O presente estudo tem como objetivo em geral, investigar as condições de inclusão nas duas escolas: Escola Estadual de Santa Catarina e Escola Municipal de Florianópolis que propõem novas políticas e práticas de educação inclusiva de surdos. Tal estudo envolve metodologia mista (qualitativa e quantitativa descritiva) para coleta de dados com o uso de instrumentos como observação em crianças surdas na escola regular da rede pública (Estadual de Santa Catarina e Municipal de Florianópolis); questionário fechado com as crianças surdas e professoras destas crianças; levantamento de materiais, recursos e práticas que facilitam a inclusão do aluno surdo no espaço escolar inclusivo. Os sujeitos desta pesquisa são 14 alunos surdos da escola estadual e 2 alunas surdas da escola municipal. É importante lembrar que apenas 12 alunos surdos da escola estadual responderam os questionários e apenas uma aluna da escola municipal respondeu o questionário. O estudo aponta como principais resultados em relação às categorias da Escola Estadual: a) Percepção dos alunos à professora: 100% dos alunos surdos afirmam que sua professora usa a LS com eles; 75% dos alunos compreendem na comunicação com a professora; 87,18% têm relacionamento positivo com a professora que usa a LS; 80,77% têm relacionamento positivo com a instrutora surda; b) A socialização do aluno surdo na sala de aula da escola estadual: 91,66% afirmam que seus colegas surdos usam a LS e que são compreendidos; 91,74% têm participação positiva na sala de aula com seus colegas surdos e professora; 100% afirmam existir perfeita e absoluta compreensão da LS usada pelos colegas surdos; c) o sentimento do aluno surdo em relação à escola estadual de educação bilíngüe: 100% dos alunos surdos afirmam gostar da escola e que sentem felizes lá; 100% satisfeitam com presença da LS e colegas surdos na escola; 54,17% afirmam que a escola possui condições para suas necessidades tal como a presença da LS para comunicação e material especializado; 50% sentem tristes e frustrados quando não têm artefatos da cultura surda no espaço escolar; 95,83% sentem felizes e conversam bastante com seus colegas ouvintes na escola; 63,88% têm satisfação positiva com os serviços apoios que a escola oferece. Enquanto na escola municipal aponta como principais resultados em relação às seguintes categorias: a)percepção da aluna surda à escola municipal: aluna afirma que sua professora usa a LS e que compreende na comunicação com a professora; aluna surda afirma ter uma relação com a professora; aluna surda compreende na comunicação com a ILS, mas sente nervosa quando relaciona com a mesma; aluna surda diz ter uma relação normal com a instrutora ouvinte e que sente segura com ela; b) socialização da aluna surda na sala de aula da Escola Municipal: aluna surda afirma que seus colegas usam LS, mas não são compreendidos; aluna surda diz ter participação negativa na discussão do grupo e que não consegue acompanhar o grupo; aluna surda depende da ILS para entender as perguntas dos alunos à professora; aluna surda diz compreender o que seus colegas ouvintes falam/sinalizam; c) Sentimento da aluna surda em relação à escola municipal: aluna diz que gosta da escola e que sente feliz lá; ela diz que todos da escola usa mais ou menos a LS; aluna surda diz que acompanha bem o material que a escola oferece, mas sente perdida quando não tem o uso da LS; aluna surda diz que satisfeita em estudar na sala de ouvintes e que satisfeita com os serviços de apoio que a escola oferece. Conclui-se que a escola estadual apresenta mais condições em relação à cultura surda fazendo com que os alunos surdos possam se sentir bem, felizes e interagir bem com o grupo, com a professora e todos da escola, enquanto a escola municipal a aluna surda apresenta dificuldades, atrasos na linguagem e que não consegue acompanhar o grupo. Fica comprovado que no espaço de ensino bilíngüe na presença do professor bilíngüe e pares surdos, a interação e aprendizado do aluno surdo apresentam pontos positivos e que ajudam no desenvolvimento cultural, cognitivo e lingüístico. This study aims to, in general, investigate the new policies and practices of inclusive education of the deaf, as well as the conditions for inclusion of deaf students in the two systems that make up the public schools network of Santa Catarina: the State School of Santa Catarina and the Municipal School of Florianopolis. This study involves mixed methodology (qualitative and quantitative descriptive) for data collection with the use of instruments such as observation in deaf children in regular public schools network (the State School of Santa Catarina and the Municipal School of Florianopolis); closed questionnaire with the deaf children and their teachers; survey of materials, resources and practices that facilitate the inclusion of deaf students at school. The subject of this research are 14 deaf students from the State School and 2 deaf students from the Municipal School. It is important to remember that only 12 deaf students of the State School answered the questionnaires and only one deaf student of the Municipal School answered the questionnaire. It pointed as the main results for categories of State School: a) Perception of students to teacher: 100% of deaf students say that their teacher uses the Sign Language with them; 75% of students understand in communication with the teacher; 87.18% have positive relationship with the teacher who uses the Sign Language; 80.77% have positive relationship with the deaf instructor; b) Socialization of deaf students in the classroom of the State School: 91.66% say that their deaf colleagues use the Sign Language and are included; 91.74% have positive participation in the classroom with their deaf colleagues and the teacher, 100% say there is perfect and absolute understanding of the Sign Language used by the deaf colleagues; c) The feeling of deaf students in relation to State School of bilingual education: 100% of deaf students like the school and say they feel happy there; 100% are satisfied with the Sign Language and deaf colleagues at school; 54.17% say that the school has conditions for their needs as the Sign Language for communication and specialized equipment; 50% feel sad and frustrated when they have no artifacts of deaf culture at school; 95.83% feel very happy and chat with their fellow listeners at school; 63.88% are positively satisfied with services support that the school offers. While at the Municipal School it points as main results of the following categories: a) Perception of the deaf student to the Municipal School: the student says that her teacher uses the Sign Language and she understands in communication with the teacher; the deaf student affirms to have a good relationship with the teacher; the deaf student understands in communication with the Sign Language Interpreter, but feels nervous when related to the same; deaf student said to have a normal relationship with the listener instructor and feels safe with her; b) Socialization of the deaf student in the classroom of the Municipal School: the deaf student says that her colleagues use Sign Language, but they are not included; the deaf student says she has a no involvement in the group discussion and that she can not follow the group; the deaf student depends on the Sing Language Interpreter to understand the questions of students to teacher; the deaf student says that she understands what her listener colleagues speak / sign; c) Feeling of the deaf student in relation to the Municipal School: student says she enjoys school and she feels happy there; she says that everybody at school uses the Sign Language more or less; deaf student says that she follows well the material that the school offers, but she feels lost when there is no use of the Sign Language; deaf student says she is satisfied studying in listeners' room and that she is pleased with the support services that the school offers. It follows that the State School has more conditions related to deaf culture so that deaf students can feel good, happy and interact well with the group, with the teacher and everybody at school, while in the Municipal School the deaf student presents difficulties, delays in language and she can not follow the group. It is proven that in the area of bilingual education in the presence of bilingual teacher and deaf peers, the interaction and learning of deaf students have good points and that help in cultural, cognitive and linguistic development.

Abstract:
En aquest treball es caracteriza per primera vegada la capacitat de coordinació metàl·lica d'una metal·lotineïna (MT) de planta i es proposa un model de plegament per a les MTs de planta en general. Els resultat mostren que aquestes proteïnes poden tenir un paper molt important en la regulació de l'estat redox de les cèl·lules, probablement a través de la coordinació a Cu.
Les MTs de planta són proteïnes molt desconegudes. Es postula que participen en l'homeòstasi del Cu i en la protecció contra l'estrès oxidatiu, però es desconeix la capacitat de coordinació metàl·lica i el plegament.
En aquest treball s'han estudiat una metal·lotioneïna d'alzina surera, QsMT, aïllada d'una llibreria de cDNA de fel·lema. Els objectius concrets han estat: (1) estudiar l'expressió de QsMT i la resposta a l'estrès oxidatiu; (2) determinar la capacitat de coordinació metàl·lica i la funcionalitat in vivo; (3) fer una aproximació al plegament de les MTs de planta.
L'expressió del gen s'ha estudiat mitjançant hibridació in situ en plàntules i en embrions d'alzina surera. QsMT s'expressa majoritàriament en cèl·lules amb fort estrès oxidatiu, associat a la síntesi de polifenols (suberització i lignificació) i a la senescència. També s'expressa en cèl·lules meristemàtiques, cèl·lules en divisió molt activa on la funció de les MTs podria estar relacionada amb el manteniment de l'estat redox. L'aplicació d'estrès oxidatiu exogen (H2O2 i paraquat) incrementa fortament l'expressió de QsMT en teixits amb expressió constitutiva, confirmant la regulació de l'expressió del gen per estrès oxidatiu.
Per l'estudi de les propietats de coordinació metàl·lica es va expressar QsMT en cèl·lules d'E. coli en medi de cultiu suplementat amb Cu, Zn o Cd. Es van aïllar els agregats metàl·lics corresponents i es van analitzar mitjançant tècniques espectroscòpiques i espectromètriques (ICP-OES, ESI-MS i CD). Els resultats mostren que QsMT coordina de forma estable Cu (8 ions metàl·lics/molècula), Zn (4 ions de Zn/molècula) i Cd (6 ions de Cd/molècula), i adopta una estructura especialment quiral en coordinació a Cu. L'elevada capacitat quelant de la proteïna i la quiralitat de l'estructura indiquen que QsMT possiblement té preferència metàl·lica pel Cu i per tant una funció relacionada amb aquest metall in vivo. Estudis de complementació en llevat demostren que QsMT coordina Cu de forma funcional in vivo. En coordinació a Cd QsMT presenta una peculiaritat no observada fins ara en altres MTs: la participació d'ions sulfur en la formació de l'agregat metàl·lic incrementant la capacitat de coordinació metàl·lica (6 ions metàl·lics divalents de Cd enlloc de 4 ions de Zn). A més QsMT coordina Cd de forma funcional en llevat, i per tant la seva funció també podria estar relacionada amb la destoxicació de Cd en la planta.
QsMT s'ha utilitzat com a model per fer una aproximació al plegament de les MTs de planta. Amb aquest objectiu vam dissenyar tres pèptids mutants derivats de QsMT: N25 corresponent a la zona rica en cisteïna en posició amino-terminal, C18 corresponent a la zona rica en cisteïna en posició carboxil-terminal, i N25-C18 corresponent a les dues zones riques en cisteïna enllaçades per 4 glicines substituint la zona central de 39 aminoàcids. Es van expressar i estudiar aquests pèptids per les mateixes tècniques utilitzades en l'estudi de QsMT. Els resultats indiquen que QsMT es plega formant un sol agregat metàl·lic per la interacció de les dues zones riques en cisteïna. En aquest model la zona central d'enllaç, típica de les MTs de planta, no participa en la coordinació metàl·lica però és imprescindible per a la funció de la proteïna. El paper de la zona central podria variar en funció del metall que coordina, participant en el plegament i estructura de la proteïna quan coordina Zn i Cd i en la seva regulació i estabilització quan coordina Cu.
In this work we characterize for the first time the metal coordination capacity of a plant metallothionein (MT), and propose a model for the protein folding of plant MTs in general. Results show that these proteins can play a very important role in the regulation of the cellular redox, probably through its copper-coordination ability.
Plant MTs are largely unknown proteins. It is postulated that they play a role in copper homeostasis and protection against oxidative stress, but precise function of these proteins, as well as its metal coordination capacity remain unknown.
In this work we have studied a cork oak metallothionein, QsMT, isolated from phellem cDNA library. Our main objectives were: (1) to study the QsMT gene expression and its response to oxidative stress; (2) to determine the protein metal coordination properties and the functionality in vivo; (3) to estimate a type of folding for plant MTs.
The sudy of the gene expression has been carried out by in situ hybridization in cork oak plants and embryos. QsMT is mainly expressed in cells with high oxidative stress associated to both polyphenol synthesis (suberization and lignification) and senescence. QsMT is also expressed in meristematic cells, where its function could be related to the redox regulation. The oxidative treatment (H202 and paraquat) highly increases gene expression in tissues with constitutive expression, confirming its regulation by oxidative stress.
To characterize the protein metal coordination properties we expressed QsMT in E. coli cells in culture medium supplemented with Cu, Zn or Cd. We isolated the corresponding metal aggregates and analyzed them using spectroscopic and spectrometric techniques (ICP-OES, ESI-MS, CD). Results show that QsMT effectively binds Cu (8 ions/molecule), Zn (4 ions/molecule) and Cd (6 ions/molecule), and presents highest chirality when folds with copper. The high binding capacity and chirality of the protein in the presence of copper suggest that QsMT may play a function related to this metal in vivo. Functional complementation studies confirm that QsMT functionally binds copper in vivo. When coordinating Cd QsMT presents a peculiarity not seen in other MTs: the participation of sulfur ions in the formation of the metal aggregate, this increasing the coordination capacity (6 Cd/molecule in contrast to 4 Zn/molecule). Moreover, QsMT functionaly binds Cd in yeast, suggesting that the protein function could also be realted to a Cd detoxification role in plant.
We have also used QsMT as a model for the study of the folding of plant MTs. With this aim we designed three mutant peptides derived from QsMT: N25 corresponding to the amino Cys-rich domain; C18 corresponding to the carboxy Cys-rich domain; and N25-C18 corresponding to the two Cys-rich domains linked by 4 glycines in substitution to the central 39 amino acid region. Peptides were expressed and studied using same techniques used for the study of QsMT. Results point to a protein folding in one metal cluster formed by the interaction of the two Cys-rich domains. In this model the central region, typical from plant MTs, does not participate in metal coordination but is indispensable for the protein function. The role of the central region could depend on the metal it coordinates, playing a role in the protein folding and structure when it binds Zn or Cd and a role in protein regulation and stability when it binds Cu.

