Abstract:
SNAP-25 is a synaptosomal associated protein localized at the plasma membrane of nerve terminals. SNAP-25 associates with syntaxin 1 and vesicle-associated membrane protein-2 (VAMP-2) and is thought to form a complex essential for neurotransmitter release. We have identified syndet, a novel protein related to the family of SNAP-25 isoforms. Like SNAP-25, syndet has regions with high probability of forming coiled coils, a cysteine rich-domain, and lacks a signal sequence or transmembrane domains. Syndet is tightly bound to membranes, possibly by acylation within the cysteine-rich domain. Syndet is expressed in non-neuronal tissues. In adipocytes, syndet is found at the plasma membrane and in an intracellular compartment. The identification of syndet supports the hypothesis that multiple SNAP-25 related proteins ensure specificity of vesicle fusion at the cell surface.

Purpose This paper aims to propose a statistical and context-aware feature reduction algorithm that improves sentiment classification accuracy. Classification of reviews with different granularities in two classes of reviews with negative and positive polarities is among the objectives of sentiment analysis. One of the major issues in sentiment analysis is feature engineering while it severely affects time complexity and accuracy of sentiment classification. Design/methodology/approach In this paper, a feature reduction method is proposed that uses context-based knowledge as well as synset statistical knowledge. To do so, one-dimensional presentation proposed for SentiWordNet calculates statistical knowledge that involves polarity concentration and variation tendency for each synset. Feature reduction involves two phases. In the first phase, features that combine semantic and statistical similarity conditions are put in the same cluster. In the second phase, features are ranked and then the features which are given lower ranks are eliminated. The experiments are conducted by support vector machine (SVM), naive Bayes (NB), decision tree (DT) and k-nearest neighbors (KNN) algorithms to classify the vectors of the unigram and bigram features in two classes of positive or negative sentiments. Findings The results showed that the applied clustering algorithm reduces SentiWordNet synset to less than half which reduced the size of the feature vector by less than half. In addition, the accuracy of sentiment classification is improved by at least 1.5 per cent. Originality/value The presented feature reduction method is the first use of the synset clustering for feature reduction. In this paper features reduction algorithm, first aggregates the similar features into clusters then eliminates unsatisfactory cluster.

SummaryAccording to the classical genetic analysis in Paramecium caudatum by Tsukii & Hiwatashi (1983), the E mating type of each syngen is expressed when the cell bears alleles specific for syngen at the Mt locus. The O mating type is expressed when cells are homozygous for the null allele, mt, at the Mt locus. In such mt/mt cells the O syngen specificity is determined by alleles at two other loci called MA and MB. Inthe study reported here, macronucleoplasmic transplantation technique was used to test the above hypothesis. When macronucleoplasm of type E3 (mating type E of syngen 3) was injected into a macronucleus of type O12 (mating type O of syngen 12), some recipients changed to type E of the donor syngen but some others changed to type E of the recipient syngen. Thus, syngen specificity of donor macronucleoplasm controlling type E was converted into that of the recipients, even though the latter has no gene that controls type E. When this transformant expressing type E of the recipiexnt syngen was re-injected back into E of the other syngen, the expression of the converted mating type in some way continued in the recipient. This suggests that syngen specificity of gene Mt of the donor was changed to that of the recipients by intersyngenic transplantation of macronucleoplasm. We also obtained results suggesting that the gene dosage ratio of Mt to mt or Mt to MA and MB may be important for syngen specific expression of type E.

Application of reverse transcription–PCR to total RNA prepared from TNF-α (tumour necrosis factor-α)-stimulated HUVECs (human umbilical vein endothelial cells) revealed that the syndecan-2 mRNA was up-regulated by this inflammatory stimulus. By immunoprecipitation using an anti-syndecan-2 antibody on TNF-α-stimulated HUVEC lysates, inflammation-induced interleukin-8 was found to be an interaction partner of this HS (heparan sulphate) proteoglycan, but not of any other syndecan on these cells. The glycosylated [Syn2ect(+HS)] and non-glycosylated [Syn2ect(−HS)] forms of Syn2ect (the syndecan-2 ectodomain) were purified from a stably transfected human cell line and from a bacterial expression system respectively. By CD spectroscopy, Syn2ect was found to adopt an all-β secondary structure. The dissociation constant of Syn2ect(+HS) with respect to interleukin-8 binding was determined by isothermal fluorescence titrations to be 23 nM. Despite its lack of HS chains, Syn2ect(−HS) exhibited significant binding to the chemokine, with a Kd of >1 µM. Thus, in addition to glycosaminoglycan binding, protein–protein contacts might also contribute to the chemokine–proteoglycan interaction.

