Abstract:
The recent introduction of iridium (Ir) into the catalytic converter has resulted in an increase of Ir levels into the environment, especially ambient air and soil. These observations suggested the need to evaluate potential toxicity due to Ir exposure. Since Ir compounds have been previously shown to cause immune sensitization in humans, the effects of Ir via drinking water for 90 days was assessed in adult female Wistar rats with respect to selected immune parameters. The Ir exposure induced dose-dependent decrease (p < .01) in T helper 1 (Th1) cytokines and increase (p < .001) in a T helper 2 (Th2) cytokine. The findings show that the Ir exposure affects an immune imbalance with a skewing toward a Th2 bias, a risk factor for asthma.

Abstract How environmental cues distinctly shape CD4+ T cell differentiation in response to specific bacterial infections remains largely unknown. Here we tested whether the route of infection determined which cytokines were produced by specific memory CD4+ T cells. Listeria monocytogenes and Streptococcus pyogenes were engineered to express the 2W1S antigenic peptide (LM-2W1S and SP-2W1S) and used to infect C57BL/6 mice. Endogenous 2W1S-specific CD4+ T cells were tracked using a 2W1S:I-Ab tetramer and magnetic bead enrichment. Infection with either LM-2W1S or SP-2W1S via the intranasal route resulted in splenic 2W1S:I-Ab-specific CD4+ memory cells that produced interleukin-17A (IL-17A) but not interferon-γ (IFN-γ) upon in vivo challenge, whereas infection with LM-2W1S or SP-2W1S via the intravenous route resulted in splenic 2W1S:I-Ab-specific memory cells that produced IFN-γ, but not IL-17A. These results suggest that the lymphoid environment of primary antigen encounter is an important factor in determining the type of CD4+ T cell memory generated and indicate that the route of immunization should be a major consideration when designing vaccines to improve cell-mediated immunity. Abstract T effector subsets, such as Th1 or Th2 cells, are key players in inflammatory reactions. It is not known whether chemokines are able to recruit these subsets differentially, as has been shown for memory vs naive T cells. Here we demonstrate that Th1 and Th2 cells differ in their intrinsic migratory properties and their chemotactic responsiveness toward distinct chemokines. While the CC-chemokines macrophage inflammatory protein (MIP)-1α, MIP-1β, and RANTES were efficient chemoattractants for Th1 cells, inducing a dose-dependent transmigration, Th2 cells were not attracted by these chemokines. Another CC-chemokine, JE/monocyte chemoattractant protein (MCP)-1, and a CXC-chemokine, stromal cell-derived factor (SDF)-1α, exerted chemotactic effects on both Th1 and Th2 cells, but differences in sensitivity and the percentage of responding cells were recorded between both subsets. These results indicate that chemokines play a distinct role in the regulation of local immune reactions by influencing the local balance between proinflammatory and antiinflammatory T cell subsets.

Abstract:
Abstract The differentiation of CD4+ T cell subsets in response to infection has been studied extensively, however the epigenetic programs that regulate these processes remain poorly understood. Active demethylation by tet methylcytosine dioxygenases of CpG dinucleotides within DNA is a key component of epigenetic programing that promotes lineage specific gene expression and contributes to cellular differentiation and function. Here we report that Tet2 acts to restrict the differentiation of T follicular helper (Tfh) cells in CD4+ T cells responding to viral infection. Using an adoptive transfer model of virus-specific CD4+ cells we found that Tet2-deficient CD4+ T cells skew away from the Th1 lineage and instead preferentially differentiate into highly functional germinal center (GC) Tfh cells that provide enhanced help for B cell responses. We found that the impact of Tet2-mediated programing on CD4+ T cell differentiation is cell intrinsic and the shift in lineage differentiation occurs as early as 2 days post infection. Using genome-wide expression, DNA methylation and transcription factor binding analyses, we found that Tet2 coordinates with multiple transcription factors to mediate the demethylation and expression of their target genes following activation. Our findings establish Tet2 as an important regulator of Tfh cell differentiation and reveal pathways that could be targeted to enhance GC responses against infectious disease.
