Abstract:
The whole genome duplication (WGD) event is an intriguing mechanism from an evolutionary perspective. Such an event may be the source of new genes, functions or species. Traces of WGD event have been detected in the genome of all four eukaryotic kingdoms: plants, animals, fungi and protists. In fungi, an ancestor of Saccharomyces cerevisiae underwent an event of WGD, about 100 million years ago, after diverging from the Kluyveromyces lineage. In S. cerevisiae, only ten percent of the resulting duplicated genes survived as duplicates. In particular, some of these duplicates encodes for transcription factors in several nutrient sensing pathways.

The main subject of this thesis’s work is the external amino acid sensing system in S. cerevisiae. The detection of extracellular amino acids in yeast begins with a transporter homologue devoid of any uptake activity, the Ssy1 sensor. The binding of extracellular amino acids to Ssy1 leads to the successive activation of Ptr3 and the Ssy5 endoprotease. This endoporotease catalyses the processing of two transcriptions factors: Stp1 and Stp2. The Stp factors, released from their N-terminal cytoplasmic-anchored domains, are then translocated into the nucleus, where they activate the transcription of several amino acid permease genes (e.g. AGP1 and DIP5). Starting this work, the Stp factors were considered as functionally redundant.

We first determined that the STP1 and STP2 genes derivate from the event of WGD. The conservation of these two genes in S. cerevisiae was accompanied by a functional divergence of their products at several levels: processing sensibility, transcriptional activation capacity, target genes, cellular abundance level and stability. The Stp2 factor with its high abundance in the cells and its higher Ssy5-processing sensibility is specialized towards induction of the AGP1 gene when the external amino acid signaling is weakly stimulated. Under strong stimulation conditions, the amino acids induce cleavage-triggered destabilization of Stp2 through the proteasomal pathway and the induction of AGP1 is mediated mainly by the Stp1 transcription factor. Unlike Stp2, the Stp1 factor is characterized by its high transcriptional activation capacity and weaker sensitivity towards Ssy5-processing. The Stp factors differ also by their genetic targets. Indeed, only Stp2 regulates the expression of DIP5. Finally, we determined that the processing sensibility and the transcriptional activation capacity of each Stp factors is directly linked to their N- and C-terminal domains, respectively.

The phosphorylation states and the degradation of the Stp2 factor were also examined. The event of degradation concerns only the processed forms of this factor and takes places principally in the nucleus. Some data indicate that such an event might be important to limit the activation capacity of this factor. The role of the Stp2 phosphorylation in the external amino acid signaling pathway is still unknown but this event might be important for the Stp2 degradation or its transcriptional activity.

The unique Stp factor from Kluyveromyces lactis (Kl-Stp), a pre-WGD species, was also studied. The Kl-Stp factor shares at least two characteristic with the S. cerevisiae Stp2 factor: high sensibility towards processing and high levels of degradation. This observation leads us to conclude to that the STP genes may have been conserved after WGD though a mechanism called neofunctionalization (one of the duplicate obtained after duplication retains the ancestral function while the other evolves to perform a novel function).

Finally, a new model for the external amino acid signaling pathway that brings together all the data obtained during this thesis’s work is proposed.

This report is a part of a larger project with the aim of developing a proof-of-concept prototype. The project was started on the initiative of Company X and is a collaboration between them and the University of Borås. Company X are a logistics consultant firm specialized in warehousing operations. Through this project Company X hope to strengthen their standing on the market and offer their customers new and innovative solutions. The purpose of this project is to, with the assistance of AI, calculate in what cluster within a warehouse a product should be placed in order to create more efficient picking routes and to research the market in order to evaluate the business potential for this kind of service. This specific report aims to conduct a market analysis for the potential service. It will do so by conducting interviews and creating a theoretical framework consisting of relevant literature. When performing the market research and analysis of the potential IT-service there are a few limitations caused by a number of factors that could potentially affect the significance of the end result. The service is not yet developed and cannot be described in detail but only as an idea, which could make it difficult for the interview respondents to fully understand the service. The report will also be limited for certain practical reasons such as the Covid-19 pandemic. Because of this, the report has not had the opportunity to conduct the interviews with potential customers on site but have been conducted online, via link. The theoretical framework has two different main focus areas. The first area addresses basic warehouse logistics and difficulties in selling high technology products, while the second area includes marketing strategies and strategic marketing planning. The market analysis will consist of a SWOT-analysis and an STP-analysis. The report has concluded that the service has potential and suggests that company type B is the best suited potential customer. Personal and demand driven sales are also considered helpful factors when selling high-tech products and solutions.
Detta arbete är en del av ett större projekt med syftet att utveckla en proof-of-concept prototyp. Projektet, som är ett samarbete mellan ”Företag X” och Högskolan i Borås, startades på initiativ av Företag X. Företaget är specialiserat på logistiklösningar och lagerverksamhet. Förhoppningen med projektet från Företag X perspektiv är att stärka sin marknadsposition och kunna erbjuda sina kunder nya och innovativa lösningar. Projektets syfte är med hjälp av AI beräkna i vilket kluster inom ett lager en produkt ska placeras för att skapa effektivare plockrundor samt att undersöka marknaden för att kunna utvärdera affärspotentialen för denna typ av tjänst. Detta arbete syftar till att genomföra en marknadsanalys för den potentiella tjänsten som underlag för forskningsprojektet. Marknadsanalysen baseras på kvalitativa intervjuer med personer utvalda genom ett experturval samt ett teoretiskt ramverk. Marknadsanalysen har vissa begränsningar som potentiellt kan påverka resultatet. Bland annat det faktum att tjänsten inte är utvecklad än och därför bara kan beskrivas som idé vilket kan bidra till svårigheter för intervjurespondenterna att förstå tjänsten fullt ut. Även Covid-19 pandemin anses vara en begränsning rent praktiskt då intervjuerna inte har kunnat ske på plats utan har fått ske digitalt istället. Det teoretiska ramverket har två huvudfokus, det ena berör lagerlogistik och svårigheter med att sälja högteknologiska produkter medan det andra behandlar marknadsföringsstrategier och strategisk marknadsplanering. Marknadsanalysen består av en SWOT-analys och en STP-analys. Arbetet har kommit fram till att tjänsten har potential och föreslår företagstyp B som den mest optimala kunden att rikta försäljning av denna typ av tjänster mot. Personlig och efterfrågestyrd försäljning anses vara viktiga aspekter av försäljning av högteknologiska produkter och tjänster.

