Diplomová práce se zabývá analýzou trhu práce, vytvářením pracovních příležitostí a nezaměstnaností v okrese Kolín. První kapitola je věnována ekonomické teorii trhu práce a výkladu důležitých termínů, druhá pak sociokenomickým charakteristikám regionu, které svým způsobem předurčují míru nezaměstnanosti. Třetí kapitola rozebírá zaměstnanost v okrese Kolín, její vývoj a současnou strukturu. Součástí této kapitoly je i určení nejvýznamnějších zaměstnavatelů v okrese a stručné představení dvou z nich. V následující kapitole se zabývám analýzou nezaměstnanosti, jejím vývojem a strukturou. Poslední část mé práce je věnována společnosti Toyota Peugeot Citroen Automobile Czech s.r.o. Cílem této kapitoly je postihnout vliv této investice na regionální trh práce.

Centra měst jsou přeplněna automobily, je tedy nutné prosazovat šetrné druhy dopravy, jako je například cyklodoprava. V teoretické části je popsáno fungování veřejného sektoru, přiblížena problematika veřejných statků a veřejných zakázek. Jsou zde přiblíženy cíle Národní strategie cyklistické dopravy a nástroje BYPAD+ na podporu cyklodopravy ve městech. Dále jsou popsány vlivy a přínosy tohoto druhu alternativní dopravy a možnosti financování. Cílem diplomové práce je analýza stávajícího stavu cyklistické dopravy v Kolíně a jeho okolí, stanovení hlavních priorit a dílčích cílů strategie cyklistické dopravy a návrh vývojového programu, který by umožnil zhodnotit politiku cyklodopravy v tomto městě a přinést návrhy na zlepšení této politiky do budoucna.

The demographical transition continued in Bohemia until the end of 19 th century with changing populations and reproductive behaviour. The political district of Kolín belonged among Czech areas with fairly early asserted changes. That was because of the regions economical maturity and also closeness of Prague, whence these new tendences of reproductive behaviour spread. INTRODUCTION From the nineties of the twent5r's century [l], when appeared the first references about the new type ofthe stationary phases designated for HPLC called monoliths, these became to be the object of interest of several scientific groups [2-4]. Their uncontested advantage in comparison with classical columns is their high stability in pH range from 2 to 12 where there is no possibility to use the classical columns. other advantages are their easy and cheap preparation and the absence of frits on the ends of the separating medium because the monolith is covalently bound to the inner side of the capillary [5]. On the other hand, their disadvantage remains to be worse in reproducibility of the stationary phase preparation than in case of classical columns. This PhD Thesis is focused on monoliths based on organic polymers, mainly the methacrylate monoliths. For the first time, this ýpe of monoliths was shown by Peters et al. [6] and since then there have been made several monolithic phases based on different separating features 17_101. Methacrylate monoliths are convenient for separation of the low molecular substances L7l as well as the high molecular substances like the proteins, oligonucleotides or saccharides n0-121.

This thesis is focused on polymorphism of MDR1 gene c.-6-180T>G, which causes phenobarbital resistance. Phenobarbital is one of the most commonly used drug against epilepsy, which is one of the most common neurological disease in dogs with frequency between 0,5-5 % according to breed. This mutation has so far been detected only in the border collie breed, which manifests up to 30 percent resistance to the treatment of epilepsy. In the text is subscribed border collie breed, dog epilepsy and gene MDR1. Methodical part is focused on detection of the polymorphism in model population of 82 dogs. The results agree with previous studies and confirm frequency of mutant alleles 32 % within the population.

3 Abstract Title: Athletics at elementary schools in Kolín district Aims of work: The aim of my thesis is to collect information about present education of athletics at second grade of elementary schools from children's point of view. We would like to find out the poularity of component athletic disciplines among the pupils, standard of material equipment at schools for athletics classes and if there is an athletic club at school from the inquiry. The method: I used an inquiry method, where the respondents answered questions created by myself. Pupils from second degree of elementary schools in Kolín district were the representative file. Results: We reached the conclusion that popularity of athletics among the children at elementary schools is not very great from aquired and elaborated data. We also found out the popularity of component athletic disciplines among the pupils and school equipment for athletics classes. Our theories were validated only in part. The Title of diploma thesis: Use of core-shell columns in the Analysis of Drugs I The aim of this thesis was to find suitable conditions for HPLC analysis of fenbendazole and its metabolite oxfendazole and to search a suitable internal standard. Several types of columns, mobile phase compositions and temperatures were examined in the thesis. Ascentis® Express F5 10 cm x 3,0 mm, 2,7µm column was proved to be suitable. The mobile phase composition was formic acid 0,03 mol/l and acetonitrile, and it was necessary to set a gradient elution due to significantly different lipophilicities of analytes. The increase of column temperature to 50řC improved the shape of chromatographic peaks. I managed to choose suitable internal standard - reduced flubendazole. The new method has been successfully tested in the analysis of the real biological samples.

