Full textIn gene therapy, decoy strategy consists in using short nucleic acid containing a sequence specificallyrecognized by a transcription factor in order to trap it and in turn to inhibit the expression of genesunder its control. The therapeutic efficacy of this strategy is related to the biostability of the nucleicacid and the possibility to target several transcription factors that cooperate to induce cellderegulations responsible in human diseases. We have developed a technology to product in vitroDNA minicircle less than 250 base pairs which can interact with several proteins, chemicallyfunctionalizable and resistant to exonucleases. A dual targeting of the transcription factor NF-κB by aminicircle allows inhibition in cellulo of a NF-κB-dependent transcription reporter system consistentwith a decoy activity induced by the minicircle.Cisplatin is an anticancer drug whose pharmacological target is DNA. In several cancer cell lines, thedecrease of transcriptional activity of NF-κB has been linked to an increase in the sensitivity of cells tocisplatin. We studied in cellulo whether the mechanism of inhibition of NF-κB by cisplatin could dependon the platination of its consensus sequence using a novel application of the decoy strategy. We showthat cisplatin adducts, unlike those of transplatin (inactive isomer), decreases recognition of NF-κB toits consensus sequence in vitro and in cellulo.Decoy strategy with minicircle DNA will be developed to make the proof of concept of this approach atthe cellular level and further to demonstrate its biological effectiveness

Full textMyotonie dystrophy types 1(DM1) and 2 (DM2) are autosomal dominant multisystem diseases with a strong neuromuscular component. They are characterized by progressive myotonia, muscle weakness, cognitive impairment, and cardiac conduction defects. These diseases are caused by abnormal amplification of C(C)TG repeat sequences located in the 3'UTR region of the DMPK gene and in the intron of the CNBP gene, respectively. These expansion-containing sequences are transcribed and retained in the nucleus as riboprotein aggregates. The presence of these toxic C(C)UG RNAs induces sequestration of the MBNL family of RNA-binding proteins, leading to their loss of function and deregulation of alternative splicing events, many ofv/hich are associated with clinical symptoms in patients. There is currently no1reatment for DM. In this thesis, I have developed a gene therapy tool based on a modification of the MBNL1 protein. This C- terminal truncated MBNL derivative (MBNLΔ) acts as a decoy to release endogenous MBNL proteins sequestered by mutant RNAs. Our approach was validated in muscle cells from DM1 patients and in a mouse model of the disease after AAV virus injection. Treatment with MBNLΔ allows the delocalisation of endogenous MBNL proteins from the foci, modifies the foci dynamics, corrects the transcriptome and myotonia, which is maintained 1 year after injection

Full textResearchers in psychology have shown that we use several strategies to accomplish cognitive tasks. A strategy is a procedure or a set of procedures for achieving a higher level goal or task. One of the crucial issues concerns how we choose a strategy among several available strategies to solve each problem. To further our understanding of strategy selection, the overall objective of this thesis was to investigate processes involved in strategy selection, and their evolution with aging. To achieve this goal, we adopted both neuroscience and cognitive approach. Data reported in our fMRI studies indicate that a specific network was associated with strategy selection. This particular network includes prefrontal regions known to be engaged in tasks involving executive functions. Thus, our data suggest that, contrary to the assumptions of theoretical models, executive functions may be involved in the strategy selection. The lack of activation in these regions in older adults may help us to understand why the ability to choose the better strategy decreases during aging. Interestingly, we also show that participants can use different strategy selection processes based on the difficulty of problems. Finally, in the last section, we discuss the implications of our results for theoretical models of strategy selection, and for cognitive aging

