ABSTRACT Sterigmatocystin (ST) and aflatoxin B1(AFB1) are two polyketide-derived Aspergillusmycotoxins synthesized by functionally identical sets of enzymes. ST, the compound produced by Aspergillus nidulans, is a late intermediate in the AFB1 pathway of A. parasiticus and A. flavus. Previous biochemical studies predicted that five oxygenase steps are required for the formation of ST. A 60-kb ST gene cluster in A. nidulanscontains five genes, stcB, stcF,stcL, stcS, and stcW, encoding putative monooxygenase activities. Prior research showed thatstcL and stcS mutants accumulated versicolorins B and A, respectively. We now show that strains disrupted atstcF, encoding a P-450 monooxygenase similar to A. parasiticus avnA, accumulate averantin. Disruption of either StcB (a putative P-450 monooxygenase) or StcW (a putative flavin-requiring monooxygenase) led to the accumulation of averufin as determined by radiolabeled feeding and extraction studies.

Abstract:
Stanniocalcin (STC) is a calcium- and phosphate-regulating hormone produced in bony fish by the corpuscles of Stannius, which are located close to the kidney. It is a major antihypercalcemic hormone in fish. As the corpuscles of Stannius are absent, and antihypercalcemic hormones are basically not necessary, in mammals, the discovery of a mammalian homolog, STC1, was surprising and intriguing. STC1 displays a relatively high amino acid sequence identity (∼50%) with fish STC. In contrast to fish STC, STC1 is expressed in many tissues, including kidney. More recently, a human gene encoding the second stanniocalcin-like protein, STC2, was identified. STC2 has a lower identity (∼35%) with STC1 and fish STC. Similar to STC1, STC2 is also expressed in a variety of tissues. Research into the functions of STCs in mammals is still at an early stage, and the ultimate physiological and pathological roles of STCs have not yet been established. A few studies indicate that STC1, similar to fish STC, stimulates phosphate absorption in the kidney and intestine, but the function of STC2 is still unknown. However, several interesting findings have been reported on their cellular localization, gene structure, and expression in different physiological and pathological conditions, which will be clues in elucidating the functions of STCs in mammals. STC1 expression is enhanced by hypertonicity in a kidney cell line or by ischemic injuries and neural differentiation in the brain. STC1 expression in the ovary is also enhanced during pregnancy and lactation. Calcitriol upregulates STC1 and downregulates STC2 expression in the kidney. Interestingly, STC1 and STC2 are expressed in many tumor cell lines, and the expression of STC2 is enhanced by estradiol in breast cancer cells. STC2 is also expressed in pancreatic islets. These results suggest that the biological repertoires of STCs in mammals will be considerably larger than in fish and may not be limited to mineral metabolism. This brief review describes recent progress in mammalian STC research.