Foram utilizadas carcaças residuais da filetagem industrial de tilápias (Oreochomis sp.) para obtenção de surimi. Amidos de diferentes fontes, milho ceroso, milho ceroso modificado e mandioca, e o polissacarídeo carragena foram usados como aditivos no surimi e estudados seus efeitos no comportamento do gel de surimi. Uma comparação através de análises histológicas do músculo íntegro de tilápia e de surimi de tilápia, indicou que algumas estruturas celulares (núcleos) e fibras musculares permaneceram intactas durante o processamento do surimi, no entanto, o diametro e a disposição das fibras musculares foram alterados, devido ao processamento e adição de aditivos crioprotetores. O estudo por Calorimetria Diferencial de Varredura (DSC) dos sistemas gelificados amido/água e amido/carragena/água demonstrou ter a carragena exercido uma diminuição na quantidade de calor (H cal/g) fornecida ao sistema, por outro lado a temperatura de transição sol-gel não foi alterada, sendo também monitorada por amilograma Brabender. O estudo por DSC do surimi de tilápia apresentou três transições térmicas: 57,11°C, 61,62°C e 65,66°C, devido à presença do complexo actomiosina. A termogravimetria (TGA) mostrou ter o surirni, entre os sistemas estudados, a mais alta capacidade de retenção de água sob aquecimento, seguido pelo sistema surimi/fécula de mandioca/carragena e surimi/fécula de mandioca. A análise instrumental de textura apresentou um efeito fortalecedor dos amidos no gel de surimi, sendo esse efeito proporcional à viscosidade dos amidos, por outro lado, tanto os amidos como a carragena apresentaram uma diminuição da viscoelasticidade do gel de surimi.

Abstract:
Abstract The Surma basin is a part of the Bengal Basin situated in northeastern Bangladesh. The presence of eight gas fields and one oil field makes this an area that is interesting both economically and geologically. In spite of detailed geological and geophysical investigations, information available on palynostratigraphy and geochemistry for the area is scanty. The aim of the present work was to investigate the palynological assemblages, mineralogy and geochemistry of the Surma Group (SG) sequences in Surma Basin, Bangladesh. Core samples (n = 188) were gathered from the wells following: Patharia well-5, Rashidpur well-1, Atgram well-IX, Habiganj well-1, Kailastila well-1 and Fenchuganj well-2. They were provided by BAPEX (Bangladesh Petroleum Exploration Company). X-ray Fluorescence (XRF), Atomic Absorption Spectrometry (AAS), Loss of Ignition (LOI), X-ray diffraction (XRD), Transmission Electron Microscopy (TEM) and Scanning Electron Microscopy (SEM) were used for geochemical and mineralogical study of shale samples. In the palynological study, the distributions of pollens and spores were determined. For data analysis, SPSS computer programme was used. Palynological assemblages of the Surma Group of sedimentary sequence of Bangladesh include taxa range in age from the lower Miocene to the Upper Miocene which can be potentially used in dating and correlation. The Lower Miocene interval is correlated with the Simsang Palynological Zone IV of Meghalaya, India and the Bengal Palynological Zone (BPZ) V. The Upper Miocene is correlated with the Simsang Palynological Zone IV of Meghalaya, India and the BPZ Zone V of Bengal. They were deposited in two types of paleoenvironments ranging from the brackish type to shallow marine to brackish. The sequence contains reworked palynomorphs of BPZ IV and III namely Meyeripollies naharkotensis, Polypodiesporites Oligocenecus, Palmepollenities Eocencus and ornamented Tricolpate pollen of the Eocene-Oligocene age which are mainly encountered in the lower Miocene sediments indicative of increased tectonic activity in the area. Geochemical ratios (SiO2/ Al2O3, Cu/Zn, Maturity = K2O+ Al2O3/Na2O+MgO, Rb/K2O, K2O/Na2O, Cr/Rb, Zr/Rb, V/Rb, Th/U etc.) were useful for determining grain size, maturity, tectonics and environment of deposition. High Ba enrichment was detected in the Patharia well-5 and showed high surface water productivity and diagenetic mobilisation. Tectonic descrimination was achieved using SiO2 and K2O/Na2O ratio. XRD analysis revealed the minerals kaolinite, illite, chlorite, illite/smectite (I/S) and kaolinite/smectite (K/S) mixed layers. Kaolinite/Smectite here reported for the first time in Bangladesh. Clay mineral analyses provided evidence for diagenesis. Smectite diagenesis and dehydration have contributed to the generation of overpressure in the Bhuban Formation in the Patharia well -5. Geochemical ratios of the present study from the Surma Basin is undoubtedly a powerful technique and can be applied to any sedimentary basin analysis to infer the palaeoenvironment, palaeoclimate and palaeotectonics.

Esta dissertação, vinculada aos Estudos Culturais em Educação e aos Estudos Surdos, procura responder à pergunta “Que representações sobre a diferença surda circulam nas produções literárias dos alunos do curso de graduação em Letras-Libras?”. Objetiva descrever o material empírico utilizado na pesquisa; investigar as recorrências e singularidades nas narrativas sobre as experiências de ser surdo; e analisar as representações sobre a diferença surda que circulam nas produções literárias dos alunos do curso de graduação em Letras-Libras, através da análise de trinta e sete (37) vídeos, entre poesias, fábulas, piadas e outras narrativas, elencadas a partir da disciplina de Literatura Surda. Tendo como conceitos-ferramenta para as análises aqui postas, a representação (HALL, 1997), identidade e diferença (WOODWARD e SILVA, 2009) percebo a literatura surda como produtiva no sentido de constituir modos de ser surdo e de experienciar a surdez. Fica evidente, durante a análise, o papel dessas produções em demarcar espaço de resistência aos modos de narrar os surdos a partir da falta e da deficiência, denunciando práticas excludentes, dando ênfase ao papel da comunidade e da língua de sinais para a constituição do sujeito surdo; o valor em registrar as narrativas da comunidade surda em Libras, oportunizando aos surdos dizerem sobre si e sobre suas relações com os diferentes. Ainda, observa-se uma exaltação da língua de sinais para além do valor comunicativo, como um meio de produção e registro cultural, assim como principal fonte para uma mudança de olhar sobre os sujeitos surdos.
This dissertation, linked to the Cultural Studies in Education and to the Deaf Studies, tries to answer the question "What representations of deaf difference circulate in literary productions of students of the Languages-Libras undergraduatestudents?".The goals are to describe the empirical material used in the research; to investigate recurrences and singularities in the narratives about the experiences of being deaf; and to analyze the representations of deaf difference that circulate in literary productions of students of the Languages-Libras undergraduate students, through the analysis of thirty-seven (37) videos, like poems, fables, jokes and other narratives, listed from Deaf Literature class. Representation (HALL, 1997), identity and difference (WOODWARD and SILVA, 2009) are concepts used as tools for the analysis of materials; also I consider deaf literature as productive to establish ways of being deaf and to experience the deafness. It becomes evident, during the analysis, the role of these productions to mark a space of resistance to narratives that view the deaf from a perspective of absence and disability, denouncing exclusionary practices, emphasizing the role of the community and of sign language for the constitution of the deaf subject; the value in registering narratives of the deaf community in Brazilian Sign Language, providing the deaf people opportunities to signs about themselves and their relationships with different ones. There is also an exaltation of sign language in addition to the communicative value as a means of cultural production and registration, as well as the main source for a change of view about deaf people.

Abstract:
Aquesta tesi es centra en la caracterització funcional d'una proteïna de xoc de calor de baix pes molecular (Small Heat Shock Protein - sHSP) de classe I de surera pel que fa a la seva capacitat per protegir les cèl·lules de l'estrès i per estabilitzar les membranes. Les sHsps són proteïnes que s'expressen en condicions d'estrès cel·lular. Encara que certs aspectes funcionals de les sHsps són ben coneguts, el nostre treball aporta informacions noves sobre el paper de les diferents regions de la proteïna, especialment de la regió N-terminal.
L'objectiu concret d'aquest treball és determinar la funció termoprotectora de QsHsp17.4-CI, una sHsp de classe I oobtinguda a partir de les cèl·lules de fel·lema d'alzina surera, en un model bacterià i analitzar la importància de les diferents regions de la proteïna en aquesta funció. Amb aquesta finalitat s'han dissenyat dues proteïnes parcials derivades de QsHsp17.4-CI: una a la que li falta la regió N-terminal (C105) i una altra amb pràcticament tot el domini -cristal·lí deleccionat (N61), i una tercera, derivada de QsHs10-CI, a la que li falta la meitat del domini -cristal·lí (Hsp10). També s'estudia la possible capacitat estabilitzadora de membranes i la capacitat de modificar l'expressió d'altres Hsps quan s'expressa de forma heteròloga.
Els nostres resultats demostren que l'expressió de QsHsp17.4-CI protegeix a les cèl·lules d'E.coli de l'estrès tèrmic alhora que la regió N-terminal i la regió consens II del domini -cristal·lí són imprescindibles per aquesta funció de protecció.
En relació a un possible paper en les membranes, els estudis de localització subcel·lular mostren que QsHsp17.4-CI colocalitza amb la fracció membranes i que la regió N-terminal de la proteïna és responsable d'aquesta colocalització. No s'ha pogut demostrar, però, que la localització amb la membrana estigui associada a un efecte protector d'aquesta: en cap cas la sobrexpressió de les proteïnes modifica la composició d'àcids grassos i només N61, que no té acció termoprotectora, altera l'estat fisico-químic de la membrana.
En estudis d'expressió de novo en E.coli s'ha observat que, a diferència de les altres proteïnes heteròlogues, N61 activa l'expressió de la majoria de Hsps d'E.coli fent pensar en una possible relació entre l'estat físic de la membrana i l'activació de la resposta a l'estrès.
En resum, en aquest treball hem provat la capacitat protectora de QsHsp17.4 i aportem noves dades sobre la importància de la regió N-terminal i la regió consens II del domini -cristal·lí en aquesta funció. Per altra banda, es suggereix que QsHsp17.4 podria interaccionar amb la membrana d'E.coli i que la regió N-terminal seria imprescindible per aquesta interacció. Finalment hem determinat que les proteïnes que provoquen variacions en l'estat de fluïdesa de la membrana poden activar la resposta al xoc de calor per part de la cèl·lula bacteriana.
This thesis is focused in the functional studies of a Small Heat Shock Protein (sHsp). sHsps are expressed under stress conditions. Although some functional aspects of these proteins are known, our work aport new data about the role of the different protein regions, especially the N-terminal region.
The aim of this work is to demonstrate a thermotolerance effect of QsHsp17.4-CI in bacterial cells and to analyze the importance of the protein regions in this function. To achieve this objective two deletion mutants derived from QsHsp17.4-CI were designed: a protein lacking the N-terminal region (C105) and a protein where the entire -cristallin domain is missing (N61) and a third mutant, derived from QsHsp10-CI, that bears half of the -cristallin domain (Hsp10). To better understand the functional mechanism of sHsps we study the membrane stabilizing capacity of QsHsp17.4-CI as well as its capacity to modify other Hsps expression.
Our results demonstrate that the expression of QsHsp17.4-CI protects E.coli cells from a heat shock and that the N-terminal region and the consensus region II of the -cristallin domain are necessary for this protective function.
Related to a possible role in membranes, location studies suggest that QsHsp17.4-CI colocalizes with cell membrane fraction and that N-terminal region is important for this location. However, no relation between membrane localization and a protective effect has been demonstrated: Protein overexpression does not modify membrane fatty acid composition and only N61, which has no thermoprotection, changes membrane physical state.
Studies of E.coli de novo synthesis show that, unlike the other recombinant proteins, the overexpression of N61 activates the expression of almost all E.coli Hsps suggesting a possible relation between membrane physical state and the activation of the heat shock response.
As summary, in this work we have demonstrated the thermoprotective capacity of QsHsp17.4-CI and we contribute with new data about the importance of N-terminal region and consensus region II of -cristallin domain for this function. On the other hand, we suggest the possibility that QsHsp17.4-CI interacts with membrane and that N-terminal region is important for this interaction. Lastly, we have observed how changes in membranes fluidity state can activate heat shock response in bacterial cells.

