Abstract:
Background: Metastatic cancer is associated with an invariably fatal outcome. However, DeltaRex-G, a tumor-targeted retrovector encoding a gene-edited dominant-negative CCNG1 inhibitor gene, has induced long term (>10 years) survival of patients with chemo-resistant metastatic pancreatic adenocarcinoma, malignant peripheral nerve sheath tumor, osteosarcoma, B-cell lymphoma, and breast carcinoma. Objective: To evaluate the level of CCNG1 expression in tumors as a potential biomarker for CCNG1 (Cyclin G1-blocking) inhibitor therapy. Methods: CCNG1 RNA expression levels that were previously measured as part of whole genome molecular profiling of tumors (TCGA, N=9161), adjacent “tissues” (TCGA, N=678) and GTEx normal tissues (N=7187) across 22 organ sites were analyzed. Differential expression of CCNG1 and Ki-67 in primary (N= 9161) vs metastatic (N= 393) tumors were also compared in primary (N=103) vs. metastatic (N=367) skin cancers (i.e., melanoma). Statistical Analysis: To detect systematically differential expression of CCNG1 and Ki-67 expression between populations (e.g. tumor vs. normal), unpaired Student's t-tests were performed. Results: Enhanced CCNG1 RNA and Cyclin G1 protein expression were noted in tumors compared to normal analogous counterparts, and CCNG1 expression correlated significantly with that of Ki-67. Moreover, CCNG1 expression tended to be higher than that of Ki-67 in metastatic vs primary tumors. Conclusions: Taken together with the emerging Cyclin G1 / Cdk / Myc / Mdm2 / p53 Axis governing Cancer Stem Cell Competence, this supportive data indicates: (1) CCNG1 expression is frequently enhanced in tumors when compared to their normal analogous counterparts, (2) CCNG1 and Ki-67 expressions are higher in metastatic vs primary tumors, (3) CCNG1 expression is significantly correlated with that of Ki-67, and (4) CCNG1 may actually be a stronger prognostic marker of stem cell competence, chemo-refractoriness, and EMT/metastasis than Ki-67. Phase 2 studies are planned to identify patients most likely to respond favorably to CCNG1 inhibitor therapy.

Abstract:
Prostate Cancer is the most commonly diagnosed cancer in men. The incidence of prostate cancer has risen over the last two decades, although mortality rates have not increased to the same extent. For many men with localised prostate cancer the choice of treatment lies between radical surgery or radiotherapy and active surveillance. Evidence suggests that the survival benefits related to radical treatment are small, and that conservative management may be appropriate for many men. A strategy of focal therapy in prostate cancer aims to treat tumour foci within the prostate, without treating the whole gland, with a resultant reduction in side-effects. Photodynamic therapy (PDT) is the use of a photosensitising drug, which when activated by light can cause cellular damage. Tookad is a new-generation photosensitiser, which is activated in vasculature. Phase I and II clinical trials of Tookad PDT in men with recurrent prostate cancer following radiotherapy have been performed. These studies show that it is possible to ablate prostate tissue with Tookad PDT. This thesis describes a phase I/II clinical trial evaluating the safety and efficacy of Tookad PDT in men with primary prostate cancer. Treatments were performed under general anaesthetic, using 2mg/kg Tookad and interstitial light delivery via cylindrical diffuser fibres. 15 patients were included in part A (single-fibre, light-dose-escalation), 19 patients in part-B (multi-fibre, focal and whole-gland ablation). Follow-up was for 12-months, with assessment of treatment effect made on MRI. Post-treatment imaging demonstrated that prostate tissue can be ablated in a reproducible and controlled manner with Tookad PDT. Although genitourinary side-effects were relatively few, significant cardiovascular complications in this study lead to early termination of the clinical trial and withdrawal of the drug. Results suggest that Tookad PDT could potentially deliver either focal or whole-gland therapy. Further studies with modified photosensitsers are underway.

