Human embryonic stem cells (hESCs) are known by their ability to either self-renewal and differentiate into any adult cell type. These properties confer to hESCs a huge applicability for cell therapy, tissue engineering and drug screening. However, successful implementation of hESCs-based technologies requires the production of large numbers of well characterized cells and their efficient long-term storage. In this study, alginate microencapsulation technology was used in order to develop an efficient, scalable and integrated 3D culture system for expansion and cryopreservation of pluripotent hESCs. Three strategies were outlined: microencapsulation of hESCs as single cells, cell aggregates and cells immobilized on microcarriers. Encapsulation of hESCs immobilized on microcarriers was the best strategy to expand and cryopreserve pluripotent hESCs. The culture of encapsulated hESCs-microcarriers in spinner vessels assured an approximately 20-fold increase in cell concentration. Moreover, this strategy improved twice cell survival after cryopreservation by a slow-freezing rate procedure, comparatively with non-encapsulated culture. Microencapsulation also protected hESC aggregates from damage caused by stirring, allowed the control of aggregates size and the maintenance of cells pluripotency for two weeks. This work demonstrates that microencapsulation technology is a powerful tool to enhance growth and post-thawing recovery of pluripotent hESCs. The 3D culture systems developed herein represent a promising vehicle to assist the transition of hESCs to the clinical and industrial fields.

Simulation has overcome critical obstacles to become a valuable method for obtaining insights about the behavior of complex systems. George Boxs famous assessment that all models are wrong, some are useful referred to statistical models, but should now be reimagined to reflect that many simulation models are right enough to aid in decision making for important practical problems. Over the past fifty years, simulation has transformed from its beginnings as a brute-force numerical integration method into an attractive and sophisticated option for decision makers. This is due, in part, to the exponential growth of computing power. Although other analytic approaches also benefit from this trend, keyword searches of several scholarly search engines reveal that the reliance on simulation is increasing more rapidly. A descriptive analysis paints a compelling picture simulation is frequently a researchers preferred method for supporting decision makers and may often be the first resort for complex real world issues.

Recent crisis and economic recession caused in many countries problems with drop in inflation and overall downturn in economic growth. The Central Banks found themselves in so called zero lower bound. This thesis describes the situation of deflation, zero lower bound and liquidity trap. These conditions called for the use of alternative monetary policy tools. The main goal of the thesis is to evaluate the use of foreign exchange rate intervention as unconventional tool in a small open economy with inflation targeting regime represented by the Czech Republic in the analytic part. Thesis examines other non-standard instruments and provides reasons for the Czech National Bank decision. The analysis shows the complexity of the issue and provides the evidence that exchange rate commitment was not displayed to the desired price level. The tool could lead to expected repercussion, but the effect is impacted by factors, which Central Bank cannot control. The effect of the CB way to fight the decreasing inflation will be better shown after the CB exit from the commitment.

CoolVent, a software design tool for architects, has been improved. The work of Maria- Alejandra Menchaca-B. and colleagues has been improved to include a more robust and intuitive building and window dimensioning scheme, feedback on the validity of user inputs and thermal comfort modeling (custom, ASHRAE and adaptive). These results now allow the architect to understand how their design choices have not only affected the temperature and airflow in each of the zones of their building design but also how their design choices have affected the overall comfort throughout the zones of their building. From this, architect's can now easily understand whether their building design can be successful as a naturally ventilated building and if not, they can use the provided interface to gain insight into how their design can be modified to make their building more sustainable. A user study has been conducted to test the effectiveness of the tool.

This dissertation examines the scope and limitations of port state jurisdiction with respect to the enforcement of international conservation and management measures established under international and national instruments. The current fisheries regulatory regimes rely primarily on flag states to enforce these measures against foreign vessels. The aim of this dissertation is to recommend an expansion of port state enforcement jurisdiction over foreign vessels when fishing in the high seas. The expansion of port state jurisdiction supplements the role of flag states in enforcing the provisions of international conventions and agreements applicable in the high seas and also fills in where flag of convenience fishing vessels have failed to do so. To support this view, the dissertation will compare the issue of illegal unreported unregulated fishing with the issue of marine pollution. The significance of this comparison is to show how provisions in regard to marine pollution, have legitimized the expansion of port state jurisdiction to the extent that port states have enforcement jurisdiction over foreign flagged vessels which violate the provisions of LOSC relating to marine pollution in the high seas.

AbstractWith its low thermal conductivity and high thermal capacity, polymer concrete has excellent thermal properties that when used in machine tools ensure increased accuracy of the manufactured parts. Polymer concrete has been used successfully in machine tool construction for many years in the form of machine beds. The aforementioned thermal properties in combination with a low density, high damping and a lower primary energy requirement also make polymer concrete interesting for use as a structural component. However, the comparatively low tensile strength and creep tendency of the material pose a challenge here. One approach to increase the tensile strength of the material is the integration of prestressed carbon fibres into the material. In order to clarify the suitability of this hybrid material, its temperature behaviour is investigated in this paper. The focus is on the investigation of residual stresses that arise during heating, which result from the combination of the positive thermal expansion coefficient of polymer concrete with the negative thermal expansion coefficient of the carbon fibres. In addition, the flexural properties of pure polymer concrete and of pre-stressed fibre-reinforced polymer concrete are determined at different test temperatures within the scope of this paper. It is shown that the prestressing of fiber-reinforced polymer concrete leads to an improvement of the flexural strength and the stiffness of polymer concrete.

