Tujuan artikel ini adalah untuk menjabarkan proses kerja produksi karya musik The Story of Tapa Malenggang.Tapa Malenggang merupakan sastra lisan yang berasal dari Kabupaten Batanghari, Desa Tanjung, Maruo Provinsi Jambi. Tapa Malenggang mempunyai nama asli yaitu Mambang Diawan yang hidup dipintu langit merupakan sebuah ritual. Dalam ritual Tapa Malenggang, terdapat dendang yang dihadirkan sebagai bentuk syukur dan terima kasih atas kebaikan Tapa Malenggang yang telah menjaga kedamaian masyarakat disekitar sungai Batanghari. Pengkarya tertarik untuk mengangkat cerita Tapa Malenggang dengan mengadodpsi sepenggal melodi dendang ke dalam konsep musik elektro-akustik dan menggunakan beberapa teknik-teknik sound design. Elektro-akustik merupakan suara atau bunyi yang terdengar, tidak lagi terlihat sumber aslinya. Metode yang di gunakan dalam artikel ini adalah pengolahan elektro-akustik menggunakan beberapa teknik sound design seperti audio synthesis yang meliputi wave shape, LFO dan volume envelope. Hasil yang diperoleh dalam artikel ini adalah gambaran proses kerja dalam produksi karya musik The Story of Tapa Malenggang.ABSTRACTThe purpose of this article is to describe the work process of producing the musical work The Story of Tapa Malenggang. Tapa Malenggang is an oral literature originating from Batanghari Regency, Tanjung Village, Maruo, Jambi Province. Tapa Malenggang has a real name, namely Mambang Diawan, who lives at the door of the sky as a ritual. In the Tapa Malenggang ritual, there is a song that is presented as a form of gratitude and thanks for the kindness of Tapa Malenggang who has kept the peace of the community around the Batanghari river. The authors are interested in bringing up the story of Tapa Malenggang by adopting a piece of dendang melody into the concept of electro-acoustic music and using several sound design techniques. Electro-acoustic is a sound or sound that is heard, no longer seen the original source. The method used in this article is electro-acoustic processing using several sound design techniques such as audio synthesis which includes wave shape, LFO and volume envelope. The results obtained in this article are an overview of the work process in the production of the musical work The Story of Tapa Malenggang.

Rono tapa (teri panggang) adalah ikan teri yang dibungkus daun pisang lalu dipanggang. Rono dan tapa, adalah dua suku kata dari bahasa Kaili. Rono artinya ikan teri dan Tapa artinya panggang. Penelitian ini bertujuan untuk mengetahui kualitas kimia mie basah rono tapa, sedangkan manfaatnya adalah untuk dapat memberikan informasi ilmiah mengenai kualitas kimia mie basah rono tapa. Penelitian ini dilaksanakan pada bulan Agustus 2023. Pembuatan Mie Basah Rono Tapa dilaksanakan di Laboratorium THP STPL Palu. Analisis protein, kadar air, kadar abu dan kalsium dilaksanakan di Laboratorium Kimia FMIPA Universitas Tadulako Palu. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) yaitu pengujian kadar air dan kadar abu yang terdiri dari 4 perlakuan dan 3 ulangan sehingga diperoleh 12 unit percobaan. sedangkan pengujian kalsium dan protein menggunakan deskriptif dengan formulasi mie basah empat taraf perlakuan, yaitu 0%, 15%, 25%, dan 35% rono tapa setiap perlakuan dilakukan 3 ulangan dan dilanjutkan dengan uji Anova. Hasil dari pengujian analisis didapatkan diantaranya; rata-rata kadar kalsium tertinggi adalah pada perlakuan P3 yaitu 0,035 % dan yang terendah terdapat pada perlakuan P0 yaitu 0,020 %. Rata-rata kadar air tertinggi adalah pada perlakuan P3 yaitu 44 % dan yang terendah terdapat pada perlakuan P0 yaitu 38 %. Rata-rata kadar protein tertinggi adalah pada perlakuan P3 yaitu 7,975 % dan yang terendah terdapat pada perlakuan P0 yaitu 4,706 %. Rata-rata kadar abu tertinggi adalah sama untuk 3 perlakuan yaitu 3 % dan yang terendah terdapat pada perlakuan P0 yaitu 2 %. Analisis kimia pada mie basah rono tapa yang terbaik adalah pada perlakuan P3 yaitu kadar kalsium 0,035%. kadar air 44%. kadar protein 7,975%. Kadar abu adalah sama untuk 3 perlakuan yaitu 3%.

