Abstract:
Developing inhaled drugs requires knowledge of lung anatomy, cell biology, respiratory physiology, particle physics, and some plumbing. Although dose makes the poison, in the context of an inhaled drug, the “dose” is not easily defined. This lack of clarity around dose poses issues and challenges in the design of inhalation toxicology programs. To better understand dose, the influence of ventilation is discussed as are the perturbations in pulmonary function observed with inhalation exposure that can affect dose. Methods for determining inhaled drug deposition to arrive at an estimate of lung dose are examined. Equally important to understanding dose are the techniques used to deliver aerosols to animals. With a better understanding of dose and inhalation exposure, species-specific histopathologic lesions, both common background and toxicologically significant lesions, are reviewed. Finally, insight into how regulators synthesize and evaluate these complex findings to assess clinical safety risks is presented.

The article deals with an optimisation of the relation between production and non-production functions of agriculture. In the model, a two-product production process is presupposed, the result of which is agricultural production and a complex of other effects arising in agriculture. To obtain them, two aggregated inputs – capital and labour – are used. To maximise the summary economic result, the procedure of derivation of the optimal extent of both production processes proceeding as well as non-production effects in minimisation of both inputs in their summary is worked out in the article. In the article, the knowledge of the research gained in framework of solution of the institutional research intention MSM: 411100013 “Efficient integration of the Czech agrarian sector into the frame of the European structures – presumption of sustainable development” is included.

In the steel industry Sn is always regarded as harmful impurity element, one of "Five harmful elements", which affects the welding property and equality of steel product especially the equality of continuously cast bloom and causes hot brittleness, temper brittleness, crack and fracture. While Sn element also has a vital influence on free-cutting steel, electrical sheet, cast iron and stainless steel. NSSC developed ferritic stainless steel containing low Sn and Ni content improving the corrosion resistance performance and the workability significantly. So the Sn element, as cheap alternative adding element, can substitute for Cr and Ni. It's helpful to study the effect of tin in the iron and steel and develop new stainless steel products of high equality conserving chromium resources. In this way it will promote the variety and technique of stainless steel, ensure the independence on resources and conserve resources remarkably.

In biomedical research summary measures such as the curve maximum (Cmax)are frequently used to describe and analyze unimodal response curves. However, if the true curve is perturbed by autocorrelated noise the calculation of summary measures from raw data can be arbitrary and misleading due to high peaks produced by the correlated errors. A possible solution is to fit suitable nonlinear functions to the response curves and estimate the summary measures from these functions. In this paper formulas are derived providing a way to estimate important summary measures of unimodal curves by means of the parameters of the lognormal function. The method is illustrated by application to pharmacodynamic data.

Abstract:
Bayesian linear regression is an approach to linear regression where statistical analysis depend of Bayesian inference. The Bayesian model on big data uses a summary of data statistics as input; Statistical summary can be calculated from each subset, then a statistical summary of the full dataset is obtained from the sum of the summary statistics for each subset. Recent developments in data science and research, produce large datasets that are too large to be analyzed as a whole due to the limitations of computer memory or storage capacity. To overcome this, a program package was introduced from R namely BayesSummaryStatLM for the Bayesian linear regression model with the Markov Chain Monte Carlo implementation that overcomes this limitation. Then the program package from R, ff is used to read data in large datasets while calculating statistics summary. In this study Bayesian linear regression model used with several choices of prior distribution for unknown model parameters, and illustrates in simulation data and real datasets for flight delay data in US 2008. The application of simulation data and flight delay data produces a plot of density functions for the β parameters has a shape resembling a plot of Normal distribution density function, whereas for plot parameters the density function has a shape resembling the plot of Inverse Gamma distribution density function. In the simulation data, the estimator for each parameter produced has a value that approach to the value of the specified parameter (True Value). This is also indicated by the narrow credible interval for each parameters.

