MINNELLA, MARCO LORENZO.

Abstract:

Abstract:
We consider propagation of high-energy earth-skimming taus produced in interactions of astrophysical tau neutrinos. For astrophysical tau neutrinos, we take generic power-law flux, E-2 and the cosmogenic flux initiated by the protons. We calculate tau energy loss in several approaches, such as dipole models and the phenomenological approach in which parametrization of the F-2 is used. We evaluate the tau neutrino charged-current cross section using the same approaches for consistency. We find that uncertainty in the neutrino cross section and in the tau energy loss partially compensate giving very small theoretical uncertainty in the emerging tau flux for distances ranging from 2 to 100 km and for the energy range between 10(6) and 10(11) GeV, focusing on energies above 10(8) GeV. When we consider uncertainties in the neutrino cross section, inelasticity in neutrino interactions and the tau energy loss, which are not correlated, i.e. they are not all calculated in the same approach, theoretical uncertainty ranges from about 30% and 60% at 10(8) GeV to about factors of 3.3 and 3.8 at 10(11) GeV for the E-2 flux and the cosmogenic flux, respectively, for the distance of 10 km rock. The spread in predictions significantly increases for much larger distances, e.g., similar to 1, 000 km. Most of the uncertainty comes from the treatment of photonuclear interactions of the tau in transit through large distances. We also consider Monte Carlo calculation of the tau propagation and we find that the result for the emerging tau flux is in agreement with the result obtained using analytic approach. Our results are relevant to several experiments that are looking for skimming astrophysical taus, such as the Pierre Auger Observatory, HAWC and Ashra. We evaluate the aperture for the Auger and discuss briefly application to the other two experiments.

Abstract:
A search for the neutral Higgs bosons predicted by the Minimal Supersymmetric Standard Model (MSSM) is presented. The analysis is performed on data from proton-proton collisions at a centre-of-mass energy of sqrt{s}=8 TeV at the Large Hadron Collider recorded by the ATLAS detector. The data were collected in 2012, correspond to an integrated luminosity of 20.3 fb-1 and represent the full 8 TeV dataset. This search is performed in the tau-tau final state, with one tau lepton decaying leptonically and the other hadronically. The analysis is optimized in three categories, addressing low-mass Higgs bosons which are accompanied by a bottom quark induced jet or not and high-mass Higgs bosons. No deviations from Standard Model predictions are observed, and therefore no evidences of new Higgs bosons are found. Exclusion limits are set on the cross-section times branching fraction of the Higgs bosons and for parameters m_A and tan{beta} of MSSM benchmark scenarios

Abstract:
Metod: Sekundärdata samlades in från databasen PubMed.

Abstract:

Abstract:
In this dissertation we expand on the study of Τn-factorizations or generalized integer factorizations introduced by D.D. Anderson and A. Frazier and examined by S. Hamon. Fixing a non-negative integer n, a Τn-factorization of a nonzero nonunit integer a is a factorization of the form a = Λ.a1.a2...at where t ≥ 1, Λ= 1 or -1 and the nonunit nonzero integers a1,a2,...,at satisfy a1 ≡ a2 ≡ ... ≡ at mod n. The Τn-factorizations of the form a = a1,a2,...,at (that is, without a leading -1) are called reduced Τn-factorizations. While similarities exist between the Τn-factorizations and the reduced Τn-factorizations, the study of one type of factorization does not elucidate the other. This work serves to compare the Τn-factorizations of the integers with the reduced Τn-factorizations in Z and the Τn-factorizations in N. One of the main goals is to explore how the Fundamental Theorem of Arithmetic extends to these generalized factorizations. Results regarding the Τn-factorizations in Z have been discussed by S. Hamon. Using different methods based on group theory we explore similar results about the reduced Τn-factorizations in Z and the Τn-factorizations in N. In other words, we identify the few values of n for which every integer can be expressed as a product of the irreducible elements related to these factorizations and indicate when one can do so uniquely.

