Abstract:
DNA binding with one finger (Dof) proteins are plant-specific transcription factors with crucial roles in plant growth and stress response. Even so, little is known about them in wheat. In this study, 108 wheat Dof (TaDof) genes across 21 chromosomes were detected. Although variable in sequence length, molecular weight, and isoelectric point, all TaDof proteins contained conserved zinc-finger structures and were phylogenetically divided into 7 sub-groups. Exon/intron and motif analyses suggested that TaDof structures and conserved motifs were similar within sub-groups but diverse among sub-groups. Many segmental duplications were identified and Ka/Ks and inter-species synthetic analyses indicated that polyploidization was main reason for increased number of TaDofs. Prediction and experimental confirmation revealed that TaDofs functioned as transcription factors in the nucleus. Expression pattern profiling showed that TaDofs specifically affected growth and development, and biotic and abiotic stress responses. Wheat miRNAs and cis-regulator were predicted as essential players in molding TaDofs expression patterns. qRT-PCR analysis revealed that TaDofs were induced by salt and drought stresses. Customized annotation revealed that TaDofs were widely involved in phytohormone response, defense, growth and development, and metabolism. Our study provided a comprehensive understanding to wheat TaDofs.

Abstract:
Although results obtained with the Tadoma method of speechreading have set a new standard for tactual speech communication, they are nevertheless inferior to those obtained in the normal auditory domain. Speech reception through Tadoma is comparable to that of normal-hearing subjects listening to speech under adverse conditions corresponding to a speech-to-noise ratio of roughly 0 dB. The goal of the current study was to demonstrate improvements to speech reception through Tadoma through the use of supplementary tactual information, thus leading to a new standard of performance in the tactual domain. Three supplementary tactual displays were investigated: (a) an articulatory-based display of tongue contact with the hard palate; (b) a multichannel display of the short-term speech spectrum; and (c) tactual reception of Cued Speech. The ability of laboratory-trained subjects to discriminate pairs of speech segments that are highly confused through Tadoma was studied for each of these augmental displays. Generally, discrimination tests were conducted for Tadoma alone, the supplementary display alone, and Tadoma combined with the supplementary tactual display. The results indicated that the tongue-palate contact display was an effective supplement to Tadoma for improving discrimination of consonants, but that neither the tongue-palate contact display nor the short-term spectral display was highly effective in improving vowel discriminability. For both vowel and consonant stimulus pairs, discriminability was nearly perfect for the tactual reception of the manual cues associated with Cued Speech. Further experiments on the identification of speech segments were conducted for Tadoma combined with Cued Speech. The observed data for both discrimination and identification experiments are compared with the predictions of models of integration of information from separate sources.

Abstract:
In the Tadoma method of communication, deaf-blind individuals receive speech by placing a hand on the face and neck of the talker and monitoring actions associated with speech production. Previous research has documented the speech perception, speech production, and linguistic abilities of highly experienced users of the Tadoma method. The current study was performed to gain further insight into the cues involved in the perception of speech segments through Tadoma. Small-set segmental identification experiments were conducted in which the subjects' access to various types of artieulatory information was systematically varied by imposing limitations on the contact of the hand with the face. Results obtained on 3 deaf-blind, highly experienced users of Tadoma were examined in terms of percent-correct scores, information transfer, and reception of speech features for each of sixteen experimental conditions. The results were generally consistent with expectations based on the speech cues assumed to be available in the various hand positions.

Abstract Summary Proteins with highly similar tandem domains have shown an increased propensity for misfolding and aggregation. Several molecular explanations have been put forward, such as swapping of adjacent domains, but there is a lack of computational tools to systematically analyze them. We present the TAndem DOmain Swap Stability predictor (TADOSS), a method to computationally estimate the stability of tandem domain-swapped conformations from the structures of single domains, based on previous coarse-grained simulation studies. The tool is able to discriminate domains susceptible to domain swapping and to identify structural regions with high propensity to form hinge loops. TADOSS is a scalable method and suitable for large scale analyses. Abstract:
This study reports on the linguistic abilities of 3 adult deaf-blind subjects. The subjects perceive spoken language through touch, placing a hand on the face of the speaker and monitoring the speaker's articulatory motions, a method of speechreading known as Tadoma. Two of the subjects, deaf-blind since infancy, acquired language and learned to speak through this tactile system; the third subject has used Tadoma since becoming deaf-blind at age 7. Linguistic knowledge and productive language are analyzed, using standardized tests and several tests constructed for this study. The subjects' language abilities prove to be extensive, comparing favorably in many areas with hearing individuals. The results illustrate a relatively minor effect of limited language exposure on eventual language achievement. The results also demonstrate the adequacy of the tactile sense, in these highly trained Tadoma users, for transmitting information about spoken language sufficient to support the development of language and learning to produce speech.