Skills Development through education and training has always formed an integral part of the development of individuals and the productivity and competitiveness of institutions. Education in South Africa, especially in the Apartheid Era, was a hotly contested area. Since the African National Congress (ANC) came into power in 1994, a new skills strategy was introduced and three important pieces of legislation were passed into law, namely the South African Qualifications Authority (SAQA) Act, the Skills Development Act and the Skills Development Levies Act. The Skills Development Levies Act in effect forces organisations to provide their employees with training and provides for the reimbursement of their skills levies in the form of grants only if certain training criteria are met. The financial implications of the training and development of employees in South Africa comprise of costs and benefits. Human resource managers are under increased pressure to validate the, often substantial, expenses and the effect they have on the bottom line of the company. In an environment where competition is ever increasing and in the light of the financial impact of the new legislation, the financial viability of an investment in training and development is being questioned. A corporate pharmacy group raised a similar question. The general objective of this research was to determine the financial viability of the training and development of pharmacist's assistants in this corporate pharmacy group. The study consisted of a literature study and an empirical survey by means of a survey questionnaire to measure the impact of training as perceived by pharmacist's assistants and their supervising pharmacists and a questionnaire used during structured interviews with the key personnel of the corporate pharmacy group. The results of the interviews and an examination of the financial records of the pharmacy group were used to calculate the financial viability of the training programme for pharmacist's assistants. The results showed that the training programme for pharmacist's assistants in the corporate pharmacy group is financially viable for the group. Furthermore, a performance evaluation on the training programme indicated that the programme adds value to the group by yielding a return in excess of the required rate of return. Based on the results of this study, it is recommended that the pharmacy group should continue providing the pharmacist's assistants with in-house training because it is not only financially viable, but provides other intangible benefits. It is further recommended that the personnel of the pharmacy group have to be informed about the benefits of determining the return on investment of a training programme and trained in the implementation thereof. More effort should be put into the measuring of improvement as a result of the training programme at different stages of implementation of the programme. Further attention should also be given to the allocation of training costs within the pharmacy group.

Abstract:
Eight samples of varying qualities and ages of surimi were used in the development of a new method for testing the quality of surimi. The effects of salt, pH, concentration, heating temperatures and times, and cooling times were observed. These results were used in the development of the McRae-Manning Test. By employing this method, it was possible to determine the difference between fresh high quality surimi, fresh low quality surimi, and old surimi. For this test, surimi was mixed at a 15% concentration, heated at 90 C for 20 minutes in plastic syringes, cooled and evaluated. The surimi was evaluated by emptying the samples onto prepared transparent sheets and measuring how far the sample spreads with time. The transparent sheets had circular measurements which indicated the amount of spread. Best results were obtained when the sheets were elevated at one end.

Although Attention-Deficit/Hyperactivity Disorder (ADHD) is one of the most prevalent childhood disorders, occurring in about three to five percent of the school-going population, there is a dearth of information in literature concerning ADHD learners' intellectual strengths and weaknesses and concomitant learning preferences. An abundance of literature sources can, however, be traced dealing with ADHD learners' behavioural and scholastic problems. Because of this predominantly negative focus on ADHD, these learners are often misunderstood and didactically neglected by teachers in regular classrooms. In 1983, Howard Gardner proposed a new model for understanding intelligence, namely the Theory of Multiple Intelligences (Ml). He stated that a person can be intelligent in more than one way and identified eight intelligences, namely linguistic-verbal, logical-mathematical, visual-spatial, musical, bodily-kinaesthetic, interpersonal, intrapersonal and naturalist. Ml theory provides teachers with a positive model for understanding, supporting and accommodating ADHD learners better in classrooms. The aims of the research were to determine: - what the Ml profiles of ADHD learners reveal in terms of their intellectual strengths and weaknesses; - whether the Ml profiles of ADHD and non-ADHD learners differ significantly; and - what the implications of ADHD learners' Ml profiles are for their teaching and learning. With a view to achieve these aims, a literature study and an empirical investigation were undertaken. The literature study focussed on ADHD, Ml theory and its implications for the teaching and learning of ADHD learners. \n the empirical section of the research, a self-report questionnaire (MIDAS-KIDS) was administered to a group of ADHD and non-ADHD learners who attend five Section 21 primary schools (ex-model C schools) in the D12 school district (Roodepoort) of the Gauteng Province. The data was statistically analysed and the following findings were made: - The Ml profile of the ADHD learners revealed no visible intellectual strengths and weaknesses. - With the exception of Writing and Reading, no differences of real practical significance were observed in the Ml profiles of the ADHD and non-ADHD participants. The following conclusions were drawn, based on these findings: - The MIDAS-KIDS is a measure of perceived intellectual disposition and because factors such as positive illusory bias (PIB) may cause disparities between ADHD learners' perceived and demonstrated intellectual competence, Ml profiles need to be reviewed and interpreted carefully against the backdrop of other diagnostic information when decisions are made with regard to ADHD learners' intellectual strengths and weaknesses. - Although with the exception of Writing and Reading, no differences of real practical significance were observed in the Ml profiles of the ADHD and non-ADHD learners, it is still important that teachers and other professionals take cognisance of Ml theory and its application potential for the optimal intellectual development of ADHD and other learners in classrooms.

Desde el Campo de los Estudios Culturales, Estudios Sordos, esta tesis considera la persona sorda en la condición de diferencia lingüística, teniendo como referencia de sus relaciones con el mundo, con los demás y consigo mismo, la experiencia de ser hablante de una lengua de señas posible a través de la experiencia visual. Busca producir un alejamiento de la tradición de Educación Especial y pone en condiciones, por las prácticas discursivas, piensar la sordera como "invención". Intenta, por el análisis de los materiales producidos por los sordos en el Proyecto de Extensión "Producción de Artefactos de la Cultura Sorda en la Frontera" 2015, configurar la constitución de una Comunidad Sorda de la Frontera que no está representada como una unidad, pero en su movimiento singular, al hacerlo "compartida". Se puede pensar, en esta comunidad "compartida" en vista de un encuentro de sordos fronterizos; territorialmente ubicados en la frontera entre Brasil y Uruguay, en Santana do Livramento y Rivera ciudades gemelas. Esta investigación sistematiza una materialidad: - conjunto de narrativas sordas entre otras, materiales realizados para este trabajo - para analizar las formas como los sordos enuncian sus identidades culturales para que se constituyan como Comunidad Sorda de la Frontera. Tomo estas narrativas de la noción de una experiencia visual compartilhada / compartida que ha producido modos de vida y voluntad de ser sordo" en la frontera y ser sordo en una comunidad de sordos. Esta materialidad analizada produce una red discursiva de una comunidad sorda que no consigue decirse de Brasil o de Uruguay. Una comunidad de sordos compuesta por personas con identidades producidas en un contexto de negociación de y en la frontera entre lo que es ser sordo en su país de nacimiento y la necesidad de reconocimiento cultural de pertenencia a algo que es del orden vivido en las reuniones en esta "zona de contacto". En este análisis fue posible identificar cinco aspectos que están elaborando esta comunidad sorda: un movimiento diaspórico movido por la búsqueda de los sordos por la escolarización, considerando la especialidad de la experiencia visual como constitutiva del sordo. Los espacios y lugares de encuentro de la comunidad, entre ellos las instituciones de enseñanza en la frontera, como espacios que proporcionan el encuentro y la articulación de esta comunidad; una agenda de luchas, siendo las manifestaciones políticas movilizadas en conjunto en defensa de la lengua de señas e de las identidades sordas; un sentimiento de pertenencia a la comunidad como un discurso de resistencia e extrañeza en que los sordos se narran fuera de un "padrón sordo de ser" y una última categoría, una nueva constitución que es propia de esta Comunidad Sorda de la Frontera, siendo esta del orden del acontecimiento, producida en el compartir y lejos del entendimiento de consenso, un "artefacto cultural lingüístico" una lengua de señas compartida en la frontera, produciendo en este intersticio la diferencia.
No Campo dos Estudos Culturais em Educação, Estudos Surdos, este trabalho de dissertação toma o sujeito surdo na condição da diferença linguística, tendo como referência de relação com o mundo, com os outros e consigo mesmo, a experiência de ser falante de uma língua de sinais possível através da experiência visual. Busca produzir um afastamento da tradição da Educação Especial e colocar-se na condição de, pelas práticas discursivas pensar a surdez como invenção . Intenta, pela análise de materiais produzidos pelos surdos no Projeto de Extensão Produção de Artefatos da Cultura Surda na Fronteira 2015, dar configuração a constituição de uma Comunidade Surda da Fronteira não representada como unidade, mas em seu movimento singular, num fazer-se compartilhado. É possível pensar nessa comunidade compartida tendo em vista o encontro de surdos fronteiriços, localizados territorialmente na fronteira entre o Brasil e o Uruguai, nas cidades-gêmeas de Santana do Livramento e Rivera. Para tal empreendimento sistematiza-se uma materialidade conjunto de narrativas surdas, dos materiais formados para este trabalho para analisar a partir delas os modos como os surdos enunciam suas identidades culturais para que se constituam nessa Comunidade Surda da Fronteira. Tomo essas narrativas a partir da noção de uma experiência de ser falante de uma língua de sinais possível através da experiência visual compartilhada/compartida que tem produzido modos de vida e vontades de ser surdo na fronteira e de ser surdo em uma comunidade surda. Essa materialidade analisada produz uma rede discursiva de uma comunidade surda que não consegue se dizer brasileira ou uruguaia, uma comunidade surda composta por pessoas com identidades produzidas num contexto de negociação da/na fronteira entre o que é ser surdo em seu país de nascimento e a necessidade de reconhecimento cultural, de pertencimento a algo que é da ordem vivida nos encontros nesta zona de contato . Na análise foi possível identificar cinco aspectos que estão inventando esta comunidade surda: um movimento diaspórico movimentado por uma busca dos surdos pela escolarização, considerando a especificidade da experiência visual como constitutiva do surdo; os espaços e lugares de encontro da comunidade, entre eles as instituições de ensino na fronteira como espaços que proporcionam o encontro e articulação desta comunidade; uma agenda de lutas, sendo as manifestações políticas mobilizadas em conjunto em defesa da língua de sinais e das identidades surdas; um sentimento de pertencimento a comunidade como um discurso de resistência e estranheza em que os surdos se narram fora de um padrão surdo de ser , e, uma última categoria, uma nova constituição que é própria dessa Comunidade Surda da Fronteira, sendo essa, da ordem do acontecimento, produzida no compartilhar e longe do entendimento de consenso, um artefato cultural linguístico , uma língua de sinal compartilhada na fronteira, produzindo neste interstício a diferença.

