Abstract:
The Master Thesis is focused on potential consequences of toll implication for vehicles under 3,5 tonnes. The main goal of this paper is to evaluate these consequences and propose a recommendation to the government whether to imply the toll or not. Quantitative research methods in a form of questionnaire were used in order to estimate the potential consequences of 0,4 CZK per km toll for vehicles under 3,5 tonnes. Moreover, was analysed the publicly accessible data of transportation, car accidents and deaths on roads in case of the Czech Republic. Theoretical part is focused on the currently used system on highways, the history of highways, system used in European countries and aspects of cost-benefit analysis. Three frequently used highways were examined in the practical part of this paper. A numerous of calculations were used in order to quantify all possible costs of toll implication. Toll implication costs and additional costs that arise due to avoiding expensive highways by some drivers represented the main aim of these calculations. Conclusion of this paper sums up key points of this paper and points out possible negative consequences of toll implication.

Abstract:
The ability to manage a fish stock relies on an understanding of life history characteristics and basic biology of the species. Numerous age-growth studies are facilitated by the relative ease of ageing fishes through hard-part analyses. Determining reproductive parameters for fish populations is equally important for stock assessments and management, and histological examination of gonads provides the most accurate determination of fecundity and spawning periods. Coastal pelagic fishes are often targeted commercially and recreationally due to their easy access by private vessels. However, there are few studies researching the biology and reproduction of recreational fishes in the waters of Southeastern Florida that would lead to a better understanding for management practices. The objective of this study was to provide baseline reproductive data for two fishes particularly important to Florida fisheries: the coastal pelagic scombrids Blackfin Tuna Thunnus atlanticus and Little Tunny Euthynnus alletteratus. Archived gonad samples from 2010-2014 for these two species were evaluated, and GSI values and histological examination indicated one spawning season for both species. Little Tunny spawn April through August, and Blackfin Tuna spawn May through June. Additionally both species have asynchronous oocyte development, and are batch spawners. Postovulatory follicles were used to estimate spawning frequency; for Blackfin Tuna, mean spawning occurs once every 1.49 days, and for Little Tunny, mean spawning occurs once every 1.47 days. Comparison of otolith age data to these results indicates that Little Tunny mature at a smaller size and younger age than Blackfin Tuna. Size at 50% maturity for male Blackfin Tuna was 435.2 mm TL, for female Blackfin Tuna was 392.3 mm TL, and for male Little Tunny was 347.77 mm TL. Age at 50% majority for male Blackfin Tuna was 0.66 years, and for male Little Tunny was 0.50 years. In addition to providing important baseline data for fisheries management, this study collaborated with previous research to improve accuracy of reproductive age assessments. Finally, reproductive parameter studies of fishes in Florida and the Greater Caribbean area commercial and recreational fisheries provide information important for future ecosystem based management.

Abstract:
Understanding the life history of a species is essential for fully understanding its role within an ecosystem. However, many of the fish species of high ecological value have not been studied due to their less prominent roles in local recreational and commercial fisheries in comparison to other targeted species. This study describes the age and growth patterns of three small tuna species inhabiting South Florida waters: blackfin tuna Thunnus atlanticus, little tunny Euthynnus alletteratus, and skipjack tuna Katsuwonus pelamis. Tuna specimens were collected via donations obtained from various fishing tournaments and charter captains in the areas of the Florida Straits as well as hook-and-line by the Nova Southeastern University Oceanographic Center. Age was described via sagittal otolith deposition patterns. They were removed, dried, sectioned, and rings were counted as well as measured. Validation of the timing of ring deposits was done by marginal increment analysis. Growth parameters were determined by comparison of fish fork length to count measurements. This comparison via the Von Bertalanffy growth equation produced a growth rate for each species: blackfin, L∞ = 95.34 cm, K = 0.28, and t0 = -1.53; little tunny, L∞ = 77.93 cm, K = 0.69, and t0 = -0.69; and skipjack, L∞ = 112.76 cm, K = 0.24, and t0 = -1.70. The curves indicate an average size of an individual of a given species at a certain age. They also give an estimation of a maximum length (L∞) of each species, in addition to specific growth rate, which is indicated by the slope. Parameters of each resulting Von Bertalanffy equation were compared among species. Results were also compared with growth rates currently used in stock assessments by fisheries management organizations, such as the International Commission for the Conservation of Atlantic Tunas (ICCAT).