Abstract:
O uso de silagens de forrageiras tropicais é mais uma alternativa para minimizar a escassez de forragem no período seco, dentre as forrageiras que podem ser adotadas o capim elefante merece destaque. Nesse contexto, o objetivo do trabalho foi avaliar os aspectos bromatológicos e qualidade fermentativa de silagem de capim elefante aditivada com torta de semente de cupuaçu. O experimento foi realizado em delineamento inteiramente casualizado com cinco tratamentos e quatro repetições, denominados de: SCE= 100% silagem de capim elefante puro; STC20 = silagem 80% capim elefante + 20% de torta de cupuaçu; STC40 = silagem 60% capim elefante + 40% de torta de semente de cupuaçu; STC60 = silagem 40%capim elefante + 60% de torta de semente de cupuaçu e STC80 = silagem 20% capim elefante + 80% de torta de semente de cupuaçu. Foram utilizados silos de PVC com 50 cm de comprimento, 10 cm de diâmetro e capacidade de 3,5kg, após 60 dias de fermentação os silos foram abertos, pesados novamente, homogeneizadas e retiradas amostras para determinar os teores de: MS, PB, FDN, FDA, MM, EE, CEL, HEM, LIG, MM, NDT, PMS, PE, PG e pH da silagem. Em termos de composição bromatológicas a silagem STC80 foi a que apresentou os melhores resultados, além de diminuir as perdas de MS, as perdas por efluentes e por gases. Portanto, recomenda-se a inclusão de 80% da torta de semente de cupuaçu na silagem com capim elefante, por melhorar o perfil fermentativo e a composição bromatológica da silagem. Abstract:
Stanniocalcin (STC), a secreted glycoprotein, was first studied in fish as a classical hormone with a role in regulating serum calcium levels. There are two closely related proteins in mammals, STC1 and STC2, with functions that are currently unclear. Both proteins are expressed in numerous mammalian tissues rather than being secreted from a specific endocrine gland. No phenotype has been detected yet in Stc1-null mice, and to investigate whether Stc2 could have compensated for the loss of Stc1, we have now generated Stc2−/− and Stc1−/−Stc2−/− mice. Although Stc1 is expressed in the ovary and lactating mouse mammary glands, like the Stc1−/− mice, the Stc1−/−Stc2−/− mice had no detected decrease in fertility, fecundity, or weight gain up until weaning. Serum calcium and phosphate levels were normal in Stc1−/−Stc2−/− mice, indicating it is unlikely that the mammalian stanniocalcins have a major physiological role in mineral homeostasis. Mice with Stc2 deleted were 10–15% larger and grew at a faster rate than wild-type mice from 4 wk onward, and the Stc1−/−Stc2−/− mice had a similar growth phenotype. This effect was not mediated through the GH/IGF-I axis. The results are consistent with STC2 being a negative regulator of postnatal growth.

Abstract:
Stanniocalcin 1 (STC1) and stanniocalcin 2 (STC2) are two recently identified mammalian peptide hormones. STC1 plays a role in calcium and phosphate homoeostasis, while the role of STC2 is unknown. We examined a human fibrosarcoma cell line, HT1080, that has high steady-state STC1 and STC2 mRNA levels, to determine whether these proteins are secreted. Following incubation of HT1080 cells with 32P, labelled STC1 and STC2 were found to be secreted into the medium. STC1 was phosphorylated in vitro by protein kinase C (PKC). In vitro and in vivo phosphorylation both occurred exclusively on serine and the phosphopeptide maps were similar, suggesting that PKC might be the in vivo kinase. STC2 was phosphorylated in vitro by casein kinase II (CK2), in vitro and in vivo phosphorylation were exclusively on serine and the phosphopeptide maps were indistinguishable. Phosphorylation of STC2 in intact cells resulted from the action of an ecto-protein kinase, since exogenous STC2 was phosphorylated by HT1080 cells and no phosphorylated STC2 was detectable inside the cells. The ectokinase activity was abolished by heparin and GTP could substitute for ATP as the phosphate donor, indicative of an ecto-CK2-like activity. The in vitro CK2 phosphorylation site was shown by matrix-assisted laser-desorption ionization–time-of-flight MS to be a single serine located between Ser-285 and Ser-298 in the C-terminal region of STC2. This is the first report of the secretion of STC1 or STC2 from mammalian cells. We conclude that these human fibrosarcoma cells express both STC1 and STC2 as secreted phosphoproteins in vivo, with STC2 being phosphorylated by an ecto-CK2-like enzyme.

