Full text
Abstract:
Five compounds of mono para-hidroxy chalcon were synthesized (TC1, TC2, TC3, TC4, and TC5) and tested their cytotoxicity against HeLa cell and Raji cell. The difference in substituent of TC1 (R4 =H), TC2 (R4 = OCH3), and TC3 (R4 = F), showed the difference of their citotoxicity against HeLa cell. The citotoxicity of TC1 (LC50 = 16.08 µg/mL) ≈ TC3 (LC50 = 13.37 µg/mL), but the substituent difference of TC2 (LC50 = 147.43 µg/mL), decreasing it citotoxicity 10 times. Like wise their citotoxicity against Raji cell of TC1 (LC50 = 36.44 µg/mL) ≈ TC3 (LC50 = 30.46 µg/mL), but the substituent difference of TC2 (LC50 = 468.94 µg/mL), decreasing it citotoxicity activity 15 times. Nevertheless the strength of citotoxicity TC4 (LC50 = 98.74 µg/mL) and TC5 (LC50 = 110.97 µg/mL) against Raji cell are stronger than the citotoxicity of two of them against HeLa cell (LC50 of TC4 = none, LC50 of TC5 = 576.53 µg/mL). Abstract:
Aim. To evaluate the safety issues and adverse effects of using TCHP regimen (docetaxel, carboplatin, trastuzumab, and pertuzumab) versus TCP regimen (docetaxel, carboplatin, and trastuzumab) in older postmenopausal women with nonmetastatic HER2-positive breast cancer. HER2 overexpressed in 20–25% of breast cancer signals an aggressive form of breast cancer and is treated with trastuzumab and pertuzumab. Methods. The patient record database was accessed to identify all postmenopausal women in the Punjab Care hospital who were above 65 years old, with stages 1–3 HER2-positive breast cancer and treated with neoadjuvant TCHP and neoadjuvant TCP from 2013 till 2016. Results. In TCH-P group and TCH group, mild fatigue (34% versus 36%) and diarrhea (48% versus 49%) were most common toxicities. Fever in TCH-P group and TCH group (12% versus 13%) was common. Anorexia affected 21% and 16% of patients receiving TCH and TCHP regimen, respectively. Febrile neutropenia was higher in TCH-P group 13% (3/23) versus 4.5% (1/22) in TCH group. Also 27.2% (6/22) of TCH-P group was hospitalized for treatment related toxicities versus 21.7% (5/23) of TCH group. Conclusion. Comparing neoadjuvant TCP and neoadjuvant TCH-P showed TCH-P regimen had an acceptable toxicity profile. Severe cardiac dysfunction was not observed. Using TCH-P regimen can be considered as relatively safe therapeutic option for elderly postmenopausal women with nonmetastatic HER2-positive breast cancer.

Abstract:
Contamination with 2,4,6-trichloroanisole (TCA) often causes taste and odor (T&O) problems in drinking water due to its low odor threshold concentration. Microbial O-methylation of the precursor 2,4,6-trichlorophenol (TCP) would be the dominant mechanism for TCA formation. Simple and rapid measurement of TCP in the low concentration range is necessary to control the problems induced by TCA. In this study, the combination of microbial conversion and instrumental analysis was proposed as a method of TCP quantification. Fungi and bacteria were isolated from various water samples and examined for their ability to produce TCA from TCP. As a result, a strain exhibiting quantitative TCA production and a high growth rate was obtained and named Mycolicibacterium sp. CB14. The conversion rate of TCP to TCA by this strain was found to be high and stable (85.9 ± 5.3%), regardless of the applied TCP concentration, although within the range of 0.1–10 µg/L. The limits of detection and quantification for TCP by this proposed method were determined to be 5.2 ng/L and 17.3 ng/L, respectively. By improving the methods, Mycolicibacterium sp. CB14 could be used for the quantification of TCP at very low concentration levels, which is sufficient to manage the T&O problem caused by TCA.

