Abstract:
ABSTRACT Rhodococcus equi causes severe pyogranulomatous pneumonia in foals and in immunocompromised humans. Replication of virulent isolates within macrophages correlates with the presence of a large plasmid which encodes a family of seven virulence-associated proteins (VapA and VapC to VapH), whose functions are unknown. Although cell-mediated immunity is thought to be crucial in eliminating R. equi infection, antibody partially protects foals. The antibody response to both VapA and VapC was similar in six adult horses and six naturally exposed but healthy foals, as well as in eight foals with R. equi pneumonia. The immunoglobulin G (IgG) subisotype response of pneumonic foals to Vap proteins was significantly IgGb biased and also had a trend toward higher IgGT association compared to the isotype association of antibody in adult horses and healthy exposed foals. This suggests that in horses, IgGb and IgGT are Th2 isotypes and IgGa is a Th1 isotype. Furthermore, it suggests that foals which develop R. equi pneumonia have a Th2-biased, ineffective immune response whereas foals which become immune develop a Th1-biased immune response. Pneumonic foals had significantly more antibody to VapD and VapE than did healthy exposed foals. This may indicate a difference in the expression of these two Vap proteins during persistent infection. Alternatively, in pneumonic foals the deviation of the immune response toward VapD and VapE may reflect a bias unfavorable to R. equi resistance. These data indicate possible age-related differences in the equine immune response affecting Th1-Th2 bias as well as antibody specificity bias, which together favor the susceptibility of foals to R. equi pneumonia.

Abstract:
ABSTRACT The virulence plasmids of the equine virulent strainsRhodococcus equi ATCC 33701 and 103 were sequenced, and their genetic structure was analyzed. p33701 was 80,610 bp in length, and p103 was 1 bp shorter; their sequences were virtually identical. The plasmids contained 64 open reading frames (ORFs), 22 of which were homologous with genes of known function and 3 of which were homologous with putative genes of unknown function in other species. Putative functions were assigned to five ORFs based on protein family characteristics. The most striking feature of the virulence plasmids was the presence of a 27,536-bp pathogenicity island containing seven virulence-associated protein (vap) genes, includingvapA. These vap genes have extensive homology to vapA, which encodes a thermoregulated and surface-expressed protein. The pathogenicity island contained a LysR family transcriptional regulator and a two-component response regulator upstream of six of the vap genes. The vap genes were present as a cluster of three (vapA, vapC, and vapD), as a pair (vapE andvapF), or individually (vapG;vapH). A region of extensive direct repeats of unknown function, possibly associated with thermoregulation, was present immediately upstream of the clustered and the paired genes but not the individual vap genes. There was extensive homology among the C-terminal halves of all vap genes but not generally among the N-terminal halves. The remainder of the plasmid consisted of a large region which appears to be associated with conjugation functions and a large region which appears to be associated with replication and partitioning functions.

Abstract:
Vaping-associated pulmonary injury (VAPI) is a newly evolving medical condition caused by inhalation of substances including tetrahydrocannabinol- or nicotine-containing products via vaping. Vaping is the act of inhaling and exhaling an aerosol, which can contain fine particles that have been identified as toxic chemicals, produced by an e-cigarette or similar device. Presenting symptoms of VAPI include respiratory symptoms (dyspnea, chest pain, cough, and hypoxia), gastrointestinal symptoms (nausea, emesis, diarrhea, and abdominal pain), and constitutional symptoms (fever, chills, weight loss, and fatigue). The diagnosis and management of VAPI are being studied and it is considered a diagnosis of exclusion. Most patients are young, with male predominance, and history of vaping. The severity of presenting symptoms varies. Radiographic findings vary in severity with chest x-rays showing bilateral infiltrates, although those findings may be absent early in the disease course. Computed tomography of the chest can show bilateral ground-glass opacities with pleural effusions and pneumomediastinum. Management of VAPI is based upon clinical presentation with empiric antibiotics for CAP coverage and steroids if indicated for reducing lung inflammation.