Abstract:
Given the proven zoonotic potential of the intestinal protozoan Blastocystis sp., a fast-growing number of surveys are being conducted to identify potential animal reservoirs for transmission of the parasite. Nevertheless, few epidemiological studies have been conducted on farmed animals in Egypt. Therefore, a total of 1089 fecal samples were collected from herbivores (sheep, goats, camels, horses, and rabbits) in six Egyptian governorates (Dakahlia, Gharbia, Kafr El Sheikh, Giza, Aswan, and Sharqia). Samples were screened for the presence of Blastocystis sp. by real-time PCR followed by sequencing of positive PCR products and phylogenetic analysis for subtyping of the isolates. Overall, Blastocystis sp. was identified in 37.6% of the samples, with significant differences in frequency between animal groups (sheep, 65.5%; camels, 62.2%; goats, 36.0%; rabbits, 10.1%; horses, 3.3%). Mixed infections were reported in 35.7% of the Blastocystis sp.-positive samples. A wide range of subtypes (STs) with varying frequency were identified from single infections in ruminants including sheep (ST1–ST3, ST5, ST10, ST14, ST21, ST24, ST26, and ST40), goats (ST1, ST3, ST5, ST10, ST26, ST40, ST43, and ST44), and camels (ST3, ST10, ST21, ST24–ST26, ST30, and ST44). Most of them overlapped across these animal groups, highlighting their adaptation to ruminant hosts. In other herbivores, only three and two STs were evidenced in rabbits (ST1–ST3) and horses (ST3 and ST44), respectively. The greater occurrence and wider genetic diversity of parasite isolates among ruminants, in contrast to other herbivores, strongly suggested that dietary habits likely played a significant role in influencing both the colonization rates of Blastocystis sp. and ST preference. Of all the isolates subtyped herein, 66.3% were reported as potentially zoonotic, emphasizing the significant role these animal groups may play in transmitting the parasite to humans. These findings also expand our knowledge on the prevalence, genetic diversity, host specificity, and zoonotic potential of Blastocystis sp. in herbivores.

Abstract:
Halotolerant plant growth-promoting rhizobacteria (HTPGPR) are beneficial microbes that can be exploited to mitigate the negative effects of soil salinity on crops. In the current investigation, eight saline soil samples collected from 2 mangrove ecosystems of Tamil Nadu (Chidambaram and Thanjavur) during December 2022 were used to isolate saline tolerant bacterial cultures at the Department of Agricultural Microbiology, Tamil Nadu Agricultural University, Coimbatore, Tamil Nadu, India. Using 5 different bacterial growth media, 48 rhizobacterial isolates (ST1 to ST48) were obtained. Salt tolerance ability (0, 5, 10, 15 and 20% NaCl) of these 48 isolates under in vitro conditions indicated their potential to tolerate up to 10% NaCl (1.71 M). None of the isolates could grow at 20% NaCl (3.42M). Bacterial isolates such as ST11, ST13, ST18, ST20 and ST27 showed minimum growth at 15%NaCl (2.57M). 33 isolates which could grow well at higher salt concentrations were selected. Among 33 isolates, 18 isolates with higher concentration of intracellular sodium content (ST1, ST2, ST3, ST7, ST8, ST9, ST11, ST12, ST13, ST17, ST18, ST20, ST27, ST30, ST32, ST34, ST39, and ST40) were selected and characterized qualitatively for their ability to mineralize phosphate, potassium, and zinc, and to produce HCN. Potential of these18 bacterial isolates to tolerate other abiotic stress factors such as pH and temperature was also studied. Among 18 isolates, the isolates ST7, ST17, and ST30 were found to be multimineral solubilizers. Bacterial isolate ST17 was found to prefer alkaline (pH 9.0) and mesophilic temperature (35ºC) for its growth at both saline (5% NaCl) and non saline condition.

Stanniocalcin 1 (STC1) and STC2 are secreted, homodimeric glycoproteins that share 30% amino acid sequence identity. Breast tumour gene profiling studies have demonstrated significantly upregulated STC2 expression in hormone-responsive positive breast tumours; therefore, the purpose of this study was to investigate STC2 hormonal regulation and function in breast cancer cells. Here we report that STC2 is expressed in a number of human breast cancer cell lines, regardless of their oestrogen (E2) and progesterone (P4) receptor status, and its expression is readily detectable in human and mouse mammary gland tumours. Besides E2, retinoic acid (RA) and P4 play an important role in the regulation of STC2 expression, not only in MCF-7 but also in other breast cancer and non-breast cell lines. The expression of the related hormone, STC1, is not affected by the above hormones in breast and endometrial cancer cell lines implying a fundamental difference in regulation in cancer cell lines. The induction of STC2 expression by E2 and RA occurs at the transcriptional level but through intermediary transcription factors. The STC2 proximal promoter region is not responsible for hormonal induction, but exhibits a high basal transcriptional activity. Constitutive STC2 expression in human breast cancer cell lines resulted in significant impairment of cell growth, migration and cell viability after serum withdrawal. In conclusion, STC2 is a downstream target of E2, P4 and RA signalling pathways. In hormone receptor negative cell lines it can function in a paracrine/autocrine fashion to reduce cell proliferation.