Abstract:
ABSTRACT Cork taint is a musty or moldy off-odor in wine mainly caused by 2,4,6-trichloroanisole (2,4,6-TCA). We examined the role of 14 fungal strains isolated from cork samples in the production of 2,4,6-TCA by O methylation of 2,4,6-trichlorophenol (2,4,6-TCP). The fungal strains isolated belong to the genera Penicillium (four isolates); Trichoderma (two isolates); and Acremonium, Chrysonilia, Cladosporium, Fusarium, Mortierella, Mucor, Paecilomyces, and Verticillium (one isolate each). Eleven of these strains could produce 2,4,6-TCA when they were grown directly on cork in the presence of 2,4,6-TCP. The highest levels of bioconversion were carried out by the Trichoderma and Fusarium strains. One strain of Trichoderma longibrachiatum could also efficiently produce 2,4,6-TCA in liquid medium. However, no detectable levels of 2,4,6-TCA production by this strain could be detected on cork when putative precursors other than 2,4,6-TCP, including several anisoles, dichlorophenols, trichlorophenols, or other highly chlorinated compounds, were tested. Time course expression studies with liquid cultures showed that the formation of 2,4,6-TCA was not affected by a high concentration of glucose (2% or 111 mM) or by ammonium salts at concentrations up to 60 mM. In T. longibrachiatum the O methylation of 2,4,6-TCP was catalyzed by a mycelium-associated S-adenosyl-l-methionine (SAM)-dependent methyltransferase that was strongly induced by 2,4,6-TCP. The reaction was inhibited by S-adenosyl-l-homocysteine, an inhibitor of SAM-dependent methylation, suggesting that SAM is the natural methyl donor. These findings increase our understanding of the mechanism underlying the origin of 2,4,6-TCA on cork, which is poorly understood despite its great economic importance for the wine industry, and they could also help us improve our knowledge about the biodegradation and detoxification processes associated with chlorinated phenols.

Abstract:
Combined chemical and sensory methods were used for the source identification of musty off-flavours in water; the methods were based on stripping enrichment, gas chromatography (GC) with both instrumental and sensory detection, and preparative GC. Two case studies of drinking water derived from groundwater (one chlorinated and one non-chlorinated) showed that 2,4,6-trichloroanisole (TCA) was the major odorous compound. Furthermore, the concentration of TCA increased significantly in the distribution system. Studies of surface waters showed that TCA is widespread in Sweden, and in one small forest lake, it was found that TCA made a significant contribution to the total odour of the water. The formation of TCA during distribution was probably caused by microbial methylation of 2,4,6-trichlorophenol (TCP). TCP is formed during chlorine disinfection of drinking water and has recently also been identified as a natural halogenation product. In the present study, fungi (of the genera Phialophora, Acremonium and Penicillium) and actinomycetes isolated from drinking water were capable of producing TCA by methylating added TCP.

Abstract:
Elution of bisphenol A (BPA), chlorinated BPAs (i.e., 3-chlorobisphenol A, sum of 3,5-dichlorobisphenol A and 3,3′-dichlorobisphenol A, 3,3′,5-trichlorobisphenol A, and 3,3′,5,5′-tetrachlorobisphenol A) and 2,4,6-trichlorophenol (TCP) from lined pipes coated with two epoxy resins were investigated in a 24-month continuous test passing tap water. BPA, chlorinated BPAs, and TCP were not detected in the tap water at the outlet of lined pipes in most cases. However, all of these chemicals were detected in the tap water after 16 h of retention. The sums of BPA and chlorinated BPAs in the retained tap water were usually high when the residual chlorine levels were low. The residual chlorine was related to temperature, residual chlorine in the tap water at the inlet of lined pipes, and the elapsed time. From the results of leach tests, it was indicated that no marked changes in BPA concentration over the 24 months of the continuous test. Chlorinated BPAs and TCP were accumulated on the surface of lined pipes during the continuous test.