Background: SIRT1 is a nicotinamide adenine dinucleotide-dependent deacetylase belonging to the class III histone deacetylases. Abundantly expressed in the kidney, especially in the renal medulla, SIRT1 is closely involved in renal physiology and pathology. Summary: SIRT1 targets both histone and nonhistone proteins, participates in many important signaling pathways and mediates the regulation of longevity, metabolic homeostasis, acute stress response and DNA integrity. With regard to the kidney, SIRT1 attenuates diabetic albuminuria, reduces blood pressure and related cardiovascular diseases, resists acute kidney injury, delays kidney fibrogenesis, promotes cyst formation and benefits renal ageing. Key Messages: This review summarizes the biology of SIRT1 and focuses on the latest studies concerning SIRT1 as a potential therapeutic target for kidney diseases.

Abstract This paper examines how native speakers (NS) and non-native speakers (NNS) of Italian approach topic organisation (topic shift, topic closure, digressions, topic recovery, and summary) in oral interactions. The research focuses on which discourse markers (DMs) are used when speakers try to organise discourse topics, and the differences between NS and NNS when performing such metadiscourse functions. The analysis is based on data from a spoken corpus designed to study conversational strategies in Spanish learners of L2 Italian. It reveals that the acquisition of metadiscourse functions progresses at different rates depending on the function: whereas learners have a good pragmatic competence in using DMs for the introduction of new topics in conversation, they have difficulties with other functions, such as topic closure or summary. In addition, the function of topic recovery after a digression is explicitly marked by NNS by DMs which are not found in native varieties.

In the first section, we prove several theorems about open sets in real norm space, which are needed in the proof of the inverse function theorem. In the next section, we define a function to exchange the order of a product of two normed spaces, namely 𝔼 ↶ ≂ (x, y) ∈ X × Y ↦ (y, x) ∈ Y × X, and formalized its bijective isometric property and several differentiation properties. This map is necessary to change the order of the arguments of a function when deriving the inverse function theorem from the implicit function theorem proved in [6]. In the third section, using the implicit function theorem, we prove a theorem that is a necessary component of the proof of the inverse function theorem. In the last section, we finally formalized an inverse function theorem for class of C 1 functions between Banach spaces. We referred to [9], [10], and [3] in the formalization.

Summary This paper develops Bayesian function estimation on compact Riemannian manifolds. The approach is to combine Bayesian methods along with aspects of spectral geometry associated with the Laplace-Beltrami operator on Riemannian manifolds. Although frequentist nonparametric function estimation in Euclidean space abound, to date, no attempt has been made with respect to Bayesian function estimation on a general Riemannian manifold. The Bayesian approach to function estimation is very natural for manifolds because one can elicit very specific prior information on the possible symmetries in question . One can then establish Bayes estimators that possess built in symmetries. A detailed analysis for the 2–sphere is provided.

Second-order investigation based on Ripley's <i>K</i>-function and related statistics (i.e., <i>L</i>- and pair correlation function <i>g</i>) is widely used in ecology to develop hypothesis on underlying processes by characterizing spatial patterns of vegetation. The aim of this study was to demonstrate effects of underlying heterogeneity of wild pistachio (<i>Pistacia atlantica</i> Desf.) trees on the second-order summary statistics of point pattern analysis in a part of Zagros woodlands, Iran. The spatial distribution of 431 wild pistachio trees was accurately mapped in a 40 ha stand in the Wild Pistachio & Almond Research Site, Fars province, Iran. Three commonly used second-order summary statistics (i.e., <i>K</i>-, <i>L</i>-, and <i>g</i>-functions) were applied to analyse their spatial pattern. The two-sample Kolmogorov-Smirnov goodness-of-fit test showed that the observed pattern significantly followed an inhomogeneous Poisson process null model in the study region. The results also showed that heterogeneous pattern of wild pistachio trees biased the homogeneous form of <i>K</i>-, <i>L</i>-, and <i>g</i>-functions, demonstrating a stronger aggregation of the trees at the scales of 0&ndash;50 m than actually existed and an aggregation at scales of 150&ndash;200 m, while regularly distributed. Consequently, we showed that heterogeneity of point patterns may bias the results of homogeneous second-order summary statistics and we also suggested applying inhomogeneous summary statistics with related null models for spatial pattern analysis of heterogeneous vegetations.

