Abstract:

Human hydroxysteroid dehydrogenase-like protein 2 (HSDL2) is a member of the short-chain dehydrogenase/reductase (SDR) subfamily of oxidoreductases and contains an N-terminal catalytic domain and a C-termianl sterol carrier protein type 2 (SCP-2) domain. In this study, the C-terminal SCP-2 domain of human HSDL2, including residues Lys318–Arg416, was produced inEscherichia coli, purified and crystallized. X-ray diffraction data were collected to 2.10 Å resolution. The crystal belonged to the trigonal space groupP3121 (orP3221), with unit-cell parametersa=b= 70.4,c= 60.6 Å, α = β = 90, γ = 120°. Two protein molecules are present in the asymmetric unit, resulting in a Matthews coefficient of 2.16 Å3 Da−1and an approximate solvent content of 43%.

Abstract:

Abstract Relying on the high current temperature rise test, starting from three aspects of different materials, different structural strengths, and different contact conditions, the research on the current-carrying-temperature rise characteristics of the joint terminals of ±800kV UHVDC converter stations under different conditions is carried out. The results show that, for joint terminals of the same structure type and contact area, the difference in resistivity of joint terminals of different materials has a significant impact on the temperature rise level. When the material is the same, the temperature rise level can be reduced to a certain extent after the silver (tin) layer is added to the contact surface of the terminal. When the contact resistance of the joint terminal and the current density are the same, after increasing the contact area, the temperature rise and decrease level of the aluminum alloy plate lap type joint terminal is significantly better than other types of joint terminals. The contact resistance and temperature rise of the joint terminal decrease with the increase of the bolt tightening torque, and become stable when it reaches more than 80%. When the bolt tightening torque reaches more than 90%, the surface finish of the contact surface has no obvious effect on the contact resistance of the joint terminal. By controlling the material properties, structural strength, contact conditions, significantly reduces the temperature rise of the level of the joint terminal, effectively ensuring the safety ±800kV UHVDC converter station stable operation.

Abstract:

This study aims to describe and analyze (1) the Type B Passenger Terminal function in the Southeast Sulawesi Province and (2) the strategy for developing the function of the Type B Passenger Terminal in the Southeast Sulawesi Province. Data analysis, namely with qualitative descriptive analysis and SWOT analysis. The research was carried out in Kendari City in August - December 2022. The results of the study concluded that (1) the terminal function has not been running optimally and, (2) strategies that must be carried out, namely a) Utilizing the terminal's strategic position by encouraging public transport companies using large terminal land, b) Partnering with private parties in accordance with regulations to utilize large terminal land to build terminal supporting facilities, c) Make effective officers at each terminal to direct drivers and passengers to the terminal and control the shadow terminals that disrupt the function of existing terminals, d) Synergize with city/district government in the synergy of the city/regency route network with the location of the type B passenger terminal and e) Encouraging the improvement of the quality of Transportation Human Resources, especially terminal management so that the implementation and management of the terminal runs according to the specified SOP.

Abstract:

Many people are interested in using the shadow terminal area on Jalan Raya Cibeber as a place to wait for public transportation and an interaction point between modes and routes. The purpose of this study is to evaluate the existence of shadow terminals around Pasir Hayam Terminal, focusing on the factors that influence passenger preferences in choosing shadow terminals compared to official terminals. This research method uses a quantitative approach, where Importance Performance Analysis (IPA) is used to evaluate public transportation users' satisfaction with the performance of shadow terminals and main terminals. In addition, the SWOT Analysis Method is used as a tool to help structure the problem, especially in analyzing the strategic environment, which consists of two aspects, namely the internal and external environment. The results of calculations and analysis using the IPA method show that comfort and convenience are the main factors that influence passenger preferences in choosing the Shadow Terminal. Another significant factor is the availability of clean and complete dining and toilet facilities. Through SWOT analysis, Pasir Hayam Terminal is in quadrant 2 which indicates a diversification strategy. In this strategy, the focus will be on optimizing the strengths of the terminal, improving existing weaknesses, taking advantage of open opportunities, and overcoming threats that may be faced. In addition, this strategy includes efforts to increase the advantages of Pasir Hayam Terminal over Terminal Bayangan and utilize the potential of Terminal Bayangan as a strength.

