Abstract:
Abstract. Soil information (e.g., soil texture and porosity) from existing soil datasets over the Tibetan Plateau (TP) is claimed to be inadequate and even inaccurate for determining soil hydraulic properties (SHP) and soil thermal properties (STP), hampering the understanding of the land surface process over TP. As the soil varies across three dominant climate zones (i.e., arid, semi-arid and subhumid) over the TP, the associated SHP and STP are expected to vary correspondingly. To obtain an explicit insight into the soil hydrothermal properties over the TP, in situ and laboratory measurements of over 30 soil property profiles were obtained across the climate zones. Results show that porosity and SHP and STP differ across the climate zones and strongly depend on soil texture. In particular, it is proposed that gravel impact on porosity and SHP and STP are both considered in the arid zone and in deep layers of the semi-arid zone. Parameterization schemes for porosity, SHP and STP are investigated and compared with measurements taken. To determine the SHP, including soil water retention curves (SWRCs) and hydraulic conductivities, the pedotransfer functions (PTFs) developed by Cosby et al. (1984) (for the Clapp–Hornberger model) and the continuous PTFs given by Wösten et al. (1999) (for the Van Genuchten–Mualem model) are recommended. The STP parameterization scheme proposed by Farouki (1981) based on the model of De Vries (1963) performed better across the TP than other schemes. Using the parameterization schemes mentioned above, the uncertainties of five existing regional and global soil datasets and their derived SHP and STP over the TP are quantified through comparison with in situ and laboratory measurements. The Steiner tree problem (STP) aims to determine some Steiner nodes such that the minimum spanning tree over these Steiner nodes and a given set of special nodes has the minimum weight, which is NP-hard. STP includes several important cases. The Steiner tree problem in graphs (GSTP) is one of them. Many heuristics have been proposed for STP, and some of them have proved to be performance guarantee approximation algorithms for this problem. Since evolutionary algorithms (EAs) are general and popular randomized heuristics, it is significant to investigate the performance of EAs for STP. Several empirical investigations have shown that EAs are efficient for STP. However, up to now, there is no theoretical work on the performance of EAs for STP. In this article, we reveal that the (1+1) EA achieves 3/2-approximation ratio for STP in a special class of quasi-bipartite graphs in expected runtime [Formula: see text], where [Formula: see text], [Formula: see text], and [Formula: see text] are, respectively, the number of Steiner nodes, the number of special nodes, and the largest weight among all edges in the input graph. We also show that the (1+1) EA is better than two other heuristics on two GSTP instances, and the (1+1) EA may be inefficient on a constructed GSTP instance.

Abstract:
This research aims at investigating the realization of thematic progression (TP) patterns, which is defined as “the way in which the theme of a clause may pick up, or repeat a meaning from a preceding theme or rheme” (Paltrigde, 2006, p. 148), and their effects on text quality of online IELTS Writing Task 2 samples. Investigated in the light of systemic functional approach, TP is classified into five patterns, namely Simple Linear Progression (SLP), Constant Progression (CP), Split Rheme Progression (SRP), Split Theme Progression (STP) and Derived Hypertheme Progression (DHP) (McCabe, 1999). In this study, the corpus consisting of 24 sample essays derived from an English-learning website was analyzed using both quantitative and qualitative methods. The results of this study highlight that SLP is the most frequently employed pattern in the corpus, followed by CP, SRP, STP and DHP patterns respectively. The analysis also shows that proper employment of TP overall is a contributing factor to the coherence and cohesion of the text, with each pattern exerting different effects. SLP, with its cross-referential links, yields a constantly developing and cohesive text whilst CP keeps a strong thematic focus. Meanwhile, SRP and STP patterns are both found to offer a planned text development and DHP creates cohesion with hypernym-hyponym relation. Problems related to the lack of TP, inappropriate theme positioning and the overuse of the theme “It” and “There” are also reported in the corpus.

