Full textIt is now widely accepted that stress plays an important role in modern societies. It impacts the body and the mind at several levels and the association between stress and disease has been observed in several studies. However, there is no consensual definition of stress yet, and therefore there is no consensual way of assessing it either. Thus, although the quality of assessment is a key factor to build robust stress detection solutions, researchers have to choose among a wide variety of assessment strategies. This heterogeneity impacts the validity of comparing solutions among them. In this thesis, we evaluate the impact of several assessment strategies for stress detection. We first review how different fields of research define and assess stress. Then, we describe how we collected stress data along with multiple assessments. We also study the association between these assessments. We present the behavioural and physiological features that we extracted for our experiments. Finally, we present the results we obtained regarding the impact of assessment strategies on 1) data normalization, 2) feature classification performance and 3) on the design of machine learning algorithms. Overall, we argue that one has to take a global and comprehensive approach to design stress detection solutions

Full textPancreatic cancer, also known as pancreatic ductal adenocarcinoma (PDA) is the 4th cause of cancer related death in industrialized countries. The absence of specific clinical symptoms of this disease leads to a late diagnosis, mostly at a metastatic stage. Moreover, the current classification of PDA patients does not allow personalized patient management, which may explain the lack of efficient treatment, resulting in a poor treatment response rate and a 5-year survival rate not exceeding 7%. The important genomic heterogeneity between tumors and the high abundance of the stroma hinder the analysis of the pancreatic carcinogenesis process, and make particularly complex the targeting of a specific pathway. Different classifications of PDA tumors based on mutational landscape and genome wide transcriptomic analysis emerged principally two subtypes "basal-like" and "classical", correlating with patient overall survival but not to a particular sensitivity to a molecule class. Nonetheless, several research groups including ours, demonstrated the presence of a high deregulation in mRNA translation mechanisms in PDA, showing that the abundance of mRNA and the level of the associated protein do not always correlate. My work allowed the identification of a tumor subgroup with a particular translational signature, undetectable at the transcriptional level, through the translatome analysis of 28 pancreatic patient-derived xenografts (PDX) by unsupervised bioinformatics approaches. The Integrated Stress Response (ISR) pathway is constituvely activated in these tumors, in association with a global protein synthesis attenuation. This particularity seems to contribute in increasing resistance to stress, including ISR-activating chemotherapies, and could be explained by an increased resistance to apoptosis in these cancer cells. Nonetheless, the characterization of this subtype revealed an important dependency on a specific amino acid metabolism. As a consequence, targeting this metabolic pathway might be an interesting therapeutic approach for these pancreatic tumors

Full textTraditional procedures for designing structures against fatigue are grounded upon the use of so-called safety factors in an attempt to ensure structural integrity while masking the uncertainties inherent to fatigue. These engineering methods are simple to use and fortunately, they give satisfactory solutions with regard to safety. However, they do not provide the designer with the structure’s safety margin as well as the influence of each design parameter on reliability. Probabilistic approaches are considered in this thesis in order to acquire this information, which is essential for an optimal design against fatigue. A general approach for probabilistic analysis in fatigue is proposed in this manuscript. It relies on the modelling of the uncertainties (load, material properties, geometry, and fatigue curve), and aims at assessing the reliability level of the studied structure in the case of a fatigue failure scenario. Classical reliability methods require a large number of calls to the mechanical model of the structure and are thus not applicable when the model evaluation is time-demanding. A family of methods named AK-RM (Active learning and Kriging-based Reliability methods) is proposed in this research work in order to solve the reliability problem with a minimum number of mechanical model evaluations. The general approach is applied to two case studies submitted by SNECMA in the frame of the ANR project APPRoFi

Full textIf has been stated that several psychological and physiological symptoms are generally induced by the stressful conditions of a polar winter-over (Strange & Klein, 1973 ; Palinkas, 1992 ; Rivolier, 1989). According to the stress episode model (Lassarre, 2002), these reactions appear when individual's health and well-being are severely tested. Consequently, the study of psychological adaptation in Antarctica has great relevance in order to allow the early detection of the winterers who express adaptation difficulties. The study of the Antarctic and sub-antarctic winterers is made with an observation grid. This grid lists the stress reactions that may appear during the course of the winter-over, and is based on a comparative study of different adaptation assessment tools in Antarctica (Rivolier, 1981). The grid is completed every two weeks by the mission's doctor who has to note the presence or the absence of each stress reaction for each winterer. The data collected during 12 different missions between 1998 and 2002 confirm the satisfactory psychometric qualities of the grid. The study of the regroupings of stress reactions leads to a five-factor-classification that describes the five most frequent "adaptive behaviours" of the winterers. The results also allow being more specific about the modalities of appearance of the "third quarter phenomenon" (Bechtel & Berning, 1991). The results are discussed in order to study the contribution of the observation grid for the study of psychological adaptation on the one hand, and to propose different classification of stress reactions to the polar environment on the other hand

