Abstract:
This article describes how the encryption algorithm (called Tu-vera) depends on the transformation of a phrase written in English into a sequence of propositional logic formulas which can be understand by a human receiver. This happens if the receiver has a set of reserved words and he/she knows the level of unfolding manipulation that the receiver needs to perform in the transformation of the phrase/sentence. The Tu-vera algorithm requires several steps like a) to give a phrase; b) to re-order words of the given phrase in order to form a propositional logic formula; c) to make use of background knowledge by performing substitutions; d) to answer questions in general subjects (like literature, biology and so forth); e) to change synonyms/antonyms (if this is feasible); f) to perform actions in order to give value to both or one operand of the logic formula and g) to conclude the final answer of the logic formula (true or false) depending of the logic values of the operands in the logic formula. Finally, a working example, in the subject of universal history is introduced.

Abstract:
To enhance the bioavailability of testosterone undecanoate (TU) and overcome the current problem of soft capsules (Andriol Testocaps®), Nano-structured lipid carriers (NLC) for TU was developed. First, suspension of TU-loaded NLC (TU-NLC) was prepared by high pressure homogenization; then adsorbent or a protective agent β-cyclodextrin was used to solidify the suspension through a vacuum system; finally, the solid powder of TU-loaded NLC (solid TU-NLC) was filled into hard capsules. The characteristics of solid TU-NLC, were investigated in vitro and vivo. The particle size of TU-NLC was about 273.3 nm, the potential was 0.156±0.04. Transmission electron microscope (TEM) revealed that the NLC was spherical and uniform. Differential scanning calorimetry (DSC) suggested the drug had been encapsulated into NLC lipid matrix. The drug release proved that solid TU-NLC showed a higher dissolution in vitro. The CaCO-2 cell permeability showed that solid TU-NLC could enhance trans-membrane absorption of the TU. Moreover, the AUC of solid TU-NLC formulations (4304±550.50 μg/L*min) was higher than commercial product Andriol Testocaps® (3075±372.50 μg/L*min). In conclusion, solid TU-NLC could enhance the rate of dissolution, and had a relatively higher bioavailability than Andriol Testocaps® in vivo Graphical Abstract.

Abstract:

Abstract:
The aim of the work was evaluate the effects of testosterone undecanoate (TU) treatment combined with moderate physical training on: the estrous cycle, body weight (BW), motor behavior (MB), and the morphohistology of the reproductive system, the liver and kidney in rats. Female Wistar rats (180 g - 250 g) were divided as follows: sedentary + TU (S + TU), trained + TU (T + TU), sedentary + vehicle (S + V), trained + vehicle (T + V). The rats swam 50 min/Day, strapped with a 5% BW load, for 4 weeks. During this training, (BW) was monitored daily as well as the estrous cycle (EC) by vaginal smear. The TU (15 mg/kg s.c) was administered 3 times/week for 4 weeks. At the end of the study, data on MB, BW and morphohistopathological changes in viscera were compiled. The (T + TU) group had on average, a higher (BW) in the fourth week compared to the first week, and (BW) higher than (S + V) and (S + TU) groups. We noted an interruption in the EC and a decrease in weight of ovaries in animals treated with TU. In addition, there was an increase in the relative weight of the heart in groups (T + V) and (T+ TU), and kidneys in group (T + TU). Histopathological analysis showed periportal congestion and isolated foci of hepatic necrosis in rats with TU. Thus, TU combined with training abolished the EC, promoted ovarian atrophy, liver necrosis, cardiac hypertrophy and a decrease in motor activity.

Abstract:
The eucaryotic elongation factor Tu (eEF-Tu) is a single polypeptide with an approximate Mr of 53,000. During protein synthesis eEF-Tu promotes the binding of aminoacyl-tRNA to the ribosome. To study the expression of the gene(s) for this factor, a genomic clone was isolated that contains a mouse eEF-Tu gene. We screened a phage genomic library with a synthetic oligonucleotide probe complementary to a region of the Saccharomyces cerevisiae and Artemia sp. eEF-Tu genes which codes for an area that is highly conserved between both yeast and Artemia sp. eEF-Tu. From approximately 75,000 phage plaques we obtained five isolates with apparently identical inserts. All five clones contained a 3.8-kilobase EcoRI fragment that hybridized to additional oligonucleotide probes corresponding to different conserved regions of eEF-Tu. We sequenced the 5' end of one genomic clone and determined the length of the cloned fragment that was protected by eEF-Tu mRNA in S1 nuclease protection assays. A quantitative S1 nuclease protection assay was used to compare the relative steady-state levels of eEF-Tu mRNA in total mRNA in total RNA isolated from hexamethylene-bisacetamide-induced murine erythroleukemia cells. The results show a dramatic reduction in the steady-state level of eEF-Tu mRNA as differentiation proceeds. A similar reduction in transcription of eEF-Tu mRNA was observed in isolated nuclei. Finally, we examined the in vivo synthesis of eEF-Tu during differentiation and found that it declined in a manner parallel to the decline in the steady-state level of eEF-Tu mRNA. In addition, we have isolated and sequenced a cDNA clone for mouse eEF-Tu. The derived amino acid sequence is compared with sequences from other eucaryotes.

Minerals 10, no. 9 (September 15, 2020): 811. http://dx.doi.org/10.3390/min10090811.

