e17551 Background: Few studies have addressed the carcinogenic process of laryngeal cancer from its premalignant phases. Our aim was to compare mutational status of laryngeal dysplasias (LD) evolving to laryngeal cancer (ELC) and not evolving to LC (NELC). Methods: Retrospective study of LD diagnosed between 2007 and 2011. A customized 15-gene NGS panel (DICER, IRF6, NOTCH1, NOTCH2, NOTCH3, NOTCH2NLA, PIK3CA, PTEN, RB1, RIPK4, SYNE1, SYNE2, TP53, TP63, CASP8) was used for mutational analysis with Truseq Custom Amplicon (Illumina) performed in FFPE LD samples and results compared between LD-ELC and LD-NELC. Results: Sixty-four patients (pts) with LD were identified. LD-ELC (N = 23) and LD-NELC (N = 41) were scored as mild (N = 40; NELC: 32 / ELC:8), moderate (N = 8; NELC: 4/ ELC: 4) and severe (N = 16; NELC:5 / ELC: 11). Prior or current moderate-to-heavy tobacco smoking ( > 10 pack-year): 53/64 (83%). Males: N = 50 (ELC: 21; NELC: 29); Female: N = 14 (ELC: 2; NELC: 12). Median time to cancer among 23 LD-ELC: 8 m (range: 0-46). Forty-seven gene variants were detected in ELC not found in NELC, of which 27 were pathogenic/likely pathogenic: Frameshift: 8 (1 in NOTCH2, 1 in PTEN, 2 in RB1, 2 in SYNE1 y 2 in SYNE2); Stop-gained: 3 (1 in NOTCH2NL, 2 in SYNE2); Splicing: 3 (1 in RB1, 1 in RIPK4 y 1 in TP53); Missense: 12 (1 in IRF6, 2 in NOTCH1, 1 in RB1, 2 in RIPK4, 6 in SYNE1). Conclusions: LD from pts ELC showed a different mutational profile than LD-NELC. These results show promise for identifying pts at higher risk for developing LC and should be validated in a larger, prospective study.

Abstract:
Syne-2 (also known as Nesprin-2) is a member of a family of proteins that are found primarily in the outer nuclear membrane, as well as other subcellular compartments. Syne-2 contains a C-terminal KASH transmembrane domain and is part of a protein network that associates the nuclear envelope to the cytoskeleton via the binding to actin filaments. Syne-2 plays a role in nuclear migration, nuclear positioning during retinal development, and in ciliogenesis. In a previous study, we showed a connection between Syne-2 and the multifunctional scaffold protein Pericentrin (Pcnt). The elimination of the interaction of Syne-2 and Pcnt showed defects in nuclear migration and the formation of outer segments during retinal development, as well as disturbances in centrosomal migration at the beginning of ciliogenesis in general. In this study, the Syne-2 KO mouse model Nesprin-2△ABD (Syne-2tm1Ngl, MGI) with special attention to Pcnt and ciliogenesis was analyzed. We show reduced expression of Syne-2 in the retina of the Syne-2 KO mouse but found no significant structural—and only a minor functional—phenotype. For the first time, detailed expression analyses showed an expression of a Syne-2 protein larger than 400 kDa (~750 kDa) in the Syne2/Nesprin-2 KO mouse. In conclusion, the lack of an overt phenotype in Syne-2/Nesprin-2 KO mice suggests the usage of alternative translational start sites, producing Syne-2 splice variants with an intact Pcnt interaction site. Nevertheless, deletion of the actin-binding site in the Syne-2/Nesprin-2 KO mouse revealed a high variability in scotopic oscillatory potentials assuming a novel function of Syne-2 in synchronizing inner retinal processes.

Abstract:
Addressing the critique that the global city discourse should tackle the important issue of migration, this article utilizes Sydney as a case study to substantiate and advance the debates. It is a comprehensive literature review on the global Sydney discourse and migration in global Sydney. Concerning global Sydney, it addresses Sydney's rise as a global city and the economic, social, and spatial transformations. Concerning migration, it addresses its increased scale and complexity, spatial settlement and movement, and social polarization and spatial inequality. The article identifies two gaps in the literature: Missing migration in the global Sydney discourse, and failure to address the interplay between global Sydney and migration. Based upon the thematic patterns of the literature, and the identified gaps, some critical reflections are made to suggest a future research agenda — utilizing global Sydney as a meaningful spatial scale for contemporary migration analysis, and developing the analytical frameworks and methodologies that integrate both global Sydney and migration research.