Tadoba lake is the major lake in Tadoba Andhari Tiger Reservoir (TATR) in the Chandrapur district, in eastern Maharashtra which represent a unique habitat for wildlife in central India and oldest National Park of the state. The lake is free from human activities. The present investigation was carried out to evaluate the population of planktonic algae of the lake from June-2006 to May-2007. The study of phytoplanktonic diversity revealed the presence of 35 species belonging to four major groups, Chlorophyceae, Bacillariophyceae, Myxophyceae and Euglenophyceae among which Bacillariophyceae was dominant. The rich floral diversity indicates high productive nature of pond.

Abstract:
Blast disease resulting from Magnaporthe oryzae fungal infection reduces annual rice yield by up to 30% globally. The wheatwin2 (wwin2) is a pathogenesis-related (PR) gene that encodes for a PR-4 protein with chitinase properties that is capable of degrading chitin, a major constituent of certain fungal cell walls. However, the potential for wwin2 to contribute to M. oryzae resistance in rice is unclear. This study reports the construction of a pMDC140 vector carrying the wwin2 gene and its Agrobacterium-mediated transformation into the Tadong rice genome. In brief, the wwin2 gene was synthesized and integrated into a pMDC140 vector using Gateway cloning technology and was transformed into the Tadong rice genome. Our results show a promising high transformation rate, with more than 90% of the transformed rice calli expressing β-glucuronidase (GUS), the reporter gene marker. The expression of the wwin2 gene in transformed rice calli was further confirmed using quantitative real-time polymerase chain reaction. In conclusion, a pMDC140-wwin2 vector was constructed, which had a high transformation rate and could consistently induce expression of the GUS and wwin2 genes in Tadong rice. Data of this study is beneficial for subsequent in vitro and M. oryzae-infected field experiments to confirm the defense mechanism of the wwin2 gene towards blast disease in rice.

This study attempts to create a checklist of moths recorded from two different parts of Tadong in Sikkim, located in the northeastern Himalaya of India. Out of 160 photographed specimens of moths, 133 species were identified and classified. Sixteen families of moths were recorded out of which Erebidae (30.83%) had the highest number of species followed by Geometridae (24.81%), and Crambidae (18.05%) while the other families comprised of 26.30% of the total species.

Hydrothermal activity played an important part in the formation of the Ordovician dolostone reservoirs in Tadong area (eastern Tarim Basin). By applying the technologies of thin rock section microscopic identification, cathodoluminescence analysis, carbon and oxygen isotope analysis, trace element, rare earth element analysis, among others, the mineralogical characteristics and geochemical characteristics of the hydrothermal process are studied. It can have seen that the Ordovician hydrothermal process has represented by dolomite in various forms in the aspect of petrology and mineralogy. It is mainly composed of medium-megacryst crystalline dirty dolomite and powder-megacryst crystalline cleaner dolomite. The geochemical characteristics are the hydrothermal origin, medium-megacryst crystalline dirty dolomite with lower δ18O value, high Mn-Fe content, and positive Eu anomaly. The powder-megacryst crystalline has cleaner dolomite, with higher δ18O value, the Mn content low, and the negative Eu anomaly. Provide a reference for the Ordovician carbonate in Tadong area oil and gas exploration in the study.

