Abstract:
Abstract The increased quality and data availability of large-scale transcriptomic, genomic, and proteomic studies require a pan-cancer integrated proteogenomic approach to define tumor molecular signatures accurately and identify new therapeutic targets. We comprehensively investigate more than 1000 samples across 12 cancer types from the Clinical Proteomics Tumor Analysis Consortium (CPTAC) and the International Cancer Proteogenome Consortium (ICPC). The types are comprised of breast (BR), colorectal (CO), and ovarian (OV) cancers, clear cell renal cell (ccRCC), head and neck squamous cell (HNSCC), lung squamous cell (LSCC), hepatitis B virus (HBV)-related hepatocellular (HCC), and endometrial (EC) carcinomas, lung adenocarcinoma (LUAD), pancreatic ductal adenocarcinoma (PDAC), and glioblastoma (GBM). In particular, we examine 8 Tumor Signature Associated Phenotypes (TSAPs), namely aristolochic acid (AA), aging, microsatellite instability (MSI), homologous recombination deficiency (HRD), POLE, APOBEC, smoking, and ultraviolet (UV) light exposure. This study is the first to report proteomic markers associated with these TSAPs on a pan-cancer level. In addition to genetic alterations and mutational signatures, we utilize multi-omics data of high-resolution proteome, phosphoproteome, acetylome, and gene expression to infer expression signatures of TSAPs by defining the most critical changes in the transcriptome and proteome accompanying the transitions to these TSAPs, especially markers that were uniquely found in proteomic data. We consolidated multi-omic data and calculated the novel quantitative Tumor Signature Associated Phenotypes (TSAPs) score to predict the TSAP status. For example, the use of proteomic markers for MSI-TSAP scoring can improve clinical testing of MSI status. We further study environmental exposure-related tumor proteogenomic signatures, immune proteogenomic signatures, and the association between the immune subtypes and TSAPs. Smoking strongly influences the tumor immune microenvironment and disease prognosis. We show that expression signatures can facilitate the prediction of TSAPs and help to uncover their underlying molecular mechanisms. By connecting these findings with druggable databases, we provide a link to actionable therapies, identify putative TSAP-related targets, and offer novel cues to optimize therapeutic options for patients, such as how additional targeting of genes up-regulated in PARP1 inhibitor-treated HRD tumors may overcome resistance. This will promote the identification not only of unique druggable targets, but also to determine putative novel therapeutic targets using integrated approaches. Citation Format: Yize Li, Nadezhda V. Abstract:
Abstract STEAP3/TSAP6 encodes a ferrireductase that is involved in the acquisition of iron by developing erythroblasts and steap3/tsap6 null-mice display severe microcytic anemia. We report a family in which 3 siblings born to healthy parents display transfusion-dependent hypochromic anemia. A nonsense STEAP3/TSAP6 was identified in the siblings at the heterozygous state. This mutation was inherited from their father while no mutation was found in their mother. A large variability of expression was found between normal alleles in a control population, confirming a previous report that STEAP3/TSAPS6 is an expressed quantitative trait locus (e-QTL). Determination of the relative allele expression showed that the “normal” allele was expressed at a significantly higher level in the father than in the affected siblings relative to the shared mutated allele. The blood level of STEAP3/TSAP6 mRNA was severely reduced in the siblings, while both parents were in the lower range of normal controls. The STEAP3/TSAP6 protein was also reduced in lymphocytic cell lines from the patients. Collectively, our data support the hypothesis that STEAP3/TSAP6 deficiency leads to severe anemia in the affected siblings and results from the combination of a mutated allele inherited from their father and a weakly expressed allele inherited from their mother.

Abstract:
La titulación de Técnico Superior en Audiología Protésica (TSAP) nació en España en el año 2001. Desde entonces, han surgido títulos propios de posgrado en audiología. Sin embargo, no existe aún en España el grado universitario de audiología general. El objetivo de este estudio fue examinar los lugares de trabajo, los servicios ofrecidos y la percepción de la formación de los TSAP con y sin formación universitaria y si éstos ejercían labores propias del audiólogo general propuesto por la Federación Europea de Sociedades Audiológicas (EFAS). Un cuestionario en línea fue completado por 255 TSAP. Se utilizaron análisis de varianza para comparar las respuestas entre TSAP sin formación complementaria, TSAP con grado de una disciplina afín a la audiología, TSAP con posgrado en audiología, y TSAP con ambos complementos. Independientemente de su nivel educativo, la mayoría de los TSAP indicaron desempeñar su labor profesional en centros auditivos adaptando audífonos a pacientes adultos, para lo cual percibieron estar suficientemente formados. Los TSAP con mayor nivel educativo indicaron trabajar en centros más especializados, así como atender a pacientes pediátricos con más frecuencia, y percibieron que su formación en implantes cocleares era mejor que los TSAP. En general, los TSAP no reciben formación ni ofrecen servicios en áreas que están dentro de las competencias del audiólogo general, tales como la programación de implantes cocleares o el diagnóstico de problemas vestibulares. Los estudios futuros deberán enfocarse en conocer la formación que reciben los profesionales que ofrecen este tipo de servicios audiológicos en España.

