Abstract:
The burden of human infections with influenza A and B viruses is substantial, and the impact of influenza B virus infections can exceed that of influenza A virus infections in some seasons. Over the past few decades, viruses of two influenza B virus lineages (Victoria and Yamagata) have circulated in humans, and both lineages are now represented in influenza vaccines, as recommended by the World Health Organization. Influenza B virus vaccines for humans have been available for more than half a century, yet no systematic efforts have been undertaken to develop high-yield candidates. Therefore, we screened virus libraries possessing random mutations in the six “internal” influenza B viral RNA segments [i.e., those not encoding the major viral antigens, hemagglutinin (HA) and neuraminidase NA)] for mutants that confer efficient replication. Candidate viruses that supported high yield in cell culture were tested with the HA and NA genes of eight different viruses of the Victoria and Yamagata lineages. We identified combinations of mutations that increased the titers of candidate vaccine viruses in mammalian cells used for human influenza vaccine virus propagation and in embryonated chicken eggs, the most common propagation system for influenza viruses. These influenza B virus vaccine backbones can be used for improved vaccine virus production.

The relevance of the article. Ukrainian society is experiencing a difficult period of existence, in 2022 began a full-scale Russian aggression against our country, which aggravated many complex issues of physical survival and psychological endurance of Ukrainians. Current life of Ukrainian citizens is characterized by instability, threats of physical destruction, information disorientation, unpredictability, the need to simultaneously solve many vital problems, which is a significant stress factor. Under these conditions, Ukrainian librarians, despite the destruction of many library premises, actively engaged in helping the society in all possible directions. The purpose of the article. To prove the availability of information and resource potential of public libraries of Ukraine as centers of national resistance through diversification of their basic functions, attraction of innovative instruments of activity and channels of communication. The methodology. The article considers the activity of Ukrainian libraries as centers of national resistance and psychological support of the society during the Russian military aggression against Ukraine. The results. The article presents an innovative view on the content and organization of the basic directions of socio-cultural activity of Ukrainian libraries. The need to strengthen the information impact on users through social networks, use of communication potential of copywriting and Storytelingu was emphasized. The availability of powerful capabilities and reserves of critical thinking, media literacy and information hygiene skills for libraries has been proved. The scientific novelty. The article presents an innovative view on the content and organization of the basic directions of socio-cultural activity of Ukrainian libraries. The practical significance. Abstract:
We studied a nesting aggregation of the primitively eusocial sweat bee Halictus ligatus near Victoria in southern Ontario during the summers of 1984, 1990, and 1991. Differences in local weather patterns from year to year had marked effects on bee demography and behaviour, belying previous conclusions about "typical" social organization in this aggregation. In 1990, comparatively cool, rainy weather resulted in high nest-failure and low brood-survival rates, while in 1984 and 1991, relatively dry, warm weather had the opposite effect. In 1984 and 1990, spring nest initiation was synchronous and the emergence periods of the first (worker) and second (reproductive) broods were temporally distinct. In 1991, exceedingly warm spring weather caused asynchrony in the timing of nest initiation, accelerated brood and colony development, and continuous brood production. In 1984 and 1990, a few males were produced in the first brood but most were produced in the second brood several weeks later. In 1991, continuous brood production meant that production of males represented the transition between production of workers and of gynes (second-brood females). Patterns of demographic and social variation exhibited by H. ligatus at Victoria parallel those observed on a continent-wide geographic scale. This suggests that primitively eusocial sweat bees maintain a variety of reproductive options, adjusting their social behaviour in response to local environmental conditions.