ABSTRACTThis study aims to describe the packaging of teaching materials on the material of Appreciation and Expression of the Tapa-tapa Dance through audio visual to be taught to students of class X of High Schools in North Sumatra. In addition, it describes the steps and results in making Tapa-tapa Dance packaging through audio visual to be taught to high school students in North Sumatra. The theories used are Setyosari's (2015) packaging theory, namely packaging steps which include planning, compilation, production, and evaluation. The theory of teaching materials Prastowo (2015) is a teaching material that is arranged systematically and used for the learning process in the form of audio visual. The method used in this study is a qualitative research method. The instrument used to test the feasibility of audio visual teaching material (VCD) is to use the Audio Visual Product Validation Test (VCD) questionnaire and the Material Validation Test Questionnaire, this questionnaire consists of aspects of assessment which are based on competency standards. This research resulted in an audio visual teaching material product VCD for Tapa-tapa dance material. Starting from (1) Planning by making teaching materials, selecting dancers, determining the location of taking photos and videos of products, and preparing facilities and infrastructure, (2) Preparation, where all teaching materials for KD 3 appreciation and KD 4 expressions covering background behind the Tapa-tapa dance, the Tapa-tapa dance function, the basic movements of the Tapa-tapa dance, cosmetology, fashion, musical instruments, tutorials and video performances of the Tapa-tapa dance performance that can be used as teaching material in dance learning, (3) Production is editing of images and sound results using Wondershare Filmore, (4) Evaluation, conducted by 2 validators for material validation and 1 validator for audio visual validation (VCD) by providing a validation questionnaire. In the validation stage, the results of validation by material experts with a score of 4.6 and validation by audio visual product (VCD) experts with a score of 4.4. From the score the product of teaching material in the form of audio visual (VCD) got a very good category applied to students of class X of High School in North Sumatra. Keywords: Packaging, Teaching Materials, Audio Visual (VCD) ABSTRAKPenelitian ini bertujuan untuk mendeskripsikan pengemasan bahan ajar pada materi Apresiasi dan Ekspresi Tari Tapa-tapa melalui audio visual untuk diajarkan kepada siswa kelas X Sekolah Menengah Atas di Sumatera Utara. Selain itu mendeskripsikan langkah-langkah dan hasil dalam pembuatan pengemasan Tari Tapa-tapa melalui audio visual untuk dapat diajarkan kepada siswa Sekolah Menengah Atas di Sumatera Utara. Teori-teori yang digunakan adalah teori pengemasan Setyosari (2015) yaitu langkah-langkah pengemasan yang meliputi perencanaan, penyusunan, produksi, dan evaluasi. Teori bahan ajar Prastowo (2015) yaitu materi ajar yang disusun sistematis dan digunakan untuk proses pembelajaran dalam bentuk audio visual. Metode yang digunakan dalam penelitian ini adalah metode penelitian kualitatif. Instrumen yang digunakan untuk menguji kelayakan bahan ajar audio visual (VCD) adalah dengan menggunakan angket Uji Validasi Produk Audio Visual (VCD) dan Angket Uji Validasi Materi, angket ini terdiri dari aspek penilaian yaitu berdasarkan standar kompetensi. Penelitian ini menghasilkan suatu produk bahan ajar audio visual VCD materi tari Tapa-tapa. Dimulai dari (1) Perencanaan dengan pembuatan materi bahan ajar, pemilihan penari, penetapan lokasi pengambilan foto dan video produk, serta penyiapan sarana dan prasarana, (2) Penyusunan, dimana semua materi bahan ajar kegiatan KD 3 apresiasi dan KD 4 ekspresi yg meliputi latar belakang tari Tapa-tapa, fungsi tari Tapa-tapa, gerak dasar tari Tapa-tapa, tata rias, tata busana, alat musik, tutorial serta penampilan video pertunjukan tari Tapa-tapa yang dapat digunakan sebagai bahan ajar pada pembelajaran tari, (3) Produksi yakni penyuntingan hasil gambar dan suara dengan menggunakan Wondershare Filmore, (4) Evaluasi, dilakukan oleh 2 validator untuk validasi materi dan 1 validator untuk validasi audio visual (VCD) dengan cara pemberian angket validasi. Pada tahap validasi, dengan hasil validasi oleh ahli materi dengan skor 4,6 dan validasi oleh ahli produk audio visual (VCD) dengan skor 4,4. Dari skor tersebut produk bahan ajar dalam bentuk audio visual (VCD) mendapat kategori sangat baik diterapkan untuk siswa kelas X Sekolah Menengah Atas di Sumatera Utara.Kata Kunci: Pengemasan, Bahan Ajar, Audio Visual (VCD)