Type IV pili (T4P) are ubiquitous and versatile bacterial cell surface structures involved in adhesion to host cells, biofilm formation, motility, and DNA uptake. In Gram-negative bacteria, T4P pass the outer membrane (OM) through the large, oligomeric, ring-shaped secretin complex. In the β-proteobacteriumNeisseria gonorrhoeae, the native PilQ secretin ring embedded in OM sheets is surrounded by an additional peripheral structure, consisting of a peripheral ring and seven extending spikes. To unravel proteins important for formation of this additional structure, we identified proteins that are present with PilQ in the OM. One such protein, which we name T4P secretin-associated protein (TsaP), was identified as a phylogenetically widely conserved component of the secretin complex that co-occurs with genes for T4P in Gram-negative bacteria. TsaP contains an N-terminal carbohydrate-binding lysin motif (LysM) domain and a C-terminal domain of unknown function. InN. gonorrhoeae, lack of TsaP results in the formation of membrane protrusions containing multiple T4P, concomitant with reduced formation of surface-exposed T4P. Lack of TsaP did not affect the oligomeric state of PilQ, but resulted in loss of the peripheral structure around the PilQ secretin. TsaP binds peptidoglycan and associates strongly with the OM in a PilQ-dependent manner. In the δ-proteobacteriumMyxococcus xanthus, TsaP is also important for surface assembly of T4P, and it accumulates and localizes in a PilQ-dependent manner to the cell poles. Our results show that TsaP is a novel protein associated with T4P function and suggest that TsaP functions to anchor the secretin complex to the peptidoglycan.

Thermo-sensitive polyurethane (TSPU) was synthesized from poly(ε-caprolactone) and 4,4’-methylenebis (phenyl isocyanate) by a two-step process with 1,4-butanediol as the chain extender. Following that, a novel temperature-sensitive material was created by the strategy of IPN from TSPU and PNIPAAm in the method of in situ polymerization. The chemical structure and thermo properties of the semi-IPN were characterized with FT-IR and DSC. The results prove that intensive inter-molecular interactions exist between TSPU and PNIPAAm chains, which have significant influence on the phase transition behaviors of the material. According to these results, the transition temperature of the semi-IPN can be adjusted in the range of 30~40 °C by controlling the composition of TSPU and the semi-IPNs.

The influence of head-out water immersion (WI) (37 degrees C) on baroreflex control of heart rate was studied in five trained, instrumented, conscious dogs. Arterial pressure was raised and lowered using occluder cuffs implanted around the aorta and inferior vena cava. Function curves relating transmural systolic arterial pressure (TSAP = systolic arterial pressure-pleural pressure) to heart rate (HR) were constructed to compare responses in air and during WI. The resting TSAP in air [142 +/- 8 mmHg (mean +/- SE) at 78 +/- 6 beats/min] increased significantly during WI (161 +/- 9 mmHg at 109 +/- 9 beats/min). During WI, the saturation TSAP at the bradycardia plateau of the relation increased significantly, by 19 mmHg, whereas the average gain (slope) of the relation decreased significantly, from -1.426 to -0.752 beats.min-1.mmHg-1. Therefore, WI elicits both a resetting and a decrease of the average gain of the TSAP-HR relation. The heart rate range increased during WI as well. After cholinergic blockade with atropine, WI did not elicit a resetting of the relation and the change in average gain was abolished. However, after beta 1-blockade with metoprolol, the resting TSAP increased significantly during WI and resetting persisted, but the decrease of average gain was abolished. Therefore, the alteration of the TSAP-HR relation in WI is achieved via a modulation of both adrenergic and cholinergic regulation of HR.

Abstract:
Objective. We recently reported on the expression and clinical role of molecules that mediate exosome secretion in high-grade serous carcinoma. In the present study, the biological role of these molecules was analyzed. Methods. OVCAR8 and ES-2 ovarian carcinoma cells were studied using a combination of CRISPR/Cas9 technology and two 3D in vitro models—spheroids emulating effusions and alginate scaffolds representing solid lesions. Isolation of exosomes was validated by electron microscopy. TSAP6, NSMASE2, RAB27A, and RAB27B mRNA and protein levels were analyzed using qRT-PCR and Western blotting, respectively. Tumor aggressiveness was studied in vitro using scratch assay, invasion assay, and matrix metalloproteinase (MMP) activity assay and in vivo using a mouse model. Results. In OVCAR8 cells, mRNA expression of TSAP6 and RAB27A was significantly higher in spheroids compared to scaffolds, whereas the opposite was true for NSMASE2 mRNA. In ES-2 cells, TSAP6 and RAB27B mRNA expression was significantly higher in spheroids versus scaffolds. In addition, nSMase2 and TSAP6 protein expression was significantly higher in scaffolds compared to spheroids. CRISPR-edited cells with silencing of NSMASE2, TSAP6, and RAB27A/B had reduced migration, invasion, and MMP activity. Additionally, knockout (KO) of these molecules resulted in significantly diminished exosome secretion. In vivo assay showed that when injected to mice, OVCAR8 RAB27A/B KO cells, as opposed to control OVCAR8 cells, did not form ascites or visible tumor lesions and had reduced MMP expression. Conclusion. The present study provides evidence that different models for culturing ovarian carcinoma cells affect the expression of molecules mediating exosome secretion and that these molecules have a tumor-promoting role. Silencing these molecules may consequently have therapeutic relevance in this cancer.