Abstract:
Commercial capture-based aquaculture of the Atlantic bluefin tuna (ABT), Thunnus thynnus (L.), has been prominent in the Mediterranean for over a decade. Owing to several limitations encountered in working with the species, including its high commercial value, there has been little research carried out relating to this species. The objective of this study was to examine several health parameters of captive ABT. These included an examination of coronary artery lesions, haematology, plasma biochemistry, assessment of immune function and changes in fatty acid (FA) flesh content through the on-growing period. Arteriosclerosis in fish is a pathologic condition of uncertain etiology and involves the main coronary artery in teleosts. Apart from reports of their widespread occurrence in salmonids, they have been described from a restricted number of wild ABT specimens but have not received further attention. This investigation analysed the effect of size and period of net-pen rearing on the prevalence and severity of arteriosclerotic lesions in ABT. Coronary arteries from wild and captive fish were investigated and prevalence was 100 %, but increasing structural degradation was observed with increasing fish size, suggesting that lesions progress throughout the life of the fish. Due to the limited availability of wild specimens, the effect of captivity on arteriosclerosis in ABT could not be adequately quantified, although observations suggest that the farming process has no major effect on arteriosclerotic lesions in ABT. Studies on the haematology, plasma biochemistry and immunology of ABT are limited. Haematological and plasma biochemical indices are useful in animal health assessment but use of these requires the establishment of species-specific ranges. Blood was collected from captive ABT specimens of varying weight (61-361 kg) and the major haematological (n = 45), plasma biochemical (n = 30) and immunological parameters (n = 45) were quantified. Size-based differences were found in haematological indices between experimental sub-groups including increased erythrocyte number and haemoglobin level in smaller ABT. No differences were found in immunological parameters except for total IgM levels, which were higher in the smaller individuals. Preliminary investigations indicated that disease prevalence in captive ABT is very low. Epidermal mucus is an important interface between fish and their environment and comprises immunological components which act as a first barrier against pathogen entry or colonisation. Mucus was collected from captive ABT and analysed for innate immune components. The presence of IgM was detected in the mucus of ABT by an enzyme-linked immunosorbent assay and several different enzymes were detected with an API-ZYM kit assay. Zymography experiments confirmed the presence of protease-like enzymes in the mucus, while enzyme assays quantified alkaline phosphatase, protease, esterase and cathepsin B activities. Lysozyme levels were high. The mucus agglutinated sheep erythrocytes but did not demonstrate complement or bacteriolytic activity. There is restricted information on the fatty acid composition of farmed ABT or how this is influenced when the fish are held under commercial aquaculture conditions. This study investigated the FA composition of farmed ABT, its variation by dorsal muscle region and the correlation between dietary FA composition with that of the fish. Analysis of flesh samples retrieved from farmed ABT did not reveal significant differences in the FA composition of experimental sub-groups irrespective of size, time held in captivity or diet. These results indicate that FA metabolism in ABT is substrate-selective. Gene expression measurements from several organs of ABT showed that expression of Δfad5 and elovl5, genes involved in FA metabolism, were highest in the brain followed by the liver but no expression of these genes was detected in the spleen. The findings of this research address aspects of health evaluation and nutritional status in farmed ABT and are discussed in terms of farming practice. Conclusions from some of these studies suggest that the practice of holding wild-captured stock in cages for periods of up to 18 months does not result in significant impact on ABT.

Abstract Based on the Romer and Romer (1989) methodology this paper analyzes behavior of the US Federal Reserve System (FED) and the Czech National Bank (CNB) during the recent crisis. It explores minutes and press releases from the meetings of the Bank Board of the CNB and the Federal Open Market Committee of the FED, i.e. on one hand from the side of a small open economy that has been hit mainly through decline in foreign demand, on the other from the side of a big closed economy, in which the recent crisis has originated. It compares reaction in interest rate and adequacy of unconventional measures with a prediction of a simple 'Taylor rule' (Taylor, 1993) and tries to evaluate the adequacy of the overall reaction of mentioned central banks to the crisis.

Då datorer i alla dess former har kommit att bära betydande mängder relevant information för diverse forensiska utredningar så krävs en ingående kunskap gällande hur denna data lämpligast infångas. En typ av klient som nyligen har gjort sitt kommersiella intåg på marknaden är den internetbaserade tunna klienten. Det existerar i dagsläget begränsat med underlag gällande huruvida en sådan klient bär information som är relevant för en IT-forensisk utredning. Inom denna uppsats har därför en internetbaserad tunn klient av typen Chromebook varit mål för undersökning. Syftet har varit att kartlägga vilka sårbarheter som existerar, samt om möjligt huruvida dessa kan brukas som underlag vid en forensisk utredning. För detta ändamål har klientens lagrings- samt kommunikationsmönster analyserats. För genomförandet av detta brukades experimentella metodiker i form av penetrationstest samt IT-forensisk undersökning. Det har även kartlagts huruvida programvaror avsedda för operativsystemet Linux kan exekveras på klienten. Ändamålet med detta var att fastställa om klientens volatila minne kan utvinnas med hjälp av verktyg för just Linux. Rapportens resultat påvisar att det inom en klient av denna typ finns forensiskt värdefull information att inhämta. Det påvisas även att Chrome OS från en användares perspektiv kan anses som säkert.
Computers in various forms have come to carry a significant amount of relevant information for various forensic investigations. Therefore, detailed knowledge is required regarding how this data is best acquired. One type of client that has recently made its entry onto the commercial market is the Web Thin Client. As it is today, only a limited amount of relevant information can be found regarding in what capacity such a client contains data that is relevant to an IT-forensic investigation. For this reason, a Web thin client of the model Google Chromebook has been evaluated in this paper. The purpose of this examination has been to identify the vulnerabilities that currently exist, and if possible whether these can be used as a basis for a forensic investigation. To achieve this, the client's storage and communication patterns have been analyzed by implementation of experimental methodologies. The methods used for the purpose are penetration testing and an IT forensic investigation. Tests have also been performed to evaluate whether software for the Linux operating system can be executed on the client. The purpose of this was to determine whether the client's volatile memory can be extracted using tools created for Linux. The results presented in this paper demonstrate the fact that within a client of this type, valuable forensic information can be located. It is also established that Chrome OS from a user's perspective can be considered as safe.

