Full textEn esta tesis se han estudiado y demostrado distintos mecanismos moleculares involucrados en la diferenciación osea. Los microRNAs emergieron hace pocos años como reguladores post-transcripcionales presentes en todos los procesos biológicos. En esta tesis se describe el microRNA-322 como un miRNA reprimido durante el tratamiento con BMP-2 en células osteoblásticas y se detalla cómo éste actúa sobre el extremo 3'-UTR del mRNA de Tob2, produciendo su degradación. TOB2 es una proteína que forma parte de la familia de proteínas antiproliferativas TOB2/BTG. Tob2 se ha descrito como mediador de modificaciones post-traduccionales regulando la deadenilación general de los mRNAs. A demás, se ha visto que participa acelerando específicamente la degradación de determinados mRNAs al unirse a proteínas CPEBlike (CPEB2-4). En la presente tesis se describe el mecanismo por el cual el miR-322 degrada específicamente Tob2 y lo retiene de ejercer su función específica sobre Osterix. El uso de inhibidores específicos y los estudios con ratones mutantes existentes hasta el momento sugerían un papel importante para la proteína PI3-quinasa en el desarrollo y mantenimiento óseo. En esta tesis se describen los fenotipos derivados de la deleción específica en osteoblastos de las isoformas más abundantes en hueso (p110alpha, p 1 10beta y p 1 10alpha/beta). La ausencia de cualquiera de las subunidades catalíticas de PI3K se traduce en una reducción de masa ósea cuando los fémures de dichos ratones son analizados por microtomografia computerizada (micro-CT). El estudio in vitro de osteoblastos primarios obtenidos de los mismos ratones muestran una disminución de los principales genes osteogénicos a nivel de mRNA y proteína, concluyendo que la deficiencia de PI3K en la célula provoca una menor diferenciación osteoblástica. PI3K regula, a través de la fosforilación de AKT, un gran número de efectores. Entre ellos está GSK3, quinasa que se fosforila e inactiva en respuesta a PI3K. Estudios anteriores a esta tesis describen cómo GSK3 fosforila la región "linker" de Smadl (proteína efectora de la vía de BMPs), marcándola para degradación. En concordancia con dichos trabajos, tanto los ratones como los osteoblastos primarios defectivos en PI3K presentan menor cantidad de SMAD1 y de SMAD1 fosforilado, constituyendo así una mecanismo en el cual la falta de PI3K, a través de una menor inactivación de GSK3, aumentaría la degradación de Smads. Dicho mecanismo describe un eje PI3K-GSK3-SMAD en el que la deficiencia de PI3K conlleva a un desarrollo óseo deficitario.

