Abstract:
W artykule została scharakteryzowana metoda wykonania tunelu z wykorzystaniem technologii TBM. Na przykładzie linii metra warszawskiego zaprezentowany został schemat wykonywania prac ta metoda oraz monitoring bezpieczenstwa przylegajacej infrastruktury. Metoda ta jest równie‚ z powodzeniem wykorzystywana do dra‚enia tuneli drogowych, wodociagowych, kanalizacyjnych, hydroenergetycznych czy wielozadaniowych. Za pomoca technologii TBM utworzono ju‚ 369 projektów tuneli metra o długosci ponad 880 km, 440 km tuneli kolejowych i 171 km drogowych. Łacznie ta technologia wydra‚ono ponad 1900 km tuneli na całym swiecie [7]. Przewa‚nie jest ona bardziej ekonomiczna i efektywna w stosunku do metod tradycyjnych (odkrywkowych). Nie powoduje nadmiernych utrudnien w komunikacji transportu ulicznego, a w ciagu doby może powstac nawet trzykrotnie dłuższy tunel, aniżeli ten wykonany w tradycyjny sposób. Ponadto, co istotne z punktu widzenia ekonomicznego, całkowity koszt wydrażenia metra technologia TBM jest niższy [7].

Povod tomu bila je zamjena kabelskoga voda za dizalo koje će povezivati mrežu podzemnih tunela i utvrdu Kaštel. Istraživanjem je utvrđena fazna izgrađenost kule: kasnoantička kula poligonalnoga oblika, čija je struktura vidljiva u današnjoj hodnoj razini, nalegla je na antičku kulu kružnoga oblika, koja je temeljena na četvrtastoj kuli iz najranije faze formiranja pulskoga gradskog obrambenog sustava. Na samom ulazu u tunel otkriveni su ostaci kasnoantičkoga bedema. Istraživanjem je utvrđen način spoja kasnoantičkoga bedema na poligonalnu kulu: preslojavanjem četvrtaste kule i sloja popločenja te oslanjanjem na okruglu kulu koja je pridodana potezu najranijega gradskog bedema.

Abstract:
Background: Many studies done in past literature focuses on gonadal toxicity of Cyclophosphamide, but very less studies are done on the epididymis post exposure to this drug. Hence a sincere attempt is made in present study to look into the cellular changes in the epididymis through histology and TUNEL Assay. Subjects & Methods: Healthy male Swiss albino adult mice, were obtained from Central Animal Research Facility, Manipal were used for the study. The adult as well as prepubertal male mice were divided into control and test groups. In test group, the mice were injected with Cyclophosphamide at variable doses. From each group certain no. of mice was sacrificed on different days. A control group was kept for each intervals. The epididymal tissue was then extracted and used in histological and Tunnel Assay study. Results: Even though there was no significant change in the number of basal cells, the apical cells decreased marginally at higher doses of CP. A TUNEL assay was performed in the epididymal sections to understand its effect on the DNA integrity. Up to 100 mg/kg, CP did not induce any significant increase in the TUNEL positive tubules. However, at 200 mg/kg dose and above, there was an increase in the number of tubules with TUNEL positive cells indicating its DNA damaging effect. However, this increase was statistically not significant Conclusion: The present study highlights the need for looking into the cellular damage of the cells after exposure to one of the commonly used anticancer drug- cyclophosphamide. Abstract:
TUNEL (terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling) is a method of choice for rapid identification and quantification of the apoptotic cell fraction in cultured cell preparations. However, TUNEL application has been restricted to a narrow spectrum of sample conditions, and only detergents have been proposed as labeling enhancers. This study was aimed at extending TUNEL to variously fixed cells and improving TUNEL sensitivity by optimized pretreatments, the specificity being assessed by reference to the apoptotic morphology. Comparative TUNEL was performed with three protocols on CEM-C7 cells, a model of glucocorticoid-induced apoptosis. Samples were submitted to six modalities of fixation and TUNEL was performed after each of the following conditions: no pretreatment; detergent permeabilization; proteolytic digestion; microwave irradiation; and a recently published combination of the latter two. The proportion of TUNEL-stained elements within the cell fraction, with and without apoptotic morphology, was quantified. Our results showed that: (a) with an adequate pretreatment, reliable TUNEL can be obtained after each fixative tested; (b) detergent was inefficient in improving sensitivity; (c) whatever the fixation, microwave pretreatment provided the best TUNEL sensitivity without notable loss of specificity; (d) under adaptive technical conditions, TUNEL can be associated with detection of various proteins by double labeling; and (e) the existence of a limited population of intensely TUNEL-positive cells that lacked apoptotic morphology contributes to the current debate about a preapoptotic state.

Terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) assay is a long-established assay used to detect cell death-associated DNA fragmentation (3’-OH DNA termini) by endonucleases. Because these enzymes are particularly active in the kidney, TUNEL is widely used to identify and quantify DNA fragmentation and cell death in cultured kidney cells and animal and human kidneys resulting from toxic or hypoxic injury. The early characterization of TUNEL as an apoptotic assay has led to numerous misinterpretations of the mechanisms of kidney cell injury. Nevertheless, TUNEL is becoming increasingly popular for kidney injury assessment because it can be used universally in cultured and tissue cells and for all mechanisms of cell death. Furthermore, it is sensitive, accurate, quantitative, easily linked to particular cells or tissue compartments, and can be combined with immunohistochemistry to allow reliable identification of cell types or likely mechanisms of cell death. Traditionally, TUNEL analysis has been limited to the presence or absence of a TUNEL signal. However, additional information on the mechanism of cell death can be obtained from the analysis of TUNEL patterns.

This article is an analysis of Tunel (The Tunnel), the novel by Magdalena Parys, a Polish author living in Germany. The action of the book focuses on the fictional construction of an escape tunnel built under the border wall between the western and eastern part of Berlin in the early 1980s. This, somehow, predestines Parys’s work to be read in the context of transgression, and crossing limitations. Starting with the writer’s non-literary inspirations, the article shows the multiplicity of literal and metaphorical meanings of the term “border” and examines how the motif functions in the presented world.

One hallmark of apoptosis is the degradation of chromosomal DNA. We cloned the Caenorhabditis elegans gene nuc-1, which is involved in the degradation of the DNA of apoptotic cells, and found that nuc-1 encodes a homolog of mammalian DNase II. We used the TUNEL technique to assay DNA degradation in nuc-1 and other mutants defective in programmed cell death and discovered that TUNEL labels apoptotic cells only during a transient intermediate stage. Mutations in nuc-1 allowed the generation of TUNEL-reactive DNA but blocked the conversion of TUNEL-reactive DNA to a subsequent TUNEL-unreactive state. Completion of DNA degradation did not occur in the absence of cell-corpse engulfment. Our data suggest that the process of degradation of the DNA of a cell corpse occurs in at least three distinct steps and requires activities provided by both the dying and the engulfing cell.

Abstract: Dehydration is a necessary procedure prior to exposing seeds to long term storage, but this is associated with metabolism-linked injury mediated by cell injury. In order to assess cellular alterations during re-establishment of desiccation tolerance (DT) in C. fissilis germinated seeds and their relation to DNA damage, we verified the occurrence of DNA fragmentation through the TUNEL test and its evidence through the cytological analyses. To re-establish DT, germinated seeds were incubated for 72 h in polyethylene glycol (PEG, -2.04 MPa) before dehydration in silica gel (at 10% moisture content) followed by rehydration. The moisture content changes during the reestablishment of the desiccation tolerance was accomplished. (DT)TdT-dUPT terminal nick-end labeling (TUNEL) was used to assess rates of cell death. TUNEL staining was performed using Click-iT-TUNEL Alexa Flour imaging assay. The TUNEL test showed a consistent DNA fragmentation in the 2 and 5 mm long radicles. Moreover, nuclear and chromosomal alterations were observed in the 5 mm meristematic root cell cycle, contributing to the identification of diagnostic markers of cell death.

Resumo: Túneis de congelamento são equipamentos utilizados pela industria alimentícia para o condicionamento térmico de produto tais como iogurte, sorvete, leite, carne e derivados. Assim, geralmente estão no fim do processo de produção de produtos altamente perecíveis. Quando um túnel representa m gargalo do processo, ele causa transtornos à produção que podem até interrompê-la. Este trabalho visa a abordagem do problema de operação eficiente e túneis de congelamento quanto ao aspecto de carregamento. Primeiramente abordamos o problema estático e suas relaxações e restrições. No problema estático determinístico, o instante de chegada das bandejas de produtos e o tempo de exposição necessário para seu condicionamento são conhecidos de antemão. Deve-se, então, programar por completo o carregamento do refrigerador definindo, para cada bandeja, em que nível do túnel deve ser colocada ou se deve ser rejeitada. Resultados teóricos são obtidos, particularmente para o caso (relaxado) do refrigerador de gavetas. Várias possibilidades de abordagem do problema estático são discutidas. Em seguida abordamos o problema dinâmico, onde não se dispõe do conhecimento prévio dos parâmetros de cada bandeja. Estes só ficam definidos a partir da chegada de cada bandeja, quando então a decisão quanto ao carregamento ou rejeição da bandeja deve ser tomada. Para o problema dinâmico são apresentadas várias políticas, que são testadas num caso real e em cenários hipotéticos nele baseados. Para avaliação de desempenho das políticas usamos limites' inferiores calculados com base no desenvolvimento teórico do problema estático. Os resultados são muito satisfatórios para uma classe de políticas a que chamamos de encadeamento.
Abstract: Freezing tunnels are food industry facilities for conditioning of meat, ice-cream, milk, yogurt, etc. They are very unwished botlenecks because they are positioned at the end of the production process of highly perishable items (low in-process inventory). The freezing tunnel loading problem is that of maximizing its production in terms of the amount of frozen trays of products. The purpose of this study is to model this problem and develop methods for solving its static and dynamic versions. In the static problem, the arrival time and the conditioning time of each tray are known in advance for the entire loading period. One should then completely schedule the loading of the tunnel. Each tray must be assigned to a position in the tunnel or be rejected. . We discuss several approaches and show some theoretical results for relaxed problems. In the dynamic problem, the arrival and the demand of trays are not known in advance. In this case, the decisions must take place at the arrival times. Several policies are developed. Some real case based scenarios are used for testing the policies. The study shows the potential gains with the application of some policies, mainly those ofthe class we named 'chain policies'.