Abstract:
Tuna are the major marine fishery in Sri Lanka, and yellowfin tuna (YFT) (Thunnus albacares) and skipjack tuna (SJT) (Katsuwonus pelamis) represent 94% of all tuna caught. The tuna catch in Sri Lanka has increased rapidly over recent years and this is true generally for the Indian Ocean. Tuna are a major animal protein source for 20 million people in Sri Lanka, while marine fisheries provide the main income source for most Sri Lankan coastal communities. While the importance of the fishery will require effective stock management practices to be employed, to date no genetic studies have been undertaken to assess wild stock structure in Sri Lankan waters as a basis for developing effective stock management practices for tuna in the future. This thesis undertook such a genetic analysis of Sri Lankan T. albacares and K. pelamis stocks. Samples of both YFT and SJT were collected over four years (2001 - 2004) from seven fishing grounds around Sri Lanka, and also from the Laccadive and Maldive Islands in the western Indian Ocean. Partial mitochondrial DNA (mtDNA) ATPase 6 and 8 genes and nuclear DNA (nDNA) microsatellite variation were examined for relatively large samples of each species to document genetic diversity within and among sampled sites and hence to infer stock structure and dispersal behaviour. Data for YFT showed significant genetic differentiation for mtDNA only among specific sites and hence provided some evidence for spatial genetic structure. Spatial Analysis of Molecular Variance (SAMOVA) analysis suggests that three geographically meaningful YFT groups are present. Specifically, one group comprising a single site on the Sri Lankan west coast, a second group comprising a single site on the east coast and a third group of remaining sites around Sri Lanka and the Maldive Islands. Patterns of variation at nDNA loci in contrast, indicate extensive contemporary gene flow among all sites and reflect very large population sizes. For SJT, both mtDNA and nDNA data showed high levels of genetic differentiation among all sampling sites and hence evidence for extensive spatial genetic heterogeneity. MtDNA data also indicated temporal variation within sites, among years. As for YFT, three distinct SJT groups were identified with SAMOVA; The Maldive Islands in the western Indian Ocean comprising one site, a second group comprising a single site on the east coast and a third group of remaining sites around Sri Lanka and the Laccadive Islands. The mtDNA data analyses indicated two divergent (M^ = 1.85% ) SJT clades were present among the samples at all sample sites. SJT nDNA results support the inference that multiple 'sub populations' co-exist at all sample sites, albeit in different frequencies. It appears that variation in the relative frequencies of each clade per site accounts for much of the observed genetic differentiation among sites while effective populations remain extremely large. Based on combined data sets for management purposes therefore, there is no strong evidence in these data to indicate that more than a single YFT stock is present in Sri Lankan waters. For SJT however, evidence exists for two divergent clades that are admixed but not apparently interbreeding around Sri Lanka. The identity of spawning grounds of these two clades is currently unknown but is likely to be geographically distant from Sri Lanka. Spawning grounds of the two distinct SJT clades should be identified and conserved.

Abstract:
Tuna are arguably the world’s most valuable, versatile, yet vulnerable fishes. With current landings over 4 million tonnes annually, all species of tuna from all three major ocean basins are caught, traded, and consumed at various intensities around the globe. Understanding the implications of such an extensive industry is paramount to protecting the long-term health and sustainability of both the tuna fisheries as well as the ecosystems in which they operate. Given that the Pacific Ocean accounts for roughly two-thirds of the global commercial tuna catch, this thesis assesses the trends and ecological impacts of commercial tuna fishing at both the artisanal and industrial scale in this ocean. To observe the importance of tuna fisheries at a local scale, a case study of the Galápagos Islands is presented. In this context, it was observed that over-fishing and the subsequent depletion of large, low fecund serranids has resulted in a high level of ‘fishing down’ within the near-shore ecosystem. Consequently, as fishers are forced to expand to regions off-shore, tuna and coastal scombrids are becoming increasingly targeted. With regard to industrial fishing, tuna vessels (especially distant-water longliners) are known to generate a substantial amount of associated bycatch and discards. The second component of this thesis quantified the amount of bycatch (retained and discarded) generated by Pacific tuna fishing fleets from 1950 to 2010. Unreported retained bycatch amounted to 1.4 million t; the total discarded catch associated with tuna fishing was 3.6 million t of target species and 7.9 million t of non-target species; sharks were the most commonly discarded species. These totals represent about 14% of the reported landings during this time. Lastly, an analysis of the applicability of the ‘Catch-MSY’ method developed by Martell and Froese (2012) in the context of large pelagic fishes is presented. It was observed that this method produces MSY estimates highly correlated to those produced by complete stock assessments. Collectively, the results of this thesis suggest that the tools to adequately manage tuna exist; however, proper data collection is rare, and the implementation of adequate sustainable fishing measures by fisheries managers is still wanting.

Nous avons essaye de montrer par des analyses litteraires de periodes diverses, les caracteristiques de la dependance culturelle, c'est a dire, le rapport qui s'etablit entre le retard et le progres et qui anime notre vie intellectuelle depuis le xvieme siecle. Ainsi, la litterature coloniale, analysee a partir des lettres jesuites, correspondrait au stade du simulacre et le probleme de la dependance pourrait etre expliquee par une metaphore - celle de table rase la periode qui suit, situee entre le romantisme et le debut du xxeme siecle est l'une des plus fertiles et des plus contradictoires par rapport a la question de la dependance. Nous avons pris comme exemple l'oeuvre de joaquim nabuco, minha formacao concue entre 1893 et 1899. La troisieme periode analysee concerne l'avant-garde des annees soixante, representee par deux mouvements esthetiques : le neo- concretisme et le cinema novo, a travers son theoricien et cineaste glauber rocha. Le mouvement qui oriente la critique contemporaine semble etre le renversement total de la question de la dependance. Au lieu de la considerer comme un phenomene typique du sous-developpement latino-americain, on en vient a la considerer comme une specificite de ces societes. C'est pourquoi nous pouvons parler de la reflexion sur la dependance culturelle, s'elaborant, paradoxalement a l'interieur des societes dites dominees comme un contre-discours
We have tried to show through literary analyses of various periods of time, the particularties of cultural dependancy, which is the relationship between backwardness and progress which characterizes our intellectual life since the xvie century. Therefore, colonial literature, analysed througgh jesuites letters could correspond to simulacre stage and the problem of dependancy can be explained by a metaphore -tabula rasa. The following part, between the romantic mouvement and the early xxe century is one of the most fertile and most contradictory as far as the question of dependancy is concerned. Example : the works of joaquim nabuco, minha formacao, written between 1893-1899. The third period we have analysed deals with the "avant-garde" of the 1960's represented by two esthetic movements : the neo-concretism and the cinema novo through the work of film makes such as glauber rocha. Instead of considering the question of dependancy as a tipical phenomenon of under-development of latin america societies, we consider it as specific of these dominated societies and therefore, paradoxally, assert our reflexions a "contre-discours"

