Abstract:
This study aims to explore the implementation of attitude measurement tool known as Test of Science-Related Attitudes (TOSRA) in Sindh. 1097 higher-secondary level students from Hyderabad division of Sindh participated in the study. The alpha reliability values of different scale of TOSRA were analysed and enhanced to acceptable and good level using statistical techniques. The alpha coefficients for adapted TOSRA scales were observed as; Social Implications of Science (0.707), Adoption of Scientific Attitudes (0.801), Classroom Enjoyment (0.779), Leisure Interest in Science (0.767) and Career Interest in Science (0.701). The overall alpha coefficient for adopted (five-scale) TOSRA questionnaire administered in this study was 0.912. Similarly, the values of discriminant validity of the scales were deduced using correlation techniques for these scales where mean correlation between the scales ranged from 0.49 to 0.57. The adopted version of TOSRA was developed based on the statistical analyses including Cronbach alpha, Pearson’s correlation, and factor analysis, and showed sufficient validity and reliability to be implemented in subsequent research to measure students’ attitude towards science in province of Sindh, Pakistan. Abstract:
In the present study, the effects of STEM based robotics applications on students' creativity and scientific attitudes in the Electricity Unit of 7th grade have been investigated by using the nested pattern of the mixed method. Sixty students, 30 of whom are the experimental group and the other 30 constitute the control group, attending a post-school course in Istanbul in the academic year of 2018-2019, participated 2 weeks of pre-applications and 4 weeks of applications. TOSRA,to measure attitude towards science, and “Torrance Creative Thinking Test”,to measure creativity, were applied as pre and posttest. The data gained from the tests were analyzed with SPSS 21. Semi-structured interviews' data were analyzed by using content analysis. As a result, it was observed that STEM based robotics applications significantly increased students' creativity and attitudes towards science. Interview findings show that students enjoy using STEM applications that contain applications instead of theoretical knowledge. Using robotic and complex software materials to solve daily life problems, they felt like scientists during the practices and the applications affected their future career choices. Keywords STEM; creativity; robotic application; attitude; TOSRA

Abstract:
This study was intended to investigate students’ cognitive and attitude of secondary school in learning global warming using Student Team Achievement Division (STAD) based on gender. This study was conducted at one of bilingual boarding school in Bandung. This study uses the quasi-experimental method with matching pretest-posttest comparison group design while the number of subject in the girl class is 15 (N=15) and in the boy class is 15 (N=15). Data was gained through a cognitive test which measured by Bloom's taxonomy cognitive level (C1-C6), a questionnaire which measured by Test of Science Related Attitude (TOSRA). The result shows that student who learns global warming using STAD has an improvement on their cognitive score, especially in the girl class. Meanwhile, girl class has higher students’ attitude result in three scales of TOSRA that are; attitude to scientific inquiry, adoption of a scientific attitude, and enjoyment of science lessons. Along with the students’ cognitive result, girl class also has a better science-related attitude than the boy class. Thus, it can be concluded that there is a significant difference between students’ cognitive and attitude at the girl and boy class in learning global warming using STAD.

Abstract:
Decisions about instruction, research, or policy often require the interpretation of student assessment scores. Increasingly, attitudinal variables are included in an assessment strategy, and it is important to ensure that interpretations of students' attitudinal status are based on instrument scores that apply similarly for diverse students. In this study, a shortened version of the Test of Science Related Attitudes (TOSRA) was used to gather validity evidence based on the internal structure of the instrument in an introductory chemistry course. Using measurement invariance analysis by sex and race/ethnicity, it was found that the internal structure holds by sex, but it did not hold by race/ethnicity in our sample. Further analysis revealed problems with the normality scales for Black students in our sample. Also, this study examined the relationship between the scales of the Shortened TOSRA, achievement in chemistry, and prior math knowledge. Using Structural Equation Modeling (SEM) it was found that two of the scales, attitude toward inquiry and career interest in science, have a small but significant influence on students' achievement in chemistry. This study highlights the importance of examining if the scores apply similarly for different groups of students in a population, since the scores of these assessments could be used to make decisions that will affect the students.