We report on the improved operational performance and energy conversion efficiency of a 5-kW solar chemical reactor for the combined ZnO-reduction and CH4-reforming SynMet process. The reactor features a pulsed vortex flow of CH4 laden with ZnO particles, which is confined to a cavity-receiver and directly exposed to solar power fluxes exceeding 2000kW/m2. Reactants were continuously fed at ambient temperature, heated by direct irradiation to above 1350°K, and converted to Zn and syngas during mean residence times of 10 seconds. Typical chemical conversion attained was 100% for ZnO and up to 96% for CH4. The thermal efficiency was in the 15–22% range; the exergy efficiency reached up to 7.7% and may be increased by recovering the sensible and latent heat of the products. The SynMet process avoids emissions of greenhouse-gases and other pollutant derived from the traditional fossil-fuel-based production of zinc and syngas, and further converts solar energy into storable and transportable chemical fuels.

AbstractThe alignment of WordNet and Wikipedia has received wide attention from researchers of computational linguistics, who are building a new lexical knowledge source or enriching the semantic information of WordNet entities. The main challenge of this alignment is how to handle the synonymy and ambiguity issues in the contents of two units from different sources. Therefore, this paper introduces mapping method that links an Arabic WordNet synset to its corresponding article in Wikipedia. This method uses monolingual and bilingual features to overcome the lack of semantic information in Arabic WordNet. For evaluating this method, an Arabic mapping data set, which contains 1,291 synset–article pairs, is compiled. The experimental analysis shows that the proposed method achieves promising results and outperforms the state-of-the-art methods that depend only on monolingual features. The mapped method has also been used to increase the coverage of Arabic WordNet by inserting new synsets from Wikipedia.

The article looks at the central theme of Kaikki synnit, forgiveness, as an affective counterpart of suspicion.The story world of the series is set in the fictional North Ostrobothnian town of Varjakka, where the majority of the population are conservative Laestadians. For Nordic noir, in a genre-specific way, the story of the series arises from a topical sociopolitical issue, which in the TV series Kaikki synnit is gendered violence and sexual harassment. The theoretical background of the analysis of the series is formed by recent affect theory, where affect is understood as a social and cultural phenomenon.The music in Kaikki synnit TV series draws on affect as a key factor in the viewing experience. In the story world of the series, the affectiveness of the community is represented as bonds and breaks, which is echoed in the music of the series. Natural language processing is a subfield of linguistics concerned with the interactions between computers and human language, specifically in how to program computers to process and analyze large amounts of text data (natural language data). WSD, word sense disambiguation in natural language processing, is the task of determining the correct annotation of the pun word in given context. This paper describes about the endeavor in using cosine similarity method for detection of a single homographic pun in given context, its location, and the correct annotation with respect to helping words in the context. This paper includes two approaches: BIT_SYS1 and BIT_SYS2. The first contains the words having synset count one as it cannot be pun but it can serve as helping word to the pun, and in the later words with synset count one is eliminated and the concept of helping word is abandoned. Performance of BIT_SYS2 is better than BIT_SYS1 as F1 score of BIT_SYS2(0.8571, 1.0000, 1.0000) is higher than BIT_SYS1(0.8439, 0.8648, 0.8648) in pun detection task, pun location task, and pun annotation task.