Nesta tese, desenvolvo a temática sobre a experiência das mãos literárias de sujeitos surdos, geradores de valores culturais e vinculados à Literatura Surda. Tenho como base contribuições de pesquisas desenvolvidas no campo dos Estudos Culturais em Educação e dos Estudos Surdos, tais como as constribuições de Wrigley (1996), Hall (2007 e 2011), Karnopp, Klein e Lunardi-Lazzarin (2011), Ladd (2013), Silva (2014), Larrosa (2015) e outros. A partir da análise de entrevistas realizadas com representantes da comunidade surda, que de alguma forma estão relacionados com a Literatura Surda, investigo suas experiências literárias em línguas de sinais. O problema de pesquisa que apresento para investigação é o seguinte: como vêm sendo narradas as experiências de surdos com as mãos literárias? Tenho como finalidade investigar as representações de Literatura Surda, a partir dessas entrevistas. Deste modo, como objetivo geral pretendo analisar a experiência de surdos com a Literatura Surda, e como sua trajetória influenciou sua constituição como produtor de literatura. A pesquisa tem uma abordagem qualitativa e utiliza entrevistas semi-estruturadas realizadas com nove surdos brasileiros e três estrangeiros. Nos resultados das análises, destaco a forma como os entrevistados utilizam o conceito de Literatura Surda e Literatura em Língua de Sinais, com evidência de utilização das mãos literárias entre os surdos em encontros, festivais e escolas residenciais. As experiências dos entrevistados ocorreram por meio de sua formação literária, que foram sendo absorvidas pela sua identificação cultural e pertencimento à comunidade surda. Por fim o papel da Literatura Surda é narrado como uma forma de compartilhamento em diferentes espaços, tanto em ambientes formais quanto informais, com um público diversificado. Através da análise dos dados foram registradas suas experiências literárias em línguas de sinais. Com esta pesquisa espera-se dar visibilidade à Literatura Surda e às experiências de surdos em mãos literários, contribuindo para a educação de surdos.
The purpose of this thesis is to develop the theme of the experience of the literary hands of Deaf individuals, here seen as the generators of cultural values who are connected to Deaf Literature. As a base, we reference research carried out in the field of Cultural Studies in Education as well as Deaf Studies, from authors such as Wrigley (1996) Hall (2007 e 2011), Karnopp, Klein and Lunardi-Lazzarin (2011), Ladd (2013), Silva (2014), and Larrosa (2015), among others. Through an analysis of interviews with representatives of the Deaf community, we investigate the signed literary experiences of these members who, in some way, are associated with Deaf Literature. The research question that we pose for investigation is the following: how are literary hands narrating Deaf experiences? Our final objective is to examine representations of Deaf Literature, based on these interviews. Accordingly, our general goal is to analyze Deaf experiences with Deaf Literature and how their individual trajectories influenced their formation as producers of literature. Our approach is qualitative and includes semi-structured interviews with nine Brazilians and three foreign, all of whom are Deaf. Among the results of our analysis, we highlight that way in which those interviewed use the concepts of Deaf Literature and Signed Literature, evidenced by their use of literary hands among the Deaf at meetings, festivals and residential schools. The experiences of the interviewees, channeled through their literary formation, are also absorbed into their cultural identification and belonging to the Deaf community. Finally, the role of Deaf Literature is narrated as a form of sharing in different spaces, whether formal or informal, and among a diverse public. Through an analysis of the data, we registered their signed literary experiences. With this research, we hope to give visibility to Deaf Literature as well as to the Deaf experiences signed by literary hands, and in this way contribute to the education of the Deaf.

A dissertação com o tema "Cultura surda e educação escolar Kaingang" tem como foco principal o registro baseado nos sinais Kaingang, desenvolvidos na comunicação dos surdos na escola, em casa e na Aldeia. A pesquisa foi realizada na Escola Indígena de Educação Básica Cacique Vanhkre, situada na Aldeia Sede, município de Ipuaçu, oeste de Santa Catarina. A pesquisa assumiu um caráter participante pois, a pesquisadora desenvolveu um papel triplo, atuando também como docente no período vespertino (1ª a 4ª série) e intérprete de Língua Brasileira de Sinais junto aos surdos Kaingang incluídos numa 6ª série no período matutino. Sem priorizar uma análise Lingüística, buscou-se estudar o processo cultural de criação e uso de signos pertinentes aos significados da cultura Kaingang, na interação dos surdos da comunidade com a Língua Brasileira de Sinais, também conhecida como Libras e utilizada como língua oficial das comunidades surdas no Brasil urbano. Com o olhar voltado a esta comunicação (os sinais Kaingang e a LSB), a pesquisa buscou identificar os elementos culturais que constituem a identidade dos surdos Kaingang e analisar os contextos em que os sinais lingüísticos surdos próprios à cultura Kaingang se legitimam e se entrelaçam com a LSB. O contexto para qual não poderia ter sido mais adequado: a escola. Ambiente propício ao registro, a interpretação, a análise contextual dos sinais e das formas de comunicação entre surdos na comunidade Kaingang. O que possibilitou o estudo das representações que se fazem destes surdos na comunidade, assim como os processos de identificação cultural "Surdo/Índio". A lógica de análise fundamenta-se no reconhecimento das diferenças como alteridade, o que requer uma visão de cultura, de povo, de língua e de humano que supere a lógica bipolar (certo ou errado, feio ou bonito, superior e inferior), que ao longo da história tem servido como chave de interpretação das relações interculturais. The thesis "Deaf culture and Gaingang scholar education " has its main focus based on the analysis about Kaingang's Sign Language used by deaf members from the tribe aiming to communicate at home, school and also in their local community. The research took place at their aboriginal school "Escola Índigena Estadual Básica Cacique Vanhkre", in Ipuaçu, a town in the west of Santa Catarina. The researcher took on a triple role - besides being a researcher, she was also an interpreter during the mornings for some deaf children included in a Junior High Group and used to teach a Junior Group composed by deaf students during the afternoons. That is why this work has assumed a partaker nature. It is important to mention that this work is not a Linguistic Analyze. Herein the researcher considers the interface between signs belonging to Kaingang Culture and Brazilian Sign Language, also known as "Libras" (from "Língua Brasileira de Sinais"), used as the official language for deaf communities from the urban Brazil. Focusing on this particular communication (Kaingang signs and Libras), this work identifies some cultural elements responsible for building up Kaingang deaf's identity and its interaction with Libras according to the real context in which these specific signs, belonging to Kaingang culture, take place. The setting chosen for developing this research could not fit better: the school. There, it was possible to follow some abilities as writing, interpretation, contextual analyzes and different possibilities of communication between deaf people among themselves and among others into their local community and their processes of cultural identity "Deaf/Aborigine" as well. The logical process of this analyze is based on differences as alterity, for example. What needs an overall view about culture, people, language and also about some concepts of human that overcomes the bipolar logical (right or wrong, ugly or pretty, superior and inferior). It means some thoughts that have been used as the keystone for interpreting intercultural relationships.

Myofibrillar proteins, the main components that impart functional properties to muscle foods, can undergo denaturation and aggregation during frozen storage. The overall objective of this research was to study the changes in protein structure that are associated with the preparation and frozen storage of surimi. In addition, the relative cryoprotective effects of whey protein concentrate, whey protein isolate, soy protein isolate, flaxseed meal and flaxseed protein were assessed in surimi during storage.
Raw surimi was prepared by repeatedly washing Alaska pollock flesh with chilled water. The product was either slowly frozen or underwent rapid freezing using liquid air; in either case it was then subjected to frozen storage at -20°C for 24 months. Protein structural changes were monitored using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), native-PAGE, Fourier transform infrared/attenuated total reflectance (FTIR/ATR) spectroscopy, and differential scanning calorimetry (DSC).
FTIR/ATR spectroscopy showed that during preparation of surimi the alpha-helix content increased with increased number of washing cycles. DSC results revealed a shift in the thermal transition of actin to a higher temperature during surimi preparation. All electrophoresis, FTIR/ATR spectroscopy and DSC results revealed a loss of myofibrillar proteins from surimi after three washing cycles, suggesting that three washing cycles were adequate to prepare surimi.
Native-PAGE showed no major changes in surimi after 24 months storage at -20°C. SDS-PAGE showed relatively minor changes in protein subunit structure with some loss of the myosin light chains (MLC); myosin heavy chain (MHC), actin and tropomyosin were found to be relatively stable. FTIR/ATR spectroscopy indicated a significant decrease in alpha-helix relative to beta-sheet structure in surimi after 2 years of storage at -20°C. The loss of alpha-helical content was more significant in slowly frozen surimi compared to rapid-frozen surimi samples. DSC results revealed a shift in the thermal transition of actin to lower temperatures during frozen storage of surimi.
Changes in the ratio of alpha-helix to beta-sheet structures suggested that flaxseed protein was the most effective cryoprotectant, followed by whey protein isolate and soy protein isolate, for maintaining protein structure stability during frozen storage. Whey protein concentrate and flaxseed meal showed the least cryoprotective ability. After 15 days storage at 4°C, the SDS-PAGE results showed that flaxseed protein was the only cryoprotectant that prevented the degradation of myosin heavy chain, actin and myosin light chains.

Resumo: A década de 1950 é um dos momentos-chave da história da música popular brasileira. Nesse período, diferentes ramos da incipiente indústria cultural se modernizam e começam a se articular de modo mais orgânico. Além disso, recrudesce a segmentação da produção musical, formando-se audiências específicas para cada gênero e estilo em circulação nos principais meios de comunicação. É nesse contexto que surge o Trio Surdina, conjunto ligado num primeiro instante a um programa de rádio que tencionava reproduzir a sonoridade e o ambiente intimista das pequenas boates que então proliferavam na zona sul do Rio de Janeiro. Sua produção se insere na chamada era da "Alta Fidelidade" - tradução literal de High-Fidelity, mais conhecida pela abreviação Hi-Fi -, um momento marcado pela introdução de novos suportes, equipamentos e técnicas de gravação no âmbito da indústria fonográfica. A partir do estudo dos elementos e procedimentos constituintes das performances registradas nos LPs do Trio Surdina, este trabalho investiga em que medida a sonoridade do conjunto informa a existência de uma nova sensibilidade auditiva, de um novo "ouvido social" que se configura entre as décadas de 1940 e 1950, ligado no caso a uma fração da classe média que esteve no centro da vida sociocultural da zona sul carioca. Conforme indicam os discursos da época, certos agentes sociais estavam em busca de um tipo de música mais intimista, "moderna" e de "bom gosto", pautada pela economia de elementos, pelo tratamento "suave" e por uma "técnica aprimorada" de interpretação. A sonoridade concebida pelo Trio Surdina parecia corresponder a tais expectativas, destacando-se no segmento dos chamados "conjuntos de boîte". Em síntese, procurou-se compreender de que modo as obras musicais traduzem e condensam em suas estruturas experiências estéticas, sociais e históricas mais amplas. Nessa direção, destacou-se, ao mesmo tempo, o conteúdo propriamente original e específico da produção do Trio Surdina, isto é, aquilo que, nas músicas, escapa tanto aos padrões e convenções de estilo, quanto aos valores e juízos intrínsecos aos discursos do seu tempo, permanecendo em estado de potência como possibilidades expressivas elaboradas a partir da linguagem da música instrumental
Abstract: The 1950s are one of the key moments in the history of Brazilian popular music. At that time, different branches of the incipient cultural industry modernize and begin to articulate in a more organic way. Moreover, there is an increased segmentation of music production, emerging specific audiences for each style in the main media. It is in that context that is formed Trio Surdina, a musical group associated initially to a radio program that intended to reproduce the sonority and the atmosphere of small nightclubs which then proliferated in the southern area of Rio de Janeiro. The group¿s production is closely associated with an innovation of the recording industry at that time, the "High Fidelity", better known by the abbreviation "Hi-Fi". From the formal study of the Trio Surdina¿s work, this thesis investigates how the sonority of this musical group reflects a new "social listening", which seems to emerge between the 1940s and the 1950s linked to a fraction of the middle class that was in the center of social and cultural life of the southern area of Rio de Janeiro. As indicated by discourses of that time, certain social agents were looking for a kind of more intimate, "modern" and "tasteful" music, characterized by the economy of elements, "soft" treatment and a "refined technique" of interpretation. The sonority conceived by Trio Surdina seemed to correspond to such expectations, especially in the segment of the so-called "boîte groups." In short, this research aims to understand how the music of Trio Surdina translates and condenses into its structures aesthetics, social and historical experiences. At the same time, we emphasize the original and specific content of the production of Trio Surdina; in other words, the content that goes beyond the standards and stylistic conventions, as well as the intrinsic values and judgments to the discourses of that time, remaining as potential expressive possibilities elaborated from instrumental music¿s language

A revisão da literatura do presente trabalho apresenta o desenvolvimento e aceitabilidade de novos produtos, o aproveitamento de produtos pesqueiros e massas alimentícias. O objetivo deste trabalho foi determinar a aceitabilidade do macarrão a base de surimi que foi desenvolvido utilizando a carpa, peixe de baixo valor comercial, processada na forma de surimi, elaborando-se com este uma farinha que foi a matéria-prima protéica do macarrão. Foram desenvolvidos dois produtos: o produto 1 contendo 50% de farinha de surimi não alcançou o índice de aceitabilidade de 70%, o teste foi realizado no Colégio Estadual Leonor de Barros com uma população de 350 crianças com faixa etária entre 7 e 14 anos; o produto 2 alterando-se a proporção de farinha de surimi (20%) e adição de semolina ao invés de farinha de trigo, além da introdução de um desodorizador na última água de lavagem da carne de peixe triturada, proporcionou um produto com elevado índice de aceitabilidade (96%), o qual foi testado na Escola Básica Municipal Osmar Cunha com uma população de 450 crianças com faixa etária entre 7 a 14 anos. O macarrão a base de surimi pode ser uma alternativa a mais para a alimentação institucional, além de possibilitar uma maior diversificação da indústria pesqueira.

