Different dog sizes are associated with variations in digestive physiology, mainly related to the large intestine and its resident microorganisms. This gut microbiota plays a key role in animal health, supporting nutritional, immunological and physiological processes. Nevertheless, diseases or antibiotherapy can disturb microbial equilibrium and induce a perturbated state called dysbiosis. To restore microbiota eubiosis, new restorations strategies have been developed such as pre-, pro- or postbiotics. However, very few studies have evaluated their effects on gut microbiota in the context of antibiotherapy. This joint PhD between the Microbiology, Digestive Environment and Health unit from Université Clermont Auvergne and the two compagnies Lallemand Animal Nutrition and Dômes Pharma, aimed to investigate the impact of body weight and antibiotic disturbance on canine colonic microbiota, as well as the potential of microbial restoration strategies, using in vitro gut models.This thesis started by evaluating the impact of different methods for faecal sample storage (48-h freezing -80°C, 48-h -80°C with glycerol or lyophilization with maltodextrin/trehalose) on the kinetics of microbiota colonization and metabolic activities in the Mucosal Artificial Colon (M-ARCOL). Compared to fresh stools, inoculating with raw frozen stool without cryoprotectant was the best option among those tested. Second, thanks to a large literature review, the M-ARCOL model was adapted to reproduce the main nutritional, physicochemical and microbial parameters specific from small, medium and large size conditions in a new model called Canine M-ARCOL (CANIM-ARCOL), further validated through in vitro-in vivo comparisons. This adaptation allowed to reproduce in vitro the increase in Bacteroidota and Firmicutes abundances and higher main short-chain fatty acid (SCFA) concentrations observed in vivo. Then, we used the CANIM-ARCOL to perform a mechanistic study, which revealed that nutritional and physicochemical parameters are enough to shape microbiota activity according to dog size, but faecal inoculum was necessary to reproduce size-related microbiota composition. The next step was to adapt the CANIM-ARCOL to diseased situation, focusing on antibiotic-induced dysbiosis. In accordance with in vivo data, antibiotherapy induced an increase in Enterobacteriaceae, Streptococcaceae and Lactobacillaceae relative abundances while alpha-diversity and SCFA production decreased. Similar but lower effects were observed in mucus-associated microbiota. Lastly, we evaluated the effect of the live probiotic yeast Saccharomyces boulardii CNCM I-1079 and the heat-inactivated bacteria Lactobacillus helveticus HA-122 on microbiota resistance during antibiotic treatment and resilience afterwards. Of interest, both microbial strategies decreased the Enterobacteriaceae bloom during antibiotherapy and allowed, in the first two days, a quicker recovery of microbiota composition and activity, in both the luminal and mucosal compartments.This PhD work provided pioneering and significant insights into the impact of dog size and antibiotherapy on canine colonic luminal and mucus-associated microbiota composition and activity, filling gaps in knowledge in these fields. This work also contributed to a better understanding of microbiota resilience in response to antibiotic disturbance. In a near future, in accordance with the European 3R's rules aiming to reduce at a maximum animal experiments, our in vitro approaches could be used for mechanistic studies on the interactions between nutrients, feed additives or veterinary products and canine colonic microbiota. Such experiments could be performed under healthy but also disturbed gut microbial situations (including obesity, inflammatory bowel diseases or chronic enteropathies), always considering interindividual variabilities to move towards personalized nutrition and medicine

This thesis studies the relationships between diplomatic activities and the publication of books, journals, and pamphlets in the mid-17th century in the context of the initial development of Portuguese Restauration diplomacy in France. It is about examining in detail the goals, field of action, and initiatives of the new monarch’s representatives who intervene in the circulation of political information and in the world of books and typography workshops, giving exclusive attention to the circumstance of the first mission sent to Paris in 1641 and to some aspects of actions of the following embassies, until the return to Lisbon of the Marquis of Niza’s delegation in 1649.It will be possible, firstly, to distinguish a series of circumstances and publications prior to the arrival of the Portuguese envoys and then observe visits and contacts that they establish in France, as well as all kind of daily occupations linked to the diffusion of information, writing, and the circulation of hand-written and printed pieces. This narrower time scale will allow, in addition, investigations on the literary, rhetorical, and informational practices that were part of the editorial landscape found in France by John IV’s agents. All these studies contribute to a meticulous reading of discursive and material elements present in the publications that the embassy produced, or had hoped to incentivize, with the goal of reconstituting as much as possible the intentions of its writers and editors, or even the most immediate reflections and reactions that these publications could arouse in readers and in a substantial part of French society

Heat, induced by fire, is particularly devastating for cultural heritage objects as it causes the destruction of all or part of these witnesses of the past. In this study, we focused on leather, a material largely present in heritage collections. Until now, no restoration method has been developed to treat the damaging effects of heat.The first aim of our study was to evaluate the effects of dry heat on leather samples through a systematic characterization. Model samples from a calf skin vegetable tanned in known conditions were used and the consequences of heat exposure was characterized at different structural scales using a range of physical, chemical and biochemical methods.Besides the visible browning and shrinkage of leather, heat induces many changes including a loss of mass, the melt of the crystalline regions, an increase in both hydrophobicity and rigidity. Some of these changes result from the protein aggregation induced by exposure to heat and evidenced by our research.Our second goal was to develop an innovative restoration method based on the use of biological molecules in order to respect the nature of the object. Enzymes such as proteases, able to hydrolyze protein aggregates, were used. One of the challenges was to provide the water necessary for the enzyme activity without wetting the leather surface in order to avoid further damage of the leather. Several enzyme supports were tested. The use of an enzymatic emulsion reveals neither darkening nor retraction and in some cases a flexibility gain is observed. Encouraging results were also obtained in the case of an ancient book cover made from calfskin and dated from the nineteenth century. Additional measurements lead to attribute its effect mainly to the emulsion itself, however longer-term measurements appear to show a positive effect of the enzyme on the flexibility gain. Although further characterizations on the long term are required, the treatment may constitute a new support for leather bio-restoration.Keywords: leather – thermal denaturation – protein aggregation – bio-restoration - proteases

Adopted by article 2 from act nr 2016-1087 concerning the reconquest of biodiversity, nature and landscape of August 8th 2016, the principle of ecological solidarity calls “for taking into consideration the interactions of ecosystems, living creatures and natural or developed environments in all public decisions having a notable impact on the environment of the territories concerned”.This general principle of environmental law inscribed in article L110 1 of the environmental code is designed to preserve the interactions of ecosystems and ecological processes as well as to improve the environmental management of the territories. The duality of its objective renders it a highly valuable principle which should be applied to numerous decisions. Various supranational foundations, within jurisprudence and in international and European laws, can thus be assigned to it. In the context of ecological interdependence the evolution of the international and European dimension of the principle remains essential in order to see the benefits concerning the safeguarding of biodiversity. It is greatly enhanced, as regards internal law, by the fact that it is at the very center of the environment's equilibrium. It justifies the right to a balanced environment and extends the constitutional principles of prevention and sustainable development. Confronted with the crisis of world-wide biodiversity extinction that threatens our survival, the principle of ecological solidarity emerges as the last chance to preserve it. Two major groups of systems allow us to measure the dynamics of the principle of ecological solidarity: the green and the blue line belt network and the Ocean and coastline Governance Framework. They constitute a basis on which to develop ecological solidarity and represent the beginnings of new legislation. The principle of ecological solidarity requires them to be reinforced and more generally to advance all decisions concerned by the principle
