Abstract:
OTUB1 is a deubiquitinating enzyme that cleaves Lys-48–linked polyubiquitin chains and also regulates ubiquitin signaling through a unique, noncatalytic mechanism. OTUB1 binds to a subset of E2 ubiquitin-conjugating enzymes and inhibits their activity by trapping the E2∼ubiquitin thioester and preventing ubiquitin transfer. The same set of E2s stimulate the deubiquitinating activity of OTUB1 when the E2 is not charged with ubiquitin. Previous studies have shown that, in cells, OTUB1 binds to E2-conjugating enzymes of the UBE2D (UBCH5) and UBE2E families, as well as to UBE2N (UBC13). Cellular roles have been identified for the interaction of OTUB1 with UBE2N and members of the UBE2D family, but not for interactions with UBE2E E2 enzymes. We report here a novel role for OTUB1–E2 interactions in modulating E2 protein ubiquitination. We observe that Otub1−/− knockout mice exhibit late-stage embryonic lethality. We find that OTUB1 depletion dramatically destabilizes the E2-conjugating enzyme UBE2E1 (UBCH6) in both mouse and human OTUB1 knockout cell lines. Of note, this effect is independent of the catalytic activity of OTUB1, but depends on its ability to bind to UBE2E1. We show that OTUB1 suppresses UBE2E1 autoubiquitination in vitro and in cells, thereby preventing UBE2E1 from being targeted to the proteasome for degradation. Taken together, we provide evidence that OTUB1 rescues UBE2E1 from degradation in vivo.

Abstract:
Single-nucleotide polymorphisms (SNPs) in RNF213, which encodes a 591-kD protein with AAA+ ATPase and RING E3 domains, are associated with a rare, autosomal dominant cerebrovascular disorder, moyamoya disease (MMD). MMD-associated SNPs primarily localize to the C-terminal region of RNF213, and some affect conserved residues in the RING domain. Although the autosomal dominant inheritance of MMD could most easily explained by RNF213 gain-of-function, the type of ubiquitylation catalyzed by RNF213 and the effects of MMD-associated SNPs on its E3 ligase activity have remained unclear. We found that RNF213 uses the E2-conjugating enzymes UBE2D2 and UBE2L3 to catalyze distinct ubiquitylation events. RNF213-UBED2 catalyzes K6 and, to a lesser extent, K48-dependent poly-ubiquitylation in vitro, whereas RNF213-UBE2L3 catalyzes K6-, K11-, and K48-dependent poly-ubiquitylation events. MMD-associated SNPs encode proteins with decreased E3 activity, and the most frequent MMD allele, RNF213R4810K, is a dominant-negative mutant that decreases ubiquitylation globally. By contrast, MMD-associated RNF213 SNPs do not affect ATPase activity. Our results suggest that decreased RNF213 E3 ligase activity is central to MMD pathogenesis.

Abstract:
In this study, we investigated the potential prognostic value of ubiquitin-conjugating enzyme E2D1 (UBE2D1) RNA expression in different histological subtypes of non-small-cell lung cancer (NSCLC). A retrospective study was performed by using molecular, clinicopathological, and survival data in the Cancer Genome Atlas (TCGA)—Lung Cancer. Results showed that both lung adenocarcinoma (LUAD) (N=514) and lung squamous cell carcinoma (LUSC) (N=502) tissues had significantly elevated UBE2D1 RNA expression compared to the normal tissues (p<0.001 and p=0.036, respectively). UBE2D1 RNA expression was significantly higher in LUAD than in LUSC tissues. Increased UBE2D1 RNA expression was independently associated with shorter OS (HR: 1.359, 95% CI: 1.031–1.791, p=0.029) and RFS (HR: 1.842, 95% CI: 1.353–2.508, p<0.001) in LUAD patients, but not in LUSC patients. DNA amplification was common in LUAD patients (88/551, 16.0%) and was associated with significantly upregulated UBE2D1 RNA expression. Based on these findings, we infer that UBE2D1 RNA expression might only serve as an independent prognostic indicator of unfavorable OS and RFS in LUAD, but not in LUSC.

