Abstract:
The “tubulin code” combines different α/β-tubulin isotypes with several post-translational modifications (PTMs) to generate microtubule diversity in cells. During cell division, specific microtubule populations in the mitotic spindle are differentially modified, but only recently, the functional significance of the tubulin code, with particular emphasis on the role specified by tubulin PTMs, started to be elucidated. This is the case of α-tubulin detyrosination, which was shown to guide chromosomes during congression to the metaphase plate and allow the discrimination of mitotic errors, whose correction is required to prevent chromosomal instability—a hallmark of human cancers implicated in tumor evolution and metastasis. Although alterations in the expression of certain tubulin isotypes and associated PTMs have been reported in human cancers, it remains unclear whether and how the tubulin code has any functional implications for cancer cell properties. Here, we review the role of the tubulin code in chromosome segregation during mitosis and how it impacts cancer cell properties. In this context, we discuss the existence of an emerging “cancer tubulin code” and the respective implications for diagnostic, prognostic and therapeutic purposes.

Diversity in cytoskeleton organization and function may be achieved through variations in primary sequence of tubulin isotypes. Recently, isotype functional diversity has been linked to a “tubulin code” in which the C-terminal tail, a region of substantial sequence divergence between isotypes, specifies interactions with microtubule-associated proteins. However, it is not known whether residue changes in this region alter microtubule dynamic instability. Here, we examine recombinant tubulin with human β isotype IIB and characterize polymerization dynamics. Microtubules with βIIB have catastrophe frequencies approximately threefold lower than those with isotype βIII, a suppression similar to that achieved by regulatory proteins. Further, we generate chimeric β tubulins with native tail sequences swapped between isotypes. These chimeras have catastrophe frequencies similar to that of the corresponding full-length construct with the same core sequence. Together, our data indicate that residue changes within the conserved β tubulin core are largely responsible for the observed isotype-specific changes in dynamic instability parameters and tune tubulin’s polymerization properties across a wide range.

In the more divergent alpha 2 gene, a leucine replaces an arginine at amino acid 308, and a valine replaces a glycine at amino acid 366. The results predicted that two alpha-tubulin proteins with different net charges are produced as primary gene products. The predicted amino acid sequences are 86 and 70% homologous with alpha-tubulins from rat brain and Schizosaccharomyces pombe, respectively. Each gene had two intervening sequences, located at identical positions. Portions of an intervening sequence highly conserved between the two beta-tubulin genes are also found in the second intervening sequence of each of the alpha genes. These results, together with our earlier report of the beta-tubulin sequences in C. reinhardi, present a picture of the total complement of genetic information for tubulin in this organism.

Microtubules are cytoskeletal filaments that are dynamically assembled from α/β-tubulin heterodimers. The primary sequence and structure of the tubulin proteins and, consequently, the properties and architecture of microtubules are highly conserved in eukaryotes. Despite this conservation, tubulin is subject to heterogeneity that is generated in two ways: by the expression of different tubulin isotypes and by posttranslational modifications (PTMs). Identifying the mechanisms that generate and control tubulin heterogeneity and how this heterogeneity affects microtubule function are long-standing goals in the field. Recent work on tubulin PTMs has shed light on how these modifications could contribute to a “tubulin code” that coordinates the complex functions of microtubules in cells.