AbstractThis chapter traces the use of religious governance in England’s early attempts to colonise Virginia between 1606 and 1624. It assesses how, in the initial steps to establish English authority abroad, religious governance was influenced by the political and governmental characters of successive company leaders such as Thomas Dale, Thomas Gates and John Smith. This explains why the Virginia Company embraced multiple forms of religious governance that would later be used as separate and distinct models of governance by successive companies. The Virginia Company experimented with religious governance to secure their control over English personnel abroad. Moreover, it became an instrumental tool in the companies’ attempts to expand their jurisdictional authority over Native American leaders, such as Powhatan, Pocahontas and her uncle Uttamatomakkin. By doing this company leaders hoped to establish governmental control over Native American peoples, and traditions, such as those Smith writes about in Generall Histoirie of Virginia, traditionally considered beyond the bounds of English governance. Finally, it examines how the experiences and memories of religious governance in the Virginia Company provided the groundwork for future forms of corporate religious governance to evolve.

AbstractPersistent organic pollutants (POPs) were identified in humans who have not been dealing with these chemicals intentionally—from organochlorine pesticides towards industrial chemicals, brominated and fluorine containing POPs. This chapter provides a brief overview of major developments in POPs monitoring in human milk and depicts a gradual broadening of the knowledge underpinned by advances in the instrumentation for chemical analysis as well as expansion of range of analytes that warranted attention. The chapter also shows how, in the course of the past 70 years, human milk monitoring has become an efficient and cost-effective non-invasive biomonitoring tool to evaluate the internal human exposure to POPs and the resulting body burden.

AbstractDuring metal machining, a large amount of heat is generated in the cutting zone, which has a negative impact on machining accuracy due to the thermal expansion of the materials. To reduce the temperature in the cutting zone, liquid coolants are used which increase the costs and can have a negative impact on the environment. This problem is being studied using Computational Fluid Dynamics (CFD) to better understand the behavior of the coolant flow in the cutting zone, which will allow optimization of the use of liquid coolants and the development of a correction method for thermal errors, resulting in more accurate machining with reduced resource and environmental footprints. However, due to the complexity of multiphase CFD simulations, the simulation model must be simplified as much as possible. This is particularly important for the process heat generation, as combining flow simulation of coolant flow around the rotating cutting tool with structural simulation of the milling process, including chip formation, would require excessive computational power. In following paper an alternative method of tool heating by electromagnetic induction is presented and the measurement dependencies required to determine the heat flux induced into the cutting tool are described. This can be further applied as a boundary condition for the numerical simulation as a verification method for the coupled Fluid-Structure Interaction FSI simulation model of the thermally induced deformations of the cutting tool and its holder.

AbstractThis paper investigates the thermal modeling challenges of high-speed motorized spindles up to 40,000 rpm. The thermal expansion and deflection of motorized spindles are critical determinants for the resulting tool center point displacement and the achievable machining accuracy of machine tools. In order to compensate, finite element and reduced physical models (digital twins) therefore require an accurate understanding of the thermal boundary conditions. In motorized spindles, heat sources are of great significance to the resulting temperature field. However, it is very difficult to accurately quantify the heat sources in motorized spindles. This is a particular challenge for high-speed applications exceeding 20,000 rpm, where commonly used boundary conditions are not validated. While the power loss of the electric motor could be quantified with reasonable accuracy, the calculation approaches for air and bearing friction proved to be inadequate. This paper introduces approaches to quantify the air and bearing friction of motorized spindles with improved accuracy for applications up to 40,000 rpm. The method was verified based on a coupled thermal/fluid-mechanical spindle simulation model. The mean absolute temperature difference between the model and the test bench was 1.5 K.

Abstract The importance of predicting and reducing thermal expansion errors in workpieces is becoming greater as better precision machining processes are developed. An artificial neural network model to estimate the workpiece thermal expansion errors in real-time during precision machining operations is developed and compared with experimental results. A finite element model of workpiece thermal expansion has been created to predict expansions in a thin cylinder undergoing a turning process. The neural network has been trained using finite element model solutions over a range of conditions to allow for changing machining parameters. To realize “on-line” capability, the measurable values of heat flux into the workpiece, surface heat transfer coefficient, and tool location are used as inputs and the expansion as the output for the neural network. The estimations of the network are compared with experimental results from a turning process on a large diameter aluminum cylinder. There is reasonable agreement between measured and estimated expansions with an average error of 18%. The neural network has not been trained at the cutting conditions used during the experiment. The speed of the neural network estimation is much greater than the solution to the finite element model. The finite element model required over 15 minutes to solve on a Pentium 133Mhz computer. The neural network calculated the expansions easily at 1 Hz during the experiment on the same computer. With real-time estimation using measurable data, compensation can be made in the tool path to correct for these errors. The application of this method to precision machining processes has the capability of greatly reducing the error caused by workpiece thermal expansions.