Abstract:
Translationally controlled tumor protein (TCTP) is cytoplasmic and structurally related to guanine-nucleotide free chaperones. TCTP (also called histamine-releasing factor) has been described previously as a secreted protein that participates in inflammatory responses by promoting the release of histamine. How TCTP is eventually exported out of the cell to promote such activities is unknown. Here we show that TCTP secretion was insensitive to either brefeldin A or monensin, suggesting that it proceeds via an endoplasmic reticulum/Golgi-independent or nonclassical pathway. Moreover, our analyses also suggest that secreted TCTP originates from pre-existing pools. TSAP6, a p53-inducible 5–6 transmembrane protein, was found to interact with TCTP in a yeast two-hybrid hunt. GST pull down assays confirmed their direct interaction, and immunofluorescence analysis revealed their partial co-distribution to vesicular-like structures at the plasma membrane and around the nucleus. Functionally, the overexpression of TSAP6 consistently leads to enhanced secretion of both endogenously and exogenously expressed TCTP. Finally, we found TCTP in preparations of small secreted vesicles called exosomes, which have been suggested as a possible pathway for nonclassical secretion. Overexpression of TSAP6 also increased TCTP levels in exosome preparations. Altogether, these data identify a novel role for TSAP6 in the export of TCTP and indicate that this multipass membrane protein could have a general role in the regulation of vesicular trafficking and secretion.

Abstract. The plasma levels of thermolabile (TLAP) and thermostable (TSAP) alkaline phosphatase were investigated in adult male white-tailed deer. Distinct seasonal variation of TLAP (with highly elevated levels in July) were observed, whereas TSAP exhibits low concentration with a small increase in October. No circadian rhythm was found for TLAP or TSAP. A close correlation (r = 0.98) between TLAP activity and antler weight was found. Administration of ACTH or dexamethasone were ineffective in influencing AP activity. On the other hand, variations of triiodothyronine (T3) levels in plasma induced by thyroxine (T4) injections correlated well with concentration of TLAP.

Heritage towns in many countries are facing the pressure of redevelopment due to economic needs and also in the name of tourism. It is unfortunate that many a times, the conservation process is separated from development planning cum redevelopment thus destroying the urban fabric. Taiping is one such town that has been identified to be developed for heritage tourism under the Special Area Plan development plan upon its declaration as a Heritage Town in 1999. The Taiping Special Area Plan (TSAP 2010) was launched in 2010 with seven development plans set within the Taiping Town and the Lake Gardens Conservation Zone. The objective of this study is to explore the level and type of change to the façade characteristics of buildings located within Taiping Town Conservation Zone (TTCZ) since the implementation of TSAP 2010 and the conformance of these buildings with change to the TSAP 2010 guidelines. The research adopted a case study strategy with a field survey of the entire building units located within TTCZ followed by a document review of the TSAP 2010 guideline. The TTCZ photo inventory was cross-analysed with archival data to firstly identify the level of change based on a numerical rating scale. The data for building units with change were then analysed further to identify the type of change followed by its adherence to the TSAP 2010 guidelines. The findings revealed that there were changes to the building’s façade characteristics even though the overall magnitude of change was low. The changes were found to be mainly on the shop house typology as opposed to other typologies and the shop houses with changes did not adhere to the TSAP 2010 guidelines. The study concluded that the façade characteristics of building units has changed prior to 2010 and the TSAP 2010 guidelines may have not been implemented fully though this may need to be studied further. Edge computing is becoming increasingly commonplace, as consumer devices become more computationally capable and network connectivity improves (e.g., due to 5G). With the rapid development of edge computing and Internet of Things (IoT), the use of edge-cloud collaborative computing to provide service-oriented network application (i.e., task) in edge-cloud IoT has become an important research topic. In this paper, we present an edge-cloud collaborative computing framework and our resource deployment algorithm with task prediction (RDAP). Based on our paradigm, tasks in the cloud service center are predicted using the two-dimensional time series, and task classification aggregation and delay threshold determination are combined to optimize task resource deployment of edge servers. A task scheduling algorithm with Pareto improvement (TSAP) is also proposed. At the edge servers, the Pareto progressive comparison is conducted in two stages to obtain the tangent point or any intersection point of the two objective curves of user’s quality of service and effect of system service to optimize task scheduling. The experimental results show that for varying user task scales and different Zipf distribution α parameters, combining RDAP and TSAP (RDAP-TSAP) can improve the average user task hit rate. In addition, the average task completion time of users, the overall system service effect, and the total task delay rate of RDAP-TSAP are better than TSAP and the benchmark algorithms for task scheduling.