Abstract:

The projection specificity of retinal ganglion cells and the morphology of their terminals were studied in the plethodontid salamander Plethodon jordani. In an in vitro approach, ganglion cells were stained with biocytin and reconstructed by means of light microscopy. Single retinal ganglion cells often have multiple terminal structures in the thalamus, pretectum, and tectum. The projection pattern in the diencephalic neuropils is related to the depth of the terminal arbor within the tectal fiber layer. Terminal arbors in the tectum differ in location, size, and branching pattern. The following types could be distinguished: The most superficial of the optic terminals in layer 1 are relatively small with a diameter of about 100 μm. With the exception of a few varicosities (beads) in the pretectal neuropils, their stem axons have no further collaterals or terminal arbors in the diencephalic neuropils. Intermediate terminals in layer 2 fan out to form a dense plexus with a medio-lateral extent of 180 μm on average. Some terminals in this layer show obvious antenna-like fibers reaching toward the surface of the tectum. The axons of layer 2 projecting neurons have additional collaterals and terminal arbors in the thalamus and pretectum. The deep layer 3 terminals spread out over a diameter of 400 μm on average and their degree of branching is moderate. The axons of layer 3 projecting ganglion cells have dense additional terminal arbors in the thalamus and pretectum. The deepest retinal terminals in the tectum are found within the predominantly efferent fiber layers. This type consists of an unbranched, but beaded axon which runs rostro-caudally with several bends and loops. The stem axon has an additional very dense terminal arborization in the neuropil of the nucleus Bellonci pars medialis and additional sparse collaterals in the pretectal area.

Abstract:

The design and crash test results of a median barrier version of the Flared Energy-Absorbing Terminal, known as FLEAT-MT, are presented. This energy-absorbing terminal is designed for use with a W-beam, strong-post median barrier. The FLEAT-MT terminal uses two standard FLEAT terminals, one for each of the two W-beam rail elements. The energy-absorbing capability of the FLEAT-MT terminal is based on the sequential kinking concept, similar to that used with the Sequential Kinking Terminal and FLEAT guardrail terminals. Three full-scale vehicle crash tests were conducted to evaluate the impact performance of the FLEAT-MT terminal in accordance with guidelines set forth in NCHRP Report 350: Test 3-35—pickup truck redirection test (Test No. FMT-1), Test 3-31—pickup truck head-on test (Test No. FMT-2), and Test 3-39—pickup truck reverse-direction test (Test No. FMT-3M). The terminal performed as designed. The FLEAT-MT terminal meets all evaluation criteria for a Test Level 3 median barrier terminal set forth in NCHRP Report 350. The FLEAT-MT terminal is being evaluated by FHWA for approval to be used on the National Highway System.

Abstract:

Permasalahan sanitasi yang ada di negara berkembang disebabkan beberapa faktor diantaranya adalah minimnya perhatian dan prioritas yang diberikan oleh pemerintah atau dinas terkait pada sektor sanitasi. Penelitian ini bertujuan untuk menggambarkan sanitasi lingkungan di terminal Kota Luwuk Kabupaten Banggai. Jenis penelitian ini yaitu deskriptif. Populasi dalam penelitian ini adalah seluruh terminal yang ada di Kota Luwuk Kabupaten Banggai tahun 2020 yaitu berjumlah 2 terminal, adapun teknik pengambilan sampel secara total populasi. Analisis data menggunakan SPSS versi 2.0. Hasil penelitian menunjukan bahwa dari keseluruhan variabel yang di teliti sebagian besar belum memenuhi syarat. Saran bagi petugas terminal dan masyarakat untuk selalu menjaga dan merawat fasilitas yang ada di terminal agar fasilitas tersebut masih layak digunakan sebagaimana mestinya. Sanitation problems that exist in developing countries are caused by several factors, including the lack of attention and priority given by the government or related agencies in the sanitation sector. This study aims to describe environmental sanitation in Luwuk City terminal, Banggai Regency. This type of research is descriptive. The population in this study were all terminals in Luwuk City, Banggai Regency in 2020, which amounted to 2 terminals, as for the total population sampling technique. Data analysis using SPSS version 2.0. The results showed that of the overall variables studied, most of them did not meet the requirements. Suggestions for terminal officers and the public to always maintain and care for existing facilities at the terminal so that these facilities are still suitable for use as they should.