The wastewater resulting from milk production processes with animals at free stall barns has huge organic matter concentration and requires treatment before being disposed at the environment. At this study, the dairy cattle wastewater from free stall barns was treated in horizontal tubular anaerobic reactors with continuous flow and 9.6 liters volume each, made with polyvinyl chloride (PVC). The material was collected at Iguaçu Farm, with geographic coordinates latitude 25°02'43.07" south, longitude 53°46'51.85" west and 641 meters altitude, sieved at 2 mm mesh and stored at -18 °C, conditioned in polyethylene terephthalate (PET) packs. The material was put in room temperature before being added to the reactor each day. Different loads were used, resulting in retention times equals to were 8, 10, 15 and 20 days. Two temperatures were used: 30 and 35 °C; with 4 replications. Statistical data were evaluated by factorial crossover design 4 x 2 x 4, 4 factors of hydraulic retention time (HRT), 2 temperature factors and 4 replicates, with the average calculated by the Tukey method. They were evaluated the chemical oxygen demand (COD) removal, by colorimetric method, total solids (TS) and volatile solids (VS), by gravimetric methods. The best treatments were met with 20 days HRT and 30 °C, with average 83.30% 76.16% and 86.23% of COD, TS and VS removed. The biogas production was measured relatively to COD, TS and VS removals, resulting in 1.32 m³ of biogas kg-¹ DQO at 5 days of HRT and 35 ° C. At the 8 days HRT and 30 ° C, they resulted in 1.05 and 1.01 m³ of biogas · kg-¹ of TS and VS removed from the system, respectively.
A água residuária resultante dos processos das atividades de produção intensiva de leite com animais em regime de estabulação completa possui elevada concentração de matéria orgânica e necessita de tratamento antes de ser disposta no ambiente. Neste estudo, a água residuária da criação e produção de bovinos leiteiros em regime de estabulação completa foi tratada em reatores anaeróbios tubulares horizontais de fluxo contínuo, em escala laboratorial, de volume útil de 9,6 litros cada, construídos em cloreto de polivinila (PVC). O material foi coletado na Fazenda Iguaçu, com coordenadas geográficas latitude 25°02 43,07 sul, longitude 53°46 51,85 oeste e altitude 641 metros, sendo peneirado em malha de 2 milímetros e armazenado a 18 °C abaixo de zero, em embalagens de polietileno tereftalato (PET). O material foi levado à temperatura ambiente antes de ser acrescentado aos reatores a cada dia. Foram utilizadas diferentes cargas, de forma que os tempos de retenção do efluente foram de 8, 10, 15 e 20 dias. Foram empregadas 2 temperaturas: 30 e 35 °C; com 4 repetições. Os dados estatísticos foram avaliados através de delineamento fatorial cruzado 4 x 2 x 4, sendo 4 fatores de tempo de retenção hidráulica (TRH), 2 fatores temperatura e 4 repetições; com as médias calculadas pelo método de Tukey. Foram avaliados a remoção de demanda química de oxigênio (DQO), a partir da metodologia colorimétrica, sólidos totais (ST) e sólidos voláteis (SV), com métodos gravimétricos. Os melhores tratamentos ocorreram com 20 dias de TRH e 30 °C com médias de 83,30%, 76,16% e 86,23% de DQO, ST e SV removidos, respectivamente. A produção de biogás foi avaliada em relação à DQO removida e aos ST e SV retirados do sistema, com 1,32 m³ de biogás·kg-¹ DQO em 15 dias de TRH a 35 °C e 8 dias com 30 °C, 1,05 e 1,01 m³ de biogás·kg-¹de ST e SV retirados do sistema, respectivamente.

Abstract:
Tubular Joints play quite a vital role in most of the marine structures. It is obvious that having full control about the capacity and strength of tubular joints would be very worthy to have good knowledge about the total performance of the structure. In recent years the application of the finite element method has become very popular in the analyses of different types of welded circular tubular joints. The rapid development of FE programs and computation facilities has extended the analyses in the study of stress distribution and load-displacement behavior of a joint to the assessment of the ultimate capacity. This thesis presents a numerical investigation using open source finite element analysis on tubular joints that are the main structural components of steel offshore structures. The studied joint is reinforced and unreinforced T-joint. In nowadays, several types of reinforcement are available to enhance the strength of tubular joints. Thus, for a better understanding of the static strength of joints, two common types of reinforcement plate (doubler and collar) were investigated. In this regards twelve T-Joint specimens subjected to axial brace load (6 in compression and 6 in tension) have been studied, with the different geometric, material property, and reinforced plate. In addition, different types of shell and solid finite elements are considered in this investigation. The shell elements are the linear triangular and quadrilateral elements and the solid elements are the linear tetrahedral and hexahedral elements. Also, this study was performed to investigate and report the usefulness of the open-source software programs Code\_Aster and Salome-Meca in implementing static finite element modeling of tubular joints.