The purpose of this research is to describe the efforts to maintain the Tado language in Lindu District, Sigi Regency as a form of preserving regional language education. This research method uses a descriptive qualitative approach. The data sources of this research are native speakers who use the Tado language with data collection techniques carried out are interviews, observation, and documentation. The data analysis technique used descriptive analysis. The results of this study indicate that the maintenance of the Tado language in Lindu District, Sigi Regency cannot be separated from the role of the community, namely with various creative efforts to maintain the Tado language, including carrying out cultural activities and also producing works using the Tado language with publish it on social media, the government also has a role in maintaining tado language by including tado language in local content subjects at school.

The population of this study were all students of class VII UPT.SMP Negeri 1 Laut Tador, totaling 130 students. While the sample used in this study was UPT.SMP Negeri 1 Laut Tador class VII-1 students, totaling 26 students. This study uses a type of quantitative research with a research design that is one-group pretest-posttest design. The data analysis technique used in this study is the normality test using the Kolmogorov Smirnov test and hypothesis testing using the t test. the data tested will be processed using the SPSS program. the results of the research that has been carried out show that the value of the g factor ranges from -10.75 (low category) to 49.5 (high category). With the percentage of the g factor which is in the low category which is 26.6% and the percentage of the g factor which is in the high category which is 73.3%. Whereas in the medium category and the false category is 0% and from the data it can be obtained that the average value of the g factor is 9.71%, so it is included in the high-quality category. So, it can be concluded that the use of the Two Stay Two Stray model is effective on student learning outcomes in Mathematics at UPT.SMP Negeri 1 Laut Tador TP. 2022/2023

Abstract Rice (Oryza sativa L.) is the most important staple food for many countries including Malaysia. But the country is still relying on the import to meet its domestic demand. Though significant public resources allocated for the rice industry, rice cultivation is still perceived to be uneconomical and associated with poverty. Therefore, a hydroponic system is seen as an alternative rice cultivation system that possibly be able to overcome common problems encountered in conventional and traditional paddy cultivation such as irrigation systems, limited cultivation area, control of nutrient inputs, control of pests, and loss of community interest particularly the new generation towards paddy cultivation. This net house study which one of the objectives was conducted to examine the vegetative growth of two local rice varieties under hydroponic condition. The experiment was a split plot design with nutrient solution concentration as the main plot, and the subplot is a local rice variety consisting of low land variety TQR-8 Sri Aman and upland rice variety called as ‘Tadong’. Each treatment was replicated in four plants or pots. A two-way analysis of variance (ANOVA) was performed to determine the effect of nutrient solution concentration and variety on rice growth cultivated using hydroponic system. The results revealed that the hydroponically grown rice of TQR-8 Sri Aman has greater tillering ability compared to Tadong for both NCSs. On the other hand, Tadong variety showed higher plant height than TQR-8 Sri Aman grown under hydroponic system. In addition, interaction of variety and nutrient solution concentration was significantly difference on tiller number and plant height. Further analysis should be conducted to evaluate the yield and feasible of the rice grown using hydroponic system.

Abstract:
Tarim Basin is the largest Petroliferous basin in China, while its shale gas development potential has not been fully revealed. The organic-rich black shale in middle Ordovician Heituao Formation from Tadong low uplift of Tarim Basin has been considered as an important source rock and has the characteristic of large thickness, high organic matter content and high thermal maturity degree. To obtain its development potential, geochemical, mineralogical and mechanics research is conducted based on Rock-Eval pyrolysis, total organic carbon (TOC), X-ray diffraction (XRD) and uniaxial compression experiments. The results show that: (1) the TOC content ranges between 0.63 and 2.51 wt% with an average value of 1.22 wt%, the Tmax values are 382–523 °C (average = 468.9 °C), and the S2 value is relatively low which ranges from 0.08 to 1.37 mg HC/g rock (averaging of 0.42 mg HC/g rock); (2) the organic matter of Heituao shale in Tadong low uplift show poor abundance as indicated by low S2 value, gas-prone property, and post mature stage (stage of dry gas). (3) Quartz is the main mineral component in Heituao shale samples, accounting for 26–94 wt% with an average of 72 wt%. Additionally, its Young’s modulus ranges from 20.0 to 23.1 GPa with an average of 21.2 GPa, Poisson’s ratio ranges between 0.11 and 0.21 (average = 0.15); (4) the fracability parameter of brittleness index (BI) ranges between 0.28 and 0.99 (averaging of 0.85), indicating good fracability potential of Heituao shale of Tadong low uplift and has the potential for shale gas development. This study reveals the shale gas accumulation potential in middle Ordovician of the Tarim Basin, and beneficial for future exploration and production practice.

