Abstract:
ABSTRACT Bacteroides thetaiotaomicron, a gram-negative obligate anaerobe, utilizes polysaccharides by binding them to its cell surface and allowing cell-associated enzymes to hydrolyze them into digestible fragments. We use the starch utilization system as a model to analyze the initial steps involved in polysaccharide binding and breakdown. In a recent paper, we reported that one of the outer membrane proteins involved, SusG, had starch-degrading activity but was not sufficient for growth on starch. Moreover, SusG alone did not have detectable starch binding activity. Previous studies have shown that starch binding is essential for starch utilization. In this paper, we report that four other outer membrane proteins, SusC through SusF, are responsible for starch binding. Results of 14C-starch binding assays show that SusC and SusD both contribute a significant amount of starch binding. SusE also appears to contribute substantially to starch binding. Using affinity chromatography, we show in vitro that these Sus proteins interact to bind starch. Moreover, protease accessibility of either SusC or SusD greatly increased when one was expressed without the other. This finding supports the hypothesis that SusC and SusD interact in the outer membrane. Evidence from additional protease accessibility studies suggests that SusC, SusE, and SusF are exposed on the cell surface. Our results demonstrate that SusC and SusD act as the major starch binding proteins on the cell surface, with SusE enhancing binding. SusF's role in starch utilization has yet to be determined, although the fact that starch protected it from proteolytic attack suggests that it does bind starch.

Abstract:
ABSTRACT An early step in the utilization of starch by Bacteroides thetaiotaomicron is the binding of starch to the bacterial surface. Four starch-associated outer membrane proteins of B. thetaiotaomicron that have no starch-degrading activity have been identified. Two of these, SusC and SusD, have been shown by genetic analysis to be required for starch binding. In this study, we provide the first biochemical evidence that these two proteins interact physically with each other. Both formaldehyde cross-linking and nondenaturing gel electrophoresis experiments showed that SusC and SusD interact to form a complex. Two other proteins encoded by genes in the same operon, SusE and SusF, proved not to be essential for starch utilization and actually decreased starch binding when they were present along with SusC and SusD. Consistent with this, nondenaturing gel analysis revealed that in a strain producing SusC, SusD, and SusE, the SusCD complex was partially destabilized. The strain producing SusC, SusD, and SusE also grew more slowly on starch than a strain producing SusC, SusD, SusE, and SusF (μmax, 0.29 and 0.37/h, respectively). Thus, SusE appears to interact with the SusCD complex. SusE also interacts with SusF, because SusE was less susceptible to proteinase K digestion when SusF was present, and nondenaturing gel analysis detected a complex formed by these two proteins. Our results indicate that SusC, SusD, SusE, and SusF form a protein complex in the outer membrane but that SusE and SusF are dispensable members of this complex.

Abstract:
Offline Arabic Handwriting Recognition (OAHR) has recently become instrumental in the areas of pattern recognition and image processing due to its application in several fields, such as office automation and document processing. However, OAHR continues to face several challenges, including high variability of the Arabic script and its intrinsic characteristics such as cursiveness, ligatures, and diacritics, the unlimited variation in human handwriting, and the lack of large public databases. In this paper, we introduce a novel context-aware model based on deep neural networks to address the challenges of recognizing offline handwritten Arabic text, including isolated digits, characters, and words. Specifically, we propose a supervised Convolutional Neural Network (CNN) model that contextually extracts optimal features and employs batch normalization and dropout regularization parameters. This aims to prevent overfitting and further enhance generalization performance when compared to conventional deep learning models. We employ a number of deep stacked-convolutional layers to design the proposed Deep CNN (DCNN) architecture. The model is extensively evaluated and shown to demonstrate excellent classification accuracy when compared to conventional OAHR approaches on a diverse set of six benchmark databases, including MADBase (Digits), CMATERDB (Digits), HACDB (Characters), SUST-ALT (Digits), SUST-ALT (Characters), and SUST-ALT (Names). A further experimental study is conducted on the benchmark Arabic databases by exploiting transfer learning (TL)-based feature extraction which demonstrates the superiority of our proposed model in relation to state-of-the-art VGGNet-19 and MobileNet pre-trained models. Finally, experiments are conducted to assess comparative generalization capabilities of the models using another language database , specifically the benchmark MNIST English isolated Digits database, which further confirm the superiority of our proposed DCNN model.

Abstract:
In this paper we describe an educational program for the development of the study method for students with specific learning disorders (SLD) and a first Italian application on a small sample of fifth grade students. This experience was aimed at collecting useful data precisely to review and improve the above-mentioned program. A particularly relevant problem was identified looking at the anthropological dimension of the capability learning and the need for students to acquire the learning to learn ability. Specifically, it is about the low levels of text comprehension in Italian schools and its consequences on the study activities. Within the inclusive perspective and taking into account the effective didactic strategies in the evidence-based education (EBE) perspective, the project created and tested a teaching kit (SUST) for the fifth classes of the primary school, at-tended by students with SLD. This experimental framework was conducted following the Design Based Research methodology. It also focused on the training of teachers within the intervention classes. The positive results of SUST application open a constructive scenario for the definition of intervention programs aimed at improving text comprehension and study skills of students with SLD. This program should be tested in a wider context of schools.