Abstract Genetic variation in the enzymes that catalyze posttranslational modification of proteins is a potentially important source of phenotypic variation during evolution. Ubiquitination is one such modification that affects turnover of virtually all of the proteins in the cell in addition to roles in signaling and epigenetic regulation. UBE2D3 is a promiscuous E2 enzyme, which acts as an ubiquitin donor for E3 ligases that catalyze ubiquitination of developmentally important proteins. We have used protein sequence comparison of UBE2D3 orthologs to identify a position in the C-terminal α-helical region of UBE2D3 that is occupied by a conserved serine in amniotes and by alanine in anamniote vertebrate and invertebrate lineages. Acquisition of the serine (S138) in the common ancestor to modern amniotes created a phosphorylation site for Aurora B. Phosphorylation of S138 disrupts the structure of UBE2D3 and reduces the level of the protein in mouse embryonic stem cells (ESCs). Substitution of S138 with the anamniote alanine (S138A) increases the level of UBE2D3 in ESCs as well as being a gain of function early embryonic lethal mutation in mice. When mutant S138A ESCs were differentiated into extraembryonic primitive endoderm, levels of the PDGFRα and FGFR1 receptor tyrosine kinases were reduced and primitive endoderm differentiation was compromised. Proximity ligation analysis showed increased interaction between UBE2D3 and the E3 ligase CBL and between CBL and the receptor tyrosine kinases. Our results identify a sequence change that altered the ubiquitination landscape at the base of the amniote lineage with potential effects on amniote biology and evolution.

Abstract:
The aim of the present study was to determine a set of reference genes in granulosa cells of dominant follicles that are suitable for relative gene expression analyses during maternal and follicular aging. Granulosa cells of growing and preovulatory dominant follicles were collected from aged and young cows (maternal aging study) and from FSH-stimulated follicles developing under different durations of FSH treatment (follicular aging study). The mRNA levels of the two commonly used reference genes (GAPDH, ACTB) and four novel genes (UBE2D2, EIF2B2, SF3A1, RNF20) were analysed using cycle threshold values. Results revealed that mRNA levels of GAPDH, ACTB, EIF2B2, RNF20, SF3A1 and UBE2D2 were similar (P > 0.05) between dominant follicle type, age and among follicles obtained after FSH-stimulation, but differed (P = 0.005) due to mRNA processing (i.e. with versus without amplification). The stability of reference genes was analysed using GeNorm, DeltaCT and NormFinder programs and comprehensive ranking order was determined using RefFinder. The mRNA levels of GAPDH and ACTB were less stable than those of UBE2D2 and EIF2B2. The geometric mean of multiple genes (UBE2D2, EIF2B2, GAPDH and SF3A1) is a more appropriate reference control than the use of a single reference gene to compare relative gene expression among dominant and FSH-stimulated follicles during maternal and/or follicular aging studies.

Abstract All-trans retinoic acid (ATRA) has been widely used in differentiation therapy for acute promyelocytic leukemia (APL). ATRA binds to retinoic acid receptor (RAR) and triggers the formation of the transcription coactivator complex, which leads to changes in gene expression, APL cell-cycle arrest and differentiation, and clinical remission. The mechanisms responsible for ATRA's beneficial effects are still ill-defined. Here, we conducted a large-scale, unbiased short hairpin RNA (shRNA) screen aiming to identify mediators of ATRA-induced differentiation and growth arrest of APL cells. Twenty-six proteins were identified. They cover a wide range of cellular functions, including gene expression, intracellular signaling, cell death control, stress responses, and metabolic regulation, indicating the complexity of ATRA-induced cell growth control and differentiation in APL. One of these proteins, the ubiquitin-conjugating enzyme UBE2D3, is up-regulated in ATRA-treated acute promyelocytic NB4 cells. UBE2D3 is physically associated with cyclin D1 and mediates ATRA-induced cyclin D1 degradation. Knocking down UBE2D3 by RNA interference (RNAi) leads to blockage of ATRA-induced cyclin D1 degradation and cell-cycle arrest. Thus, our results highlight the involvement of the ubiquitin-mediated proteolysis pathway in ATRA-induced cell-cycle arrest and provide a novel strategy for modulating ATRA-elicited cellular effects.