Abstract:

The Indium Gallium Arsenide (InGaAs) based MOSFETs have been widely used in the research of high-speed devices with higher frequency. It has some application in the designing areas of power amplifiers. The InGaAs mainly have greater electron mobility and the lesser band gap in their compound makes them more suitable for developing high-speed devices. The Indium Gallium Arsenide compound-based MOSFETs are designed using the source/drain grown on a passive layer of Indium Phosphide substrate. This helps in reducing the power budget of the MOSFET and thereby reduces source and drain resistance. The re-grown layers over the bulk have serious issues such as parasitic capacitance and greater electrical field at the terminals of the gate along with the drain terminal. This results in a larger leakage current along with the terminals and thereby induces the degradation of the frequency of the application amplifiers. The high-ƙ dielectric along the gate terminal makes the device immune to leakage current for lesser frequency applications. The optimum material for the dielectric may be Hafnium (IV) Oxide – HfO2 which has been used as a sidewall in the proposed InGaAs MOSFET design. The device simulation was carried out in a way to evaluate the characteristics of the proposed designs. The results were submissive to the conventional MOSFETs in terms of output capacitance over the source and drain terminals, leakage current in the drain terminal, and improved frequency parameters. The results also suggested that the sidewall design over the gate terminal constitutes the frequency improvement without losing the power and current characteristics.

Abstract:

Tanjung Priok Port is one of Indonesia's largest and busiest ports, playing a vital role in goods trade and passenger transportation. This research focuses on enhancing the infrastructure at the Nusantara 1 and 2 passenger terminals to increase the port's competitiveness and value, particularly in the passenger terminal area. Using GAP Analysis, literature review, interviews, observations, and documentation, the study aims to provide solutions for improving and expanding passenger terminal facilities to enhance passenger service and value. The findings indicate that optimal maintenance of port infrastructure significantly enhances the port's value and passenger comfort. Observations reveal a yearly increase in passengers at the PURA 1 & 2 terminals, reaching 353,005 passengers in 2022. The temporary waiting room accommodates 1,600 passengers, while the general waiting room holds 3,666 passengers. Despite the good maintenance of these facilities, further improvements are necessary to meet the growing demands.

Abstract:

Single crystals of the the title compound, 2K+·C58H56O18 2−·4CH3OH, were obtained from a methanol solution. The anion is centrosymmetric; consequently the asymmetric unit contains one-half of the salt and two methanol molecules. Each polyether chain contains one K+, which is coordinated by eight O atoms belonging to the carboxylate, terminal hydroxy and six ether groups. The structure of the entire anion is such that the two K+ are enclosed in the shape of an S. One methanol molecule forms hydrogen bonds with the O atom of the carboxylate group that does not participate in the coordination. The other methanol molecule forms hydrogen bonds with the other O atom of the carboxylate group that coordinates with K+ and also with the terminal hydroxy group of an adjacent salt unit.

Abstract:

The article is a review of the transport problems that appear in a marine intermodal terminal. The importance of intermodal terminals has been increasing since the 1980s as a result of the development of containerized international trade. This increase in the handling of containers cause problems connected with securing adequate space for container storage or efficient usage of available terminal loading equipment such as cranes, AGVs, straddle carriers or terminal tractors with trailers. The equipment of a terminal depends mainly on the size of containers flow. The use of this equipment determine the terminal technology and work organization. That is why the classification of intermodal container terminals is presented in chapter 2. This classification was made according to different criteria. To understand transport problems, the identification of processes performed in the marine intermodal terminal is presented in chapter 3. For the purpose of research an example of an intermodal marine terminal with its individual functional areas is presented. Finally, the literature review on research conducted on the transport processes performed in the terminal are presented in chapter 4. These processes refer to quay side and land side horizontal transport operations. Additionally, the arrangement of containers at the storage yard was investigated.