This study aimed to evaluate the productive performance of dairy cows fed with sugarcane treated with 5g kg-1 of calcium oxide (CaO) or hydroxide [Ca(OH)2]. Eight Holstein cows with 638.01±12.52kg of body weight and milk yield of 20.32±1.5kg d-1 were randomly assigned into two 4x4 Latin squares, fed with the following diets composed of corn silage (CS), fresh sugarcane (FS), sugarcane treated with calcium oxide (STCO) or calcium hydroxide (STCH) as only forage. Data collection lasted five days, after 15 days of adaptation to diets and facilities. The dry matter intake (% of body weight) was higher in diets with CS (3.08) compared to those with FS (2.67), STCO (2.73) or STCH (2.73), which did not differ. Diets with CS determined milk production adjusted for 4% fat (20.05kg d-1) similar to diets containing STCO and STCH (18.01 and 17.89kg d-1, respectively) and higher than those with FS (17.33kg d-1). The experimental diets did not alter the composition of milk. The use of sugarcane treated with Ca(OH)2 is a viable option for feeding Holstein cows with average genetic potential for milk production because it allows production and composition similar to milk dairy cows fed with corn silage, besides benefiting the logistics of feeding in the rural properties.

Abstract:
6-shogaol is a natural and the most potent bioactive vanilloid in dried Zingiber officinale rhizomes. Many scientific studies have reported the diverse biological activities of 6-shogaol. However, the major drawback of 6-shogaol is its instability at room temperature. We synthesised new shogaol thiophene compounds (STCs) by replacing the pentyl group in the sidechain with thiophene derivatives. The STCs were tested for their nuclear factor erythroid 2-related factor 2 (NRF2) activation ability in murine hepatoma cells (Hepa1c1c-7) by determining their NAD(P)H quinone oxidoreductase 1 (NQO1) inducing ability and expression of NRF2-associated antioxidant genes. The anti-inflammatory activity of STCs was determined in Escherichia coli lipopolysaccharide (LPSEc)-stimulated NR2-proficient and -silenced mouse microglial cells (BV-2) by measuring the inflammatory markers, cytokines, and mediators. The modes of action (interacting with the Kelch domain of KEAP1, covalent bonding with cysteines of KEAP1, and inhibition of GSK-3b enzyme activity) of NRF2 activation by STCs were determined using commercially available kits. The in vitro metabolic stability of the STCs in liver microsomes (humans, rats, and mice) was also investigated. The molecular docking and molecular dynamics studies were conducted to identify the binding poses, stability, and molecular interactions of the STCs in the binding pockets of Kelch and BTB domains of KEAP1 and GSK-3b enzyme. The new STCs were synthesised in good yields of > 85%, with a purity of about 95%, using a novel synthesis method by employing a reusable proline–proline dipeptide catalyst. The STCs are more potent than 6-shogaol in activating NRF2 and reducing inflammation. The nature of substituents on thiophene has a profound influence on the bioactivity of the STCs. Phenylthiophene STC (STC5) is the most potent, while thiophenes containing electron-withdrawing groups showed weaker bioactivity. The bioactivity of 6-shogaol is in the micromolar range, whereas STC5 showed bioactivity in the sub micromolar range. The STCs showed anti-inflammatory effects via NRF2-dependent and NRF2-independent mechanisms. The STCs improved NRF2 activity through multiple (KEAP1-independent and -dependent) mechanisms. The STCs showed decreased reactivity with thiols than 6-shogaol and thus may possess fewer side-effects than 6-shogaol. The STCs were more metabolically stable than 6-shogaol.

