Written during his American ‘exile’ and depicting an alternative story of the prophet Moses, T. Mann’s little studied novella The Tables of the Law [Das Gesetz] (1943) is compared with S. Freud’s Moses and Monotheism [Der Mann Moses und die monotheistische Religion] (1939), his last work completed before his death. The author traces a number of significant parallels: in particular, Moses’s origins in the highest classes of Egyptian society. Mann sees the figure of Moses in The Tables of the Law as one of the purest and most vivid embodiments of creative principles, going back to his own period of ‘playing Goethe.’ Moses is yet another of his powerful, harmonious and optimistic characters from the 1930s–1940s (Joseph in the four-part novel Joseph and His Brothers [Joseph und seine Brüder] or Goethe in Lotte in Weimar), with roots in the majestic Dutchman Peeperkorn from The Magic Mountain [Der Zauberberg] (1924). Instead of engaging in a direct polemic with corresponding ideological dogmata, the two books written by the giants of Germanspeaking culture in Nazi times reveal a profound examination of not fully recognized and therefore still dangerous sources of Nazi ideology on the mythological level in mankind’s collective unconscious.

Abstract Introduction: T cell activation is critical in the initiation and potentiation of anti-tumor immune responses. Hematopoietic Progenitor Kinase 1 (HPK1/MAP4K1) is a member of the MAP4K family whose activity has been demonstrated to restrain T cell activation through phosphorylation of SLP-76 at Serine 376 leading to TCR disassembly. Mediators generated in the tumor microenvironment (TME) such as adenosine, PGE2 and TGFβ can dampen T cell activity and present a significant barrier to cancer therapy. HPK1 inhibition may allow for enhanced T cell activation under such suppressive conditions. Herein, we describe studies to assess the effects of HPK1 inhibition on T cell activation and in alleviating suppression. We also test the ability of HPK1 inhibition in combination with adenosine receptor antagonism to further amplify T cell activation. Methods: CD8+ T cells were isolated from human blood and RNA was isolated for qPCR analysis of MAP4K1-7 expression. Cells were activated using CD3/28 stimulation and supernatants were assayed for IL-2, IFNɣ and TNFα using CBA. CRISPR knockout of HPK1 in CD8+ T cells and reference or in-house HPK1 inhibitors used to characterize HPK1 function. We used a flow cytometry-based assay to quantify activation markers CD69 and CD25, and phospho-SLP-76 in isolated CD8+ T cells and in human and mouse whole blood. Results: Using publicly available gene expression databases, we identified that HPK1 is primarily expressed in immune cells, with highest expression in T cells, B cells, and dendritic cells. In-house gene expression analyses demonstrated that HPK1 is the predominant MAP4K family member in human T cells. CRISPR knockout in primary human CD8+ T cells was used to demonstrate that HPK1 but not MAP4K3 and MAP4K4 negatively regulates T cell activation. Consistent with this, reference and in-house HPK1 kinase inhibitors reduced phosphorylation of SLP-76 in both isolated T cells and human and mouse whole blood. In human T cells, HPK1 inhibition dose-dependently increased IL-2, IFNɣ and TNFα secretion, highlighting the negative role of HPK1 in T cell activation. Using adenosine, PGE2 and TGFβ, expression of CD69 and T cell cytokine secretion is diminished. HPK1 inhibition restored T cell activation to unsuppressed levels. Consistent with these results, HPK1KO CD8+ T cells displayed significant resistance to adenosine, PGE2 and TGFβ-induced immunosuppression. Finally, upon suppression using adenosine, combining HPK1 inhibition with adenosine receptor antagonism further elevated T cell activation above individual inhibitor treatments alone. Conclusion: These data demonstrate that HPK1 plays a significant role in dampening T cell activation and provide a strong therapeutic rationale for targeting HPK1 to relieve T cell immunosuppression in the TME and amplify anti-tumor immune responses. This thesis addresses the impact of religious conversion on the later works of Eliot and Auden, and the manner in which they responded to each other as they developed a Christian poetics. Following an introduction which discusses the nature of their relationship as well as their basic theological positions, Chapter One examines their postconversion criticism, and particularly their stance on what is typically formulated as "the problem of belief in poetry," which focuses on how ideology influences a work's creation and reception. Chapter Two considers their transitional poetry, wherein their new religious beliefs figure prominently and their anxiety over the potential conflict between artistic and spiritual values is most acute. Chapter Three looks at their major postconversion poems and specifically at how Eliot's and Auden's understanding of the Incarnation informs their views on time, history, language, and literature, as embodied by these works. Chapter Four centers on their drama, initially comparing their early plays---written when Eliot was a Christian but Auden was not---to show how they employed similar techniques to further different ends, before turning to an examination of Eliot's later verse plays and Auden's libretti. I investigate the ideological motivation behind the adoption of these different dramatic forms, as well as the specific ways in which they affect how belief is conveyed. Throughout the dissertation, the effects of Eliot's and Auden's conversion upon their reputations and the difficulties facing modern Christian artists in general are given particular consideration.