Робота присвячена обґрунтуванню параметрів технологічних процесів виготовлення деталей спеціальних приводів машин і механізмів односторонньої дії. Це приводи трубчатих конвеєрів, їх ланцюгових передач, приводи довбальних пристроїв та інших металорізальних верстатів, прямолінійні шліцьові поверхні зубчастих зачеплень валів односторонньої дії. Розроблена динамічна модель процесу фрезерування опуклих і увігнутих зовнішніх профілів привідних зірочок і досліджено динамічні навантаження на елементи системи верстат – пристрій – заготовка – фреза. Розроблені теоретичні передумови оброблення односторонніх прямолінійних шліцевих поверхонь валів черв’ячними фрезами методом обкатування і конструкція спеціальної фрези. Розроблено три схеми технологічних процесів виготовлення привідних зірочок з випуклими і ввігнутими радіусними профілями механізмів односторонньої дії для серійного і крупносерійного виробництва. Розроблено технологічний процес виготовлення приводних зірочок на основі багатофакторного експерименту ПФЕ33 з виведенням рівнянь регресії при обробленні їх зі сталей 45 і 40Х. Спроектовано і виготовлено трубчатий конвеєр для дослідження силових, конструктивних і технологічних параметрів. Розроблено спеціальні конструкції багатошпиндельної головки і свердлильного кондуктора для цієї головки. Особливістю конструкції є те, що в якості кондукторних втулок використовують радіально-упорні підшипники, у внутрішні кільця яких запресовують змінні втулки, які у процесі свердління обертаються разом із свердлами з можливістю зміни міжцентрових віддалей для оброблення різних типів деталей.
Работа посвящена обоснованию параметров технологических процессов изготовления деталей приводов механизмов одностороннего действия и технологического оснащения. К которым относится: трубчатые конвейеры приводы долбежных приспособлений и других металлорежущих станков, прямолинейные шлицевые поверхности зубчатых зацеплений валов одностороннего действия. Разработана динамическая модель технологического процесса фрезерования выпуклых и вогнутых наружных профилей приводных звездочек и исследовано динамические нагрузки на элементы системы станок – приспособление– заготовка – фреза. Разработаны технологические предисловия обработки односторонних прямолинейных шлицевых поверхностей валов червячными фрезами – методом обкатывания и конструкция специальной фрезы. Теоретически обосновано ударные контактные напряжения долбежных резцов в приспособлениях для изготовления внутренних винтовых шлицов. Разработан технологический процесс изготовления приводных звездочек трубчатого конвеера на основе многофакторного эксперимента ПФЕ33 с выводом уравнений регрессии при изготовлении их из сталей 45 и 40Х Спроектировано и изготовлено трубчатый конвеер для исследования силовых, конструктивных и технологических параметров элементов приводов одностороннего действия. Разработаны специальные конструкции многошпиндельной свердлильной головки и кондуктора для этой головки. Особенностью ее конструкции является то, что в качестве кондукторных втулок использовали радиально-упорные подшипники, в внутренние отверстия которых запресованы сменные втулки, которые в процессе сверления вращаются вместе со сверлами или другими инструментами с возможностью изменения межцентровых расстояний между шпинделями при обработке различных типов деталей. Разработаны прогрессивные конструкции технологического оснащения для изготовления деталей приводов машин одностороннего действия которые в значительной мере дополняют и расширяют функциональные возможности металорежущих станков и технологического оборудования. Предложенные кондукторные втулки обеспечивают повышение эксплуатационной надежности и долговечности оснастки и повышения качества продукции. Проведено технико-экономическое обоснование разработки новых технологических процессов и технологической оснастки. Основные результаты работы частично внедрено в производство в виде конструкторских разработок, оснастки, технологических процессов, методик проектирования и расчетов.
The thesis is dedicated to substantiating the parameters of technological process of manufacturing the elements of special machines’ drives and mechanisms of one-sided action. These are drives of tubular conveyers, their chain transfers, drives of metal-cutting tools’ grooving devices, rectilinear slot surfaces of toothed gearings of one-sided action cams. The analytical dependences for determining the parameters of contact steps of flexible traction non-core chains of tubular conveyers’ drives are developed as well as their access fields. The theoretical preconditions of processing the one-sided rectilinear slot surfaces of cams using the method of winding by means of worm cutters are defined. The impact contact stresses of grooving cutters in a device when manufacturing the internal screw slots are theoretically substantiated. The technological process of manufacturing the drive stars on the basis multi-fact experiment with the development of regression equation when processing them using the steels 45 and 40 X is worked out. The tubular conveyer for researching the power, structural and technological parameters is designed and manufactured. Special constructions of multi- arbor head and milling conductor to be used with this head are developed. The main feature of this construction is that the radial supporting bears are use as the conductor cores. In the internal rings of these bears the removable bushings are pressed which rotate during the milling together with drills with the possibility of change between central wholes for processing different types of details.