Full textSpinocerebellar ataxia with axonal neuropathy (SCAN1) is a rare recessive neurodegenerative syndrome associated with cerebellar atrophy and peripheral neuropathy. It is caused by a homozygous missense mutation in the tyrosyl-DNA phosphodiesterase-1 (TDP1) gene (A1478G). This results in a substitution of histidine for arginine-493 (H493R) in the TDP1 catalytic site, leading to a reduced TDP1 activity. TDP1 hydrolyses the bond between a DNA 3’-end and a tyrosyl moiety within a trapped topoisomerase I cleavage complex (TOP1cc). TDP1 not only excises trapped TOP1ccs but also processes other 3’-end-blocking lesions, including 3’-phosphoglycolates that result from oxidation. Even so, how TDP1 H493R mutation promotes the SCAN1 phenotype, which is associated with the death of post-mitotic neurons, is unclear. DNA double-strand breaks (DSBs) are infrequent but among the most harmful genomic lesions. Their defective repair can induce cell death, and they have been implicated in the pathogenesis of several human diseases, including neurodegenerative syndromes. Hence, my Ph.D. objective was to investigate whether the SCAN1 phenotype could be related to an accumulation of DSBs in non-replicating cells harboring the H493R mutation of TDP1. The only available models to study the impact of TDP1 H493R mutation were lymphoblastoid cell lines derived from SCAN1 patients compared to those of healthy individuals. Hence, we have generated models of osteosarcoma U2OS cells homozygous for TDP1 H493R or TDP1 KO employing the CRISPR-Cas9 technique. We have also generated primary lung WI38 hTERT fibroblasts TDP1 KO. We found that both TDP1 H493R and TDP1 KO cells accumulate endogenous DSBs, primarily in the G1 phase of the cell cycle compared to S phase. A similar increase of DSBs was observed in quiescent WI38 hTERT cells following depletion of TDP1 with siRNA, suggesting the replication-independent nature of DSBs. Treatment of TDP1 H493R and TDP1 KO cells with camptothecin to induced trapped TOP1ccs, further suggests that accumulation of DSBs could be related to the defective removal of TOP1ccs. Next, we asked whether DSB accumulation in those cells could be related to an increase in DSB production and/or a defect in their repair. Notably, R-loop structures that form co-transcriptionally can induce DSBs in non-replicating cells. We found that TDP1 deficiency modulated R-loop levels at some gene loci, raising the possibility of their implication in DSB formation. Analysis of DSB repair following camptothecin treatment revealed that both TDP1 H493R and TDP1 KO cells were defective in the repair of DSBs in G1 but not in S, with TDP1 H493R having the most pronounced effect. These results suggest that DSBs would accumulate specifically in TDP1-deficient cells that do not undergo replication, due to a defective repair of those breaks. Together, our results provide insights on the etiology of the SCAN1 neurodegenerative syndrome. This work was supported by a PhD fellowship under the French-Italian University VINCI Program 2016

Full textThis paper analyzes morphosyntactic strategies that encode evidentiality and epistemic modality in eastern and western Toba (Formosa, Argentina), from the functional typological perspective (Chafe 1986; Bybee et al. 1994; Givón 2001; De Haan 2001, 2005; Aikhenvald 2004; etc.). In eastern Toba, a reportative derived from a grammaticalyzed ‘say’-verb, and the use of two epistemic adverbs –ʔegoʔ~ʔekoʔ ‘apparently’ and yataqta ‘certainly’– are described. In western Toba, demonstrative roots plus the prefix ho-, after the modified element (noun, verb, adverb, or clause), are analyzed as a strategy of encoding epistemic modality, specifically low subjective certainty.

Full textParallelisms between stories of biblical inspiration and myths that describe the origins of the human beings, their corporal transformations and the differences between diverse types of beings can be found in contemporary narrations of the Toba people (Qom) of the Argentinean Gran Chaco. Events described in the Bible are usually spun with elements of the mythical past of Toba people, therefore generating a biblical reading of the indigenous past and new readings on the human origins, of the differences between beings and of the importance of the body in the constitution of human specificity. This paper examines some of these narrations where myths and Biblical stories are interlaced giving rise to new notions of body and person.

Full textDamaging mRNA, the molecules carrying the genetic information, or altering the mechanisms responsible for their synthesis, their translation or their degradation can be responsible for initiating diseases. It is thus important to understand the mechanisms impacting mRNA, especially their degradation. The latter is initiated by the deadenylation reaction characterized by the progressive shortening of the mRNA 3’ poly(A) tail. One of the complexes responsible for deadenylation is CCR4-NOT. BTG/Tob proteins are able to bind the CAF1 deadenylase of CCR4-NOT via their APRO domain. In this thesis, interaction of BTG2 with PABPC1, the factor binding poly(A) tail, was analyzed. This binding stimulates CAF1 deadenylase activity in vitro and in cellulo. The role of this interaction on the antiproliferative properties of BTG2 and the impact of BTG2 on translation were also investigated. A model explaining deadenylation activation by BTG2 is proposed