Huvudfrågeställningen i studien var om Sjöfartshögskolan i Kalmar kursplaner uppfyller de krav som ställs i STCW del A-III/1 på utbildningsprogrammen för sjöingenjörer. Syftet med arbetet var att verifiera Sjöfartshögskolan i Kalmars kursplaners uppfyllelse av STCW-kraven enligt del A-III/1 för sjöingenjörer. Metoden som användes var en kvalitativ litteraturstudie där de i STCW uppställda kraven enligt del A-III/1 studerades och jämfördes med kursplanerna inom utbildningsprogrammet för sjöingenjörer på Sjöfartshögskolan i Kalmar.  Resultatet som framkom var att 72 % av kraven i A-III/1 var uppfyllda i kursplanerna för Sjöingenjörsprogrammet i Kalmar. Min rekommendation är att kursplanerna ses över och justeras för att bättre stämma överens med STCW-kraven, för att säkerhetsställa att kvaliteten på utbildningen är hög och konstant.

Abstract:

The purpose of this Master’s Thesis is to produce a specification for the aircraft conflict alert system STCA, and implement a prototype as a module in the air traffic surveillance system NOVA9000.

The specification is constructed based on functional requirements from EUROCONTROL and describes a system using a nominal trajectory method, where the future paths of aircraft are estimated. The trajectory is created using a probabilistic approach, where future positions are described with probability fields.

The prototype is implemented using the specification with some simplifications. The prototype is evaluated using recorded traffic from a heavy air traffic region surrounding an airport with parallel runways. 15 alerts were induced in 1,5 hour of morning traffic; this is far too much to be acceptable. Improvements are proposed and explanations to the high rate of alerts are made.

Under hösten 1997 och våren/hösten1998 har ett pilotprojekt bedrivits i Linköpings kommun. Det är drivet av Kungliga Vetenskapsakademien och Kungliga Ingenjörsakademien och handlar om ett amerikanskt undervisningsmaterial inom naturvetenskap och teknik. På engelska heter det"Science and Technology for Children"vilket givit namnet STC. Det är ett temaarbete med 24 olika teman att använda under de 6 första skolåren.

För att använda materialet i skolan har vissa utvalda delar översatts till svenska. Det är endast en direktöversättning och inget speciellt anpassat till den svenska skolan. Detta arbete har jag gjort för att undersöka hur det går att använda STC materialet Motion and Design enligt våra svenska styrdokument. Jag har även tittat på varför lärare fastnar för det.

Resultatet visar att det fungerar relativt bra utifrån de förutsättningar materialet hade när det provades. I framtiden kan vissa förändringar ske utifrån utvärderingar och respons från lärare. Materialet är väldigt tilltalande enligt konceptet att allt kommer i en låda och är förberett.

Resumo: A Stanniocalcina-1 (STC1) humana é uma glicoproteína homóloga a Stanniocalcina (STC) originalmente identificada como um hormônio regulador da homeostase de cálcio em peixes. A STC1 humana secretada atua em diferentes processos fisiológicos incluindo a angiogênese, a hipóxia e, principalmente, a carcinogênese, demonstrando assim uma atividade abrangente. Atualmente não se conhece o receptor da STC1 e pouco se sabe sobre o mecanismo de ação e de entrada nas células dessa proteína. Assim, o objetivo desse trabalho foi investigar um candidato a receptor de membrana dessa proteína, o receptor de transferrina (TfR1), uma proteína transmembrana responsável pela absorção de ferro nas células. Esse receptor é provavelmente expresso por todas as células em diferentes níveis, em destaque em células do sistema hematopoiético, em células em divisão celular e células neoplásicas. Assim, avaliou-se por citometria de fluxo o efeito do tratamento com STC1 em células não transfectadas e células transfectadas superexpressando o receptor de transferrina. Células tratadas com STC1 demonstraram um efeito semelhante ao tratamento com transferrina, um conhecido ligante desse receptor, no qual ambos diminuíram o número de células positivas para a marcação da superfície com transferrina conjugada com fluorocromo (transferrina-Alexa Fluor® 488 - Life Technologies). Em outro conjunto de experimentos de Western Blot foi demonstrado que a STC1 adicionada no sobrenadante das culturas de células é internalizada nas células e detectável no lisado celular, principalmente as células transfectadas para a superexpressão do receptor de transferrina. Complementarmente, em experimentos de localização subcelular por imunofluorescência a STC1 foi detectada em uma forma pontual e espalhada no citoplasma. Em conjunto, todos esses experimentos sugerem que STC1 e transferrina interferem na localização do receptor de transferrina na superfície celular e que possivelmente esse receptor está envolvido em mecanismos de internalização da própria STC1
Abstract: Human Stanniocalcin 1 (STC1) is the mammalian homologue of STC, which was originally identified as a calcium-regulating hormone in bony fishes. The human secreted Stanniocalcin acts on different physiological processes, including angiogenesis, hypoxia and especially carcinogenesis, facts that demonstrate their activity is wide. Currently there are few data on the mechanism of action of this protein or how it enters the cell. Thus, the aim of this study was to investigate transferrin receptor (TfR1) as a candidate to membrane receptor protein of STC1. This receptor is a membrane protein responsible for the iron uptake in cells. This receptor is probably expressed by all cells especially by cells in division and cancer cells, but its expression level may vary. We evaluated by flow cytometry the effect of STC1 treatment in non-transfected cells and cell with TfR1 overexpression. The treatment with STC demonstrated a similar effect to treatment with transferrin, a known ligand for receptor, which decreased the number of positive cells for staining with fluorochrome (transferrin conjugated to Alexa Fluor® 488 - Life Technologies). We also demonstrated by Western Blot that STC1 added to the supernatant of cultures of cells, especially cells that overexpress transferrin receptor, is internalized into the cells and detectable in the cell lysate. Additionally, in subcellular localization experiments by immunofluorescence STC1 was detected in a timely manner and scattered in the cytoplasm. Together all this information suggests that STC1 and transferrin interferes with the localization of the transferrin receptor in the cellular surface and perhaps this receptor is involved in the mechanism of internalization of STC1