Abstract:
Salmonella enterica serovar Typhimurium genome encodes 13 fimbrial operons. Most of the fimbriae encoded by these operons are not produced under laboratory conditions but are likely to be synthesized in vivo. We used an in vivo expression technology (IVET) strategy to identify four fimbrial operons, agf, saf, sti, and stc that are expressed in the spleen. When any three of these operons were deleted, the strain retained wild-type virulence. However, when all four operons were deleted, the resulting strain was completely attenuated, indicating that these four fimbriae play functionally redundant roles critical for virulence. In mice, oral doses of as low as 1 x 10(5) CFU of the strain with four fimbrial operons deleted provided 100% protection against challenge with 1 x 10(9) CFU of wild-type S. Typhimurium. We also examined the possible effect of these fimbriae on the ability of a Salmonella vaccine strain to deliver a guest antigen. We modified one of our established attenuated vaccine strains, chi 9088, to delete three fimbrial operons while the fourth operon was constitutively expressed. Each derivative was modified to express the Streptococcus pneumoniae antigen PspA. Strains that constitutively expressed saf or stc elicited a strong Th1 response with significantly greater levels of anti-PspA serum IgG and greater protective efficacy than strains carrying saf or stc deletions. The isogenic strain in which all four operons were deleted generated the lowest anti-PspA levels and did not protect against challenge with virulent S. pneumoniae. Our results indicate that these fimbriae play important roles, as yet not understood, in Salmonella virulence and immunogenicity. IMPORTANCE Salmonella enterica is the leading cause of bacterial food-borne infection in the United States. S. Typhimurium is capable of producing up to 13 distinct surface structures called fimbriae that presumably mediate its adherence to surfaces. The roles of most of these fimbriae in disease are unknown. Identifying fimbriae produced during infection will provide important insights into how these bacterial structures contribute to disease and potentially induce protective immunity to Salmonella infection. We identified four fimbriae that are produced during infection. Deletion of all four of these fimbriae results in a significant reduction in virulence. We explored ways in which the expression of these fimbriae may be exploited for use in recombinant Salmonella vaccine strains and found that production of Saf and Stc fimbriae are important for generating a strong immune response against a vectored antigen. This work provides new insight into the role of fimbriae in disease and their potential for improving the efficacy of Salmonella-based vaccines.

This thesis report presents a method to analyze the communication architecture for the Intelligent Transportation Systems (ITS). The report also includes a case study on ASN.1 notation and analysis of its encoding rules. Included in the report is also: (i) accompanying instruction on how to use ASN.1 compilers to produce the C/C++ message encoder/decoder, and (ii) analysis of Non-IP communications of Communication Access for Land Mobiles (CALM-FAST) protocol stack in ITS. The thesis is a part of the research project entitled “SCOOP”, a joint project between SCANIA CV AB and KTH. The purpose of this thesis is to contribute to the ultimate goal, which is to equip a vehicle with necessary hardware and software technology to provide a platooning behavior in the GCDC 2011 competition. This goal is achieved by the means of wireless communication system for both vehicle to vehicle and vehicle to road side units communications in the platoon. Overall, this thesis introduces the important usage of ASN.1 in implementation of cut-edge telecommunication systems especially in V2V and V2I communication; and clarifies the CALM-FAST protocol stack in mobile nodes.

The present work introduces a methodology to obtain a discrete time state space representation of an electrical network using the nodal [G] matrix of the Electromagnetic Transients Program (EMTP) solution. This is the first time the connection between the EMTP nodal analysis solution and a corresponding state-space formulation is presented. Compared to conventional state space solutions, the nodal EMTP solution is computationally much more efficient. Compared to the phasor solutions used in transient stability analysis, the proposed approach captures a much wider range of eigenvalues and system operating states. A fundamental advantage of extracting the system eigenvalues directly from the EMTP solution is the ability of the EMTP to follow the characteristics of nonlinearities. The system's trajectory can be accurately traced and the calculated eigenvalues and eigenvectors correctly represent the system's instantaneous dynamics. In addition, the algorithm can be used as a tool to identify network partitioning subsystems suitable for real-time hybrid power system simulator environments, including the implementation of multi-time scale solutions. The proposed technique can be implemented as an extension to any EMTP-based simulator. Within our UBC research group, it is aimed at extending the capabilities of our real-time PC-cluster Object Virtual Network Integrator (OVNI) simulator.

IP Multimedia Subsystem (IMS) is the next generation of telecommunication systems. The system is based on an IP network and uses technologies from the Internet. The IMS system is designed to evolve from a telephone system into a general information and communication system. It is meant to include television, Video on Demand (VoD), interactive services etc, etc. It is designed to simplify the implementation of newservices in telecom networks. This report investigates security aspects of VoD services when merging an IP Television (IPTV) system with IMS. The investigation covers security functions in IMS, transitionsolutions for authentication of the Set-Top-Box (STB) in IMS, and identifies problems inthe integration of IPTV and IMS. The report concludes that IMS has good solid Authentication, Authorization, and Accounting (AAA) functions that will provide security and billing functionality. One problemis found in the media control between the STB and the streaming server. This interface lacked specification at the time of investigation, and some problems have been identified. These problems have to be solved before a system can be brought into service and beregarded as secure.

The purpose of this research was to evaluate biomechanical differences in alternate stair descent strategies to determine the mechanisms by which these strategies modulate task mechanics Seventeen healthy subjects (9 males, 8 females) performed ten trials in four experimental conditions; forwards (FD), backwards (BD), step-by-step lead (SBSL) and trail (SBST) limbs. Results showed that peak ankle plantiflexor powers and work in BD and SBSL were similar in magnitude (p>0.05) and reduced relative to FD at initial contact (p<0.001) Similarly, the midstance peak plantiflexor moments, powers and work in FD and SBST were greater than both BD and SBSL (p<0.001). The plantiflexor moment and work in BD were larger than those at the SBSL limb (p<0.01), whereas power magnitudes were not significantly different (p>0.05). Lastly, peak moments, powers and work at push-off were greater in FD compared to all other conditions (p<0.001) and further, were larger in SBST relative to BD and SBSL (p<0.001). Peak moments, powers and work were similar in BD and SBSL conditions (p>0.05). Peak knee moments, powers and work were reduced during the eccentric midstance burst. Knee moments, powers and work were similar in FD and SBST (p>0.05), but were greater than BD and SBSL (p<0.001). Peak moments, powers and work were greater in BD compared to the SBSL limb (p<0.001). Thus, the step-by-step descent strategy is more appropriate for individuals with unilateral knee problems, whereas backwards descent is more suitable for bilateral quadriceps weakness. Peak hip powers and work were larger in BD relative to FD (p <0.001) during initial contact. Similarly, the extensor moment in BD was significantly larger than the flexor moment demonstrated in FD. No consistent stair or condition effects were observed for the concentric hip flexor burst at push-off in FD, SBSL and SBST conditions. Finally, this study considered foot clearance in stair descent. Clearance was greater in BD relative to FD, SBSL and SBST in the mid staircase region, but was similar at step 1, which represented the transition from standing to stair descent. No foot contacts, trips or stumbles were recorded, demonstrating the likelihood of contacting a stair edge in all patterns was nonexistent for these healthy participants. The present results have implications to improve functionality in stair descent for individuals that find the task difficult or painful. Future studies will likely consider the strategies employed during the initiation of stair descent by elderly and clinical populations to gain insight on the strategies used to minimize mechanical burden on both the joints and overlying musculature. Lastly, research should consider the effects of handrail use on alternate stair descent strategy mechanics, such as backwards and step-by-step descent.