Abstract This paper investigates the novel labeling diversity (LD) technique with two transmit antennas and receive antennas applied in the Reed-Solomon coded cooperative (RSCC) system to propose a novel RSCC-LD scheme over the Rayleigh quasi-static fading and frequency-flat fast-fading channels. Two separate RS codes over the same Galois field are deployed at the source and relay terminals, respectively with the relay RS code providing more extra redundancy. To optimize the use of the information contained in two RS codes, a novel joint decoding approach is developed at the destination terminal. The results of numerical simulation demonstrate that the proposed RSCC-LD has an obviously better performance than the non-cooperative RS coding scheme and RSCC scheme without LD. Furthermore, the performance of the RSCC-LD reveals the superiority of some other existing schemes.

Abstract:

The utility of the container terminal has become quite remarkable all over the world. It is vital to maximize the utility of the container yard and the terminal gate system. With the ever-increasing importance of containerization in the world, more than 60% of cargo is transported by containers, this ratio even reaches 100% in some highly economic developed and political stable places. It is estimated that, the total quantity of containerization in the world will increase by 10% every year before 2020. The aim of this work is to be used as a decision support system tool when designing and constructing container terminals. The article focuses on optimizing the staging of containers within a terminal gate system. A Port within the main Land China is being used as a test bed to demonstrate the Simulation Model based on Queuing Theory. The analysis considers both the facility and the setting of the terminal gate system by considering the Discrete Event System Theory and the terminal gate system in order to make the daily operation more suitable for the terminal. The research combines the computer simulation method and the actual complex discrete event system, and identifies the most suitable setting and the theoretical maximum capacity for the terminal. Abstract:

Regulation of the epithelial Na+ channel (ENaC) by ubiquitylation is controlled by the activity of two counteracting enzymes, the E3 ubiquitin-protein ligase Nedd4-2 (mouse ortholog of human Nedd4L) and the ubiquitin-specific protease Usp2-45. Previously, Usp2-45 was shown to decrease ubiquitylation and to increase surface function of ENaC in Xenopus laevis oocytes, whereas the splice variant Usp2-69, which has a different N-terminal domain, was inactive toward ENaC. It is shown here that the catalytic core of Usp2 lacking the N-terminal domain has a reduced ability relative to Usp2-45 to enhance ENaC activity in Xenopus oocytes. In contrast, its catalytic activity toward the artificial substrate ubiquitin-AMC is fully maintained. The interaction of Usp2-45 with ENaC exogenously expressed in HEK293 cells was tested by coimmunoprecipitation. The data indicate that different combinations of ENaC subunits, as well as the α-ENaC cytoplasmic N-terminal but not C-terminal domain, coprecipitate with Usp2-45. This interaction is decreased but not abolished when the cytoplasmic ubiquitylation sites of ENaC are mutated. Importantly, coimmunoprecipitation in HEK293 cells and GST pull-down of purified recombinant proteins show that both the catalytic domain and the N-terminal tail of Usp2-45 physically interact with the HECT domain of Nedd4-2. Taken together, the data support the conclusion that Usp2-45 action on ENaC is promoted by various interactions, including through binding to Nedd4-2 that is suggested to position Usp2-45 favorably for ENaC deubiquitylation.