Abstract:
Mestrado em Biomedicina Molecular
Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by the presence of two histopathological hallmarks: the extracellular amyloid plaques (APs) composed of beta-amyloid protein (Abeta) and intracellular neurofibrillary tangles (NFTs), containing hyperphosphorylated tau protein. Therefore, Abeta and tau are important molecules associated with AD and evidence suggests that Abeta may initiate the hyperphosphorylation of tau, which by disrupting neuronal network leads to the process of neurodegeneration. In the present study, using rat primary cortical and hippocampal neuronal cultures, it was shown that exposure to aggregated Abeta1-42 for prolonged periods decreased tau phosphorylation at Ser202 and Thr205 residue, but in contrast increased at Ser262 residue. Tau hyperphosphorylation in AD may be related to alterations in signal transduction pathways involving tau phosphorylation, such as an imbalance in the regulation of protein kinases (PKs) and protein phosphatases (PPs). Thus it is also important to determine which specific PKs and PPs are involved in this process. We observed the involvement of PP1 in the dephosphorylation of tau at Ser202 and Thr205, and the involvement of PP1 and PP2A at the Ser262 residue. An important aspect of tau metabolism are its binding proteins, and to date many such proteins have already been described both in vitro and in vivo. The interactome of tau is shaped by its phosphorylation and so is crucial to map the crosstalk between normal and pathologically hyperphosphorylated tau. By co-immunoprecipitation we intend to identify proteins that interact with tau and more specifically with phosphorylated tau (p-Tau). Furthermore the effect of Abeta on this interactome should be forthcoming, which is relevant for AD tau pathology.

Dickson, John Robert. "The Role of the Human Tau 3'-Untranslated Region in Regulating Tau Expression." Thesis, Harvard University, 2013. http://dissertations.umi.com/gsas.harvard:11070.

Full text
Abstract:
The microtubule-associated protein tau forms pathological neuronal filaments in Alzheimer's disease (AD) and other neurodegenerative disorders, known collectively as tauopathies. Previous studies in transgenic mouse models of AD suggest that reducing tau expression may be safe and beneficial for the prevention or treatment of AD and possibly other tauopathies. As a first step toward identifying novel therapeutic strategies to reduce tau levels, the studies presented in this dissertation aim to investigate the role of the human tau 3'-untranslated region (3'-UTR) in regulating tau expression. Tau expresses two 3'-UTR isoforms, long and short, as a result of alternative polyadenylation. The exact sequence of these two 3'-UTR isoforms was determined by rapid amplification of cDNA 3'-ends (3'-RACE), and the two 3'-UTR isoforms were cloned into a luciferase reporter vector. Using these reporter constructs, the expression of these isoforms was found to be differentially controlled in human neuroblastoma cell lines M17D and SH-SY5Y by luciferase assays and quantitative PCR (qPCR). Through an unbiased screen of tau 3'-UTR deletions and fragments using luciferase reporter constructs, several regions in the long tau 3'-UTR isoform that contain regulatory cis-elements were identified. Additionally, several microRNAs were computationally identified as candidates that might bind the long tau 3'-UTR and thereby differentially control the expression of long versus short tau 3'-UTR isoforms. Screening these candidate microRNAs via luciferase reporter assay identified miR-34a, which was subsequently shown to repress the expression of endogenous tau protein and mRNA in M17D cells using Western blot and qPCR, respectively. Conversely, inhibition of endogenously expressed miR-34 family members leads to increased endogenous tau expression. Taken together, these studies suggest that the expression of the two tau 3'-UTR isoforms is differentially regulated and that this differential regulation is due to the presence of regulatory cis-elements found only in the long tau 3'-UTR isoform, including a binding site for miR-34 family members. Improved understanding of the regulation of tau expression by its 3'-UTR may ultimately lead to the development of novel therapeutic strategies for the treatment of Alzheimer's disease and other tauopathies.