Full textIn this thesis, we analyse the relationships between space, social relations and political action between the Nation and the indigenous of the Argentinean Chaco. We consider the Chaco's space as a heterogeneous space, formed by the existence of multiple societies and spaces. By revealing the force of the utopian thought in the consolidation of the Argentinean nation and in the bases of the relationships with the indigenous people, we study the constitution of the Argentinean Nation in the Chaco region. The focus on the construction of the Nation in the Chaco and on the historical constitution of Toba peoples, is carried out by exceeding the idea of the existence of an a-historical condition among indigenous people. We study the contemprorary poach in territories of political action, the fabrication of collectives identities and the cracks of the universalism of some profects (as the citieznship, the indigenism of the class's social movements)

Full textMany uniform total order broadcast protocols have been designed in the last 30 years. They can be classified into two categories: those targeting low latency, and those targeting high throughput. Latency measures the time required to complete a single message broadcast without contention, whereas throughput measures the number of broadcasts that the processes can complete per time unit when there is contention. All the protocols that have been designed so far make the assumption that the underlying network is not shared by other applications running. This is a major concern provided that in modern data centers (aka Clouds), the networking infrastructure is shared by several applications. The consequence is that, in such environments, uniform total order broadcast protocols exhibit unstable behaviors.In this thesis, I provide two contributions. The first contribution is MDC-Cast a new protocol for total order broadcasts in which it optimizes the performance of distributed systems when executed in multi-data center environments. MDC-Cast combines the benefits of IP-multicast in cluster environments and TCP/IP unicast to get a hybrid algorithm that works perfectly in between datacenters.The second contribution is an algorithm designed for debugging performance in black-box distributed systems. The algorithm is not published yet due to the fact that it needs more tests for a better generalization

Full text... llega a los oídos de una manera que no da tregua. Ese es uno de los principales recuerdos de la que debe ser una de las películas de terror más mentadas de toda la historia. Y su creador ha muerto hace poco. Justo cuando empezábamos a asimilar la partida de George A. Romero, ese gran creador del cine grotesco, se nos va otra pieza clave del gore. Es por eso que en CineScrúpulos hemos querido rendir homenaje a dos de nuestros directores más queridos en estas ediciones pequeñas de verano. Pero no vaya a pensarse que Hooper es un creador coherente que hilvanó a la perfección una obra que podría codearse con otros grandes del séptimo arte. Lamentablemente, quizás porque se trató de un chispazo, porque no contó con los recursos necesarios o porque simplemente la actividad cinematográfica se convirtió en una mera excusa que le permitía comer, la filmografía del buen Tobe está plagada de algunos plomazos insufribles y aventuras desteñidas en la pantalla chica. Sin embargo, eso no debe ser motivo para olvidar piezas claves que cualquier fanático del terror con dos dedos de frente debería haber visto y revisto mientras se relame con gusto. Ahí están The Texas chain saw massacre, Eaten alive, Poltergeist o Lifeforce para demostrarlo. Pero esta edición de CineScrúpulos también se engalana con un texto acerca de los acercamientos de Tim Burton al cine de invasión extraterrestre en esa oda a la exageración que es Marcianos al ataque. Y si con ello no basta, pues nos acercamos un poco a la televisión para dar cuenta de hasta qué punto David Fincher es capaz de mantener sus constantes estéticas durante sus aventuras en las series que llevan su firma. Todo está sazonado de tal manera que la edición número 16 de CineScrúpulos sea de tu agrado. Y debido a ciertos cambios en la estructura de los trabajos finales del curso, pero sobre todo gracias al enorme talento de los estudiantes del presente ciclo, tenemos material para más y hemos decidido publicar este verano dos ediciones de nuestra pequeña revista. De ese modo está todo lo que debe estar y tú no pierdes la oportunidad de deleitarte con la sana y amena lectura del equipo de redacción. Como si estuvieras a medianoche en tu casa y de pronto escucharas que el abuelo está con ganas de sentirse matarife mientras Leatherface estrena rostro nuevo y tú te preguntas dónde demonios dejaste el cadáver de la abuela. Ya sabes: lo más importante es la familia.

