Full textQuesta tesi tratta l'analisi automatica di immagini a colori del fondo dell'occhio. In particolare, essa si focalizza sul progetto di una fundus camera ad ottica adattiva per l’aquisizione di fundus retinici e sull’implementazione di software per l’analisi e la valutazione quantitativa delle retinopatie, prevalentemente ipertensiva e diabetica come naturale ricaduta dei metodi sviluppati. Sia l'ipertensione che il diabete colpiscono, anche se con modalità e decorso temporale differente, il microcircolo sanguigno. La retinopatia è una delle conseguenze di tale danno circolatorio. La retina ed i vasi retinici sono assai sensibili a cambiamenti nella microcircolazione: è stato dimostrato che i singoli segni della retinopatia hanno un alto valore prognostico per infarto, sclerosi carotidea e danno coronarico. Inoltre, la retinopatia è una malattia sociale, con una ricaduta economica (diretta ed indiretta) elevata: la perdita o la diminuzione della capacità visiva porta infatti ad una ridotta capacità lavorativa e all’impossibilità di condurre una vita indipendente. Nonostante anche altri organi siano sensibili ad alterazioni del microcircolo sanguigno, la retina ha il grande vantaggio di essere facilmente disponibile ad un controllo non invasivo. Tale caratteristica suggerisce un modo efficiente ed efficace per seguire il decorso di malattie sistemiche associate alla retinopatia. Inoltre, con riguardo alla prevenzione della perdita della vista, il riconoscimento della retinopatia al suo insorgere è il punto più critico per evitare che degeneri in cecità. Ciò è particolarmente importante nella retinopatia diabetica in cui, allo stato attuale della farmacologia, i danni alla retina non recedono con il trattamento farmacologico o col controllo del diabete. Sfortunatamente, le fasi iniziali della retinopatia sono quasi asintomatiche. Un programma di screening potrebbe evitare alla maggior parte della popolazione a rischio lo sviluppo di retinopatie che minaccino la vista. Allo stesso tempo, nel mondo occidentale non ci sono abbastanza risorse, sia in termine di tempo che soprattutto in termine di disponibiltà di oftalmologi esperti, per organizzare uno screening di tal genere. E’ dunque forte il bisogno di uno strumento che valuti automaticamente la retina, per diagnosticare la presenza e la severità dell'eventuale retinopatia. Le attuali fundus camera sono in grado di compensare l'aberrazione di fuori fuoco introdotta dall'occhio tramite sistemi mobili di ottiche interne, ma non possono correggere l'astigmatismo oppure le aberrazioni di ordine superiore. Nei primi capitoli della presente tesi si descriverà la realizzazione del prototipo di un sistema di acquisizione che consenta di compensare anche quel tipo di aberrazioni, permettendo così in linea teorica di ottenere immagini di elevata qualità del fondo retinico, anche in pazienti soggetti ad aberrazioni elevate. Le immagini digitalizzate ad alta risoluzione del fondo retinico saranno poi elaborate da algoritmi d’analisi, descritti nella seconda parte della tesi, dedicati alla diagnosi automatica di alcune patologie dell'occhio. Il concetto di base dello strumento d’acquisizione sviluppato nei nostri laboratori è quello di inserire lungo il cammino ottico di una fundus camera un'ottica adattiva DM (Deformable Mirror). Il principale problema delle fundus camera convenzionali è che l'informazione per adattare la forma del DM per compensare le aberrazioni presenti è fornita da un complesso e costoso analizzatore di fronte d’onda. La soluzione proposta consiste nel proiettare un pattern noto sul fondo della retina e di acquisirne l’immagine con un sensore dedicato. Analizzando in tempo reale quest’immagine con opportuni