В статье рассматриваются особенности социального развития Ливерпуля в контексте его роли во внутренней политике Великобритании в эпоху королевы Виктории. Крупнейший город и порт Соединённого Королевства Великобритании и Ирландии одним из первых ощутил на себе все негативные последствия промышленного переворота, урбанизации и миграции населения. Как и многие другие города Великобритании, Ливерпуль фигурировал в прессе, медицинских, статистических и парламентских отчётах, как город подвалов, центр массовой миграции, трущоб, высокой смертности населения, отсутствия санитарно-технических норм и антисанитарии. Парламентские расследования и пресса указали на Ливерпуль, а также Вулверхемптон, Глазго, Дублин, Лидс, Лондон, Манчестер, Шеффилд, как города, где необходимо первоочередное проведение социальных реформ. В середине XIX в. в эпоху королевы Виктории Ливерпуль израсходовал значительные суммы денег на решение проблемы антисанитарии, уборки и мощения города, водоснабжения и освещения, жилищную реформу, организацию прачечных, общественных бань, библиотек, парков. Пионерами муниципализации и здравоохранения Ливерпуля стали — доктор Уильям Данкен, С. Хольм, Дж. Тинн. The article examines the features of Liverpool's social development in the context of its role in the domestic politics of Great Britain in the era of Queen Victoria. The largest city and port of the United Kingdom of Great Britain and Ireland was one of the first to feel all the negative consequences of the industrial revolution, urbanization and population migration. Like many other cities in the UK, Liverpool appeared in the press, medical, statistical and parliamentary reports as a city of basements, a center of mass migration, slums, high mortality, lack of sanitary standards and unsanitary conditions. Parliamentary investigations and the press have pointed to Liverpool, as well as WolverHampton, Glasgow, Dublin, Leeds, London, Manchester, Sheffield, as cities where social reforms are needed as a priority. In the middle of the XIX century in the era of Queen Victoria, Liverpool spent significant amounts of money on solving the problem of unsanitary conditions, cleaning and paving the city, water supply and lighting, housing reform, the organization of laundries, public baths, libraries, and parks. The pioneers of municipalization and health care in Liverpool were Dr. William Duncan, S. Holm, J. Thinn. Prior to the introduction of the National Public Health Law of 1848, Liverpool initiated the Institute of inspection and sanitary doctors, housing reform, and systematic state intervention in solving social problems.

AbstractCancer is a major health problem and a leading cause of death worldwide. Early cancer detection and continuous changes in treatment strategies have improved overall patient survival. The recent development of targeted drugs offers new opportunities for personalized cancer treatment. Nevertheless, individualized treatment is accompanied by the need for biomarkers predicting the response of a patient to a certain drug. One of the most promising breakthroughs in recent years that might help to overcome this problem is the organoid technology. Organoid cultures exhibit self-renewal capacity, self-organization, and long-term proliferation, while recapitulating many aspects of their primary tissue. Generated patient-derived organoid (PDO) libraries constitute “living” biobanks, allowing the in-depth analysis of tissue function, development, tumor initiation, and cancer pathobiology. Organoids can be derived from all gastrointestinal tissues, including esophageal, gastric, liver, pancreatic, small intestinal and colorectal tissues, and cancers of these tissues. PDOs are amenable to various techniques, including sequencing analyses, drug screening, targeted therapy testing, tumor microenvironment studies, and genetic engineering capabilities. In this review, we discuss the different applications of gastrointestinal organoids in basic cancer biology and clinical translation.

Abstract:
Teaching basic swimming, water safety and rescue skills is recommended by the World Health Organization for all school-aged children. However, there is a lack of evidence on effective pedagogies to develop swimming competency and the success of swimming lessons as a drowning prevention intervention. This study used a self-report questionnaire and practical testing procedures to examine the effectiveness of a 10-week student-centred aquatic programme designed for children aged 10–12 years. The study also determined whether the non-traditional swimming programme was accepted by swim teachers, school teachers and principals, and parents from a range of schools from different geographical regions in Victoria, Australia. A total of 204 students were enrolled in the programme. The pre-programme results indicated a good level of swimming, water safety and aquatic knowledge, but low swimming ability. Swimming ability significantly improved from pre-programme to post-programme, with no significant post-programme ability differences between male and female children or for participants from different programmes. Qualitative feedback collected through questionnaires, interviews and/or focus groups from students ( n = 73) and parents ( n = 69), school teachers and principals ( n = 14), swim teachers and swim school managers ( n = 21) indicated strong support from principals and swim teachers for the student-centred pedagogy, and all stakeholders valued the focus on survival swimming competencies. This research highlights the importance of including stakeholders when designing and implementing aquatics programmes. The study has resulted in a well-founded, effective programme with tailored resources and instructional materials that are available for swim centres and schools that would enable schools globally to adopt and implement this programme.