Full textAbstract : This descriptive research work is born out of a question from a teacher/researcher point of view: What strategies and pedagogical actions should a CEGEP science teacher emphasize in order to stimulate the motivation of the students and get them engaged in their learning activities ? The pedagogical stakes are high in the CEGEP sector; scientific studies point to a large drop out rate, a weak level of motivation and students engagement towards their studies, and a success rate below intended targets. The teacher can play a primary role in promoting student engagement towards their studies, be it through the choice of pedagogical strategies, the selection of pedagogical activities or through the support that can be given to the learner. The distinctiveness of the CEGEP system where both pre-university and technical programs are found side by side has yet to be studied from a comparative perspective on motivational dynamics as it relates to student engagement in a scientific curriculum. The general goal of this study is to describe the perceptions of students, as they relate to motivation stemming from different pedagogical actions and strategies utilised by teachers in two CEGEP level programs, the pre-university Science program and the Laboratory Techniques program, Biotechnology profile. Each of the 16 participating teachers in Biology and Physics within both programs were observed during one or two teaching periods totalling 58 hours of teaching for 29 groups of students, so as to identify the general pedagogical strategy of the period and the pedagogical actions that were enacted in the classroom. Students then completed an online survey regarding their perceptions as they relate to their motivation with regards to the strategies and pedagogical actions. Surveys from 272 respondents were treated, analysed and interpreted so as to portray the motivational dynamics along comparison groups through statistical analyses and a qualitative analysis of the respondents’ spontaneous comments. Biotechnology students’ perceptions favor motivation because they attach more value to the pedagogical activities than Science respondents. Their desire for engagement is also higher. As for Biotechnology male students, they feel significantly more competent than all other respondents, regardless of gender or program. They find it easier to participate in pedagogical classroom activities, and rarely mention negative emotions related to said activities. Students in both programs show a large variety of perceptions regarding learning activities and pedagogical strategies; they can even be of opposite nature among students of a given teaching period, indicating that using a variety of pedagogical acts is to be encouraged in order to stimulate the largest proportion of students.

Full textThe globalization of the economy, and of the markets; the development of new techniques of information and communication; the disappearance of the geographical and time barriers, has led to the creation of new forms of organizations in the setting up of strategies within the network and also to new forms of management and of control. The multinational business and the intra organizational relations between the subsidiaries and the parent company gave place to many works. The specificity of the multinational business lies in the fact that it is submitted to a dual problem: on the one hand it must manage the business in its whole and therefore set up the tools of integration, and on the other hand, it undergoes the pressures of the parent company. The present research intends to integrate the problematic of the influence that the evolution of the organizational structures of the multinational business can have on their localization strategies. It is in the perspective of this evolution that the organization of the parent-subsidiary links appear in our research. To do so, first, we will demonstrate how the organizational structures of the multinational business evolve and how they influence the strategies of expansion. Second, we will analyse the interactions of the organizational structures and of the multinational business with their anagement systems by assuming that the latter are both the expression of the decision making process and of the power relationships regulated by the organizational structures. Through the general setting provided by the strategies of expansion according to the evolution of the organizational structures, there is an order of power rule which helps to direct and to ensure the coordination and the control of the organization activities. It is what we mean by monitoring systems. To describe these reports better, we have examined the systems of power and the parent-subsidiarylinks implied in the concept centralization and decentralization

Full textPleiotrophin (PTN) and Midkine belong to the Heparin Binding Growth Factors (HBGF) family, and are known to promote cells proliferation and angiogenesis along tumorigenesis process. We first evaluated a PTN inhibitor: the PTNΔ111-136 peptide, in breast cancer model. This peptide appeared to be an efficient inhibitor of tumor growth in vitro and in vivo. It is also able to inhibit in vitro the proliferation and migration of human endothelial cells. This peptide acts on two different important steps of angiogenesis : tumor cells proliferation, and tumor angiogenesis. We then evaluated a MDK inhibtor: the MDKΔ81-121 peptide, in tumors derived from neural crests: neuroblastoma and neurofibrosarcoma. The peptide induces the inhibition of endothelial cells proliferations, and of tumoral angiogenesis, and also inhibits tumor cells proliferation in vitro and in vivo

It uses an empirical research which, through the process of triangulation, to explore the role of stakeholders in the management of their territory.A theoretical analysis of main concepts related to the territorial governance will highlight a conceptual framework adapted to the study of tools available to actors to promote a territorial governance. Indeed, this study suggests an approach that integrates the various theories to determine all the factors that allow players to register in a context of territorial governance. The thesis introduced the project in terms of issues and methodology, through a survey of the 20 municipalities of the Meurthe and Moselle and a content analysis of various reports and internal memoranda relating to a company located in a department of the North eastern France. Research suggests that the territorial model of territorial governance can not be considered as universal. However, it can be invested through certain characteristics related to rationality, cooperation and decision-making process