The mini-scale pneumatic conveying ring dryer (PCRD) type sago starch dryer with a capacity of 80 kg / day has been applied to the processing of sago starch to produce dry sago starch. To increase the production capacity of the PCRD dryer, a recirculation pipe was modified. The modified pipe section is the venturi pipe diameter and the upriser vertical pipe increased. The diameter of the vertical upriser pipe is 2.5 times larger than that of the ubend pipe and the downcomer vertical pipe so that a buffer is formed. In addition, the difference in the diameter of the recirculation pipe can increase the residence time of the material. The purpose of this study was to simulate using ansys fluent to determine the temperature profile, air flow velocity, and pressure in the pneumatic conveying ring dryer (PCRD) type sago starch dryer pipe with a capacity of 1 ton per day. Simulations were carried out using the Computational Fluid Dynamics (CFD) technique using ansys fluent software package. The simulation results show that the temperature along the pipe has decreased by about 2oC at various variations of the input air velocity and variations in the outlet at the boundary conditions. Likewise, the air velocity at the end of the outlet pipe (vertical downcomer pipe) increases due to the difference in diameter with the inlet pipe. The pressure on the vertical upriser pipe is higher than the pressure on the ubend pipe and downcomer vertical pipe. The simulation results show that the recirculation pipe design is very well used so that it can be continued for the manufacture of PCRD-type sago starch dryer on a scale of 1 ton per day.

This study explores relocated algorithmic governance through a qualitative study of the Ubenwa health app. The Ubenwa, which was developed in Canada based on a dataset of babies from Mexico, is currently being implemented in Nigeria to detect birth asphyxia. The app serves as an ideal case for examining the socio-cultural negotiations involved in re-contextualising algorithmic technology. We conducted in-depth interviews with parents, medical practitioners and data experts in Nigeria; the interviews reveal individuals’ perceptions about algorithmic governance and self-determination. In particular, our study presents people’s insights about (1) relocated algorithms as socially dynamic ‘contextual settings’, (2) the (non)negotiable spaces that these algorithmic solutions potentially create and (3) the general implications of re-contextualising algorithmic governance. This article illustrates that relocated algorithmic solutions are perceived as ‘cosmopolitan data localisms’ that extend the spatial scales and multiply localities rather than as ‘data glocalisation’ or the indigenisation of globally distributed technology.

Mitochondrial E3 ubiquitin ligase 1 (MUL1) is located in the mitochondrial outer membrane and regulates various biological processes, including apoptosis, cell growth, mitophagy and mitochondrial dynamics. The C-terminal region of MUL1 faces the cytoplasm and contains the RING domain (MUL1-RING) where the Ub~E2 thioester binds. Unlike most RING-type E3 enzymes, MUL1-RING alone does not have an additional region that recruits a substrate protein, yet is still able to ubiquitylate the substrate, the p53 protein. Nevertheless, the exact mechanism of the ubiquitylation of p53 by MUL1-RING has not yet been elucidated. In order to understand this novel ubiquitylation mechanism, it is necessary to determine the three-dimensional structures of MUL1-RING and of its complex with the cognate E2 enzyme. Here, Ube2D2 was validated as a functional E2 enzyme for the ubiquitylation of the p53 transactivation domain (p53-TAD) by MUL1-RING, and purification and crystallization processes for MUL1-RING and the MUL1-RING–Ube2D2 complex are reported.