Machine Therapy is a new practice combining art, design, psychoanalysis, and engineering work in ways that access and reveal the vital, though often unnoticed, relevance of people's interactions and relationships with machines. Machine Therapy will be illustrated through the construction of several systems including re-appropriated domestic devices such as Blendie, wearble apparatuses such as ScreamBody, and body-signal-based companion machines - Umo, Amo, and Omo - that function through visceral interactions including breathing and non-verbal sounds. These systems will be used to explore themes of human-machine relations in terms of visceral, cathartic, and reflexive expressions and communications. This work incorporates elements from my technical research in digital signal processing, machine learning, mechanical engineering, and sensor design. Combining these areas of research and practice, I have been able to help manifest new objects and relationships that are unique in some aspects while maintaining quotidian familiarity in other aspects. These apparatuses enable unusual explorations of what we interact with when we interact with machines. I hypothesize that the answer will turn out to be much more than the machine itself, and will include our sense of self, agency in the interpersonal and political world, and our shared psychological, emotional, cultural, and perceptual approaches to the world. The importance of the parapractic elements and also the therapeutic properties of the Machine Therapy machines will be evaluated in studies of participants' interactive engagements with the machines as well as their affective responses to the machines.

De esta forma, ahorran tiempo, dinero y esfuerzo en trasladar a sus hijos a sus terapias, recibiendo las mismas desde la comodidad de su hogar. En atención a esta necesidad se creó Edukids Therapy, una plataforma web que brinda servicios de terapia del lenguaje para niños en edad escolar, generando en ellos la seguridad para hablar en público y tranquilidad en los padres al advertir el normal desenvolvimiento de sus hijos. La ventaja competitiva de Edukids Therapy es ofrecer un servicio virtual a través de una plataforma web durante el estado de emergencia producido por la pandemia del virus Covid-19, una vez superada esta situación se ofrecerá el servicio de forma presencial y online. Este documento busca exponer ante el lector y/o potenciales inversionistas, un modelo de negocio atractivo para la inversión, ya que demuestra la demanda, viabilidad y rentabilidad de invertir en el mismo, atendiendo un mercado no atendido. Por ello, esta investigación presenta, de forma estructurada, desde la investigación y validación de la demanda del servicio en el mercado, hasta la rentabilidad y periodo de recuperación de la inversión.
This research seeks to present the possibility of a business project supported by a web platform that offers speech therapy services. It is aimed at parents with school-age children who have speech problems, difficulties staying on their children's speech therapies, and fear of catching the Covid-19 virus. In this way, they save time, money and effort in taking their children to their therapies, receiving them from the comfort of their home. In response to this need, Edukids Therapy was created, a web platform that provides language therapy services for school-age children, generating in them the security to speak in public and tranquility in the parents when they notice the normal development of their children. The competitive advantage of Edukids Therapy is to offer a virtual service through a web platform during the state of emergency produced by the Covid-19 virus pandemic, once this situation is overcome, the service will be offered in person and online. This document seeks to present to the reader and / or potential investors, an attractive business model for investment, since it demonstrates the demand, viability and profitability of investing in it, serving an unattended market. For this reason, this investigation presents, in a structured way, from the investigation and validation of the demand for the service in the market, to the profitability and payback period of the investment.

El proyecto consiste en dar solución viable, rentable y novedosa que no existe actualmente en el mercado. El problema que se ha identificado, es que existen aproximadamente 40 mil niños menores de 14 años que viven en Lima Metropolitana y padecen de algún tipo de discapacidad o limitación física y/o mental. Asimismo, la capacidad de atención del sistema de salud a nivel nacional en el Perú es menor al 50%, con solo una sesión de rehabilitación por paciente al mes. Para atender esta necesidad, se desarrolla el proyecto Therapy Kids, que ofrece terapias de rehabilitación para el desarrollo de las capacidades de estos niños. Los apoderados podrán solicitar servicios a través de los diversos canales de atención, en los que se les ofrecerá precios, horarios accesibles y promociones, con el valor diferenciado de que el servicio será brindado únicamente a domicilio y con una frecuencia de tres veces por semana. Las fuentes de ingreso para Therapy Kids serán las terapias a domicilio y las consultas del médico terapista., en el primer año, se espera un ingreso de S/5 682 600 sin IGV y la inversión a realizar en dicho año para lograr los objetivos trazados asciende a S/ 285,202.82, de los cuales el 73% provendrá de los accionistas y se buscará un financiamiento por el 27% restante. Therapy Kids busca evitar el malestar de hacer largas colas y viajes para conseguir una cita con un especialista. Asimismo, no solo busca apoyar a los padres de familia y generar utilidades para la empresa, si no también contribuir considerablemente en el desarrollo del sector salud del país.