Tapa Pepe merupakan sebuah metode penyatuan antara Atman dengan Brahman melalui metode meditasi dengan memusatkan kepada sumber matahari. Tapa Pepe memfokuskan konsentrasinya pada “Sapta Cakra” yang ada dalam tubuh manusia. Untuk mengaktifkan cakra-cakra tersebut dengan tujuan mendekatkan diri dengan Tuhan. Ketika cakra-cakra dalam tubuh manusia terbuka, maka manusia akan banyak mendapatkan manfaat baik secara medis dan non medis, sekala dan niskala. Tapa Pepe merupakan tradisi Hindu yang telah dilaksanakan para leluhur pendahulu kita di tanah Jawa sebagai cara mendekatkan diri dalam proses menyatu dengan alam yang tiada lain adalah Tuhan itu sendiri sesuai dengan tujuan agama Hindu yaitu “mokshartam jagadhita ya ca iti dharma” dimana proses penyatuan diri seseorang melalui laku tertentu ibarat seperti seseorang melaksanakan laku upawasa (puasa) yang juga secara otomatis juga menyucikan dirinya sebagai jalan kepada Tuhan. Ritual Tapa Pepe yang dilakukan oleh Umat Hindu Jawa di Pura Mandira Seta Kraton Surakarta tidak jauh berbeda dengan ritual tapa pepe dibeberapa tempat. Perbedaanya adalah tempat pelaksanaannya dimana pada umumnya dilakukan di tempat terbuka, halaman rumah dan lain-lain. Berbeda dengan Tapa Pepe yang berada di Pura Mandira Seta dilakukan di dalam areal pura, dimana laku tapa pepe ini dilakukan di pelataran mandala tengah pura atau madya mandala, tepatnya di pelataran Arca Bhatari Dhurga Mahisasura Mardini. Tujuan dari Tapa Pepe di Pura Mandira Seta yaitu menyatukan Atman dengan Brahman yang berada dalam diri melalui metode menatap matahari sebagai sarana konsentrasinya. Konsentrasi pada Tapa Pepe atau Yoga Matahari berpusat pada cakra-cakra yang ada dalam tubuh. Tujuh cakra itu yaitu : Cakra Sahasrara (Cakra Mahkota), Ajna Cakra (Cakra Mata Ketiga/kening), Wisudha Cakra (Cakra Tenggorokan), Anahata Cakra (Cakra Jantung), Manipura Cakra (Cakra Pusar), Swadistana Cakra (Cakra Kelamin), Muladara Cakra (Cakra Tulang Ekor). Ketika cakra-cakra itu aktif, maka seseorang yang melakukan Tapa Pepe akan mendapatkan banyak manfaat seperti tidak mudah sakit karena menyerap energi matahari terbukti sekarang dilakukan banyak orang dimasa pendemi covid 19 ini.

The making and gifting of koloa faka-Tonga (women’s valuables, also known as koloa) has been an integral part of Tongan identity and culture for as long as anyone can remember. Working collaboratively on tapa and ngatu provides space for discussions and understanding, and reflections of Tongan identity and culture. The practice of gifting koloa has continued outside of the Kingdom of Tonga as Tongans migrate for new opportunities, including Aotearoa New Zealand (henceforth Aotearoa NZ). Using two different examples, this article will explore, through talatalanoa, how Tongan family partnerships working with tapa/ngatu in contemporary ways are sites of intergenerational knowledge sharing through art practices in Aotearoa NZ. Sulieti Burrows and Tui Emma Gillies are a mother-daughter partnership of tapa artists who work and reside in Aotearoa NZ and use their time together to share stories and make tapa works depicting what concerns them in contemporary society. Also residing in Aotearoa NZ, Lavinia and Fire Fonua are a mother-son partnership who turn koloa into contemporary personal adornment, alongside Fire’s wife, Sonia. In this article, the four Tongan authors’ reflections on inter-generational knowledge sharing and practices related to koloa making and gifting are described and illustrated using examples of their work. Their diverse stories also reflect on Tongan material culture, and Tongan identity, and demonstrate how working on practices that centre koloa provides opportunities to consider what it means to be Tongan in Aotearoa NZ, and how Tongan ways of being, knowing, and doing are valued as tu’atonga.

Abstract TAPA-1 is a 26-kDa integral membrane protein expressed on many human cell types. Antibodies against TAPA-1 induce homotypic aggregation of cells and can inhibit their growth. The murine homologue of TAPA-1 was cloned from both cDNA and genomic DNA libraries. A very high level of homology was found between human and mouse TAPA-1. The 5' untranslated region of the TAPA-1 gene resembles housekeeping gene promoters with respect to G + C content and the presence of potential Sp1 binding sites. The chromosomal localization of human and murine TAPA-1 genes was determined by Southern blot experiments using DNA from somatic cell hybrids. The genes were found to be part of a conserved syntenic group in mouse chromosome 7 and the short arm of human chromosome 11. The organization of the TAPA-1 gene and the projection of the exon boundaries on the proposed protein structure are presented.