Abstract:
Nine families of bread wheat (TC5, TC6, TC7, TC8, TC9, TC10, TC14, 5395-(243AA), and 5395) with resistance to barley yellow dwarf virus and containing putative translocations between wheat and a group 7 chromosome of Agropyron intermedium (L1 disomic addition line, 7Ai#1 chromosome) induced by homoeologous pairing or tissue culture were analyzed. C-banding, genomic in situ hybridization (GISH), and restriction fragment length polymorphism (RFLP) in combination with repetitive Agropyron-specific sequences and deletion mapping in wheat were used to determine the relative locations of the translocation breakpoints and the size of the transferred alien chromatin segments in hexaploid wheat–Agropyron translocation lines. All homoeologous compensating lines had complete 7Ai#1 or translocated 7Ai#1–7D chromosomes that substitute for chromosome 7D. Two complete 7Ai#1 (7D) substitution lines (5395-(243AA) and 5395), one T1BS–7Ai#1S∙7Ai#1L addition line (TC7), and two different translocation types, T7DS–7Ai#1S∙7Ai#1L (TC5, TC6, TC8, TC9, and TC10) and T7DS∙7DL–7Ai#1L (TC14), substituting for chromosome 7D were identified. The substitution line 5395-(243AA) had a reciprocal T1BS∙1BL–4BS/T1BL–4BS∙4BL translocation. TC14 has a 6G (6B) substitution. The RFLP data from deletion mapping studies in wheat using 37 group 7 clones provided 10 molecular tagged chromosome regions for homoeologous and syntenic group 7 wheat or Agropyron chromosomes. Together with GISH we identified three different sizes of the transferred Agropyron chromosome segments with approximate breakpoints at fraction length (FL) 0.33 in the short arm of chromosome T7DS–7Ai#1S∙7Ai#1L (TC5, TC6, TC8, TC9, and TC10) and another at FL 0.37 of the nonhomoeologous translocated chromosome T1BS–7Ai#1S∙7Ai#1L (TC7). One breakpoint was identified in the long arm of chromosome T7DS∙7DL–7Ai#1L (TC14) at FL 0.56. We detected some nonreciprocal translocations for the most proximal region of the chromosome arm of 7DL, which resulted in small duplications. Key words : C-banding, genomic in situ hybridization (GISH), physical mapping, translocation mapping, RFLP analysis.

Abstract:
A gradient bioceramic composite coating was prepared by wide-band laser cladding technique on TC4 alloy surface. The influence of rare earths oxide CeO2 on microstructure of bioceramic coating was studied. The experimental results indicated that CeO2 plays an important role in inducing HA + β-TCP formation. There is almost no HA+β-TCP in bioceramic coating without CeO2. When CeO2 content is higher than 0.2 wt.%, the amount of HA+β-TCP catalyzed by CeO2 gradually increases. The amount of HA+β-TCP becomes largest when CeO2 content is up to 0.4wt%. However, when CeO2 content ranges from 0.6 wt.% to 0.8 wt.%, the amount of synthesizing HA+β-TCP conversely goes down. Through cell culture experiment in vitro, the effect of bioceramic coating with different CeO2 contents on the expression of characteristic protein is investigated. The results show that the largest amount of expression of hydroxyproline(Hyp) at 2d and alkaline phosphatase(ALP) at 6d on coating is complied with 0.4wt.% CeO2, The result indicates that bioactivity of bioceramic coating is dependent on the amount of HA + β-TCP catalyzed by different CeO2 contents.

Abstract:
The concentrations of organophosphoric acid triesters (OPEs) in water samples from Maizuru Bay were in the range of 3.0–62 ng/l. In general, the concentrations of OPEs were found in the order of TBXP > TDCPP > TCEP > TBP > TCP > TPP > TEP. The organophosphorous pesticides (OPPs) diazinon, fenitrothion, iprobenfos and chlorpyrifos were detected in water samples. The concentrations of OPEs in sediment from Maizuru Bay were in the range of <0.5–56 µg kg−1 dry weight (dw). Among OPPs, diazinon in sediment samples were in the range of 1.8–71 µg kg−1 dw. However, the detection frequencies of fenitrothion and chlorpyrifos in sediment were low. The concentrations of OPEs in mussels from Maizuru Bay were in the range of <1–34 µg kg−1 wet weight (ww). The concentrations of OPEs were found in the order of TBP > TDCPP > TCP > TBXP = TPP = TCEP. Pesticides were detected in mussels, but these concentrations were lower than the acceptable daily intake (ADI) values. The partition coefficients between water and sediment (Kws) of diazinon and fenitrothion were 200–1300 and 200–300, respectively and the partition coefficients between water and biological samples (Kwb) of diazinon and fenitrothion were 700–3300 and 450–700, respectively, suggesting that these pesticides accumulate in biological samples at higher rates than in sediment.

Abstract:

Abstract:
Transmission Control Protocol (TCP) is working well in wired networks. TCP needs modification to work well in wireless networks. This work analyses the performance of the proposed two TCP cross layer flavours, namely the TCP-AL and TCP-WPAL. The cross layer interaction (TCP-WPAL) produces better performance than the TCP-AL. The PDR (Packet Delivery Ratio) of the TCP-WPAL is increased, and the delay and jitter of the TCP-WPAL are decreased in multihop wireless networks. Abstract:
The structures of the charged porphyrins and their dimers have been investigated with computational methods. Dimers have been formed based on electrostatic attraction of the opposite charges on two different porphyrin monomers, tetra ammonium porphyrin (TAP) and tetra carboxy porphyrin (TCP). Semi-empirical quantum mechanical calculations have been employed to explore the most stable ground-state structures of TCP, TAP and their hetero-dimers. Dimeric structures analyzed are all in face-to-face fashion indicating the strong electrostatic attraction between the two porphyrin rings. Calculations have also predicted that the protons transfer from [Formula: see text]; groups to -COO- groups when the interplanar separation is shorter than 3.7 Å.