Full textThe European regulation CE 1126/2007 dictates the maximal mycotoxins contents allowed in cereals. Their direct measurement with the reference methods is long and tedious. Furthermore, it is destructive and cannot be used at silo. Alternative tools such as infrared spectroscopy are studied. The objective of the first part of the study is to apply near infrared spectroscopy to identify and discriminate Fusarium isolates, grown on solid culture medium, without preparation of the sample. This approach should allow discrimination of Fusarium species most abundant in the corn : Fusarium graminearum, Fusarium proliferatum, Fusarium subglutinans, Fusarium verticillioides. The infrared spectra of 58 strains belonging to these four species were collected on a spectrometer. A model based on artificial neural networks was developed for the species discrimination. With this model, the correct classifiaction on the external validation set was very good (98. 8%). The objective of the second part is to sort the corn samples regarding their deoxynivalenol and fumonisins contents. More than 2000 samples were used in this study. Their infrared spectra were collected on a near spectrometer, and they were referenced for their mycotoxins contents with chromatography methods. The performances of the infrared models developed to quantify the deoxynivalenol (DON) and the fumonisins (FUM) contents are not good enough to be used in the field, even if the support vector machines approach gives interesting results. Thus, qualitative models were developed to sort the samples in three classes : 'no risk for DON and FUM', 'risk for DON and/or FUM' and a middle class 'samples to be analysed by reference method'. The objective of the last part is to study the link between contents of ergosterol, fumonisins and fungal biomass (Colony Forming Units-CFU) in 117 corn sapmles. A fungal cell count was also done for 34 species. The near infared spectra of the corn samples were collected and used to predict the fungal biomass and the ergosterol contents

Full textThe general objective of this thesis was to study the impact of past resources of individuals on perceived memory compensation (Study 1 and Study 2) and on objective external memory compensation (Study 3). In order to operationalize the evaluation of perceived memory compensation strategies, Study 1 involved the French validation and normalization of the Memory Compensation Questionnaire (F-MCQ) of 749 participants aged from 18 to 92 years. In order to adapt this tool to different contexts, we created a shorter version (BF-MCQ) and conducted his validation on 260 participants aged from 50 years to 85 years in study 2. The results of these two works show a satisfactory internal validity for these two scales, and normalization reports an effect of age and gender on the perception of memory compensation strategies that an individual thinks that he use in everyday life. The external validity of these two scales showed the links between stress, self-memory efficiency and personality with the five compensation strategies measured by these tools. Our third study tested a) the impact of the personality trait "Openness" and of controlled processes on the use of external aid and on its effectiveness in prospective recall situation with 80 participants aged from 50 to 92 years and b) the link between behavioral observations of participants and their own perception of behavior during the experiment. The results show that only the controlled processes are involved in the mobilization of external strategies and in their effectiveness. Although seniors reported using external aids daily and frequently in the French Memory Compensation Questionnaire, we did not find this behavior to occur significantly in situation. In the end, these three studies allow us to consider the external memory compensation as a process that depends on self-initiation of cognitive resources, and to highlight the impact of various emotional and psycho affective factors in the perception that individuals have about their daily compensatory mode

Full textIn search of new bioactive compounds, marine bacteria constitute a considerable source of innovative molecules. The bacterial exopolysaccharide (EPS), produced by the deep-sea hydrothermal vent strain Alteromonas infernus, is a high molecular weight, highly branched and anionic heteropolysaccharide with a nonasaccharide repeating unit. This macromolecule and its low-molecular-weight derivatives obtained through a chemical process have previously displayed interesting GAG-like properties such as anti-metastatic and anti-coagulant (heparin-like) ones; they can also improve stem cell differentiation.To investigate the molecular bases of the biological activity and to facilitate the structural analysis of the bioactive derivatives (~20 000 g/mol), fragments of the native EPS molecule have been prepared by organic synthesis and enzymatic depolymerisation.The first part of this thesis is dedicated to the synthesis of the disaccharides composing the repeating unit of the EPS. The synthetic pathways to acceptor and donor glycosyls have been developed and two protected disaccharides have been synthetized.The second part is devoted to the preparation of oligosaccharides and low molecular weight derivatives by enzymatic depolymerization of the native EPS. A new generation of low molecular weight derivatives has been produced and a new bisulfated octasaccharide has been characterized by mass spectrometry