: Arjuna Tapa New Creation Dance is a new creation inspired by Arjuna Tapa's story. It is about the sending of Arjuna by Yudhisthira to meditate to Mount Indrakila in the hope of obtaining a powerful weapon, which can be used in the face of a major war against the Kauravas. In the hermitage Arjuna received many temptations such as the temptation of a number of angels from heaven, the temptation of the big pig incarnate Momosimuka, and the temptation of the hunter Kirata. But all the temptations can be overcome properly and finally because of Arjuna's determination, Lord Shiva gave Pasupati's Arrow as a gift that would later be able to destroy his enemies. This dance is performed by seven male dancers, carrying arrows. These seven dancers in certain scenes will be divided into two characters, namely five male characters who play the five pandavas, namely Yudistira, Bima, Arjuna, Nakula, and Sahadewa. While the other two dancers play female characters as Dewi Kunti and Dewi Drupadi. The creation process went through the stages of exploration, experimentation, and formation. The structure of the work consists of four parts, namely pepeson, pengawak, pengecet, and pekaad. The dance moves refer to the line dance movements. This dance is accompanied by a gamelan gong that lasts 12 minutes. The message conveyed in Arjuna Tapa New Creative Dance is that a true warrior will succeed in doing his job well and smoothly if he can overcome all the obstacles and temptations he faces in achieving his goals.

Koridor Kyai Tapa merupakan koridor jalan yang berpotensi sebagai koridor unggulan Jakarta Barat, karena posisinya yang strategis menghubungkan wilayah Jakarta Barat dengan Jakarta Pusat. Pada kondisi eksisting koridor Kyai Tapa, terdapat beberapa titik ruang terbuka hijau yang bersifat pasif dan minim fasilitas yang bisa dimanfaatkan oleh masyarakat. Sehingga perlu dilakukan evaluasi pemanfaatan RTH pada koridor Kyai Tapa untuk mendukung peningkatan kualitas RTH Kota Jakarta. Metode penelitian yang digunakan adalah metode deskriptif dengan analisis kualitatif dan kuantitatif. Data dan analisis menunjukan pemanfaatan RTH Kyai Tapa masih rendah. 60% responden yang berkegiatan di RTH hanya sekedar lewat saja. Hal ini dipicu karena sangat minim atraksi dan penerangan pada malam hari sedangkan pengguna mulai aktif disekitar RTH pada sore hari. Pemanfaatan RTH pada koridor Kyai Tapa sangat perlu ditingkatkan mengingat potensi yang dimiliki dengan Penambahan fasilitas, atraksi dan peningkatan keamanan.

Adapun ilmu pengetahuan dan teknologi yang ditrasfer adalah: 1) Pelatihan pembuatan olahan ikan tapa seperti pembuatan Uta Dada Ikan Tapa dan Sambal Ikan Tapa, 2) Pelatihan cara pengasapan produk menggunakan sistem drum vertikal 3) Tata Cara produksi pangan yang baik, Manajemen Kewirausahaan dan Pengemasan. Untuk menunjang operasionalisasi kegiatan usaha kecil ikan tapa demi meningkatkan hasil produksi dan kualitas serta ketahanan kualitas, mitra diberi beberapa peralatan pengolahan diantaranya alat pengasapan drum vertikal dan renovasi rumah asap tersebut dapat memberikan kemudahan dan mengefisienkan waktu dalam hal pengolahan, selain itu mitra memperoleh pengetahuan mengenai perbedaan kualitas ikan asap yang dilakukan oleh mitra sebelumnya yaitu secara tradisional. Pengemasan ikan tapa merupakan bagian terpenting untuk menjaga kualitas dari olahan sehingga pengetahuan mengenai cara pengemasan yang baik juga diberikan kepada mitra yaitu dengan menggunakan alat pengemas (sealer). Pengolahan ikan tapa dengan beberapa variasi telah dilatihkan kepada Mitra antara lain sambal ikan tapa, utadada ikan tapa dan olahan lainya. Pemanfaatan olahan menjadi makanan ringan merupakan pengetahuan dan keterampilan baru bagi mitra yang diharapkan dapat meningkatkan pendapatan serta dapat diproduktifkan kinerjanya dalam usaha produksi rumahan yang sifatnya sambilan dengan mengolah menjadi beraneka ragam jenis produk camilan yang sifatnya inovatif untuk membuka lapangan pekerjaan baru dalam menunjang pemasukan kebutuhan sehari-hari. Prosedur dan tata cara produksi pangan yang baik juga dilatihkan kepada mitra yang diharapkan dapat menjadi bekal dalam pengurusan izin khususnya ikan tapa untuk menyatakan produk makanan yang diproduksi memenuhi standar keamanan pangan.