Denna rapport är en jämförande studie av sjukvårdsutbildningen MedicalCare och hur den fungerar i verkligheten. Till sjöss är ”läkaren” en sjöman, med endast ett fåtal veckors sjukvårds-utbildning bakom sig och en erfarenhet som inte går att jämföra med den samlade sakkunskap som finns på ett sjukhus iland. Det ställs snarlika krav på sjömannen ombord som på läkaren iland vad gäller omhändertagande av den skadade. Internationellt sett finns det en konvention som styr hur MedicalCare-utbildningen skall vara konstruerad. Dessa direktiv är satta för att sjömän världen över skall ha samma riktlinjer att utbilda sig efter och få en likvärdig kompetens när kursen är genomförd. Studien har använt sig av en kvalitativ intervjustudie för att samla in information. Resultatet av denna undersökning visar att MedicalCare-kursen skiljer sig åt mellan skolorna, trots att alla arbetar efter samma regler.
This study is a comparison of how the MedicalCare-education works in reality. At sea the “Doctor” is a sailor with only a few weeks of medical training, with an experience not comparable with the expertise found in a hospital ashore. However he or she is faced with almost the same demands and responsibilities when it comes to nurse for the wounded person. Internationally there is a convention that states how the MedicalCare-education shall be constructed. These directions are set because all the sailors’ world wide shall have an equivalent education and the same guidelines to the structure of their training, for them to reach equal competence when the education is fulfilled. This study has collected information by conducting qualitative interviews This survey shows that although the demands are the same world wide, the education differs between the countries.

Transmission Control Protocol (TCP) is the dominant transport protocol today and likely to be adopted in future high‐speed and optical networks. A number of literature works have been done to modify or tune the Additive Increase Multiplicative Decrease (AIMD) principle in TCP to enhance the network performance. In this work, to efficiently take advantage of the available high bandwidth from the high‐speed and optical infrastructures, we propose a Stratified TCP (STCP) employing parallel virtual transmission layers in high‐speed networks. In this technique, the AIMD principle of TCP is modified to make more aggressive and efficient probing of the available link bandwidth, which in turn increases the performance. Simulation results show that STCP offers a considerable improvement in performance when compared with other TCP variants such as the conventional TCP protocol and Layered TCP (LTCP).