Abstract:
In order to investigate gender and ethnicity-based differences in scientific attitudes among Fijian students, the widely studied Test of Scientific Related Attitudes (TOSRA) was administered to 1401 senior secondary Fijian students (Years 11–13; approximately 15–18 years of age). Students generally had a positive attitude towards science overall in Years 11–13, with females showing a more positive attitude than males. By Year 13, the attitudes of females towards science had become more negative. The attitude of iTaukei students towards science started out lower than other ethnicities in Year 11 and increased during Year 12, before falling to below the starting attitude levels in Year 13. Fijian students of Indian descent generally had a positive attitude towards science that remained consistent throughout Years 11–13, with an increase in leisure and career interest in science in Year 13. A strong correlation was found between the ethnicity of a student and their first language. Continued science outreach programmes, particularly in Year 12, are important to achieve and retain scientific interest and attitudes among Fijian secondary students.

<p style="text-align: justify;">It should be noted that learning outcomes are not students’ only achievement; attitude is also the main output in learning. This research explores students’ attitudes toward science learning based on gender and the grade level of schools in Aceh, Indonesia. The participants are 1,023 students from the sixth grade of primary schools and the eighth grade of secondary schools. The total sample includes 16 schools spread across the province. The data have been collected using TOSRA. By using the Likert scale, this questionnaire is useful for obtaining descriptions of the students’ attitudes and assigning scores for a certain group of participants. Based on gender, the results show females reflect more positive attitudes toward science than male students do. According to the grade level of the schools, the data reflect the equality of students’ attitudes toward science between primary and secondary schools. Nevertheless, when primary school students enter secondary school, the majority of students enjoy learning science less. This fact is meaningful feedback for science teachers. This result supports the scholars seeking ways to avoid the gender gap in learning activities. Pedagogical implications are also discussed.</p>

Abstract:
This study proposes to assess the relationship between student perceptions on teacher interpersonal behavior and student attitude toward Science in Indonesian Primary school. This research engaged 143 students from SDN 47 Jambi, Indonesia. To gather the data, two types of questionnaires were used; Test of Science Related Attitude (TOSRA) and the Indonesian version of the Questionnaire Teacher Interpersonal (QTI). SPSS program were applied to process the data statistically. The result shows that the questionnaires are relatively reliable for Indonesian primary students. Acceptability of Cronbach alpha coefficient was found in both actual and preferred situation. The findings also reveal that from the students’ view, their teachers have relatively good Leadership and Understanding in the classroom, less Uncertainty, Dissatisfaction and Admonishment toward students. However, the students perceive to some extent unfavorable perceptions on their Freedom and teacher Strictness. Another analysis describes that there are significant differences between actual and ideal perceptions on the whole scale of QTI except on Uncertainty. Furthermore, between male and female perceptions, this study finds that there are significant differences on Helping, and Strictness. Lastly, the multiple correlation analysis explaines that there is a correlation between the scale of QTI and Enjoyment toward science, in which Uncertainty and Admonishment scales are significantly negative affect students’ enjoyment in science classroom.

