Abstract Repression of GAL1 expression during growth on glucose is mediated in part by cis-acting promoter elements designated URSG. We show that oligonucleotides containing sequences from two regions of URSG confer glucose repression upon a heterologous promoter. Repression caused by URSG is dependent on trans-acting factors of the glucose repression pathway and is independent of orientation or location within a promoter, suggesting that URSG contains binding sites for a glucose-activated repressor protein(s). Genetic analysis identified three apparently novel genes (URR1, URR3 and URR4) that are specifically required for URSG-mediated repression and may encode such repressor proteins. Mutations in the URR genes suppress the defect in URSG derepression caused by a snf1 mutation.

In fact Urry's discussion blends the analysis of globalization and complexity theory so thoroughly that he makes a convincing case that globalization is inexplicable without complexity theory. The book is long on big (well-conceived) ideas, description and anecdotal evidence and a bit short on specific applications and direction for proceeding with a complexity research agenda. Taken as a whole, Urry provides a compelling case for complexity and Global Complexity should be considered an important entry in both the globalization and complexity theory literatures.

The word tourist is loaded with negativity, especially, as MacCannell and Urry argue, for those of us that travel ourselves. In this exploration, the author takes a deep dive into the primary theories of MacCannell's search for authenticity and Urry's tourist gaze while recounting a series of journeys throughout Cambodia through her own Western epistomologic lens while also considering feminist, postcolonialist and decolonialist extensions and counterarguments. The purpose of this paper is to critique the portrayal of tourists as the monolithic characters created by MacCannell and Urry through an exploration of the author's experience and offer varying viewpoints and considerations that are applicable to the theorists and not. The conclusion is a call to action for further research where the tourist and destination intertwine.

Abstract:
This thesis shows that illicit drug consumers in Vietnam who administer product via injection are vulnerable actors in a paradoxical global/glocal phenomenon rooted in historical complexities of globalisation. Therefore, responsibility for HIV risks should be shifted upstream from the level of individuals toward institutional processes that manufacture environments of harm. At the global level, the UN Milennium Development Goals do not provide the required level of leadership on HIV prevention for drug injectors. Association between globalisation, opiates and blood-born disease in Vietnam is not new and is inseparable from historical transportation, migration and trade processes. As a key locale in the Cold War, after 1975, and 1979 in particular, Vietnam was 'at distance' from increasing intra-regional trade flows across its western frontiers and northern border. As a consequence, it was hermetically sealed to nearby HIV sub-epidemics unfolding among heroin cons umers. A latent HIV risk environment awaited Vietnam should geopolitical grievances be resolved and it became re-integrated among Mekong sub-regional flows. Neo-liberal financial flows returned to Vietnam in 1993 and the Mekong was spanned in 1994. In 1995 it normalised relations with the United States, joined ASEAN and announced the resurrection of transportation linkages across the northern border with China. Mid-decade, its borders were made more porous at the same time as local opium production was reduced as part of the UN global programme against drugs. Exploiting enhanced trans-boundary mobilities intended for goods, opiate traffickers quickly transformed Vietnam into a transit nation and a marke for high-quality heroin well suited to a youthful population experiencing socio-economic change including new consumerism. Following traditional pathways, a radical transformation in the fluidity of drug consumption environs ensued, enabling more widespread and efficient flows of blood across complex boundari es. Analysis reveals that a spatio-temporal confluence of structural factors has created conditions which enabled this process. These factors are overlapping and they range from global influences, such as the collapse of the USSR, to micro-economic reform such as privatisation and modernisation of the domestic pharmaceutical sector. The transformation in opiate consumption from injecting opium to heroin injecting occurred faster than expert-driven prevention systems responded, even in time and space where this was most forseeable. Although the opiate transformation was highly predictable, there has been a time-lag of almost a decade between risk transformations and policy responses equated with harm reduction principles. The thesis shows that blame for HIV sub-epidemics in Vietnam should not be attributed to vulnerable youths and young adults. Expert-driven economic transition associated with global inegration has manufactured circumstances in which drug availability has risen dramatically at a time when emp loyment growth has been insufficient and a commercial sex industry has expanded. This research confirms the cimportance of new methods of risk environment analyses, particularly in relation to trans-boundary hazards associated with global flows, including trade and human mobilities.