Abstract:
A detailed kinetic analysis of pyrolysis processes of Tennis string polyurethane (TSPU) waste and waterborne polyurethane (WPU) waste was carried out in the present paper. Non-isothermal pyrolysis characterizations of two wastes were acquired through thermogravimetric determinations under the constant heating rates of 5, 10, 15 and 20 K/min. Experimental results showed that the pyrolysis processes of TSPU and WPU were mainly characteristic of three stages and two stages, respectively. Two pyrolysis performance indices, the devolatilization index (DI) and heat-resistance index (HRI), were used to interpret the heating rate effect on the pyrolysis features and different thermal dependences of TSPU and WPU. Isoconversional kinetic analysis was thoroughly performed with model-free and model-fitting methods. By using Starink, Vyazovkin–Dollimore and Coats–Redfern methods, the activation energies thus obtained were in the range of 103.3~148.3 kJ/mol and 92.5~204.3 kJ/mol, respectively, for TSPU and WPU, over the entire pyrolysis process. Their respective pre-exponential factor lnA values were determined to be within 17.94~33.42 min−1 and 16.56~20.82 min−1. Thermodynamic parameters in terms of ΔG#, ΔH# and ΔS# throughout the entire pyrolysis process were also calculated. Finally, by means of the model-fitting Coats–Redfern method, the most appropriate mechanism functions were ascertained for, respectively, describing multi-stage pyrolysis degradations of TSPU and WPU waste. These results may offer meaningful support for designing any industrial pyrolysis reactor to dispose of polyurethane wastes.

Abstract STEAP3 (Six-Transmembrane Epithelial Antigen of Prostate 3) is the major ferrireductase in the erythroblast. Also named TSAP6 (Tumor Supressor Activated Pathway 6) after it had been found to play a role in cancer, its total ablation in the mouse leads to severe microcytic and hypochromic red cells with moderate anemia. The protein function appears conserved among mammals, as patients carrying a nonsense mutation in the TSAP6/STEAP3 gene have been reported with hypochromic anemia. Here, we investigated the mechanism leading to the anemia. In the present study, using the TSAP6/Steap3 knockout mice, we undertook a comprehensive hematologic characterization of the red cell compartment. Red cell indices derived using ADVIA 120 blood counter confirmed the hypochromic microcytic anemia phenotype with a marked reduction in the mean corpuscular volume (MCV; 21.5fL ± 1.3fL in knockout vs 45.2fL ± 1.5fL in wild-type X ± SD, p<0.001) and mean cell hemoglobin content (MCH; 5.8pg ± 0.1pg vs 14.5pg ± 0.1pg, p<0.001). The reticulocyte count was marginally elevated (5.7% ± 1.2% vs 2.6% ± 0.4%, p<0.05) indicating that the anemia is proliferative. By phase contrast microscopy and transmission electron microscopy, we observed marked anisocytosis as well as the presence of fragmenting erythrocytes. Consistent with this observation, we found by ektacytometry decreased membrane mechanical stability of knockout red cells. Interestingly, we were unable to document significant changes in the expression levels of the major skeletal and transmembrane proteins to account for this decrease in the membrane stability. As defects in either the production or destruction of red cells can lead to anemia, we measured red cell survival and erythropoiesis in these mice. No differences in red cell survival could be documented using biotin labeled red cells implying that decreased survival cannot account for the anemia. However, when we monitored erythropoiesis using Ter119, CD44 and Forward Scatter (FSC) as markers of terminal differentiation, we found a decreased number of proerythroblasts in the bone marrow of TSAP6/Steap3-/- animals (2.20% ± 0.49% vs 4.07% ± 0.77%, p<0.01). In addition, progression from the proerythroblastic to the orthochromatic stage was also affected, with accumulation of cells at the polychromatic stage. These findings imply that ineffective erythropoiesis is the dominant cause of anemia in these mice and that TSAP6/Steap3 may play a role during erythropoiesis beyond its role as ferrireductase. It is interesting to note that while the phenotype of microcytic red cells with decreased hemoglobin content in the TSAP6/Steap3 null mouse is similar to red cells in thalassemic and porphyria mice, there are distinct differences in the etiology of the hematologic phenotype. In order to match the solid food oxidation during logistics and storage process under severe high temperature, a double-switch temperature-sensitive controlled release antioxidant film embedded with lyophilized nanoliposomes encapsulating rosemary essential oils (REOs) was prepared. The double switch temperature at 35.26 and 56.98 °C was achieved by development of a temperature sensitive polyurethane (TSPU) film. With biaxially oriented polyethylene terephthalate (BOPET) as a barrier layer, the intelligent complex film was prepared via coating the TSPU embedded with lyophilized nanoliposomes encapsulating REOs on BOPET. The results indicate that the REO is well encapsulated in nanoliposomes with encapsulation efficiency (EE) of 67.3%, high stability and lasting antioxidant effect during 60 days. The incorporation of lyophilized nanoliposomes containing REOs into TSPU remains the double-switch temperature-sensitive characteristic of the prepared TSPU. In agreement with porosity and WVTR results, the diffusion coefficient (D) of the antioxidant complex film sharply increases respectively at two switching temperatures, indicating that the intelligent double-switch temperature-sensitive controlled release property is functioning. Furthermore, compared with films directly added with REO, the lower Ds of films added with lyophilized nanoliposomes encapsulating REOs provides a longer-lasting antioxidant activity. Thus, the acquired controlled release antioxidant film sensitive to temperature at 39.56 and 56.00 °C can be potentially applied for protection of solid food during distribution and storage process under severe high temperatures.