algoritmi, sarà possibile ottenere le indicazioni su come l'ottica adattiva debba essere opportunamente deformata per ridurre l'aberrazione introdotta dall’occhio e permettere di acquisire immagini ad alta definizione e senza distorsioni anche in pazienti affetti da aberrazioni non compensabili con le fundus camera ora disponibili. Nonostante test di simulazione su banco ottico abbiano fornito risultati preliminari incoraggianti, la realizzazione di una fundus camera adatta all’utilizzo clinico/diagnostico ha incontrato limitazioni di carattere tecnico relative alla prototipazione dello strumento in ambienti non controllati. L’incidenza dei riflessi spuri, introdotti nel sistema ottico dall’ambiente circostante, sul rapporto segnale-rumore del segnale acquisito sarà materia di ricerca del prossimo futuro. Le immagini del fondo oculare, ricavate da dataset da noi collezionati e da altri già utilizzati per indagini scientifiche da altri gruppi di ricerca, saranno analizzate con algoritmi sviluppati ad-hoc, in grado di ricavare importanti parametri clinici per la diagnosi delle retinopatie, con particolare attenzione a quelle ipertensiva e diabetica. Nell’ambito di questa tesi saranno prese in esame le features dell’apparato vascolare: l’identificazione del reticolo dei vasi (arterie e vene), consentirà la misurazione i suoi principali descrittori geometrici (lunghezza, direzione, calibro, presenza di biforcazioni, etc.); da questi saranno calcolati specifici indici di rilievo diagnostico, che forniranno ai clinici informazioni sul grado complessivo di retinopatia del paziente, la cui significatività è molto importante per la diagnosi delle patologie retiniche. Gli algoritmi presentati in questa tesi rendono possibile pensare ad uno strumento utilizzato sia in ampi programmi di screening delle retinopatia ipertensiva e diabetica, sia per il controllo nel tempo del progredire della malattia. La loro utilià è triplice. In primo luogo, potrebbero essere uno strumento diagnostico di aiuto alla pratica clinica. In secondo luogo, i dettagli quantitativi sulle struttura vascolare della retina potrebbero essere utili nella ricerca medica e per meglio caratterizzare gli sviluppi della retinopatia. Infine, nell’ambito della ricerca farmaceutica, si renderebbe disponibile l'utilizzo di una misura quantitativa e riproducibile dell'evoluzione della retinopatia durante un trattamento farmacologico. Il software riguardante l’identificazione delle strutture vascolari retiniche è al momento oggetto di valutazione clinica presso il Department of Ophthalmology and Visual Sciences, University of Wisconsin, USA, il cui Fundus Photograph Reading Center ha espressamente dichiarato la sua volontà nell’acquisire il programma come standard per la determinazione di features diagnostiche, quali gli indici CRAE, CRAE e AVR, come indicatiori del “generalized arteriolar narrowing”. Sempre per quanto riguarda i metodi di identificazione della struttura vascolare della retina, la corrente collaborazione con il Department of Twin Research & Genetic Epidemiology, del King’s College London Division of Genetics and Molecular Medicine, St Thomas' Hospital, UK, si è recentemente concretizzata nell’accordo che prevede che un numero superiore a 3500 immagini di fundus retinico (più 10 nuove immagini a settimana) vengano analizzate per scopi clinici con il software da noi sviluppato. I risultati conseguiti nelle sperimentazioni effettuate e le collaborazioni internazionali in atto con gruppi clinici e di ricerca di rilievo ci rendono abbastanza confidenti riguardo la qualità delle metodologie sviluppate e i potenziali successi del suo impiego, con l’auspicio che futuri miglioramenti possano ampliarne l’utilizzabilità.