Bu durum İbâziyye mezhebinin fıkhî bir mezhep olarak bilinmesinin önüne geçmiştir. Bundan ötürü İbâziyye mezhebi daha çok mezheplerler tarihi ve itikâdî görüşlerinden ötürü de kelam alanında çalışılmıştır. Bu yönelim de mezhebin fıkhî yönünün ihmal edilmesine sebep olmuştur. Oysa ki İbâziyye mezhebi zengin fıkhî müktesebatı ve literatürüyle müstakil bir ameli mezheptir. Hatta İbâzîlere göre kendi mezhepleri kronolojik olarak teşekkül eden ilk fikhî mezheptir. İbâziyye mezhebi Abdullah b. İbâz’a nispet olunmakla birlikte İbâziler Câbir b. Zeyd’i müessis olarak kabul ederler. İbâzîler hicrî 3. asrın sonralarına kadar kendilerini İbâzî olarak isimlendirmekten kaçınmışlardır. Çünkü İbâzîler fıkhî mezheplerin, kurucularına nispet edilmesini gerektiğini, mezhep kurucularının da Cabir b. Zeyd olduğunu söylerler. Mezheplerinin Abdullah b. İbâz’a nispet edilmesinin siyasi saikler nedeniyle olduğunu ısrarla vurgularlar. Bundan ötürü İbâzîler mezheplerinin müessisi olarak Câbir. b. Zeyd’i görürler. Câbir tâbiî alimlerinden olup Basra müftüsü olmasıyla da şöhret bulmuştur. İbâziyye mezhebinin Câbir’den sonra en önemli ismi olarak kabul edilen Ebû Ubeyde Muslim b. Ebî Kerime mezhebin teşekkülünde ve sistemleşmesinde çok önemli bir yere sahiptir. Ebû Ubeyde Gerek Emevîler gerekse de Abbâsîler döneminde İbâzîlere yönelik baskılara rağmen mezhebi bir arada tutarak Basra dışına özellikle de Kuzey Afrika’da yayılmasını sağlamıştır. İbâziyye mezhebinin hicrî 2. asrın ikinci çeyreğinden itibaren farklı coğrafyalara taşınması bu mezhebin sistemleşmesi açısından önemli bir merhale olmuştur. Ayrıca İbâziyye mezhebinin Basra dışındaki bölgelere yayılması İbâzî fıkıhla ilgili tedvinlerin artmasını sağlamıştır. İbâzî fıkhının en önemli eserleri arasında ilk sıralarda olan Ebû Ğânım el-Horasâni’ye ait olan el-Müdevvetü’l-Kübrâ bu dönemin mahsulüdür. İbâziyye mezhebi sünnî mezheplerin fıkhî meselelerde başvurdukları istinbât ve istidlâl yönteminden farklı bir yöntem kullanmamıştır. Kur’an, Sünnet, İcmâ, Kıyas ve diğer İstidlâl yöntemleri İbâzî fakihlerce de başvurulan yöntemler olmuştur. Dolayısıyla İbâzîlerin kullanmış olduğu usûl yöntemlerinin diğer mezheplerin usûl yöntemiyle bire bir aynı olmasa da bir birine çok yakın olduğunu söyleyebiliriz. İbâzîlerin istidlali deliller kısmında diğer mezheplerden farklılaşması ilham konusunda olmuştur. İbâzilerde tartışmalı olmakla birlikte ilham istidlali delillerden biri olarak zikredilmiştir. Bu çalışmamızda mezhebin teşekkülü esnasında Haricîlerle olan bağlantısı, mezhebin ortaya çıkışı, fıkhı ve usülüne değineceğiz.

Abstract:
Patients with diabetes are physiologically frail and more likely to suffer from infections and even life-threatening sepsis. This study aimed to identify and verify potential biomarkers of diabetes-related sepsis (DRS). Datasets GSE7014, GSE57065, and GSE95233 from the Gene Expression Omnibus were used to explore diabetes- and sepsis-related differentially expressed genes (DEGs). Gene set enrichment analysis (GSEA) and functional analyses were performed to explore potential functions and pathways associated with sepsis and diabetes. Weighted gene co-expression network analysis (WGCNA) was performed to identify diabetes- and sepsis-related modules. Functional enrichment analysis was performed to determine the characteristics and pathways of key modules. Intersecting DEGs that were also present in key modules were considered as common DEGs. Protein-protein interaction (PPI) network and key genes were analyzed to screen hub genes involved in DRS development. A mouse C57 BL/6J-DRS model and a neural network prediction model were constructed to verify the relationship between hub genes and DRS. In total, 7457 diabetes-related DEGs and 2606 sepsis-related DEGs were identified. GSEA indicated that gene datasets associated with diabetes and sepsis were mainly enriched in metabolic processes linked to inflammatory responses and reactive oxygen species, respectively. WGCNA indicated that grey60 and brown modules were diabetes- and sepsis-related key modules, respectively. Functional analysis showed that grey60 module genes were mainly enriched in cell morphogenesis, heart development, and the PI3K-Akt signaling pathway, whereas genes from the brown module were mainly enriched in organelle inner membrane, mitochondrion organization, and oxidative phosphorylation. UBE2D1, IDH1, DLD, ATP5C1, COX6C, and COX7C were identified as hub genes in the PPI network. Animal DRS and neural network prediction models indicated that the expression levels of UBE2D1 and COX7C in DRS models and samples were higher than control mice. UBE2D1 and COX7C were identified as potential biomarkers of DRS. These findings may help develop treatment strategies for DRS.