Esta dissertação intenciona investigar a construção do cômico, mediado pela tecnologia, em vídeos remix publicados na plataforma YouTube. Os trabalhos analisados propõem o riso a partir da manipulação de tempo e pitch do áudio de vídeos virais, possibilitada por softwares como Auto-Tune e Melodyne. Entendemos remix conforme autores como Lev Manovich, Eduardo Navas e Lawrence Lessig, que afirmam que vivemos em uma \"Cultura Remix\", uma época que tem como base cultural o sampling, atividade que consiste na troca inteligente entre fragmentos de diversas mídias. Analisaremos a emergência do riso nesse contexto, a partir de reapropriações de materiais preexistentes na rede. Autores como Henri Bergson e Michail Bakhtin nos fornecem o suporte conceitual para investigarmos o riso como um recurso dramático e da cultura tanto na vida cotidiana quanto nas plataformas digitais, que facilitaram, por um lado, o acesso e reciclagem de \"detritos midiáticos\", e, por outro, potencializaram as possibilidades de manipulação desses materiais
This dissertation intends to investigate the construction of the comic, mediated by technology, in video remix published on the YouTube platform. The analyzed works propose the laughter from the manipulation of time and pitch of the audio of viral videos, made possible by softwares like Auto-Tune and Melodyne. We understand remix by authors such as Lev Manovich, Eduardo Navas and Lawrence Lessig, who claim that we live in a \"Culture Remix\", an era that is based on cultural sampling, an activity that consists of the intelligent exchange between fragments of various media. We will analyze the emergence of laughter in this context, from re-appropriation of pre-existing materials in the network. Authors such as Henri Bergson and Michail Bakhtin provide us with conceptual support for investigating laughter as a dramatic and cultural resource in everyday life as well as on digital platforms that facilitated access to and recycling of \"media debris\" on the one hand, and , on the other, potentialized the possibilities of manipulation of these materials.

This thesis deals with the optimization of the production technology of the cap tube made by injection molding. Series production is set at 5 000 000 units per year; the minimum time of the production is 5 years. A literary study is developed, focusing on the technology of injection molding in molds including the possibilities of the optimization of the production by changing the structure of the mold. The practical part of the thesis is devoted to the optimization of existing mold with cold runner system for the production of the cap tube, using the hot runner system supplied by Synventive. The change of the structure of the mold involves changing the three plate mold to a two plate mold. It is about changing the fixed part of the mold in which the hot runner system is placed; it is composed of standards parts of the company HASCO. Based on the technological parameters the injection machine Krauss Maffei KM 80-380 CX is chosen. There is a technical-economical evaluation of the optimization of the production by using the hot runner system at the end of the thesis.

There are many challenges when automating industrial processes. Successful automation projects can lead to several benefits such as lower manufacturing costs, improved quality and reduced lead times. The purpose of this study is to identify the biggest challenges when automating processes within heavy industry and then, find ways to solve these. The study has an abductive approach, where focus has shifted between literature and empirical data. Empirical data has been gathered through a case study. The case study is carried out at a company that operates within heavy industry. It’s a pre-study before an automation project. The case study has followed methods presented in literature. Challenges that proved most difficult during automation within heavy industry were: Adapting a product for automatic manufacturing and product variations. The conclusion drawn from the results and experiences during the case study, is that the most comprehensive solution to these challenges is continuous improvements of products and processes.

The Iron Age cemetery at Tuna in Badelunda parish, Västmanland, is a complex and unique burial ground used for only 69 graves during a period of roughly 700 years, between 300 - 1050 AC. The individuals buried at Tuna show an impressive variety of gravegods as well as stonesettings formed above the grave. Who where these people that were cremated at Tuna, among the mysterious women in the boats and the rich women in grave X? As we study the graves of the cremated individuals we reach a new understanding of the cemetery; from its social structure down to every individual. Through the analysis of the stonesettings, gravegods and bones we see a how these subjects, when analysed, show a picture of the individuals and social structure of the cemetery. It was a cemetery for the higher society and unique individuals rather then the average people.