This work was focused on the comparison of the separation of ambroxol and its three impurities with a column contained fully porous particles (Acquity BEH Shield RP18) and with two core-shell columns (Kinetex EVO C18 a Kinetex C18). Different pH values of buffer (borate buffer with pH = 9.70 and a phosphate buffer with pH = 7.00 and pH = 3.00) and different gradient programs were tested for separation of analytes. Compared to the column with fully porous particles (Acquity BEH Shield RP18), core-shell columns provided shorter analysis time. Lower backpressure and different selectivity depend on a separation system. The best separation conditions were obtained with the column containing fully porous particles (BEH Shield RP18) and gradient according to program Method 2 with the borate buffer. Analysis time was 10 minutes and the last peak had the retention time about 9 min. Autosampler was tempered at 10 ř C and injection volume was 2 μl. The column was tempered at 30 řC. Detection was set at wavelength of 247 nm. Component A contained 10 mM borate buffer with pH = 9.70 and component B was acetonitrile. This thesis deals with new found neolithic settlement in Kolín, which is dated from Neolithic - Stroked Pottery Culture. On the site were found also the Lengyel Culture imports. The thesis brings the results of the pottery and the chipped industry analyses. The relative chronology of the settlement and some pits was based upon the pottery analysis. There were some settlement pits, which were situated outside and inside the circle, and circular ditch chosen for it. Marginally is here solved the question of interpretation of the function of objects and intercultural relations. The work brings especially the findings about next neolithic site with circular ditch and Lengyel culture imports in Bohemia. The solid phase extraction (SPE) is an effective, reliable and selective modern method for sample preparation of biological material before chromatographic analysis. In this graduation thesis the new SPE method for isolation of vitamin D2 (ergocalciferol), D3 (cholecalciferol) and its metabolite 25 - (OH) D3 (calcidiol) simultaneously with vitamin A (retinol) and E (α-tocopherol) in human serum was developed. The vitamins were subsequently determined by validated high performance liquid chromatography method (HPLC) using tocol as internal standard. The developed SPE procedure: Firstly 250 l of human serum was pipetted to glass tube and 1000 l of cool ethanol (4řC) was added. Sample was shaken and left 10 minutes in a refrigerator at 4řC for deproteinisation. After centrifugation (4 000 x g, 15 minutes, 4řC) all supernatant was carefully removed and applied into pre-treatment SPE column (firstly washed with 1 ml of methanol, then with 1 ml of distilled water). During these two steps the sorbent must not be dried up. The vitamins were eluted by 1.5 ml of methanol and then 2.0 ml of n-hexane. The organic solutions were evaporated in the vacuum concentrator at a temperature of 45 ř C. The residue was dissolved in 250 l of methanol and analyzed by using HPLC method. The Lucembourg court attracted several noble families that had a oportunity to get a good status by serving a king and also hold offices. For such a service were ideal loyal nobles of newly arrived noble families, who didn't own any property in Bohemia. Lords of Colditz and lords of Bergov also belonged to them. The king rewarded them for their loyal service by giving them fiefs (especially in north-west Bohemia: Krupka and Bílina). The Luxembourg dynasty enabled these noble families a social rise. Lords of Colditz were connected to royal court since their beginnings in Bohemia. On the other hand some of the lords of Bergov hold offices, but they didn't hesitate to leave the court at a convinient moment. More likely they participated in newly emerging estates. During the reign of Wenceslav IV. lords of Colditz remained faithful to him, on the other hand lords of Bergov belonged to the main figures of his opposition. At the same time they changed the royal fiefs (Bílina was bought by lords of Colditz) for allod (Trosky, Turnov, Chlumec upon Cidlina). The overall theme of this thesis is the electronic training of employees (volunteers) in Czech non-school organizations. The main objects are the organizations' satisfaction with e-learning methods, benefits and obstacles that organizations face while implementing and using e-learning and forms of e-learning that are distinctive for Czech non-school environment. The theoretical part of the thesis is dedicated first to explaining the term "e-learning" and followed by its brief history. Subsequently, different approaches to classification of e-learning are summed up. One of the chapters follows the work of David A. Kolb - especially his theory of experiential learning including learning styles and their use in the development of e-learning in organization. In the practical part of the thesis the current state of e-learning in the Czech non-school organizations is explored (with the use of a survey). First, the methodology is presented, followed by an analysis of collected data and answering of seven stated research questions with the goal to clarify different aspects of the use of e-learning in Czech non-school organizations. This diploma thesis summarizes knowledge of significant genetically contitioned deseases occurring in border collies. There is described a total of 14 diseases, some with the location of causal mutation not yet known. Primary focus of this thesis is g.4411956_4411960delGTTT mutation of gene VPS13B causing Trapped Neuthrophil Syndrome (TNS), MDR1 gene's mutation AF045016.1: c.227_230delATAG associated with multidrug resistance (MDR1) and CUBN gene's mutation c.8392delC which causes intestinal malabsorption of cobalamin by another name ImerslundGräsbeck syndrome (IGS). A genotype analysis of 89 border collies with a proof of origin was performed. The DNA was extracted from buccal mucosal swabs, the isolation of DNA was performed by Chelex-100 from the native material. The analysis was proceeded by optimized PCR-RFLP method using restrictive MboI (MDR1) and Msl I (IGS) enzymes. There were detected 4 g.4411956_4411960delGTTT mutation vectors causing TNS. As for the MDR1 and IGS there wasn't detected any affected case.