Les étapes critiques du procédé de fabrication du surimi de chinchards (Trachurus trachurus) ont été identifiées. Ainsi, des solutions techniques et chimiques ont été proposées afin de limiter le développement des réactions d'oxydation des lipides au cours du procédé. Pour ce faire, les méthodes d'analyse des lipides et de leur produits d'oxydation ont été adaptées aux produits étudiés, une méthode de dosage des hydroperoxydes a été mise au point et la technique de fluorescence en mode frontal testée pour évaluer l'altération du surimi. Les cinétiques d'oxydation des lipides ont été déterminées au cours de la conservation des chinchards sous glace ou à 17ʿC. La formation des composés d'oxydation des lipides a été suivie au cours de la fabrication du surimi et de sa conservation à l'état congelé. L'impact de l'état de fraîcheur de la matière première sur l'évolution des réactions d'oxydation des lipides au cours de la fabrication et de la conservation du surimi a également été déterminé
Critical steps of the process of horse mackerel surimi manufacturing were identified. This led to propose technical and chemical solutions in order to limit development of lipid oxidation reactions. In this way, analytical methods of lipids and lipid oxidation compounds were adapted to the studied products. A method to determine hydroperoxyde concentrations was developed. The front face fluorescence technique was tested to evaluate the degree of surimi alteration. Lipid oxidation kinetics were determined during horse mackerel storage on ice or at 17ʿC. The formation of lipid oxidation products was followed during manufacturing and horse mackerel surimi frozen storage. The impact of raw material freshness on the development of lipid oxidation during surimi manufacturing and storage was also determined

Tiivistelmä. Tämän tutkimuksen tarkoitus on selvittää, millä keinoilla lasten kuvakirjojen kuvat ilmaisevat kuolemaa ja surua. Kuolema mielletään usein arkaluontoiseksi aiheeksi, jota vanhempien ja opettajien on vaikea käsitellä lasten kanssa. Siitä on kuitenkin olemassa useita lapsille suunnattuja kuvakirjoja, joita on helppo käyttää apuna kuoleman käsittelemisessä. Olemme rajanneet tutkimuksemme koskemaan vain kuvia, sillä kuvakirjan teksti nähdään usein tärkeämmässä roolissa tarinan kannalta. Haluamme herätellä lukijoita kiinnittämään enemmän huomiota myös kuvakirjan kuviin, jotka voivat kertoa tarinaa myös itsenäisesti. Olemme huomanneet, että lapset selailevat kuvakirjoja myös itsekseen, jolloin he lukevat kirjasta vain kuvia luoden mielikuvaa tarinasta. Kyseessä on teoreettinen tutkimus, jossa on mukana empiirinen osio. Tutkimuksen viitekehyksessä olemme määritelleet kuvaa, kuvakirjaa ja visuaalista lukutaitoa eri teoreetikkojen kautta. Kuva on kaikille tuttu ilmiö, jolla on oma tekstistä eroava tapansa ilmaista asioita. Kuvakirjan lukijan on tärkeä ymmärtää kuvan visuaalisia viestejä, mikä tapahtuu visuaalinen lukutaidon kautta. Tutkimuksen empiirisessä osiossa tutkimme Marjo Räsäsen semioottisen kuva-analyysin keinoin kolmea kuolemaa ja surua ilmaisevaa lasten kuvakirjojen kuvaa. Analyysistä nousi esiin kolme selkeää keinoa, joilla kuva ilmaisee kuolemaa ja surua: symbolit, värit sekä kirjan henkilöiden ja muiden olentojen kehonkieli. Lisäksi löysimme joukon muita keinoja, jotka omalta osaltaan muokkaavat kuvan sanomaa ja tunnelmaa. Tuloksia pohtiessamme nostimme esiin myös näkökulman, että kuvalla on aina lähettäjä ja vastaanottaja. Kuvittajalla ja lukijalla on omat henkilökohtaiset kokemusmaailmansa, joiden kautta he tekevät ja tulkitsevat kuvia. Tutkimus tuo esille sen, että kuvakirjojen kuvat ovat monimerkityksellisiä ja niiden eteen on tehty paljon työtä. Kuvittaja on käyttänyt apunaan monia tarkkaan harkittuja keinoja, joiden avulla hän välittää tunteita kuvan katsojalle. Kuvien merkitystä kuvakirjojen valinnassa lapsille tulisi korostaa. Jo kolme kuvaa tarkastelemalla käy ilmi, että kuoleman kaltaisia rankkoja teemoja on kuvitettu ajatuksella huomioiden lapsi kuvan tulkitsijana.

Esta dissertação objetiva analisar as interações da criança surda em escola comum, a fim de entender o papel das interações no contexto escolar, bem como sua importância para a constituição da linguagem e da aprendizagem. Essa pesquisa, caracterizada como estudo de caso, de cunho exploratório, tem como sujeito uma criança surda, com 10 anos de idade, filha de pais ouvintes, frequentadora do 3º ano do Ensino Fundamental de uma escola pública do Rio Grande do Sul. A definição por uma escola de ensino comum como lócus de investigação justifica-se pela Política Nacional de Educação Especial na Perspectiva da Educação Inclusiva, instituída em 2008, que pretende assegurar a inclusão de alunos com algum tipo de deficiência na rede regular de ensino. Para amparar teoricamente este estudo, é utilizado como norteador o paradigma histórico-cultural de Vigotski. A construção dos dados baseia-se em dois níveis de fontes de evidências: as prioritárias, que consistem nas entrevistas semiestruturadas com os professores e mãe da menina, e o diário de campo, composto por registros das observações diretas em sala de aula; e as de apoio, que são os vídeos registrados nas visitas à escola e os registros de acompanhamento da vida escolar da estudante. Os resultados evidenciam a necessidade de mudanças acerca da visibilidade da criança surda na escola comum. O entendimento da surdez como diferença linguística é imprescindível, pois é por meio desse entendimento que a comunicação e os processos de ensino e aprendizagem serão conduzidos. É possível constatar que a precariedade na comunicação entre a menina e seus interlocutores compromete a qualidade das interações entre eles, assim como interfere de forma prejudicial os processos de aprendizagem da estudante. Os resultados apontam ainda para a necessidade de transformação dos moldes da escola atual, bem como direcionam reflexões importantes quanto à importância da interação, do diálogo e do espaço para as diferenças. O cenário encontrado confirma a necessidade de novas pesquisas e discussões acerca dos aspectos que permeiam a educação, dita inclusiva, dos estudantes surdos.

This Master`s dissertation aims to analyze the interaction of deaf children in regular school environments so as to understand not only their interactions in the school context, but also the importance it takes on the acquisition of language and earning. This research, which is characterized as an exploratory case study, has, as it main subject, a ten-year old child, daughter of hearing parents, who is attending the 3rd grade of a public elementary school in Rio Grande do Sul, Brazil. The choice of a regular education school as the focus of the investigation was based on the National Policy on Special Education from the Perspective of Inclusive Education, implemented in 2008, which is intended to ensure the inclusion of students with special education needs in regular schools. To theoretically support this study, the Vogotski`s cultural-historical paradigm was used as a guide. The collection of data is based on two levels of sources of evidence: the priority areas, consisting of semi-structured interviews with teachers and the girl`s mother, as well as field journal, made up of records of direct observations in classroom; and the support areas, consisting of videos recorded during visits to the school and follow-up records of the student`s daily life at school. The results show the need for changes in deaf children`s visibility in regular schools. Understanding deafness as a linguistic difference is crucial, since it is through this understanding that communication and the teaching-learning process will take place. It was possible to see that the poor communication between the girl and her peers compromised the quality of their interactions, also seriously interfering with the student`s learning process. In addition, the results not only demonstrate that there is a need to modify the mainstream programs of current schools, but also point to the need of serious reflections on the importance of interaction, dialogue and space for differences. The scenario that has been found confirms the need for new investigation and discussions on the aspects permeating education – said as an Inclusive one – and deaf students.

Geóloga
En la XV región de Chile existe un déficit hídrico que, sumado a la creciente demanda de agua, hace necesario estudiar el recurso hídrico. Este estudio se focaliza en la subcuenca endorreica altiplánica del Salar de Surire, y busca comprender el origen las aguas de la cuenca, sus procesos hidrogeoquímicos, y su calidad. Esta cuenca se encuentra constituida principalmente por formaciones volcánicas y por depósitos de clastos volcánicos. En su base se forma un salar con ocasionales cuerpos lacustres. El clima es desértico-subdesértico, en donde en el verano austral ocurren los mayores eventos de precipitación. La escorrentía del salar se encuentra marcada por el Río Surire, vertientes frías menores a lo largo del perímetro del salar, y por una zona de manifestaciones termales en el costado SE. Se analizaron 16 muestras que cumplieron el BI, distribuidas en 3 campañas: 6 muestras de noviembre del 2015, 5 muestras de mayo del 2016 y 4 muestras de agosto del 2016. Su temperatura fluctúa entre los 2,3°C a 46°C, siendo la mayoría de las muestras clasificadas como agua termal. El pH es neutro/alcalino, variando de 6,5 a 9,5. La CE es lo que más varía, desde los 100 μS/cm a los 71.500 μS/cm El tipo de agua varía de bicarbonatadas cálcico-magnésico sódicas a aguas cloruradas sódicas, siendo las primeras de menor mineralización y lejanas al salar, y las segundas de mayor mineralización y cercanas o sobre el salar. A partir del contexto geológico y del análisis de diagramas binarios, se sugiere la presencia de procesos de disolución de carbonatos y sulfatos de calcio y magnesio en la química del agua analizada. Las muestras presentan un exceso de sulfato que puede indicar un aporte diferente al mencionado. Las concentraciones de sílice están directamente relacionadas con la temperatura sugiriendo que la interacción agua/roca puede ser el origen de este elemento. Los datos isotópicos indican que las muestras poseen, en general, un origen meteórico, y algunas presentan procesos de fraccionamiento isotópico asociado a evaporación. Las simulaciones en PHREEQC corroboraron los procesos de interacción agua/roca y disolución evaporítica. Los resultados de las simulaciones geoquímicas permitieron diferenciar las probables litologías en contacto con las muestras estudiadas. Esto, junto con una definición de unidades hidrogeológicas a partir de trabajos anteriores, logró resumir que: depósitos aluviales y fluviales Pa, Ha, y tobas fracturadas de OMI(b), poseen capacidad acuífera; la de los depósitos glaciales Qg es media/baja; la de estructuras volcánicas es variable; y, finalmente, los depósitos evaporíticos del salar pueden poseer capacidad acuífera ya sea a partir de permeabilidad primaria o secundaria. Finalmente, las aguas no cumplen con la norma de agua potable y siete de las ocho muestras tampoco cumplen con la norma de riego NCh1333. La muestra que sí cumple, es posible utilizarla para ese fin con un monitoreo constante asociado.