Abstract. The synergetic aerosol retrieval method SYNAER (Holzer-Popp et al., 2002a) has been extended to the use of ENVISAT measurements. It exploits the complementary information of a radiometer and a spectrometer onboard one satellite platform to extract aerosol optical depth (AOD) and speciation (as a choice from a representative set of pre-defined mixtures of water-soluble, soot, mineral dust, and sea salt components). SYNAER consists of two retrieval steps. In the first step the radiometer is used for accurate cloud screening, and subsequently to quantify the aerosol optical depth (AOD) at 550 nm and spectral surface brightness through a dark field technique for different pre-defined aerosol mixtures. In the second step the spectrometer is applied to choose the most plausible aerosol mixture through a least square fit of the measured spectrum with simulated spectra using the mixture-dependent values of AOD and surface brightness retrieved in the first step. This method was developed and a first case study evaluation against few (15) multi-spectral ground-based AERONET sun photometer observations was conducted with a sensor pair (ATSR-2 and GOME) onboard ERS-2. Due to instrumental limitations the coverage of SYNAER/ERS-2 is very sparse. Therefore, SYNAER was transferred to similar sensors AATSR and SCIAMACHY onboard ENVISAT. While transferring to the new sensor pair significant improvements in the methodology were made based on a thorough evaluation of the methodology: (1) an update of the aerosol model, (2) improved cloud detection in the tropics and sub tropics, and (3) an enhanced dark field albedo characterization. This paper describes these improvements in detail and assesses their combined impact on the results. After a brief assessment of atmospheric noise impact on comparisons of pixel and station measurements a validation against ground-based measurements establishes error bars for the SYNAER/ENVISAT method version 2.0. A theoretical analysis of the information content with regard to aerosol composition (second retrieval step) is presented to quantify the potential and limitations of this new capability provided by the SYNAER method. Building on this analysis, first seasonal and monthly composition results calculated by applying SYNAER version 2.0 to AATSR and SCIAMACHY are shown to demonstrate the potential of the approach. An inter-comparison to earlier results of SYNAER version 1.0 is made for both the validation and the example datasets.

Løgstrup’s thoughts move in two different dimensions, but with the same intention of demonstrating that it was not the capacity of man to create culture that first gave significance to the world. But where Grundtvig speaks about history, Løgstrup speaks about »phenomena«, »nature«, and »universe«.While Grundtvig was largely unaffected by Kant, the latter - with his concepts of the selfexistent subject and the idea of the faculty of cognition as productive - became a challenge to Løgstrup. Kant heralds an era whose relationship with the universe is characterized as a »marginal existence«. Our culture became an emancipatory culture which was all to the good, but the era lost its sense of the .pre-cultural. structures in which life is »encased«.The era has also emancipated itself from Grundtvig’s historical view. But a history on the premisses of relativism is no history. Or, in Løgstrup’s words, there is no other history than the history of what is essential in life. Therefore, in reality, Løgstrup’s phenomenological and philosophical endeavours become a defence of history. Grundtvig’s view of nature was determined by his radical prioritization of history. He prefers to view nature as part of the historical life of man, which again determines his use of nature images. In Grundtvig there is no religious interpretation of any experience or perception of nature in spite of the fact that everything in the Creation is to be understood as images of the eternal.In Løgstrup there is no such cautions attitude towards nature. Here nature and sense perception are liberating, but as is the case with Grundtvig, nature is seen as the foundation of man’s life, as immediate experience.Grundtvig’s radical prioritization of history colours his view of art. Abstract:
Despite new developments in machine learning classification techniques, improving the accuracy of spam filtering is a difficult task due to linguistic phenomena that limit its effectiveness. In particular, we highlight polysemy, synonymy, the usage of hypernyms/hyponyms, and the presence of irrelevant/confusing words. These problems should be solved at the pre-processing stage to avoid using inconsistent information in the building of classification models. Previous studies have suggested that the use of synset-based representation strategies could be successfully used to solve synonymy and polysemy problems. Complementarily, it is possible to take advantage of hyponymy/hypernymy-based to implement dimensionality reduction strategies. These strategies could unify textual terms to model the intentions of the document without losing any information (e.g., bringing together the synsets “viagra”, “ciallis”, “levitra” and other representing similar drugs by using “virility drug” which is a hyponym for all of them). These feature reduction schemes are known as lossless strategies as the information is not removed but only generalised. However, in some types of text classification problems (such as spam filtering) it may not be worthwhile to keep all the information and let dimensionality reduction algorithms discard information that may be irrelevant or confusing. In this work, we are introducing the feature reduction as a multi-objective optimisation problem to be solved using a Multi-Objective Evolutionary Algorithm (MOEA). Our algorithm allows, with minor modifications, to implement lossless (using only semantic-based synset grouping), low-loss (discarding irrelevant information and using semantic-based synset grouping) or lossy (discarding only irrelevant information) strategies. The contribution of this study is two-fold: (i) to introduce different dimensionality reduction methods (lossless, low-loss and lossy) as an optimization problem that can be solved using MOEA and (ii) to provide an experimental comparison of lossless and low-loss schemes for text representation. The results obtained support the usefulness of the low-loss method to improve the efficiency of classifiers.