University students often share that studying abroad was a life-changing experience. We explored how one short-term, faculty-led study abroad, Tanzania Study Abroad Program (TSAP), contributed to the participants’ lives after their experience. Through a web-based survey, we collected responses from 82 former TSAP participants. We used corpus and thematic analysis to determine prominent themes of participants’ memories and perceptions of how TSAP impacted their lives. Respondents reflected on the people they interacted with throughout their study abroad experiences, both with those abroad and their peers. Furthermore, the program leaders’ intentional actions in the planning of the context, curriculum, and community engagement informed participants’ opportunities to develop relationships that were non-transactional in nature. Participants also reported that their cross-cultural relationships with Tanzanians positively influenced their subsequent interactions with people from diverse communities. Prioritizing relationships with host community members and peers on study abroad shows promise for supporting participants’ intercultural development.

Подробно описана работа студентов ТГПУ в Центре учебной помощи по оказанию консультационных услуг предметного и технического характера школьникам и родителям при выполнении домашних заданий, а также отряда цифровых волонтеров, выезжающих в школы и помогающих учителям в освоении новых образовательных технологий. The work of TSPU students in the Center for Educational Assistance in providing consulting services of a subject and technical nature to schoolchildren and parents when doing homework, as well as the Detachment of digital volunteers who travel to schools and help teachers in mastering new educational technologies are described in detail. The projects of the Association TSPU “Technovolunteers of Education” are socially significant. They are aimed at the formation of new pedagogical competencies associated with the use of the modern digital environment of a single educational space in the educational process and based on the concept of joint activities. In addition to the main projects of the association “Technovoloters Education” TSPU, volunteers can be involved as organizers’ assistants in various events held by TSPU or its partners.

Abstract Heart valve tissue engineering aims at creating living valves through colonization of scaffolds with patient’s own cells. Various cell sources have been explored focusing mainly on endothelialization of the scaffold surface. Endothelial like cells, such as endothelial progenitor cells (EPCs), which can be isolated from peripheral blood or bone marrow could be a suitable option. In this study we investigated cell colonization potential of ovine EPCs (OEPCs) on thermoplastic silicone-based polyurethane (TSPU) polymer scaffolds. TSPU nonwovens with and without vascular endothelial growth factor (VEGF) functionalization were used. SEM images showed that by day 3 the cells were growing as patches on the surface of both polymer groups. The cell patches continued growing and started covering more of the nonwoven surface. On day 7 the cells had almost covered the scaffold surface. The cells were more uniformly distributed as monolayer on the functionalized TSPU compared to the non-functionalized nonwovens. Live/Dead staining provided bright green fluorescence on the samples, indicating metabolically active alive cells. These static cell seeding experiments demonstrated that functionalized TPSU nonwovens support endothelialization. The feasibility of TPSU nonwovens as heart valve prosthesis scaffold could be further explored with animal studies in an ovine model.

It is common practice to dilute food products in 0.1% peptone before microbiological analysis. However, this diluent may not be appropriate for detection of injured organisms present in acidic foods. Shelf-stable unclarified apple juice (pH 3.6) was inoculated with approximately 1 × 107 CFU/ml of Escherichia coli O157:H7 and held at 23 ± 2°C (control) or frozen to −20 ± 2°C for 24 h to induce injury before sampling. Unfrozen or frozen and thawed juice was diluted 1:1 or 1:10 in 0.1% (wt/vol) peptone (pH 6.1) or 0.1 M phosphate buffer (pH 7.2). Juice samples were plated onto tryptic soy agar with 0.1% (wt/vol) sodium pyruvate (TSAP) to measure survival or onto sorbitol MacConkey agar (SMA) to indicate injury. Counts on TSAP or SMA were the same for control samples held in peptone or phosphate buffer for up to 45 min. However, populations of E. coli in frozen and thawed samples declined rapidly upon dilution in 0.1% peptone. Within 20 min, E. coli underwent a &gt;1-log10 CFU/ml reduction in viability as measured on TSAP and a &gt;2-log10 CFU/ml reduction to below the limit of detection (1.6 or 2.3 log10 CFU/ml) on SMA. In contrast, populations of E. coli in frozen and thawed samples diluted in phosphate buffer did not decrease significantly on TSAP and decreased by &lt;0.6 log CFU/ml on SMA during a 45-min holding period. The acidity of apple juice appears to interfere with the recovery of freeze–thaw-injured E. coli O157:H7 during sampling. Using 0.1 M phosphate buffer (pH 7.2) as a diluent results in superior recovery of these organisms on both selective and nonselective plating media.

ABSTRACT A study was done to determine the performance of differential, selective media for supporting resuscitation and colony development by stressed cells of Enterobacter sakazakii. Cells of four strains of E. sakazakii isolated from powdered infant formula were exposed to five stress conditions: heat (55°C for 5 min), freezing (−20°C for 24 h, thawed, frozen again at −20°C for 2 h, thawed), acidic pH (3.54), alkaline pH (11.25), and desiccation in powdered infant formula (water activity, 0.25; 21°C for 31 days). Control and stressed cells were spiral plated on tryptic soy agar supplemented with 0.1% pyruvate (TSAP, a nonselective control medium); Leuschner, Baird, Donald, and Cox (LBDC) agar (a differential, nonselective medium); Oh and Kang agar (OK); fecal coliform agar (FCA); Druggan-Forsythe-Iversen (DFI) medium; violet red bile glucose (VRBG) agar; and Enterobacteriaceae enrichment (EE) agar. With the exception of desiccation-stressed cells, suspensions of stressed cells were also plated on these media and on R&F Enterobacter sakazakii chromogenic plating (RF) medium using the ecometric technique. The order of performance of media for recovering control and heat-, freeze-, acid-, and alkaline-stressed cells by spiral plating was TSAP > LBDC > FCA > OK, VRBG > DFI > EE; the general order for recovering desiccated cells was TSAP, LBDC, FCA, OK > DFI, VRBG, EE. Using the ecometric technique, the general order of growth indices of stressed cells was TSAP, LBDC > FCA > RF, VRBG, OK > DFI, EE. The results indicate that differential, selective media vary greatly in their abilities to support resuscitation and colony formation by stressed cells of E. sakazakii. The orders of performance of media for recovering stressed cells were similar using spiral plating and ecometric techniques, but results from spiral plating should be considered more conclusive.