This diploma thesis deals with the new trends in the monetization of the video gaming industry with emphasis on crowdfunding, free-to-play model, selling games by "pay what you want" bundles and sales on Steam and other digital distribution stores. The purpose of this diploma thesis is to describe how the players on personal computers react to these trends in the Czech Republic. First, the situation in Czech gaming industry and the situation of players in the Czech Republic is described. Then, the new trends are analyzed thoroughly using foreign researches. The practical part analyses the effects of the new trends on Czech players with the results of an online questionnaire. First, the methodology is presented and subsequently the collected data is analyzed. Gradually, the thesis are answering the three research questions which are clarifying various aspects of how the Czech players respond to the new trends in the video gaming industry monetization. This thesis examines the views and daily life of the employees of the company and residents of Kolín. The work provides an overview of the history and development of the company Tatra. Represents district Kolín and focuses primarily on the description in the period of. Normalization, which touched city, business, residents but also the whole of Czechoslovakia. The thesis is based mainly on archival materials, historical periodicals and interviews, which were conducted by the method oral history with contemporary witnesses. It connects the working environment, personal lives, leisure activities, social culture of the city and residents who were part of a large company and history of the city of Kolín. At the same time, the work seeks to highlight the positives and negatives period of so-called normalization and current relationship of people to the place where they lived more or less part of their lives.

This diploma thesis deals with development and optimization of conditions of HPLC method for a suitable pre-concentration and determination of two pesticides - fenoxycarb and permethrin in surface waters. The on-line SPE HPLC with column switching system was used for method development and determination. To validate the optimal conditions there was chosen analytical column Supelco Ascentis Express Phenyl - Hexyl (100 x 4.6 mm), particle size of 2.7 microns. Detection was carried out at wavelength 225 nm and column temperature was set on 45 řC using gradient elution with a mobile phase of acetonitrile - water at a flow rate of 1,0 ml/min. Prior to analysis, the sample was pre-concentrated using an extraction precolumn Ascentis Express C-18 (5 x 4.6 mm), particle size of 2.7 micron with washing mobile phase of 30% acetonitrile in water at a flow rate of 0.5 ml/min. The injected sample volume was optimized to 1500 μl of water sample. An original method, which allows the determination of two selected pesticides in real matrix surface waters without difficult sample pre-treatment was developed.