Abstract:
ABSTRACTA heterogeneous subset of extraintestinal pathogenicEscherichia coli(ExPEC) strains, referred to as uropathogenicE. coli(UPEC), causes most uncomplicated urinary tract infections. However, no core set of virulence factors exists among UPEC strains. Instead, the focus of the analysis of urovirulence has shifted to studying broad classes of virulence factors and the interactions between them. For example, the RTX nonfimbrial adhesin TosA mediates adherence to host cells derived from the upper urinary tract. The associatedtosoperon is well expressedin vivobut poorly expressedin vitroand encodes TosCBD, a predicted type 1 secretion system. TosR and TosEF are PapB and LuxR family transcription factors, respectively; however, no role has been assigned to these potential regulators. Thus, the focus of this study was to determine how TosR and TosEF regulatetosAand affect the reciprocal expression of adhesins and flagella. Among a collection of sequenced UPEC strains, 32% (101/317) were found to encode TosA, and nearly all strains (91% [92/101]) simultaneously carried the putative regulatory genes. Deletion oftosRalleviatestosArepression. Thetospromoter was localized upstream oftosRusing transcriptional fusions of putative promoter regions withlacZ. TosR binds to this region, affecting a gel shift. A 100-bp fragment 220 to 319 bp upstream oftosRinhibits binding, suggesting localization of the TosR binding site. TosEF, on the other hand, downmodulate motility when overexpressed by preventing the expression offliC, encoding flagellin. Deletion oftosEFincreased motility. Thus, we present an additional example of the reciprocal control of adherence and motility.

This study aims to investigate the student perception on teacher interpersonal behavior and their attitude toward science subject. 207 respondents were involved, consisted of 200 students in 10-12 grade and 7 science teachers of public high school in Aceh. Two types of questionnaires were used namely the Indonesian version of the questionnaire of teacher interaction (QTI) and test of science related attitude (TOSRA). SPSS program were applied to process the data statistically. First, the reliability of questionnaires is measured using descriptive statistics of all and each scale of QTI. Second, the difference between students’ perceptions on the actual and ideal teacher interpersonal behavior was computed by Paired Sample t-tests. Third, the relationship between students' perception on teachers’ interpersonal behavior and students' attitudes towards science subjects was compared using multiple regression analysis-standardized regression coefficient β. The finding showed that the Indonesian version of questionnaire of teacher interaction (QTI) is reliable (αC = 0.86), to be applied to the high school students in the regency. It is generally seen that the students consider their teachers demonstrate more positive interpersonal behavior than the negative. However, it is clearly seen that between actual and ideal perceptions on the whole scale of QTI is significantly different. Last, from the three scales of teacher interpersonal behavior, helping/friendly, dissatisfaction, and admonishing appeared influential, only the helping/friendly scale significantly correlate (at the .05 level) with student attitudes toward science subject.

Abstract:
Urinary tract infections (UTIs) are a major burden to human health. The overwhelming majority of UTIs are caused by uropathogenicEscherichia coli(UPEC) strains. Unlike some pathogens, UPEC strains do not have a fixed core set of virulence and fitness factors but do have a variety of adhesins and regulatory pathways. One such UPEC adhesin is the nonfimbrial adhesin TosA, which mediates adherence to the epithelium of the upper urinary tract. Thetosoperon is AT rich, resides on pathogenicity islandaspV, and is not expressed under laboratory conditions. Because of this, we hypothesized thattosAexpression is silenced by H-NS. Lrp, based on its prominent function in the regulation of other adhesins, is also hypothesized to contribute totosoperon regulation. Using a variety ofin vitrotechniques, we mapped both thetosoperon promoter and TosR binding sites. We have now identified TosR as a dual regulator of thetosoperon, which could control thetosoperon in association with H-NS and Lrp. H-NS is a negative regulator of thetosoperon, and Lrp positively regulates thetosoperon. Exogenous leucine also inhibits Lrp-mediatedtosoperon positive regulation. In addition, TosR binds to thepapoperon, which encodes another important UPEC adhesin, P fimbria. Induction of TosR synthesis reduces production of P fimbria. These studies advance our knowledge of regulation of adhesin expression associated with uropathogen colonization of a host.