This study was undertaken to determine whether pretreating inoculated Gala apple slices with metabisulfite or acidic solutions enhanced the inactivation of Salmonella during dehydration and storage. Apple slices inoculated with a five-strain mixture of Salmonella (7.6 log CFU/g) were pretreated, dried for 6 h at 60°C, and stored aerobically at 25°C for 28 days. Predrying treatments included (i) no treatment, (ii) 10 min of immersion in sterile water, (iii) 10 min of immersion in a 4.18% sodium metabisulfite solution, (iv) 10 min of immersion in a 3.40% ascorbic acid solution, and (v) 10 min of immersion in a 0.21% citric acid solution. Samples were plated on tryptic soy agar with 0.1% pyruvate (TSAP), brilliant green sulfa (BGS) agar, and xylose lysine tergitol 4 (XLT4) agar for the enumeration of bacteria. Populations were not significantly (P &gt; 0.05) reduced by immersion in water but were reduced by 0.7 to 1.1 log CFU/g by immersion in acidic solutions. Immersion in the sodium metabisulfite solution reduced populations by 0.4, 1.3, and 5.4 log CFU/g on TSAP, BGS agar, and XLT4 agar, respectively. After 6 h of dehydration at 60°C, populations on untreated and water-treated slices were reduced by 2.7 to 2.8, 2.7 to 2.9, and 4.0 to 4.2 log CFU/g as determined with TSAP, BGS agar, and XLT4 agar, respectively. In contrast, populations on slices treated with sodium metabisulfite, ascorbic acid, and citric acid were reduced after 6 h of dehydration by 4.3, 5.2, and 3.8 log CFU/g, respectively, as determined with TSAP; by 4.7, 5.5, and 3.9 log CFU/g, respectively, as determined with BGS agar; and by 5.5, 5.7, and 5.6 log CFU/g, respectively, as determined with XLT4 agar. Bacteria were still detectable by direct plating after 28 days except on slices treated with ascorbic acid. Immersion in metabisulfite or acidic solutions prior to dehydration should enhance the inactivation of Salmonella during the dehydration and storage of Gala apple slices.

Esta revisão de estudos teve como objetivo analisar como é avaliada a performance dos atletas nos desportos coletivos e individuais através dos instrumentos Game Performance Assessment Instrument (GPAI) e Team Sport Assessment Procedure (TSAP). Para tal, efetuou-se um levantamento de todos os instrumentos de avaliação utilizados nos últimos 25 anos e analisaram-se as modalidades onde foram usados. A revisão dos estudos mostra uma maior utilização destes instrumentos nos desportos coletivos (21estudos), sendo o Futebol a modalidade mais estudada (9 estudos). Nos desportos individuais, o Tênis, com 2 estudos, e o Badminton com 1 estudo, foram as modalidades mais abordadas. Conclui-se que o GPAI foi o instrumento mais utilizado, isto devido à adaptação que pode ser feita na análise das componentes técnicas e táticas, as quais têm aplicações práticas nos desportos coletivos e individuais.

Les études de Richard, Godbout et Gréhaigne (1998 et 2000) ont regardé différentes facettes de son intégration aux programmes d’éducation physique dans les écoles primaires du Nouveau- Brunswick (5e à 8e année). La présente étude a pour but de continuer le processus de validation de cette procédure d’évaluation dans différentes activités d’apprentissages souvent exploitées par les programmes d’éducation physique (soccer et volley-ball) et de concevoir des versions modifiées favorisant une intégration graduelle des élèves dans le processus d’évaluation (évaluation par les pairs). Les résultats obtenus démontrent que le TSAP et ses versions modifiées fournissent des mesures valides permettant de discriminer la performance offensive globale au soccer et au volley-ball.

BACKGROUND: Several tactical assessment tools have been proposed to evaluate players’ tactical performance in more representative contexts. AIM: Investigate the associations between the results from different tactical assessment tools, TSAP and FUT-SAT, in youth soccer players. METHOD: Twenty-seven male U-11 soccer players participated in the study (mean age = 11.02 ± 0.78 years). The players' technical-tactical performance was analyzed using the Team Sports Performance Assessment Procedure - TSAP. The players' tactical performance and behavior were assessed using the Football Tactical Assessment System - FUT-SAT. Pearson's correlation test was used to analyze the association between the performance indexes obtained through both assessment tools. RESULTS: With the results obtained, it was possible to verify that both assessment tools' performance indexes did not present a significant correlation. CONCLUSION: Through these results, it is possible to conclude that it is extremely necessary for the correct choice of the assessment tools to be used, considering their characteristics and limitations, as well as their relation with tactical content to be emphasized during the learning process.