Purpose Text mining is growing in importance proportionate to the growth of unstructured data and its applications are increasing day by day from knowledge management to social media analysis. Mapping skillset of a candidate and requirements of job profile is crucial for conducting new recruitment as well as for performing internal task allocation in the organization. The automation in the process of selecting the candidates is essential to avoid bias or subjectivity, which may occur while shuffling through thousands of resumes and other informative documents. The system takes skillset in the form of documents to build the semantic space and then takes appraisals or resumes as input and suggests the persons appropriate to complete a task or job position and employees needing additional training. The purpose of this study is to extend the term-document matrix and achieve refined clusters to produce an improved recommendation. The study also focuses on achieving consistency in cluster quality in spite of increasing size of data set, to solve scalability issues. Design/methodology/approach In this study, a synset-based document matrix construction method is proposed where semantically similar terms are grouped to reduce the dimension curse. An automated Task Recommendation System is proposed comprising synset-based feature extraction, iterative semantic clustering and mapping based on semantic similarity. Findings The first step in knowledge extraction from the unstructured textual data is converting it into structured form either as Term frequency–Inverse document frequency (TF-IDF) matrix or synset-based TF-IDF. Once in structured form, a range of mining algorithms from classification to clustering can be applied. The algorithm gives a better feature vector representation and improved cluster quality. The synset-based grouping and feature extraction for resume data optimizes the candidate selection process by reducing entropy and error and by improving precision and scalability. Research limitations/implications The productivity of any organization gets enhanced by assigning tasks to employees with a right set of skills. Efficient recruitment and task allocation can not only improve productivity but also cater to satisfy employee aspiration and identifying training requirements. Practical implications Industries can use the approach to support different processes related to human resource management such as promotions, recruitment and training and, thus, manage the talent pool. Social implications The task recommender system creates knowledge by following the steps of the knowledge management cycle and this methodology can be adopted in other similar knowledge management applications. Originality/value The efficacy of the proposed approach and its enhancement is validated by carrying out experiments on the benchmarked dataset of resumes. The results are compared with existing techniques and show refined clusters. That is Absolute error is reduced by 30 per cent, precision is increased by 20 per cent and dimensions are lowered by 60 per cent than existing technique. Also, the proposed approach solves issue of scalability by producing improved recommendation for 1,000 resumes with reduced entropy.

Abstract. The synergetic aerosol retrieval method SYNAER (Holzer-Popp et al., 2002a) has been extended to the use of ENVISAT measurements. It exploits the complementary information of a radiometer and a spectrometer onboard one satellite platform to extract aerosol optical depth (AOD) and speciation (as choice from a representative set of pre-defined mixtures of water-soluble, soot, mineral dust, and sea salt components). SYNAER consists of two retrieval steps. In the first step the radiometer is used to do accurate cloud screening, and subsequently to quantify the aerosol optical depth (AOD) at 550 nm and spectral surface brightness through a dark field technique. In the second step the spectrometer is applied to choose the most plausible aerosol type through a least square fit of the measured spectrum with simulated spectra using the AOD and surface brightness retrieved in the first step. This method was developed and a first case study evaluation against few (15) multi-spectral ground-based AERONET sun photometer observations was conducted with a sensor pair (ATSR-2 and GOME) onboard ERS-2. However, due to instrumental limitations the coverage of SYNAER/ERS-2 and the AERONET network in 1997/98 is very sparse and thus only few coincidences with AERONET were found. Therefore, SYNAER was transferred to similar sensors AATSR and SCIAMACHY onboard ENVISAT. While transferring to the new sensor pair a thorough evaluation of the synergetic methodology and its information content has been conducted, which led to significant improvements in the methodology: an update of the aerosol model, an improved cloud detection, and an enhanced dark field albedo characterization. This paper describes the information content analysis and these improvements in detail and presents first results of applying the SYNAER methodology to AATSR and SCIAMACHY.