Born in Sydney in 1911, Sydney Dattilo Rubbo was educated at Sydney Boys? High School and the University of Sydney (BSc, 1934) before travelling to London to further his studies. He obtained a diploma in bacteriology from the London School of Hygiene and Tropical Medicine (1935) and was awarded a scholarship for microbiological research at the University of London (PhD, 1937). Returning to Australia in 1937, Rubbo took up an appointment as a senior lecturer in the Department of Bacteriology at the University of Melbourne where he taught students of medicine, dentistry, science and agricultural science. A ?brilliant and provocative lecturer?, he inspired a generation of students. He also studied and completed a medical degree (MB, BS, 1943) and in 1945, at the age of 33, was appointed Professor of Bacteriology (Microbiology from 1964), a position he held until 1969.

Abstract Using a qualitative instrumental case study research design, this study examines the strategies and carbon reduction measures implemented by Sydney Airport to achieve their goal of being a carbon neutral airport by 2025. The study period was from 2013 to 2019. The qualitative data was analyzed using document analysis. Sydney Airport has implemented a wide range of carbon reduction measures that underpin its strategy to become a carbon neutral airport. Sydney Airport’s annual emissions intensity per passenger declined in each year examined in study. Sydney Airport has participated in the Airports Council International Airport Carbon Accreditation Program since 2014 and currently holds Accreditation Level 3: Optimization. Sydney Airport’s goal is to be awarded Accreditation Level 3+: Carbon Neutrality by 2025. Sydney Airport has a carbon offsets agreement in place with a not-for-profit organization.

The Sydney Harbour waterway modelling suite examines the changes in water quality in the harbour estuary and its tributaries associated with stormwater runoff and wet weather sewage overflows from the upstream catchments, in Sydney Australia. This paper discusses the development and performance of the numerical models. The models have been used to investigate the spatial variability of catchment pollutant loads and the impacts of sewer overflows on the water quality in the Sydney Harbour estuary. The scenario modelling results demonstrate that sewer overflows have a minimal impact on the Sydney Harbour estuary water quality, with stormwater dominating most changes in water quality.

AbstractThis study investigated the generalisability of the Rapid Screen of Concussion (RSC; a series of brief computerised tests) and the Digit Symbol Substitution Test (DSS; Wechsler, 1981) for discriminating between concussion (mTBI) and orthopaedic patient groups in two different centres, the Royal Brisbane Hospital, Brisbane (N = 177) and St. Vincent's Hospital, Sydney (N = 211). Group differences and interactions were assessed between test centre (Sydney or Brisbane) and injury type (mTBI or orthopaedic control) on a composite variable of the RSC and DSS subtests. Age, education and number of standard drinks of alcohol consumed were covaried for these analyses. While the mTBI patients obtained lower scores than orthopaedic patients, there were no significant differences between Brisbane and Sydney participants in performance on the RSC and DSS. There was no significant interaction between injury type and test centre. Similarities of correct classification rates of the RSC and DSS for Brisbane and Sydney were examined using a double cross-validation technique. When cross-validating from the Brisbane to Sydney sample it was revealed that sensitivity was 73% in Brisbane and 71% in Sydney, while specificity for the two samples was 76% and 67%, respectively. When cross-validating from the Sydney sample to the Brisbane sample sensitivity was 64% and 63%, respectively. Specificity was 81% in Sydney and 80% in Brisbane. These results indicated that correct classification rates were sufficiently similar between the cities and that the RSC and DSS could be generalised. When samples were combined, sensitivity was found to be 66% while specificity was 80%. The RSC and DSS are recommended as adjunct procedures to medical diagnoses of mTBI.

Urban planning policies in New South Wales (NSW), Australia are continuously being reformed, in order to make them more economic development friendly. These reforms are concerned with making development approvals easier and faster. The implementation of these reforms and their outcomes in Greater Sydney, NSW, vary according to the local socio-economic conditions. The affluent communities in Greater Sydney are very concerned about these reforms and actively resist their application in their areas. They are successful in avoiding the application of reformed urban planning policies. However, the lower socio-economic parts of Greater Sydney in the outer areas are not able to engage with these urban policy issues. The reformed urban policies are fully applied in the poorer areas, often resulting in excessive and poor-quality urban development. Past research on urban planning policy development, application and outcomes in Sydney has not investigated selective planning policy application and its differential outcomes. This paper analyses the selective application of some recent urban planning policy reforms as they relate to socio-economic division in Greater Sydney. The research argues that the selective application of urban planning policy in Greater Sydney is reinforcing socio-economic division there.