There are several weaknesses in the popular transport protocol TCP (Transmission Control Protocol). A possible replacement to TCP would be the newer protocol SCTP (Steam Control Transmission Protocol). This thesis presents three different proposed implementations of SCTP: HTTP over SCTP, online games over SCTP and IP mobility over SCTP. The proposed implementations are analyzed, based on relevant literature, and recommendations are issued on the importance of moving forward with them. The result of the thesis is that HTTP over SCTP is recommended. SCTP features such as multi-streaming, multi-homing and the four-way handshake addresses the inherent weaknesses with using TCP for HTTP traffic. IP mobility over SCTP is also recommended since it results in lower delay in the handover process compared to MIPv6 (Mobile IPv6). Online games over SCTP, however, is not recommended since the existing implementations of SCTP results in poor latency for the kind of traffic online games produce.

This research examines information flow in online, interest-based networks to determine if existing models of information dissemination are adequate to describe the communication processes that occur in online publics. This study finds that a small number of primary influencers from within online communities are central to information collection, collation, and distribution in online, interest-based networks. This finding is inconsistent with one-step, two-step, and multi-step flow models, which privilege mass media as the central source of information. To more accurately depict online information flow in interest-based networks, this study introduces the radial model of information flow. Furthermore, the results of this study show that communication processes in online publics are best explained using a combination of the transmissive paradigm of communication, on which information flow models are based, and a ritual view of communication. This research also contributes to the ongoing development of the situational theory of publics by identifying organized publics as a key subgroup of active publics. Organized publics are networks of individuals within active publics who frequently and consistently communicate on a shared interest or concern. Organized publics form active online communication networks and prepare for advocacy related to a shared interest, making them of particular interest to public relations professionals. Using a case study approach, this dissertation uses online network analysis and qualitative cluster analysis to study the role of community influencers in information flow and cultural development within the online young adult cancer community. Instead of focusing exclusively on social media as channel for message dissemination, the results of this study indicate that successful relationship building can best by achieved by public relations practitioners who work to develop authentic presences in online communities. This research shows that embracing a participatory model of public relations that actively engages primary influencers in the planning and campaign implementation processes can promote authentic online presences.

Abstract:

One of the central motivators behind genetic research is to understand how genetic variation relates to human health and disease. Recently, there has been a large-scale effort to find common genetic variants associated with many forms of disease and disorder using single nucleotide polymorphisms (SNPs). Several genome-wide association (GWAS) studies have successfully identified SNPs associated with phenotypes. However, the effect sizes attributed to individual variants is generally small, explaining only a very small amount of the genetic risk and heritability expected based on the estimates of family and twin studies. Several explanations exist for the inability of GWAS to find the "missing heritability."

The results of recent research appear to confirm the prediction made by population genetics theory that most complex phenotypes are highly polygenic, occasionally influenced by a few alleles of relatively large effect, and usually by several of small effect. Studies have also confirmed that common variants are only part of what contributes to the total genetic variance for most traits, indicating rare-variants may play a significant role.

This research addresses some of the most glaring weaknesses of the traditional GWAS approach through the application of methods of polygenic analysis. We apply several methods, including those that investigate the net effects of large sets of SNPs, more sophisticated approaches informed by biology rather than the purely statistical approach of GWAS, as well as methods that infer the effects of recessive rare variants.

Our results indicate that traditional GWAS is well complemented and improved upon by methods of polygenic analysis. We demonstrate that polygenic approaches can be used to significantly predict individual risk for disease, provide an unbiased estimate of a substantial proportion of the heritability for multiple phenotypes, identify sets of genes grouped into biological pathways that are enriched for associations, and finally, detect the significant influence of recessive rare variants.

Abstract:
The yeast Saccharomyces cerevisiae utilises many transport pathways for the efficient an accurate segregation of proteins to the compartments of the cell in which they function. Of these, the secretory pathway is responsible for the localisation of proteins to the endoplasmic reticulum (ER), Golgi apparatus, endocytic and vacuolar compartments and the cell surface. The first step along this pathway is the targeting of nascent polypeptides to the ER membrane. For many proteins, the signal recognition particle (SRP), a cytosolic ribonucleoprotein, and its cognate receptor (SR) on the ER membrane are responsible for this step. SRP and the SR direct proteins to the translocon, which forms an aqueous channel through which the nascent proteins are translocated or from which they are then integrated into the ER membrane. The aim of this study was to dissect incompletely understood interactions that occur at the ER membrane between SRP, SR and the translocation and to reconstitute the SRP-dependent translocation reaction with purified proteins. In particular, through 2-hybrid analysis and pull-down assays a novel interaction was identified between SR and the major translocon component Sec61p. To facilitate the study of this and other interactions, attempts were made to reconstitute the SRP-dependent targeting pathway with SR and translocon purified from yeast. It was demonstrated that SRP-dependent translocation could be reconstituted with solubilised yeast ER membrane proteins. SR was purified and shown to be functional, and the translocation reaction was shown to be stimulated by the presence of the ER-lumenal chaperone Kar2p. Preliminary experiments were also carried out that suggested that the purified translocon was active; indicating that the goal of reconstituting SRP-dependent translocation with purified components is attainable.

There is a strong need for powerful unified statistical methods for discovering underlying genetic architecture of complex traits with the assistance of omics information. In this paper, two methods aiming to detect novel association between the human genome and complex traits using intermediate omics data are developed based on statistical mediation modeling. We demonstrate theoretically that given proper mediators, the proposed statistical mediation models have better power than genome-wide association studies (GWAS) to detect associations missed in standard GWAS that ignore the mediators. For each ofthe modeling methods in this paper, an empirical example is given, where the association between a SNP and BMI missed by standard GWAS can be discovered by mediation analysis.