One of the major problems of modern Internet is the upcoming depletion of addressing space in IPv4 protocol. This master thesis focuses on the study of network protocol IPv6, the possibilities of expanding this protocol in the Internet using the appropriate tunelling method and ways of tunnel management, especially the TIC protocol. Major goal of this work is using knowledge of protocols in a design and implementation of the server part of tunnel broker system, which uses mentioned tunneling method and is compatible with the open source client AICCU. Second part of this work deals with testing and pilot deployment of this system. Main benefit consists in creation an open source tunel broker system. Real deployment of this system is particularly advantage for Czech users as a low latency tunnel IPv6 Internet.

Resumo: o objetivo do presente estudo foi desenvolver um modelo matemático para um secador industrial de túnel para fi'utas, simulá-Ia e otimizar sua operação considerando a qualidade do produto como restrição principal na formulação do problema de otimização. Os secadores operam em forma semi-contínua e têm carrinhos e bandejas. O período de cada ciclo é determinado fazendo cumprir as especificações da umidade inicial e final do produto, de forma que cada carrinho com um número determinado de bandejas com fruta úmida seja alimentado em ciclos ao túnel enquanto outro que atingiu a umidade final é retirado do outro extremo do túnel. As configurações consideradas no estudo foram concorrente e contracorrente. O modelo que foi desenvolvido a partir dos balanços simultâneos de calor e massa para a fase sólida e gasosa, tanto para as bandejas (carrinhos) quanto para o túnel, é simples e apropriado para efeitos de otimização onde o tempo computacional é muito importante. O modelo foi aplicado para simular a secagem de uva Itália usando dados experimentais da cinética de secagem, ajustando-se os parâmetros de acordo aos requerimentos das equações do modelo. Os códigos de simulação e otimização foram desenvolvidosem FORTRAN 90 de Microsoft Fortran PowerStation. A não linearidade das equações do modelo e da restrição de qualidade da fruta, representada pela degradação de ácido ascórbico, conduziram ao desenvolvimento de um algoritmo para a solução do problema de otimização cuja função objetivo foi minimizar o consumo de energia por unidade de sólido seco, tempo do ciclo e temperatura máxima atingida pela fruta. Os resultados mostram as condições de operação ótimas para um número fixo de carrinhos no túnel e indicam que a configuração contracorrente é mais eficiente quando se avalia os requerimentos de calor por kg de sólido seco para uma degradação de ácido ascórbico aproximadamente constante. Igualmente, que se reduz o consumo de energia em ambas configurações quando comparado aos valores simulados sem otimizar.
Abstract: The non linearity of the equations of the model and the quality of the fruit, represented by the ascorbic acid degradation, leaded to develop an algorithm for the solution of the optimization problem which objective function was to minimize the energy consumption, cycle time and maximum temperature achieved by the fruit. Results show the optimal operation conditions for a fixed number of trucks in the tunnel and indicate that countercurrent configuration is more efficient when heat requirements per kg of dry solids is evaluated for approximately constant ascorbic acid degradation. Furthermore, energy consumption for both configurations is reduced when compared to non optimized simulation results.

