The Venous Thromboembolism (VTE) risk factors are environmental and genetic. The well established risk factors are anti-thrombin, protein C, protein S deficiency, Factor V Leiden and factor II mutation and ABO gene, with A1 and B allele increasing the risk of VTE. While an important part of VTE heritability remains unexplained, contemporary studies fail to discover new susceptibility genes with weaker effects. In order to increase the discovery power, I searched for genetic geterminism of two intermediary phenotypes of VTE : Factor VIII plasmatic activity (FVIII) and von Willebrand factor antigenemia (vWF)First, I performed a linkage study of FVIII and vWF from a sample of 5 large pedigrees (N=255). Four loci have been identified. One included ABO gene. I searched for candidate genes located in the others loci by studying in silico results from o Genome Wide Association Study (GWAS) of the VTE including 419 cases and and 1228 controls. témoins. Two candidate genes were identified : STAB2 et BAI3. Then I performed association studies of five SNPs in BAI3 with FVIII and vWF. One of them was associated to vWF (in a sample of 108 nuclear families and 916 VTE patients), and associated to VTE in two case-controls samples (respectively 916 cases and 801 controls, and 250 cases et 607 controls).Second, I performed a meta-analysis of three GWAS of FVIII and vWF from the same 5 pedigrees and two samples of VTE (N=972 and 570) adjusted on ABO blood group. No polymorphisms were significant after Bonferoni correction (p<10-7). Nevertheless, among 11 genes carrying polymorphisms with a p<10-5, interestingly was STAB2. Futhermore, this study allowed to confirm newly discoverd association with VWF, STXBP5 et STX2

Soil treatment, especially with hydraulic binders or lime, is a widespread technique to improve the mechanical characteristics of poor-quality soils. After their construction, engineering structures are subjected to environmental solicitations that can potentially alter the effects of the treatment, and thus lead to the degradation of the performance of the treated soil. In this context, the objective of this thesis was to study the impact of the accumulation of hydric cycles on the mechanical behaviour of a cement-treated sand. A multi-scale and multi-physics methodology was set up to understand and quantify the degradation of the macroscopic mechanical behavior of cement-treated sands during drying/wetting cycles.Short-term mechanical characterization of the sand was performed by conducting a triaxial testing campaign. The bonding ratio, η_bondmax, defined according to the stress-dilatancy theory, was introduced to assess the mobilization and the degradation of cementitious bonds during the triaxial tests. In order to characterize the durability of the treated sand, the impact of two types of hydric cycles of different intensity was evaluated. Following the cycles, triaxial tests were performed constituting a database of 130 test results. The bonding ratio monitoring allowed to explicitly quantify the degradation of the cemented specimens as a function of the type and number of cycles. The main effect of the hydric cycles is to alter the cementitious bonds and subsequently to decrease the mechanical performances. This alteration depends on the cement content but also on the intensity of the cycles. Type I cycles lead to a more important degradation than type II cycles. The kinetics of the alteration as well as its extent depend on the cement content.The investigation of the processes occurring at the microscopic scale after 24 cycles of the two types via microscopic observations (SEM and TEM) and physicochemical analyses (XRD and GTA) has highlighted intense mineralogical transformations including carbonation of the cementitious phases to various degrees depending on the intensity of the cycle as well as the formation of ettringite needles in the pore-space. However, in terms of macromechanical behaviour, a stabilization of mechanical performance after a moderate decrease (-20 to -30% of the deviatoric stress) was observed after 12 hydric cycles. This approach has shown that relating mineralogical transformations to durability is not sufficient. It is necessary to evaluate the contribution of each phase to the strength since intense mineralogical transformations are not necessarily associated with strong degradation of the treated soils.A constitutive law to model the observed deviatoric behaviour of treated sand was proposed by combining the Hardening Soil Model for the pre-peak hardening and an inverse sigmoid function for the post-peak softening. This approach showed a satisfactory accuracy to simulate the behaviour and in particular the softening of treated soils under relatively high confining pressures. Under low confining pressures an exponentially decreasing function was however more suitable. In addition, parameters optimization determined the most appropriate values for 5 model parameters (ψ, Rf, m, λ, and ecrit) as a function of cement content. The modeling permitted to complete the experimental study and broughtsome reflexions on the way to simulate the post-peak behaviour.This study allowed a mechanical quantification from macroscopic observations coupled with a physicochemical quantification of the various processes occurring during hydric cycles. A conceptual framework allowing to take into account the effect of cementing in the behaviour of treated soils was proposed