Tuna are of significant global economic importance and a prime food source. Increased levels of fishing have resulted in many stocks being under threat and a number of species are considered to be overfished. The South African tuna industry has had limited management attention from the South African fisheries management agency. A recent development has been an increase in the number of tuna caught by small vessels that target the fresh tuna market in South Africa and overseas. This has highlighted the importance of developing a holistic management plan for the sector and creating an awareness, among vessel owners, of the importance of compliance with initiatives such as the Marine Stewardship Council (MSC). The South African tuna fishery comprises three sectors: baitboat, sport and longline, all of which are currently in need of acquiring more biological and fisheries data. This project was initiated to collate existing information and to collect additional information where possible. This encompassed a qualitative and quantitative assessment of the size and shape of the tuna fishing industry, which included estimates of total catch, effort, catch-per-unit-of-effort (CPUE) and stock structure (obtained through comparative estimates of age and growth), as well as socio-economic and economic information. A technique involving an examination of specific vertebrae was used to obtain age-growth information for T. albacares. These data were used to estimate von Bertalanffy (VBGF) growth parameters: F 2 1. , k 0.1 , and t₀ -0. 1 year. No significant differences in growth parameters were found in yellowfin tuna (Thunnus albacares) from different localities around the South African coast, i.e. from the south eastern Atlantic and the south western Indian Ocean. In addition, growth did not differ between South Africa and other regions (Draganic and Pelzcarski 1984, Fonteneau 1980, Gascuel et al. 1992, LeGuen and Sakagawa 1973, Lehodey and Leroy 1999, Lessa and Duarte-Neto 2004, Shuford et al. 2007, Stequert et al. 1996, Wild 1986, Yang et al. 1969). Further substantiation of the above-mentioned observations was found by recording differences in the sizes of fish caught in the inshore (baitboat and sport fishery vessels) and offshore (large pelagic longline vessels) sectors of South African tuna fisheries. A significant difference between the regions — in terms of the size of fish caught inshore — was noted, with mostly-juvenile fish being caught in KwaZulu-Natal (5.4 ± 3.5kg), sub-adult fish in the Eastern Cape (26.2 ± 13.4kg), and adult fish in the Western Cape (42.3 ± 14.4kg). Since mostly-adult fish were caught offshore by longliners, with no significant differences between regions, it is however possible th at adult fish predominantly inhabit the offshore region. Yellowfin tuna caught by the large pelagic longline fishery in the three managerial zones (A, B and C) were predominantly adult fish of similar size, namely Zone A: 38.9 ± 6.9kg; Zone B: 28.7 ± 4.6kg, and Zone C: 36.0 ± 5.1kg. The recreational ski boat sport fishery has remained stable, in terms of participation, consisting mostly of white middle aged males in the top 25% of household income distribution, having either permanent occupational status or being retired. Fishers within this sector are willing to incur great expense to partake in the fishery and they provide an important economic contribution to coastal towns, particularly in the Eastern Cape. The total catch (of 83t) of yellowfin tuna by the competitive sport fishery within the Western and Eastern Cape regions was considerably lower than that of commercial tuna baitboat catches, which amounted to 186t, and the large pelagic longline sector that caught t in 200. It is however likely that the competitive sport fishery's total yellowfin tuna catch (of 83t in 2009) of the Eastern and Western Cape competitive sport fishery was considerably less than the total yellowfin tuna catches of the whole South African deep-sea sport fishery. Longfin tuna are the primary target species of South Africa‘s baitboat fisheries, comprising an average of 86% of the total catch and generating ZAR49 million in employment income in 2002. South Africa was responsible for 20% of the total longfin tuna annual yield in 2004 in the southern Atlantic Ocean, behind Taiwan with 59%. However, yellowfin tuna only contributes a small percentage towards total catches (8.4 ± 8.2% between 1995 and 2009), generating ZAR1.3 million in employment income in 2002. Of the four vessel categories comprising the tuna baitboat fishery, ski boats had the highest yellowfin tuna CPUE in 2009 (117 ± 62 kg.vessel⁻¹.day⁻¹) and the lowest effort. The ski boats sector is the most opportunistic fishery as they are only active when either longfin or yellowfin tuna are in high abundance. At such times catches are guaranteed, so can be expected to offset expenses. In 2009 the CPUE for yellowfin tuna for 15–19m vessels and freezer deckboats was 12 ± 20kg.vessel⁻¹.day⁻¹ and 3 ± 6kg.vessel⁻¹.day⁻¹, respectively. These vessels specifically target longfin tuna when they are in abundance. Since the start of the tuna baitboat fishery in 1995, there has been a substantial increase in the number of new entrants. In 2002 this sector had a fleet size of 82 vessels with a capital value of ZAR163 million and a total employment income of ZAR58 million, employing 2 173 fishers, of which 87% were black African. The commercial tuna baitboat fleet has subsequently grown to 200 vessels and 3600 crew, with 110 active vessels fishing for a combined fleet average of 46 days per year. There are a high number of owner-operated vessels. Since 2007 the fishery's profit to cost ratio has been low due to the low abundance of tuna stocks off the coast of South Africa, which has resulted in poor catch returns, placing economic pressure on the fishery. The baitboat industry is a low-profit-margin fishery with a total net catch value worth ZAR90 million in 2009 (Feike 2010). The abundance of yellowfin tuna influences profit margins, with very high profits being made when abundance and catches are high. The large pelagic longline fishery has a total allowable effort of 43 vessels of which only 30 vessels fished during 2009, when a reported 766t of yellowfin tuna were caught, representing a total tonnage far in excess of that obtained by the combined effort of the baitboat and sport fishery. It is, however, assumed that considerable underreporting of catches takes place within this fishery, which means that the estimated total net catch value of ZAR100 million could, in fact, be much higher. Such underreporting of catches is of great concern for this fishery, as is the high bycatch of Chondricthians spp. that significantly outweighs imposed regulatory limits (DEAT 2007). The present study demonstrates the current lack of comprehensive catch and effort data for the sport, baitboat and longline fisheries as well as the serious limitations and flaws associated with current databases. Results from the present study have drawn attention to a number of high-priority research needs, as outlined below. (1) A major lack of comprehensive catch and effort data for the sport fishery, which can be rectified by focussing on obtaining more competition data, as well as high-quality catch and effort and socio-economic information, as opposed to relying on information from non-club anglers (Gartside et al. 1999, Williams 2003, Cass-Calay 2008). Acquisition of such data is relatively inexpensive: the location of organized clubs and their frequent competition meetings provide widespread coverage along the Southern African coastline. Such data acquisition efforts have the potential to provide reliable information on spatial catch trends. (2) Validation of vessel catch return data is required for commercial fisheries and on some recent data that has emerged from studies of catch rates and trends for target species, particularly in the longline fishery. In this context it should be noted that the most recent peer-reviewed publications on this exploratory fishery were published more than a decade ago (Kroese 1999, Penny and Griffiths 1999). Additional studies need to be undertaken and journal articles published on the current stock status of South African catches of yellowfin and bigeye tuna and swordfish.