Abstract CD19 is a member of the Ig superfamily expressed on the surface of B lymphocytes that may be involved in the regulation of B cell function. Immunoprecipitation studies with B cell lines solubilized by digitonin have shown CD19 to be part of a multimolecular complex that includes CD21 (CR2) and other unidentified proteins. In this study, two of the CD19-associated proteins were identified as TAPA-1, which is expressed on most cell types, and Leu-13, which is expressed on subsets of lymphoid cells. TAPA-1 and Leu-13 are physically associated in many cell lineages. CD19 and CD21 mAb each specifically coprecipitated proteins of the same size as those precipitated by TAPA-1 and Leu-13 mAb from B cell lines and cDNA-transfected K562 cell lines. Western blot analysis with a TAPA-1 mAb verified the identity of TAPA-1 in CD19 and CD21 immunoprecipitated materials. In addition, when TAPA-1 or Leu-13 were crosslinked and patched on the cell surface, all of the CD19 comigrated with TAPA-1 and some of the CD19 comigrated with Leu-13. Furthermore, mAb binding to CD19, CD21, TAPA-1, and Leu-13 on B cell lines induced similar biologic responses, including the induction of homotypic adhesion, inhibition of proliferation, and an augmentation of the increase in intracellular [Ca2+] induced by suboptimal cross-linking of surface Ig on B cell lines. Together, these data suggest that TAPA-1 and Leu-13 are broadly expressed members of a signal transduction complex in which lineage-specific proteins, such as CD19 and CD21, provide cell-specific functions.

Abstract TAPA-1 is a transmembrane protein that has been shown to be involved in cell growth and cellular adhesion. Our studies were aimed at determining the mechanisms of the biologic phenomena mediated by TAPA-1, which include the identification of proteins that are associated with it on the surface of lymphocytes. We and others have previously shown that Leu-13, a leukocyte Ag, is one such molecule and that in B cells TAPA-1 is associated with the CD19 Ag. Herein we identify an additional molecule, HLA-DR, that is noncovalently associated on the surface of B cells with TAPA-1. This association was first detected by immunoprecipitation by anti-TAPA-1 and by anti-HLA-DR antibodies in the presence of mild detergents. The initial observation was confirmed by 2-dimensional SDS-PAGE and by direct identification of TAPA-1 in anti-HLA-DR immunoprecipitates by Western blot analysis. The association of the two molecules on the surface of a human B cell line was shown by cocapping experiments. In addition, antibodies to both molecules can induce cellular adhesion and an antiproliferative effect. Because the tissue distribution of these two molecules only partially overlaps, with TAPA-1 being expressed on most cell types and MHC class II expressed on a more restricted group of tissue, it is possible that the TAPA-1 molecule provides a basic function that can augment a cell type specific activity. In B cells the association of TAPA-1 with CD19 and HLA-DR may increase cellular interaction and play a supporting role in the transmission of specific signals.

Abstract C33 Ag and M38 Ag had been identified by mAb inhibitory to HTLV-1-induced syncytium formation. The cDNA encoding C33 Ag had revealed that it belongs to the newly defined transmembrane 4 superfamily (TM4SF). M38 Ag was detected on virtually all human cell lines and fresh leukocytes except for most granulocytes. It was also expressed on a mouse hybrid cell clone containing human chromosome 11q23-pter. Immunoprecipitation and immunoblot analyses identified a monomeric 26-kDa protein. The M38 epitope was dependent on S-S bonding. These characteristics were very similar to those reported for TAPA-1 (the target of antiproliferative antibody-1), which also belongs to TM4SF as C33 Ag. We therefore cloned the cDNA of human TAPA-1 and expressed it in COS cells. M38 indeed reacted with COS cells expressing human TAPA-1. We concluded that M38 Ag was identical to TAPA-1. To further investigate the biologic functions of C33 Ag and M38 Ag (TAPA-1) and their roles in HTLV-1-induced syncytium formation, molecules associated with these Ag were examined in T cells. Immunoprecipitation from surface-iodinated cell lysates revealed that proteins co-precipitated by C33 and M38 were mostly common including each other. Sequential immunoprecipitation-immunoblot experiments confirmed that C33 Ag and M38 Ag (TAPA-1) were associated with each other. The association was further confirmed in BHK cells doubly transfected with human cDNA for C33 Ag and TAPA-1. We extended similar analyses and found that C33 Ag and M38 Ag (TAPA-1) were regularly associated with CD4 or CD8. The association of these Ag on the cell surface was further supported by co-modulation of M38 Ag (TAPA-1), CD4 and CD8 with C33 Ag. This is the first time that a physical association between the members of TM4SF is demonstrated. Furthermore, the regular association of C33 Ag and M38 Ag (TAPA-1) with CD4 or CD8 might indicate that they play a role in expression and/or function of the CD4/CD8 co-receptor complex.