Full textThe purpose of this research was to study the coping strategies of teenagers when they are confronted by the criticisms of their closest friends in their class about a brand choice and their effect on brand attitude and repurchase intention. We have developed a situational measurement of coping strategies of teenagers. We have found five coping strategies: the justification, the revenge, the denial, the fact of making feel guilty of criticizers and the self-reproach. The final study was carried out on 446 teenagers. A part of the hypotheses were validated: certain negative emotions felt immediately after exposure to the stressor influenced the strategies of teenagers. These latter could have an adaptative or maladaptative effect on the negative emotions which are felt after coping. Finally, these last emotions, according to their nature, could have an influence on brand attitude and/or repurchase intention. Several effects of moderating variables were also analyzed

Full textIn spite of a very rich and various literature about suicidal behaviour, research suffer from confusions between a large range of situations, acts and subjects’ characteristics, and seldom take the suicidal functions or intentions into consideration. However, this functions could explain the links between subjects’ coping skills and the parasuicides or suicide attempts. Objectives : the first objective of this research was to propose a french validation of a suicidal functions questionnaire, the RASQ (Holden et al. , 1998). The second was to try to explain why people engage in suicidal behaviour, and its relationship with coping strategies and psychopathology. The third was to study attempters according to their previous attempts, the suicidal functions of their last attempt, their coping strategies and their psychopathology, to explain why they commit and why they repeat suicidal behaviour. Method : in a cross-sectional design, 398 subjects were interviewed and were then asked to complete various self-questionnaires. Results : a two-dimensionnal structure was found in the RASQ : « agression / control » and « avoidance / relief ». Compared to suicidal ideators, attempters engaged in more avoidant coping ways, specially more distraction, and were more frequently diagnosed with Cluster B personality disorders. Distraction and female sex were the only significant independent predictors of an attempt. Attempters can’t be clearly distinguished by the number of previous attempts, but by the nature of their attempt (suicide attempt vs parasuicide), their gender, their age, and the suicidal functions of their last attempt. Hopelessness and personality disorders are the only significant independent predictors of the attempts severity and repetitions. Conclusion : Distraction, female sex, cluster B personality disorders, and hopelessness are the most involved dimensions into suicidal behavior and its repetition. Single and multiple suicide attempters don’t make two clear distinct populations and suicidal intentions could improve the qualitative understanding of the attempt. Attempts and repetitions could be better understood by follow-up and qualitative studies, and by assessing each attempt, its caracteristics and consequences, and « state-trait » coping variations

Full textThe dynamic capability of an organization is its ability to constantly change and develop its resources, skills and processes to meet the needs of, and act upon its changing environment. However, the link between dynamic capabilities (DC) and inter-organizational relationships is rarely developed in the literature. Therefore, our dissertation deals with whether the network environment, through its structural and contextual characteristics, is suitable for the deployment and the development of DC. We question the processes that enable the deployment, detection and the creation of DC to understand whether DCs operate individually or in combination, and try to understand which DCs operate. Also, we question the inter-organizational nature of dynamic capabilities in territorial networks. Can DC be built or emerge from interactions in these networks? To answer these questions, we chose to conduct in-depth interviews in different territorial networks in the PACA region in France. Results of this empirical study show the need to take into account collective actions initiated by these networks of organizations, underline the role of proximity (broadly defined) as a means to deploy dynamic capabilities, and indicate the emergence of "network dynamic capabilities". Research results allow us to confirm the role of structural and contextual characteristics of territorial networks on the deployment and the creation of these capabilities and allow us to identify them in these contexts