In order to clarify the sedimentary development law under the Cambrian Ordovician regional stratigraphic framework in Tadong area, and lay a theoretical foundation for further oil and gas exploration in the study area. The distribution, characteristics and evolution of main sedimentary facies belts of Cambrian Ordovician are studied by means of drilling core observation, cast thin section identification, logging curve feature analysis, seismic profile and well connection profile. The results show that the Cambrian middle lower Ordovician in Tadong area is equivalent to a second-order sequence and can be further divided into 12 thirdorder sequences. Each third-order sequence is mainly composed of transgressive and highstand tracts. Carbonate platform margin beach facies and Reef (mound) beach complex facies are favorable reservoir development facies belts in this area; Under the regional stratigraphic framework, three types of sedimentary facies can be identified in Cambrian Ordovician, and a total of 10 subfacies are developed; The evolution of sedimentary facies is mainly controlled by the rise and fall of sea level, which is characterized by the migration of platform margin facies and the change of platform facies.

TAD1 (Triticum aestivum defensin 1) is a plant defensin specifically induced by low temperature in winter wheat. In this study, we demonstrated that TAD1 accumulated in the apoplast during cold acclimation and displayed antifungal activity against the pink snow mold fungi Microdochium nivale. When M. nivale was treated with TAD1, Congo red-stainable extracellular polysaccharides (EPS) were produced. The EPS were degradable by cellulase treatment, suggesting the involvement of β-1,4 glucans. Interestingly, when the fungus was treated with FITC-labeled TAD1, fluorescent signals were observed within the EPS layer. Taken together, these results support the hypothesis that the EPS plays a role as a physical barrier against antimicrobial proteins secreted by plants. We anticipate that the findings from our study will have broad impact and will increase our understanding of plant–snow mold interactions under snow.

d-Propranolol eliminates the increased adenine nucleoside release from hypoperfused hearts [R. D. Wangler, D. F. DeWitt, and H. V. Sparks, Am. J. Physiol. 247 (Heart Circ. Physiol. 16): H330-H336, 1984]. To determine whether d-propranolol reduces adenosine formation or adenosine release into the vascular compartment, we measured myocardial tissue adenosine (TADO). Decreased formation would lower TADO, whereas decreased release would elevate TADO. Reduction of perfusion pressure by 50% reduced coronary flow (CF), venous oxygen tension (PVO2), and myocardial oxygen consumption (MVO2) by approximately 40, 25, and 35%, respectively. Total adenosine and inosine released during 30 min of hypoperfusion increased 10- and 5-fold, respectively. Also, TADO increased from 2.68 +/- 0.37 to 5.17 +/- 0.67 nmol/g (P less than 0.05). In the presence of d-propranolol, the same reduction in perfusion pressure caused a similar decrease in CF and MVO2. d-Propranolol eliminated the release of adenosine and inosine associated with hypoperfusion. TADO after 30 min of hypoperfusion plus d-propranolol was not significantly increased (3.27 +/- 0.40 nmol/g) and was significantly less than hypoperfused hearts. When severe hypoperfusion was created by reducing perfusion pressure 75%, adenosine release still did not increase if d-propranolol was present. When adenosine release was plotted as a function of oxygen supply-consumption, they were related in a hyperbolic fashion. Despite the severity of hypoperfusion, in the presence of d-propranolol the supply-to-consumption ratio was similar to that of the control perfusion group (no drug). We conclude that d-propranolol blocks nucleoside formation during hypoperfusion by reducing oxygen demand such that a reduction of oxygen supply no longer stimulates adenosine formation.