Abstract:
Photosynthetic eukaryots emerged as a result of several billion years of evolution between proeukaryotic cell and ancestral cyanobacteria that formed modern chloroplasts. The symbiotic relationship led to significant rearrangements in the genomes of the plastid and the nucleus: as many as 90 % of all the plastid genes were transferred to the nucleus. The gene transfer has been accompanied by the development of sophisticated regulatory signaling networks originating in the organelle (retrograde) and in the nucleus (anterograde) that coordinate development of the plastid and ensure adequate cell responses to stress signals. In this thesis I have demonstrated that transcriptional activity of PEP in the chloroplast is essential for proper embryo and seedling development in Arabidopsis thaliana. The function of PEP is dependent on the nuclear encoded PEPassociated factor PRIN2 that is able to sense the redox status of the plastid during seedling development and different stress. In response to the plastid status PRIN2 modulates the transcription activity of the PEP enzyme complex. We further established that PRIN2, as an essential component for full PEP activity, is also required to emit the Plastid Gene Expression (PGE) retrograde signal to regulate the Photosynthesis-Associated Nuclear Genes (PhANG) in the nucleus during early seedling growth via GUN1. On the other hand, regulation of PhANG expression during the High Light (HL) conditions requires functional PRIN2 and PEP activity but is GUN1-independent. Another retrograde signal produced by the developing chloroplast is associated with the tetrapyrrole biosynthesis pathway. We have established that accumulation of the chlorophyll intermediate MgProtoIX-ME in the crd mutant triggers repression of the PhANG expression, and this negative signal is mediated by a cytoplasmic protein complex containing the PAPP5 phosphatase. The nuclear targets that receive the tetrapyrrole mediated signal are GLK1 and GLK2 transcription factors that control the PhANG expression and the expression of the enzymes involved in the biosynthesis of chlorophyll.
Fotosyntetiserande eukaryoter uppstod från en endosymbiotisk interaktion under några miljarder år mellan en ur-eukaryot och kloroplastens förfader, den prokaryota cyanobakterien. Den symbiotiska händelsen ledde till att kloroplastens och kärnans genom blev väsentligt förändrade. Så småningom överförde kloroplasten så många som 90 % av dess gener till cellkärnan. För att koordinera genutrycket från de två genomen utvecklade växtcellen ett sofistikerat signalsystemen som inkluderar: plastid-kärn (retrograd) och kärn-plastid (anterograd) signalering som styr kloroplastens utveckling och förmåga att anpassa sig till stressförhållanden. Den här avhandlingen beskriver kloroplastens maskineri för genuttryck (PEP) som en nödvändig komponent för embryo- och växtutvecklingen hos Arabidopsis thaliana. PEP funktionen är beroende av det kärnkodade kloroplastproteinet PRIN2 som är associerat med PEP. PRIN2 mottar redox signaler från plastiden och förändrar genuttrycksaktivitet under kloroplastens utvecklingen eller under olika stressförhållanden. Jag visar dessutom att PRIN2 spelar en viktig roll i överföring av kloroplastens signal som kommunicerar genuttrycksaktivitet (PGE) via GUN1 till kärnan där den styr uttryck av de kärnkodade fotosyntetesgenerna (PhANG). Under högljus stressförhållanden styrs dock PhANG-uttrycket av signaler som uppstår från PEP-aktivitet och PRIN2 men som är oberoende av GUN1. Vidare finns det en annan retrograd signal som har sitt ursprung i biosyntesen av tetrapyrroler. Jag har visat att ackumuleringen av tetrapyrrolen MgProtoIX-ME i crd-mutanten framkallar nedreglering av PhANG-uttryck genom interaktion med ett fosfatas (PAPP5) i cytosolen. GLK1 and GLK2 är två transkriptionsfaktorer som tar emot den tetrapyrrole-medierade signalen i sin tur styr biosyntes av chlorofyll och PhANG uttryck.

This thesis focuses on the algorithm configuration problem. In particular, three efficient learning configurators are introduced to tune parameters offline. The first looks into metaoptimization, where the algorithm is expected to solve similar problem instances within varying computational budgets. Standard meta-optimization techniques have to be repeated whenever the available computational budget changes, as the parameters that work well for small budgets, may not be suitable for larger ones. The proposed Flexible Budget method can, in a single run, identify the best parameter setting for all possible computational budgets less than a specified maximum, without compromising solution quality. Hence, a lot of time is saved. This will be shown experimentally. The second regards Racing algorithms which often do not fully utilize the available computational budget to find the best parameter setting, as they may terminate whenever a single parameter remains in the race. The proposed Racing with reset can overcome this issue, and at the same time adapt Racing’s hyper-parameter α online. Experiments will show that such adaptation enables the algorithm to achieve significantly lower failure rates, compared to any fixed α set by the user. The third extends on Racing with reset by allowing it to utilize all the information gathered previously when it adapts α, it also permits Racing algorithms in general to intelligently allocate the budget in each iteration, as opposed to equally allocating it. All developed Racing algorithms are compared to two budget allocators from the Simulation Optimization literature, OCBA and CBA, and to equal allocation to demonstrate under which conditions each performs best in terms of minimizing the probability of incorrect selection.