Full text“Good design enables, bad design disables”1 The usual planning of public space takes several standards as reference, which relate to diferent technical and planning requirements inevitably creatng exclusion. To be inclusive, it is mandatory to consider users with variable and diferent features. Duchenne Muscular Dystrophy (DMD) is a degeneratve disease; the progression of disease phases relates to diferent needs towards the space, needing a fexible design method. A child afected by DMD meets one the most critcal phases from psychological point of view (due to percepton of his own limitatons and to consciousness of disease) during the beginning of mandatory school. In this crucial stage, the answers provided by the environment are very important. The analysis of data collected from literature and some semi-structured interviews undertaken with healthcare, teaching and technical professionals determined the choice of the feld of investgaton: the learning environment in primary school is the most frequented place during the most critcal age and becomes a symbolic place for inclusive culture producton. School is made of spaces (architecture) and services (pedagogy and didactc) that follow diferent paths to obtain inclusion. Italy has a 30-years experience on designing inclusive services in schools (L. 517/77, L. 104/92), and meanwhile inclusive design is ruled by a limited number of laws (D.M. 236 e L.13/89), contrastng the values expressed by UN Conventon 2006 on rights of people with disability and by ICF classifcaton (WHO 2001)2. The aim of the Thesis is to propose a fexible instrument which connects spaces and services in the way which is expressed by ICF, helping in managing the diferent needs of users inside primary school areas and promotng their partcipaton.

Full textObesity has become a global concern, whose numbers are increasing with not negligible social and economic costs. Bariatric Surgery (BS) is considered the best treatment for people with morbidly obesity, because it allows the loss of a large amount of weight in the short period, improving the quality of life and clinical condition. However, BS is affected by side effects and complications, mostly due to the empirical approach and the anaesthetic dose. Modelling techniques could provide a rational description of the mechanisms of BS, overcoming the main limitations. Moreover, bio-engineering tools can be useful predictors of effectiveness during bariatric planning. The aim of the Ph.D. was the definition of reliable constitutive parameters of gastric tissues, for animal and human models by applying a coupled experimental and computational approach. Experimental tests at tissue, sub-structural and structural level were carried out. Computational models of the sub-structural and structural tests were developed to tune and validate the constitutive parameters. Then, finite element models of post-surgical configurations were generated to point out the modifications induced by bariatric procedures in stomach mechanical response and the differences among the bariatric procedures. The bioengineering models described the post-surgical gastric functionalities, providing mechanical quantities (as volumetric capacity, stress and strain distribution etc.), which are hard-to-measure in vivo. The results showed the potentialities of computational methods applied to surgery, proposing the possible outputs and scenarios, and laying the foundations to clinical tools that will help surgeons in choosing and customising the operation. A multidisciplinary approach that combines clinical and engineering fields could be the key to address the main issues related to BS.

Full textThe additive manufacturing (AM) technologies allow producing components layer upon layer in a completely different way with respect to the traditional techniques. This new approach in building the parts enables unprecedented design freedom; indeed, objects with complex shapes, cellular solids, internal features, and multiple materials can be easily produced. However, even though the manufacturing technologies are ready for producing such components, the literature emphasized that the available design tools are not appropriate and do not allow taking full advantage of the AM capabilities. For instance, the geometric modeling of structures with a lot of elements requires high computational resources, Boolean operations often fail, and the methods are not robust; the modeling of multi-material parts require new approaches able to describe the model at each point of the volume, and not only on the external surface. This research project aims to overcome some of the highlighted limitations by developing new geometric modeling methods suitable for exploiting the capabilities offered by AM. To reach the objective, several research topics have been addressed. Methods for the geometric modeling and optimization were proposed; approaches for the generation of conformal wireframes and for the size and multi-objective optimization of lattice structures were presented. A geometric modeling method based on meshes and subdivision surface algorithms that allows obtaining smooth surfaces without sharp edges was developed; more, the possibility of introducing internal walls and external skins to lattice structures was implemented. These approaches were then introduced in a more comprehensive optimization workflow for AM, dealing with the embodiment design phase and including the possibility of performing size and topology optimization. The proposed geometric mesh modeling method was then numerically validated, and experimental campaigns on both bulk samples and lattice structures produced by several powder bed fusion AM technologies were conducted. The results showed that the proposed mesh modeling method, together with the subdivision surface algorithm, is suitable to design lattice structures efficiently, requiring low computational resources and, at the same time, offering a good dimensional accuracy with respect to reference models. The C2 curvature continuity of the model allows reducing the stress concentration at the nodal points of the lattice if compared to similar structures obtained by traditional CAD software. Furthermore, the possibility of introducing internal walls makes it possible to create optimized ducts and paths and can locally modify the properties of the component. Due to the versatility of AM, the outcomes of the research can be adopted in different fields, as shown by the presented test cases: in the automotive field where lightweight and energy absorption components are used for reducing the fuel consumption and increasing the performance and safety of the vehicles; in the aerospace field, where lightweight components are required; in high-performance heat exchangers, microfluidic applications, and biomedical scaffolds where fluid dynamics plays a key role; and in consumer goods such as sports apparel and equipment.