Abstract:
Expanded non-coding RNA repeats of CCUG are the underlying genetic causes for myotonic dystrophy type 2 (DM2). There is an urgent need for effective medications and potential drug targets that may alleviate the progression of the disease. In this study, 3140 small-molecule drugs from FDA-approved libraries were screened through lethality and locomotion phenotypes using a DM2 Drosophila model expressing 720 CCTG repeats in the muscle. We identified ten effective drugs that improved survival and locomotor activity of DM2 flies, including four that share the same predicted targets in the TGF-β pathway. The pathway comprises two major branches, the Activin and BMP pathways, which play critical and complex roles in skeletal development, maintenance of homeostasis, and regeneration. The Drosophila model recapitulates pathological features of muscle degeneration in DM2, displaying shortened lifespan, a decline in climbing ability, and progressive muscle degeneration. Increased levels of p-smad3 in response to activin signaling were observed in DM2 flies. Decreased levels of activin signaling using additional specific inhibitors or genetic method ameliorated climbing defects, crushed thoraxes, structure, and organization of muscle fibers. Our results demonstrate that a decrease in activin signaling is sufficient to rescue muscle degeneration and is, therefore, a potential therapeutic target for DM2.

ObjectiveDiffuse sclerosing variant papillary thyroid carcinoma (DSVPTC) is an uncommon variant of papillary thyroid carcinoma (PTC). The biological behaviors and prognostic outcomes of this variant, however, are still controversial. The aim of this systematic review and meta-analysis is to investigate the prognostic significance and outcomes of DSVPTCs in comparison with classical PTCs (cPTCs).MethodsAn electronic search was performed in five libraries: PubMed, Scopus, ISI, World Health Organization Global Health Library (WHO GHL) and Virtual Health Library (VHL) in June 2016. Published data were extracted and were pooled into odds ratios (OR), mean differences and corresponding 95% confidence intervals (CI) using random-effect model. Publication bias was analyzed using Egger’s regression test and funnel plot observation.ResultsFrom 315 articles, we included 16 articles comprising 732 DSVPTCs for meta-analysis. Overall, DSVPTC manifested more aggressive clinicopathological behaviors than cPTC such as higher rate of vascular invasion (OR: 5.33; 95% CI: 3.08–9.23), extrathyroidal extension (OR: 2.96; 95% CI: 2.04–4.30), lymph node metastasis (OR: 5.40; 95% CI: 2.82–10.35), distant metastasis (OR: 3.61; 95% CI: 1.89–6.88) and were more likely to relapse (OR: 2.83; 95% CI: 1.59–5.05). DSVPTC patients were associated with a worsened overall survival (HR: 1.89; 95% CI: 1.36–2.62).ConclusionDSVPTCs should be considered high-risk PTCs because of high propensity for tumor invasion, metastasis, relapse and mortality. Aggressiveness of DSVPTCs might be related to a different molecular pathway than that in cPTCs.