A new age has begun for telecommunication industry. Over the past decade wireless industry has grown at a remarkable pace, consequently level of technology development goes beyond the level of customer desire. Application flexibility and being highly dynamic will be the main features of beyond 3G services of interest to users. Being there all these emerging technologies in one cellular network has opened the work of designing and optimization of the networks to be viewed from a different perspective. APSEN analysis for Beyond 3G wireless networks has been discussed. “AP” stands for application layer, analysis of services and applications. “SE” stands for session layer, analysis of session management protocol and “N” for network layer, analysis for network protocols. The main purpose of our research is to focus on the challenges offered at APSEN for beyond 3G wireless network. There are a lot of research challenges in each of the different layers but focus will be on APSEN. The objective is to take a look at and familiarize with some of the major challenges offered at APSEN for beyond 3G wireless network. Qualitative approach has been used which will help us to collect wide range of information and enhance our knowledge by analyzing the perspectives of different researchers.

Internet commerce continues to grow rapidly. Over 60% of US households use the internet to shop online. A secure payment protocol is required to support this rapid growth. A new payment protocol was recently invented at IBM. We refer to the protocol as SPP or Secure Payment Protocol. This thesis presents a protocol analysis of SPP. It is essential that a thorough security analysis be done on any new payment protocol so that we can better understand its security properties. We first develop a method for analyzing payment protocols. This method includes a list of desirable security features and a list of proofs that should be satisfied. We then present the results of the analysis. These results validate that the protocol does contain many security features and properties. They also help understand the security properties and identify areas where the protocol can be further secured. This led us to extend the design of the protocol to enhance its security. This thesis also presents a prototype implementation of SPP. Three software components were implemented. They are the Electronic Wallet component, the merchant software component and the Trusted Third Party component. The architecture and technologies that are required for implementation are discussed. The prototype is then used in performance measurement experiments. Results on system performance as a function of key size are presented. Finally, this thesis presents an extension of SPP to support a two buyer scenario. In this scenario one buyer makes an order while another buyer makes the payment. This scenario enables additional commerce services.

Abstract:
Data from a Croatian isolate population are analysed in a genome-wide association study (GWAS) for a variety of disease-related quantitative traits. A novel genomewide approach to analysing pedigree-based association data called GRAMMAR is utilised. One of the significant findings, for uric acid, is followed up in greater detail, and is replicated in another isolate population, from Orkney. The associated SNPs are located in the SLC2A9 gene, coding for a known glucose transporter, which leads to identification of SLC2A9 as a urate transporter too (Vitart et al., 2008). These SNPs are later implicated in affecting gout, a disease known to be linked with high serum uric acid levels, in an independent study (Dehghan et al., 2008). Subsequently, investigation into different ways in which to use SNP data to identify quantitative trait loci (QTL) for genome-wide association (GWA) studies is performed. Several multi-marker approaches are compared to single SNP analysis using simulated phenotypes and real genotype data, and results show that for rare variants haplotype analysis is the most effective method of detection. Finally, the multi-marker methods are compared with single SNP analysis on the real uric acid data. Interpretation of real data results was complicated due to low sample size, since only founder and unrelated individuals may be used for population-based haplotype analysis, nonetheless, results of the prior analyses of simulated data indicate that multi-marker methods, in particular haplotypes, may greatly facilitate detection of QTL with low minor allele frequency in GWA studies.

IP Telephony is a technology which uses internet and signalling protocols like H.323 and SIP to setup and transfer voice signals from one destination to another. These protocols use various encoding schemes to transmit voice signals over the digital technology. The success of IP Telephony is the cost and time effectiveness; hence we studied on the call setup delay of these two signaling protocols H.323 and SIP. H.323 uses TCP and SIP uses UDP as transport protocols to set up a call. They have different scopes that distinguish them. The signalling protocols H.323 and SIP call setup delays are observed and studied successfully between the traditional telephones and IP telephones. The call setup delay for H.323 is analysed experimentally in the Cisco environment by establishing calls at various loads such as null load, medium load and heavy load. Due to practical difficulties the SIP call setup delay is studied only literally. The parameters that we considered for our experiments are the average call setup delay, bandwidth and different traffic loads. Finally, the results of both H.323 and SIP are compared with each other to find out the best signalling protocol. The comparison is done according to the call setup delay and other factors like complexity, compatibility, reliability and bandwidth utilization.

Taiwan is located in a highly seismic zone and the historical “Dieh-Dou” timber buildings, constructed without following any code or standard, are prone to collapse under earthquake. These buildings are unique and represent the culture, heritage and art of Taiwan, therefore need to be preserved while minimizing unnecessary intervention that could damage their authenticity.
This research comprises a thorough investigation on the parameters influencing the seismic vulnerability of the Dieh-Dou timber frames in Taiwan, and propose a methodology of assessment and a strategy for strengthening validated through experimental testing and numerical analysis.
After review existing literature and post-earthquake surveys, the failure modes of the buildings are identified, showing that the dislocation of the elements of the frame from the joints is the primary source of damage. An experimental investigation is carried out comprising both rotational and translational tests on full scale joint specimens which, together with a parametric study undertaken with an appropriate FE simulation, demonstrates how both the rotational and translational stiffness of the joints play a key role in defining the behaviour of these structures.
Lateral force, response spectrum, and step-by-step pushover analyses are performed and compared with the post-earthquake survey of two Dieh-Dou buildings seriously affected by the 1999 Chi-Chi earthquake. The results show that the proposed FE modelling can successfully be employed to assess the vulnerability of the frames.
Based on a damage level approach, an assessment methodology is suggested that would allow to optimisation of the strengthening strategy, permitting protection these precious structures from future earthquakes while avoiding unnecessary interventions.

The purpose of this study was to examine a sample of students who participated in a student assistance program in Southwest Virginia. Using existing data from a school system in Southwest Virginia, this sample was observed to measure changes in student academic performance. The sample was also examined to determine the extent to which they instituted positive behaviors such as school attendance and reduction of disciplinary actions taken. In addition, this study assessed differences in demographic characteristics among student participants. Also studied was the extent to which gender differences related to academic performance and behavior. Significant changes in students’ academic performance, attendance and disciplinary measures were established in the present study. Negative associations were established for those participating in the student assistance program and grade point average. Positive associations were found for those participating in SAP and attendance and disciplinary measures. No significant differences were yielded in the comparison of gender to academic performance and behavior. SAP coordinators recorded that the majority of participants did improve since referral to program and most completed or currently remained in the student assistance program. However, a small percentage of students actually entered treatment programs following recommendations made to parents from the student assistance program committee. This research assessed only the demographic and individual characteristics: gender, gifted or special education status, ethnicity and age. Therefore, other demographics such as socio economic status may offer additional explanation into academic and behavior outcomes of students involved in student assistance programs.