Full textThe field of psychoneuroimmunology is among other things dedicated to the question, whether and on what terms psychosocial stress results in disease. Human studies investigating this question often only reveal associations. Consequences regarding mechanisms, causes, and clinical relevance, are thus usually deduced from animal or in vitro studies. The aim of the present thesis was to find an approach, which provides a broader basis for interpretations of correlative findings from human studies. The investigation of patients with Addison´s disease was regarded as the most promising approach. Due to destroyed adrenal glands, these patients are not able to produce cortisol, which therefore has to be substituted. Since no experimental data regarding free cortisol levels resulting from substitution therapy as well as endocrine stress responses are available, in a first step these two question formulations were investigated. Subsequently, this approach was used to investigate two immunological question formulations. The present studies found patients with Addison´s disease to be over-treated. Psychosocial stress resulted in noradrenaline but not in cortisol or adrenaline responses. An injection of 0.03mg/kg hydrocortisone after stress was suitable to induce increases in cortisol levels and cortisol maximums comparable to healthy subjects. Healthy subjects and patients with Addison´s disease did not differ in any baseline immune measures. Stress-induced changes in lymphocyte numbers suggest cortisol being necessary for lymphocyte redistribution subsequent to stress-induced migration into peripheral blood. Without stress-induced cortisol increases, patients are at higher risk for infectious diseases. The trajectories of stimulated interleukin-6 production support the hypothesis of stress-induced cortisol levels protecting the organism against an over-reacting inflammatory immune reaction. Furthermore, in healthy subjects post-stress cortisol levels predicted stress-induced decreases in NF-´B activity, whereas in patients with Addison´s disease noradrenaline predicted such changes. The results of the present thesis supports the assumption of Addison´s disease being a promising approach to test the validity of psychoneuroimmunological in vitro and animal data in human whole organism. Additionally, the present data emphasize the astonishing adaptability of the immune system. This further emphasizes the necessity to investigate psychoneuroimmunological processes utilizing the criteria of hormone subtraction and hormone substitution also in human research. In future studies investigation of patients with Addison´s diseases will certainly provide crucial evidence regarding the clinical relevance of a normal endocrine stress response

Full textMedicare Part B is one of the federal health insurance programs available to senior citizens in the United States. Unlike Medicare Part A, Part B enrollment is not automatic, and those missing their initial enrollment period are assessed a 10% or more penalty in addition to their monthly premium rate for the rest of their lives. This problematic enrollment policy has impacted senior citizens who have missed Part B enrollment windows, creating for them an added financial burden when many are transitioning to fixed incomes. Guided by social construction theory and using a nonprobability, convenience sampling approach, the likelihood coefficient values associated with Medicare Part B enrollee awareness, stress, and income of 112 residents of a suburban city in a northeastern state who were 65 years and older were examined. Sequential Forward: LR methodology yielded a significant, negative (b = –1.21, Wald X2(1) = 7.56, OR = .298, p = .006, CI [.126, .707]) and a significant, positive (b = 2.16, Wald X2(1) = 6.29, OR = 8.678, p = .012, CI [1.60, 46.99]) likelihood of predicting Medicare Part B late enrollment penalties for awareness and stress; income was not a significant model predictor. Participants who reported higher stress levels were 8.7 times more likely to be classified in the Medicare Part B late enrollment penalty than those reporting lower stress. Participants who were aware of enrollment needs were 3.4 times more likely to have no late enrollment penalties than those who were unaware. Positive social change centers on increasing Medicare Part B consumer awareness, reducing stress of enrollment deadlines, and providing information to federal policy makers to simplify enrollment policies to reduce or end late enrollment penalties.

Abstract: The regulations of standards, standardization of farms in the welfare, and the demands of consumption are causing a major change in the production of broiler chickens in Brazil. With these changes are occurring with the development of new technological tools, which are used together in animal production systems, can aid in decision support systems, focused on the estimation of animal welfare. Thus, the vocalization of the animals was shown to be an important analytical tool, providing information in a non-invasive way, and enabling its use in an automated manner. The vocalization of the animals shows the expression of its condition, and it may occur spontaneously or as a result of some external event, e.g., pain, hunger, thirst. The objective of this study was to implement a pattern recognition analysis of broiler chickens, including sex, and to relate these patterns as an indicative of welfare. This research took place in two stages and during the first days of life of birds. The birds were divided into two groups, the first group and the second with 15 birds with 10 birds were provided food and water ad libitum. They were then placed in a semi-anechoic chamber and vocalization was recorded using a unidirectional microphone coupled to a digital recorder. In the experiment of gender, 120 chicks Cobb® and Ross® were used, and they were previously sexed using the characteristics of the feathering of the chicks. The vocalizations were observed within an average of two minutes. Analyzes were performed on the acoustic structure of vocalizations of chicks, and we calculated the "Pitch" intensity, and the 1st and 2nd formant. In the first experiment, the results showed that different birds vocalize when isolated and in groups. The chicks, when isolated, emit a high sound intensity that is identified as "call of danger". The amount of signal energy is influenced by the weight of the birds. As the results of the identification of gender, the vocalizations of both sexes were statistically different. The results of the vocalizations of day-old chicks indicated that there is difference between the vocalization of males and females

Full textThe purpose of this study was to analyze inventory inaccuracy at Haldex. In order to answer the purpose a qualitative method in the form of interviews, a survey and observations were conducted. The empirical material was then later analyzed with relevant theories regarding inventory inaccuracy. This study found that the inventory inaccuracy at Haldex mainly was a result of human errors combined with technological errors. The human errors came from a lack of knowledge in the company's ERP system AX12. The interviews found that the system had been implemented too fast and without relevant training, which later resulted in employees not knowing how to properly use it. The technological errors came from the company's weak criteria’s when it came to classify the inventory. Today Haldex uses an ABC-classification that is based on the products value, this essay suggests that lead time and importance of the product should be added. In order to minimize inventory errors this essay suggest that RFID technology needs to be added to the company's A-products.