This study aims to determine the socio-economic activities of sand miners in Tapa, Kabila District, Bone Bolango Regency in 1998-2010. The method used in this study is the historical method. Steps such as heuristics, namely collecting and seeking information from secondary sources and secondary sources. Primary sources are obtained orally, such as interviews with informants who are directly involved in the sand miners in Tapa. Primary source The results of the study describe that the sand mining activities in Tapa that have been occurring so far are very closely related to the socio-economic conditions of the people who develop in the region. The monetary crisis in 1998 had a real impact on the community by dropping the prices of the plantation commodities they planted. The beginning of its emergence was indeed as a response from the community from economic uncertainty. Entering the 2000s, sand mining activities experienced a peak with many transport trucks operating in a day. This phenomenon indicates that the community is highly dependent on this extractive activity. in 2008 the sand miners moved to the GORR bridge which is now in Tapa. The miners used traditional tools until 2008, from 2008 to 2010. They used electronic devices, namely using machines to take sand from the river.

Adapun tahapan metode penciptaan dalam mewujudkan konsep tersebut ialah tahap penjajagan (Eksplorasi), tahap percobaan (Improvisasi), dan tahapan pembentukan (Forming). Karya yang dihasilkan dari konsep tersebut adalah karya seni pewayangan modern yang mengangkat cerita "Bima Tapa Agni". Dalam karya ini, penggarap menggabungkan unusr wujud wayang tradisional Bali dengan teknologi layar lebar yang memperlihatkan latar belakang yang menarik dan teampak realistik. Proses kreatif dalam pembuatan karya ini melibatkan kolaborasi antar beberapa pemain wayang (dalang), musisi, penata suara, dan teknisi layar lebar. Mereka menggunakan berbagai teknik dan pendekatan untuk menghasilkan visual yang sesuai dengan tema cerita yang diangkat. Para dalang juga menggabungkan unsur-unsur modern, seperti penggunaan media elektronik dalam memunculkan screen latar kekinian pada media layar lebar dan teknik bermain wayang yang tidak biasa, dengan unsur-unsur tradisional pewayang gaya Bali, dalam menciptakan sebuah karya yang unik dan menarik. Secara keseluruhan, karya ini menceritakan tentang perjalanan lanjutan dari kisah Bima Nawa Ruci, di mana Bima melakukan tapa brata dengan alasan untuk mencari jatidirinya yang dilakukan di Gunung Pastika dengan sarana api.

Jugi Tapa saga is oral literature in Sawang District. North Aceh Regency, Aceh Province. This saga tells the story of the main character named Leubee Muda. Then, Leubee Muda became a heretic because he opposed his teacher named Teungku Lhok Drien. After his heresy, he was called Jugi Tapa, who became the title of this saga.One day, his teacher asked him to deliver a book to Teungku Jambo Haji. The teacher advised him not to read the book. However, Leubee Muda violated the mandate of his teacher, he read and practiced the entire contents of the book. Because of his intelligence, in a short time, he can master all the knowledge contained in the book.To perfect his magic, he must meditate and marry one hundred beautiful women, some of whom are king's wives and daughters of dignitaries. Jugi Tapa had married eighty-eight beautiful women. Amat Banta's mother, King Sadon's wife from Kuala Diwa, was his 99th wife. He needs one more woman as his hundredth wife as a condition of perfecting his power. Before obtaining a hundred women, he could not have sex with a woman, and he continued to meditate in the palace which he made with magic.His 99th wife was Putro Bunsu, the wife of King Sadon of the Kingdom of Kuala Diwa, having a son named Amat Banta. When Jugi Tapa brought his mother, Amat Banta was three days old. The Mother left a ring in the swing of her little son. Later that ring was the proof that he was her son she had left behind.Based on information from Bing Pho in Indonesian called Kepiting Uca (fiddler crab), Amat Banta looked for his mother to Blang Laka (in the current known as Bener Meriah Regency), where Jugi Tapa collected ninety-nine women, including Amat Banta's mother. With the help of Amat Banta ’s mother who had obtained the secret of Jugi Tapa's magic from Jugi Tapa's mouth, Amat Banta managed to kill Jugi Tapa.Several messages can be taken from the story of Jugi Tapa, both as symbols, indexes, and icons. First, good deeds do not always get good replies. Therefore, every good work must be done sincerely so that we are not disappointed later on. Second, women should not be kept secret. Teknik pengumpulan data dalam penelitian ini dengan menggunakan tes praktek atau perbuatan dan pengukuran, yang diawali dengan pretest dengan instrumen menggunakan Tes Kesegaran Jasmani Indonesia (TKJI) untuk usia anak 10-12 tahun, setelah diberikan pretest kemudian siswa diberikan perlakuan berupa pembelajaran permainan tradisional tapa gala setlah mendapatkan perlakuan siswa diberikan postest berupa tes yang sama dengan pretest yaitu tes TKJI. Teknik analisis data menggunakan uji t. Hasil penelitian menunjukkan tingkat kesegaran jasmani siswa kelas V SDN Sumi 1 setelah mendapatkan pembelajaran dengan permainan tradisional tapa gala meningkat sebesar 10,48% dari saat pretest. Analisis uji-t menunjukkan bahwa t hitung lebih besar dari t tabel, hal ini mengindikasikan bahwa terdapat perbedaan yang signifikan antara pre test dan post test, sehingga hal ini menunjukkan bahwa terdapat peningkatan yang signifikan kesegaran jasmani siswa kelas V SDN Sumi 1 setelah diberikan pembelajaran melalui permainan tapa gala. Pada saat pre test besarnya rata-rata adalah sebesar 13,83 dan nilai rata-rata untuk data post test adalah sebesar 15,28. Hasil ini menunjukkan tingkat imunitas tubuh siswa kelas V SDN Sumi 1 setelah belajar dengan permainan tradisional tapa gala meningkat sebesar 1,45 atau sebesar 10,48% dari saat pre test. Dalam hal ini dapat dikatakan peningkatan kesegaran jasmani siswa setelah pembelajaran dilaksanakan melalui permainan tapa gala adalah sebesar 10,48%. Simpulan dalam penelitian ini menunjukkan bahwa setelah pembelajaran dengan permainan tradisional tapa gala terdapat peningkatan imunitas tubus siswa melalui permainan tradisional tapa gala siswa kelas V SDN Sumi 1