Abstract:
Sugar transporter proteins (STPs), such as H+/sugar symporters, play essential roles in plants’ sugar transport, growth, and development, and possess an important potential to enhance plants’ performance of multiple agronomic traits, especially crop yield and stress tolerance. However, the evolutionary dynamics of this important gene family in Gramineae crops are still not well-documented and functional differentiation of rice STP genes remain unclear. To address this gap, we conducted a comparative genomic study of STP genes in seven representative Gramineae crops, which are Brachypodium distachyon (Bd), Hordeum vulgare (Hv), Setaria italica (Si), Sorghum bicolor (Sb), Zea mays (Zm), Oryza rufipogon (Or), and Oryza sativa ssp. japonica (Os). In this case, a total of 177 STP genes were identified and grouped into four clades. Of four clades, the Clade I, Clade III, and Clade IV showed an observable number expansion compared to Clade II. Our results of identified duplication events and divergence time of duplicate gene pairs indicated that tandem, Whole genome duplication (WGD)/segmental duplication events play crucial roles in the STP gene family expansion of some Gramineae crops (expect for Hv) during a long-term evolutionary process. However, expansion mechanisms of the STP gene family among the tested species were different. Further selective force studies revealed that the STP gene family in Gramineae crops was under purifying selective forces and different clades and orthologous groups with different selective forces. Furthermore, expression analysis showed that rice STP genes play important roles not only in flower organs development but also under various abiotic stresses (cold, high-temperature, and submergence stresses), blast infection, and wounding. The current study highlighted the expansion and evolutionary patterns of the STP gene family in Gramineae genomes and provided some important messages for the future functional analysis of Gramineae crop STP genes.

Abstract:
Our previous studies have shown the presence of daytime positive electron density disturb-ances during several days after the start of the recovery phase. The aim of this paper is to study after-effects of geomagnetic storms (after-storm effects), i.e. ionospher-ic effects observed on the 3–5th day after the beginning of the storm recovery phase. From numerical calcula-tions with the GSM TIP model, we have found the main mechanisms for the formation of the after-storm effects. Using Irkutsk (52° N, 104° E) and Kaliningrad (54° N, 20° E) ionosonde data, we have carried out a statistical analysis of daytime ionospheric responses to geomagnetic storms. As a result of the analysis, we obtained averaged ionospheric responses at the beginning of the storm recovery phase and for five consecutive days. The statistical analysis results received near the beginning of the recovery phase are in good agreement with the well-known ionospheric effects of geomagnetic storms obtained in previous studies. For the first time, the obtained statistics of iono-spheric responses observed on the 3–5th day after the beginning of the recovery phase allowed us to reveal the dependence of after-storm ionospheric effects on season, storm intensity, and ionosonde geomagnetic latitude. In addition, we for the first time present the interpretation of after-storm ionospheric effects from numerical simulation results.

The glass-to-metal seals are usually used in the solar thermal power (STP) and electronic devices. However, the requirement of mechanical properties in the STP is much higher than that of electronic devices, because the glass-to-metal joints used in the STP need to have anti-fatigue performance in adition to higher static tensile strength. Under the repeated fluctuating loads, damage and failures of glass-to-metal seals in the STP often lead to serious consequences. Therefore, analysis of damage evolution and fracture behavior of glass-to-metal diffusion welded joints was performed in this paper. Firstly, the finite element (FE) model of glass-to-metal welded joints was established in accordance with the STP welded structures. And damage simulation was carried out by the FE software ABAQUS. Also, the work illustrates the modeling of damage in terms of traction versus separation to simulate crack propagation and introduces the use of traction-separation law as a damage initiation and evolution criteria. The microgram of damage distribution in the glass side near the interface could be characterized by Scanning Electron Microscope (SEM), which was compared with predictions obtained by finite element method (FEM) analysis. As result, the damage criteria on the lap joints in conjunction with FM analysis were used to optimize the glass-to-metal diffusion welding technology. The above results provide the basis of design against damage and reliable estimation of glass-to-metal seals.

Many firms are increasingly cooperating in their technological undertakings. They engage in strategic technology partnerships (STP) for technological, commercial, industrial, and financial reasons. STP is deemed imperative for easing access to strands of technologies that are unknown to a company. This frequency analysis on STP is based on systematic literature review (SLR) approach and provides a thorough review of 57 articles published in highly-ranked peer-reviewed journals spanning a 26-year period from 1988 to the beginning of 2014. Research on STP is somewhat fragmented which, renders some of the research studies irreconcilable and impedes a greater understanding and consistency of the discipline. There is still a growing body of literature on the subject matter from various disciplinary perspectives which adopt various theoretical and methodological lenses at diverse levels of analysis. In the current paper, we analyze among other things the various methodological, research and theoretical issues underpinning STP and propose a research agenda. Hence, we contribute to the existing body of knowledge on STP literature by offering a state-of-the-art overview of current research and elaborating on promising areas for further investigation.