[Abstract]Market segmentation has been identified in the tourism marketing literature as an effective tool that can be used by management to meet the needs of a market more efficiently and effectively. It has been widely used by researchers seeking to develop tourism segments. Historically, most segmentation studies have developed tourism profiles at a destination using 1) a visitor questionnaire survey developed from studies in the literature or 2) secondary data. Very little research has taken a stakeholder approach to destination segmentation, despite many authors arguing the importance of utilising a stakeholder approach for destination management and marketing purposes. This research proposes a two-step approach to destination segmentation. It details this approach using one Australian destination. The two-step approach to segmentation begins with firstly understanding how tourism stakeholders at a destination segment their market before surveying tourists for the purposes of identifying segments. In contrast to previous tourist-focused segmentation studies, the two-step approach recommended and detailed in this study considers both tourism stakeholders and tourists. The segments devised from the proposed approach are then compared and contrasted with segments currently utilised by the destination marketing organisation (DMO). Step one involved interviewing 14 tourism stakeholders to determine how they segment the market. Based on these findings, a questionnaire survey was developed and data was collected from 852 tourists. This represented step two of the approach. Three tourism segments were identified through cluster analysis. Only one of these three segments was comparable with the segments defined by the DMO. The other two were not considered in the DMO segmentation. These segments represented over half of all tourists in the sample. Based on the sample in this study, the DMO segments target less than a quarter of the types of tourists visiting the destination. Contributions to theory and practice were identified. Firstly, it was recognised that different tourism stakeholders attract different tourists, not all stakeholders segment their market, and some stakeholders do not adhere to segments targeted by the DMO which may lead to an inconsistent message in the market. Secondly, the two-step approach is a new method incorporating a stakeholder view, which gives a more holistic view and a richer description for the segments obtained when compared with academic and practitioner segmentation approaches. The two-step approach can be utilised at other tourist destinations. The two-step approach to segmentation is capable of assisting tourism marketers to target more of the tourists frequenting the destination. This study suggests that many dollars may be wasted targeting tourists that are not likely to travel to the destination and not targeting those who would. Future research should be conducted at alternative destinations to further the understanding of the recommended two-step approach to segmentation.

Les strongles gastro-intestinaux, dont Haemonchus contortus constituent un problème majeur pour l'élevage des ovins allaitants. Ils entrainent des pertes de production et le recours aux anthelminthiques est remis en question par l'apparition de souches de vers résistantes. La sélection d'ovins plus résistants fait partie des stratégies complémentaires de lutte les plus sérieuses. Cependant sa mise en oeuvre requiert une meilleure compréhension des mécanismes sous-jacents. Cette thèse vise à identifier les régions du génome ovin impliquées dans la résistance aux strongles gastro-intestinaux. Une analyse statistique d'association entre des marqueurs génétiques et des mesures de résistance d'un troupeau d'ovins croisés Martinik Black-belly x Romane a mis en évidence un nombre limité de régions d'intérêt. Parmi celles-ci, un segment du chromosome 12 a été choisi pour effectuer des accouplements raisonnés et valider son rôle dans la résistance à H. contortus. L'effet de cette région a été validé chez les descendants issus d'accouplements assistés par marqueurs génétiques. Cette région semble limiter fertilité des vers femelles tout en contribuant à une réponse immunitaire plus forte. Le rôle d'une région du chromosome 21 dans la variation de concentration plasmatique en pepsinogène, un marqueur de lésions abomasales, a également été confirmé. Un gène candidat sous-jacent est en cours de séquençage et l'analyse des polymorphismes devrait contribuer à la validation de son rôle. Deux autres gènes très proches pourraient également être impliqués et mériteraient une considération future. Ces travaux illustrent à la fois la variation génétique disponible pour les caractères de résistance à H. contortus et la complexité des mécanismes mis en jeu. Des études complémentaires de séquençage et d'étude d'expression par séquençage devrait contribuer à une meilleure compréhension des fonctions des gènes impliqués et de leurs interactions
Gastro-intestinal nematodes, among which Haemonchus contortus are a major threat to the meat sheep industry. They are responsible for production losses and the apparition of worm populations resistant to drugs limits their use as worm control strategy. Breeding more resistant sheep is among the most practicable alternative strategy. However its implementation requires a deeper understanding of underlying mechanisms. This PhD aims at identifying regions of the ovine genome affecting resistance to gastro-intestinal nematodes. A statistical analysis of existing associations between genetic markers and resistance traits of a Martinik Black-belly x Romane cross-bred sheep flock unraveled a limited number of key players. Among these, a fragment of the chromosome 12 was chosen to perform marker-assisted matings and to validate its role in resistance to H. contortus. The effect of this region was validated in the progenies born from matings. It seems this chromosomic fragment limits female worms fertility and is associated to a stronger immune response. The putative role played by a fragment of the chromosome 21 in plasmatic pepsinogen concentration (a biomarker of abomasal lesions) was also confirmed in this work. A candidate gene underlying this region has been sequenced and the analysis of the detected polymorphisms should confirm its role. Further, two other genes in its vicinity could also play a role in this biological phenomenon and they should also deserve future considerations. This work illustrated both the existing genetic variation for resistance to H. contortus and the associated complexity of underlying mechanisms. Additional sequencing and gene expression sequencing studies should help understanding gene functions and interactions

The advancements in Whole Genome Sequencing (WGS) have increased the amount of genomic information available for epidemiological analyses. WGS opens many avenues for investigation into the tracking of pathogens, but the rapid advancements in WGS could soon lead to a situation where traditional analytical techniques might become computationally impractical. For example, the traditional method to determine the origin of an isolate is to use phylogenetic analyses. However, phylogenetic analyses become computationally prohibitive with larger datasets and are best for retrospective epidemiology. Therefore, I investigated if there might be less computationally demanding methods of analysing the same data to obtain similar conclusions. This thesis describes a proof-of-principle method for evaluating if such alternative analysis techniques might be viable. In this thesis Methicillin resistant Staphylococcus aureus (MRSA) was used, and single nucleotide polymorphism (SNP) and insertion/deletion (indel) genomic variation. I move away from whole genome analysis techniques, such as phylogenetic analysis, and instead focus on individual SNPs. I showed that genetic signals (such as SNPs and indels) can be utilised in novel ways to rapidly produce a summary of the possible geographic origin of an isolate with a minimal demand on computational power. The methods described could be added to the suite of analytical epidemiological tools and are a promising indication of the viability of developing cheap, rapid diagnostic tools to be implemented in healthcare institutions. Furthermore, the principles behind the development of the methods described in this thesis could have much wider applications than just MRSA. This implies that further work based on the principles described in this thesis on alternative pathogens could prove to be promising avenues of investigation.