The first part is focused on the constitution of the patrimony, the feudal rights the Borghese disposed upon and the management of the fiefs. In the second part, I show the the extent of the juridictional powers of the Borghese -which denies the thesis of an efficient administrative centralization in the papal state- as well as the strategies of the feudal officers and the recourse of the vassals to the feudal jurisdiction. In the third part, I analyse the changing relationships between the lords and the other institutions, the conflicts and alliances between officers and the local factions and the transformations of this situation in the 18th century, when the control of the papal magistratures over the fiefs becomes more efficient

Full textThe main purpose of this thesis is to study the role and the place of the financial instruments (known in France as "quasi-fonds propres") in the CEO's power and managerial entrenchment. The financial theory seems to be unable to explain the use of this these financial instruments by the companies. The contractual theories give these instruments the role of regulation. A comparison of these theories with the entrenchment theories supports the idea that these instruments can be used for the power and the entrenchment of the managers. A quantitative method on a sample of French listed companies for a period between 1990 and 2003 enables us to study the relation between the se instruments and the managerial entrenchment. The results of this study confirm the role of these instruments in the managerial entrenchment and the influence of managerial entrenchment on the decision of issuing such instruments

Full textLoss of control situations are innumerable (e.g., global pandemic, romantic break-up) and most often give rise to attempts to restore the perception of control. These attempts, in the form of often unconscious defensive strategies, restore meaning and predictability to events. Anthropomorphism, or the attribution of human traits to non-human objects including social robots, is conceived in the literature as a compensatory strategy for a lack of (perceived) control. This process would allow the application to robots of social scripts typically deployed in human interactions, making them more familiar, predictable and controllable. The objective of this thesis is to examine anthropomorphism as a compensatory strategy for a lack of control and its impact on acceptability. It is divided into two steps: 1. to identify the conditions of application of this strategy, 2. to examine the capacity of anthropomorphism to structure, give meaning and make coherent the social environment. To these ends, the thesis articulates three major concepts: perception of control, anthropomorphism, and acceptability of social robots. Using a scoping review and a state of the art, the acceptability of social robots and possible barriers to their adoption are examined. Then, the development and validation of a tool, a correlational pilot study and an experimental pilot study initiate the first relationships between these three concepts. In the extension, three experimental studies test in turn three types of operationalization of control (on life events, on the robot, on the task) and allow to identifysome causal links between control, anthropomorphism and acceptability of social robots. Finally, two experimental studies test the structuring quality of anthropomorphism. All these studies show that the perception of the social robot changes according to the type of control induced. Similarly, anthropomorphism (i.e., superficial aspects of the robot such as appearance, behavior, etc.) and anthropomorphization (i.e., essential aspects such as mental states, emotions, etc.) influence differently its usage intention. Full textRecursive movement patterns, by which an individual returns to already visited sites, are very common. Memory use, hypothesized to be advantageous when the environment is predictable, could underlie the emergence of these patterns. However, our understanding of the memory-movement interface has been limited by two knowledge gaps. We still lack appropriate methodologies and theoretical knowledge of the advantages of memory use and of the patterns that emerge from it. During this PhD project, I aimed at filling in some of these gaps. I present here three new frameworks for the analysis of recursive movement patterns. The first one delimits the areas most frequently revisited by an individual, the second one detects periodic revisit patterns, and the third one formally defines and quantifies routine movement behaviour in terms of movement sequence repetitiveness, and presents an algorithm that detects the sub-sequences that are repeated. Using an individual-based model, we show that memory use, when the environment is predictable, is very energetically advantageous compared to foraging strategies that do not use memory, including in a situation of competition, and that it leads to the emergence of stable Home Ranges and spatial segregation between individuals. Memory use invalidates several hypotheses very commonly made in population studies, by leading to a stronger environmental depletion, to a higher equilibrium population size, and to a nonlinear relationship between the total population size and the individually-experienced intensity of competition. Therefore, my PhD thesis contributes to a better understanding of the consequences of memory use for the fitness of individuals, for movement patterns, and for population dynamics. It offers innovative methodologies that quantify and characterize recursive movement patterns that can emerge from its use. These methods should open new opportunities for the comparison of the movements of individuals from different populations and species, and thus the testing of hypotheses about the pressures that select for memory use