Structured personal accounts of contemporary events and happenings composed in the Irish language are exceedingly rare in the pre-Revival period; that is to say, in the period prior to the founding of the Gaelic League or Conradh na Gaeilge in Dublin in 1893. The first such composition and the main focus of this paper is the account of the Ulster Earls’ journey from Rathmullan in Co. Donegal to Rome in 1607–8. It was written by Tadhg Ó Cianáin, a member of the native learned class, who accompanied the Earls on their journey. Tadhg Ó Cianáin’s account of the Earls’ journey to Rome includes many instances where specific reference is made to conversation as an accomplishment and pleasurable social activity. There would seem to be no doubt but that this reflects Ó Cianáin’s observation of a practice which was being cultivated on the Continent for societal and aesthetic purposes, and regarded as being conducive to family, kindred and community cohesion. Nonetheless, the examples of conversation-in-being culled from Irish literature more generally and cited in this article bear witness to the ongoing engagement with conversation, discourse and storytelling which has always permeated Irish culture.

Three <em>TaDL</em> homoeologous genes (<em>TaDL1</em>, <em>TaDL2</em>, and <em>TaDL3</em>) were isolated and found to be located on chromosomes 4AS, 4DL, and 4BL, respectively. The putative <em>TaDL1</em>, <em>TaDL2</em>, and <em>TaDL3</em> protein were 22.4, 22.6, and 22.3 kDa, with theoretical pI of 9.04, 9.04, and 8.98 and containing two distinct domains, namely, a zinc-finger domain in the N-terminal region and a C-terminal YABBY domain. The genome sequence of the three homologous genes consisted seven exons and six introns. All the introns, except for intron VI, varied in length and sequence composition. Phylogenetic analysis revealed that <em>TaDL</em> was most closely related to the <em>Brachypodium distachyon</em> <em>BdDL</em> and <em>Oryza sativa</em> <em>DL</em> genes. The expression pattern analysis indicated that <em>TaDL1</em>, <em>TaDL2</em>,<em> </em>and <em>TaDL3</em> have a higher expression levels in PS and P; however, the three genes are very rare in S. Thus, <em>TaDL</em> gene may contribute to pistil development. Moreover, overexpression of <em>TaDL</em> gene in stamens may cause complete or partial homeosis of stamens into pistils in wheat.</p>

ABSTRACT Toluate dioxygenase (TADO) of Pseudomonas putida mt-2 catalyzes the dihydroxylation of a broad range of substituted benzoates. The two components of this enzyme were hyperexpressed and anaerobically purified. Reconstituted TADO had a specific activity of 3.8 U/mg with m-toluate, and each component had a full complement of their respective Fe2S2 centers. Steady-state kinetics data obtained by using an oxygraph assay and by varying the toluate and dioxygen concentrations were analyzed by a compulsory order ternary complex mechanism. TADO had greatest specificity for m-toluate, displaying apparent parameters of KmA = 9 ± 1 μM, k cat = 3.9 ± 0.2 s−1, and K m O2 = 16 ± 2 μM (100 mM sodium phosphate, pH 7.0; 25°C), where K m O2 represents the K m for O2 and KmA represents the K m for the aromatic substrate. The enzyme utilized benzoates in the following order of specificity: m-toluate > benzoate ≃ 3-chlorobenzoate > p-toluate ≃ 4-chlorobenzoate ≫ o-toluate ≃ 2-chlorobenzoate. The transformation of each of the first five compounds was well coupled to O2 utilization and yielded the corresponding 1,2-cis-dihydrodiol. In contrast, the transformation of ortho-substituted benzoates was poorly coupled to O2 utilization, with >10 times more O2 being consumed than benzoate. However, the apparent K m of TADO for these benzoates was >100 μM, indicating that they do not effectively inhibit the turnover of good substrates.

Seán Ó Neachtain (c. 1640–1729) and his son Tadhg (c. 1671–c. 1752) were at the centre of an extensive circle of Gaelic scholars in the city of Dublin in the early part of the eighteenth century. Seán Ó Neachtain composed a broad range of creative literature. Although primarily written in Irish, his works include examples of Irish/English code-mixing as well as pieces composed entirely in English. His son, Tadhg Ó Neachtain, is credited with having written over 25 surviving manuscripts. He makes considerable use of English sources and of English itself in a number of these manuscripts, which are either pedagogical in nature, devoted to geography and history, or are characterised by frequent commonplace entries referring to contemporary events. This paper examines the interaction of the two languages in these manuscripts, exploring (1) the use of English language sources (textbooks and Dublin newspapers), (2) the content of the English portions of the manuscripts in question, and (3) the relationship of the English material to the Irish in the immediate compositional context. The paper seeks to assess whether the permeating bilingualism of these manuscripts is merely indicative of the contemporary socio-linguistic milieu in which the Ó Neachtains functioned, or can be regarded as harbinger of the subsequent community language change from Irish to English.