Abstract:
Compact heterochromatin is essential for genome stability and hence cell survival. Studies in many organisms including humans underline the importance of pericentromeric heterochromatin in centromere function. Fission yeast centromeres share a common structural organisation with those of their metazoan counterparts. The fission yeast model has been pivotal in understanding many key events in the pathway leading to the assembly of pericentromeric heterochromatin. In particular, studies in this system have revealed that the RNA interference (RNAi) pathway connects with the chromatin modification machinery to impart proper heterochromatin formation. Transcription of the pericentromeres by RNA polymerase II (Pol II) produces double stranded RNA (ds RNA) which is processed by Dicer(Dcr1) into small interfering RNAs (siRNAs). These siRNAs are loaded onto the Argonaute protein Ago1, and target the Ago1- containing RITS (RNA-Induced Transcriptional Silencing) complex to the pericentromeres via complementary base-pairing of the siRNA to the nascent centromeric transcript. RITS then recruits the sole Histone H3-K9-methyl transferase, Clr4, as part of the Clr4-complex, CLRC. The resulting H3K9-methyl marks further result in the recruitment of downstream chromatin binding proteins including the HP1- homolgue Swi6 which plays a key role in cohesin retention. Additionally, the H3K9- methyl marks are required for stabilising the association of CLRC and RITS, thereby promoting a reinforcing loop within the RNAi-mediated heterochromatin pathway. Thus crosstalk between RITS and CLRC is important in establishing and maintaining silent chromatin at the pericentromeres. Stc1 has been proposed to act as a critical link that connects the RITS and CLRC complexes. Stc1 is required for heterochromatin establishment and maintenance at the pericentromere and association of RITS with CLRC is lost in the absence of Stc1. Moreover, Stc1 directly interacts with Ago1 and is essential for siRNA production. These and other previous observations (Bayne et al. 2010) highlight the key role played by Stc1 in the RNAi-mediated heterochromatin pathway. To understand how Stc1 mediates the specific cross-talk between RNAi and chromatin modification, I have investigated the nature of Stc1 interactions with the RNAi and chromatin modification machineries. Using in-vitro binding assays, I found that Stc1 directly interacts with the CLRC subunits Dos2 and Clr4. I also identified the RITS subunit Tas3 as a potential interactor of Stc1, in addition to Ago1. A collaborating research group elucidated the structure of Stc1 using NMR (He et al. 2013) and my study provides evidence for interactions via the distinct domains of Stc1. Stc1 utilises its disordered C-terminus to bind to Dos2 while the N-terminus, which contains a tandem zinc finger domain, acts as a multi-protein interaction interface binding the CLRC subunit Clr4 and RITS subunits Ago1 and Tas3, opening up possibilities for Stc1-containing distinct-complexes. My work provides new insights into the role of Stc1 and opens up future avenues of research key to understanding how heterochromatin domains are defined and maintained.

Den svenska skolan har sedan en tid haft bristande natur- och teknikundervisning i de lägre årskurserna. Den Kungliga Vetenskapsakademien gav vissa av Linköpings skolor uppdraget att i lärarlag arbeta med ett nytt material, för att kunna se dess fördelar och brister. Materialet heter Science and Technology for Children (STC) och är framtaget av den amerikanska organisationen National Science Resource Center. Sedan utvärderingen av STC har läromedlet bytt namn till NTA (Natur och Teknik för Alla).

Den Kungliga Vetenskapsakademien bad också ett antal lärarkandidater på Linköpings lärarhögskola att hjälpa till med utvärderingen. I denna uppsats behandlas ett av de tre teman som utvärderas i Linköping, nämligen Plant growth and Development, ett tema där man får följa en växts hela livscykel.

Uppsatsen är en studie av de utvärderande lärarlagens arbete. En enkät har delats ut för att kartlägga lärarnas attityder till läromedlet. Vissa åsikter har registrerats vid personliga samtal. Syftet är att se om lärarna tycker att STC är något för den svenska skolan, och om användandet av STC kan motiveras av läroplanen, Lpo94.