Abstract:
The Compact Muon Solenoid (CMS) detector is located at the CERN Large Hadron Collider. It is a general purpose detector designed to test predictions of the standard model (SM) and search for the Higgs boson, as well as to look for signatures of new physics beyond the SM. The analyses described in this thesis use proton-proton collision data recorded by CMS during 2011 and 2012. Searches for neutral Higgs bosons decaying into tau pairs are presented. The searches are performed separately for the SM Higgs boson or the neutral Higgs bosons of the minimal supersymmetric standard model (MSSM). The MSSM searches consist of two different analyses. The first is a direct search for any one of the MSSM neutral Higgs bosons in the mass range from 90 GeV to 1 TeV decaying directly into taus. The second searches for the signature from a heavy neutral Higgs boson of the MSSM decaying into two light MSSM Higgs bosons with SM-like properties (H → hh) in the final state with two b-quarks and two taus. The SM and MSSM ττ analyses use 4.9 fb−1 of data collected at 7 TeV and 19.7 fb−1 collected at 8 TeV, while the MSSM H → hh analysis uses only the 8 TeV data. In the SM analysis, an excess of events in data above the background expectation is observed with an observed (expected) local significance of 3.2 (3.7) standard deviations at 125 GeV. This excess is found to be consistent with the 125GeV Higgs boson of the SM in mass and coupling strengths. No significant excess is observed in either MSSM search, and so upper limits are set on cross-section times branching ratio for the Higgs production processes at the 95% confidence level. Limits are also set in mA-tanβ parameter space of several interesting MSSM benchmark scenarios.

In this thesis, I perform a systematic resolution study for di-tau pairs at the Compact Muon Solenoid (CMS) experiment at the Large Hadron Collider (LHC) at CERN. I performed this analysis using Monte Carlo simulated events with a Higgs boson which decays into two tau particles with initial mass mH = 90 - 500 GeV/c ² . The two daughter taus were required to then decay leptonically. I determined the dependence of the input mass on the mass resolution and total energy contribution of the electron/muon visible mass peak. For mH 120 GeV/c², a mass resolution of =/< 25% and total energy fraction of =/> 50% can be achieved.

Abstract:
Immunotherapy for Alzheimer\'s disease has emerged as a promising approach for clearing pathological tau protein conformers. To explore this kind of treatment we tested an active immunization with pseudo-phosphorylated tau fragments in P301L tangle model mice that develop neuronal tau aggregates as observed in frontotemporal dementia and Alzheimer’s disease. We found that an immunization reduces neurodegeneration in α-motor neurons in the spinal cord and slows progression of the tangle-related behavioral phenotype. Performance on behavioral assays correlated with tau pathology at the corresponding spinal cord level. Interestingly, a slowed progression of these tauopathy related characteristics were only seen in mice that received a specific immunization with pseudo-phosphorylated tau fragments, not in animals that received a non-specific activation of the immune system. An immunization witch pseudo-phosphorylated tau fragments may be a valuable therapeutic option in targeting one of the major hallmarks of Alzheimer’s disease and frontotemporal dementia.

A measurement of standard model Higgs pair production in [beta][beta][tau][tau] final states at the High Luminosity LHC is investigated. Higgs pair production can be used to measure the Higgs trilinear coupling constant, which uniquely determines the shape of the Higgs potential. The doubly hadronic, hadron-muon, and hadron-electron di-[tau] final states are considered, with a shape analysis on either the stransverse mass (doubly hadronic) or a BDT discriminant (hadron-muon, hadron-electron) distribution performed to extract expected significances. The expected 95% CL upper limit on the cross section times branching ratio from a combination of all three channels is 2.2 times the SM value, with an expected +1[sigma] uncertainty on the measured cross section of 67%, indicating this measurement is feasible.

Tan, Heng-Ru May. "Neural representation of movement tau." Thesis, University of Edinburgh, 2008. http://hdl.handle.net/1842/2675.

Full text
Abstract:
A fundamental aspect of goal‐directed behaviour concerns the closure of motion‐gaps in a timely fashion. An influential theory about how this can be achieved is provided by the tautheory (Lee, 1998). Tau is defined as the ratio of the current distance‐to‐goal gap over the current instantaneous speed towards the goal. In this work we investigated the neural representation of tau in two sets of experiments. In one study we recorded neuromagnetic fluxes (using magnetoencephalography, MEG) from the whole brain of human subjects performing discrete hand movements aimed to targets in space, whereas the other study involved recordings of single cell activity from prefrontal and posterior parietal areas of a behaving monkey during geometrical shape‐copying tasks. These two studies provided complementary information, for the former covered the whole brain (at the cost of weak localization), whereas the latter used the finest neural grain (at the expense of limited brain regions). However, the two studies together yielded valuable information concerning the dynamic, time‐varying neural representation of tau, with respect to both integrated synaptic events in neuronal ensembles (recorded by MEG) and neural spike outputs (recorded by microelectrodes). The relations between neural signals and tau were analyzed using a linear regression model where the time‐varying neural signal (magnetic field strength in fT or spike density function) was the dependent variable and the corresponding value of movement tau and speed were the independent variables. In addition, the model included an autoregressive term to account for the expected correlated errors, given the time series nature of the data. The neurophysiological study revealed a statistically significant (p < 0.05) relation of spike density function to tau (in the presence or absence of a significant speed effect) in 17% of cells in the posterior parietal cortex (N = 399) and 8% of cells in the prefrontal cortex (N = 163). These results are in accord with previous findings in an interception task. The MEG study revealed that a mean of 21.98 (± 6.08) % of sensor signals had a statistically significant (p < 0.05) relation to tau across all subjects. These effects were distributed predominantly over the left parietal‐temporo‐occipital sensor space, with additional foci over the frontal sensorimotor regions. Altogether, these findings demonstrate a specific involvement of neurons and neuronal ensembles with the tau variable and pave the way for further studies on predictive tau control.

