. Using the MD method, the drug combination was examined to explore the drug effect on time course of 5-HT release in DRN and PFC. Based on the experiment results, a mechanism-based PD model was developed. Phoenix WinNonlin ® and Berkeley Madonna™ were used for model estimation, external validation with secondary data set, and simulation. The result supports the possibility of a 5-HT 2A /AMPA feedback control circuit that originates in the PFC and modulates DRN and PFC 5-HT levels through feedback coupling of 5-HT. The time-course profiles of 5-HT in both DRN and PFC was well modeled and model parameters were estimated with good precision (CV% ranged from 1.37% to 35.03%). The mechanism model was developed to characterize and better understand the neurotransmitter mechanisms, providing estimations of various parameters of the disease related receptor system.

Syftet med denna undersökning var att ta reda på vilken förståelse elever i skolår 5 och 6 har om begreppen HIV och AIDS. Vidare ville vi undersöka vilken skillnad det fanns mellan de två skolåren. Eftersom vi valde att göra en kvantitativ undersökning, blev enkät som underökningsmetod vårt val. 106 elever deltog i undersökningen, 50 elever i skolår 5 och 56 elever i skolår 6. Resultatet visade att många elever i skolår 6 hade förståelse för begreppen men endast en elev i skolår 5 hade den förståelse som vi sökte.

This paper presents a method for the Jacobian derivation of 5-DOF 3R2T PMs (parallel mechanisms), where 3R denotes three rotational DOFs (degrees of freedom) and 2T denotes two translational DOFs. First the mobility analysis of such kind of parallel mechanisms is reviewed briefly. The Jacobian matrix of the single limb kinematic chain is obtained via screw theory, which is a 6 × 5 matrix. Then it is shown that the mobility analysis of such kind of PM is important when simplifying the 6 × 5 matrix into a 5 × 5 Jacobian matrix. After obtaining the 5 × 5 Jacobian matrix for each limb, a 5 × 5 Jacobian matrix for the whole mechanism can be established.

Tax amnesties have taken centre stage as a compliance tool in recent years. The OECD estimates that since 2009 tax amnesties in 40 jurisdictions have resulted in the collection of an additional €102 billion in tax revenue. A number of African countries have introduced tax amnesties in the last decade, including Nigeria, Namibia, South Africa and Tanzania. Despite their global popularity, the efficacy of tax amnesties as a tax compliance tool remains in doubt. The revenue is often below expectations, and it probably could have been raised through effective use of regular enforcement measures. It is also argued that tax amnesties might incentivise non-compliance – taxpayers may engage in non-compliance in the hope of benefiting from an amnesty. This paper examines the administration of tax amnesties in various jurisdictions around the world, including the United States, Australia, Canada, Kenya and South Africa. The paper makes a cost-benefit analysis of these and other tax amnesties – and from this analysis develops a model tax amnesty, whose features maximise the benefits of a tax amnesty while minimising the potential costs. The model tax amnesty: (1) is permanent, (2) is available only to taxpayers who make a voluntary disclosure, (3) relieves taxpayers of penalties, interest and the risk of prosecution, but treats intentional and unintentional non-compliance differently, (4) has clear reporting requirements for taxpayers, and (5) is communicated clearly to attract non-compliant taxpayers without appearing unfair to the compliant ones. The paper then focuses on the Ugandan tax amnesty introduced in July 2019 – a Voluntary Disclosure Programme (VDP). As at 7 November 2020, this initiative had raised USh16.8 billion (US$6.2 million) against a projection of USh45 billion (US$16.6 million). The paper examines the legal regime and administration of this VDP, scoring it against the model tax amnesty. It notes that, while the Ugandan VDP partially matches up to the model tax amnesty, because it is permanent, restricted to taxpayers who make voluntary disclosure and relieves penalties and interest only, it still falls short due to a number of limitations. These include: (1) communication of the administration of the VDP through a public notice, instead of a practice note that is binding on the tax authority; (2) uncertainty regarding situations where a VDP application is made while the tax authority has been doing a secret investigation into the taxpayer’s affairs; (3) the absence of differentiated treatment between taxpayers involved in intentional non-compliance, and those whose non-compliance may be unintentional; (4) lack of clarity on how the VDP protects the taxpayer when non-compliance involves the breach of other non-tax statutes, such as those governing financial regulation; (5)absence of clear timelines in the administration of the VDP, which creates uncertainty;(6)failure to cater for voluntary disclosures with minor errors; (7) lack of clarity on VDP applications that result in a refund position for the applicant; and (8) lack of clarity on how often a VDP application can be made. The paper offers recommendations on how the Ugandan VDP can be aligned to match the model tax amnesty, in order to gain the most from this compliance tool.