En los años 90 el Perú experimentó una de las más grandes reformas en el sector eléctrico, pasando de un mercado bajo características de un “monopolio estatal verticalmente integrado” a uno de “competencia mayorista”, debido a que el anterior modelo poseía numerosas deficiencias se vio por necesario abrir camino a uno que fomente la inversión privada y que pueda alinear los interés privados con los estatales mediante la regulación. Es así que nace el COES como una entidad operadora del sistema encargada principalmente de ordenar el despacho diario de energía al mínimo costo entre las generadoras y, posteriormente, determinar y valorizar las inyecciones de potencia y energía activa realizada al sistema. De esta manera, en el presente informe analizaremos el Expediente N° 66-2012-STCOSINERGMIN la cual aborda una problemática surgida en el ámbito de ejercicio de funciones del COES como operador del sistema y OSINERGMIN como Organismo Regulador del sector eléctrico. Por lo cual, recurrimos al análisis tanto de la normativa aplicable durante los hechos acontecidos como de doctrina autorizada a efectos de tener un panorama completo del caso y poder concluir que las valoraciones emitidas por el COES son actos administrativos vinculantes a CASAPALCA y que el mecanismo para hacer efectivo el derecho de crédito creado a favor de ELECTROPERÚ no es la vía de solución de controversias ejercida por OSINERGMIN.
In the 90s, Peru underwent one of the largest reforms in the electricity sector, moving from a market with the characteristics of a "vertically integrated state monopoly" to one of "wholesale competition" because the previous model had numerous deficiencies. It was necessary to open the way to one that encourages private investment and can align private and state interests through regulation. Thus, the COES was born as an operating entity of the system in charge of ordering the daily dispatch of energy at minimum cost among the generators and, later, determining and valuing the injections of power and active energy made to the system. In this way, in this report, we will analyze Resolution No. 66-2012-STC-OSINERGMIN which addresses a problem that arose in the field of exercise of functions of the COES as system operator and OSINERGMIN as Regulatory Body of the electricity sector. Therefore, we resort to the analysis of both the applicable regulations during the events that occurred and the authorized doctrine to have a complete overview of the case and to be able to conclude that the assessments issued by the COES are administrative acts binding on CASAPALCA and that the mechanism for making the credit right created in favor of ELECTROPERU effective is not the means of dispute resolution exercised by OSINERGMIN.

The major driver for broadband wireless communications has been reliable, high-data rate services. In wireless communication, the multipath fading constitutes a bottleneck for increasing data rates and causes performance degradation. To combat fading, we can use diversity. Wireless systems with multiple antennas at the transmitter and receiver have much larger capacity in fading channels than standard wireless systems. The objective of this thesis is to investigate the transmission scheme provided by matrix A and B in the 802.16 standard and show how it can be implemented. The research focuses on using maximal-ratio combining (MRC) to demodulate the transmitted symbols. Modifications to the existing matrix by using more frequency bands were introduced; this reduces the number of transmitting antennas and uses fewer time slots to transmit the same number of symbols. The modulator and demodulator design is also discussed. The performance of orthogonal and non-orthogonal space time codes (STC) are evaluated.

STCW 78 as amended is the most comprehensive source of information regarding mariners' knowledge, understanding, and professions (KUPs), and is a reliable reference for curriculum design in maritime education and training institutions (METIs). However, the changing regulatory and technological shipping landscape requires continuous assessing the adequacy of current training requirements. Therefore, this study reviews safety, quality, and environmental management systems (SQEMS) from two large shipping companies. From the review, the expected seafarers’ skills are deduced, then compared with the STCW 78 as amended requirements. The results of this comparison reveal some insufficiencies in the KUP requirements mainly due to the STCW attitude, approach, and focus. STCW attitude refers to the low reactivity of STCW, lack of details in expressing KUPs, and unbalanced details for different KUPs. The STCW approach to defining KUPs for different ranks is based on the ship's functions. Although this approach is comprehensive, its rigidity results in some skills in the interface of different ship functions being overlooked or not being sufficiently emphasized. STCW focus is primarily on safety, by highlighting the details of safety-related codes and conventions, procedures, and equipment, while in response to environmental features, it mentions only "protection of the marine environment", without explicit reference to environmental conventions, machinery, inspections, or procedures. Consequently, the study highlights KUPs weaknesses and recommends changes to improve training.