Sebuah fenomena tentang terapi untuk kesehatan banyak kita jumpai dalam kehidupan masyarakat kita. Salah satu dari berbagai metoda terapi tersebut adalah terapi menggunakan pasir panas (Tapa Sweda). Fenomena tersebut dijadikan objek penelitian dengan judul penelitian: Terapi Pasir Panas(Tapa Sveda) Untuk Kesehatan Perspektif Ayurweda. Penelitian ini menggunakan metode kwalitatif, dengan pendekatan Ayurweda. Ayurweda dipakai sebagai pendekatan karena usada Bali memiliki prinsip dasar pengobatan yang sama dengan Ayurweda yaitu dengan bahan alam. Dari hasil pembahasan terhadap rumusan masalah maka didapat simpulan bahwaTapa Sveda adalah sebuah terapi dengan menggunakan media pasir panas sebagai sebuah terapi berkeringat (Svedacaraniya cikitsita) yang bertujuan mengeluarkan ama (racun tubuh). Terapi pasir panas(Tapa sweda)dapat mengatasi beberapa penyakit, khususnya penyakit yang disebabkan oleh unsur vata dan kapha, yaitu penyakit yang disebut Ama Vata atau rematik akut.

Immunoprecipitation studies revealed that the antibody reacted with a 26-kilodalton cell surface protein (TAPA-1). A diverse group of human cell lines, including hematolymphoid, neuroectodermal, and mesenchymal cells, expressed the TAPA-1 protein. Many of the lymphoid cell lines, in particular those derived from large cell lymphomas, were susceptible to the antiproliferative effects of the antibody. TAPA-1 may therefore play an important role in the regulation of lymphoma cell growth. A cDNA clone coding for TAPA-1 was isolated by using the monoclonal antibody to screen an expression library in COS cells. Analysis of the deduced amino acid sequence indicated that the protein is highly hydrophobic and that it contains four putative transmembrane domains and a potential N-myristoylation site. TAPA-1 showed strong homology with the CD37 leukocyte antigen and with the ME491 melanoma-associated antigen, both of which have been implicated in the regulation of cell growth.

ABSTRACT Porphyromonas gingivalis is one of the most etiologically important microorganisms in periodontal disease. We found in a previous study that PG1385 (TprA) protein, a tetratricopeptide repeat (TPR) protein, was upregulated in P. gingivalis wild-type cells placed in a mouse subcutaneous chamber and that a tprA mutant was clearly less virulent in the mouse subcutaneous abscess model (M. Yoshimura et al., Oral Microbiol. Immunol. 23:413-418, 2008). In the present study, we investigated the gene expression profile of tprA mutant cells placed in a mouse subcutaneous chamber and found that 9 genes, including PG2102 (tapA), PG2101 (tapB), and PG2100 (tapC) genes, were downregulated in the tprA mutant compared with those in the wild type. Expression of a cluster of tapA, tapB, and tapC genes of the mutant was also downregulated in an in vitro culture with enriched brain heart infusion medium. The TprA protein has three TPR motifs known as a protein-protein interaction module. Yeast two-hybrid system analysis and in vitro protein binding assays with immunoprecipitation and surface plasmon resonance detection revealed that the TprA protein could bind to TapA and TapB proteins. TprA and TapB proteins were located in the periplasmic space, whereas TapA, which appeared to be one of the C-terminal domain family proteins, was located at the outer membrane. We constructed tapA, tapB, and tapC single mutants and a tapA-tapB-tapC deletion mutant. In the mouse subcutaneous infection experiment, all of the mutants were less virulent than the wild type. These results suggest that TprA, TapA, TapB, and TapC are cooperatively involved in P. gingivalis virulence.

