This paper has analyzed the morphophysiological and biochemical responses in sugarcane plants (Saccharum spp) and cactus pear (Opuntia stricta Haw) in function of exposition to various kinds and concentrations of cytokinins during these species’ micropropagations. The cytokinins are growth regulators which are largely used in the multiplication phase to induce breaking of the apical dominance, stimulating cell division and budding of lateral buds. The 6-benzylaminopurine (BAP) is the most used cytokinin in the micropropagation industry due to its effectiveness and accessibility; however it may incur abnormal growth in some plants. A kind of aromatic, non-conventional cytokinin, the meta-topolin (mT), has been an alternative to BAP for the purpose of reducing occurrence of morphophysiological disorders. Morphophysiological anomalies like fasciation in sugarcane and hyperhydricity in cactus pear, are problems caused by excessive exposure to BAP. The work was done in the Laboratório de Cultura de Tecidos Vegetais/UFRPE with collaboration of the Biofábrica Governador Miguel Arraes/CETENE. Sugarcane plants of the RB98710 variety were grown in static liquid system in various cytokinin concentrations (6,66 μmol L-1 of BAP; 5 μmol L-1 of BAP or mT). Buddings of cactus pear from variety Orelha de Elefante Mexicana, growth in vitro were used in the experiment. The treatments consisted in two kinds of cytokinins, BAP and mT in concentrations of 2,22 μmol L-1 of BAP, 1,11 μmol L-1 of BAP or 1,11 μmol L-1 of mT in static system in liquid or solid medium. Analysis were realized for multiplication ratio, fresh matter, height, analysis for enzymatic activity of superoxide dismutase (SOD), ascorbate peroxidase (APX) and catalase (CAT), as well as quantification in hydrogen peroxidase (H2O2) and malondialdehyde (MDA). In sugarcane it was observed that the mT promoted the higher rate on multiplication and higher height of plants and tillers. The role of hydrogen peroxyde in plant growth and development was observed. The plants submitted to higher BAP concentration displayed a higher MDA content. In the cactus pear growth it was observed that high concentrations of BAP favored the reduction of fresh matter, as well as buddings’ height. Formation of buds in liquid medium happened, in low amounts compared to solid medium. The hyperhydricity characteristics were more intense in buds grown with BAP. The hyperhydric buddings displayed a purple coloring, primarily in the podaries. From this paper it was verified that the sugarcane variety RB98710 responded positively to mT addition. The use of mT in cactus pear micropropagation in MS solid medium leads to promising results for bud production.
A presente pesquisa visou avaliar respostas morfofisiológicas e bioquímicas em plantas de cana-de-açúcar (Saccharum spp) e em plantas de palma forrageira (Opuntia stricta Haw) em função da exposição a diferentes tipos e concentrações de citocininas durante a micropropagação dessas espécies. As citocininas são reguladores de crescimento que são amplamente empregadas na fase de multiplicação por induzir a quebra da dominância apical, estimulando a divisão celular e a brotação de gemas laterais. A 6-benzilaminopurina (BAP) é a citocinina mais utilizada na indústria de micropropagação devido à sua efetividade e acessibilidade, porém pode acarretar crescimento anormal em algumas plantas. Um tipo de citocinina aromática não convencional, a meta-topolina (mT), tem sido alternativa ao uso do BAP a fim de reduzir o aparecimento de desordens morfofisiológicas. Anomalias morfofisiológicas como fasciação em cana-de-açúcar e hiperidricidade, em palma forrageira, são problemas atribuídos ao efeito do excesso ou tempo de exposição prolongado ao BAP. O trabalho foi desenvolvido no Laboratório de Cultura de Tecidos Vegetais/UFRPE com colaboração da Biofábrica Governador Miguel Arraes/CETENE. Plantas de cana-de-açúcar variedade RB98710 foram cultivadas em sistema estático líquido em diferentes concentrações de citocininas (6,66 μmol L-1 de BAP; 5 μmol L-1 de BAP ou mT). Brotações de palma forrageira, cultivar Orelha de Elefante Mexicana cultivadas in vitro foram utilizadas na obtenção de unidades propagativas. Os tratamentos consistiram em dois tipos de citocininas, BAP e mT nas concentrações de 2,22 μmol L-1 de BAP, 1,11 μmol L-1 de BAP ou 1,11 μmol L-1 de mT em sistema estático em meio líquido ou sólido. Foram realizadas avaliações da taxa de multiplicação, matéria fresca, altura, avaliação da atividade enzimática da superóxido dismutase (SOD), ascorbato peroxidase (APX) e catalase (CAT), como também a quantificação dos níveis de peróxido de hidrogênio (H2O2) e malondialdeído (MDA). Em cana-de-açúcar foi observado que mT promoveu maior taxa de multiplicação e maior altura de plantas e perfilhos. Foi observado o papel do peróxido de hidrogênio no crescimento e desenvolvimento vegetal. As plantas submetidas a maior concentração de BAP apresentaram maior teor de MDA. No cultivo da palma forrageira foi constatado que altas concentrações de BAP favoreceu para a redução da matéria fresca, bem como da altura das brotações. Houve formação de brotos em meio líquido, mesmo que em número reduzido quando comparado com o meio sólido. As características de hiperidricidade foram mais intensas em brotos provenientes do cultivo com BAP. As brotações hiperídricas apresentaram uma coloração púrpura, principalmente nas podarias. A apartir desse estudo foi constatado que a variedade de cana-de-açúcar RB98710 respondeu de forma favorável à adição de mT. O uso de mT na micropropagação da palma forrageira em meio MS sólido apontam resultados promissores para a produção de mudas em larga.escala.