The work is devoted to the development of a fundamentally new way of modeling the ionospheric D-region – deterministic-probabilistic. The results of Ne calculations using this technique are analyzed. Research of this kind is of fundamental importance, related to the rejection of a purely deterministic description of a continuously changing environment such as the ionosphere. In this work, the electron density is calculated using a five-component system of ionization-recombination cycle equations. Probability density functions (PDFs) of input parameters of the model are used to solve the system. The most important sources of the D-region ionization are taken into account to calculate PDFs of the ionization rate. The necessary number of iterations is determined by the convergence of PDFs of the electron density from 50 km to 85 km at midlatitudes under different heliogeophysical conditions. Theoretical Ne PDFs have been shown to be in good agreement with two experimental databases on electron density, especially at large D-region heights. The next important stage of modeling is the thorough verification of Ne PDFs from experimental radiophysical data on VLF–LF propagation.

Metal acquisition is vital to pathogens for successful infection within hosts. Staphylopine (StP), a broad-spectrum metallophore biosynthesized by the major human pathogen, Staphylococcus aureus, plays a central role in transition-metal acquisition and bacterial virulence. The StP-like biosynthesis loci are present in various pathogens, and the proteins responsible for StP/metal transportation have been determined. However, the molecular mechanisms of how StP/metal complexes are recognized and transported remain unknown. We report multiple structures of the extracytoplasmic solute-binding protein CntA from the StP/metal transportation system in apo form and in complex with StP and three different metals. We elucidated a sophisticated metal-bound StP recognition mechanism and determined that StP/metal binding triggers a notable interdomain conformational change in CntA. Furthermore, CRISPR/Cas9-mediated single-base substitution mutations and biochemical analysis highlight the importance of StP/metal recognition for StP/metal acquisition. These discoveries provide critical insights into the study of novel metal-acquisition mechanisms in microbes.

Recently, the concept of a Sliding Track Panel (STP) system, which allows the bridge to slide in the longitudinal direction, has been proposed for continuous welded rail method laid on long span non-ballast bridges, such as truss railway bridges. The performance of the STP system is determined based on the ability to prevent the lateral buckling of STP. This study attempts to derive the optimum interval distance and required stiffness of the lateral support member to be installed in the STP system through buckling analysis. According to the analysis, the interval distance of lateral support member, which satisfies the strength of STP, is less than 2.8 m and the rigidity is more than 800 N/mm.

Abstract:
The whole genome duplication (WGD) event is an intriguing mechanism from an evolutionary perspective. Such an event may be the source of new genes, functions or species. Traces of WGD event have been detected in the genome of all four eukaryotic kingdoms: plants, animals, fungi and protists. In fungi, an ancestor of Saccharomyces cerevisiae underwent an event of WGD, about 100 million years ago, after diverging from the Kluyveromyces lineage. In S. cerevisiae, only ten percent of the resulting duplicated genes survived as duplicates. In particular, some of these duplicates encodes for transcription factors in several nutrient sensing pathways.

The main subject of this thesis’s work is the external amino acid sensing system in S. cerevisiae. The detection of extracellular amino acids in yeast begins with a transporter homologue devoid of any uptake activity, the Ssy1 sensor. The binding of extracellular amino acids to Ssy1 leads to the successive activation of Ptr3 and the Ssy5 endoprotease. This endoporotease catalyses the processing of two transcriptions factors: Stp1 and Stp2. The Stp factors, released from their N-terminal cytoplasmic-anchored domains, are then translocated into the nucleus, where they activate the transcription of several amino acid permease genes (e.g. AGP1 and DIP5). Starting this work, the Stp factors were considered as functionally redundant.