Abstract:
It is important to remember that learning outcomes are not the only thing students accomplish, but attitude is also a major learning output. The study purpose was to investigate students’ attitudes, especially regarding the attitude toward nonlinear pedagogy (ATNP), according to gender and grade level in Yogyakarta. Materials and methods. The study involved 1,160 kids who were in fifth grade in primary schools and in eighth grade in secondary schools. A total of 18 schools from around the province were included in the study. The Test of Science Related Attitudes (TOSRA) was used to gather data. This questionnaire is effective for gathering descriptions of students’ attitudes toward nonlinear pedagogy scores for a specific set of participants, since it uses the Likert scale. Results. The study findings show students have quite different levels of excitement for nonlinear science learning. The male students in primary school have a higher positive attitude than the female students do, (M = 36.75; p < 0.001). The male students exhibit a more eager attitude than the female students do in junior high schools (M = 35.78; p < 0.001). The data revealed that the male students in secondary schools have more positive attitudes than the female students do (8.23, M>F). According to the grade level of the school, the male students’ ATNP at the elementary level is more positive than in junior high schools. Conclusions. The statistics show that students’ opinions toward nonlinear teaching are similar in elementary and secondary schools regardless of the grade level of the schools. Even still, most secondary school students find physical education less enjoyable than it was in elementary school. This fact provides instructors of physical education with user input. This finding is encouraging for students looking for strategies to bridge the gender gap in classroom settings. The effects of nonlinear pedagogy are also explored.

Abstract:

Abstract:
Item analyses and confirmatory factor analyses on the Test of Self‐Conscious Affect (TOSCA), in a student (N = 723) and an adult (N = 891) sample, supported the theorized four factor structure of proneness to reparation, negative self‐evaluation, externalizing blame and unconcern. However, two‐fifth of the items did not empirically differentiate between two or more factors. Differential TOSCA scales, including only differentiating TOSCA items, were constructed and related to measures of long‐term affect, depression, anxiety, and anger. Both the pattern and size of correlations of the original and the differential TOSCA scales were almost identical. Results of this study support the interpretation of TOSCA guilt as a measure of a tendency to reparation associated with guilt and TOSCA shame as a measure of a tendency to global negative self‐evaluation. Abstract:
DYT1 dystonia is caused by an in-frame deletion of a glutamic acid codon in the gene encoding the AAA+ ATPase TorsinA (TorA). TorA localizes within the lumen of the nuclear envelope/endoplasmic reticulum and binds to a membrane-spanning cofactor, lamina associated polypeptide 1 (LAP1) or lumenal domain like LAP1 (LULL1), to form an ATPase; the substrate(s) of TorA remains ill-defined. Here we use budding yeast, which lack Torsins, to interrogate TorA function. We show that TorA accumulates at nuclear envelope-embedded spindle pole bodies (SPBs) in a way that requires its oligomerization and the SUN (Sad1 and UNc-84)-domain protein, Mps3. We further show that TorA physically interacts with human SUN1/2 within this system, supporting the physiological relevance of these interactions. Consistent with the idea that TorA acts on a SPB substrate, its binding to SPBs is modulated by the ATPase-stimulating activity of LAP1. TorA and TorA-ΔE reduce the fitness of cells expressing mps3 alleles, whereas TorA alone inhibits growth of cells lacking Pom152, a component of the nuclear pore complex. This genetic specificity is mirrored biochemically as TorA, but not TorA-ΔE, binds Pom152. Thus, TorA–nucleoporin interactions might be abrogated by TorA-ΔE, suggesting new experimental avenues to interrogate the molecular basis behind nuclear envelope herniations seen in mammalian cells lacking TorA function.