Structured data has been a major component of web resources since the very beginning of the web. Metadata that was originally mostly meant for display purposes gradually expanded to incorporate the semantic content of a page. Until now semantic data on the web has mostly focused on factual knowledge, namely trying to capture “what humans know”. This thesis instead focuses on procedural knowledge, or in other words, “how humans do things”, and in particular on step-by-step instructions. I will present a semantic framework to capture the meaning of sets of instructions with respect to their potential execution. This framework is based on a logical model which I evaluated in terms of its expressiveness and it compatibility with existing languages. I will show how this type of procedural knowledge can be automatically acquired from human-generated instructions on the web, while at the same time bridging the semantic gap, from unstructured to structured, by mapping these resources into a formal process description language. I will demonstrate how procedural and factual data on the web can be integrated automatically using Linked Data, and how this integration results in an overall richer semantic representation. To validate these claims I have conducted large scale knowledge acquisition and integration experiments on two prominent instructional websites and evaluated the results against a human benchmark. Finally, I will demonstrate how existing web technologies allow for this data to seamlessly enrich existing web resources and to be used on the web without the need for centralisation. I have explored the potential uses of formalised instructions by the implementation and testing of concrete prototypes which enable human users to explore know-how and collaborate with machines in novel ways.

Abstract:
This thesis is a report  of a master thesis project work. The project was performed at Volvo IT, Gothenburg, from March to September of 2013. This study targets SAP ERP applications.  SAP AG is one of the biggest enterprise resource planning software corporations  and the ERP system is one of the most famous products of its. As one of world's leading manufactures in the field of trucks, buses, construction requirement and marine and industries engines, Volvo Group has more than 50,000 users are working with SAP applications.  The purpose of this project is to evaluate the current SAP application  in Volvo and propose several strategies for a new interface. This usability study is consisted of observation, interview and questionnaire. The result is based on a total number of eight interviews and twelve questionnaires. Proposals are given in the end for five aspects. They are a combination of ideas of my own and the new technologies published by SAP AG.

Abstract:
This thesis investigates the use of the step frequency waveform, its design and analysis using the ambiguity function. The step frequency waveform consists of a series of N pulses each with a pulse width of z, and whose frequency is increased from pulse to pulse in steps of delta f. A design procedure for detection of small targets with a surface (land or sea) based step frequency radar employing a high pulse repetition frequency (PRF) waveform is developed. The proposed method determines the waveform parameters of given radar specifications. A simple graphical implementation as well as a computer implementation are presented. The theoretical dimensions of the step frequency waveform are defined and verified for some waveforms with parameters similar to the waveform of interest. Finally, the ambiguity function is used to analyze the step frequency waveform.

Abstract:
The work described in this thesis is concerned with the development of laser desorption laser photoionisation mass spectrometry (L2MS) towards spatially resolved analysis of real complex molecular systems. A broad overview of the main elements of the technique is presented. In addition, the experimental procedures and equipment used to carry out this work are described in some detail. Photoionisation mass spectra recorded for a series of azo dyes and porphyrin pigments revealed a marked wavelength dependence in their ionisation and fragmentation channels. The relationship between this behaviour and the known photochemical and photophysical properties of these molecules is discussed. The photochemistry of these molecules has been exploited to aid the differentiation between isomeric species. The selectivity inherent in the L2MS technique has been exploited for in situ studies of a number of real systems. Polymer additives, such as antioxidants and ultraviolet stabilisers, have been successfully detected directly from their host polymer matrices without recourse to extraction, separation or pre-concentration. The technique has been shown to be surface specific, suggesting that the long-term goal of spatially resolved analysis to monitor, for example, additive aggregation and migration to the surface are feasible. In further in situ studies, polycyclic aromatic hydrocarbons, an important class of priority pollutants, adsorbed onto aerosol particulates have been detected. Electrochemically polymerised indoles, known to form conducting films, have also been identified directly from the electrode surface. Finally, current limitations of the L2MS technique are discussed. It is suggested that many of the problems identified are inextricably linked to the laser desorption process. It is shown that energy imparted to neutral molecules during the desorption event can lead to fragmentation. This has implications for both the ionisation of high mass molecules and for quantitative studies. Possible ways of circumventing these problems are discussed. The future outlook for the technique, both for fundamental studies, and as an analytical tool, is also discussed.

Because of “Cape No.7”, “Black and White” and “Monga”, marketing the city by movies and TV drama is more popular in Taiwan. There are many successful example of city-marketing by movies and TV dramas, especially Seoul and Hangzhou. The purposes of the research are understanding the theory of city-marketing, discovering the successful experiences of Seoul and Hangzhou and analyzing the city-marketing strategy for Taipei by movies and TV dramas. This is a qualitative research. We discover the city-marketing experiences of Seoul and Hangzhou, and analyze the critical success factors. In order to understand the experiences and opinions about the city-marketing by movies and TV dramas for Taipei, we contact the government-related organizations, industry workers and the opinion leaders by in-depth interview method. When we observe and analyze the cases of Seoul and Hangzhou, we can discover the successful city-marketing program is a set of co-ordination of plans and policies. Although every city has different strengths and features, we can generalize about the successful factors: government role, the environment of the movies and TV dramas industry and “market “concept. From clear policy directions and objectives, the formulation of relevant policies and regulations to providing administrative assistance and subsidies, the role of government affect the content and effectiveness of the city-marketing policies. Improving and completing the environment of the audio-visual industry can not only create cluster effect to strengthen the competitive advantages, but also promote the development of related industries. The most important factor is “market ”concept. In the planning stage, “market” concept make the movies and TV dramas touch the audiences’ heart, create the economic benefits and enhance the city image. In Taipei audio-visual city-marketing strategy, we use geographic location, features and advantages of Taipei City's and the limited resources, as the basis of market segmentation. we choose Japan, mainland China and Hong Kong and Macau, Southeast Asia and other regions in Asia and Europe and America as the target markets, and attract the backpackers who love depth and multi-cultural travel because of the characteristics and advantages of Taipei .in the end, set the appropriate city-marketing strategy to match the needs of different customers, and build Taipei into a “Chinese Cultural Center of Asia”, “the depth of cultural backpacking” and “the window to Asian Chinese Culture”.