Abstract:
Nos travaux sur 26 gènes de la famille des WDR a permis d’en identifier sept (Atg16l1, Coro1c, Dmxl2, Herc1, Kif21b, Wdr47, Wdr89) associés à des anomalies cérébrales majeures. Cette grande famille de protéines reste pourtant peu explorée quant à ses rôles dans le développement du système nerveux central. Nous avons choisi d’étudier WDR47, dont la fonction est totalement inconnue en dépit d’une très grande similarité structurale avec LIS1, protéine à l’origine de la lissencéphalie. En combinant trois modèles expérimentaux (souris, siRNA et levure), nous avons démontré que Wdr47 est essentiel pour la survie de l’organisme et est impliqué dans la coordination motrice et le maintien de l’homéostasie énergétique avec une origine probablement centrale. Au niveau cellulaire, Wdr47 assure un rôle clé dans la dynamique des microtubules et la stabilisation du cône de croissance au travers d’interaction protéiques avec Reelin et SCG10. En outre, Wdr47 est aussi impliqué dans la prolifération neuronale et la macroautophagie. Ces résultats ont permis d’établir un lien de causalité entre une duplication de 200 kb contenant Wdr47 et des troubles de coordination motrice et une obésité hyperphagique chez un jeune patient
WD40-repeat (WDR) proteins are one of largest eukaryotic family, however little is known about their role in neurodevelopment. We investigated 26 WDR genes, and found 7 (Atg16l1, Coro1c, Dmxl2, Herc1, Kif21b, Wdr47, Wdr89) with a major impact in brain structure when inactivated in mice. We chose WDR47 for further investigation, as it is a completely unknown protein that shares striking domain similarity with LIS1. Using three independent model systems (mice, siRNA and yeast), we found an essential role of Wdr47 in survival, and key neuronal processes involving microtubule dynamics such as proliferation, autophagy and growth cone stabilization. Next we identified Reelin and superior cervical ganglion 10 (SCG10) as top interacting proteins of WDR47. Interestingly, a 200-kb duplication encompassing WDR47 was linked to poor coordination in one patient, recapitulating mouse behavioural anomalies. Together our data help unravel for the first time a key role of Wdr47 in brain

Abstract:
Oxymonads are a group of heterotrophic flagellates living in low oxygen environment. These protists inhabit mainly the gut of xylophagous insects (cockroaches, termites), with an exception of the genus Monocercomonoides, which was described from the intestinal contents of many vertebrates. On the basis of molecular data, Oxymonadida are classified into the supergroup Excavata (Cavalier-Smith, 2002; Simpson et al., 2006, Hampl et al. 2009). This thesis was focused on the diversity of genus Monocecomonoides from the morphologically simplest family Polymastigidae. The main goal of our work was to gather sequence data from strains isolated from a wide spectrum of hosts. We have obtained 26 partial sequences of the gene for the SSU rDNA in total, of which two belonged to another oxymonad, apparently genus Oxymonas. Our phylogenetic analysis indicated that the representatives of the genus Monocercomonoides form one group, however with a low bootstrap support. On the basis of published data about the presence of non-canonical genetic code in some oxymonads (Keeling and Leander, 2003; de Koning et al., 2008), we decided to explore this rare phenomenon in representatives of the genus Monocercomonoides. Pyridine is a heterocyclic molecule with a nitrogen atom that is often found in nature. As a prosthetic group taking part in redox processes in the biological system, it plays an important function in many enzymes of the living system. Pyridine is an important pharmacophore, a privileged scaffold, and a superior heterocyclic system in drug development, with various applications in anticancer research because of its ability to work on significant receptors. Typically, it is the core of several currently available medicines. In the fight against cancer, many pyridine derivatives have been shown to inhibit kinases, androgen receptors, tubulin polymerization, topoisomerase enzyme, human carbonic anhydrase, and several other targets. Researchers are now concentrating on developing pyridine novel entities with other moieties for cancer therapy. This section presents pyridine derivative synthesis and biological expansions, as well as their target receptor sites.