Purpose The purpose of this paper is to present a methodology based on the transactional asset pricing approach (TAPA) and to illustrate the application of TAPA within the context of professional property valuation. Design/methodology/approach The TAPA is a novel analytical valuation methodology recasting the traditional derivations of the income approach techniques, including DCF, from a transactional perspective based on the principle of inter-temporal transactional equity, instead of the conventional investor-specific view originating from I. Fisher (1907, 1930). Findings The authors present DCF analysis as a specific case of a more general TAPA approach to valuation under the income method. This also leads to novel analytical derivations of the Direct income capitalization, Gordon, Inwood, Hoskold and Ring models. Based on the TAPA framework, the authors also research the value-enhancing effects of benchmark market volatility on the subject property value and conclude that such effects can be statistically significant depending on the DCF analysis period. Research limitations/implications The research has a direct bearing on time-variable discount rate forecasting capabilities, as it uses a time-variant structure for the discount rates. Practical implications Using the US Case-Shiller and BLS rental indices as a valuation benchmark, the paper contains an example of applying the general TAPA framework to value a notional property under a TAPA’s DCF version. Such property valuations can be easily replicated in practice – especially in the context of equitable/fair value determination under the International Valuation Standards Council valuation standards. Social implications TAPA is a deductive principles-based theory of asset valuation especially fit for the transactional and illiquid asset valuation contexts – thus enabling a more efficient pricing for such assets in a sense of reflecting the transactional interests of the parties more closely than achievable under the conventional valuation methods. Originality/value TAPA is an original filiation of research with roots going as far back as Aristotelian Catallactics. It contains analytical formalizations of certain transactional equity principles.

This article discusses about the hidden meaning of tapa wuda that had performed by Kalinyamat Queen as a symbol of Islamic education. This ritualistic behavior recorded in the Babad Tanah Jawi text which is often invites controversy about its meanings. Many people interpret tapa wuda textually as "asceticism without wearing clothes". Scientific studies with a hermenautic approach will reveal what means in this fragment of the Babad Tanah Jawi text. The method that is used in this journal is a descriptive qualitative method which will present data and findings in the form of descriptive writing. The results show that tapa wuda is a symbol that has meanings of the importance of an educational environment, patience, asceticism, and strong determination and purpose in seeking knowledge to gain wisdom in thinking and acting. Thin anodic porous alumina (tAPA) was fabricated from a 500 nm thick aluminum (Al) layer coated on silicon wafers, through single-step anodization performed in a Teflon electrochemical cell in 0.4 M aqueous phosphoric acid at 110 V. Post-fabrication etching in the same acid allowed obtaining tAPA surfaces with ≈160 nm pore diameter and ≈80 nm corresponding wall thickness to be prepared. The tAPA surfaces were made SERS-active by coating with a thin (≈25 nm) gold (Au) layer. The as obtained tAPA–Au substrates were incubated first with different thiols, namely mercaptobenzoic acid (MbA) and aminothiol (AT), and then with phospholipid vesicles of different composition to form a supported lipid bilayer (SLB). At each step, the SERS substrate functionality was assessed, demonstrating acceptable enhancement (≥100×). The chemisorption of thiols during the first step and the formation of SLB from the vesicles during the second step, were independently monitored by using a quartz crystal microbalance with dissipation monitoring (QCM-D) technique. The SLB membranes represent a simplified model system of the living cells membranes, which makes the successful observation of SERS on these films promising in view of the use of tAPA–Au substrates as a platform for the development of surface-enhanced Raman spectroscopy (SERS) biosensors on living cells. In the future, these tAPA–Au-SLB substrates will be investigated also for drug delivery of bioactive agents from the APA pores.

This research was conducted at SMPN X Tapa. The purpose of this research was to determine the relationship between self-concept and coping mechanisms in adolescent smokers at SMPN X Tapa. The research method uses an analytical design with a cross sectional approach. Cross sectional survey is a study to study the dynamics of the correlation between risk factors and effects, by approach, observation or data collection all at once (point time approach). The sample of this research was 88 students and the sampling was done by using purposive sampling technique. Purposive sampling is a sampling technique with certain considerations. Where the probability value is in the range of 0.088 (> 0.05) and the correlation coefficient is -0.183. This research shows that the self-concept of adolescent smokers at SMPN X Tapa is mostly in the moderate category, namely 53 people, while the coping mechanisms of adolescent smokers at SMPN X Tapa are classified as maladaptive, namely 48 people. It can be concluded about the data analysis carried out between self-concept variables and coping mechanisms in adolescents at SMPN X Tapa, where the probability value is in the range of 0.088 which means > 0.05 and the correlation coefficient is -0.183. The results of the study, seen from the existence of a Spearmen rank correlation hypothesis test, showed that there was no negative relationship between self-concept and coping mechanisms in adolescent smokers at SMPN X Tapa.