1. The effect of presynaptic, axoaxonal inhibition, that exerts its action by producing a local conductance increase, on the behavior of action potentials at postsynaptic axon terminals is analyzed computationally. The significance of the location and strength of the presynaptic inhibition, as well as the morphology and membrane properties of the axonal terminals, are considered. 2. Keeping the specific properties of terminal membrane and axoplasm constant, the critical "silent" steady-state conductance change (gcrit) that blocks propagation is linearly scaled with the terminal diameter raised to the 3/2 power. At the midpoint of a 5 lambda long, 1 micron diameter axon that has the standard Hodgkin and Huxley (1952) kinetics at 18 degrees C (and an input conductance of 8.7 nS), gcrit is 72 nS. At 0 degrees C, gcrit = 200 nS, whereas at 30 degrees C gcrit = 30 nS. 3. The critical conductance change that blocks propagation depends steeply on the density of excitable channels (gNa) at the terminal. For a geometrically uniform terminal at 18 degrees C, the action potential can not be blocked by a local shunt when gNa greater than 600 mS/cm2. 4. An axoaxonal synapse in the proximity of the postsynaptic release site has graded control over the spike amplitude (and, therefore, over the postsynaptic output) at that site. Presynaptic inhibition located remotely from the release site has an all-or-none effect at the release site. 5. Inhibition is more effective in attenuating the spike at the terminal when it impinges onto a passive terminal rather than on an excitable one. 6. The detectability of the conductance increase as well as the change in action potential amplitude associated with the presynaptic inhibition is poor at only a short distance from the axoaxonal synapse. The presence of bottlenecks and varicosities at some axonal terminals enhances this decoupling effect even more. Thus presynaptic inhibition may reduce the potential near the output site of the affected axon significantly, without producing any noticeable conductance or voltage change at a distance of only several tens of micrometers (a few tenths of a space constant) away from it. 7. Varicosities and bottlenecks typically found along axonal terminals are the optimal loci for presynaptic inhibition to be most effective in attenuating the action potential at the terminal. Even with high density of excitable channels, propagation with such terminal geometry is insecure, and one, or few, quanta of inhibitory transmitter (a conductance change of several nanosiemens) can block propagation there.(ABSTRACT TRUNCATED AT 400 WORDS)

Abstract:

To characterize the "reliability" or "robustness" of a network, more information is required than simply the system failure rate. In particular the length of the downtimes can be of great relevance. Hence, in this paper we present two methods for computing the system downtime distribution in 2-terminal network problems. The first method is an importance sampling simulation which allows simulations to capture the tail of the distribution with much greater precision than simple Monte Carlo methods. Hence the statistics of unusually long failures can be investigated by simulation. The second method is an approximate analytical method whereby system failures due to 1 or 2 or … or N component failures are characterized exactly. The error in using N=2 and neglecting greater than three simultaneous component failures is shown to be negligible in many cases of interest. This method can be extended to handle N>2 component failures but the resulting calculations escalate rather quickly and are omitted here. This technique makes it possible to provide approximate failure time distributions very rapidly for arbitrary networks.

By immunoreactivity analysis using monoclonal antibodies, we showed that the C-terminal domain [R415–631; R is residue] of progelatinase A [pro-matrix metalloproteinase-2 (proMMP-2); EC 3.4.24.24] affected the immunoreactivity of a one-step sandwich enzyme immunoassay (sandwich EIA) for tissue inhibitor of metalloproteinases-2 (TIMP-2) in exactly the same way as does proMMP-2 [Fujimoto, Zhang, Iwata, Shinya, Okada and Hayakawa (1993) Clin. Chim. Acta 220, 31–45], confirming that the C-terminal domain (‘tail’ portion) of TIMP-2 participates in the binding with the C-terminal domain of proMMP-2. We also demonstrated that not only the C-terminal domain but also the N-terminal domain (R1–417) of proMMP-2 bound to TIMP-2 in a 1:1 molar ratio. The binding of each individual domain to TIMP-2, however, was weak enough that either domain could be fully replaced by proMMP-2 itself, suggesting that either terminal domain binds to TIMP-2 through the same binding sites as does proMMP-2, and also that the high-order structure of proMMP-2 allows a more stable binding to TIMP-2. We further confirmed that TIMP-2 complexed with the N-terminal domain of proMMP-2 had fully inhibitory activity against the collagenolytic activity of MMP-1. We also demonstrated that either the interstitial collagenase–TIMP-2 complex or the gelatinase B (MMP-9)–TIMP-2 complex was able to form a ternary complex with proMMP-2 in a 1:1 molar ratio, clearly indicating that there are two distinct binding sites, one specific for proMMP-2 and the other for active MMPs, on the TIMP-2 molecule. The C-terminal domain was able to bind to the MMP-9–TIMP-2 complex, but the binding seemed to be less stable than the binding with TIMP-2 alone. Even in the presence of a 10-fold molar excess of the N-terminal domain, ternary complex formation was not observed between the N-terminal domain and the MMP-9–TIMP-2 complex. These clear differences might be ascribed to some significant conformational change(s) evoked in the TIMP-2 molecule, or hindrance of a part of the N-terminal domain binding site of TIMP-2 by complex formation with MMP-9.