This thesis presents archaeological finds from the site "U silnice", in municipality Plaňany (district Kolín), where a settlement from roman period has been found during excavations in 2016 and 2017. An assemblage of excavated finds consists mostly of ceramic, animal bones and daub. The aim of the thesis is to describe qualitative and quantitative attributes of these finds. Results will be used to validate and compare properties of individual archaeological contexts. Main goal is to place the site into regional network of settlements and cemeteries from roman period. Potential of the site rises from the fact that it is the only settlement from roman period excavated by modern methods in the region and therefore it represents a valuable source of new information. The circadian clock generates circadian rhythms, which participate on regulation of a number of signalling pathways. Disruption of the circadian regulatory mechanism is linked to a development and a progression of certain types of cancer including colorectal tumorigenesis. Progression of tumorigenesis depends on the cell cycle machinery related to cell proliferation and apoptosis. MiRNAs play a role in initiation and progression of tumorigenesis because they interfere in regulatory pathways associated with tumorigenesis. The aim of the thesis was to determinate existence of circadian rhytms in clock controlled genes (Tef, Dbp), miRNAs (miR-1-3p, miR-16-5p, miR-34a-5p, miR-155-5p, miR-192-3p) and genes of the cell cycle machinery (Ccnd1, Ccne1, Ccna1, Ccnb1) and apoptosis (Casp3, Bcl2, Bad). Further, to compare detected circadian rhythms during aging and neoplastic transformation of colon by quantitative RT-PCR. We have observed circadian expression of Tef, Dbp, Ccne1, Ccna1, Ccnb1, Casp3 and Bcl2 in young mice colon, Tef, Dbp, miR-1-3p, Ccne1, Ccna1 in old mice colon and Tef and Dbp in colorectal tumors. RNDr. Dalibor Šatínský, Ph.D. Title of Diploma Thesis: Column switching chromatography for mycotoxines determination in Czech beer samples A new fast and sensitive method of high performance liquid chromatography for simultaneous determination of mycotoxins ochratoxin A and citrinin using column- switching system for on-line sample pretreatment was developed. 100 µl of beer was injected directly into the chromatographic system. Isolation of analytes was performed on guard column Ascentis Express RP-C18 (5 x 4.6 mm, 2.7 µm) washing out with methanol - water solution of acetic acid 0.5% (30:70, v/v), at a flow rate 2 ml/min; time of valve switch was set on 2 minutes. The separation was performed on Ascentis Express Phenyl-Hexyl column (100 x 4.6 mm, 2.7 µm) with mobile phase of composition acetonitril - water solution of acetic acid 0.5% at a flow rate 1 ml/min. This part of the diploma thesis acquaints with potential leisure centres and presents various forms of leisure activities for children with moderate and severe mental retardation. The practical application part identifies specific leisure activity options and in a collaboration with leisure centres it provides a comprehensive overview of leisure activities for children with moderate and severe mental retardation in Kolin and Nymburk region. It also captures parental interest in these leisure activities.

The following analytical columns were tested: Kinetex PFP particle size of 1,7 µm and 2,6 µm (Phenomenex) and Ascentis Express Phenyl-Hexyl particle size of 2,7 µm (Sigma- Aldrich), which are columns with porous shell particles and Acquity BEH Phenyl column with totally porous particles size of 1,7 µm. To compare the properties of the columns, 7 estrogen steroids were used using PDA detector at a wavelength of 280 nm for detection. Measurements were carried out using isocratic elution. The Mobile phase consisted of ultrapure water and acetonitrile. Columns were evaluated in terms of efficiency, peak symmetry, resolution and the back pressure, which was generated on the columns. The aim of this diploma thesis is to find out whether selected children aged 8-15 years are sufficiently educated about the course of the planned surgery in the Department of Pediatric Surgery at Kolín Hospital. In the theoretical part, I have described possible ways of communication with children aged 8 to 15, play therapy and the possibilities of its use, and perioperative care for children in childhood surgery at the Kolín hospital. I used the method of structured interviews with selected children between the ages of 8 and 15 who were hospitalized in the department of pediatric surgery for the planned surgery from June to September 2017. The practical part describes interviews with selected children. I used the interview results to prepare educational materials for children agend 4 to 10 years.

AABBSSTTRRAACCTT The molecular circadian clock enables anticipation of environmental changes. In mammals, clocks are ubiquitously present in almost all tissues and they are comprised of transcriptional-translational feedback loops of the so-called clock genes. The central clock represents the intrinsic pacemaker which is located in suprachiasmatic nuclei (SCN) of hypothalamus and synchronizes peripheral clocks. Clockwork system in alimentary tract and its regulatory link to intestinal functions are poorly understood. Therefore the objective of the thesis was to characterize molecular clock in particular parts of the rat intestine and to elucidate its link to the intestinal transport, regulation of cell cycle and neoplastic transformation in colonic tissue. We used quantitative RT-PCR (qPCR) to determine circadian profiles of mRNA expression of clock genes in the epithelium of duodenum, jejunum, ileum, and colon of rat. Furthermore, we analysed the expression of genes coding sodium chloride transporters and channels as well as cell cycle regulators in colon. To focus more precisely on different structures of intestinal epithelia we used laser capture microdissection. The quality of soil intended for the housing development is the topic of diploma thesis. Frequently, the local soil of the highest quality is built on. For the purpose of this thesis there were 5 model areas defined: these are the areas of Roudnice, Říčany, Slaný, Vlašim, and Kolín. The thesis describes the quality of soil which is built on in these areas. The extent of housing development in these areas is specified on the basis of landscape plans. As such, it is possible to determine the quality of soil intended for the future housing development. Moreover, the thesis deals with the legislative protection of soil.