Full textIn this thesis I analyse how the Toba people of the argentinean Chaco define the person and the body. I examine the body as the space where the relations are established and as the condition for the creation of the person. The idea of the body as the centre of the fulfilment of interpersonal relations has given rise to the notion of the multiple person, which manifests itself adopting different forms of of corporeity. The corporeal connections are realized through the circulation of elements conceived of as "extensions" of persons. These connections are the indirect ways through which relations are made possible. First, the mythological body has served as the starting point for the exploration of entities of the cosmos with the aim of increasing the importance of the body in the processes of differentiation. Second, considering the body from the perspective of corporeal substances, I analyze the links between emotions and cognition

Produtos biológicos, tais como soros hiperimunes e vacinas, podem ser definidos como produtos derivados ou produzidos a partir de organismos vivos. Isso implica que suas características podem variar de um lote para outro. Conseqüentemente, um minucioso controle da qualidade na liberação de lotes é requerido para garantir que esses produtos sejam seguros e eficazes. O ensaio de potência com animais é um dos ensaios preconizados para garantir a qualidade das vacinas e soros hiperimunes. Somente para realização do ensaio de potência para o produto final de vacinas e soros antidiftéricos (SAD) e antitetânicos (SAT), são utilizados no INCQS cerca de 5.000 animais por ano, desses aproximadamente 4.500 animais são utilizados na etapa de soroneutralização e o restante na etapa de imunização. Neste contexto, e baseado nos princípios dos 3Rs, o objetivo deste trabalho foi padronizar e validar o método imunoenzimático de inibição da ligação da toxina (ToBI) capaz de avaliar a potência de SAD, SAT e de vacinas bacterianas combinadas: tétano-difteria uso adulto (dT) e uso infantil (DT) e tétano-difteria-pertussis (DTP), como alternativa da etapa de soroneutralização do ensaio de controle de potência. Os resultados encontrados indicam que o método proposto apresenta linearidade, especificidade e precisão (intra e inter-ensaio) satisfatórias e que é um ensaio confiável, rápido e conveniente para avaliar a potência de componentes diftérico e tetânico presentes em vacinas e soros hiperimunes. Além disso, reduz drasticamente o número de animais usados, o tempo na liberação de laudos e os custos associados ao controle da qualidade destes produtos. O método ToBI apresenta potencial para a substituição do ensaio em animais na determinação de potência de vacinas e soros antidiftéricos e antitetânicos...

Full textThe tonneau1 and fass mutants display considerable morphologic alterations associated with abnomalities in microtubules organisation at the plant cell’s cortex. These are the only mutants that are unable to form the preprophase band (PPB), a ring of microtubules that appears at G2/M transition and forecasts the future division site. TON1 proteins interact with a new family of 31 proteins named TIM, among which some, as the TIM1 protein, are MAPs (Microtubulus associated proteins). TON1 localization on cortical arrays seems to depend on TIM1. The FASS gene encodes a protein that shares similarities with a regulatory subunit of phosphatase PP2A complex. We showed that FASS localizes at the PPB, interacts with TON1 in vivo, and is targeted to the cytoskeleton via its interaction with TON1 and TIM1. FASS also interacts with RCN1, a scaffolding subunits of PP2A complex in A. Thaliana. RCN1 is targeted to the cytoskeleton through its interaction with FASS. Our working model is that the PP2A complex targeted to the cytoskeleton controls the phosphorylation status of MAPs and regulated microtubules organisation. The study of TON1 homolog in P. Patens reveals that TON1 function in the organisation of cortical microtubules arrays is conserved and reveals that PpTON1 is implicated in gravi-perception, a function not described in A. Thaliana. Ours results help to clarify cortical cytoskeleton regulation events and highlight their conservation among land plants and suggest that despites the evolutive divergence, functions that are implicated in microtubules organisation, similar to those usually associated with animal’s centrosome are conserved