Finite element method is applied to calculate the stress distribution and the maximum stress location of the tubesheets, tubes and other parts of a tubular heat exchanger in a petrochemical plant. Comparison between FE results and TEMA results is discussed. The temperature and stress distribution of tubesheets and other important parts can be accurately calculated by FE method, also the safety margin of the parts in the heat exchanger can be clearly given by FE method. For multi-pass heat exchanger, FE method is more useful. By analyzing FE results, it is easily to know whether the stresses in exchangers are over the allowable value. Therefore, FE method is a good measure to aid heat exchanger design when standard of TEMA is used.

The challenges in designing high performance combustion systems have not changed significantly over the years, but the approach has shifted towards a more sophisticated analysis process. A technical discussion on combustion technology status and needs will show that the classic impediments that have hampered progress towards near stoichiometric combustion still exist. Temperature rise, mixing, liner cooling, stability, fuel effects, temperature profile control and emissions continue to confront the aerodynamic and mechanical designers with a plethora of engineering dilemmas and trade-offs. The process of combustion chamber design has taken a new meaning over the past several years as three dimensional codes and other advanced design and validation tools have finally changed the approach from a "cut and burn" technique to a much more analytical process. All of these new aspects are now integral elements of the new equation for advanced combustor design that must be fully understood and utilized. Only then will the operable, high temperature capable or low emission combustor systems needed for future military and civil aircraft as well as for stationary gas turbines can be developed. The present paper is an attempt to analyze the flow patterns within the combustion chamber of a 20 kW gas turbine engine using a CFD code CFX. It summarize the CFD simulation of the complete combustion chamber including primary zone, intermediate zone and dilution zone and finally attempts to achieve temperature distribution in the entire combustion chamber. These CFD results are then compared with experimental readings which not only validates analytical results but leads towards improved design.

Abstract. This paper presents an experimental investigation on the behaviour of concrete filled steel tube columns under axial compression. The steel columns were filled with self-compacting and self-curing concrete instead of normal conventional concrete. A test program consisting of square column, circular column and rectangular column was firstly conducted. The behaviour of three concrete filled steel tubular sections (CFSTs) under axial load is presented. The effect of steel tube dimensions, shapes and confinement of concrete are also examined. Measured column strengths are compared with the values predicted by Euro code 4 and American codes. Euro code 4, gives good estimation of self-compaction concrete. However, lower values as measured during the experiments were predicted by the American Concrete Institute (ACI) equation. Also, the effect of thickness of steel tubes, concrete cube strength and steel percentage is also studied. In addition to CFST column the steel tube also acts as confinement for concrete.

The tubular bracing members of offshore structures may sustain collision damages from the supply ships, which lead to the deterioration of the load carrying capacity of tubular bracing members. This paper presents a numerical simulation of the ultimate strength of damaged tubular bracing members under axial compression with the nonlinear finite element code ABAQUS, based on previous experimental investigations. Parametric studies are conducted to investigate the load capacity of damaged tubular bracing members, by considering the effects of diameter (D), wall thickness (H), pipe length (L) and the damage positions on the ultimate strength of tubular members. It is found that lateral damage can cause great reduction of the axial load capacity of tubular members. In addition, an approximate equation to predict the ultimate strength of tubular members based on the given damage depth is proposed.

This paper proposes a new design method for concrete-filled tubular buckling-restrained braces (CFT-BRBs) by incorporating the confinement effect on pre-buckling rigidity. A series of experiments are performed to investigate the effects of concrete strength and sectional dimension on the initial stiffness, ultimate strength, and energy dissipation behaviors. Experimental results indicate that the confined concrete plays an important role in the energy dissipating capacity of CFT-BRBs. On the other hand, the sectional dimensions of the steel tube and core are influential factors governing the ultimate failure modes of CFT-BRBs. The findings in study provide technical supports to optimize the design methods for ductile seismic performance of CFT-BRBs in low-rise and high-rise steel buildings.