En baloncesto, la falta de un instrumento válido y fiable para evaluar de forma objetiva el nivel de conocimiento táctico, es el problema principal en los procesos de formación y en las comunidades de entrenamiento. Team Sport Assessment Procedure fue ideado por Grahaigne, Godbout &amp; Bouthier, brindando a los profesores datos objetivos sobre las actuaciones ofensivas de los estudiantes. Dos jugadores profesionales fueron observados mediante análisis de video por tres observadores, a fin de determinar la validad del TSAP en baloncesto táctico en situación de partido. El valor del coeficiente k de Cohen (k = 0,88) revela un acuerdo entre observadores casi perfecta lo que significa que los observadores registraron las mismas acciones y los interpretan de la misma manera. Los resultados muestran que el TSAP es una herramienta válida y fiable para evaluar elementos ofensivos en juego del partido y fácilmente utilizable por entrenadores y jugadores.</p><p> </p>

В качестве примера применения данного алгоритма проведено исследование процессов конформационной динамики комплексов [Tm(H2O)(DOTA)]–, обусловленной взаимопревращением двух конформеров SAP и TSAP. Aims and Objectives: Motor control is described as the internal control and regulation processes which facilitate movements. Scientists divide motor control into the function of stability and the movement to maintain balance in locomotion. The FMS is used to analyze the motor behavior of muscle slings on the one hand, and to estimate injury risk on the other hand. Yet, the true extent of the FMS’ diagnostic information remains unclear. The low intensity level of the FMS items causes low tension within the motor network. Based on Hennemen’s size principle, this is a clear indicator for the activation of slow-twitch fibers in the muscle. A possible relationship between movement and stability function of muscle slings has yet to be examined. Therefore, the aim of this investigation was to shed light on the motor behavior of dynamic and isometric muscle activity as well as the examination of average muscle activity when performing the items deep squat (DS) and trunk stability pushup (TSPU). Materials and Methods: Data from 16 collegiate athletes (22.1±1.5yrs) with various sports histories was analyzed. The data analysis was based on Kendall’s tau correlation coefficient. The items DS and TSPU were rated via an ordinal ranking system ranging from 0 (pain) to 3 (adequate). Muscle activity was recorded with a new EMG device at a rate of 25 Hz (Myontec Ltd, Kuopio, Finland). In a concurrent pilot study the reliability of this device was examined with a progressive loaded squat protocol (50 up to 125% BW). It indicated almost perfect reliability for the quad/ham balance, quadriceps distribution (r< 0.9, CV:3-5%) and high reliability for the hamstrings (r >0.8, CV:<10%). Results: Strong relationships between the quad/ham balance (r=0.7, p<0.01) and quadriceps distribution (r=0.6, p<0.03) in DS and TSPU were identified. The connection between hamstring muscle activity balance was low (r< 0.2, p>0.5). In both items, the muscle contractions were dominated by the activation of the quadriceps (79% vs. 21% hamstrings). Additionally, the average muscle activity was 4 times higher in a loaded squat (100% BW) than it was in DS. Generally, DS was rated as follows: score 3 (3 times) and score 2 (13 times). For the item TSPU score 3 was given 14 times and score 2 was given 2 times. Conclusion: The relationship between dynamic and static motor behavior of the quadriceps supports the connection between the stabilizing and movement function of muscle slings. The low intensity corresponds with the slow EMG activity in DS, which is an indicator for the activation of mainly slow twitch muscle fibers. This scientific innovation is relevant for practitioners as it establishes the diagnostic value of the FMS. Based on this evidence, the FMS provides information with regards to low tension motor behavior. It is unlikely that this information is specific enough to estimate injury risk or performance because when it comes to return to sport decisions, it is also important to obtain information of motor control associated with fast-twitch fibers.

AbstractDynamic stability is an essential physical component for team sport athletes. Certain Functional Movement Screen (FMS) exercises (deep squat; left- and right-leg hurdle step; left- and right-leg in-line lunge [ILL]; left- and right-leg active straight-leg raise; and trunk stability push-up [TSPU]) have been suggested as providing an indication of dynamic stability. No research has investigated relationships between these screens and an established test of dynamic stability such as the modified Star Excursion Balance Test (mSEBT), which measures lower-limb reach distance in posteromedial, medial, and anteromedial directions, in team sport athletes. Forty-one male and female team sport athletes completed the screens and the mSEBT. Participants were split into high-, intermediate-, and low-performing groups according to the mean of the excursions when both the left and right legs were used for the mSEBT stance. Any between-group differences in the screens and mSEBT were determined via a one-way analysis of variance with Bonferroni post hoc adjustment (p < 0.05). Data was pooled for a correlation analysis (p < 0.05). There were no between-group differences in any of the screens, and only two positive correlations between the screens and the mSEBT (TSPU and right stance leg posteromedial excursion, r = 0.37; left-leg ILL and left stance leg posteromedial excursion, r = 0.46). The mSEBT clearly indicated participants with different dynamic stability capabilities. In contrast to the mSEBT, the selected FMS exercises investigated in this study have a limited capacity to identify dynamic stability in team sport athletes.