Nous avons ciblé les interactions entre Pin1 et la protéine Tau comme modèle de substrats pour une étude détaillée des mécanismes intervenant à l'échelle moléculaire, sur base de substrats peptidiques, qui permettraient d'expliquer le rôle fonctionnel de Pin1. L'interaction avec les substrats au travers des motifs Ser/Thr-Pro phosphorylés est double : un domaine de liaison WW permet la liaison du substrat et un domaine catalytique PPIase (peptidyl-prolyl isomérase) catalyse l'isomérisation cis/trans des prolines. Un criblage par RMN des différents motifs phospho-Ser/Thr-Pro au sein de la protéine Tau a permis de déterminer un nouveau site d'interaction centré autour du motif Thr212-Pro213, phosphorylé uniquement dans la forme pathologique de Tau.
Nous avons étendu l'investigation des interactions avec Pin1 à l'échelle de la protéine Tau entière. Comme pour la plupart des régions protéiques impliquées dans les interactions avec Pin1, la protéine Tau se caractérise par une absence de structure globale qui limite considérablement les études par RMN. Un fragment peptidique de 40 acides aminés comprenant les sites Thr231 et Thr212 phosphorylés a permis de montrer un rôle régulateur du domaine WW dans l'activité enzymatique. Une première étude avec une protéine mutante mimant l'état phosphorylé de Tau a montré une interaction avec le domaine catalytique de Pin1 et a nécessité la mise au point préalable d'une technique d'attribution de la protéine Tau par RMN que nous avons appelé « mapping peptidique ».
La phosphorylation du domaine WW de Pin1 est associée à l'inhibition de la liaison des substrats et joue un rôle dans la régulation de l'activité de Pin1 in vivo. La forme non phosphorylée active de Pin1 est retrouvée majoritairement dans les cellules cancéreuses et la forme phosphorylée inactive dans les cellules saines. Nous avons envisagé de cibler les interactions entre Pin1 et les phospho-peptides avec la synthèse de molécules organiques mimant le dipeptide phosphoThr-Pro et la mise en œuvre d'un test de criblage par RMN pour l'obtention d'inhibiteurs ciblant le domaine WW de Pin1 qui pourraient mimer la forme inactive de la protéine.

The recent advancement in MRI systems and the increase of the static magnetic field strength were accompanied by a strong concern about the effect of RF electromagnetic fields on patients. The effect can be increased by the use of conductive elements inside the volume coil as in the case of endoluminal coils used to analyze digestive walls. These coils lead to an increase of the local SAR which is induced by RF electric field in the presence of the coaxial cable connecting the coil to the MR system, resulting in strong local heating. Giving that these coils have the potential to present a real benefit to the patient, it worth to overcome these limitations. Accordingly, the first objective of the thesis is the development of a fully optical endoluminal receiver coil. An optical active detuning system has been developed and characterized. The NMR images show a signal-to-noise ratio distribution similar to that obtained with conventional detuning techniques, thus validating the efficiency of the optical detuning. Concerning the electro-optical conversion and the optical transmission of the NMR signal, experiments were performed to overcome constraints related to the use of waveguide for electro-optical conversion by Pockels effect. Moreover, the importance of monitoring global and local SAR during MRI exams remains a need which is not limited only to the endoluminal coils. The second objective of the thesis is then the experimental validation of an electro-optical probe for real-time measurements of RF electric field. This probe can measure the RF electric field in air and in biological media at 3 T and 4.7 T MRI systems and allows the estimation of the local SAR
