Full textThis work begins from the big interest about School and orientation. It starts with a little story of School and orientation to formulate some questions, to guide the survey. The survey is about the orientation choice for secondary school, in Lomellina. It begins from a questionnaire to parents, than continues with focus-groups with teachers. To end, it have been interviewed some parents, after about 18 months from the questionnaire. The results show that families prefer choice without external suggest. Parents pay a lot of attention to the secondary school presentation, only based on external image. Both teachers than parents have some difficulties to help pupils in decision making, if they haven't good performance at school.

Full textThe emergence of writing has changed the way we live, providing a tool for structuring thinking; transferring what is in memory to an external medium; dialoguing and debating with those who share the same culture and conventions. On the other hand, writing is not a simple task, especially for the beginner student: cognitively, because it requires knowing how to manage several processes and strategies; linguistically, because it is necessary to use several linguistic structures to maintain surface compactness and to ensure text continuity; socially, because it needs to adapt to the recipients. If writing is difficult for all learners, it's even more so for first or second-generation foreigners, whose presence is massive in many schools. For these reasons, the school environment is renewing its methods and proposals with an ever-greater push. This research focuses on one of these educational proposals: the project Osservare l'interlingua (Observing interlanguage). The area of intervention is language education, with a particular focus on writing. The Project was born in 2007 to promote an active, inclusive, and difference-respectful education, so that everyone can acquire the necessary strategies to communicate effectively. In the early years, the Project's activities were aimed only at multilingual pupils, to encourage their linguistic and affective inclusion. Subsequently, the educational objectives were extended to entire classes, to propose even more inclusive and participatory activities. The proposed educational paths, combining the interlanguage approach to language teaching with the principles of cooperative learning and process writing, educate students to perceive writing as an active and ingenious process, to acquire responsibility towards classwork and more abilities in communicating with classmates. The Project not only aims to linguistic development but also refers to transversal objectives, such as the setting up of positive and motivated attitudes towards writing and the construction of a serene and welcoming class climate and good relationships between students. Given both Project’s linguistic and transversal objectives, this research aims to analyze the effects this has produced on classes that have experienced it over a long period (from three to five years). The sample includes experimental classes (five 5th grade and two 8th grade classes) and control classes (four 5th grade and two 8th grade classes), for a total of 226 students. The groups were compared on writing quality, attitudes and motivation towards writing, and class well-being and cohesion. The instruments used were narrative texts written by the students (analyzed through holistic and analytical procedures), four self-perception questionnaires (three on motivation and writing and one on class belonging and well-being), and a sociometric questionnaire (analyzed through structural indices of Social Network Analysis). Given that the Project promotes students' inclusion from its very beginning, the data were analyzed in three formats: whole groups, multilingual students only (active speakers of one or more languages besides Italian), and monolingual students only (Italian-only speakers). The intention is to observe whether the results achieved in entire classes reappear in the two separate subgroups, particularly the multilingual pupils, and to understand if the Project’s methodology is effective in reducing inequalities and generating inclusion.

Full textIn this work, four new neutral cycle peptide , scutianenes W (210), X (211), Y (212) and Z (213), have been isolated from Scutia buxifolia Reiss (Rhamnaceae) , along with seven know compounds, four cylopeptide alkaloids, scutianine B (99), scutianine C (100), scutianine D (101), and scutianine E (102), two triterpenoids , ursolic acid (215) and betulic acid (216), and two steroids, β-sitosterol (214) and β-sitosterol glycoside (217). The structures of compounds isolated were established by various spectroscopic techniques, including 1H and 13C NMR, COSY, NOESY, DEPT, HMQC and HMBC experiments, and comparison with authentic samples. Scutianenes W, X and Y are three diastereoisomeric compounds, with a 3- hydroxileucine as a β-hydroxiamino acid unit, which is connected to the styryl fragment via an ether bridge, and a β-phenylserine as a common ring-bonded amino acid. Attached to the amino group of the 3-hydroxiamino acid unit is a side chain trans CO-CH=CH-PH. In its turn, scutianene Z (213) has a phenylserine as the β-hydroxiamino acid unit, isoleucine as a common ring-bonded amino acid and the same side chain trans CO-CH=CH-PH. The stereochemistry of the four new compounds was established from coupling constants between H3/H4 and H-7/H-28, and by differences in chemical shifts of CH-3, CH-4, CH-7 and CH-28, on the 1H and 13C NMR spectra. The absolute stereochemistry of the diastereoisomeric scutianenes X (211) and Y (212), were confirmed by X-ray diffraction analysis of its O-acetyl derivatives. In addition, extracts and compounds isolated were evaluated for its antimicrobial activity against Gram-positive and Gram-negative bacteria and yeasts, and for its Acetylcholinesterase inhibitory activity.