We propose a technique for synthesizing bidirectional programs from the corresponding unidirectional code plus a few input/output examples. The core ideas are: (1) constructing a sketch using the given unidirectional program as a specification, and (2) filling the sketch in a modular fashion by exploiting the properties of bidirectional programs. These ideas are enabled by our choice of programming language, HOBiT, which is specifically designed to maintain the unidirectional program structure in bidirectional programming, and keep the parts that control bidirectional behavior modular. To evaluate our approach, we implemented it in a tool called Synbit and used it to generate bidirectional programs for intricate microbenchmarks, as well as for a few larger, more realistic problems. We also compared Synbit to a state-of-the-art unidirectional synthesis tool on the task of synthesizing backward computations.

e21547 Background: Gene mutations in DNA damage repair (DDR) pathway were reported to affect the clinical response to immune checkpoint inhibitors (ICIs) by driving mutagenicity. Previous studies showed that RECQL5 is involved in RAD51-mediated strand invasion for homologous recombination DNA damage repair (HR-DDR). RECQL5 encoding RecQ protein-like 5 (RECQL5) is a member of RecQ helicase family and was discovered to play a tumor-suppressive or oncogenic role in various cancers. However, the association between RECQL5 mutation and ICI efficacy has not been revealed. Methods: Data of nine publicly independent cohorts of NSCLC, melanoma, and pan-cancer were retrieved (Rizvi, MSKCC, OAK/POPLAR, Van Allen, Hugo, Synder, Miao and Samstein cohorts) to investigate the correlation between RECQL5 mutations and clinical events including overall survival (OS), progression-free survival (PFS), objective response rate (ORR), and clinical benefit rate (CBR). Wilcoxon test was used for comparing tumor mutational burden (TMB) between RECQL5-mutated patients and their wild-type counterparts. In addition, the correlation between infiltration of immune cells and RECQL5 mutation status was also analyzed by QUANTISEO using data of SKCM cohort (n=466) from TCGA database. Statistical significance was set at P = 0.05. Results: RECQL5 mutations were most commonly seen in melanoma patients and the detection rate was 4%-8% in melanoma patients from cohort Van Allen, Hugo, Synder, and Miao. RECQL5 mutation was not detected in other cancers except for one lung cancer case. In the three melanoma cohorts (Van Allen, Synder, Miao) with OS data available, the OS of the RECQL5-mutated patients was 2 to 3 times longer than that of the RECQL5-wt patients. In Synder cohort, the difference in OS between these two subsets was statistically significant (53.9 months vs 25.0 months, P = 0.045). In addition, the occurrence of RECQL5 mutations was correlated with higher CBR both in Miao ( P = 0.017) and Synder cohorts ( P = 0.011). No difference was observed in the PFS or ORR between these two subsets. Notably, RECQL5 mutations were associated with higher TMB levels both in Miao (32.6 mut/Mb vs 7.9 mut/Mb, P = 0.016) and Synder cohorts (1124 mut/Mb vs 358 mut/Mb, P = 0.033). Moreover, RECQL5 mutations were found to be correlated with increased infiltration of CD4+ T cells (no-regulatory) ( P = 0.037), B cells ( P = 0.13) and decreased infiltration of regulatory CD4+ T cells ( P = 0.02) in RECQL5-mutated tumors over RECQL5-wt tumors. Conclusions: RECQL5 mutations can serve as a potential predictor for a durable response to ICIs in melanoma. Moreover, the occurrence of RECQL5 mutations was correlated with higher TMB level as well as infiltration of immune cells, indicating the underlying mechanism of its predictive effect.