Gray, Will. "T.S. Eliot among the Metaphysicals." Thesis, University of St Andrews, 2011. http://hdl.handle.net/10023/3123.

Full text
Abstract:
Eliot's admiration for the poetry of the seventeenth century is well known. However, the several documents that explore the subject (thinly scattered across decades) fail to constitute a full account. Drawing on manuscript and print sources, and tracing particularly Eliot's prose poetics, this thesis redresses the scholarly need for a nuanced account of Eliot's role among the Metaphysical poets. The relationship ran in both directions, most famously in Eliot's championing of the poets and his urging that they find a new readership. His part in the revival of Metaphysical poetry, though, has been greatly exaggerated and the record is here faithfully adjusted. He was not in any way responsible for that revival, though he is its most important product, as is shown by a careful reconstruction of turn-of-the-century transcontinental publishing and reception. Eliot's criticism tells its own, largely unexplored story about the Metaphysicals and their influence on his critical and poetic sensibility. Most scholars, for instance, know that Eliot loved Donne, but few know the origin of that interest, let alone its brief nature or the personal reasons that drove him to appreciate the poet's audacity. Most also know the Modernist dicta of Tradition, objective correlative and the dissociation of sensibility, but not the fact that each owes something to Eliot's thinking about Donne. Engaging with Harvard class notes, under-consulted textbooks and a close study of Eliot's articles from the 1910s, two separate chapters investigate his education and early prose, along with their delicate dance between impersonality and confessional criticism. 1921-1926 marks a crucial stage in Eliot's writing, both for his poetry and his criticism. The Metaphysicals provide the clearest barometer of that change as well as the space where he approached conversion. This thesis is the first to trace the poets throughout Eliot's criticism, one of the first to engage with his Metaphysical-themed Clark Lectures, and the first to move far past Eliot's conversion, interpreting George Herbert as typical of his late mindset. In 1961 Eliot claimed no one had been as influenced by the Metaphysical poets as he had been. What this thesis offers is not only a more nuanced portrait of that influence but also a glimpse into the educational, critical and reading cultures of the early 1900s.

Abstract:
The present thesis is about aspects of Wagnerism in the works of T.S. Eliot, in terms of both influence and affinities. The opening chapter offers a brief historical background to Eliot's familiarity with and relation to Wagner, as well as an account of the principal issues to be discussed in the following chapters. The two artists' volumes of criticism mostly show in theory how they work in practice. One key theme there is the maintenance of the idea of tradition and at the same time that of revolutionizing the arts (poetry, opera and drama). Eliot's interest in music, notably in the use of the Wagnerian leitmotif, is one of the highlights of this study. Then, Eliot's concern for myths: his 'mythical method' is discussed in a separate chapter for comparison with Wagner's way of handling myths in his operas. Other important topics feature drama, the Greeks and Shakespeare, and poetry, especially Dante's, the Romantics' and that of the French Symbolists. Certain themes common in both (like salvation and love) are tackled in more than one chapter because of their relevance throughout, but a selection of motifs is singled out in a separate chapter. The study also investigates Eliot's and Wagner's relation to art and life from religious (Buddhism and Christianity) and philosophical (Schopenhauer and Nietzsche) paints of view. Where relevant, some biographical data shedding light on their arts are touched upon--e.g. their personal (including marital) experiences and their anti-Semitism. The concluding chapter rounds off the subject by mainly offering some possible reasons for Eliot's obscure and neglected Wagnerism.