Neste trabalho, quatro novos ciclopeptídeos neutros foram isolados de Scutia buxifolia Reiss (Rhamnaceae), scutianenos W (210), X (211), Y (212) e Z (213), juntamente com sete compostos conhecidos, quatro alcalóides ciclopeptídicos, scutianina B (99), scutianina C (100), scutianina D (101) e scutianina (102), dois triterpenos, ácido ursólico (215) e acido betulínico (216) e dois esteróides β-sitosterol (214) e β-sitosterol glicosilado. As estruturas dos compostos isolados foram determinados por várias técnicas espectroscópicas, incluindo experimentos RMN 1H e 13C, COSY, NOESY, DEPT, HMQC e HMBC, e por comparação com amostras autênticas. Scutianenos W, X e Y são três compostos diastereoisoméricos, tendo a com 3-hidroxileucina como a unidade de �-hidroxiaminoácido, que está ligado ao fragmento estirilamina através de uma ligação fenol- éter, e uma unidade β-fenilserina como sendo o β- aminoácido do macrociclo. Ligado ao grupo amino temos uma unidade da cadeia lateral [-COCH = CHPh]. Por sua vez, o scutianeno Z (213) possui uma fenilserina como unidade de β- hidroxiaminoácido, isoleucina como um aminoácido ligado ao macrociclo, ligado ao grupo amino temos uma unidade da cadeia lateral [-CO-CH = CHPh]. A estereoquímica dos quatro novos compostos foi estabelecida a partir de constantes de acoplamento entre H3/H4 e H-7/H-28, e pelas diferenças de deslocamentos químicos do CH-3, CH-4, CH-7 e CH-28, nos espectros de RMN de 1H e 13C. A estereoquímica absoluta dos scutianenos diastereoisoméricos X (211) e Y (212), foram confirmados por análise de difração de raios-X de seus derivados O-acetil. Além disso, extratos e todos os compostos isolados foram avaliados para atividade antimicrobiana frente a bactérias Gram-positivas e gram-negativas e leveduras, e determinada a atividade de inibição da acetilcolinesterase.

Full textThe phytochemical investigation of the stem bark of the specie Scutia buxifolia (Rhamnaceae) led to the isolation of four cyclopeptide alkaloids: scutianine C (14) and three diastereoisomer of scutianine H (19), isolated and identified for the first time, coded as P06, P11 and P12 and called scutianines -N (37), -O (38) and -P (39), respectively. The metabolites had their structures elucidated on the basis of spectral data 1H and 13C NMR uni and bidimensional and high resolution mass spectrometry. The diasteroisomeric alkaloids P06, P11 and P12 had the stereochemistry of β-hydroxyamino acid (β-hidroxileucine) established from data 1H and 13C NMR by the coupling constants between H-3/H-4 and chemical shifts of C-3 and C-4. By NOESY technique spatial correlations of the hydrogens were verified and suggested stereochemical for phenylserine unit. The crude extract, the acidic and basic fractions and isolated compounds were tested as to their antimicrobial activity and all demonstrated antimicrobial potential.