Abstract:
The thiourea (TU) leaching of gold from refractory ores can be considered an alternative to cyanidation. However, the high reagent consumption causes an increase in cost, which seriously limits its use. In order to effectively reduce the TU consumption, it is necessary to analyze the influencing parameters of gold recovery and TU consumption and apply them to the prediction of the TU leaching process. This paper investigated six potential influencing parameters and used grey relational analysis (GRA) to analyze the relational degree between each parameter and gold recovery and TU consumption. Then, the artificial neural network (ANN) model was established to simultaneously predict the gold recovery and TU consumption in the TU gold leaching process. The results of the GRA indicated that the leaching time, initial pH, temperature, TU dosage, stirring speed, and ferric iron concentration were all well related to the gold recovery and TU consumption. Therefore, the incorporation of these parameters can significantly improve the ANN model validation. The predictive results noted that the prediction accuracy of gold recovery varied from 94.46% to 98.06%, and the TU consumption varied from 95.15% to 99.20%. Thus, the predicted values corresponded closely to the experimental results, which suggested that the ANN model can accurately reflect the relationship between the operational conditions and the gold recovery and TU consumption. This prediction method can be used as an auxiliary decision-making tool in the TU gold leaching process, and it has broad engineering application prospects in engineering.

Abstract:
Abstract Context: In rodents and monkeys, a combination of hormonal and physical agents accelerates germ cell death. Objective: A “proof of concept” study was performed to investigate whether addition of heat exposure or a progestin to an androgen induces germ cell death and more complete and rapid spermatogenesis suppression. Design and Settings: A randomized clinical trial was performed at academic medical centers. Participants: We treated four groups of healthy male volunteers (18 per group) for 18 wk: 1) testosterone undecanoate (TU) 1000 mg im (first dose), followed by 500 mg im every 6 wk; 2) submersion of scrota at 43 C in water for 30 min/d for 6 consecutive days; 3) TU plus heat; and 4) TU plus oral levonorgestrel (LNG) 250 μg/d. Main Outcome Measures: Semen parameters, testicular histology, and germ cell apoptosis were the main outcome measures. Results: Heat alone and TU plus heat suppressed sperm counts more than TU alone by wk 6. By wk 9, recovery began in the heat only group, whereas spermatogenesis remained suppressed in the TU plus heat group. Oral LNG plus TU suppressed spermatogenesis earlier and more severely than TU alone. At wk 2, significantly greater germ cell apoptosis occurred in heat and heat plus TU subjects, but not in subjects without heat treatment, compared with pretreatment subjects. By 9 wk, markedly smaller seminiferous tubule diameters and fewer spermatocytes and spermatids were noted in all 12 biopsies from men receiving TU, TU plus LNG, with most dramatic differences for the TU plus heat group, whereas no differences from pretreatment biopsies were observed in men who received heat treatment only. Conclusions: Heat causes a rapid and transient suppression of spermatogenesis. TU plus heat resulted in low-sperm output that was maintained by continuous treatment with TU. Addition of an oral progestin accelerated spermatogenesis suppression by TU alone. Increased germ cell apoptosis contributed to suppression of spermatogenesis.

Abstract:
Male hypogonadism is often treated by testosterone (T) replacement therapy such as oral administration of the ester prodrug, testosterone undecanoate (TU). However, the systemic exposure to T following oral TU is very low due to esterase-mediated metabolism, particularly in the small intestine. The aim of this work was to examine the esterase-inhibitory effect of natural fruit extract of strawberry (STW) on the intestinal degradation of TU as a potential approach to increasing the oral bioavailability of T. Herein, the hydrolysis of TU was assessed in fasted state simulated intestinal fluid with added esterase activity (FaSSIF/ES) and Caco-2 cell homogenates in the presence of STW extract. It is noteworthy that STW substantially inhibited the degradation of TU in FaSSIF/ES and Caco-2 cell homogenates at concentrations that could be achieved following oral consumption of less than one serving of STW fruit. This can significantly increase the fraction of unhydrolyzed TU in the intestinal lumen as well as in enterocytes. In addition, it was demonstrated that TU has high intestinal lymphatic transport potential as the association of TU with plasma-derived human chylomicrons was in the range of 84%. Therefore, oral co-administration of TU with STW could potentially increase the intestinal stability of TU and consequently the contribution of lymphatically delivered TU to the systemic exposure of T in vivo.

Abstract:
A conundrum has long lingered over association of cytosol elongation factor Tu (EF-Tu) with bacterial surface. Here we investigated it withAcinetobacter baumannii, an emerging opportunistic pathogen associated with a wide spectrum of infectious diseases. The gene forA. baumanniiEF-Tu was sequenced, and recombinant EF-Tu was purified for antibody development. EF-Tu on the bacterial surface and the outer membrane vesicles (OMVs) was revealed by immune electron microscopy, and its presence in the outer membrane (OM) and the OMV subproteomes was verified by Western blotting with the EF-Tu antibodies and confirmed by proteomic analyses. EF-Tu in the OM and the OMV subproteomes bound to fibronectin as detected by Western blot and confirmed by a label-free real-time optical sensor. The sensor that originates from photonic crystal structure in a total-Internal-reflection (PC-TIR) configuration was functionalized with fibronectin for characterizing EF-Tu binding. Altogether, with a novel combination of immunological, proteomical, and biophysical assays, these results suggest association ofA. baumanniiEF-Tu with the bacterial cell surface, OMVs, and fibronectin.
