Abstract:
Les chimères Bcr-Abl sont dues à une translocation réciproque entre les chromosomes 9 et 22, entrainant la fusion des gènes bcr et abl. La seule différence structurale entre p190bcr﷓abl, associé avec la Leucémie Aigue Lymphoblastique, et p210bcr-abl, responsable de la Leucémie Myéloïde Chronique, réside dans le domaine DH/PH présent uniquement dans p210bcr-abl. Nous avons montré précédemment que Rac1 était activée dans les cellules exprimant p190bcr-abl et p210bcr-abl alors que RhoA n’est activé que dans les cellules exprimant p210bcr-abl (Harnois et al. , Oncogene, 2003). Les cellules Ba/F3 parentales ne sont pas spontanément mobiles contrairement aux cellules Ba/F3p210 and Ba/F3p190. Les cellules exprimant p210bcr-abl présentent des mouvements amoeboïdes typiques alors que les cellules exprimant p190bcr-abl utilisent une mobilité de type roulement (rolling-type). Les résultats obtenus en utilisant des mutants des domaines GEF de Vav ou p210bcr-abl nous ont permis de proposer un modèle dans lequel Vav, en complexe avec Bcr-Abl, est responsable à travers l’activation de Rac1 du déclenchement de la motilité des cellules exprimant Bcr-Abl. Le GEF de p210bcr-abl, spécifique de l’activation de RhoA, est indispensable pour la formation des mouvements amoeboïdes dans les cellules Ba/F3, mais ne joue pas de rôle dans le déclenchement de la mobilité spontanée de ces cellules (Daubon et al. , Oncogene, 2008). Nous avons ensuite caractérisé une voie passant par Rac1/PAK1/LIMK1/cofiline1 dans laquelle chaque élément est essentiel pour le déclenchement de la motilité dite rolling-type. Nous avons montré que la voie classique RhoA/ROCK/MLC était seulement partiellement impliquée dans l’induction de la motilité amoeboïde dans les cellules Ba/F3p210. De façon surprenante, nous avons trouvé que RhoA et ROCK1 induisent l’activation de l’isoforme ADF/destrine de la famille des « Actin Depolymerizing Factors », à travers la régulation négative de la LIMK2 au niveau transcriptionnel mais aussi par une dégradation protéasome-dépendante. Cette voie de signalisation originale permet une meilleure caractérisation des mécanismes moléculaires menant aux mouvements amoeboïdes en 3 dimensions
Bcr-Abl chimeras are produced by a reciprocal t(9;22) chromosomal translocation that fuses varying amounts of the bcr with the abl genes. The only structural difference between p190bcr-abl, associated with acute lymphoid leukaemia, and p210bcr-abl, responsible for chronic myelogenous leukaemia, resides in a DH/PH domain only present in p210bcr-abl. We previously showed that Rac1 was activated in both p190bcr-abl- and p210bcr-abl-expressing cells whereas RhoA was activated in p210bcr-abl-expressing cells only (Harnois et al. , Oncogene, 2003). Ba/F3 cells were not spontaneously motile in 3D matrigel while Ba/F3p210 and Ba/F3p190 cells moved spontaneously. P210bcr-abl-expressing cells presented typical amoeboid movements in contrast to 190bcr-abl-expressing cells, which showed a rolling-type mobility. The results obtained using mutated GEF domain of Vav or p210bcr-abl allowed to propose a model in which Vav, in complex with Bcr-Abl, is responsible through Rac1 activation for triggering the motility of Bcr-Abl expressing cells. The GEF of p210bcr-abl specific of RhoA does not trigger motility by itself, but RhoA activation is crucial for committing leukaemic cells to amoeboid movements (Daubon et al. , Oncogene, 2008). Bcr-Abl-expressing cells represent then a powerful model to explore the still un-understood amoeboid motility. Then, we characterized a Rac1/PAK1/LIMK1/Cofilin pathway in which each element is critical for triggering the rolling-type motility. We also showed that the classical RhoA/ROCK/MLC pathway was only partially involved in the amoeboid motility of the Ba/F3p210 cells. Surprisingly, we found that RhoA and ROCK1 induced the activation of the ADF/destrin isoform of the Actin Depolymerizing Factor family, through the specific down-regulation of LIMK2 both at the transcriptional level and by proteasome-dependent degradation. This original pathway drives to characteristic amoeboid movements. Taken together, these results on the Ba/F3 cells motility improve the understanding of the molecular mechanisms leading to these 3 dimensional motility modes