We first determined that the STP1 and STP2 genes derivate from the event of WGD. The conservation of these two genes in S. cerevisiae was accompanied by a functional divergence of their products at several levels: processing sensibility, transcriptional activation capacity, target genes, cellular abundance level and stability. The Stp2 factor with its high abundance in the cells and its higher Ssy5-processing sensibility is specialized towards induction of the AGP1 gene when the external amino acid signaling is weakly stimulated. Under strong stimulation conditions, the amino acids induce cleavage-triggered destabilization of Stp2 through the proteasomal pathway and the induction of AGP1 is mediated mainly by the Stp1 transcription factor. Unlike Stp2, the Stp1 factor is characterized by its high transcriptional activation capacity and weaker sensitivity towards Ssy5-processing. The Stp factors differ also by their genetic targets. Indeed, only Stp2 regulates the expression of DIP5. Finally, we determined that the processing sensibility and the transcriptional activation capacity of each Stp factors is directly linked to their N- and C-terminal domains, respectively.

The phosphorylation states and the degradation of the Stp2 factor were also examined. The event of degradation concerns only the processed forms of this factor and takes places principally in the nucleus. Some data indicate that such an event might be important to limit the activation capacity of this factor. The role of the Stp2 phosphorylation in the external amino acid signaling pathway is still unknown but this event might be important for the Stp2 degradation or its transcriptional activity.

The unique Stp factor from Kluyveromyces lactis (Kl-Stp), a pre-WGD species, was also studied. The Kl-Stp factor shares at least two characteristic with the S. cerevisiae Stp2 factor: high sensibility towards processing and high levels of degradation. This observation leads us to conclude to that the STP genes may have been conserved after WGD though a mechanism called neofunctionalization (one of the duplicate obtained after duplication retains the ancestral function while the other evolves to perform a novel function).

Finally, a new model for the external amino acid signaling pathway that brings together all the data obtained during this thesis’s work is proposed.

This report is a part of a larger project with the aim of developing a proof-of-concept prototype. The project was started on the initiative of Company X and is a collaboration between them and the University of Borås. Company X are a logistics consultant firm specialized in warehousing operations. Through this project Company X hope to strengthen their standing on the market and offer their customers new and innovative solutions. The purpose of this project is to, with the assistance of AI, calculate in what cluster within a warehouse a product should be placed in order to create more efficient picking routes and to research the market in order to evaluate the business potential for this kind of service. This specific report aims to conduct a market analysis for the potential service. It will do so by conducting interviews and creating a theoretical framework consisting of relevant literature. When performing the market research and analysis of the potential IT-service there are a few limitations caused by a number of factors that could potentially affect the significance of the end result. The service is not yet developed and cannot be described in detail but only as an idea, which could make it difficult for the interview respondents to fully understand the service. The report will also be limited for certain practical reasons such as the Covid-19 pandemic. Because of this, the report has not had the opportunity to conduct the interviews with potential customers on site but have been conducted online, via link. The theoretical framework has two different main focus areas. The first area addresses basic warehouse logistics and difficulties in selling high technology products, while the second area includes marketing strategies and strategic marketing planning. The market analysis will consist of a SWOT-analysis and an STP-analysis. The report has concluded that the service has potential and suggests that company type B is the best suited potential customer. Personal and demand driven sales are also considered helpful factors when selling high-tech products and solutions.

Abstract:
Salmonella enterica serovar Typhimurium genome encodes 13 fimbrial operons. Most of the fimbriae encoded by these operons are not produced under laboratory conditions but are likely to be synthesized in vivo. We used an in vivo expression technology (IVET) strategy to identify four fimbrial operons, agf, saf, sti, and stc that are expressed in the spleen. When any three of these operons were deleted, the strain retained wild-type virulence. However, when all four operons were deleted, the resulting strain was completely attenuated, indicating that these four fimbriae play functionally redundant roles critical for virulence. In mice, oral doses of as low as 1 x 10(5) CFU of the strain with four fimbrial operons deleted provided 100% protection against challenge with 1 x 10(9) CFU of wild-type S. Typhimurium. We also examined the possible effect of these fimbriae on the ability of a Salmonella vaccine strain to deliver a guest antigen. We modified one of our established attenuated vaccine strains, chi 9088, to delete three fimbrial operons while the fourth operon was constitutively expressed. Each derivative was modified to express the Streptococcus pneumoniae antigen PspA. Strains that constitutively expressed saf or stc elicited a strong Th1 response with significantly greater levels of anti-PspA serum IgG and greater protective efficacy than strains carrying saf or stc deletions. The isogenic strain in which all four operons were deleted generated the lowest anti-PspA levels and did not protect against challenge with virulent S. pneumoniae. Our results indicate that these fimbriae play important roles, as yet not understood, in Salmonella virulence and immunogenicity. IMPORTANCE Salmonella enterica is the leading cause of bacterial food-borne infection in the United States. S. Typhimurium is capable of producing up to 13 distinct surface structures called fimbriae that presumably mediate its adherence to surfaces. The roles of most of these fimbriae in disease are unknown. Identifying fimbriae produced during infection will provide important insights into how these bacterial structures contribute to disease and potentially induce protective immunity to Salmonella infection. We identified four fimbriae that are produced during infection. Deletion of all four of these fimbriae results in a significant reduction in virulence. We explored ways in which the expression of these fimbriae may be exploited for use in recombinant Salmonella vaccine strains and found that production of Saf and Stc fimbriae are important for generating a strong immune response against a vectored antigen. This work provides new insight into the role of fimbriae in disease and their potential for improving the efficacy of Salmonella-based vaccines.