We present a small analytical framework, built in the Balance of Payments Constrained Growth (BPCG) tradition, tailored to identify how specific financial and structural vulnerabilities of emerging economies (EMEs) restrict their expansion. This framework is applied to shed light on the impact of global shocks and of major macro policy changes in developed countries on the growth path of six economies. As we show, this impact is conditioned by the vulnerabilities and dynamics of their insertion on international trade and capital markets and their exposure to policy changes in the developed centers. Our work shows that the BPCG-approach is a relevant, simple tool to identify EMEs´ challenges in the post-covid 19 era.

The COVID-19 pandemic has placed unprecedented strain on public finances across developing countries. Large fiscal responses and weak economic activity have widened deficits and increased public debt, which for many will be unsustainable over the medium-term. It is important for policymakers to consider policies for mobilising additional revenue. To this end, this policy brief discusses reform options across four types of taxes: wealth, property, consumption, and corporate income. Wealth taxes are underutilised as a policy tool. There is an opportunity to leverage them in response to COVID-19 as they can help reduce inequality and pave the way for equitable recovery. Linked to this option is the expansion of taxes on land and property, targeting the most visible form of wealth in most countries. The brief also outlines policy recommendations on more conventional tools: consumption and corporate taxes, which can be used more effectively to raise revenue by, for example, rationalising tax expenditures and using technology.

As electric mobility is expanding at a rapid pace, the standardized availability of gas stations compared to a scarcity of charging stations continues to be the greatest challenge for electric vehicles. With cities, university campuses and businesses promoting electric vehicle infrastructure and incentives, it is necessary to develop key performance metrics and visualizations that can track the utilization of the charging infrastructure. This study performs a manual data collection at dedicated plug-in charging stations across Purdue University to assess their utilization. Approximately 2,800 observations were conducted over 50 days across seven level 2 plug-in charging stations. Results showed that for large portion of the observations, vehicles were parked at the spots (40%) but not plugged in. Vehicles plugged in to charging stations accounted for 34% of observations. Charging station spots were vacant for 25% of observations indicating that current infrastructure meets the demand. There were 74 unique vehicles that used the spots, of which 27% were plugged in more than 10 times. Illegally parked vehicles accounted for less than 1% with only 4 repeat offenders who used these spots more than once. As electric deployment continues to increase, performance metrics will be an integral tool for agencies and decision makers to help with the maintenance and expansion of electric vehicle infrastructure.

Професійна компетентність фахівців різних галузей стає неможливою без володіння ними міжкультурною компетентністю. Поняття «міжкультурна компетентність» розглядається в роботі як міждисциплінарний феномен та досліджується з позицій філософії, культурології, соціології, психології, педагогіки та лінгводидактики. Особливу увагу зосереджено на зв’язку культури та мови, оскільки мовні знання є інструментом пізнання іншої культури та важливою умовою міжкультурної комунікації. The article raises the problem of importance of intercultural competence forming, in particular among students of philology. The realization of intercultural competence is considered as one of the conditions for establishment of the European standards of language education. Modern globalization processes, expansion of intercultural contacts demand the Ukrainian education to be focused on the new level specialist – the subject of multicultural surrounding. Professional competence of specialists of different fields is impossible without possessing intercultural competence. The concept «intercultural competence» is considered as multidisciplinary phenomenon which is studied from different points of view: philosophical, cultural, sociological, psychological, pedagogical and linguodidactic. The main attention is focused of the connection of culture and language because language competence is the tool for another culture cognition and an important condition of intercultural communication.

Abstract:
Electronic monitoring (EM) has emerged as a popular tool for curbing the growth of large prison populations. Evidence on the causal effects of EM on criminal recidivism is, however, limited and it is unclear how this alternative to incarceration affects the labor supply of offenders and the outcomes of their family members. We study the country wide expansion of EM in Sweden in 1997 where in offenders sentenced to up to three months in prison were granted the option to substitute incarceration with EM. Our difference-in-differences estimates, which compare the change in the prison inflow rate of treated offenders to that of non-treated offenders with slightly longer sentences, show that the reform significantly decreased the number of incarcerations. Our main finding is that EM not only lowers criminal recidivism but also increases labor supply. Additionally, EM improves the educational attainment and early-life earnings of the children whose parents were exposed to the reform. The primary mechanisms through which EM operates appear to involve the preservation of offenders’ ties to the labor market, by reducing the barriers to both finding a job and changing employers. Our calculations suggest that the social benefits stemming from EM are about seven times larger than the fiscal savings associated with reduced prison expenditures, implying that the welfare gains from EM could be much greater than previously acknowledged.