Abstract:
ABSTRACT Rhodococcus equi is a facultative intracellular pathogen that causes pneumonia in foals but does not induce disease in adult horses. Virulence of R. equi depends on the presence of a large plasmid, which encodes a family of seven virulence-associated proteins (VapA and VapC to VapH). Eradication of R. equi from the lungs depends on gamma interferon (IFN-γ) production by T lymphocytes. The objectives of the present study were to determine the relative in vivo expression of the vap genes of R. equi in the lungs of infected foals, to determine the recall response of bronchial lymph node (BLN) lymphocytes from foals and adult horses to each of the Vap proteins, and to compare the cytokine profiles of proliferating lymphocytes between foals and adult horses. vapA, vapD, and vapG were preferentially expressed in the lungs of infected foals, and expression of these genes in the lungs was significantly (P < 0.05) higher than that achieved during in vitro growth. VapA and VapC induced the strongest lymphoproliferative responses for foals and adult horses. There was no significant difference in recall lymphoproliferative responses or IFN-γ mRNA expression by bronchial lymph node lymphocytes between foals and adults. In contrast, interleukin 4 (IL-4) expression was significantly higher for adults than for foals for each of the Vap proteins. The ratio of IFN-γ to IL-4 was significantly higher for foals than for adult horses for most Vap proteins. Therefore, foals are immunocompetent and are capable of mounting lymphoproliferative responses of the same magnitude and cytokine phenotype as those of adult horses.

Abstract:
ABSTRACT Rhodococcus equi causes severe pyogranulomatous pneumonia in foals. This facultative intracellular pathogen produces similar lesions in immunocompromised humans, particularly in AIDS patients. Virulent strains of R. equi bear a large plasmid that is required for intracellular survival within macrophages and for virulence in foals and mice. Only two plasmid-encoded proteins have been described previously; a 15- to 17-kDa surface protein designated virulence-associated protein A (VapA) and an antigenically related 20-kDa protein (herein designated VapB). These two proteins are not expressed by the same R. equi isolate. We describe here the substantial similarity between VapA and VapB. Moreover, we identify three additional genes carried on the virulence plasmid, vapC, -D, and -E, that are tandemly arranged downstream of vapA. These new genes are members of a gene family and encode proteins that are approximately 50% homologous to VapA, VapB, and each other. vapC, -D, and -E are found only in R. equi strains that express VapA and are highly conserved in VapA-positive isolates from both horses and humans. VapC, -D, and -E are secreted proteins coordinately regulated by temperature with VapA; the proteins are expressed when R. equi is cultured at 37°C but not at 30°C, a finding that is compatible with a role in virulence. As secreted proteins, VapC, -D, and -E may represent targets for the prevention of rhodococcal pneumonia. An immunologic study using VapA-specific antibodies and recombinant Vap proteins revealed no evidence of cross-reactivity despite extensive sequence similarity over the carboxy terminus of all four proteins.

To our knowledge, this is the first description of R. equi and/or mycobacteria species identified in the lymph nodes of Tayassuid specimens. Despite the absence of Vap genes, the identification of R. equi, as well as nontuberculous and saprophytic mycobacteria highlight the risk of transmission of these pathogens from Tayassuidae species to humans by means of meat or meat products contaminated with lymph node contents, since R. equi plasmidless type and mycobacterial species described here have been reported as causes of pulmonary and extrapulmonary infections both in immunocompetent and immunocompromised humans.

Abstract:

Outcome of a pioneering experience in Brazil, the Creative Writing postgraduate program of Pontifícia Universidade Católica do Rio Grande do Sul (PUCRS), this dis-sertation presents a two level composition: a fictional story, Mônica Vai Jantar, un-published and written during the master’s degree period, and a complementary reflexive essay, here serving as an afterword. Mônica Vai Jantar is a novelette, having stream of consciousness as literary basis, or, specifically, a narrated monologue, containing a sin-gle long sentence, in which I use only commas as punctuation mark, along with rare question marks. It tells the mind confusion of a character that, while preparing for a party, finds out her husband was masturbating inside a bus. The essay, in its own turn, never a step-by-step follow up, is a short narrative route of my influences, doubts, deci-sions, difficulties, authorial references, theoretical references, fictional and personal conversations that helped me through the writing of this work.