Title: Values, Desirability and Ideology in CNN Travel -- a Discourse Analysis on Travel Stories Author: Emmi Laine Course: Journalistikvetenskap, Kandidatkurs, H13 J Kand (Bachelor of Journalism, Fall 2013), JMK, Stockholm University, Sweden Aim: The aim is to examine which values and ideologies CNN Travel fulfills in their stories. Method: Qualitative discourse analysis. Summary: This Bachelor ́s thesis asks what is desirable, which are the values of CNN Travel, the major U.S. news corporation CNN ́s online travel site. The question has been answered through a qualitative discourse analysis on 20 chosen travel stories, picked by their relevancy, diversity, and their expressive tone. Due to the limited space and the specific textual method, the analysis was restricted to the editorial texts of these stories. The chosen method was discourse analyst Norman Fairclough ́s model of evaluation, which revealed the explicit and implicit ways the media texts suggest desired characteristics. These linguistic devices took the readers ́ agreement for granted, as they imposed a shared cultural ground with common values, which is a base for a mutual understanding. After identifying the explicit and implicit evaluations, they were organized according to some major discursive themes found in the texts, and finally analyzed in order to expose their underlying values. The results showed how these certain values brought forth certain ideologies, to some extent in keeping with recent research of tourism and travel journalism. As the study has been put into a larger context of related research, the following pages will first explain some larger concepts of discourse analysis, such as representation, cultural stereotypes, ideology and power. A cross-section from older to more contemporary theories in culture studies has been utilized; moving from Edward Said ́s postcolonial classic Orientalism, an example of cultural stereotyping, to the more recent topics of ‘promotion culture’ and consumerism, and tourism researcher John Urry ́s ideas about the consumption of places and the ‘tourist gaze.’ In the end, the study considers what kind of power does travel journalism possess over the represented tourism destinations. Finally, when questioning the travel journalists ́ legitimacy and power to represent the travel destinations, poststructuralist Michel Foucault ́s theory about the ‘regime of truth,’ as well as Antonio Gramsci ́s ideas of ‘hegemony,’ theory of dominance through consent, were discussed and confirmed.

The search for life on Mars is one of today’s most exciting and far-reaching scientific challenges. The European Space Agency’s ExoMars mission will renew the pursuit of signs of past or present life equipped with a range of analytical instruments: one such instrument will likely be Urey, a tool that will extract and analyze organic molecules. This thesis first seeks to contextualize the search for life through the use of analogue sites on Earth and ultimately works to optimize the spectrofluorometric detection of relevant organic molecules. Organic host analogues – sites on Earth where life faces similar challenges as potential martian life forms – aid in mission planning and data analysis. With these dual purposes in mind and the understanding that collaborative analogue use benefits all stakeholders, the Imperial College Mars Analogue Repository (ICMAR) was established. Samples from Iceland and the Middle Atlas mountains of Morocco were gathered to start the collection. As it grows, mission planners and scientists will have a centralized, well-characterized set of samples for astrobiological studies. Optimizing Urey’s spectrofluorometric detection of amino acids and polycyclic aromatic hydrocarbons (PAHs) was another primary aim of this study. Standard solutions were tested across a range of instrumental parameters and an ideal excitation wavelength was identified to optimize the detection of any organics present at the sampling site. The fluorescent interaction between molecules was also investigated to evaluate the instrument’s ability to detect and quantify complicated, multicomponent samples. Finally, natural organic-rich samples from England’s Blue Lias formation were tested to evaluate spectrofluorometric analysis of natural samples – a capability that will be crucial for in situ martian analysis. By highlighting the role of organic Mars analogues and optimizing the Urey instrument, this thesis enhances ExoMars’ ability to identify organic molecules and sets the stage for continued martian exploration.