Budaya belis menjadi beban ekonomi bagi masyarakat karena sudah bergeser dari makna aslinya yang juga berpengaruh pada perubahan pembentukan karakter masyarakatnya. Tujuan penelitian ini adalah untuk mengetahui praktik pelaksanaan budaya belis, makna budaya belis dan nilai-nilai karakter yang terkandung dalam budaya belis. Penelitian ini menggunakan metode kualitatif deskriptif. Teknik analisis data dilakukan dengan cara reduksi data, penyajian data dan penarikan kesimpulan. Informan dalam penelitian ini adalah tokoh adat, tokoh masyarakat (guru), kepala desa, dan masyarakat yang pernah pernah terlibat langsung dalam pelaksanaan budaya belis. Tempat penelitian dilakukan di Desa Benteng Tado. Hasil penelitian menunjukan bahwa: proses pelaksanaan budaya belis dilakukan melalui 3 tahap yaitu (1) pra pernikahan: karong salang, cumang tau ata tua, tukar kila, turuk empo, (2) pernikahan: ngo ba paca, wagal, (3) pasca pernikahan: podo, curu/ roko, gerep ruha. Makna budaya belis bagi masyarakat desa Benteng Tado adalah sebagai bentuk penghargaan terhadap perempuan dan untuk membalas jasa orang tua dan keluarga perempuan. Dari hasil penelitian mengenai budaya belis sebagai kearifan lokal, teridentifikasi 18 nilai-nilai karakter kehidupan yang terkandung dalam budaya belis pada perkawinan adat di Desa Benteng Tado. Nilai-nilai kehidupan tersebut berketerkaitan dengan seluruh dimensi pembentuk karakter yaitu: nilai religius, nilai jujur, nilai toleransi, nilai disiplin, nilai kerja keras, nilai kreatif, nilai mandiri, nilai demokratis, nilai rasa ingin tahu, nilai semangat kebangsaan, nilai cinta tanah air, nilai menghargai prestasi, nilai bersahabat/komunikatif, nilai cinta damai, nilai gemar membaca, nilai peduli lingkungan, nilai peduli sosial, dan nilai tanggung jawab. This research was motivated by changes in the traditional marriage system that used belis in the village of Fort Tado. Belis culture is an economic burden for the community because it has shifted from its original meaning which also has an effect on changing the formation of the character of its society. The purpose of this study is to determine the practice of implementing belis culture, the meaning of belis culture and character values contained in belis culture. This research uses descriptive qualitative methods. Data analysis techniques are carried out by means of data reduction, data presentation and drawing conclusions. Informants in this study are traditional leaders, community leaders (teachers), village heads, and communities who have been directly involved in the implementation of belis culture. The place where the study was conducted in the Village of Benteng Tado. The results showed that: the process of implementing belis culture was carried out through 3 stages, namely (1) pre-marriage: karong salang, cumang tau ata tua, tukar kila, turuk empo, (2) marriage: ngo ba paca, wagal, (3) post-wedding: podo, curu/ roko, gerep ruha. Makna belis culture for the people of Fort Tado village is as a form of appreciation for women and to repay the services of women's parents and families. From the results of research on belis culture as local wisdom, 18 life character values contained in belis culture were identified in traditional marriages in Benteng Tado Village. These life values are related to all character-forming dimensions, namely: religious values, honest values, tolerance values, discipline values, hard work values, creative values, independent values, democratic values, the value of curiosity, the value of the spirit of nationality, the value of love for the homeland, the value of appreciating achievements, the value of friendly / communicative, the value of peace-loving, the value of love of reading, the value of caring for the environment, the value of caring for the environment, the value of respecting achievements, the value of friendly/communicative, the value of peace-loving, the value of love to read, the value of caring for the environment, the value of caring for the environment, the value of value social care, and the value of responsibility.