As crucial international trade and global logistics players, container terminals worldwide handle more than 80% of the global merchandise trade. After analyzing and summarizing the previous studies, we use container terminal companies as the research object to fill the gap left by previous studies. Based on the above research status, this study analyzes the operational efficiency and total factor productivity of 32 container terminal companies in China using the super -efficiency DEA–SBM model and the Malmquist index method. The results show that (1) the operational efficiency level of 32 container terminals in China from 2017 to 2020 has a huge gap, and 15 container terminal companies have operational efficiency below 0.6, which indicates that most container terminals have excess inputs and a waste of resources. (2) The container terminals in the Bohai Rim, Pearl River Delta and Yangtze River Delta regions have higher operational efficiency. This shows that the development of container terminals cannot be separated from the economic hinterland of the cities where the ports are located. (3) The Malmquist index analysis shows a 2.8% decrease in total factor productivity, a 3.2% increase in the composite technical efficiency index and a 5.8% decrease in the technological progress index, which indicates that most container terminal companies have imperfect management practices and decision making. Based on the study’s results, this research provides relevant and feasible recommendations for policymakers who formulate policies for the development of the shipping industry to promote high quality and sustainable development of the shipping industry and the economy.

The purpose of the article is to develop a refined methodology for the general design of a container terminal. The difference between the given design methodology and the existing ones is that a decision is proposed to change its throughput (annual container flow passing through it), without changing the dimensions of the container yard and the way containers are stored, that is, for the same capacity of the container yard. Such cases occur, for example, when it is necessary to reconstruct a container terminal. Cargo terminals for containers are a specific field of logistics and cargo delivery system, where cargo is processed in consolidated transport modules - containers. Considered the main stages of designing a container terminal as a complex stochastic technical system: setting the task (it should be a single one); selection and determination of elements of technological zones and the structure of the container terminal, as various connections between system elements; analysis of system functioning (including its modeling); account of interaction of the system with the environment; consideration of the results of the container transport and technological system and operation. It is determined that this general methodology can be used during the creation or analysis of any other transformation of a technical or social system that receives a stream of some objects with one set of parameters, processes it through its structure and outputs them from other parameters. This method of formalizing the functioning of the container terminal can be used for its calculations, modeling, research and optimization. In the processes of interaction with environmental systems, the container terminal passes from one state to another, which is characterized by the types and volume of technological operations that must be performed at a certain moment, cargo that must be stored, delivered to and from the terminal, assembled, sorted, business employee processes, forklifts, cranes and other equipment in use. The functioning of the container terminal (part of the warehouse system) as a procedure of transitions from one state to another can be represented in the form of a graph or transition probability matrix.