La premiere partie est consacree a la presentation de chaque piece : il s'agit du lieu et de la date de la premiere representation, du sujet principal ainsi que des sources de la creation des heroines. La deuxieme partie concerne l'analyse des personnages feminins. C'est-a-dire, leur comportement, leur passion et leurs types de caractere sont etudies. Nous deduisons egalement l'interet dramaturgique issu de ces roles feminins. Dans la derniere partie, nous etudions la nature de la femme d'apres les enonces trouves dans les pieces. Nous examinons quel genre d'idees en general apparait dans ces pieces et si ces idees correspondent a l'image des heroines. Par ailleurs, nous etudions la condition feminine de l'epoque depeinte a travers celle des personnages dramatiques. Dans la conclusion, nous voyons l'evolution des heroines depuis les pieces ecrites avant oedipe et supposons les raisons de cette evolution
We study in this thesis the cornelian heroines in his last dramatic works written between 1659 and 1684 except la toison d'or and psyche. The first part of this study is destinated to the presentation of each drama. It means the information about its first performance, its story and the source of the heroines' creation in each work. In the second part, we analyze properly the heroines : their comportment, their passions and their characters. At the same tine we considerate their dramatic value in each work. The last part is given to the study of the woman's nature and her social condition. We see what kind of opinion about the woman's nature are presented in cornelian last dramas and how the dramatist use these opinions in the creation of the heroines. After all we try to find out if the womans' condition in france of the seventeenth century is reflected in these dramas. In the conclusion, we see the evolution of heroines from the first dramas written before ours

A educação escolar das crianças surdas deve ser ofertada nas escolas inclusivas, ou não? Esta pesquisa, cujo título é “Políticas Públicas na Educação dos Surdos: o que se diz, o que se faz, o que os surdos querem”, analisou três eixos da educação dos surdos, na educação infantil e nos anos iniciais do ensino fundamental: O que diz (as políticas públicas de educação inclusiva, vigentes); O que faz (como estas políticas se materializam nas escolas da Rede Municipal de Ensino de Belo Horizonte); O que os surdos querem (o ponto de vista, político e educacional, dos surdos, envolvidos nesse processo). Esse estudo empregou a abordagem qualitativa e utilizou, como procedimentos metodológicos, a pesquisa bibliográfica, documental e entrevistas com profissionais da educação, dando ênfase nos depoimentos das pessoas surdas. Para analisar os dados, buscou-se confrontar os discursos apresentados pelas partes envolvidas, verificando as contradições existentes entre o que se diz, o que se faz e o que as pessoas surdas almejam. Os resultados evidenciaram que a Instância Estadual diz subsidiar a educação dos surdos considerando, principalmente, as diretrizes educacionais indicadas pela Política Nacional de Educação Especial na Perspectiva da Educação Inclusiva (2008). A Rede Municipal de Belo Horizonte, que segue a mesma Diretriz, diz que promove o ensino para as crianças surdas, por meio de ações que subsidia a oferta do ensino da Libras e em Libras; oferta de Atendimentos Educacionais Especiais - AEE, propiciando o apoio educacional complementar necessário para atender às necessidades educacionais desses alunos. Os profissionais surdos revelam que as ações implementadas (o que se faz) não atendem às especificidades desses alunos e, por fim, indicam que os surdos almejam uma educação bilíngue. A pesquisa apontou que há necessidade de se repensar as políticas públicas para a educação dos surdos, considerando o ponto de vista, político e educacional, apresentado pela Comunidade Surda, buscando ressignificar o espaço educacional das crianças surdas, assegurando-lhes o direito à educação que reconheça as diferenças linguísticas, pedagógicas e culturais, ou seja, a oferta de uma educação bilíngue.

Esta pesquisa tem como objetivo pensar de uma forma responsiva e responsável como os sujeitos surdos da Grande Vitória constituem os termos cultura surda e identidade surda nos seus discursos. Através, principalmente, das discussões levantadas pelos Estudos Culturais, a comunidade surda buscou transformar o olhar sobre o sujeito surdo, tradicionalmente concebido pelo viés da medicina, do assistencialismo, da deficiência e da falta de audição, para um viés cultural, em que se valoriza o sujeito crítico, com identidades e língua próprias, ou seja, as identidades surdas e a língua de sinais. Por meio desses estudos criam-se os Estudos Surdos, com o objetivo de fomentar novos conceitos e ideias a respeito da pessoa surda. Como proposta de desenvolvimento nesses estudos, buscamos o pensamento da transculturalidade para incrementar essa pesquisa e trazer novas perspectivas culturais em vista de ainda existirem tantos episódios de intolerância e ódio entre as pessoas. A ideia de uma cultura superior, fechada e que se divide entre “nós” e “eles” muitas vezes acaba por produzir mais segregação e preconceito. Por isso, a proposta da nossa pesquisa é pensar a cultura surda através do transculturalidade, do dialogismo, da escuta do outro e, além disso, repensar os conceitos de identidades e cultura surdas, visando contribuir para a construção de uma sociedade mais sensível à escuta e receptiva ao outro. O corpus desta pesquisa é formado por entrevistas realizadas com representantes da comunidade surda da Grande Vitória que estão inseridos na comunidade acadêmica sobre questões como: identidade surda e cultura surda, que são termos comumente utilizados/pesquisados por estudiosos dos Estudos Surdos. A hipótese elencada é a de que a comunidade surda tem assumido um posicionamento fechado nos conceitos de cultura e identidade surda. Para analisarmos se essa hipótese se sustenta, utilizamos uma metodologia dialógica da escuta com a pretensão de compreender as vozes que perpassam os sujeitos envolvidos nesse processo da pesquisa.

Abstract:
Els organismes responen a la temperatura i a molts altres estressos sintetitzant un grup de proteïnes anomenat proteïnes de xoc de calor (HSPs). En plantes les sHsps, d'entre 15 i 30 kDa formen el grup més abundant i divers, classificat en funció de la seva localització subcel.lular i homologia en: mitocondrials, cloroplàstiques, de reticle endoplasmàtic i citoplàsmiques de classe I i II. Les sHsps-CI s'ha descrit que s'indueixen per estrès tèrmic, hídric i oxidatiu (peròxid d'hidrògen, llum UV, ozó) i en resposta a algunes hormones. També s'expressen durant el desenvolupament, per exemple durant l'embriogènesi, on es creu que podrien tenir un paper protector de l'embrió enfront la dessecació. Tot i que hi ha abundants treballs que correlacionen la resistència a l'estrès i l'acumulació de sHsps-CI, els mecanismes moleculars d'aquesta activitat són poc conguts. Tot i això, per diverses sHsps-CI ha estat descrita una activitat xaperona in vitro i, més recentment, que la seva sobreexpressió augmenta la viabilitat de cèl.lules d'E.coli en condicions d'estrès tèrmic.
L'estudi de l'acumulació de sHsps-CI en surera (Quercus suber) mitjançant immunodetecció en electroforesi bidimensional mostra uns patrons d'acumulació complexos i formats per dos grups d'espècies proteiques principals, a l'entorn dels 10 i 17 kDa respectivament, que mostren una inducció diferencial en funció del teixit i l'estrès. Mentre que les espècies proteiques de 17 kDa s'indueixen per temperatura però no per estrès oxidatiu, les de ca. 10 kDa ho fan per estrès oxidatiu i no per temperatura. Ambdós grups d'espècies proteiques s'acumulen conjuntament en fel.lema. Assajos de PCR i RT-PCR han permès clonar parcialment tres noves sHsps-CI en surera: Qshsp10-CI, QshspC-CI i QshspD-CI. Aquest fet confirma la multigeneïcitat de les sHsps-CI en surera que apuntava el patró bidimensional. Dels nous clons obtinguts destaca especialment Qshsp10-CI, un gen que presenta un codó stop enmig del domini -cristal.lí que fa que a la proteïna que se'n dedueix li manqui un 55% del domini -cristal.lí i tota l'extensió C-terminal. Es tractaria de la sHsp més petita i més truncada descrita fins al moment. L'anàlisi de l'expressió de Qshsp10-CI mitjançant RT-PCR mostra expressió en plantes tractades amb H2O2 però no en les que han estat sotmeses a un xoc de calor. Aprofitant l'oportunitat que oferia aquesta sHsp-CI de ser utilitzada com a model per l'estudi de la importància del domini -cristal.lí i l'extensió C-terminal en l'activitat protectora enfront l'estrès, es va voler determinar la capacitat que tenia d'augmentar la viabilitat de cèl.lules d'E. coli en condicions d'estrès tèrmic i oxidatiu. Els resultats mostren que la proteïna recombinant QsHsp10-CI, tot i la important truncació que té, és capaç de protegir cèl.lules d'E. coli en condicions d'estrès tèrmic i, remarcablement, en condicions d'estrès oxidatiu. Tots aquests resultats indiquen que les espècies proteiques de ca. 10 kDa podrien correspondre a Qshsp10-CI i tenir un paper en les cèl.lules del fel.lema en la protecció enfront l'estrès oxidatiu.
L'estrès oxidatiu provoca lesions al DNA que poden produir errors en la replicació, transcripció o traducció i generar proteïnes aberrants. Donades les condicions d'estrès oxidatiu a les quals es troben sotmeses les cèl.lules del fel.lema, s'ha volgut estudiar la variabilitat dels seus àcids nucleics. La determinació de la taxa de mutació de la regió codificant del gen Qshsp17.4-CI en mRNA i DNA de fel.lema i àpex radicular, un teixit jove i en creixement actiu va mostrar unes taxes sorprenentment elevades en l'mRNA (1/1784 pb) i el DNA genòmic (1/1520 pb) del fel.lema. Aquestes taxes són les més altes descrites en un genoma nuclear eucariota i són similars a les dels virus d'RNA d'evolució ràpida com el virus de l'Hepatitis C. Amb aquestes taxes de mutació, un terç dels mRNAs del fel.lema de la surera contindrien missatges aberrants i la supervivència de les cel.lules es veuria compromesa. Això implica que el fel.lema hauria de ser considerat com un mosaic de cèl.lules genèticament heterogènies i, per tant, una sola seqüència no defineix en tota la seva amplitud un gen en aquest teixit. No es va detectar cap mutació en àpex de rel. Amb l'objectiu d'aprofundir en el coneixement de les mutacions que es donen en aquests dos teixits i per tal de poder fer una anàlisi qualitativa més completa que permetés especular sobre el seu origen, es va aplicar un mètode de selecció de seqüències mutants en base a la utilització d'enzims de restricció. Les mutacions detectades en fel.lema es corresponen amb les relacionades, en altres sistemes no nuclears (plasmidis, fags i DNA bacterià), amb l'estrès oxidatiu. En conseqüència, l'estrès oxidatiu al qual estan sotmeses les cèl.lules del fel.lema podria ser el causant de l'elevada taxa de mutació detectada. D'acord amb això, el tipus majoritari de productes d'oxidació de les bases del DNA que s'acumulen en brots de plàntules de surera en resposta al peròxid d'hidrògen produeixen el mateix tipus de mutacions detectades en l'mRNA del fel.lema de la surera. La major sensibilitat d'aquest nou mètode ha permès, a més, detectar mutacions en molècules d'mRNA de rel, un teixit en el qual no s'havia trobat cap mutació utilitzant el mètode de clonatge i seqüenciació directa. Tot i això, el tipus de mutacions predominants no estan relacionades amb l'estrès oxidatiu sinó amb erros en la reparació dels àcids nucleics.
Small heat shock proteins (sHsps,15-30 kDa) are the most abundant and diversified Hsps in plants. They have been classified according to its homology and cellular localisation in: mitochondrial, chloroplastic, endoplasmic reticulum and class I and II citoplasmic sHsps. Although sHsps-CI are involved in the stress response and accumulate at some stages of embryonic development and there is abundant work correlating stress resisitance and sHsps accumulation, the molecular mechanisms of their activity are not well known. However, in vivo and in vitro chaperone activity under temperature stress has been described.
2D immunodetection patterns of cork oak (Quercus suber) sHsps-CI (thermic, hydric and oxidative). At the 17 kDa region there is a set of protein species highly induced by temperature and, at least some of them correspond to QsHsp17.4-CI. On the other hand, protein species at the ca. 10 kDa region are highly induced under oxidative stress conditions and accumulate in the endogenous oxidatively stressed tissues xylem and phellem but not after a heat shock. PCR and RT-PCR assays allowed us to clone three new members of the sHsps-CI multigenic family in cork oak. Among them, Qshsp10-CI is specially interesting because codes for a truncated protein that lacks 55% of the -crystallin domain and all the C-terminal extension, being the more C-terminal truncated sHsp reported to date. Overexpression of recombinant QsHsp10-CI and a more truncated protein lacking the whole -crystallin domain in E. coli cells shows that most of the -crystallin domain and all the C-terminal extension are dispensable, but amino acids 1 to 41 of the -crystallin domain (including the consensus II region) are essential for sHsps-CI protective activity under temperature and oxidative stress conditions. The expression of Qshsp10-CI in response to oxidative but not temperature stress and its protective activity of E. coli cells under oxidative stress conditions points to a correspondence between Qshsp10-CI and the ca. 10 kDa protein species detected in 2D immunodetections and a protective activity of those in the oxidatively stressed phellem cells.
Endogenous oxidative stress of phellem cells might generate mutations accumulation in nucleic acids. Variability analyses of DNA and mRNA of phellem cells in the coding region of Qshsp17.4-CI showed a surprisingly high rate of mutation in both mRNA (1/1784 bp) and genomic DNA (1/1520 bp). These are the highest rates described for a nuclear eukariotic genome and are similar to those detected in RNA viruses. With these mutation rates one third of mRNAs of phellem cells would contain mutations and code for abnormal proteins. No mutations were detected in root tip, a normally growing young tissue. With the aim of deeping into the nature of mutations that accumulate in phellem cells, we applied a method to in vitro select mutant sequences using restriction enzymes. The types of mutation predominant in phellem cells mRNA were those related with oxidative stress in other systems (plasmids, phages and bacterial DNA). In addition, the predominant DNA lesions that accumulate in H2O2 treated cork oak plantlets, as shown by GC-MS analyses, generate the same type of mutations detected in mRNA of phellem cells. Accordingly, the high accumulation of mutations detected in phellem cells might be due to its endogenous oxidative stress. Moreover, young and actively growing tissues are also subjected to a certain degree of base lesions and mRNA mutations. However, both mutational spectrum and accumulation levels are different compared to oxidatively stressed tissues.