Abstract:
Objective. In minimally invasive spinal surgery, the treatment of lumbar spinal stenosis with microendoscopic discectomy (MED) or unilateral biportal endoscopic discectomy (UBED) shows effective results, but which is more effective is controversial. Our study aimed to evaluate the efficacy and safety of UBED versus MED in the treatment of lumbar spinal stenosis by a systematic review and meta-analysis, so as to provide reference for the promotion of UBED in clinical practice. Methods. The multiple databases like PubMed, EMBASE, Web of Science, Cochrane Library, Chinese National Knowledge Databases, Chinese BioMedical Database, and Wanfang Database were used to search for the relevant studies. Review Manager 5.4 was adopted to estimate the effects of the results among selected articles. Odds ratio (OR) and mean difference (MD) with 95% confidence intervals (CIs) were used to estimate the overall pooled effect. Subgroup analysis, forest plots, funnel plots and Egger’s test for the articles included were also conducted. Results. Three randomized clinical trials and seven cohort studies were finally retrieved, these studies included 685 and 829 patients in the UBED and MED groups, respectively. There were no differences in terms of operation time (MD = -0.92, P =0.72), estimated blood loss (MD = -26.31, P =0.08), complications (MD =0.81, P =0.38) and Oswestry Disability Index (ODI) score (P >0.05 in four subgroup) between the two groups. The visual analog scale (VAS) score of back pain in the UBED group was better than MED group only at 6 months (MD = -0.23, P =0.006) after operation, the VAS score of leg pain in the UBED group was better than that of MED group at 3 mouths (MD = -0.22, P =0.002) and 6 months (MD = -0.24, P =0.006) after operation, the UBED group had a less postoperative length of stay than the MED group (MD = -1.85, P <0.001). The bias analysis showed that there was no potential publication bias in the included literature. Conclusion. This study showed that compared with MED, UBED has the advantages of short hospital stay and good short-term curative effect, but there is no significant difference in long-term efficacy and safety, they can be replaced by each other in clinical application.

Abstract Cleavage by the endonuclease CPSF73 and polyadenylation of nascent RNA is an essential step in co-transcriptional mRNA maturation, and abnormal poly(A) site choices have been associated with human diseases, including cancer. Recent work has identified CPSF73 as a promising drug target for inhibiting the growth of specific cancers, and higher level of CPSF73 correlates to poor survival in breast cancer patients. However, our understanding of the regulation of CPSF73 is still at an early stage. Here, we report that a HECT-like E3 ligase, UBE3D, participates in stabilizing CPFS73 protein by preventing its ubiquitin-mediated degradation by the proteasome. Depletion of UBE3D in both non-cancer cells and cancer cells leads to CPSF73 downregulation, a pre-mRNA cleavage defect and dysregulated gene expression. UBE3D dysfunction or chemical inactivation of CPFS73 inhibited migration and invasion as well as stem cell renewal phenotypes in vitro in triple negative breast cancer cells. In addition, genetic overexpression of CPSF73 promoted breast cancer stemness and knocking down of CPSF73 inhibited stem cell renewal property. Together, our findings indicate that targeting the pre-mRNA processing nuclease CPSF73 has potential for breast cancer therapy. This work was supported by the NSF grant NS0224 to C. Abstract Background: The blood of cancer patients is known to contain fragments of RNA released from the tumor. The application of genomic profiling techniques to plasma RNA may allow the unbiased selection of cancer markers in the blood, but the informative value of genomic profiling of plasma RNA is currently unknown. Methods: We used cDNA microarray hybridization to perform genomic profiling of plasma RNA from colorectal cancer (CRC) patients and from healthy donors. From a list of 40 genes differentially upregulated in cancer patients, we randomly selected 4 genes for further characterization. These 4 markers were analyzed by quantitative reverse-transcription PCR in a wide set of samples including paired samples from the same CRC patients before and after surgical resection of the tumor. Results: Three of the selected markers—EPAS1, UBE2D3, and KIAA0101—were confirmed by PCR to be significantly increased in cancer compared to healthy donors. Importantly, 2 of the markers, EPAS1 and UBE2D3, showed a significant decrease after surgery, returning to the levels of healthy donors. Finally, supervised class prediction using these 3 markers correctly (77%) assigned presurgery samples to the CRC group and assigned postsurgery samples from the same patients to the healthy group. Conclusions: Our findings demonstrate the usefulness of gene expression profiling of circulating plasma RNA to find cancer markers of potential clinical value.