Beverage market in Taiwan has been growing every year and because of the scorching weather, you can see that almost everyone holds a handmade drink, walking on the street. Accordingly, living habits of Taiwanese people and the weather bear a strong correlation to the high concentration of beverage market. Drinking handmade beverages has been just as necessary as eating. What's more, people are in the habit of drinking beverages with meals and almost everyone holds one beverage when shopping. By surviving a series of food safety scandals, “Milkshop” has caught customers' attention so that it has to upgrade its service quality. Also, what raw materials are used is highly valued. This research is based on the document analysis and interviews so as to further understand the origin of Milkshop on the market. With the combination of entrepreneurship and STP analysis, Milkshop has created more diverse value. From the perspective of opportunity recognition, Milkshop has its own brand of farm fresh milk and realizes the importance of health requirements on the market. This research further explores the startup process of Milkshop .

Wireless Sensor Network (WSN) is a network consisting of a number of sensor nodes (SN). Due to the restraint of power consumption and communication range, sensor nodes communicate with each other using the multi-hop method. When the distance between some SNs is larger than their communication range, the WSN cannot be connected. One approach to resolve this problem is to place relay nodes (RN) which have better transmission power, but more expensive, than general SNs. This thesis focuses on the minimization of the relay node placement problem. This problem is called STP-MSP (the Steinerized Tree Problem with Minimum number of Steiner Points), which is defined as follows. Given a set of SNs, X={p1,p2,…,pn}, in the Euclidean plane R2, and a positive constant R, representing SNs’ communication range, STP-MSP asks for a Steiner tree T that connects all nodes in X such that each edge in T has length less than or equal to R, and the number of Steiner points is minimized. In [1], it is shown showed that the STP-MSP problem is NP-hard. In [2][3], authors presented an approximate algorithm with approximation ratio 3. In this paper, we proved some analyses and improvements on the 3-approximation algorithm. First, we analyzed the worst cases of the algorithm, which could lead a tighter bound of the approximation ratio. Second, we used the Delaunay triangulation to improve the performance of the algorithm, by which the time complexity is reduced from O(n3) to O(n log n).

Besides the traditional cathode ray tube (CRT) display , there are several display products in the FPD market, such as liquid crystal display (LCD) , Plasma Display Panels (PDP), organic light-emitter diode (OLED) and so on. Nowadays, because of the innovation of nanotechnnology, which drove the development of carbon nanotubes field effective display (CNT-FED). This new products driven by technology will confront two questions as follows : (1) The can’t understand the real scale of market. (2) They can’t forecast the time they need to spend to realize the market chance. Therefore , how to modify the new-products-entering model is an worthy problem to research . The research was done by using the ways of interviewing experts and secondary data collection. Then, we used scenario analysis to build the most possible scenario that CNT-FED may encounter in the future. Under the scenario, we used the approachs of STP strategy to develop th model that CNT-FED enters the market in the future.From the conclusion of this research, we know that CNT-FED should choose the market segment of big panel at the first time entering the market, and it will complete with PDP. Because the performance of CNT-FED is much better than PDP , we need to let the customers realizing and identifying this difference. Therefore, we can reduce the weakness of CNT-FED because of its high price , and , then, we can build the niche market of CNT-FED.

Recently, private supplementary tutoring industry has experienced extraordinary growth and emerged as a profitable business in Macau. The present thesis, on the one hand, strives to explore consumers’ level of satisfaction on the existing PST service in Macau and, on the other hand, tries to discover customer segments, targets, as well as develop a positioning strategy for a business idea, named XXX Multifarious Tutoring Centre. In the satisfaction analyses, two models were proposed to identify the factors that influence the satisfaction level of both parents and students using ordered logit. The regressions suggest that apart from traditional values related to academic performance, students value the opportunities of interacting with peers in the tutoring centre. At the same time, parents think safety as an essential factor. Regarding segmentation, latent class analyses based on attitude and behavioural variables were used to identify customers who are most willing to join the Centre. These customers were then considered as target customers, and a preliminary positioning strategy was made accordingly.
Nos últimos anos, a indústria de apoio pedagógico complementar de Macau teve um crescimento excecional e emergiu como um negócio lucrativo. A tese presente visa, por um lado, explorar o nível de satisfação dos consumidores com os serviços oferecidos pelos Centros de Explicações existentes em Macau e, por outro lado, descobrir segmentos de clientes, clientes-alvo, bem como desenvolver uma estratégia de posicionamento para um projeto empresarial, que tem o nome XXX Multifarious Tutoring Centre. Nas análises de satisfação, dois modelos foram propostos, utilizando Ordered logit, para identificar os elementos que afetem o nível de satisfação dos pais e dos alunos. Os resultados das regressões sugerem que, além dos valores tradicionais relacionados ao desempenho académico, os alunos valorizam também as oportunidades de interação com os pares nos centros de explicações. Ao mesmo tempo, a segurança é um fator importante para os pais. Em termos da segmentação, as análises de classe latente com base nos variáveis de atitude e comportamentais foram utilizadas para identificar os clientes com mais vontade de utilizar o serviço do XXX Multifarious Centre. Os respetivos clientes foram então designados como clientes-alvo do Centro e uma estratégia de posicionamento preliminar foi desenvolvido.