Abstract:
Cryopreservation of oocytes is a challenge. Studies were conducted to vitrify mouse zygotes and cumulus-intact bovine oocytes from follicles of different diameters, small (⠤ 4 mm) and medium (4 to 10 mm), using nylon mesh. The specific goals were to assess changes in apoptotic gene expression (Fas-FasL, Bax, Bcl-2, and survivin) in conjunction with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) and caspase assays. Mouse zygotes were exposed to increasing concentrations of ethylene glycol (EG), Ficoll-70 and sucrose in phosphate buffered saline (PBS) for vitrification on nylon mesh and plunged into liquid nitrogen. Warming resulted in 81.7% morphological survival. The rate of blastocyst formation was 59.9% for vitrified zygotes but, this was significantly lower than that of non vitrified embryos (66.2%). There was no difference in the hatching rates between groups. Both Fas and FasL mRNA were detected at the 4-cell and morula stages, suggesting Fas signaling was operational in early embryos. The level of expression of Bax mRNA tended to increase, while expression of survivin mRNA was not different for 2- and 4-cell embryos. Fragmented embryos showed an increase in Bax mRNA levels, while survivin mRNA level was reduced. In the second experiment, vitrification of bovine oocytes was carried out. Pre-cooled cryovials resulted in 98.9% morphological survival. The oocytes from small and medium size follicles had a significant impact on cleavage (53.7 ± 1.6% vs. 43.8 ± 1.6%, respectively) and blastocyst rates were 16.9 ± 1.0% and 11 ± 1.2%, respectively. Follicle size for oocytes had no impact on the expression of apoptotic genes. The Fas-FasL and Bax-Bcl-2 mRNA showed increased expression after vitrification, but tended to decrease after 9 h of maturation. Yet, results from TUNEL and caspase assays did not support the evidence of the downstream apoptotic signaling pathway in embryos. The semen utilized for in vitro fertilization in both vitrified and control oocytes responded differently in the 4 tested bulls than their field fertility record. The altered transcriptional activities of apoptotic genes, Fas-FasL and Bax-Bcl-2, and survivin were indicative of possible apoptotic activity in vitrified embryos and oocytes subjected to in vitro fertilization.

Genetics research often relies on experiments that require repetitive, time-consuming handling of small volumes of liquid (1 mL) and biomass (10–20 μL) such as fluorescence in-situ hybridization (FISH), β-galactosidase staining, immunohisto chemistry, skeletal and tunel assays. Often manual, these experiments are time intensive and error-prone. We report on the design, fabrication, and testing of a low-cost, two-axis, precision robot for FISH assays on whole mice embryos. The robot can complete 20 successive embryo immersions in unique isothermal solutions in minutes for 6 samples. Repeatability of the orthogonal axes is 66 and 214 μm, near the measurement uncertainty limit and sufficient for operation. Accuracy is achieved by systematic error compensation. Low-cost and precision are obtained using design and manufacturing techniques and processes, resulting in a cost of 15% of comparable instruments (e.g., InsituStain, Intavis Bioanalytical Instruments). This design demonstrates a simple, automated platform to perform a typically manual experimental genetics technique.

This paper presents and describes the results of an experimental study into the control of separated flow over a NACA0015 airfoil over a range of angles of attack at a Reynolds number of 3 × 104. The study was carried out in the LTRAC 500 mm × 500 mm water tunel. The separation control is actuated using a planar zero-net-mass-flow (ZNMF) jet at the leading edge of the airfoil. The ZNMF micro-jet flow is generated using a piston-cylinder arrangement connected to a skotch-yoke mechanisms that is driven by a computer controlled stepper motor. The parameter range for the planar ZNMF jet ranged from non-dimensional frequency of 0.5 to 1.5 and blowing coefficients up to 0.2. A six component force transducer was used to measure the lift and drag force on the airfoil. Planar laser induced fluoresence (PLIF) using Kyton red dye and Nd:YAG laser illumination was used for the flow visualisations. Multigrid cross-correlation digital PIV (MCCDPIV) with pulsed Nd:YAG laser planar illumination was employed with digital imaging of 4008 px × 2672 px to measure instantenous velocity fields.

Abstract:
Geophysical detection of clandestine tunnels is a complex problem that has been met with limited success. Multiple methods have been applied spanning several decades, but a reliable solution has yet to be found. This report presents shallow seismic data collected at a tunnel test site representative of geologic settings found along the southwestern U.S. border. Results demonstrate the capability of using compressional wave diffraction and surface-wave backscatter techniques to detect a purpose-built subterranean tunnel. Near-surface seismic data were also collected at multiple sites in Afghanistan to detect and locate subsurface anomalies (e.g., data collected over an escape tunnel discovered in 2011 at the Sarposa Prison in Kandahar, Afghanistan, which allowed more than 480 prisoners to escape, and data from another shallow tunnel recently discovered at an undisclosed location). The final example from Afghanistan is the first time surface-based seismic methods have detected a tunnel whose presence and location were not previously known. Seismic results directly led to the discovery of the tunnel. Interpreted tunnel locations for all examples were less than 2 m of the actual location. Seismic surface wave backscatter and body-wave diffraction methods show promise for efficient data acquisition and processing for locating purposefully hidden tunnels within unconsolidated sediments.