Surimi is a concentrated myofibrillar protein added with several additives. It has been used by seafood industries as an intermediate product to produce various seafood analogues. Due to the current increase in health awareness, consumer demands healthier and less sweeter surimi. The objective of this study is to investigate the rheological and textural properties of surimi added with different types of additives which is sugar, salt and sago starch. This study is also done to determine the possibility of reducing the amounts of additives added to surimi. Sago starch as an additive to improve the rheological and textural property of surimi is studied. Another concern in the surimi industry is the washing process. The 3 washing cycle procedure produces large amount of water waste. Thus, this study is done to understand the effects of reducing the washing cycle and using nanobubble water on surimi. Results obtained from this research shows that mannitol yields the best results when compared to sucrose and sorbitol. Surimi with 4% (w/w) salt concentration yields the highest gel strength. However, surimi with 1% (w/w) salt concentration which possesses a lower gel strength showed frozen stability up to 6 months. Sago starch was found to increase gel strength up to 11 0%. Washing using medium concentrated nanobubble water ( 11.15 x 1 08 bubbles/ml) was found to display a high gelling strength with only 2 washing cycle compared to using distilled water.

La conduite, le contrôle et le devenir de l'interrogatoire posthume sont des questions qui agitent les Marrons ndjuka du Tapanahoni (Surinam) depuis une trentaine d'années. Après avoir été abolie au début des années 1970, cette pratique divinatoire présentée comme ancestrale et qui vise à établir les circonstances du décès d'un individu, a récemment fait l'objet d'une volonté de réhabilitation de la part de certains clans, laissant apparaître des lignes de clivages entre les divers protagonistes de ce qui s'avera, au fil de l'enquête, être un véritable débat politique centré autour du bien-fondé de la reprise de la pratique. En adoptant un point de vue qui visait à révéler tout d'abord la nature (politique) des enjeux de cette controverse, puis à saisir les logiques de la position des villageois parmi lesquels j'ai enquêté, j'ai cherché à expliciter, de manière dynamique, les modalités de l'exercice du pouvoir chez les Marrons ndjuka. Plus largement, ce travail interroge la façon dont un système politique peut prendre appui sur des référents religieux.

This work is central to understand how problematic movements deaf as a space of struggle and resistance is redefining deaf come to building a policy of education for deaf guided joints between sign language and deaf culture. To account for this investment research some goals that guided the study: understanding the emergence of movements in deaf education scenario of the deaf; identify different strategies to fight organized by the movement deaf to understand its effects on deaf education in contemporary. To this he joined the field of Cultural Studies in Education, with crossings of the field of Deaf Studies who understand deafness and deaf education from a cultural and linguistic context specific. The empirical corpus study is divided into two groups of materials: narratives of deaf leaders and a set of documents that consider the conditions of possibility for the emergence of a discourse that has been producing movement deaf from other arrangements, other experiments. These sets of documents are: Document "Deaf Education that we Want" 1999 Project Course Letters / POUNDS, Law 10,436 / 5626 and certainly Movement Bilingual School for the Deaf. To the narratives of deaf leaders, collected through interviews, I could track the movement deaf at two, ie, in two outbreaks of analytical power. The first stage focused on the emergence of the fight for the official recognition of sign language in the constitution of deaf identities and the second in the trading strategies of deaf culture in education. You can check in emergencies and in the possibilities of movement history deaf political organization linked with the education of deaf and negotiation of deaf culture in the contemporary scenario.
O presente trabalho tem como problematização central entender como os movimentos surdos enquanto espaço de lutas e resistências surdas vêm se resignificando para a construção de uma política de educação para surdos pautada nas articulações entre língua de sinais e cultura surda. Para dar conta desse investimento de pesquisa alguns objetivos nortearam o referido estudo: entender a emergência dos movimentos surdos no cenário da educação de surdos; identificar as diferentes estratégias de luta organizadas pelo movimento surdo a fim de compreender seus efeitos para a educação de surdos na contemporaneidade. Para isso filiou-se ao campo dos Estudos Culturais em Educação, com atravessamentos do campo dos Estudos Surdos que entendem a surdez e a educação de surdos a partir de um contexto cultural e linguístico específico. O corpus empírico do estudo está dividido em dois grupos de materiais: narrativas de lideres surdos e um conjunto de documentos que considero com as condições de possibilidades para a emergência de um discurso que vem produzindo o movimento surdo a partir de outros arranjos, de outras experiências. Esses conjuntos de documentos são: Documento Educação que nós Surdos Queremos de 1999 Projeto de Curso Letras/LIBRAS, Lei 10.436/ Decerto 5.626 e Movimento Escola Bilíngüe para Surdos. Com as narrativas dos lideres surdos, coletados por meio de entrevistas, pude localizar o movimento surdo em dois momentos, ou seja, em dois focos de potência analítica. O primeiro momento centrado na emergência da luta pela oficialização da língua de sinais, na constituição de identidades surdas e o segundo nas estratégias de negociação da cultura surda no campo da educação. Pode-se verificar nas emergências e nas possibilidades da história do movimento surdo uma organização política articulada com a educação de surdos e com negociação da cultura surda no cenário contemporâneo.

O presente trabalho trata-se de uma dissertação de mestrado que tem por objetivo investigar os modos de vivência da musicalidade da pessoa surda, considerando sua cultura. entendendo o indivíduo como unidade afeto-intelecto e como ser de possibilidades, com base na perspectiva histórico-cultural de vigotski, a pesquisa visa perceber este indivíduo em sua totalidade, em inteireza. nesta perspectiva, e com base nos estudos da defectologia, a pessoa surda apresenta um defeito biológico, mas só é vista como uma pessoa com deficiência em sua relação com a sociedade, visto que a sociedade não se apresenta como um espaço social educativo. partimos também do contraponto da representação que a música apresenta na sociedade contemporânea. neste molde, ela carrega em si traços de uma concepção que tem por base uma série de ideias e ideais que a incluem para ser sentida, ouvida e percebida unicamente pelo órgão auditivo, desconsiderando a unidade do funcionamento do organismo. no universo sonoro que nos rodeia, compreendemos que o som não é somente a parte audível da música, mas uma característica dela, conforme nos sinaliza winisk. assim, entendemos que a vivência da musicalidade, por meio da atividade musical, não compreende unicamente a percepção auditiva. na compreensão desta vivência, tivemos um olhar etnográfico, baseado na etnografia sensorial, que compreende as experiências como multissensoriais e o funcionamento do organismo em sua unidade. fizemos uso, também, da etnografia em/com mídias sociais, o que possibilitou o contato desejado com os participantes.
The present study, a master's thesis, has as aim to investigate the ways of living the musicality of the deaf person, considering their culture. Considering the individual as an intellect-affect unit and as a being of possibilities, based on the historical-cultural perspective by Vygotsky, the research to perceive this individual in its wholeness, in entirety. In this perspective, and based on studies of defectology, the deaf person presents a biological defect, this person is only seen as an individual with a disability in their relationship with society, because the society does not present itself in the role of an educational social environment. Also, we assumed that the counterpoint of the representation of music reported in contemporary society. At this model, the representation of music carries itself traces of a conception based on a series of ideas and ideals that include it to be felt, heard, and perceived merely by the hearing organ, without considers the unity of the functioning of the organism. In the sonorous universe that surrounds us, we realize that sound is not only the audible part of the song but a characteristic of it, Winisk points out. Then, we comprehend that the lived of musicality, through musical activity, does not understand only the hearing perception. In the comprehension of this experience, we had an ethnographic view based on sensorial ethnography which includes the experiences as multisensory and the functioning of the organism in its unity. Nevertheless, we used the ethnography in / with social media, which enabled the desired contact with the participants.

O presente trabalho propõe-se estudar o contexto de crescimento de um grupo de jovens surdos, de modo a conhecer e compreender os factores que contribuíram, ou não, para o desenvolvimento da sua literacia. A base teórica deste estudo é constituída pela filosofia da educação bilingue e bicultural das crianças e jovens surdos e parte de dois pressupostos fundamentais: a primeira língua da criança surda é a língua gestual do seu país, essencial para o seu desenvolvimento emocional, linguístico e cognitivo, e a segunda língua é a língua falada no seu país, ensinada à criança como língua não materna, privilegiando as vertentes da leitura e da escrita, fundamentais para a sua inserção na sociedade e para o seu progresso académico e profissional. Para a realização do presente estudo, foi adoptada uma metodologia qualitativa e descritiva, tendo a recolha de dados sido realizada por intermédio de um inquérito por questionário. Os dados foram recolhidos junto de um grupo de jovens surdos do concelho de Lisboa, entre os 15 e os 30 anos. Da análise dos resultados foram retiradas algumas conclusões que confirmaram as premissas defendidas no enquadramento teórico deste trabalho: a importância de uma educação bilingue e bicultural e o envolvimento da família no desenvolvimento da literacia das crianças e jovens surdos. ABSTRACT: This paper will study the environment in which a group of young deaf grew up in order to identify and understand the aspects that did or did not contribute to their development of literacy. The theoretical basis of this study is the philosophy of a bilingual and bicultural education of deaf children and deaf youngsters. This theory has two main prerequisites: that the first language of deaf children is the sign language of their country, it is essential for their emotional, linguistic and cognitive development, and that the second language is the spoken language, taught as a non maternal language emphasising the reading and writing skills, which are fundamental for their integration into society as well as for their academic and professional careers. We adopted a qualitative and descriptive methodology in this study and we collected data with a questionnaire. Data was collected from a group of deaf youngsters, aged 15-30 years, from Lisbon. The conclusions taken from the data analysis confirmed the premise of the theory we argued for in our study: the importance of a bilingual and bicultural education and the participation of the family in developing deaf children and deaf youngsters’ literacy.

Traditionally, geometry at school starts on a formal level, largely ignoring prerequisite skills needed for formal spatial reasoning. Ignoring that geometry has a sequential and hierarchical nature causes ineffective teaching and learning. The Van Hiele theory postulates learner progression through levels of geometry thinking, from a Gestalt-like visual level through increasing sophisticated levels of description, analysis, abstraction, and proof. Progression from one level to the next does not depend on biolog~caml aturation or development only, but also on appropriate teachingllearning experiences. A higher thinking level is achieved through the application of a series of learning phases, consisting of suitable learning activities. The teacher plays an important facilitating role during this process. In accordance with the social cognitive learning perspective on self-regulated learning, geometry learners must direct their thoughts and actions while completing activities in order for effective learning to take place. Learners can be described as being selfregulated to the degree that they are metacognitively, motivationally, and behaviorally active in their own learning. The social cognitive theory assumes that students enter learning activities to acquire knowledge, learning how to solve' problems and completing learning activities. Self-regulated learners are aware of strategic relations between self-regulatory processes and learning outcomes and feel self-efficacious about using strategies. Self-regulation is similar to metacognitive awareness, which includes task and personal knowledge. Self-regulated learning requires that learners understand task demands, their personal qualities, and strategies for completing a task. A Van Hiele-based geometry learning and teaching program was designed (with a problem solving context in mind) and implemented in four Grade 7 classes (133 learners) at two schools. The study investigated factors and conditions influencing the effective learning and teaching of spatial concepts, processes and skills in different contexts. Results suggest that the implementation of a Van Hiele based geometry learning and teaching program in a problem solving context had a positive effect on the learners' concentration, when working on academic tasks, and level of geometric thought. The higher levels of geometric thought included higher categories of thought within these levels. Learners who completed the program reasoned on a higher level, ,gave more complete answers, demonstrated less confusion, and generally exhibited higher order thinking skills than their counterparts who did not take part in the program. The only prerequisite' is that the teacher should consistently teach from a learner-centered approach as the program will deliver little or no advantages if the program is presented in a teacher-centered content-based context.

