Abstract:
The demountable splicing of optical fibres uses different types of connectors which ensures accurate position of connected fibres. If the optical fibres are aligned in free space, a view from two perpendicular viewing directions is necessary for a maximum aligning accuracy. The method of direct monitoring of optical fibres provides this possibility however, it is necessary to use two imaging systems. This problem can be solved by a special microscope, which is designed in this thesis. The microscope can visualize the alignment of optical fibres from two mutually perpendicular directions by moving the objective lens and inclined mirror. The diploma thesis also describes the procedure of designing an optical simulation of the connection of optical fibres. Based on it, the microscope is designed, adjusted and tested with various optical fibres.

The aim of the paper is to introduce an optic microscope as well as microscopic photography producing. It further engages in sharpening issues and attaining a sharp image with extremely robust depth acuteness assuming using a series of images with different acuteness levels. The goal is to create a suitable algorithm to increase depth acuteness, to perform a visual processing of the series of images and to get one final image with high depth acuteness.

Abstract:
The new arrangement of the off-axis holographic module, which is using polarizationactive diffraction grating divides signal into reference and subject wave of an interferometer based on their polarization. However, current design of the module does not have a possibility to tune a length of the optical paths. Thus the inability to tune optical paths leads to a reduced quality of interference structure during observation of biological samples. The current module is only suitable for technical applicating due to this limitation. Possibility of tuning branches is key step in biological applications. Therefore a new computer-controlled module is created in order to enable use for biological samples.

Abstract:
Tato práce popisuje vznik obrazu v transmisním světelném mikroskopu s proměnnou aperturou kondenzoru. Tento způsob pořizování obrazu je výhodný při pozorování tlustých vzorků, což jsou objekty, u kterých je v mikroskopickém měřítku významná výška. Klasické transmisní mikroskopy pro tento výzkum vhodné nejsou, protože výsledný obraz, který produkují, obsahuje patrné informace z velkého objemu vzorku mimo oblast, která je vyšetřovaná. Tento problém byl vyřešen po vynálezu konfokálního mikroskopu, který je ovšem daleko dražší a má i některé nevýhody navíc. Cílem je zpracovat obrazy pořízené pomocí kondenzoru s rotující aperturou tak, aby došlo k redukci podílu nežádoucí informace a obraz se tak stal "čistším". Tato metoda zpracování obrazu nemohla být použita v minulosti, protože tehdejší počítače neměly dostatečnou paměť a výkon. Na tuto metodu se dá nahlížet jako na převod množiny výstupů z mikroskopu s vylepšenou osvětlovací soustavou na odpovídající množinu výstupů konfokálního mikroskopu.

Abstract:
During operational conditions of internal combustion turbines of turbojet engines, the impeller blades are stressed by load cycles which vary in time, temperature and stress. In the course of operation, the blades are exposed to a considerable number of degradation effects, particularly high-temperature corrosion, fatigue processes and creep. The presented work is aimed at the study of the structure cast nickel-base superalloy INCONEL 713 LC, creep tests exposured. The structural changes (degradation) was clasifikation by the help of image analyses. Operational conditions of heat parts, used in gas turbines and turbo-compressors, were simulated by creep tests of Inconel 713 LC nickel superalloy during a constant load of temperatures ranging from 750 up to 950C. Changes in the structure dependent upon time, temperature and stress applied were analysed by means of light and electron microscopy methods and image analyses after the fracture of test bars.

Abstract:
The aim of this work is to define relations between grain size and bioaktivity of oxide ceramics, specifically ZrO2, Al2O3 and HA. Ceramic materials with grain size from 100 nm up to 10 m, with various surface roughness, were tested for its bioactivity. Ceramography analysis was performed for all tested materials to precisely describe microstructures. Biological properties of the ceramic materials were tested via dilation tests directly in-vitro and by in-vitro extraction. Three cell culturing lines: osteoblast MG63, fibroblast L929, and epithelioid HeLa, were used for our testing. An influence of the grain size on the biological response was only found for the ceramic materials which had been thermally etched. The thermally etched nanocrystalline samples had larger areas covered by cells than ceramics with coarse grain microstructure. Biological tests on layered composites Al2O3×ZrO2 showed the cell selection determined by the type of material, where ZrO2 surfaces were preferably covered. Improved biological response of nanocrystalline ZrO2 was demonstrated on ceramic ZrO2, Al2O3 and SiO2 substrates with nanocrystalline coating of ZrO2. In this work a novel technological process for the formation of defect-free coatings was developed. Sintered coatings were tested using in-vitro technique with cell line HeLa, L929 and MG63 for up to 72 hours. The results of the biological tests of nanocrystalline coatings were consistent with results from the bulk nanocrystalline thermally etched ZrO2 ceramics.

13551398399831-171f83a418df200fec6f622f22b6fb3d.txt The Thesis describes new methods for automatic processing of image data in biology, physics and medicine. The developed methods reconstruct light microscopy images of growing microorganisms in intervals between observations, measure particles in atomic force microscopy images, and evaluate parameters of vocal fold vibrations in videokymographic images. All three problems have been hitherto solved primarily visually. The proposed methods allow automatic or computer-aided processing of the image data, and thus facilitate the evaluation process. Performance of the developed methods was tested on real images; the results were comparable with ground truth or results of visual evaluation. Application of the developed methods is not limited to the specific type of image data; the methods can be used in general for processing of images with similar characteristics. Page 1

Abstract:
Monocytes are a population of mononuclear lekocytes. The aim of this thesis was to select a suitable methodology for the isolation of monocytes and their cultivation into dendritic cells. For the experiment, bovine blood was taken from vena jugularis externa. This blood was used to isolate monocytes based on the density gradient of OptiPrep solution and Histopaque solution. The results showed that the method of isolation based on the density gradient of the Histopaque solution and the magnetic separation appeared to be a more efficient method. The monocyte population was cultured for 72 hours. Cells were analyzed by light microscopy and CD14 positive cells appeared to be transformed into dendritic cells. The purity of these cell population was determined by flow cytometry. Dendritic cells have a wide range of utility mainly in immunotherapy of various diseases and also play an important role in cancer therapy.

The main objective of this thesis was to create an educational website aimed at soil arthropods. It can be used primarily by teachers of biology and ecology, lecturers of extracurricular science education or by the students who are interested in this issue. This photographic atlas of soil arthropods consists of my own micro images obtained by the use of light and scanning electron microscope. The micro images are accompanied by text to each group of soil arthropods, which is divided into sections containing information about the systematic classification, anatomy and morphology, biology and importance, eventually representatives of the group. The atlas is accompanied by other materials usable in the classroom, such as didactic test, didactic game and proposals for group work, worksheet, field and laboratory work or educational presentations focused on soil arthropods. These materials can be downloaded from the website, along with the entire atlas of soil arthropods. The atlas is available on the following address: https://sites.google.com/site/pudniclenovci/. The review of the literature deals with the importance of arthropods in the soil and their mutual interactions.