SUMMARY Public Company Accounting Oversight Board (PCAOB) Auditing Standard No. 5 (AS5) allows external auditors (EAs) to rely on the internal audit function (IAF) when the internal audit activities meet certain criteria and the EAs would find efficiencies in relying on their work (PCAOB 2007). This paper reviews the extant literature on the EAs' reliance on IAF, identifies gaps in the literature, and proposes a series of research questions aimed at closing these gaps. We focus our review on research pertaining to how environmental factors and IAF-specific factors influence initial EAs' reliance decisions, the nature and extent of EAs' reliance on IAF, and the observable outcomes as a result of EAs' reliance decisions. Our review finds that the environment in which EAs must make a reliance decision is complex—involving several factors that must be considered simultaneously. Moreover, an evolving set of auditing standards introduces several necessary intermediary judgments that the EAs must process before, and during, reliance on the IAF. In addition, our review indicates that we continue to know very little about how, and to what extent, EAs are currently evaluating IAF's quality factors. Similarly, while we find that the nature and extent of EAs' reliance on IAF is influenced by account risk, inherent risk, and IAF sourcing, how the EAs choose task environments (e.g., revenue recognition versus payroll), and the types of tests to be relied upon within these task environments, is not completely understood. Finally, we find that there is a paucity of research concerning the effects of EAs' reliance on IAF in terms of external audit quality.

Abstract The empirical cumulative density function (ECDF) approach can be used to combine multiple, diverse assessment metrics into summary assessment metrics (SAMs) to analyze the results of impact experiments and preoperational implementation testing with numerical weather prediction (NWP) models. The main advantages of the ECDF approach are that it is amenable to statistical significance testing and produces results that are easy to interpret because the SAMs for various subsets tend to vary smoothly and in a consistent manner. In addition, the ECDF approach can be applied in various contexts thanks to the flexibility allowed in the definition of the reference sample. The interpretations of the examples presented here of the impact of potential future data gaps are consistent with previously reported conclusions. An interesting finding is that the impact of observations decreases with increasing forecast time. This is interpreted as being caused by the masking effect of NWP model errors increasing to become the dominant source of forecast error.

Summary Objective: There have been many studies on adenomyosis, which can impair the quality of life of a woman. There are various kinds of opinions on the pathogenesis, diagnostics and treatment of adenomyosis. The goal of this article is to present the current knowledge of adenomyosis and its impact on the endometrial function and receptivity. Methods: PubMed/Medline, Web of Sciences and Scopus were searched for the articles in English indexed until February 2021 with terms of: adenomyosis, endometrial receptivity, and infertility. Results: Recent studies on angiogenesis and epithelial-mesenchymal transition in the endometrium bring new information on the ethiology and pathogenesis of adenomyosis. In clinical practice, the main diagnostic methods of adenomyosis include transvaginal ultrasound, magnetic resonance imaging or hysteroscopy, although the definitive confirmation is set by histopathological examination. The rules of #Enzian classification of endometriosis should be applied for the classification of adenomyosis. The treatment of adenomyosis should consider individual clinical presentation and reproductive plans of a patient and should be performed in centers for the treatment of endometriosis. Conclusion: Adenomyosis affects endometrial vascularisation and epithelial-mesenchymal transition/mesenchymal-epithelial transition; thus, it can be the cause of irregular uterine bleeding or embryo implantation failure. The research and analysis of endometrial proteome could lead to the new ways of adenomyosis treatment.

Subjective cognitive decline (SCD) is complex and not well understood, especially among Japanese people. In the present study, we aimed to elucidate the relationships of subjective and objective mental and physical function with SCD among older community-dwelling Japanese adults. SCD was evaluated using the Kihon Checklist: Cognitive Function. Other parameters were evaluated using the Mini-Mental State Examination (MMSE) and the five-item version of the Geriatric Depression Scale (GDS-5), for an objective mental function other than SCD. A timed up-and-go test (TUG) and knee extension strength were used to test objective physical function, and the Mental Component Summary (MCS) and Physical Component Summary (PCS) in the Health-Related Quality of Life survey eight-item short form (SF-8) were used for subjective mental and physical functions. The results of the MMSE, GDS-5, TUG, knee extension strength, and MCS were significantly worse in the SCD group. In addition, logistic regression analysis showed that GDS-5 and MCS were associated with SCD onset. Depressive symptoms and decreased subjective mental function contribute to SCD among community-dwelling Japanese adults. These findings will be useful for planning dementia prevention and intervention programs for older Japanese adults.