Full textThe research addresses the issue of humanoid robotics through a design approach aimed at identifying elements of interest in the aesthetic and interactional aspects of such technological products. The scientific objective consists in attempting to define practices and approaches useful for the future design of robots, which can simplify the active inclusion of such figures within society. In fact, the thesis starts from the hypothesis that, given the mechanical and IT difficulties that accompany products of such complexity, one of the reasons behind the constant unfulfilled expectations regarding the presence of robots in everyday life is to be found precisely in the scant attention that has been paid to the design sector and to all those aspects proper to the human-centred approach. The argument is based on a research aimed at introducing what a robot is and where the definition comes from, i.e. the meaning that is currently attributed to the term, through an analysis that is articulated between linguistic aspects and historical evidence of figures that today would be defined as robots. Since the field of research appears to be vast and difficult to define, the thesis intends to focus solely on the category of humanoids, illustrating the reasons why it is believed that the latter may be the protagonists of future systematic use in the everyday lives of users. Even in this category, however, there is some difficulty in defining precise boundaries between the various terminologies used throughout history. For this reason, the dissertation proceeds by means of a novel taxonomy aimed at defining the sub-categories that make up the vast field of humanoid robotics. Having identified the precise limits within which the thesis moves, it proceeds with a state of the art analysis aimed at identifying recurring patterns, good practices, errors or anomalies and, more generally, information of design interest on everything that has been realised from the last century to the most recent years. This compilative part, presented in the form of an appendix, analyses the main humanoid robots made for the market or for research purposes and the main robotic characters in science fiction films, based on the hypothesis that the collective imagination that people have with respect to the field under discussion is defined precisely on the basis of these. Proposals for approaches, best practices and guidelines for the design of human-centred humanoid robots are then presented, defining how good acceptance by the target users can be achieved not so much through a mere search for a pleasing anthropomorphic appearance, but through strict adherence to the methods proper to the project, declined on the subject, and a firm awareness of what the field and the users for whom it is designed are. In other words, humanoid robots should only be realised for those scenarios that really require it and not as part of a present three project experiences carried out during the doctoral course in which an attempt was made to apply what has just been mentioned. In particular, thanks to the Ro.Sa. project, it was possible to study the long-term interaction of weak users with SoftBank's Pepper robot as part of a clinical trial aimed at combating muscle decay in pre-fragile subjects. In the Pos3D project, the participatory design of a humanoid robot was carried out together with users belonging to various categories, within a trade fair context at Maker Faire Rome 2019, succeeding in testing co-design methodologies and collecting data to validate pre-established research hypotheses. Finally, during the SiRobotics project, a service robot was designed starting with the definition of the briefing, passing through the survey with users regarding expectations and needs, carried out thanks to co-design activities, and the collection of further interaction data thanks to online questionnaires, up to the actual design of the product in its aesthetic, interactional and interface aspects. The process was taken care of and followed through to the realisation of a working prototype that was tested with reference users. As a conclusion to what has been said so far, the research analyses the objectives achieved and the answers obtained to the research questions, analysing which hypotheses turned out to be correct and which, on the contrary, were disregarded. However, the concluding chapter provocatively and almost paradoxically presents a number of hypotheses derived from a more-than-human-centred approach, suggesting how the results obtained from the doctoral course could be questioned and refuted in the light of broader ecosystem considerations. The resolution of this controversy can only be pursued through further research aimed at approaching the issue with a critical eye and through an extended approach.