Abstract:
The experiment was conducted at Hill Agricultural Research Station, Khagrachari (representing hilly areas), Agricultural Research Station, Satkhira and Jute Research Station, Patuakhali (representing saline areas) with seven varieties of jute and one variety of kenaf to find out the seed production potentiality in non-traditional areas. All varieties were sown in late July to mid August in 2009 to 2011. At hilly station white jute varieties (BJRI Deshi Pat-6, CVL-1 and BJRI Deshi Pat-5) performed better for seed production than tossa jute and kenaf varieties. At saline station (both Satkhira and Patuakhali) kenaf variety HC-95 performed better for seed yield than white and tossa jute varieties (O-9897, OM-1, BJRI Tossa Pat-4 and BJRI Tossa Pat-5). The kenaf variety (HC-95) produced the highest yield (0.86 ton/ha) in saline areas. Among the white jute varieties, BJRI Deshi Pat-5 gave the highest yield (0.66 ton/ha) and in case of tossa jute varieties, BJRI Tossa Pat-5 gave the highest seed yield (0.70 ton/ha). Therefore, BJRI Deshi Pat-5 of white jute, BJRI Tossa Pat-5 and HC-95 of kenaf are considered as the best varieties for seed production in hilly and saline areas of Bangladesh, respectively.

Abstract:
The purpose of this study was to observe the germination rate and fungal pathogens association in selected Tossa jute varieties. The experiment was conducted following Completely Randomized Design (CRD) with four replications. Truthfully labelled seeds (TLS) collected from four districts in Bangladesh, was used in the present study. The Highest germination rate (93.5%) and the lowest infection (31.25 pathogens/100 seed) were observed in BJRI Tossa Pat 5 (O-795) variety. BJRI Tossa Pat 4 (O-72) was observed as the most vulnerable against pathogen infection (121.25 pathogens/100 seed) and the showed lowest germination (83.25%). Regressions co-efficient (β) -0.82 was observed between germination (%) and total number of pathogens on seeds. Regression co-efficient (β) was -0.71 between moisture (%) and germination (%) of Tossa seeds. Regression co-efficient (β) was 0.65 between moisture (%) and total number of pathogens at Tossa jute seeds. For conducting the experiment, there was no limitation. From the study, it may be concluded that, seed-borne pathogenic fungi negatively affect the germination (%) of Tossa jute seed. SAARC J. Agric., 20(1): 67-76 (2022)

Abstract:
Tomato severe rugose virus (ToSRV) is the most important begomovirus species in Brazilian tomato production. Many weeds are associated with tomato, and some are hosts of begomoviruses. Only one species of weed, Nicandra physaloides, has been found to be infected with ToSRV. In this study, four weed species were investigated for their capacity to be infected by ToSRV and serve as a potential source of inoculum for tomato. Begomoviruses from naturally infected Crotalaria spp., Euphorbia heterophylla, N. physaloides, and Sida spp. were successfully transferred to tomato plants by biolistic inoculation. ToSRV was the major virus transferred to tomato. In contrast, other begomoviruses were transferred to weeds, such as Sida micrantha mosaic virus and Euphorbia yellow mosaic virus. Furthermore, a new strain of Sida micrantha mosaic virus is reported. We also confirmed that Crotalaria spp., E. heterophylla, and Sida spp. are infected with ToSRV but at low viral titers and in mixed infections with weed-infecting begomoviruses. Thus, it was demonstrated that weeds are potential sources of ToSRV for tomato in central Brazil.

Abstract:
TorsinA (TorA) is an AAA+ ATPase in the endoplasmic reticulum (ER) lumen that is mutated in early onset DYT1 dystonia. TorA is an essential protein in mice and is thought to function in the nuclear envelope (NE) despite localizing throughout the ER. Here, we report that transient interaction of TorA with the ER membrane protein LULL1 targets TorA to the NE. FRAP and Blue Native PAGE indicate that TorA is a stable, slowly diffusing oligomer in either the absence or presence of LULL1. Increasing LULL1 expression redistributes both wild-type and disease-mutant TorA to the NE, while decreasing LULL1 with shRNAs eliminates intrinsic enrichment of disease-mutant TorA in the NE. When concentrated in the NE, TorA displaces the nuclear membrane proteins Sun2, nesprin-2G, and nesprin-3 while leaving nuclear pores and Sun1 unchanged. Wild-type TorA also induces changes in NE membrane structure. Because SUN proteins interact with nesprins to connect nucleus and cytoskeleton, these effects suggest a new role for TorA in modulating complexes that traverse the NE. Importantly, once concentrated in the NE, disease-mutant TorA displaces Sun2 with reduced efficiency and does not change NE membrane structure. Together, our data suggest that LULL1 regulates the distribution and activity of TorA within the ER and NE lumen and reveal functional defects in the mutant protein responsible for DYT1 dystonia.