Abstract Absorbable stents made of poly-L-lactic acid (PLLA) exhibit inferior mechanical properties compared with metal based devices. Therefore, design optimization by means of finite element analysis (FEA) plays an important role in development of high performance absorbable stents. In the present study, three different meander structures made of PLLA material were compared in terms of their radial resistive force (RRF) using FEA. Material tests were performed to obtain input parameters of PLLA for the simulation. FEA was carried out to simulate the RRF and the length change of the meander structures as a function of the diameter. In addition, the numerical results were compared to the experimental outcome using corresponding meander based stents performing radial testing with a segmented head mechanism. Comparable high stresses occurred in the Ubends of the examined stent designs. In designs with a large radius of curvature less stress was observed while crimping. One meander structure developed comparatively higher structural resistance against radially acting forces, due to its curved strut design. In general, FEA revealed consistent results compared to the experimental investigations.

Abstract: Drehem, ancient Puzriš-Dagān, is well known as the place of origin of more than 15,000 cuneiform tablets from the Ur III period that were sold on the antiquities markets from 1909 onwards. The State Board of Antiquities and Heritage of Iraq undertook the first controlled excavations at the site in 2007 under the direction of Ali Ubeid Shalkham. The cuneiform texts and fragments found there not only add to the well-known royal archives dealing with cattle, treasure or shoes, but they include many records on crafts and agriculture. With this evidence, the subsistence economy behind this important administrative center and royal palace of the Third Dynasty of Ur becomes more evident. We thank the Iraqi State Board of Antiquities and Heritage, the Iraq Museum Baghdad and Mr Ali Ubeid Shalkham for the permission to publish the tablets from the excavation season of 2007. We present a case of a dural tear associated with nerve root herniation following unilateral biportal endoscopic decompression (UBED) that was successfully treated using a computed tomography-guided epidural blood patch. A 60-year-old man underwent UBED for radicular pain because of spinal stenosis at L4–5. A left partial hemilaminectomy and flavectomy were performed; however, the left dorsolateral side dura mater was torn during the procedure. TachoComb® was applied at the dural tear site, and the pain was relieved following UBED. However, 3 weeks post-UBED, the patient reported severe pain with an electric shock-like sensation in the left buttock and posterior thigh region with no other neurologic symptoms. The pain was aggravated by standing and spinal motion. Follow-up lumbar spinal magnetic resonance imaging was performed. Axial images indicated protrusion of the left S2 nerve root through the left dorsolateral side of the dura mater. The patient was further diagnosed with nerve root herniation following a dura mater tear. A computed tomography-guided epidural blood patch was performed, with successful therapeutic results. The outcome of this study indicates that a small dural tear that occurs during minimally invasive spinal surgery can be efficiently treated using an epidural blood patch prior to open surgery.

ABSTRACT DET1 (de-etiolated 1) is an essential negative regulator of plant light responses, and it is a component of the Arabidopsis thaliana CDD complex containing DDB1 and COP10 ubiquitin E2 variant. Human DET1 has recently been isolated as one of the DDB1- and Cul4A-associated factors, along with an array of WD40-containing substrate receptors of the Cul4A-DDB1 ubiquitin ligase. However, DET1 differs from conventional substrate receptors of cullin E3 ligases in both biochemical behavior and activity. Here we report that mammalian DET1 forms stable DDD-E2 complexes, consisting of DDB1, DDA1 (DET1, DDB1 associated 1), and a member of the UBE2E group of canonical ubiquitin-conjugating enzymes. DDD-E2 complexes interact with multiple ubiquitin E3 ligases. We show that the E2 component cannot maintain the ubiquitin thioester linkage once bound to the DDD core, rendering mammalian DDD-E2 equivalent to the Arabidopsis CDD complex. While free UBE2E-3 is active and able to enhance UbcH5/Cul4A activity, the DDD core specifically inhibits Cul4A-dependent polyubiquitin chain assembly in vitro. Overexpression of DET1 inhibits UV-induced CDT1 degradation in cultured cells. These findings demonstrate that the conserved DET1 complex modulates Cul4A functions by a novel mechanism.

Novi pokreti usmeravaju svoje postupke prema civilnim vlastima i političarima koji imaju moć da realizuju ciljeve pokreta, ali često postoji sukob između pokreta i vlasti, odnosno između pokreta i utvrđenog društva koje vlasti predstavljaju. Ti pokreti veruju da su establišment politike propale i da više ne mogu da predstavljaju zadovoljavajuća rešenja za teške političke probleme. Tri najvažnija pristupa društvenih teorija prepoznaju sledeće koncepte: koncept mobilizacije resursa i koncept struktura političkih oportuniteta i koncept freminga (framing), odnosno okvira. Relativni uspeh pobune protiv establišmenta zavisi, u krajnjoj liniji, od sposobnosti političkih aktera da ubede potencijalne pristalice da oni zaista stoje nasuprot elita moći, da nisu deo strukture elite moći, da nisu insajderi. Uspeh pokreta zavisi od širokog prihvatanja poruke, a prihvatanje poruke će zavisiti od toga koliko se ona uklapa u percepciju stvarnosti ili koliko se dobro percepcijom može izmanipulisati. Međutim u Srbiji se samo na početku činilo da će ovi novi pokreti biti vesnici novih političkih trendova i novog svetla u postojeći sistem. Oni su zapravo profitirali samo od slabljenja određenih opozicionih partija ili sindikata, ali kratkog daha. Nisu uspeli i nisu mogli da ustroje stalnu organizaciju i strukturu, jedinstveno članstvo i program.

Abstract:
Tertullian war durch den ganzen Zeitraum seines Lebens und Schaffens der Sprecher der Diszipłin. Er bemerkte in der christlichen Gemeinschaft diese, die leiten sie, und andere, die sind ihnen unterordnen. Bischofe, Priester und Diakone gehoren zu erster Gruppe. Die Bischofe leiten besonders die Kirche und sie sind Seelsorger. Sie bewahren auch „munera sacerdotałia". Diesem Gesetz des Bischofs verleugnete Tertullian niemals deutlich. Als zweite, separate Gruppe in der christlichen Gemeinschaft spezifiziert Tertullian die Laien. „Laici", ein mal verheiratete Manner, die zu „ordo Ecclesie" (Kirchrangordnung) nicht gehoren, haben auch die priesterliche Wurde. Die priesterliche Wurde der Glaubigen hat der allgemeine Charakter. Die priesterliche Funktionen uben in der christlichen Gemeinschaft Bischofe, Priester und Diakone, mit der Rucksicht auf der Disziplin und der Ordnung in der Kirche. In den notwendigen Zufallen uben es auch die Laien, das heiBt die Manner. Tertullian lehnt deutlich das Gesetz der Frauen die priesterliche Funktionen auszuuben ab. Trotz der deutlichen Absonderung durch Tertullian zwei Zustande in der Kirche, ist die Linie der Aufteilung zwischen Geistlichern und Laien in den Zeiten Tertullians in dem Grund der Sache genug gelaufig.