Abstract:
The first article proposes a new name for the Southern Khoisan family, based on the fact that all sufficiently attested languages show some reflex of the noun *tuu 'people'. This is a more suitable alternative to previous terms, because it not only unambiguously identifies the genealogical unit and is in line with established conventions for classificatory nomenclature, but also avoids several drawbacks of other terms, among them the heretofore unproven idea of a genealogical unit Khoisan. The second article gives more substantial and systematic evidence that Tuu alias Southern Khoisan itself is in fact a coherent genealogical entity. It first outlines basic structural features of Tuu languages showing that they constitute a robust and typologically fairly distinct language type. It goes on to show that this is associated with a sufficient amount of sound-meaning correspondences, in both grammar and lexicon, in order to warrant an interpretation in terms of inheritance from a common ancestor language. Both studies are the result of work carried out in the project 'Genetic and typological profile of the Tuu language family (alias Southern Khoisan): cataloguing and linguistic analysis of existing sources'. Full text
Abstract:
The IceCube neutrino observatory at the South Pole has confirmed the existence of a diffuse astrophysical neutrino flux. The flavor composition of astrophysical neutrinos carries information on the environments at the sites of cosmic particle acceleration as well as potential imprints of new physics acting during neutrino propagation. To measure the flavor composition the observation of the long-elusive tau neutrinos is required. Starting at an energy of ~O(100 TeV) a tau neutrino charged current interaction can produce a double cascade topology, where the two energy depositions from the tau creation and the tau decay vertices are resolvable. This topology together with the well-established track and single cascade topology is used to measure the flavor composition on Earth. In this work, high-energy events starting in IceCube's detector volume are classified algorithmically into the three topologies. In the dataset with a livetime of 7.5 years, two events are classified as double cascades for the first time, yielding multi-TeV tau-neutrino candidates. The properties of the two tau-neutrino candidates are investigated in an a-posteriori analysis. The statistical method is improved by performing a log-likelihood-ratio test using multi-dimensional probability densities. One of the double cascades is consistent with being a misclassified single cascade, while the second double cascade is found to have a misclassification probability of only 3%. The measured flavor composition nu_e:nu_mu:nu_tau = 0.20:0.39:0.42 is consistent with astrophysical neutrinos and with previously published results. The astrophysical tau-neutrino flux is measured to dPhi / dE=3.0 (-1.8,+2.2) (E / 100TeV)^(-2.87) 10^(-18) GeV^(-1) cm^(-2) s^(-1) sr^(-1) with spectral index gamma=2.87 (-0.20,+0.21), yielding the first non-zero results for the tau normalization. The absence of an astrophysical tau-neutrino flux is disfavored at 2.8 sigma.

Toda, K. "T.S. Eliot and Renaissance drama." Thesis, University College London (University of London), 2015. http://discovery.ucl.ac.uk/1463322/.

Full text
Abstract:
The crucial importance of non-Shakespearian Renaissance dramatists to T.S. Eliot is evident in both his poetry and his prose. Eliot himself drew attention to this: he credited his own ‘poetic formation’ to the ‘minor Elizabethan dramatists’, and when reviewing his ‘critical output for the last thirty-odd years’ in 1951, he confessed himself ‘surprised to find how constantly I have returned to the drama, whether by examining the work of the contemporaries of Shakespeare, or by reflecting on the possibilities of the future.’ As C.S. Lewis disapprovingly wrote, Eliot’s ‘sympathy with depraved poets (Marlowe, Jonson, Webster) is apparent’. This thesis will trace Eliot’s engagement with these dramatists; it is a topic that has been comparatively neglected despite the central role it played in the evolution of his poetic and critical sensibility. The first section, which is largely biographical, explores Eliot’s background and education for clues to the development of his great interest in Renaissance drama, as well as detailing the ways in which he pursued this interest when he moved to England. The second comprises a detailed study of Eliot’s many essays on Renaissance dramatists. The last section examines his poetry, from the juvenilia to The Waste Land of 1922, when the outward signs of his ‘saturation’ were particularly prominent. In my conclusion I discuss how his engagement with Renaissance drama evolved in his post-1922 poetry and culminated in the composition of his own verse plays. The aim of this thesis is to explore the nature of the ‘profound kinship’ Eliot shared with Renaissance dramatists; their work appealed to him because it combines erudition with emotion, refinement with savagery, levity with the macabre and squalid. This appeal was so strong that it powerfully shaped both his poetic ideals and his vision of modernity.