Abstract:
Calcium phosphates (CaPs) are synthetic bone grafting materials. CaPs are an alternative to overcome the drawbacks present with autologous bone grafting and/or xenograft materials. Among the CaPs, tricalcium phosphate (TCP) stands out as a good candidate due to its physicochemical properties. The clinical performance of β-TCP has already been proven and established. Nevertheless, the format in which TCP is delivered is also important in terms of clinical handling. This work assessed the in vivo performance of TCP-based bone grafting materials with different formats. Materials studied were a TCP paste (TCP-P), a TCP foam (TCP-F) and TCP granules (TCP-G). A sheep scapula model was used to evaluate the osteogenic performance of these bone grafting materials. All materials performed well in terms of bone regenerative capacity and material resorption. However, TCP-P and TCP-F displayed a more pronounced initial material resorption and also exhibited better handling properties compared to TCP-G. TCP-based materials with improved handling properties, such as TCP-P and TCP-F, which at the same time possess the advantageous properties of β-TCP are suitable bone substitute materials for grafting and reconstruction of bone defects in numerous clinical applications.

Abstract:
To improve the quality of bioceramic gradient coatings, wide band laser cladding technique was adopted to produce a kind of gradient composite bioceramic coating, which combines the good biocompatibility of HA and β-TCP with good toughness and strength of TC4 alloy. The rare earths oxide La2O3 was added to improve the properties of the bioceramic coating. The results indicate that when the adding of La2O3 is 0.6wt%, the ceramics coating is of the largest amount of HA and β-TCP catalyzed, and the number of cells proliferated and the ALP secreted on coatings is the biggest, which indicates that the biocompatibility and bioactivity of gradient bioceramic coating is the best. The biocompatibility and bioactivity are closely related to the amount of HA and β-TCP catalyzed by La2O3.

Abstract:
Beta-tricalcium phosphate/carboxymethyl chitin composites [TCP/CMCh] of various ratios of TCP granules and CMCh were made and their mechanical properties, handling properties and repair performance for bone defects and for osteochondral defects were investigated. Water pooling ratio of CMCh was approximately 40 times the weight itself. TCP/CMCh of a higher TCP ratio had higher stress at 50%-strain. The stress at 50%-strain of TCP/CMCh with 0, 2.5, 5.0, 7.5, 10 TCP ratios was 0.12, 0.51, 1.08, 1.46, 1.67 (MPa, n=5), respectively. The TCP/CMCh with 5.0 TCP ratios had the best total scores in handling tests. The bone repair rate of TCP/CMCh was TCP ratio 2.5< Blank= TCP ratio 7.5< TCP ratio 5.0. In the implantation test for osteochondral defects, TCP/CMCh was completely absorbed at four weeks after surgery. Regeneration of the articular cartilage was seen with TCP/CMCh and HA/CMCh but not with TCP granules, which remained eight weeks after implantation. The regenerated articular cartilage had remained 32 weeks after implantation. In conclusion, it was demonstrated that this TCP/CMCh composite was a promising material for repairing osteochondral defects.

Abstract:
TCP (Transmission Control Protocol), is a reliable connection oriented end-to-end protocol. It contains within itself, mechanisms for ensuring reliability by requiring the receiver to acknowledge the segments that it receives. The network is not perfect and a small percentage of packets are lost enroute, either due to network error or due to the fact that there is congestion in the network and the routers are dropping packets. TCP ensures reliability by starting a timer whenever it sends a segment. If it does not receive an acknowledgement from the receiver within the ‘time-out’ interval then it retransmits the segment. In this paper a review of various TCP is carried out. There are a number of TCP variants for application in the management of network efficiency in terms of network congestion and transmission efficiency. These variants include: - TCP Tahoe, TCP Reno, TCP New Reno, TCP Vegas, TCP SACK, TCP FACK, TCP Asym, TCP RBP, Full TCP and TCP CUBIC. Therefore, the main objective of this paper is to study the tcp types on the network performance variances. All have different features and advantages but with maximal throughput as main objective, which are termed as the clones of TCP, have been incorporated into TCP/IP protocol for handling congestion efficiently in different network scenarios.