Full textIn recent years, wireless networks have become an ubiquitous, affordable and well-accepted reality. This is particularly true for networks such as Bluetooth and Wi-Fi, which today are almost everywhere and are used for the most different applications. In many cases, when using an off-the-shelf chip, the designer can limit himself to work on the highest levels of the protocol stack and to simply implement the application layer. In other cases, the designer must go down through the stack and work on a lower level where he can see and control some network parameters that he cannot access from the application layer. It happens, for instance, when he has to design a real-time system, or when the required data-rate is high and the available bandwidth allocation must be accurately tailored. Existing simulators usually let the designer work only with very abstract system description, hiding some parameters (such as the packet error rate, the signal power, the corrupted packets, and so on) which many applications have to be aware of. For this reason, the goal of this thesis is to present a new kind of Bluetooth simulator which matches closely the real world implementation of the desired system. We will describe some applications, such as the real-time audio transmission, and we’ll see how these applications can be adapted for other wireless standards, such as the IEEE 802.15.4, with the introduction of a well chosen (and improved) speech vocoder. As a design-helping tool we will present a graphical design environment that let the user draw the system he wants to design, and see the waveform while the debugging is in progress.

Full textFriction is the resistance force to motion between two surfaces that slide or roll over each other. It is a common microscopic phenomenon, the origin of which is to be found in the adhesive interaction forces that are generated between the surfaces in contact. Dissipations due to friction cause huge energy and environmental costs in various sectors, including transport and industry. In fact, it is estimated that nearly a third of the energy produced by fossil fuels to power vehicles is spent on overcoming friction. Improving the tribology of surfaces is expected to ensure a 40% reduction in energy waste within the next 15 years. The study of friction requires a deep understanding of the microscopic phenomena that develop at the surface level. First principle simulations allow studying the electronic and chemical properties of materials with a high degree of accuracy, providing a way to understand tribological events. Over the years, several methods based on first-principle calculations have been developed to study the properties of materials at the atomic level. Among all, the use of these simulations in combination with a high-throughput approach has been spreading rapidly in recent decades. This method aims to simulate thousands of materials in parallel to select from a small set of candidates with specific physicochemical properties from the initial pool. The goal is to accelerate the discovery and dissemination of new materials for science and technology. This thesis explores the implementation and use of several innovative computational techniques in the field of tribology. The work mainly focuses on the description of a software designed to automate the research activity in nanotribology and carry out high-throughput studies on the properties of the interfaces. In its current implementation, the software allows you to combine any pair of crystalline materials, which can differ in both composition and orientation, and study their tribological properties, such as adhesion and shear strength. The program was used to study the tribological properties between two solid surfaces in contact by carrying out a screening on the elements of the periodic table. The study allows to estimate the relationship between adhesion and strength of an interface, also allowing to identify a way to assess the failure mode of materials. Another important result of this work is the realization of an innovative theoretical model that combines the traditional GFMD technique with first-principle molecular dynamics. This hybrid method improves the accuracy of atomistic simulations by considering the role of the elastic properties of solids and the propagation of vibrational waves in determining the friction of atomic structures. The program was used to calculate the friction properties of a diamond-diamond interface, considering different degrees of surface passivation. Finally, this work reports a study conducted on the lubricating properties of phosphorene, an innovative two-dimensional material based on phosphorus. First, two contact phosphorene layers were investigated. This made it possible to identify the presence of a super lubricating phase for a particular orientation of the layers. This work constitutes a first important step in the possible design of phosphorene-based lubricants. Subsequently, the ability of phosphorene to lubricate an iron substrate was evaluated, comparing the results obtained with calculations made for graphene and MoS2.