Abstract:
Tämän pro gradu -tutkielman aiheena on unkarilaissyntyinen psykiatri Thomas Szasz ja hänen antamansa kritiikki psykiatrialle. Szasz tuli tunnetuksi 1960-luvulla, kun hän kyseenalaisti mielisairauksien olemassaolon julkaisuillaan. Szasz myös julkisesti kritisoi psykiatrian pakottavaa luonnetta ja sen käyttämiä hoitokeinoja. Tällainen oppialan sisältä nouseva kapina omaa tieteenalaa vastaan on harvinaista. Vaikka psykiatrian historiaa on tutkittu valtavia määriä, on Szasz jäänyt tutkimuksissa vähäiselle huomiolle. Tässä pro gradu -tutkielmassa tarkastellaan Thomas Szaszin psykiatrialle antamaa kritiikkiä vuosien 1960–1990 välillä. Tarkoitukseni on ollut siis selvittää mitä Szasz psykiatriassa kritisoi ja miten hän kritiikkinsä perusteli. Myös Thomas Szaszin kritiikin taustalla oleva ajattelu on tarkastelun kohteena. Szaszin näkemykset syntyivät aikana, jolloin mielisairaiden yhteiskunnallinen asema oli heikko ja ongelmat mielisairaanhoidossa alkoivat saavuttaa yleisen tietoisuuden. Hänen psykiatrian kritiikki kytkeytyikin vahvasti oman aikansa yhteiskunnalliseen keskusteluun, johon ihmisoikeudet ja yksilönvapaus liittyivät vahvasti. Szasz ei uskonut mielisairauksien olemassaoloon, vaan piti niitä metaforisina sairauksina. Mielisairaudet eivät olleet Szaszille oikeita lääketieteellisiä sairauksia, vaan enemmänkin yhteiskunnan normeihin sopimatonta käytöstä. Psykiatria oli Szaszille pseudotiede ja yhteiskunnan sosiaalisen kontrollin keino, jonka avulla yhteiskunta hallinnoi normeista poikkeavia yksilöitä pseudotieteellisillä diagnooseilla ja hoitomenetelmillä. Szasz oli poliittiselta vakaumukseltaan libertaristi ja hän perusti kritiikkinsä psykiatriaa kohtaan libertaristisiin ihantenteisiinsa ja etenkin sen käsitykseen yksilönvapaudesta. Szasz näki kaiken yhteiskunnallisen toiminnan, joka vaaransi yksilön vapauden tai perustuslailliset oikeudet, moraalisesti vääränä tai laittomana. Szasz ei ollut valmis tinkimään yksilön vapaudesta mistään hinnasta, ei enemmistön edun nimissä, eikä edes silloin, kun yksilö ei ollut välttämättä kykenevä päättämään omasta parhaastaan. Szaszin näkemys yksilönvapaudesta oli hänen tärkein kulmakivensä psykiatrian kritiikissä.

Introdução: A flexão da anca é determinante na transição da fase de apoio para a fase oscilante na marcha e os músculos responsáveis por esta ação são o iliopsoas, reto femoral, tensor da fáscia lata e sartório. O encurtamento destes músculos gera várias alterações biomecânicas. O teste de Thomas e o teste de Thomas modificado são usualmente utilizados para avaliar a flexibilidade dos músculos flexores da anca. Objetivo: Determinar diferenças na ação cinemática entre os dois testes mencionados. Metodologia: Análise cinemática tridimensional do teste de Thomas e do teste de Thomas modificado através do sistema Qualisys Oqus Camera Series. Resultados: Diferenças estatisticamente significativas na comparação do membro em teste e do joelho contralateral apenas para o teste de Thomas e ausência de diferenças estatisticamente significativas entre o teste de Thomas e o de Thomas modificado para as variáveis cinemáticas. Conclusões: O teste de Thomas modificado parece reduzir compensações nos membros e segmentos analisados; o teste de Thomas aparenta afetar maioritariamente o reto femoral, quando comparado com os resultados obtidos na execução do teste de Thomas modificado.
Introduction: Hip´s flexion is determinant in the transition from the support phase to the oscillating phase in gait and the muscles responsible for this action are the iliopsoas, rectus femoris, tensor fascia latae and sartorius. The shortening of these muscles results in some biomechanical changes. The Thomas test and the modified Thomas test are usually used to assess the flexibility of the hip flexor muscles. Objective: Determine the differences in the cinematic action between the two mentioned tests. Methodology: Tridimensional cinematic analysis of the Thomas Test and Modified Thomas Test through the Qualisys Oqus Camera Series system. Results: Significant statistical differences in the evaluation of the tested limb and the knee of the other limb just for the Thomas Test and lack of significant statistical differences among the Thomas Test and the Modified Thomas Test for the cinematic variables. Conclusions: The Modified Thomas Test seems to reduce compensations/returns on the analyzed limbs and body segments; the Thomas Test appears to affect mostly the rectus femoris when compared with the results achieved in the execution of the Modified Thomas Test.