A investigação fitoquímica das cascas do caule da espécie Scutia buxifolia (Rhamnaceae) levou ao isolamento de quatro alcaloides ciclopeptídicos: scutianina C (14) e três diastereoisômeros de scutianina H (19), isolados e identificados pela primeira vez, codificados como P06, P11 e P12 e denominados scutianinas -N (37), -O (38) e -P (39), respectivamente. Os metabólitos tiveram suas estruturas elucidadas com base em dados espectrais de RMN de 1H e 13C uni e bidimensionais e espectrometria de massas de alta resolução. Os alcaloides diastereoisoméricos P06, P11 e P12 tiveram a estereoquímica do β-hidroxiaminoácido (β-hidroxileucina) estabelecida a partir de dados de RMN de 1H e 13C pelas constantes de acoplamento entre H-3/H-4 e deslocamentos químicos de C-3 e C-4. Pela técnica de NOESY foram verificadas as correlações espaciais dos hidrogênios e sugeridas as estereoquímicas para a unidade fenilserina. O extrato bruto, as frações ácidas e básicas e os compostos isolados foram testados quanto a sua atividade antimicrobiana e todos demonstraram potencial antimicrobiano.

Full textThe present work describes the study of the secondary metabolites of two species that belong to the Rhamnaceae family: Condalia buxifolia and Scutia buxifolia, and the study of the ratio structure/activity of seven cyclopeptidcs alkaloids and one neutral cyclopeptidc: scutianine-B (24), scutianine-C (14), scutianine-D (15), scutianine-E (16), scutianine-F (18), scutianine-M (37), condaline-A (22) and scutianene-D (38). In the bark of the bole of Condalia buxifolia three metabolites were isolated: lupeol (34), taraxerol (35) and β-sitosterol (33). In the bark of the roots was isolated the cyclopeptidc alkaloid condaline-A (22). In the bark of the roots of Scutia buxifolia the metabolites ursolic acid (36) and β-sitosterol (33) were isolated, whereas the cyclopeptidcs alkaloids scutianine-B (24), scutianine-C (14) and scutianine-D (15) were only identified in this part of the plant by comparison with authentic samples. Additionally, the antimicrobial activity of the portions and the isolated metabolites of Condalia buxifolia and Scutia buxifolia were analyzed by Bioautography test, utilizing different microorganisms: two gram positive bacteria (Staphylococcus aureus (Sa) and Staphylococcus epidermidis (Se)), three gram negative bacteria (Klebsiella pneumonia (Kp), Salmonella setúbal (Ss) and Escherichia coli (Ec)) and two fungus (Saccharomyces cerevisiae (Sc) and Candida albicans (Ca)). In this analysis it was observed that the stereochemistry of the amino acids that compose the alkaloids structure and the presence of the basic group of these structures are important to this sort of activity.
O presente trabalho descreve o estudo dos metabólitos secundários de duas espécies pertencentes à família Rhamnaceae: Condalia buxifolia e Scutia buxifolia, e ainda um estudo da relação estrutura/atividade de sete alcalóides ciclopeptídicos e um ciclopeptídeo neutro: scutianina-B (24), scutianina-C (14), scutianina-D (15), scutianina-E (16), scutianina-F (18), scutianina-M (37), condalina-A (22) e scutianeno-D (38). Da casca do caule de Condalia buxifolia foram isolados três metabólitos: lupeol (34), taraxerol (35) e β-sitosterol (33). Da casca da raiz foi isolado o alcalóide ciclopeptídico condalina-A (22). Da casca da raiz de Scutia buxifolia foram isolados os metabólitos ácido ursólico (36) e β-sitosterol (33), enquanto que os ciclopeptídicos scutianina-B (24), scutianina-C (14) e scutianina-D (15) foram apenas identificados nesta parte da planta através de comparação com amostras autênticas. Adicionalmente, foram analisadas atividades antimicrobianas das frações e dos metabólitos isolados de Condalia buxifolia e Scutia buxifolia, através da técnica de bioautografia utilizando-se diferentes microrganismos: duas bactérias Gram positivas (Staphylococcus aureus (Sa) e Staphylococcus epidermidis (Se)), três bactérias Gram negativas (Klebsiella pneumonia (Kp), Salmonella setubal (Ss) e Escherichia coli (Ec)) e dois fungos (Saccharomyces cerevisiae (Sc) e Candida albicans (Ca)). Observou-se, nesta análise, que tanto a estereoquímica dos aminoácidos que constituem a estrutura dos alcalóides, bem como a presença do grupo básico destas estruturas, são importantes para este tipo de atividade.