Abstract:
2,4,6-trichloroanisole (2,4,6-TCA) formation is often reported as a cause of taste and odor (T&O) problems in water distribution systems (WDSs). The biosynthesis via microbial O-methylation of 2,4,6-trichlorophenol (2,4,6-TCP) is the dominant formation pathway in distribution pipes. This paper attempted to utilize the reported data on the microbial O-methylation process to formulate deterministic kinetic models for explaining 2,4,6-TCA formation dynamics in WDSs. The pipe material’s critical role in stimulating O-methyltransferases enzymatic activity and regulating 2,4,6-TCP bioconversion in water was established. The kinetic expressions formulated were later applied to develop a novel EPANET-MSX-based multi-species reactive-transport (MSRT) model. The effects of operating conditions and temperature in directing the microbiological, chemical, and organoleptic quality variations in WDSs were analyzed using the MSRT model on two benchmark systems. The simulation results specified chlorine application’s implication in maintaining 2,4,6-TCA levels within its perception limit (4 ng/L). In addition, the temperature sensitivity of O-methyltransferases enzymatic activity was described, and the effect of temperature increase from 10 to 25 °C in accelerating the 2,4,6-TCA formation rate in WDSs was explained. Controlling source water 2,4,6-TCP concentration by accepting appropriate treatment techniques was recommended as the primary strategy for regulating the T&O problems in WDSs.

Abstract:
Twenty-seven L-shaped ortho-quinone analogs were designed and synthesized using a one pot double-radical synthetic strategy followed by removing methyl at C-3 of the furan ring and introducing a diverse side chain at C-2 of the furan ring. The synthetic derivatives were investigated for their cytotoxicity activities against human leukemia cells K562, prostate cancer cells PC3, and melanoma cells WM9. Compounds TB1, TB3, TB4, TB6, TC1, TC3, TC5, TC9, TC11, TC12, TC14, TC15, TC16, and TC17 exhibited a better broad-spectrum cytotoxicity on three cancer cells. TB7 and TC7 selectively displayed potent inhibitory activities on leukemia cells K562 and prostate cancer cells PC3, respectively. Further studies indicated that TB3, TC1, TC3, TC7, and TC17 could significantly induce the apoptosis of PC3 cells. TC1 and TC17 significantly induced apoptosis of K562 cells. TC1, TC11, and TC14 induced significant apoptosis of WM9 cells. The structure-activity relationships evaluation showed that removing methyl at C-3 of the furan ring and introducing diverse side chains at C-2 of the furan ring is an effective strategy for improving the anticancer activity of L-shaped ortho-quinone analogs.

Abstract:
AbstractThe growth, gut ecology and immunocompetence of Oreochromis niloticus and the resistance to Aeromonas hydrophila were investigated after been fed with diets containing dietary Curcumin longa for 12 weeks. Diets were formulated to contain 30% crude protein with diet TC1, TC2, TC3, TC4 and TC5 having 0% (control), 0.25%, 0.5%, 0.75% and 1.0% turmeric powder, respectively. Diets were allotted to groups of O. niloticus (mean weight of 1.29± 0.15 g) and replicated thrice for 84 days. Results showed that the highest mean final weight (4.79±0.04 g) was obtained in TC3 and corresponded to the treatment with the highest feed intake. A significantly high (p<0.05) specific growth rate (SGR) was observed in TC3 (0.73±0.03 %day−1) while TC4 (0.57±0.02 %day−1) gave the lowest value. The highest microbial load in the gut was observed in TC1 groups and the least in TC4 groups. Red blood cell count, hemoglobin, packed cell volume did not show significant variation (p>0.05) across treatments. However, white blood cell (WBC) count was significantly higher in TC1 (control). There was an improved immunocompetence, as aspartate aminotransferase (AST) progressively reduces in fish fed supplements. Similarly, there was a better oxidative response in the treated groups with reduced hydrogen peroxidase, increased total protein and glutathione peroxidase. Mortality ranged from 25% in TC4 to 95% in TC1 after the challenge test with A. hydrophila. This study showed that C. longa inclusion at 0.5% is more beneficial when growth and health status of O. niloticus juveniles are considered.