Sydney is an Indigenous place – Indigenous Country – infused with Indigenous stories and lore/Law. Yet as the original site of British colonisation in 1788, Sydney today is also a deeply-colonised place. Long-held narratives of Sydney as a colonial city have worked hard to erasure Indigenous peoples’ presences and to silence Indigenous stories of this place (Rey and Harrison, 2018). In recent years, however, Indigenous-led tours on Country are emerging in the Greater Sydney region, whereby Indigenous guides share with visitors stories of place, history, culture, language and connection. We write together as Indigenous and non-Indigenous researchers, in conversation with four Indigenous tour operators in the Greater Sydney region to reflect on their experiences of conducting Indigenous tours in this Indigenous-yet-deeply-colonised place. We document the kinds of ‘deep-colonising’ (Rose, 1996) narratives and assumptions the operators encounter during their tours and within the tourism industry, and highlight how Indigenous tour operators facilitate many non-Indigenous peoples in taking their first steps towards meaningful interactions with Indigenous Sydney-siders. We conclude that Indigenous tour operators undertake incredibly complex, confronting and challenging emotional labours trying to change the pervasive and deep-colonising narratives and assumptions about Indigenous peoples in the Greater Sydney region. In a world where the histories of thousands of cities ‘lie in dispossession and genocide of Indigenous peoples’ (Porter, 2020: 15), we argue for further and careful analytical attention on Indigenous tourism encounters in Indigenous – yet deeply-colonised – places.

Objective: The aim is to describe the process adopted in Northern Sydney, Sydney, Australia for developing consensus guidelines for the treatment of young people with first episode psychosis. Method: The process included academic detailing with psychiatrists working in Northern Sydney adolescent and adult mental health services, two clinician forums, and a survey. While the guidelines were based on the Australian Clinical Guidelines for Early Psychosis and published studies, the expertise of clinicians in Northern Sydney was also sought. Results: The consensus guidelines have been endorsed for use in Northern Sydney. The guidelines cover the issues of assessment and treatment of psychosis in young people, investigations, use of parenteral medication, diagnosis, and information for clients. Conclusions: The authors make several observations about the process of guideline development. Academic detailing proved to be a more effective way than questionnaires for obtaining information from clinicians. The process of developing the guidelines seemed to be, in some ways, more important than the final product.

MERV GRIFFITHS was born in Sydney on 8th July 1914, grew up in Northbridge and was educated in schools in North Sydney, including North Sydney Boys’ High School, which he attended from 1930-32. He entered what he, mischievously referred to as “The University” [University of Sydney] in 1934 to study Zoology. This period was difficult financially for all, including the University of Sydney, but in spite of the effects of underfunding and crowded conditions, Merv followed his biological interests under the tutelage of Professor W. J. Dakin and a small staff in the Zoology Department. Merv first began publishing in the scientific literature in 1936 with a paper on The colour changes in batoid fishes in the Proceedings of the Linnean Society of New South Wales.

Purpose: The aim of this study was to report early experiences with the Sydney and Endovascular Technologies (EVT) prostheses for the treatment of abdominal aortic aneurysms (AAA) deemed suitable for endoluminal tube graft repair. Methods: Consecutive endoluminal tube graft repairs were analyzed over the first 12 months in which the Sydney and EVT prostheses were used. Patients eligible for the EVT prosthesis had type I AAAs: a proximal neck length ≥ 2 cm, a distal cuff length ≥ 1.5 cm, and nontortuous iliac arteries ≥ 8 mm. Selection criteria for the Sydney device were more liberal and included AAAs that had distal cuffs < 1.5 cm. During the study period, 28 of 91 patients evaluated for AAA repair were thus selected for endoluminal grafting: 18 patients received the Sydney endograft and 10 the EVT device. Medical comorbidities were present in slightly less than one third of patients in both groups. Contrast-enhanced computerized tomography (CT) was performed preoperatively, within 10 days of operation, and at 6 and 12 months postprocedure. Results: All endografts were successfully deployed in both groups. Postprocedural CT scans revealed incomplete aneurysm exclusion in four patients with the Sydney endograft. Subsequent deployment of a second endograft sealed these “leaks” in two cases; the other two were converted to open repair (89% clinical success). No leaks were seen with the EVT device. Local/vascular complications occurred in 33% of the Sydney group compared with 20% for the EVT device (p = 0.001); systemic sequelae were more common in the EVT group (30% versus 17% in the Sydney cohort, p = 0.002). There were no deaths within 30 days; three late deaths were not procedure related. Conclusion: AAAs that are suitable for endoluminal tube graft repair may be treated with a high rate of initial success with either the Sydney or EVT prostheses. More liberal selection criteria may increase the likelihood of local/vascular complications.