This thesis report presents a method to analyze the communication architecture for the Intelligent Transportation Systems (ITS). The report also includes a case study on ASN.1 notation and analysis of its encoding rules. Included in the report is also: (i) accompanying instruction on how to use ASN.1 compilers to produce the C/C++ message encoder/decoder, and (ii) analysis of Non-IP communications of Communication Access for Land Mobiles (CALM-FAST) protocol stack in ITS. The thesis is a part of the research project entitled “SCOOP”, a joint project between SCANIA CV AB and KTH. The purpose of this thesis is to contribute to the ultimate goal, which is to equip a vehicle with necessary hardware and software technology to provide a platooning behavior in the GCDC 2011 competition. This goal is achieved by the means of wireless communication system for both vehicle to vehicle and vehicle to road side units communications in the platoon. Overall, this thesis introduces the important usage of ASN.1 in implementation of cut-edge telecommunication systems especially in V2V and V2I communication; and clarifies the CALM-FAST protocol stack in mobile nodes.

The present work introduces a methodology to obtain a discrete time state space representation of an electrical network using the nodal [G] matrix of the Electromagnetic Transients Program (EMTP) solution. This is the first time the connection between the EMTP nodal analysis solution and a corresponding state-space formulation is presented. Compared to conventional state space solutions, the nodal EMTP solution is computationally much more efficient. Compared to the phasor solutions used in transient stability analysis, the proposed approach captures a much wider range of eigenvalues and system operating states. A fundamental advantage of extracting the system eigenvalues directly from the EMTP solution is the ability of the EMTP to follow the characteristics of nonlinearities. The system's trajectory can be accurately traced and the calculated eigenvalues and eigenvectors correctly represent the system's instantaneous dynamics. In addition, the algorithm can be used as a tool to identify network partitioning subsystems suitable for real-time hybrid power system simulator environments, including the implementation of multi-time scale solutions. The proposed technique can be implemented as an extension to any EMTP-based simulator. Within our UBC research group, it is aimed at extending the capabilities of our real-time PC-cluster Object Virtual Network Integrator (OVNI) simulator.

IP Multimedia Subsystem (IMS) is the next generation of telecommunication systems. The system is based on an IP network and uses technologies from the Internet. The IMS system is designed to evolve from a telephone system into a general information and communication system. It is meant to include television, Video on Demand (VoD), interactive services etc, etc. It is designed to simplify the implementation of newservices in telecom networks. This report investigates security aspects of VoD services when merging an IP Television (IPTV) system with IMS. The investigation covers security functions in IMS, transitionsolutions for authentication of the Set-Top-Box (STB) in IMS, and identifies problems inthe integration of IPTV and IMS. The report concludes that IMS has good solid Authentication, Authorization, and Accounting (AAA) functions that will provide security and billing functionality. One problemis found in the media control between the STB and the streaming server. This interface lacked specification at the time of investigation, and some problems have been identified. These problems have to be solved before a system can be brought into service and beregarded as secure.