Abstract:
Viele Publikationen haben bereits den Einfluss verschiedener Zytokingenvariationen auf die Ensteheung einer Graft versus Host Disease untersucht. Ich habe den Einfluss verschiedener Zytokin- und Zytokinrezeptorgenvariationen des Interleukin 10-Gens auf die Ausprägung von GvHD und die Überlebensrate nach allogener Blutstammzelltranplantation bei Patienten mit einer hämatologischen Neoplasie untersucht. Die GvHD zählt zu einer gefürchteten Komplikation bei Patienten nach einer allogenen Stammzelltransplantation. Sie kann als akute Form innerhalb von 100 Tagen nach Transplantation, sowie als chronische Form im späteren Verlauf auftreten. Sowohl die akute wie auch die chronische GvHD können bis zum Tode des Patienten führen oder durch Organschäden und die langandauernde intensive Immunsuppression die Lebensqualität der Patienten sehr beeinträchtigen. Es fehlt bis heute das genaue Verständnis, warum einige Patienten eine schwerwiegende GvHD entwickeln, während andere Patienten von einer nur leichtgradig auftretenden GvHD auf Grund eines damit assoziierten GvL-Effektes sogar profitieren. Das Interleukin 10 spielt neben anderen Zytokinen aufgrund seiner antiinflammatorischen Funktion eine wichtige Rolle in der Pathophysiologie der GvHD. Durch IL 10 kommt es zu einer Herunterregulation von MHC Klasse 2 und Kostimulatorischen Molekülen auf der Zelloberfläche, womit die Fähigkeit antigenpräsentierender Zellen zur Antigenpräsentation supprimiert wird. Interleukin 10 bewirkt außerdem eine Hemmung der Produktion proinflammatorischer Faktoren wie z.B. Interferon a, oder TNF-a, sowie eine Suppression der Transkription von NF- kB. Für diese Analyse wurden die GvHD-relevanten klinischen Daten von 360 konsekutiven Patienten ausgewertet, die zwischen 2001 und 2011 in der Universitätsmedizin Göttingen eine allogene Transplantation erhielten und eine Einteilung in die Schweregrade der akuten und der chronischen GvHD vorgenommen. Es wurde eine Genotypisierung ausgewählter Single Nucleotide Polymorphisms des IL 10–Gens und des IL 10–Rezeptorgens mittels SNaPshot-Analyse und TaqMan® Genotyping Assays vorgenommen. Durch univariate, gefolgt von multivariaten logistischen Regressionsanalysen der Daten mit Bezug zum klinischen Verlauf wurden Genvarianten mit Einfluss auf das Auftreten und den Schweregrad der GvHD identifiziert. Für die Überlebenszeitanalysen wurde ein simpler Logrank–Test verwendet, um Variablen mit einem p-Wert < 0,20 herauszufiltern, mit denen anschließend eine Cox–Regressionsanalyse durchgeführt wurde. In Voruntersuchungen (v.a. Hansen et al) war der SNP IL-10 -597 Genotyp A (= IL-10 -592) im Patienten, in Kombination mit dem G-Allel im SNP IL-10Rβ K47E (=IL-10Rβ c238) im Spender, bereits als wichtiger protektiver Faktor gegen eine schwere aGvHD in Matched-related-Donor (MRD)-transplantierten Patienten beschrieben worden. Der protektive Effekt dieser Kombination war in unserem Datensatz über alle Patienten hinweg nur als Tendenz erkennbar, während sich der alleinige protektive Effekt des R K47E G-Allel im Spender bei Patienten mit malignen Lymphomen als signifikant erwies. Dieser Unterschied in unseren Analysen, kann an der inhomogenen Zusammenzusetzung des Kollektivs aus MRD- und MUD- transplantierten Patienten liegen. Der protektive Effekt der oben genannten Kombination wurde bisher nur in reinen MRD-Kollektiven nachgewiesen und war in reinen MUD-Kollektiven nicht vorhanden. Weiterhin erwies sich für die chronische GvHD der proximale SNP IL-10 -1087 Genotyp GG des Patienten als assoziiert mit einem erhöhten Auftreten von chronischer GvHD. Dieser Genotyp ist mit einer erhöhten IL 10 Produktion assoziiert. Laut Ergebnissen von Martin und Zhou et al., ist ein erhöhter IL 10–Spiegel mit einer cGvHD assoziiert. Unter den in dieser Untersuchung erstmalig analysierten Polymorphismen des IL 10-Rezeptorgens wurde ein signifikant erhöhtes Risiko für das Auftreten einer schweren akuten GvHD (Grad III und IV) für Patienten festgestellt, die mit einem Spender des Genotyps IL 10 Rα G351R AA transplantiert worden waren. Dieses Risiko war weiter erhöht für die Patienten festzustellen, die zur T-Zell-Depletion im Rahmen der Konditionierung ATG erhalten hatten. Der Einsatz des als prophylaktisch gegen das Auftreten einer schweren GvHD genutzen ATGs war bei dieser Patientengruppe also mit einer zusätzlichen Risikoerhöhung für das Auftreten einer schweren GvHD assoziiert. Der IL 10 Rezeptor besteht aus einer - und einer -Einheit. Analysen der -Einheit des IL 10-Rezeptors beziehen sich größtenteils auf den Rα G159S. Der Rα G351R wurde bisher nicht funktionell oder in genetischen Assoziationsstudien untersucht, weshalb die funktionelle Bedeutung dieses Polymorphismus bisher nicht bekannt ist. Eine funktionelle Rolle dieses Polymorphismus erscheint aber prinzipiell möglich, da er im codierenden Bereich des Gens liegt und mit einem Austausch der Aminosäure Glycin gegen Arginin verbunden ist. Die Bedeutung der in dieser Arbeit erstmalig untersuchten SNPs, insbesondere des Polymorphismus IL 10 Rα G351R AA, sollte in unabhängigen Patientenkollektiven hinsichtlich seiner klinischen Relevanz weiter überprüft werden, auch um die Assoziation mit einer höheren Inzidenz an schwerer akuter GvHD bei Patienten, die ATG erhielten, detaillierter zu untersuchen. Um den Effekt besser zu verstehen, sollte diese Genvariante auch in ihrer Funktionalität in vitro untersucht werden. Neben dem Interleukin 10 sind noch viele weitere Zytokine an der Entstehung bzw. der Verhinderung einer GvHD beteiligt und es ist wichtig, hier mit der Genotypisierung von Kandidatengenen fortzufahren, um weitere funktionell bedeutsame Genvarianten zu identifizieren. Im Erfolgsfalle wird durch die Identifizierung dieser Varianten die Erstellung eines genetischen Profils der Patienten vor der geplanten Stammzelltransplantation möglich. Mit Hilfe dieses Profils kann eine bessere Einschätzung des individuellen Risikos getroffen und gegebenenfalls die Medikation frühzeitig dementsprechend angepasst werden.

Abstract:
The present work introduces a methodology to obtain a discrete time state space representation of an electrical network using the nodal [ G ] matrix of the Electromagnetic Transients Program (EMTP) solution. This is the first time the connection between the EMTP nodal analysis solution and a corresponding state-space formulation is presented. Compared to conventional state space solutions, the nodal EMTP solution is computationally much more efficient. Compared to the phasor solutions used in transient stability analysis, the proposed approach captures a much wider range of eigenvalues and system operating states. A fundamental advantage of extracting the system eigenvalues directly from the EMTP solution is the ability of the EMTP to follow the characteristics of nonlinearities. The system’s trajectory can be accurately traced and the calculated eigenvalues and eigenvectors correctly represent the system’s instantaneous dynamics. In addition, the algorithm can be used as a tool to identify network partitioning subsystems suitable for real-time hybrid power system simulator environments, including the implementation of multi-time scale solutions. The proposed technique can be implemented as an extension to any EMTP-based simulator. Within our UBC research group, it is aimed at extending the capabilities of our real-time PC-cluster Object Virtual Network Integrator (OVNI) simulator.