Abstract:

If Swedish companies are to compete with companies in low-cost countries they have to achieve a more efficient production. The purpose of Total Productivity Maintenance (TPM) is to increase the company's profitability by increasing the productivity. TPM is a method that involves all employees and aims to prevent failures in order to increase the availability of existing equipment.

The aim of this paper is to evaluate the implementation process of TPM and give recommendations for the further implementation process. The conclusion and the analysis are based on a case study done at a production department at ABB Cewe-Controls facilities in Nyköping. The case study consists mainly of qualitative data from participation observations, surveys and interviews.

Commitment of the management is a key factor in the implementation process of TPM. The management of ABB Cewe-Control need to demonstrate their commitment in order for a change to happen. The case study shows that the commitment of both employee and management must be raised if the implementation process is to be successful. To avoid unnecessary conflicts or irritation in the further implementation process it is important to clarify responsibilities and roles in the implementation of TPU.

The case study has also shown that the training during the implementation process has not been sufficient. Proper training is needed to be able to conduct the implementation process of TPM in an effective manner. Proper training is also needed to raise the motivation of the staff.

Business Development is ABB’s methodology for the improvement work in the company. Business Development is a well-known concept in the organisation and has a well-defined connection with the company's strategic objectives. Because of this there is no reason why the improvement work and how it has been done should change. Business Development is a good foundation for the implementation of TPM and the improvement work in the future.

Abstract:
Thermoplastic polyurethane (TPU) containing hard and soft segments with variable molecular architecture are synthesized in two steps, the first one including a polyurethane pre-polymer. The microstructure of theses polymers shows phase separation occurring between soft and hard segments according to the nature of the segments. Hard segments can organize under two forms, either amorphous or crystalline. The morphology of TPU depends on the chemical structure of the hard segment involved. Incompatibility between soft and hard segments was highlighted from solubility parameters analysis, complete with characterization at various scales with microscopy (electron and AFM) and X-ray scattering technics. Theses thermoplastic polyurethanes are used to prepare bitumen-polymer blends. Interactions between the polymer segments and bitumen fractions are studied, considering solubility parameters of each of them and swelling measurements in order to study miscibility between the compounds. Model oils are used in the aims of mimicking some oily fractions of bitumen, and soft and hard segments are separately synthesized to study properties of each phases in the bitumen-polymer mixture. Multi-scale morphology of the blends is studied in connection with rheological properties and structure of the used polymer. The addition of polymer in bitumen allows to modify viscoelastic properties of bitumen beyond its glass transition due to the selective swelling of the polymer by the oily fractions. Having highlighted and analyzed the selective swelling by considering solubility parameters and interfacial tension measurements, we show that the presence of a continuous polymer-rich phase containing semi-crystalline hard segments in the blends leads to delay the flow of the bituminous material at highest temperatures. The oil composition of this polymer-rich phase will depend on its affinity with the fractions of bitumen and thus on the chemical structure of the polymer. In addition, semi-crystalline hard segment content of the polymer is also a key parameter allowing to adjust its affinity with bitumen and consequently on rheological properties of the bitumen-polymer mixture

"Differential induction and spread of tau pathology in young PS19 tau transgenic mice following intracerebral injections of pathological tau from Alzheimer’s disease or corticobasal degeneration brains." Doctoral thesis, Universitat de Barcelona, 2016. http://hdl.handle.net/10803/399674.