Abstract:
This research explores the notion of work as a calling and its impact on job performance and innovation in the context of the Catholic Church. Employing the data gained from Catholic priests in Australia and Indonesia and utilising the lens of role identification and person-environment fit theory, this study revealed that priest’s calling is multidimensional and influences different dimensions of their job performance. Additionally, the results found the importance of organisational climate and living the calling as essential factors in enhancing the effect of calling on job performance. This research extends the literature of calling, job performance, and innovation.

La renommée de Paine (1737-1809) est due à la publication de "Common sense" en 1776, puis à celle de "Rights of man" en 1791-1792. Dans le premier de ces pamphlets, Paine plaidait pour l'indépendance des colonies américaines et pour l'établissement d'une république ; dans l'autre, il proposait, dans le contexte anglais, une défense virulente de la révolution française. Paine atteignit un public large grâce à un style mordant et efficace qui se voulait démocratique. Moins connus sont ses écrits théologiques d'orientation déiste. Paine pourtant attachait un grand prix à la dimension religieuse de son oeuvre qu'il rendit publique en 1794 lorsqu'il publia "The age of reason", véritable brulôt religieux rédigé à Paris à l'heure de la déchristianisation. Le déisme de Paine est élaboré en harmonie avec sa pensée politique et participe de la même démarche iconoclaste. Dans ses écrits politiques comme dans ses écrits religieux, Paine cherche à démystifier la réalité existante afin de reconstruire un nouveau systême sur des bases simples et rationnelles. En politique, il montre que le systeme monarchique est fondé sur de faux principes ; en religion, il expose la fausseté du christianisme en attaquant la Bible. L'objectif de cet homme des lumières est de mener l'humanité au bonheur par le biais du républicanisme. Mais ce processus ne peut être accompli que si l'individu parvient à se défaire de l'emprise des "prêtres" et des églises, piliers du système monarchique et source d'obscurantisme. Tout en laissant à chacun la liberte de vénérer Dieu comme il le souhaite, il faut donc laisser émerger le déisme - la croyance en un dieu unique et rien de plus. Seul le déisme propose une conception juste de Dieu et des intentions divines. Seul le déisme élève la création divine - homme et nature - au rang qu'elle mérite
Thomas Paine's fame is mainly due to the publication of two political pamphlets, "Common sense" in 1776, a plea in favour of the independence of the American colonies, and "Rights of man" in 1791-1792, a defence of the French revolution. His theological writings are not as well-known, although Paine took a deep interest in the religious dimension of his work and actively participated in the organization of the first American deistic society, in 1804-1805. Only in 1794 did he publicize his deistic views. In his "Age of reason", he contended that deism (the belief "in one god, and no more") was the only true religion and that christianisty was, in contrast, based on false principles. To this purpose, he forcefully attacked the Bible. His religious (deistic) and political (republican) approaches were similarly oriented. In both fields, he resorted to the same effective, democratic, iconoclastic style, and endeavoured to demystify church and state in order to reconstruct a new system on simple and rational bases. He was a man of the enlightenment and his foremost object was consequently to lead mankind to happiness, a process which he assumed could only be achieved if man managed to get rid of priests and churches. He claimed that every one should be able to worship god as they wished, yet believed that deism would eventually emerge as the only possible religion of an enlightened humanity

Abstract:
Thomas Hardy wrote during a time of extraordinary growth in British education when the purposes of learning were being passionately questioned. This thesis situates Hardy’s writing both within and beyond these debates, showing how his writing avows a Victorian fascination with education while contesting its often rigid actualization in nineteenth-century society. This project places new emphasis on the range of educationalists that Hardy counted as friends. These included the dialect poet and early-Victorian schoolmaster, William Barnes; the influential architect of the 1870s board schools, Thomas Roger Smith; and the leader of late-century reforms to female teacher training colleges, Joshua Fitch. Caught between life in rural surroundings and systemized forms of education, Hardy's characters frequently endure dislocation from community and estrangement from natural environments as penalties of their intellectual development. Much previous scholarship has for this reason claimed education as a source of despair in Hardy’s writing. However, this thesis reveals the people and experiences which rigid institutions exclude, and foregrounds Hardy’s depiction of the natural environment as an alternative source of learning. Exploring Hardy's representations of education as both reflective of contemporary change and suggestive of new possibilities, chapters focus on aspects of education most resonant with Hardy's own life and central to his fiction, including the professionalization and training of schoolmistresses, the working-class movement for liberal education, educational architecture, and rural forms of education. By exploring connections between fiction and social and political concerns, the thesis demonstrates how the idea of education relates to some key characteristics of Hardy's writing, for example the observant onlooker and the native returned.