Hylsan som undersökts är av material AISI 440B och hade till syfte att verka som en förstärkt slityta i en 3D-applikator för exempelvis tätningsmedel och lim. Hylsan ska förhindra slitage i svivelmekansimen i applikatorn, och härdas för att erhålla en hård yta som kan motstå de abrasiva ämnen som transporteras genom applikatorn. Hylsan har undersökts i syfte att hitta kritiska punkter som kan optimeras för en mer konkurrenskraftig produkt. Därav gjordes en oberoende analys för hylsans hållfasthet utan hänsyn till tidigare arbeten. Information kring hur produkten brukas var knapphändig avgränsades arbetet mot förslitningar och transporter. Därför studerades endast tillverkning, montering och konstruktion. Haveriorsaker visade sig kunna vara mycket individuellt då det var svårt att bestämma när och hur ofta ett haveri kunde uppstå. Det var okänt hur lång tid produkten varit i bruk samt vilka material som flödat genom 3D-applikatorn i olika industrier. Det medförde att hylsans hållfasthet undersöktes med metoder som ”walk to gemba”, ”Finite Element Method” och visuell inspektion. FE-analysen granskade monteringsprocessens påverkan på hållfastheten samt ger en översiktlig bild av spänningsfördelningarna i hylsan. Den visuella inspektionen tillämpades för att erhålla en förståelse för sprickans uppkomst i hylsan samt att kartlägga sprickförloppet. Resultatet av undersökning och analys gav att följande kritiska parametrar påvisats: Litteraturstudien visade att härdning och bearbetning av hylsan på olika sätt kan vara kritiska för hållfastheten Hylsans rundhet riskerar att variera otillåtet vid montering Limmet riskerar att inte fylla någon funktion Sprickor kunde härledas till att starta i hålen för den undersökta hylsan. Orsak till sprickuppkomst kunde ej fastställas. FE- analysen av monteringsförloppet visade inte på några försvagningar i konstruktionen, eftersom inga spänningar beräknades överstiga brottgränsen Den avslutande diskussionen resulterade i ett antal förslag för fortsatta arbeten. Några förslag var att bland annat undersöka rundhetens påverkan i klämförband, utbyte av material och tribologi.
The examined sleeve is made of material AISI 440B and has the purpose of acting as a reinforced wear surface in a 3D applicator for example sealants and glue. The sleeve should prevent wear in the swivel mechanism of the applicator, and is therefore hardened to obtain a hard surface which can resist the abrasive substances flowing through the applicator. The sleeve has been investigated in order to find critical points that can be optimized for further competitive product. An independent analysis of the construction of the sleeve was therefore made without regard to previous work. Since the information regarding the use was scarce, the work was limited from studies of wear and transportation. Therefore, only manufacturing, assembly and construction were studied. The causes of failure were proved to be very individual as it was difficult to determine when and how often a failure could occur. It was unknown how long the product has been used as well as what kind of materials has flown through the 3D applicator in different industries. This meant that the construction of the sleeve was examined through methods such as "walk to gemba", "Finite Element Method" and visual inspection. FE analyse examined the impact of the assembly process on the strength and provides an overview of the stress distributions in the sleeve. The visual inspection was applied to obtain an understanding of the origin of the crack in the sleeve as well as mapping the crack propagation. The result of investigation and analysis showed that the following critical parameters were detected: The literature study showed that hardening and machining of the sleeve is critical in different ways for the strength The roundness of the sleeve risks varying impermissibly during assembly The adhesive risks to not performing any function The cracks could be derived to start in the holes of the examined sleeve. Causes of cracking cannot be determined. The FE analysis of the assembly process did not indicate any weakening in the construction and no stresses were calculated to exceed the breaking limit. The concluding discussion resulted in a number of proposals for further work. Some suggestions (among other things) were to investigate the effect of roundness in interference-fit, exchange of materials and tribology.

Abstract:

The domestication of tuna species has proven to be one of the most challenging endeavours in aquaculture. Among the issues yet unresolved is the occurrence of ‘puffy snout syndrome,’ a condition in which tumour-like growths form on the head. Ultimately, vision and feeding are impaired and long-term afflictions typically result in mortality. Though few mentions of puffy snout exist in the literature, evidence suggests that it is not uncommon among facilities that hold tunas in captivity. The specific aims of this study were to: a) describe pathological features of puffy snout, and b) investigate its etiology through the evaluation of conditions and protocols at facilities rearing/holding tuna.

To describe pathological features of puffy snout, clinical signs were detailed by observing captive tunas in a land-based holding system, and examination of evidence of infection by parasites, bacteria, and viral agents was conducted on tissues collected from fish with and without puffy snout. Histological examination of tissue from normal and affected fish was also conducted. To investigate etiology, a survey was developed and sent electronically to 28 tuna-holding facilities globally. The survey inquired about the prevalence of puffy snout and the husbandry conditions and protocols employed (e.g., biological characteristics, capture and transport procedures, holding system design and water quality, feeding regime). These data were compared across facilities in an attempt to couple puffy snout prevalence with holding conditions and/or protocols.

Clinical signs of puffy snout included occlusion of the eyes and mouth, followed by changes to swimming and feeding behaviours. Parasitology, bacteriology, and virology examination all indicated no commonly-found pathological agents were responsible for inducing the condition. Histology showed that puffy snout is largely characterised by the apparent degeneration of muscle tissue with the replacement of a loose collagenous fibrosis and an undetermined fluid filling the interstitial space in tissues anterior to the eye. In dorsal musculature, collagenous growth may occur in the epidermal or hypodermal regions.