Abstract:

Abstract:
In recent years, the quantity of municipal solid wastes (MSW) has increased significantly in the EU and other industrialised and developing countries raising the question of its sustainable disposal management. Within the waste management hierarchy, thermal disposal especially gasification with energy recovery is a desired and viable option. The gasification of solid fuels such as biomass is one of the most promising technologies for thermo chemical conversion. This process leads to a fuel gas suitable for efficient gas turbines feeding. The successfful design and modelling of a gasifier requires reliable kinetic data. The purpose of this work is first to study the steam gasification kinetics of chars produced by municipal wastes pyrolysis. The MSW are modelled as a mixture of four organic constituents: paper, wood, plastics, and vegetables. The various char samples are obtained by pyrolysis of each waste constituent, in a fixed bed reactor. These chars are used as raw materials in steam gasification experiments. The kinetics parameters, with respect to H20, together with the influence of the char's physical properties are experimentally determined. A kinetic expression for the gasification reaction, based on the random pore model is deduced. Then 1D isothermal and non-equimolar reaction-diffusion model is developed in order to assess the diffusional effects taking place during steam gasification in a fixed bed. The model takes into account local chemical reaction rate and effective transport properties dependant on time and position within the char bed. Inter particle diffusion and structural changes of the char bed during the gasification are taken into account. The model shows a good agreement with the experimental results, obtained at various bed heights, and point out the relevant rote played by diffusional effects within a fixed bed reactor. The kinetic results are used to estimate, in a down dyaft gasifier, the total carbon yield conversion as a fonction of botte the mass fraction of stagnant zones within the bed char and of the characteristic length of these stagnant zones. Finally, in thermal treatment main of the initial energy is transformed into tar. The spontaneous tar decomposition by thermal treaent is investigated. The thermal cracking kinetic of MSW pyrolysis vapours is established

Kuklik, Robert G. "Capital Asset Prices Modelling - Concept VAPM." Doctoral thesis, Vysoká škola ekonomická v Praze, 2008. http://www.nusl.cz/ntk/nusl-196945.

Full text
Abstract:
The key objective of this thesis is the outline of an alternative capital market modeling framework, the Volatility Asset Pricing Model, VAPM, inspired by the innovative dual approach of Mandelbrot and Hudson using the method based on synthesis of two seemingly antagonistic factors -- the volatility of market prices and their serial dependence determining the capital markets' dynamics. The pilot tests of this model in various periods using the market index as well as a portfolio of selected securities delivered generally satisfactory results. Firstly, the work delivers a brief recapitulation regarding the concepts of a consumer/investor choice under general conditions of hypothetical certainty. Secondly, this outline is then followed by a description of the "classical" methodologies in the risky environment of uncertainty, with assessment of their corresponding key models, i.e. the CAPM, SIM, MIM, APTM, etc., notwithstanding results of the related testing approaches. Thirdly, this assessment is based on evaluation of the underlying doctrine of Efficient Market Hypothesis in relation to the so called Random Walk Model. Fourthly, in this context the work also offers a brief exposure to a few selected tests of these contraversial concepts. Fifthly, the main points of conteporary approaches such as the Fractal Dimension and the Hurst Exponent in the dynamic framework of information entropy are subsequently described as the theoretical tools leading to development of the abovementioned model VAPM. The major contribution of this thesis is considered its attempt to apply the abovementioned concepts in practice, with the intention to possibly inspire a further analytical research.

Abstract:
A kind of Hilly Power Chassis with Vari-ground Clearance and Vari-wheel Track Mechanism (VII-HPC) is designed to be adapted to various types of crops grown ridge section and height. It consists of the middle part, balancing system, two driving axles and steering system. These assemblies form an H-type chassis structure where both sides of the driving axle and the middle part are connected for power transmission, ground clearance varying and wheel track adjustable function, which ensures full-time four-wheel drive in a complex road environment and confirms that the middle part is always in the angle bisector of the two driving axles. According to the various agricultural ground clearance and line spacing of different crops in the hilly areas, the ground clearance and the wheel track can be adjusted continuously and smoothly driven by hydraulic system. According to VII-HPC Structural Characteristic without Ackerman Steering Linkage, the hydraulic four wheel steering scheme has been proposed. The related analysis is presented to verify the feasibility of VII-HPC, including the relationship between the ground clearance and parameters like centroid height, and the ride analysis at different speeds on the pulse road. The results and the subsequent experimental results show the design method and the concept of VII-HPC are proved reasonable, feasible and practical.