Abstract:
Abstract. Ship-based multi-axis differential optical absorption spectroscopy (MAX-DOAS) measurements were conducted along the marginal seas of China from 19 April to 16 May 2018 to measure the vertical profiles of aerosol, nitrogen dioxide (NO2), and nitrous acid (HONO). Along the cruise route, we found five hot spots with enhanced tropospheric NO2 vertical column densities (VCDs) in the Yangtze River Delta, Taiwan Strait, Guangzhou–Hong Kong–Macau Greater Bay Area, port of Zhanjiang, and port of Qingdao. Enhanced HONO concentrations could usually be observed under high-level aerosol and NO2 conditions, whereas the reverse was not always the case. To understand the impacts of relative humidity (RH), temperature, and aerosol on the heterogeneous reaction of NO2 to form HONO in different scenarios, the Chinese Academy of Meteorological Sciences (CAMS) and Southern University of Science and Technology (SUST) MAX-DOAS stations were selected as the inland and coastal cases, respectively. The RH turning points in CAMS and SUST cases were both ∼ 65 % (60 %–70 %), whereas two turning peaks (∼ 60 % and ∼ 85 %) of RH were found in the sea cases. As temperature increased, the HONO / NO2 ratio decreased with peak values appearing at ∼ 12.5∘C in CAMS, whereas the HONO / NO2 gradually increased and reached peak values at ∼ 31.5∘C in SUST. In the sea cases, when the temperature exceeded 18.0∘C, the HONO / NO2 ratio rose with increasing temperature and achieved its peak at ∼ 25.0∘C. This indicated that high temperature can contribute to the secondary formation of HONO in the sea atmosphere. In the inland cases, the correlation analysis between HONO and aerosol in the near-surface layer showed that the ground surface is more crucial to the formation of HONO via the heterogeneous reaction of NO2; however, in the coastal and sea cases, the aerosol surface contributed more. Furthermore, we discovered that the conversion rate of NO2 to HONO through heterogeneous reactions in the sea cases is larger than that in the inland cases in higher atmospheric layers (> 600 m). Three typical events were selected to demonstrate three potential contributing factors of HONO production under marine conditions (i.e., transport, NO2 heterogeneous reaction, and unknown HONO source). This study elucidates the sea–land and vertical differences in the forming mechanism of HONO via the NO2 heterogeneous reaction and provides deep insights into tropospheric HONO distribution, transforming process, and environmental effects.

A massa obtida é igual a mD8D80 = (5.05±0.19) GeV.
Using the QCD sum rules we test if the new narrow structure, the X(4350) recently observed by the Belle Collaboration, can be described as a JPC = 1+ exotic D8D80 molecular state. We consider the contributions of condensates up to dimension eight, we work at leading order in s and we keep terms which are linear in the strange quark mass ms. The mass obtained for such state is mD8D80 =(5.05 ± 0.19) GeV. We also consider a molecular 1+, DD0 current and we obtain mDD0 = (4.92 ± 0.08) GeV. We conclude that it is not possible to describe the X(4350) structure as a 1+ D8D80 molecular state.

Abstract:
The ATLAS detector is one of the two largest experiments installed at the Large Hadron Collider at CERN, the European Organization for Nuclear Research. During the first run, the ATLAS detector recorded data at centre of mass energies of 7 TeV and 8 TeV, enabling many precision measurements and new physics searches. One important task in ATLAS is measuring the primary vertex, the interaction point of the hardest proton-proton collision in an event. In this thesis, a study of the primary vertex reconstruction performance in data and simulated events using $t\bar{t}$ and ${Z}$ events is presented. Within the statistics available, the performance in data and simulated events is found to be compatible. Motivated by the limitations of the Standard Model of particle physics, searches for supersymmetric particles are performed with the ATLAS experiment. No signal has been observed so far, and the results are used to set exclusion limits on the masses of the supersymmetric particles. As the exclusion limits are derived from analyses which each target only a single decay mode of a supersymmetric particle, the analyses might have lower sensitivity to more complex decay scenarios. In this thesis the sensitivity of one of the ATLAS searches for direct top squark pair production to models with more complex decay modes is investigated. The study concludes that the sensitivity to models where the top squark can decay via heavier charginos and neutralinos is lower than the sensitivity to models where only decays to the lightest chargino or neutralino are present.