Neste estudo, analiso as noções de doença e as crenças etiológicas entre os Kariri-Xocó. Eles distinguem pelo menos duas categorias de doenças que se diferenciam quanto aos critérios causais: as de cima para baixo e as de baixo pra cima . A primeira categoria, está relacionada a uma força superior, podendo ou não refletir uma punição divina. A segunda, tem a sua etiologia ligada ao sobrenatural. Since the 1950s, oil has been the dominant source of energy in the world. The cheap supply of oil has been the engine for economic growth in the western world. Since future oil demand is expected to increase, the question to what extent future production will be available is important.

The belief in a soon peak production of oil is fueled by increasing oil prices. However, the reliability of the oil price as a single parameter can be questioned, as earlier times of high prices have occurred without having anything to do with a lack of oil. Instead, giant oil fields, the largest oil fields in the world, can be used as a parameter.

A giant oil field contains at least 500 million barrels of recoverable oil. Only 507, or 1 % of the total number of fields, are giants. Their contribution is striking: over 60 % of the 2005 production and about 65 % of the global ultimate recoverable reserve (URR).

However, giant fields are something of the past since a majority of the largest giant fields are over 50 years old and the discovery trend of less giant fields with smaller volumes is clear. A large number of the largest giant fields are found in the countries surrounding the Persian Gulf.

The domination of giant fields in global oil production confirms a concept where they govern future production. A model, based on past annual production and URR, has been developed to forecast future production from giant fields. The results, in combination with forecasts on new field developments, heavy oil and oil sand, are used to predict future oil production.

In all scenarios, peak oil occurs at about the same time as the giant fields peak. The worst-case scenario sees a peak in 2008 and the best-case scenario, following a 1.4 % demand growth, peaks in 2018.

This study primarily aims to investigate the concept of walkability as a part of the literature on livability. To draw a wider theoretical framework for this study, it first seeks to answer the questions of what the terms of &lsquo

mean, and what components they constitute. Then, it focuses on the concept of &lsquo

as one of the major components of these terms and as a measurable notion in urban design. Thus, this research seeks to define the notion of &lsquo

and to identify a set of measures of walkability in public spaces to be used for the assessment of the &lsquo
walkability capacity&rsquo
of urban space. It should be noted that this research particularly tries to use the indicators of walkability which can directly impact on the design quality of urban space. Third, this research focuses on Mersin historic city center which has been in the process of deterioration for a while due to various current urban policies and strategies. The present thesis focuses on the analysis and interpretation of Chansons du Valet de Cur (1912) by Georges Adolphe Hue (1858-1948). In addition to a brief life story and work achievements of Georges Hue, the poem Le Valet de Cur (1908) and it’s author, Tristan Klingsor (1874-1966), are also introduced. After the background in introduction, the poem translation, the music analysis and interpretation are given. In summary, the union of both music and lyrics in found to be of most importance. Through this study, we hope to be able to give a title of contribution and better understanding of this great music work.