Based on survey data, the capture and transport process and feeding regime were unrelated to development of puffy snout. However, certain biological (e.g., fish size) and holding system (e.g., tank/pen size) parameters showed weak but non-dismissible coupling with puffy snout prevalence. Survey data and additional personal communication with field researchers confirmed that puffy snout in tuna is solely a captivity-related condition.

This paper is about an Iron Age cemetery at Tuna in Badelunda parish, Västmanland. Tuna is a complex burial ground that contains a total of 66 graves that consists of 68 individuals. The grave field was used during a period of roughly 700 years from around the year 300 AD until the year 1050 AD. Three types of grave constructions are identified in Tuna, 53 cremation graves, eight boat graves and five chamber graves. This paper will investigate the placements of the grave constructions on the cemetery to study if they are divided by sex, age and grave type. The paper will also analyse and discuss the sex/gender assessments of individuals from Tuna, both the osteological assessments of sex and/or gender assessments are based on? The usual presumptions from earlier research about Tuna are that it was a cemetery mainly for females, but the report about Tuna in Badelunda indicates a diffrent conclusion.

This thesis evaluates the utility of a sport fishery for yellowfin tuna, Thunnus albacares, in the Pemba Channel, Kenya, in providing ecological information relevant to commercial fishery assessment and management. Age, growth, reproductive status and diet are measured, together with an innovative assessment of the Indian Ocean yellowfin tuna stock. Catch/effort and weight data from the records of the sport fishery from 1963-1995 demonstrate a decline in tuna mean weight since the advent of purse seining in 1984. A periodicity of 5-6 years was detected in the Indian ocean longline fishery catches, and in the sport fishery catch/effort and mean weights. Samples were taken from tuna caught in the sport fishery from 1981-88. Sport-fished samples were more random in size, age and sex (ratio = 1: 1) than catches from any of the three major commercial fisheries. The total mortality rate of cohorts deconvoluted from weight-frequencies in the sport fishery catch over 6 years was used in a Caddy & Csirke analysis. Maximum Sustainable Yield estimates of the Indian Ocean stock were 113,000 tonnes (new method), 110-160,000 tonnes (Walters' method); compared with only 40-52,000 tonnes using the standard equilibrium method. Tuna (1653 over 7 years) were successfully aged using length frequency analysis and by a novel method based on clusters from a Principal Components Analysis of morphometric measures. Juvenile growth was fast, 3.1 cm/month, whereas adults grew at about 2 cm/month. Gonads (>1500) were staged for 5 years and histologically examined for 2 years. Female yellowfin mature earlier than males, but males become ripe earlier in the season. The spawning stock consists of females >120 cm FL. Diet was scored over 4 years and evaluated volumetrically over 2 years. Juvenile tuna eat fish, but adults opportunistically consume fish, cephalopods and crabs, depending on availability. Catchability by gear in both the sport and commercial fisheries is likely influenced by the currently abundant food.

Languages differ in the means they avail themselves of for the structural realization of information structure, where available options are word order, prosody and morphology. Turkish has long been characterized as predominantly using word order and its variation in realizing information structure, where certain positions in a sentence are associated with certain pragmatic functions related to information structure. Prosody has been proposed to play only a secondary role interacting with word order. Contrary to this widely established view, the thesis argues that the notion that sentential positions have pragmatic functions and word order variation is a syntactic means to realize these functions can be abandoned, without any loss of explanatory power, in favor of a tune-based perspective where prosody is the sole structural determinant of information structure. In this setting word order variations are argued to be prosodically motivated, in that Turkish phonology imposes some precedence constraints on intonational contours. Word order variation then turns out to be just a consequence as opposed to being a determinant in attaining the right information structure required by the discourse context. To substantiate these claims a tune-based account, based on Steedman'
s account of English information structure, is proposed for the structural realization of information structure in Turkish, whereby information structural units are directly associated with prosodic phrases intonationally marked in certain ways. Validity of the account is tried to be established by intonational analysis of recorded speech data. As for the explanatory value, the information structure phenomena that has received positional explanation in the relevant literature, are tried to be captured only in prosodic terms, without committing to positions, syntactic strategies and such.

Det vill säga, beskriva och analysera hur företag väljs ut, samt vilka företagsfonder som faktiskt får klassas som etiska. Vidare ämnar studien diskutera problematiken kring det valda området.   Metod: I detta arbete har vi valt att genomföra en kvalitativ för att generera en djupgående information om hur fondförvaltare arbetar med etiska fonder. Vi har valt att genomföra intervjuer med två representanter från fondbolag som erbjuder sina kunder etiska fonder för att kunna besvara uppsatsens frågor. Den information vi samlat in ligger till grund för vår analys och slutsats. Följaktligen tjänar de insamlade redan befintliga teorier ett jämförande syfte mot vår empiri.   Resultat & slutsats: Efter genomförandet av studien  kom vi fram till att fondbolagen har olika kriterier gällande vad som kan anses eller utmärka en etisk fond vilket leder till att även urvalsprocessen och uppföljningsprocessen ser annorlunda ut från fondbolag till fondbolag. Dessa skillnader kan skapa förvirring hos konsumenter om vad ett etiskfond är.     Förslag till fortsatt forskning: Denna studie gav oss insyn i hur fondbolag arbetar med granskningen av de etiska fonderna och problematiken  kring ämnet, vi ville ta reda på hur granskningen utförts på den Svenska marknaden.