Full textNucleosidic analogs are widely used as therapeutic agents in antitumoral chemotherapy. However, cellular resistance appears in a multifactorial manner and it seems that overexpression of an enzyme, the 5'-cytosolic nucleotidase (cN-II) may be involved in this phenomenon. We were interested in the study of this enzyme in order to design and synthesize potential inhibitors belonging to the family of nucleoside analogs in the á/â-modified phosphonate series. First, we reviewed literature data on the nucleotidase family and particularly on cN-II, then we engaged in the setting up of an evaluation test using various enzymological techniques. Thus, green malachite colorimetric assay was successful and efficient for the evaluation of the inhibitor capacity of our different derivatives. Secondly, we described the optimisation and the synthesis of alkynylphosphonate nucleoside analogs using two different pathways. These latter were tested and the cytosine analog inhibited 65% of the enzyme activity. Then, a new synthetic approach to â-modified nucleoside phosphonates was reported using a â-ketophosphonate as key intermediate. â-hydroxyphosphonate et â-oximephosphonate analogs were obtained and inhibited 50% of the enzyme activity. Finally, a novel prodrug stategy for the synthesis of bisSATE ester of â-hydroxyphosphonates (our leading series) was designed and developped

Full textSTAT family members have various modulating roles. Within this family, STAT1 and STAT3 are very similar and can regulate the same target genes but do have opposite effects. Whereas STAT1 is considered as an anti-oncogene, STAT3 is rather an oncongene. The transcription factor STAT1 modulates several pathways involved in cellular protection. STAT1 has antiproliferative and proapoptotic effects. A role for STAT1 as a modulator of the effect of genotoxic agents has been demonstrated and seems to depend on both the type of genotoxic agent and of the cell line studied. STAT3 is often activated in several types of cancers and is located in the nucleus. Anticancer strategies targeting the pro-proliferative and antiapoptotic STAT3 are promising. We have first shown that STAT1 regulated the expression of MLH1, an essential factor of the mismatch repair (MMR) system, and of c-Abl. STAT1 deficiency was associated with DNA lesions persistency, sustained G 2 /M cell cycle arrest and increased sensibility to the alkylating agent MNNG. Also, STAT1 entered into an active complex including STAT1/MLH1/c-Abl/p53 which could modulates the cell fate following MNNG exposure. Second, we have shown that inhibition of STAT3 with a decoy oligonucleotide (ODN) containing a GAS sequence could trap activated STAT3 dimers in the cytoplasm. This ODN impaired binding of STAT3 to karyopherin and induced a STAT1-dependent cell death. However, this ODN can also bind to STAT1 and hence it needs to be optimized in order to improve STAT3-specificity. The combination of a decoy-ODN strategy with a conventional chemotherapy should allow to synergize the action of anticancer drugs and to decrease their side effects

Full textThis thesis, at the crossroads of literature and linguistics, intends to study the beginning of fifty English and American great novels selected over three centuries for their enunciative richness. In order to put into evidence the triangular correlation between the narrator-author, the text and the reader, two main functions sustained our study: the poetic function (part one) and the seductive function (part two). Three literary techniques; opening the novel, presenting the plot and the characters putting the plot into action, conveyed the narrative tension within the initial, middle, and final phases of an opening and put into evidence the emergence of rhetorics for each century. Three grammatical markers known for showing this author's subjectivity in his literary creation were selected: pinpointing operators such as demonstratives and determiners, comparative structures and modal auxiliaries. Each marker has been categorized according to specific aspects of fictive openings associated with binary linguistic explorative tools. Therefore, we have considered the anaphoric and cataphoric references conveyed by pinpointing operators related to the characters, the place and the plot introduced at the beginning. As for comparative structures, we have organized them according to three standards: the extra-linguistic universal standard identified by both writer and reader, universal references that need to be proven by the rest of the plot and finally the references that cannot be identified by the reader because they depend on the fictive context. The analysis of the modal auxiliaries emphasizes the descriptive, informative and explicative elements opening a novel