Abstract:
There is increasing evidence for antigen-driven B-cell receptor (BCR) signalling in diffuse large B-cell lymphoma (DLBCL) and other lymphoid malignancies. This includes antigens from infections e.g.Helicobacter pyloriand Hepatitis C virus, but it is theorised that self-antigens may play a major role in some cases of lymphoid malignancy. IgVH4-34 demonstrates intrinsic autoreactivity to self-antigens on red cells, which appears to be largely mediated by two motifs within the first framework region (FR1); Q6W7 and A24V25Y26. These motifs work together to for a hydrophobic patch which determines red cell antigen binding and are frequently mutated away from self-reactivity in normal B cells. IgVH4-34 has been reported to be over-represented in DLBCL compared with expression in normal B cells. We therefore sought to identify IgVH4-34 DLBCL cases from a local cohort and to screen them for Q6W7 and A24V25Y26 motifs expecting them to be less frequently mutated in DLBCL compared with normal B cells.We also aimed to screen V4-34 cases for associated somatic mutations in other genes using high-throughput sequencing. DLBCL patient samples were obtained via the Haematology Research Tissue Bank (HRTB) in Canberra, Australia, and the Victoria Cancer BioBank. Forty-eight Formalin-Fixed, Paraffin-Embedded (FFPE) samples and 26 fresh frozen samples were screened. All samples were collected at the time of diagnosis. Patients were treated with standard chemoimmunotherapy approaches. IgVH 4-34 gene sequences were determined using an IgVH4 family-specific leader primer in combination with a JH consensus reverse primer. The IgVH region was then sequenced using Sanger sequencing. Sequences were analyzed using the IgBLAST database (National Centre for Biotechnology Information). DNA extracted from FFPE samples generally proved to have low concentration and fragmented DNA. Only 1 IgVH4-34 sequence was obtained from FFPE tissue. Five samples sequenced from fresh tissue were identified as using IgVH4-34. Using Hans criteria, it was possible to classify 3 of the 6 cases as germinal center (GC) and 1 as non-GC origin. Using fresh samples, we estimated the frequency of IgVH4-34 cases at 23%. Within FR1, Q6W7 was unmutated in all 6 samples. One sample had mutations in the A24V25Y26 motif resulting in a change to A24V25F26. The other 5 samples (83.3%) had unmutated AVY motifs. We extracted genomic DNA from and performed next generation sequencing on the 5 samples with unmutated Q6W7 and A24V25Y26 motifs using a customized capture library (SureSelectXT Target Enrichment System, Aqilent Technologies) covering genes involved in lymphomagenesis. The purified libraries were sequenced on the Illumina NextSeq500 platform at AGRF (Australian Genome Research Facility, Australia). Several genes (FCGR3A,NOTCH2andNOTCH2NLR) had mutations in all 5 samples.FCGR3Ais an IgG Fc receptor gene, and mutations inFCGR3Ahave previously been linked to systemic lupus erythematosus (SLE).NOTCH2pathway genes are frequently mutated in DLBCL.CREBBPwas mutated in four of the five samples. Mutations inCREBBPhave previously been linked with DLBCL development and regulation of immune responses. We identified high rates of IgVH4-34 (23%) in our cohort of fresh samples as previously reported. Further, we noted preservation of the Q6W7 and A24V25Y26 motifs in IgVH4-34-expressing DLBCL. This over-representation of unmutated FR1 motifs suggests that the ability to recognise self-antigens likely provides important ongoing BCR signalling that promotes survival in DLBCL. This study also highlights the difficulties in conducting DNA-based research on FFPE clinical samples which have not been collected for research purposes and the importance of tissue banking fresh samples. Studies are currently being conducted into the efficacy of BCR pathway inhibitors e.g. ibrutinib in the treatment of DLBCL and testing for unmutated IgVH4-34 FR1 motifs may present a method to predict patients who are more likely to respond. Mutations in genes such as FCGR3A,NOTCH2andCREBBPmay work in conjunction with the preserved QW and AVY motifs to promote lymphomagenesis in IgVH4-34-expressing B cells and may present targets for future research into treatment therapies. Abstract:
The Constitution of Kenya specifically recognizes the freedom of association to form and belong to trade unions. However, despite the adoption of the Labour Relations Act, union practice is still hampered by excessive restrictions. The EPZ companies are labor intensive requiring a large amount of labor to produce its goods or service and thus, the welfare of the employees play a key role in their functions. This study sought to determine the effect of trade union practices on employees’ welfare at export processing zones industries in Athi River, Kenya. The specific objectives sought to determine the effect of collective bargaining agreements, industrial action, dispute resolution and trade union representation on employees’ welfare at export processing zones industries in Athi River, Kenya. The study employed a descriptive research design. Primary data was collected by means of a structured questionnaire. The target population of the study was employees in EPZ companies in Athi River, Kenya with large employees enrolled in active trade unions. The unit of observation was the employees in the trade unions. The findings indicated that collective bargaining agreements had a positive and significant coefficient with employees’ welfare at the EPZ industries. Industrial action had a positive but non-significant effect with employees’ welfare at Export Processing Zones industries. Dispute resolution had a positive and significant coefficient with employees’ welfare at the EPZ industries. Trade union representation had a positive and significant coefficient with employees’ welfare at the EPZ industries. The study recommended that trade union should avoid the path of confrontation but continue dialogue through the collective bargaining process and demands should be realistic in nature with what is obtainable in the related industry. An existence of a formal two way communication between management and trade unions will ensure that right message is properly understood and on time too. Abstract:
From the future the book now veers backwards to the remote past, to the library of Ashurbanipal in Mesopotamia. Despite its antiquity, this library was organized systematically around subjects divided within different ‘shelves’, all collected in a kind of catalogue. The library was destroyed soon after it was made, but its destruction also ensured its long-term survival, by baking the tablets. 30,000 fragments survive, including some of the oldest literature in the world, such as the Epic of Gilgamesh. What does it tell us about the idea of the book today? The chapter considers the organization of knowledge in modern and post-modern libraries, and the recent destruction of libraries, such as at Mosul in Iraq in 2016, just a few miles from Ashurbanipal’s palace in Nineveh.