Electrical activity recorded intracellularly from peptidergic neurosecretory terminal dilatations in the sinus gland of crabs (principally Cardisoma guanhumi and C. carnifex) is described. Recordings were made from the neurohaemal organ in situ on the neural tissue of the isolated eyestalk and from isolated sinus gland-sinus gland nerve preparations. Verification that electrodes penetrated terminals was obtained by dye marking. Resting potentials ranged between −30 and −80mV. Overshooting action potentials of long duration (5–20 ms at 1/2 amplitude) relative to those of non-secretory axons (less than 2ms) were recorded in approximately 70% of stable penetrations. Action potentials occurred spontaneously at slow (less than 0.2s-1) rates in 75% of penetrations. Sequential intra- and extracellular recordings with the same microelectrode, on the same terminal, indicate impulse generation by the terminal itself. Extracellular stimulation of the axon tract evokes an all-or-none action potential at distinct threshold and latency. At rates of stimulation exceeding 5s-1, discrete fluctuations in the form of responses occur. Similar waveforms occur spontaneously and can be evoked by passing current through the electrode. They are interpreted to be electrotonically recorded activity of other parts of a complex axonal terminal arborization. Some, but not all, terminals exhibit impulse broadening (up to three-fold at 1/2 amplitude) during repetitive firing exceeding 1s-1. The same terminals show reduced impulse duration with hyperpolarization and broadened impulses with imposed depolarization. The changes are due to altered repolarization rates. Terminals sustain steady impulse firing at rates (up to 5s-1) linearly related to the imposed depolarizing current. Regenerative potentials, though of reduced rate of rise and amplitude, can be evoked by depolarizing current passed through the electrode during perfusion with salines having 1/2 normal [Na+], or containing tetrodotoxin (10(−6)moll-1). However, these block axonal conduction. Nominally Ca-free saline causes increased spontaneity and depolarization of about 5 mV in half the preparations examined, but reaching 20 mV in the others, with resultant inactivation of regenerative activity. Impulses in low-Ca saline show alterations of the falling phase, it being faster initially and then slower than normal. Thus, while the action potentials of neurosecretory axons are Na dependent, the terminals support regenerative impulses mediated by both Na and Ca.

The lodgepole terminal weevil, Pissodes terminalis Hopping, is the only member of its genus that consistently oviposits in the expanding terminal shoots of its hosts (Stark and Wood 1964). The white pine weevil, Pissodes strobi (Peck), oviposits in the year old shoot of spruce and pine (Wallace and Sullivan 1985) and other Pissodes species have their oviposition sites on boles, root collars, and slash (Furniss and Carolin 1977). In southern British Columbia, P. terminalis is primarily univoltine although adults may live and oviposit for 2 or more years. Although its life cycle is closely synchronized with terminal phenology, its life history is extremely variable (Cameron and Stark 1989). Adults are active in late spring to early summer, when they can be found feeding on the tissues of the developing terminal shoot and foliage. Soon after this maturation feeding, mating occurs, and oviposition punctures are excavated in the bark of the new terminal. Usually a single egg is deposited into each puncture (Drouin et al. 1963; Stark and Wood 1964; Maclauchlan 1992).

Abstract Pseudomonas putida rubredoxin-2 (Rxn2) is an essential member of the alkane hydroxylation pathway and transfers electrons from a reductase to the membrane-bound hydroxylase. The regioselective hydroxylation of linear alkanes is a challenging chemical transformation of great interest for the chemical industry. Herein, we report the preparation and spectroscopic characterization of cobalt-substituted P. putida Rxn2 and a truncated version of the protein consisting of the C-terminal domain of the protein. Our spectroscopic data on the Co-substituted C-terminal domain supports a high-spin Co(II) with a distorted tetrahedral coordination environment. Investigation of the two-domain protein Rxn2 provides insights into the metal-binding properties of the N-terminal domain, the role of which is not well understood so far. Circular dichroism, electron paramagnetic resonance and X-ray absorption spectroscopies support an alternative Co-binding site within the N-terminal domain, which appears to not be relevant in nature. We have shown that chemical reconstitution in the presence of Co leads to incorporation of Co(II) into the active site of the C-terminal domain, but not the N-terminal domain of Rxn2 indicating distinct roles for the two rubredoxin domains.

Abstract:

Optimized conditions for the synthesis of fully deprotected (2R)-aziridine containing dipeptides are described. Preparation of fully protected N- and C- terminal aziridine containing dipeptides was found to be straightforward and high yielding for the majority of compounds, whereas their full deprotection was possible only for C-terminal analogs. Deprotection of N-terminal derivatives using standard procedures of peptide chemistry was found difficult providing only mixtures of unidentifiable products. The described molecules have potential as building blocks in synthetic chemistry, in the chemical biology arena, as covalent modifiers, and as biomarkers.