Full text
Abstract:
Filamentous tau pathologies are the hallmark lesions of several neurodegenerative tauopathies including Alzheimer’s disease (AD) and corticobasal degeneration (CBD). These diseases show cell type-specific and topographically distinct tau inclusions. Growing evidence supports templated transmission of tauopathies through functionally interconnected neuroanatomical pathways suggesting that different self-propagating strains of pathological tau could account for the diverse manifestations of neurodegenerative tauopathies. In the study, the rapid induction of tau pathology and the distinct cell type-specific spread of pathological tau following intracerebral injections of CBD or AD brain extracts enriched in pathological tau in human mutant P301S tau transgenic (Tg) mice (line PS19) is analysed. At 1 mo post-injection of extracts obtained from brains with CBD pathology (CBD-Tau), tau inclusions developed predominantly in oligodendrocytes of the fimbria and white matter near the injection sites with infrequent intraneuronal tau aggregates. In contrast, injections of enriched tau extracts from brains with AD pathology (AD-Tau) in young PS19 mice induced tau pathology predominantly in neuronal perikarya with little or no oligodendrocyte involvement 1 mo post-injection. With longer post-injection survival intervals of up to 6 mo, CBD-Tau and AD-Tau induced tau pathology spread to different brain regions distant from the injection sites while maintaining the cell type specific pattern noted above. In conclusion, these experiments provide evidence for the prion-like hypothesis of disease spread and suggest that the tau pathology formed in vivo is dependent on the tau pathology in the preparation indicating that there are post-translational modifications or strains of tau protein that are responsible for the variation between diseases.

Abstract:

Abstract:
Tau is a microtubule associated protein that promotes assembly and stabilization of cytoskeleton microtubules (MT) required for morphogenesis and axonal transport. It is mostly expressed in neuronal and glial cells but it is also present in non-neuronal cells such as fibroblast, lymphocytes and lymphoblasts where it can play further functions. Six major tau isoforms are expressed in adult human brain, as a result of alternative mRNA splicing of exons 2, 3 and 10 of MAPT, a gene located on 17q21.1 where it occupies over 100 kb. Tau is subjected to different post-translational modifications, among which phosphorylation is known to negatively regulate its ability to bind to MT. In fact, in cycling cells, tau has a low level of phosphorylation and is mostly bound to MT during interphase, while at the onset of mitosis, it became highly phosphorylated reducing its affinity to MT and permitting their dinamic instability required for the rapid mitotic movements. Abnormal hyperphosphorylation of tau causes disruption of cytoskeleton and deposition of hyperphosphorylated tau filaments in neuron and glial cells, resulting in cerebral atrophy and dementia, hallmarks of Alzheimer’s disease as well as of the so-called tauopathies, adult-onset diseases that can be of genetic origin and caused by dominant mutations in MAPT, among which frontotemporal dementia (FTD) is the most frequent. Missense, deletion and silent mutations have been reported in the coding region of MAPT, with additional mutations in the intronic region close to the 5’ splice site of exon 10. Very recently it has been proposed an involvement of tau in chromosome stability, based on its localization both in the interphasic nucleus and along mitotic chromosomes and on the presence of several chromosome and chromatin aberrations in fibroblasts and lymphocytes of patients affected by FTD carrying P301L mutation (Rossi G et al., 2008). In the present study, in order to establish whether chromosomal aberrations are a general phenomenon in tauopathies rather than a unique feature of the P301L mutation, we investigated these alterations in somatic cells of patients bearing different MAPT mutations with standard (karyotype) and molecular (array-CGH) cytogenetic techniques. Furthermore, since the observed chromosome instability resembled some features of what is seen in syndromes caused by mutations in genes involved in the DNA repair systems, we tested the response of mutated lymphoblastoid cell lines to genotoxic agents, able to induce single- and double-strand breaks. Lymphocytes and fibroblasts of patients showed a significantly higher level of structural aberrations and total aneuploidy compared to controls. We observed also a high level of premature chromatid division (pcd), that is correlated with chromosome segregation defects and aneuploidies, and abnormal nuclei. Condensations defects were also widely present in the patients and not in the controls, thus a role of tau in condensation or decondensation of chromatin may be hypothesized. Analyzing lymphoblastoid cell lines treated with genotoxic agents, we have not yet demonstrated an involvement of the protein in DNA repair systems, but these results need to be confirmed. Our findings indicate that tau may be involved in chromosome segregation and stabilization of chromatin structure most likely through interactions with both microtubules and chromatin itself. The occurrence of numerical chromosome aberrations in FTD patients is likely due, at least in part, to misfunction of tau as a MT associated protein, since the mutations studied decrease the binding of the protein to MT, producing an instable spindle. Fragility and alterations in chromosome structure due to mutated nuclear tau may slowly accumulate in postmitotic cells and lead to abnormal regulation of gene expression with subsequent neuronal damage and death. The evidence of a role of tau in the functioning of mitotic spindle and stability of chromatin points to this protein as a possible component of the complex system guarding genome stability.