Full textThe thesis explores the theme of multifunctionality applied to agriculture as a means to reactivate the abandoned territory, degraded, at the risk of misuse or new construction. Agriculture has always had a fundamental relationship with the man and with the settlements. The relationship between urban and agricultural areas has been a relationship of union and opposition. In the transition from subsistence farming to intensive and specialized agriculture, have been involved spaces, techniques, tools, architectures related to agricultural activity and, finally, the relationship with the land. Agriculture, although produce essential goods, in the past decades has been considered a secondary element for economic reasons. There are clear cultural, scenic, ecological, social and economic values of agriculture but this is becoming less profitable and today agriculture is in serious difficulties involving the production, the sustainability , the role of the landscape and the social issues. The difficulties of agriculture also cause the inability of the areas to "defend" itself from the logic of land rent and use linked to immediate profit, as occurs in case of energy plants from renewable sources. All these issues should make territory planner to be careful so that he can find tools to enhance the potential of the agricultural areas. Clearly, the causes are not only economic but also linked to cultural patterns and lifestyles. Addressing the issues of rural areas, their productivity, land use and misuse, it is necessary to carry out a proper planning and sustainable points of view: not only the economic, ecological, historical and social but all of these together. The thesis explores the different European policies adopted to curb the crisis of agriculture. Studies and European documents identify the principle of multifunctionality in agriculture the tool to make the strongest agricultural land. Is useful to investigate, therefore, on the relationship between multifunctional agriculture and regional planning. In particular, we wanted to focus attention on the social aspects as a form of revitalization of the territory and, therefore, on agriculture as a service and social activity that creates cohesion. The questions that the research has set are two: multifunctional agricultural activities and their declination in particular social ones, can be a tool for economic revitalization, cultural and social development of rural areas, periurban and urban areas in a state of abandonment, deterioration, at the risk of misuse or new construction? Through what instrument or instruments such as the regional planning can use and exploit the potential of multifunctional agriculture? To answer these two questions we can't overlook the fact that, from the point of view of spatial and economic, agriculture is measurable in a relatively simple way. It is more difficult to measure the social aspects of the landscape and, therefore, need to refer to the qualitative analysis systems. In addition, the social farming purposes have rehabilitative, educational and business purposes but also related to spatial planning (environmental, economic, historical, cultural and social) and this has greatly been taken into consideration. Another aspect analyzed is the different European approaches to the forms of multifunctional and social agriculture, in the Italian context, and it investigates national and regional instruments that enhance agricultural activities multifunctional. It also explores the relationship between particular forms of social agriculture and areas confiscated from organized crime. The review of examples of multifunctional activities and the deepening of the case studies selected, showing how forms of multifunctional agriculture can revitalize the rural areas, periurban and urban areas. Finally, we arrive at the proposal of actions related to the integration of tools of territorial government, the integration of policies with traditional instruments, the spatial logic necessary to enable the potential of multifunctional agriculture can be considered service to the area and inhabitant.

Full textOccupants' safety in architectural spaces during an emergency is essentially connected to the evacuation process. Current strategies are influenced by a schematic and deterministic approach: it is supposed that building layout and wayfinding systems can directly induce individuals’ behaviors. Interventions on buildings could be enough for reducing people risk, because occupants would surely behave in “the correct way” (e.g.: using right paths). This approach seems to exclude behavioral aspects: experiments demonstrate enormous differences between theoretical and real behaviors in evacuation. Same problems are noticed at both small (building) and wide (urban) scale. Hence, this study focuses on a behavioral point of view and defines a “behavioral design” (BD) approach for increasing people’s safety in architectural spaces. BD is aimed at adapting architectural spaces depending on human behaviors! Hence, method phases include: understanding behaviors in emergency through experiments/real world events; defining and validating evacuation simulation model; analyzing emergency processes through simulator; proposing design solutions based on retrieved critical behaviors and verifying their impact by simulator or drills. The BD method effectiveness is shown by 2 cases. Firstly, an earthquake pedestrians’ evacuation simulator is developed. The model is able to represent man-environment interferences in damaged scenarios and can be used for evaluating vulnerability-reduction interventions on buildings, urban planning and rescuers’ management strategies. The second case concerns wayfinding systems definition in building heritage (theater). Systems are defined so as to address correct evacuation path choices, by reducing overall risks with no architectural modifications. This issue is considerably significant in these buildings because of preservation principles and minimum intervention criterion. This work was developed in accordance with “TAV-progetto SHELL-O.R.4.4” activities.