A presente dissertação, “Para além do Silêncio; Outros olhares sobre a surdez e a educação de surdos”, traz para à discussão algumas recorrências enunciativas possíveis de serem lidas nas produções de um dos grupos de pesquisadores que marcou a história da educação de surdos no Estado do Rio Grande do Sul. O grupo formou o Núcleo de Pesquisa em Políticas Educacionais para Surdos (NUPPES) e, através de tal Núcleo, fez parcerias com a Secretaria de Educação do Estado, assumindo a partir daí a elaboração dos currículos, bem como a formação de professores nos cursos oferecidos pelo Estado. Além disso, o NUPPES também fez distintos movimentos para que a comunidade surda começasse a ter expressão na academia através da entrada de surdos na Universidade. This group was formed the “Núcleo de Pesquisa em Políticas Educacionais para Surdos” (NUPPES – Nucleus of Research in Educational Politics for Deaf People) which made partnerships with the State Secretariat of Education, assuming the elaboration of the curriculums, as well as the formation of professors in the courses offered for the State. Moreover, the NUPPES made distinct movements so that the deaf community started to have academic expression by the entrance of deaf people in the university. Abstract:
One of the principal research themes of the supercritical fluid research group within the Centre of Separation Science and Technology (SST) at the North-West University (Potchefstroom Campus) is botanical extraction. The group produces botanical extracts from locally cultivated plants which contain substances (essential oils, natural waxes) relevant to the food, flavour, pharmaceutical, medical and cosmetic industries while utilising the advantages of sc-C& extraction over traditional steam distillation and solvent e~traction.'.~ In this study, which represents a contribution to a series of botanical extractions performed within the core programme outlined above, sc-COz extraction of artemisinin from Artemisia annua (or wormwood) was investigated. The active component is a potential cure against ma~aria.It~ w ould have been desirable to extract it with no solvent residues left in the final product to prevent side-effects on taking the medicine. The extraction by clean technology, using environmentally friendly sc-C&, could be relevant as it offers an affordable alternative for synthetically prepared medicines in the marketplace. The manipulation of the conditions (temperature and pressure, or density) for sc-C02 extraction could facilitate more selective isolation of the active component and thereby enhance the medicinal value of wormwood. The unique solvent strength and mass transport characteristics of sc-C02 offer the possibility of obtaining better results than with other solvent based extraction methods. The specific goals with the project were to extract an active component or ingredient (artemisinin) from the leaves of wormwood with sc-COz on laboratory scale by using an advanced benchtop supercritical fluid extractor and other available laboratory infrastructure; to compare the results of sc-COz extraction with those of classical extraction methods, such as solid-liquid extraction, to ascertain the advantages and disadvantages of different extraction methods; to identify and implement suitable analytical techniques (HPLC andlor GC) with which artemisinin containing extracts can be analysed qualitatively and quantitatively; to identify process parameters and to vary these according to a statistical design using a suitable software programme (Statistica for windows@) to establish optimum conditions for the extraction of the target component; to process the extraction data mathematically and/or graphically in such ways as to reveal the principal features of the extraction process, to facilitate modelling of the process and to enable calculation of mechanism directive activation parameters; to complement comparable studies performed in the research group. In addition to these specific goals, the project also served the purpose to contribute to a lesser extent to the following relevant issues: The essential oils, natural waxes and other chemical components derived from plants are low-volume high-value products and therefore have significant commercial value.' The importance of clean technology for "green" or sustainable chemistry is increasingly emphasised.6 sc-C02 is an environmentally friendly solvent with which solvent-free extracts can be derived. There are academic interest in and financial support for the development of knowledge about indigenous plants.7 The suitability of sc-COz for the isolation of plant components derived by traditional healers for centuries can help to better understand the beneficial effects of plant medicines. The relevance of supercritical fluid based processes for daily life creates science awareness and renders the research done in this investigation suitable for the popularisation and promotion of chemistry since the replacement of natural products in ordinary household products (beer, shampoo) captures the attention and imagination of the public. Finally, this investigation can help to convince industry to apply the technology despite of the negative perceptions about extreme conditions and the high capital investment needed to put up the required infrastructure.

O estudo busca inserir a discussão sobre a inclusão de alunos surdos, tendo em vista dois contextos de atuação: uma sala de aula regular com apoio de intérprete em Libras e uma classe especial de surdos com professora especializada e usuária da língua de sinais. Partimos do pressuposto de que a inclusão não se constitui um estado permanente, nem uma mera inserção física de alunos especiais na escola regular. Por isso, buscamos dar coerência ao discurso oficial, quando diz que é necessária uma mudança da sociedade para receber todos os seus cidadãos; trazendo, à discussão, os processos educacionais em geral e suas possibilidades de dar materialidade a uma Educação para Todos. A partir dessas considerações, colocamos como questões de pesquisa: Quais as necessidades que os estudantes surdos apresentam? Como a equipe é composta e como atua? A metodologia usada pela professora colabora para que os alunos surdos participem das aulas e atinjam os objetivos de aprendizagem de modo satisfatório? A professora revela habilidade para perceber as potencialidades dos alunos e criar situações de aprendizagem que os ajudem a aprender? Como a inserção do intérprete de língua de sinais interfere no processo? Como as professoras, intérprete e alunos percebem o processo educacional? Através de entrevistas com professoras, intérprete e estudantes surdos e em observações de situações de ensino-aprendizagem em salas de aula, regular e especial, de escolas da rede pública de ensino do Estado de Pernambuco, obtivemos informações que nos permitiram problematizar o trabalho realizado em sala de aula, regular e especial, com alunos surdos, desde a aplicação e desenvolvimento das atividades até as formas de participação dos envolvidos (professora, intérprete e alunos) no processo educativo; bem como as falas desses atores educacionais sobre aspectos relativos à inclusão. Foi possível notar que a atuação do profissional intérprete na sala de aula não considerava as formas peculiares de pensar, sentir e diferir dos surdos, agravado pelo fato de os procedimentos pedagógicos da professora continuarem inalterados quanto à presença desses alunos. Essa dificuldade pareceu que era minimizada na classe especial de surdos, que é um local considerado segregador. Isso indica que é necessário desenvolver um novo olhar sobre a inclusão, evidenciando a urgência em superar a discussão com foco no binômio inclusão-exclusão e em promover uma reflexão profunda sobre a importância de desenvolver práticas culturalmente referenciadas.

Abstract:
Raman spectroscopy was used to study the protein structure in raw and salted surimi from Pacific whiting, and in gels formed by setting (32°C), cooking (86°C) or setting followed by cooking. The intensity of the peaks assigned to disulfide bond stretching vibrations increased considerably in the cooked and set-cooked gels. A smaller increase was found in the gels that were subject to setting alone. Secondary structure estimation based on the amide I band indicated a change from predominantly a-helical structure in raw surimi to similar proportions of a-helical and anti-parallel p-sheet after setting. A further increase in anti-parallel p-sheet and decrease in a-helix content occurred during the kamaboko stage. The intensity of C-H stretching vibrations of the aliphatic residues decreased after salting, setting and cooking. The set and cooked gel had a better gel strength and fold score than the cooked gel which, in turn, had better properties than the set gel. Response surface methodology was used to determine the optimal setting and cooking time and temperature conditions resulting in a maximized gel strength, fold score and color, (whiteness index), and a minimized gel expressible liquid. Cooking temperature was the variable that had the strongest influence on the gel quality characteristics. Level set programming, a global optimization method, gave essentially the same results as a gradient based optimization method. The results obtained by these two methods were better than those generated by the Simplex technique. Using a formulation composed of pink salmon surimi, salt, whey protein concentrate and wheat starch, an optimal final product, kamaboko sausage, was developed. In addition, when this formulation was applied to herring surimi, the kamaboko obtained had similar gel strength and elasticity as a commercial surimi-based product. The effects of frozen storage conditions of roe herring on its gel making ability (GMA) were investigated. Frozen storage at around -45°C maintained the GMA for seventy days while storage at -83°C further increased the GMA period. The viscous properties of a salmon surimi paste were studied using a rotational viscometer. The paste behaved as a shear thinning fluid with a yield stress that increased with temperature up to 21 °C. Viscosity also increased with temperature up to 21 °C, possibly because of protein-protein interactions. The Theological data were reasonably well represented by a simple model which takes into consideration the effects of both shear rate and temperature.

The thesis hypothesis of this research was that farmed chinook salmon could be made into better quality functional kamaboko when made from solubilized frozen surimi than when made from conventional frozen surimi. An 84 day storage study compared kamaboko gel quality made from solubilized and traditional surimi. Fresh farmed chinook salmon (Oncorhynchus tshawytscha) was used to make both solubilized surimi and surimi (control). Solubilized treatments contained varying concentrations of calcium chloride, sodium chloride and water. The Random Centroid Optimization (RCO) program randomly generated concentration values of additives. All surimi treatments (solubilized and control) contained 8.3% cryoprotectants. Treatments were taken from storage on days 3, 7, 14, 28, 56 and 84 from an -8°C freezer and made into kamaboko. Solubilized treatments were diluted after frozen storage and then centrifuged to constant moisture content. All kamaboko gels had respective moisture, protein, crude fat and ash contents of 74.5 + 5.5%, 13.6 ± 1.7%, 4.4 + 2.4%, and 5.6 ± 3.1%. Salts added to solubilized treatments influenced ash content. No proximate analysis trends were observed between treatments during the storage study. Variation in protein and water concentrations within the range of this study did not appear to affect overall kamaboko quality.

Optimization of comminuting and heating conditions for surimi gel preparation obtained from three fish species: Alaska pollock (AP) (Theragra chalcogramma), Pacific whiting (PW) (Merluccius productus), and threadfin bream (TB) (Nemipterus spp.) was the focus of this study. Three parameters during comminution were separately evaluated: chopping time, chopping temperature, and salting time. Results from fracture gel analysis suggested a strong relation between the fish's environmental habitat and optimal final chopping temperature. Extending chopping time to 15 min under strictly controlled temperature at 0 ��C was preferable for cold water fish AP surimi. Even though high chopping temperature (20 ��C) for a shorter time (12 min) surprisingly resulted in strong gel texture similar to that of 0 ��C for 15 min, high chopping temperature should not be employed for AP surimi. AP could set as a gel at this temperature within a shorter time in a holding tank which could subsequently cause a problem when extruded on the cooking belt. Temperate water fish Pacific whiting, demonstrated its maximum gel strength when chopped at 15-20 ��C. The optimum comminution condition for warm water fish threadfin bream surimi was to chop the surimi until the paste temperature reached between 25-30 ��C. Prolongation of chopping once the surimi hit its threshold (optimum) temperature diminished the quality of the resulting protein gel. Cooling system connected to the chopping bowl is strongly recommended as it will allow the comminution process to be extended as long as possible until the surimi paste reaches its target temperature. Raman spectroscopy disclosed the different level of protein unfolding based on secondary structure of ��-helix and ��-sheet during various comminuting conditions. Unfolding of protein was facilitated by increased chopping temperature to a greater degree than extended chopping time. Extending chopping could denature the light meromyosin structure as it could not form a semi gel-like structure at temperatures between 32-40 ��C. Protein solubility of surimi paste in salt solution always decreased with prolonged chopping time. The decrease rate accelerated with increased chopping temperature. The formation of disulfide interchange gradually took place during chopping as observed from Raman spectroscopy. Also the surface hydrophobicity increased with extended chopping time. However, gel strength behaved differently according to the various chopping conditions indicating the lack of its relationship between salt soluble protein, disulfide formation, and surface hydrophobicity to gel strength. During extending chopping time, not only more mechanical force is applied to unfold protein structure, but proteins also have longer time to be extracted more by salt. Addition of salt at a different time during chopping process was therefore conducted using threadfin bream surimi due to its higher thermostability. Extending chopping time without salt followed by salt addition at the last step resulted in lower gel texture compared to the conventional chopping protocol where salt is always added at the early stage of comminution. Mechanical chopping could unfold protein structure; however, proteins, rather than staying solubilized, would precipitate and form a randomized structure under the chopping condition without salt. The heating condition greatly affected the gelation and rheological properties of AP surimi. The highest elastic modulus was obtained with the slowest heating rate at 1 ��C/min. Increased heating rate did not only shorten the time for proteins to unfold and form a well-organized network, it also interfered with the protein network through the vibration of water molecules as phase angle increased. This suggested that AP surimi gained more viscous properties and failed to form an elastic gel. Adjusting moisture content along with applying various frequencies did not alter the pattern of G' formation when paste was heated at different heating rates. AP surimi favored the slow heating.