This chapter provides biographical background on Jonathan Daniels. His education at the University of North Carolina, ambitions as a novelist, and publication of Clash of Angels (1930) are highlighted. The death in childbirth of his first wife, Elizabeth Bridgers Daniels, made it difficult for the grieving Daniels to complete a second, satirical novel that might have been his entry into the developing Southern Renaissance alongside his former classmate Thomas Wolfe. The liberal-minded editorials Daniels wrote after taking over from his father as editor of the Raleigh News and Observer in 1933 are contrasted with Josephus Daniels's role in North Carolina's "white supremacy campaign" of 1898 that resulted in the Wilmington massacre. Jonathan's liberalism reflected the influence of other white southern liberals such as Regionalist sociologist Howard Odum and publisher W. T. Couch. New York editor Harold Strauss encouraged Daniels to write a book about the South, resulting in his journey.

This chapter traces the history of Black conservative ideology and its controversial approaches to racial equality, self-help, Black identity, and culture. Throughout the twentieth century, conservative leaders and intellectuals, such as Booker T. Washington, George Schuyler, Joseph Jackson, Thomas Sowell, Anne Wortham, and Shelby Steele, contributed to national debates regarding the nature of inequality and racial discrimination as well as public policy. Although varied in their approaches and perspectives, Black conservatives advocate a coherent ideology that disrupts notions of a homogeneous Black intellectual tradition.

This chapter traces the emergence of the sonnet in African American literature to the pervasive influence of genteel conventions. These conventions have widely been regarded as conservative or even stultifying, but they provided black poets with various opportunities for self-assertion in the public sphere. The sonnet was a favourite genre among the genteel establishment, and poets pushed the boundaries of black expression by appropriating the form to subvert racial stereotypes, develop a black poetic subjectivity, and participate in the debate over the memory of the Civil War. In tracing these developments, the chapter repositions the outstanding poets of the period, Paul Laurence Dunbar and James Weldon Johnson, alongside their less-known contemporaries, Samuel Beadle, William Stanley Braithwaite, Joseph Seamon Cotter Jr., T. Thomas Fortune, and Henrietta Cordelia Ray.

This chapter looks at the relationship between Tony Harrison’s work and the legacies of modernism. In the dominant version of Harrison’s relationship with modernism, the poet undergoes a ‘Eureka’ moment akin to Philip Larkin’s repudiation of Yeats when the Leeds poet begins to write The School of Eloquence sequence: switching to the example of Thomas Hardy, Larkin no longer wished, he contested, to ‘jack himself up’ into poetry, just as Harrison desires to be the poet that ‘blokes in the boozer’ might read. However, as with Larkin’s deployment of symbolist verse and references to T. S. Eliot in his later poetry, Harrison’s work does not simply repudiate modernism either. Harrison’s apparently antipathetic response to modernist literature seems to be encapsulated in Desmond Graham’s chapter on Harrison’s early poetry, in which he depicts the Leeds poet at Poetry and Audience editorial meetings, mimicking Eliot’s voice and demeanour. Yet the poetry tells a different story. In his first full collection, The Loiners, Harrison engages with a number of modernist antecedents, including Arthur Rimbaud, Joseph Conrad and Charles Baudelaire. This chapter then focuses on the modernist ‘double consciousness’ of myth that Harrison then draws on, and refines, throughout his oeuvre. Writers such as James Joyce and T.S. Eliot were the first authors to herald an intensification of writing about myth that fictionalises mythic characters rather than retaining them as symbols and narrative ballast. I explore how Harrison utilises this ‘double consciousness’ in his film-poem Metamorpheus and in the poem ‘The Grilling’ from Under the Clock.