Sport-specific functional tests were used to assess the power, speed, and agility of the lower extremity for a specific sport, but comparison of the differences and association with sport injury was rare. The aim of this study was to investigate the differences in sport-specific functional tests between junior basketball and soccer athletes and analyze the sport injury risk and occurrences. Methods. All participants were evaluated using the sprint test, vertical jump (VJ) test, agility T test, and functional movement screen (FMS). There were significant intergroup differences in the sprint test, VJ test, agility T test, and FMS. Specific functional tests were compared against FMS score, either FMS ≤ 14 or FMS > 14 . The FMS subtests, namely, in-line lunge, trunk stability push-up (TSPU), and quadruped rotary stability, were also performed. In one-year follow-up, the sport injury incidence was also recorded. Results. Significant differences in sprint, agility, and FMS performance were found between the junior basketball and soccer athletes. Individual FMS scores of the in-line lunge, TSPU, and quadruped rotary stability were evaluated. No significant differences in sprint, VJ, and agility scores were found between FMS ≤ 14 and FMS > 14 . FMS total score ≤ 14 was significantly associated with high sport injury occurrence. Conclusions. The scores of sprint, agility, and FMS performance were differed between basketball and soccer athletes. The scores of sprint, VJ, and agility tests did not have differences with sport injury risks and occurrences, but the FMS score was associated with sport injury occurrence.

This paper investigates system performance in the Internet of Things (IoT) with an energy harvesting (EH) unmanned aerial vehicle (UAV)-enabled relay under Nakagami-m fading, where the time switching (TS) and adaptive power splitting (APS) protocols are applied for the UAV. Our proposed system model consists of a base station (BS), two IoT device (ID) clusters (i.e., a far cluster and a near cluster), and a multiantenna UAV-enabled relay (UR). We adopt a UR-aided TS and APS (U-TSAPS) protocol, in which the UR can dynamically optimize the respective power splitting ratio (PSR) according to the channel conditions. To improve the throughput, the nonorthogonal multiple access (NOMA) technique is applied in the transmission of both hops (i.e., from the BS to the UR and from the UR to the ID clusters). The U-TSAPS protocol is divided into two phases. In the first phase, the BS transmits a signal to the UR. The UR then splits the received signal into two streams for information processing and EH using the APS scheme. In the second phase, the selected antenna of the UR forwards the received signal to the best far ID (BFID) in the far cluster and the best near ID (BNID) in the near cluster using the decode-and-forward (DF) or amplify-and-forward (AF) NOMA scheme. We derive closed-form expressions for the outage probabilities (OPs) at the BFID and BNID with the APS ratio under imperfect channel state information (ICSI) to evaluate the system performance. Based on these derivations, the throughputs of the considered system are also evaluated. Moreover, we propose an algorithm for determining the nearly optimal EH time for the system to minimize the OP. In addition, Monte Carlo simulation results are presented to confirm the accuracy of our analysis based on simulations of the system performance under various system parameters, such as the EH time, the height and position of the UR, the number of UR antennas, and the number of IDs in each cluster.

Recommended drying treatments may not enhance destruction of pathogens that could be present on home-dried foods. In this study, the effects of traditional and modified treatments on Salmonella were evaluated during preparation, home-type dehydration (60°C for 6 h), and storage of potato slices. Potato slices inoculated with five strains of Salmonella (8.4 log CFU/g) were left untreated or were treated by steam blanching (88°C for 10 min), water blanching (88°C for 4 min), 0.105% citric acid blanching (88°C for 4 min), or 0.210% citric acid blanching (88°C for 4 min). Slices were then dried (6 h for 60°C) and aerobically stored for up to 30 days at 25 ± 3°C. Cells were enumerated on tryptic soy agar with 0.1% pyruvate (TSAP) and on xylose lysine deoxycholate agar. Salmonella populations were reduced by 4.5 to 4.8 CFU/g and by &gt;5.4 log CFU/g immediately following steam and water blanching, respectively. Populations were below the detection limit (0.80 log CFU/g) immediately following acid blanching, except for samples blanched in 0.105% citric acid and recovered on TSAP. After dehydration (6 h for 60°C), Salmonella reductions on blanched potato slices (5.3 to 5.6 log CFU/g) were significantly greater (P &lt; 0.05) than those on untreated samples (1.9 to 2.7 log CFU/g). Populations on all samples continued to decrease throughout 30 days of storage but still were 3.1 to 3.9 log CFU/g on untreated samples. In comparison, bacterial populations on blanched samples were undetectable by direct plating following 30 days of storage (regardless of blanching method). Blanching treatments used in this study improved the effectiveness of drying for inactivating Salmonella inoculated onto potato slices and, therefore, may enhance the safety of the product.

This study investigated the learning approach of tenth-grade students in SMK Negeri 1 Singaraja in the EFL online learning context. The study employed quantitative research. The sample involves 89 students in tenth grade in the Hotel Accommodation program. This study used the R-SPQ-2F questionnaire which was modified for the online learning context. The data analysis was implemented in the R-SPQ-2F questionnaire mean score analysis. The results showed that tenth-grade students in SMK Negeri 1 Singaraja tend to surface approach (tsap= 29.55 > tdap= 28.32). It is suggested students will study in-depth to build meaningful learning by understanding concepts and developing potential optimally in the implication of learning English as a foreign language in the online learning context.