Including local experience information with generic data can maximize operational flexibility and help make better informed nuclear asset management decisions. Experience data are particularly helpful in risk-informed asset management decision-making. In 2004, South Texas Project (STP) used a risk-informed study to help decide the proper timing for reactor vessel head replacement. The decision to replace the reactor vessel heads was based on a previous study. Up to the time of decision-making inspections had not been performed at STP and therefore, the likelihood of finding a J-Groove failure in an inspection was based on generic industry information in both of the studies. In the Unit 1 fall outage, a full volumetric reactor vessel head penetration inspection was performed. The inspection method is briefly described. The algorithm for optimal timing of the reactor vessel head is presented and solved for the STP case. The generic industry J-Groove failure data is updated with the recent results from the Unit 1 outage inspection and the new failure projections are summarized. The optimal timing model is again run using the updated data and compared with the previous results.

The South Texas Project (STP) balance of plant (BOP) model for production losses has been converted from an Excel-based software application that uses minimal cutsets from a single large fault tree, to a set of small fault trees used within a risk analysis tool, Riskman® [8]. The Riskman® application provides estimates of interest to plant investors and owners such as potential production losses and plant profitability, as well as production reliability and quality (loss frequency, type of loss, loss duration and standard economic risk metrics). The BOP models and methodologies developed are applicable to both fault-tree linking and event-tree linking styles of risk analysis. The STP BOP model is developed to support configuration risk management in two categories: 1) trip risk and concomitant core damage frequency; and 2) non-trip production risk. The primary purpose for the conversion work is to make the STP BOP model framework and application platform consistent with the STP probabilistic risk assessment model. In the course of the conversion, improvements to the Riskman® software have been implemented and techniques to more efficiently produce BOP modeling (fault tree) improvements were developed. Better methods to accurately calculate initiating event frequencies are studied. New data types added to support BOP modeling are described.

This paper presents a simplified model for assessing the hydroelasticity of the extended-column (e-column) for an extended-draft semi-submersible (E-SEMI) by using commercially available software. The E-SEMI comprises a second tier pontoon (STP) connected to a conventional semi-submersible by using extended-column (e-column). The purpose of attaching the STP is to increase the heave added mass, and as a result, could reduce the heaving motion and shift the natural period further away from the wave spectrum by increasing the natural period. One of the challenges of the E-SEMI is to obtain the hydroelastic response of the e-column when subjected to wave and current loadings. The simplified model models the e-column and STP using beam-column theory that has similar vibration frequencies and modes shapes of the complex model. The potential wave theory is used to model the waves and the Morrison equation is use to obtain the drag and inertia force acting on the e-column due to current loading. Hydroelasticity shall be performed on this simplified model to assess the strength performance as well as the deflection of the e-column. The bending moment obtained from the hydroelastic analysis is also significant in the connector design that keeps the e-column in place.

The loss of air conditioning in the electrical auxiliary building at South Texas Project (STP) has been shown in previous work to result in relatively rapid air temperature rise. The heat up is a concern in the Probabilistic Risk Assessment (PRA) because it may be associated with high conditional core damage probability. As a consequence, operator responses to mitigate the building heat up have been developed and proceduralized at STP. The plant’s current loss of electrical auxiliary building HVAC operator procedure was analyzed and improvements have been recommended. This was done using a commercial computational fluid dynamics (CFD) package that modeled the transient air temperature rise of the building and qualitatively assessed the effectiveness of operator actions. A possible methodology for validating the CFD results was developed using coupled energy balance equations for individual rooms. The methodology is a conservative approach to compare the effectiveness operator actions and provide steps for future higher fidelity simulation and validation. While the specific approach is applicable to STP, the overall methods and approaches described should be applicable to other sites that may be subject to excessive room heat up due to loss of air conditioning in critical rooms.

TQM is a management philosophy where every individual working in an ideal environment towards a common goal, continuously improving the processes and in turn improving the quality of product or process. An important factor for the success of an organization in implementing TQM is its ability to translate, integrate, and ultimately implement TQM behaviors into everyday practice on the job. This study was initially conducted by investigating the quality affecting factors (attributes) from different pieces of literature on quality performances and understanding the concept of total quality management. The questions in this questionnaire are distributed among the different phases that take place during the construction. Factor analysis involved identifying a total of 55 attributes that belonged to the different phases of construction, these factors were then distributed into critical success and failure attributes based on data analysis. These critical attributes affect the Quality performance in the construction of the STP project.