Abstract:
Supersymmetric extensions to the Standard Model (SM) have many interesting experimental consequences which can provide important hints to the physics beyond the SM. In this thesis, we first study the anomalous magnetic moment of the muon and show that a significant constraint on the parameter space can be obtained from its current experimental value. In the next topic, we study the CP violations in B -> phi K decays and show that the SM and the minimal supergravity model (mSUGRA) cannot account for the current experimental observation. We then show that all the data can be accommodated for a wide range of parameters in models with non-universal soft breaking left-right A terms. In our last topic, which is based on a Horava-Witten inspired model proposed by R. Arnowitt and B. Dutta, we extend their analysis to the full fermion sector of the SM and propose a new mechanism different from the usual see saw mechanism to generate small neutrino masses which are in good agreement with the current neutrino oscillation data.

Abstract:
After a brief introduction to 21st century fundamental physics suitable for the layman with a reasonable level of mathematical competence, I introduce the concept of unnaturalness in Standard Model electroweak symmetry breaking and Supersymmetry (Susy) as a potential solution. The optimally natural situation in Susy in light of the 2012 discovery of a Higgs boson is derived, namely that of almost maximal mixing, with the scalar top partners almost as light as can be. The discovery is also interpreted numerically in terms of the Next-to-Minimal Supersymmetric Standard Model, with greater emphasis placed on the visibility of the Higgs boson at the observed mass, i.e. on signal strengths. I introduce simple models of gauge-mediated Susy breaking (GMSB), and how their generalisation leads to a richer parameter space. I then investigate the role played by the mediation scale of GMSB: this is found to be as a control of the extent to which Yukawa couplings de-tune flavour-blind relations set by gauge couplings. Finally, issues relating to the discovery or exclusion of Susy at colliders are discussed. Bounds are derived for the masses of new particles from Large Hadron Collider searches for excesses of jets and missing energy without leptons, and compared to constraints arising from Higgs boson searches, for models of GMSB and the Constrained Minimal Supersymmetric Standard Model. I present a novel search strategy for new physics signatures with two neutral, stable particles, when such particles are produced by boosted decays. (Susy examples include models with light gravitinos, pseudo-goldstinos, singlinos or new photinos.) The method is shown to produce sharp mass peaks that enhance the visibility of the signal.

Abstract:
We study various aspects of type IIB supergravity backgrounds related to those of Klebanov-Strassler and Chamseddine-Volkov/Maldacena-Nunez, in the context of gauge-gravity duality. We first examine the structure which can be found in the family of supersymmetric solutions, and in particular the effect of adding flavour. We then turn to the problem of finding non-supersymmetric generalisations of the well-understood supersynnnetric solutions. Using a combination of analytic and numerical techniques we find a two-dimensional space of such solutions, corresponding to the baryonic branch of Klebanov-Strassler deformed by a SUSY-breaking irrelevant operator. We explore the parameter space in some detail, and find several interesting special cases and limits, including one in which supersymmetry is completely broken and unusual periodic behavior is present in the warp factors. Finally, we look in detail at some of the methods which can be used to probe the field theory corresponding to these kinds of backgrounds.

Hautala, J. (Jenna). "Boreaaliset suot maapallon hiilinieluina." Bachelor's thesis, University of Oulu, 2019. http://urn.fi/URN:NBN:fi:oulu-201901181080.

Full text
Abstract:
Erityisesti rahkasammaleella (Sphagnum) on tärkeä merkitys boreaalisilla soilla.

Abstract:
Climate change has become one of the world’s leading threats. Currently, the construction industry has a high environmental footprint. For this reason, the scientific and technological sector is looking for new materials to reduce the environmental consequences of this division. It is well known that the valorisation of different by-products can contribute to the reduction of the energy global consumption and CO2 emissions. Magnesium Phosphate Cement (MPC) can be obtained by using Low Grade Magnesium Oxide (LG-MgO) as a by-product from the industrial process of magnesite calcination. In this research, a Sustainable MPC (Sust-MPC) for different construction purposes is developed by using LG-MgO along with monopotassium phosphate KH2PO4 (MKP) as raw materials. The increasing use of synthetic fibres in clothing, as well as China’s competitive prices on Animal Fibres (AF) market, have led to a commercial interest fibre decrease for wool-like AF in Spain. This study aims to formulate a Sust-MPC cement with Animal Fibre (AF) to reduce the cost of the new material (Sust-MPC-AF) and to increase the thermal insulation, allowing the use of Sust-MPC-AF in several potential applications. Besides, it should be emphasized that the final pH of Sust-MPC is neutral, which allows containing natural fibres. To develop Sust-MPC-AF, some properties such as thermal conductivity, density, Modulus of Elasticity (MoE), flexural strength, and economic cost were evaluated using the Design of Experiments (DoE). The DoE studies allowed obtaining a model for further optimization considering minimum thermal conductivity and cost dosages. The formulation 30L-25EW presents the minimum conductivity (λ=0.140 W·m-1·K-1). Therefore, two optimal dosages (36L-25EW and 24L-22EW) are obtained by considering mixing variables such as AF/Cement ratio (AF/C) and AF/Extra Water ratio (AF/EW).