ZusammenfassungMit diesem Beitrag werden ausgewählte Projekte im Namen der Verkehrs- oder Mobilitätswende im ländlichen und suburbanen Raum betrachtet und diese in Bezug zum automatisierten und vernetzten Fahren gesetzt. Die in diesem Artikel entwickelte These ist, dass der automatisierte und vernetzte Verkehr das bestehende „System der Automobilität“ (Urry 2004) zwar entscheidend verändern kann, die konkreten Veränderungen jedoch im Zusammenhang mit lokal-spezifischen Herausforderungen und gegenwärtigen Konflikten und Kämpfen um Mobilitätssysteme betrachtet werden müssen. Der Beitrag nimmt eine klimakritische Sicht auf Autofahren und spezifisch die möglichen ökologischen, sozialen und städtebaulichen Folgen autonomen Fahrens ein. Autofahren wird auf Basis einschlägiger Literatur (z. B. Urry, Manderscheid, Seiler) als mediale Vision der Durchsetzung von Kapitalismus und Neoliberalismus kritisiert. Dabei wird auf einem Grundrecht auf Mobilität bestanden, welches gemeinschaftliches gelöst und nicht - wie es autonomes fahren potentiell tun könnte - als privates Problem behandelt wird. Mit Ansätzen der Mobilitätsgerechtigkeit (Mimi Sheller) wird Mobilität als wichtiges Medium von Gemeinschaften verstanden. Der Text plädiert für eine politische und keine marktwirtschaftliche Lösung des privaten Fahrens. Autonomes Fahren wird als sogenannte klimafreundliche Vision dekonstruiert. Der Beitrag endet mit einem Plädoyer für das Fahrradfahren als degrowth Technik sowie als soziale Technologie.

AbstractMobilities scholarship has paid considerable attention to the forms of presence enabled by air travel in hypermobile organisations (Elliott & Urry, 2010; Strengers, 2015; Storme et al., 2017). However, there has been less focus on the absences that these presences simultaneously generate. This chapter develops the concept of ‘absent presences’ enabled through the practices and policies of academic hypermobility. The chapter draws on qualitative interviews with 24 Australian-based academics, alongside a review of university policies that are relevant to air travel. We use these data to explore ‘absent presence’ in academic air travel. First, we suggest that there is an assumption in academia that embodied presence is required for authentic modes of knowledge sharing and networking, primarily at conferences and meetings. Yet this type of presence abroad requires that one is absent from home for extended periods. Second, we show how absent presence exists in academic policies concerning air travel. In university strategic plans, air travel is present as a means and measure of academic success. In university sustainability policies, however, air travel’s environmental impacts are often absent from consideration. We conclude by discussing the implications of absent presence in academic work life, as well as university policy and practice more broadly.

La siguiente comunicación aborda la investigación doctoral acerca de los procesos de creación de productos culturales artesanales que se fundan en la identidad cultural tradicional del Archipiélago de Chiloé y que se conciben para la industria del turismo. El Archipiélago de Chiloé (Chile) es un lugar que tiene la particularidad de mantener un marcado carácter tradicional en su cultura, y que se ve enfrentado en la actualidad al conflicto con la globalización, en la que el turismo ha jugado un rol muy importante. Por ello desarrollaremos en una primera parte los conceptos que estructuran el análisis: identidad cultural, tradición, turismo y posmodernidad (Freedman, Castells, Hall, Giddens, Bauman, Lipovetsky, McCannel, Urry) enfocándonos en proporcionar los antecedentes acerca de la realidad cultural de nuestro campo de estudio.Tras exponer esos antecedentes, se explicitará el marco metodológico que ha encauzado la investigación sobre la base de un modelo etnográfico que, a partir de la observación, el registro fotográfico y entrevistas, permitirá el análisis de los objetos artesanales y su carácter mediatizador de significados culturales tradicionales para el contexto del turismo. En una primera instancia nos abocaremos al diagnóstico del mercado de los productos de diseño artesanal que se ofrecen en Chiloé, analizándolos tanto desde una perspectiva semiótica, como desde el estructuralismo. De ello podremos evidenciar el choque o conflicto que se produce en el contexto del turismo y su impacto en los procesos de creación, generando productos ambiguos, híbridos, que se construyen desde la lógica del mercado intentando reivindicar una identidad cultural tradicional, a veces de manera inadecuada.Finalmente nos referiremos a un caso de producto de diseño de “modernidad apropiada”, concepto desarrollado por Sergio Mansilla, importante investigador sobre Chiloé, y que a modo de conclusión permitirá comprender el rol del diseño en el encuentro y diálogo entre lo local y lo global.http://dx.doi.org/10.4995/ANIAV.2017.5374