Abstract:
TorsinA (TorA) is an AAA+ (ATPases associated with diverse cellular activities) ATPase linked to dystonia type 1 (DYT1), a neurological disorder that leads to uncontrollable muscular movements. Although DYT1 is linked to a 3 bp deletion in the C-terminus of TorA, the biological function of TorA remains to be established. Here, we use the yeast Saccharomyces cerevisiae as a tractable in vivo model to explore TorA function. We demonstrate that TorA can protect yeast cells against different forms of environmental stress and show that in the absence of the molecular disaggregase Hsp104, TorA can refold heat-denatured luciferase in vivo in an ATP-dependent manner. However, this activity requires TorA to be translocated to the cytoplasm from the endoplasmic reticulum in order to access and process cytoplasmic protein aggregates. Furthermore, mutational or chemical inactivation of the ATPase activity of TorA blocks this activity. We also find that TorA can inhibit the propagation of certain conformational variants of [PSI+], the aggregated prion form of the endogenous Sup35 protein. Finally, we show that while cellular localisation remains unchanged in the dystonia-linked TorA mutant ΔE302-303, the ability of this mutant form of TorA to protect against cellular stress and to facilitate protein refolding is impaired, consistent with it being a loss-of-function mutation.

Abstract:
We have identified a novel gene, Tortoise (TorA), that is required for the efficient chemotaxis of Dictyostelium discoideum cells. Cells lacking TorA sense chemoattractant gradients as indicated by the presence of periodic waves of cell shape changes and the localized translocation of cytosolic PH domains to the membrane. However, they are unable to migrate directionally up spatial gradients of cAMP. Cells lacking Mek1 display a similar phenotype. Overexpression of Mek1 in torA− partially restores chemotaxis, whereas overexpression of TorA in mek1− does not rescue the chemotactic phenotype. Regardless of the genetic background, TorA overexpressing cells stop growing when separated from a substrate. Surprisingly, TorA–green fluorescent protein (GFP) is clustered near one end of mitochondria. Deletion analysis of the TorA protein reveals distinct regions for chemotactic function, mitochondrial localization, and the formation of clusters. TorA is associated with a round structure within the mitochondrion that shows enhanced staining with the mitochondrial dye Mitotracker. Cells overexpressing TorA contain many more of these structures than do wild-type cells. These TorA-containing structures resist extraction with Triton X-100, which dissolves the mitochondria. The characterization of TorA demonstrates an unexpected link between mitochondrial function, the chemotactic response, and the capacity to grow in suspension.

Abstract:
This article studies how the word Torah is used in the Pentateuch to designate a collection of laws. In Deuteronomy – originally an independent composition – “Torah” is used for the first time as a designation of a substantial collection of laws. When Deuteronomy was integrated into the Pentateuch this deuteronomic signification of “Torah” changed: According to Exod 24:12, God gives on Mt. Sinai to Moses. Based on the key terms and , the adressees of the Pentateuch could only understand this Sinai-Torah as refering to Exod 25:1–31:17; Lev 1–27; Deut 5:1b–26:16; the blessings and curses of Deut 28; and the song of Deut 32. The Torah containing these passages forms the basis of the covenant in Moab (Deuteronomy 29–30) and is written down by Moses according to Deut 31:9, 24. This “Moabite” Torah Book is placed in the Pentateuch next to the “Book of the Covenant” which is written down by Moses as well (Exod 24:7) and which provides the basis for the covenant formed on Mt. Sinai.

Abstract:
