Abstract:
Phosphoinositide 3-kinases (PI3Ks) are a family of proteins involved in many different aspects of cell signaling. To date, eight different human PI3K isoforms have been identified, and distinct roles are beginning to emerge for each family member. Statins, HMG co-A reductase inhibitors used clinically to lower LDL cholesterol levels, also act through the PI3K signaling pathway to regulate cholesterol independent of their lipid-lowering effects. In an effort to discover the role of pl 10f3 in mediating non-lipid lowering effects of pravastatin, a mutant of p110(3 was overexpressed in human coronary artery endothelial cells (HCAEC) to form a dominant negative model (p110(3 DN). Silence si-RNA as an alterative tool was also optimized to diminish p110(3 protein expression successfully. HepG2 3: RE was used to monitor statins function by assaying luciferase expression. Results from these studies will determine the contribution of p110f3 in mediating selective cellular responses to statin.

ENGLISH ABSTRACT: Background Statins are drugs of known efficacy in the treatment of hypercholesterolaemia. However, statin-induced myopathy, an adverse effect of statins in up to 15% of its users, has warranted a reduction in the prescription dose or discontinuation of the drug. The exact mechanism of statin-induced myopathy is unknown, but the potential of Coenzyme Q10 (CoQ10) as treatment has been recognized due to decreased human plasma CoQ10 levels found after statin use and the concomitant role of CoQ10 in muscle function. Objectives This systematic review assessed the effect of CoQ10 supplementation on: the severity of statin-induced myopathic symptoms, levels of plasma creatine kinase, intramuscular and plasma CoQ10, as well as whether any adverse effects of CoQ10 supplementation such as abdominal pain, nausea and vomiting or headaches were experienced. Search methods Two searches for studies were conducted in The Cochrane Central Register of Controlled Trials (inception to March 2011 and inception to November 2011), MEDLINE (inception to March 2011 and inception to November 2011), Web of Science (inception to March 2011 and inception to November 2011), Science Direct (inception to March 2011 and inception to February 2012), Wiley Online Library (inception to March 2011 and inception to February 2012), Springerlink (inception to April 2011 and inception to February 2012), EBSCOhost [Academic Search Premier and CAB abstracts (inception to March 2011 and inception to February 2012), CINAHL (inception to March 2011 and inception to November 2011)], Scopus (inception to March 2011 and inception to November 2011) and Google Scholar (inception to March 2011 and inception to February 2012). Reference lists of articles were hand searched for relevant clinical trials. Only trials with a full text were included in the review. Selection criteria Randomised controlled trials (RCTs) were included with adult participants (mean of 18-64.99 years) of all race/ethnic groups and gender on statin therapy with reported myopathic symptoms from an unknown cause. The intervention was in the form of a pure oral supplement of CoQ10 irrespective of dose, duration and frequency, and the control in the form of a placebo, a similar antioxidant, or no intervention. Outcomes included the severity of myopathic symptoms, levels of plasma creatine kinase (U/L), intramuscular CoQ10 (μmol/kg) and plasma CoQ10 (μmol/L), as well as adverse effects of CoQ10. Data collection and analysis The principle investigator and one independent reviewer selected the studies, extracted data and assessed for risk of bias using the Cochrane Collaboration‘s tool for assessing risk of bias. Authors of relevant clinical trials were contacted for additional information. Results Two RCTs were included in the review, totaling 76 participants. A meta-analysis could not be performed, thus the review is narrative. There were an insufficient number of RCTs to confirm whether routine supplementation of CoQ10 improves statin-induced myopathic symptoms. Conclusions More and larger RCTs are required to determine the efficacy of CoQ10 supplementation in statin-induced myopathy. Consensus needs to be reached regarding the definition and measurement instrument/s of myopathy so that results of future studies can easily be compared and synthesized.

[Truncated abstract] The metabolic syndrome is characterized by cardiovascular risk factors including dyslipidemia, insulin resistance, visceral obesity, hypertension and diabetes. The dyslipidemia of the metabolic syndrome includes elevated plasma triglyceride and apolipoprotein (apo) B levels, accumulation of small, dense low-density lipoprotein (LDL) particles and low high-density lipoprotein (HDL) cholesterol concentration. However, the precise mechanisms for this dyslipoproteinemia, specifically low plasma HDL cholesterol, are not well understood. This thesis therefore, focuses on HDL, its structure, function and metabolism. However, lipoprotein metabolism is a complex interconnected system, which includes forward and reverse cholesterol transport pathways. Hence, this thesis also examines and discusses the metabolism of apoB-containing lipoproteins. This thesis tests the general hypothesis that apolipoprotein kinetics are altered in the metabolic syndrome, and that lipid regulating therapies can improve these kinetic abnormalities. The aims were first, to compare and establish the clinical, metabolic and kinetic differences between metabolic syndrome and lean subjects; and second, to determine the regulatory effects of statin therapy, specifically, rosuvastatin on lipoprotein transport in the metabolic syndrome. Five observation statements were derived from the general hypothesis and examined in the studies described below. The findings are presented separately as a series of original publications. Study 1 Twelve men with the metabolic syndrome and ten lean men were studied in a case-control setting. ... These findings explain the HDL raising effects of rosuvastatin in the metabolic syndrome. Collectively, these studies suggest that the dyslipidemia of the metabolic syndrome results from increased production rates of VLDL and LDL particles, reduced fractional catabolic rates of these lipoproteins, together with accelerated catabolism of HDL particles. Treatment with rosuvastatin increases the catabolic rates of all apoB-containing lipoproteins and at a higher dose, decreases LDL apoB production. These effects are consistent with inhibition of cholesterol synthesis leading to an upregulation of LDL receptors. Rosuvastatin decreases the fractional catabolism of HDL particles. The effects of rosuvastatin on HDL kinetics may be related to a reduction in triglyceride concentration and cholesterol ester transfer protein activity. These findings are consistent with the general hypothesis that apolipoprotein kinetics are altered in the metabolic syndrome, and that statin therapy improves these kinetic abnormalities.

Abstract:
Background: Catheter ablation for atrial fibrillation (AF) is more efficacious than antiarrhythmic therapy. Post ablation recurrences reduce ablation effectiveness and are contributed by lesion discontinuity in the fibrotic linear ablation lesions. The anti-fibrotic role of statins in reducing AF is being assessed in current trials. By reducing the chronic pathological fibrosis that occurs in AF they may reduce AF. However if statins also have an effect on the acute therapeutic fibrosis of an ablation, this could exacerbate lesion discontinuity and AF recurrence. We tested the hypothesis that statins attenuate ablation lesion continuity in a recognised pig atrial linear ablation model. Aims: To assess whether Atorvastatin diminishes the bi-directional conduction block produced by a linear atrial ablation lesion. Methods: Sixteen pigs were randomised to statin (n=8) or placebo (n=8) with drug pre-treatment for 3 days and a further 4 weeks. At initial electrophysiological study (EPS1) 3D right atrium (RA) mapping and a vertical ablation linear lesion in the posterior RA with bidirectional conduction block were completed (Gepstein Circ 1999). Follow-up electrophysiological assessment (EPS2) at 28 days assessed bidirectional conduction block maintenance. Results: Data of 15/16 (statin=7) pigs were analysed. Mean lesion length was 3.7 ± 0.8cm with a mean of 17.9 ± 5.7 lesion applications. Bi-directional conduction block was confirmed in 15/15 pigs (100%) at EPS1 and EPS2. Conclusions: Atorvastatin did not affect ablation lesion continuity in this pig atrial linear ablation model. If patients are on long-term statins for AF reduction, periablation cessation is probably not necessary.

In this thesis, we trained a reinforcement learning agent using one of the most recent policy gradient methods, proximal policy optimization, in a first-person shooter game with a static player. We investigated how curriculum learning can be used to increase performance of a reinforcement learning agent. Two reinforcement learning agents were trained in two different environments. The first environment was constructed without curriculum learning and the second environment was with curriculum learning. After training the agents, the agents were placed in the same environment where we compared them based on their performance. The performance was measured by the achieved cumulative reward. The result showed that there is a difference in performance between the agents. It was concluded that curriculum learning can be used to increase the performance of a reinforcement learning agent in a first-person shooter game with a static player.
I denna uppsats tränade vi en agent genom förstärkt djupinlärning med hjälp av en av de senaste gradientmetoderna, nämligen proximal policy optimization, i ett förstapersonsskjutspel med en statisk spelare. Vi undersökte hur läroplanering kan användas för att öka prestandan hos en agent som tränats med förstärkt inlärning. Två agenter tränades i två olika miljöer. Den första miljön använde inte läroplanering och den andra miljön använde läroplanering. Efter att ha tränat agenterna placerades de i samma miljö. Deras prestation mättes genom deras kumulativa belöning. Reslutatet påvisade att det finns en skillnad i prestanda mellan agenterna. Genom att använda läroplanering i ett förstapersonsskjutspel med en statisk spelare kunde prestandan hos en agent som tränats med förstärkt inlärning öka.

La modellazione di agenti che tiene conto di emozioni e stati affettivi costituisce un argomento di discussione piuttosto importante nell’ambito della simulazione ad agenti, soprattutto per via di come introdurre parametri affettivi nella modellazione possa contribuire a rendere le simulazioni più realistiche. In questo ambito di ricerca, però, il modo di introdurre nei modelli parametri in grado di regolare lo stato affettivo degli agenti per da influenzarne azioni e comportamenti è spesso basato sui modelli emozionali che si trovano in letteratura, oppure sulle teorie e i modelli fisici che vengono solitamente utilizzati per la modellazione di pedoni e folle. L’approccio presentato in questo lavoro, quindi, mira ad approcciare il problema dal punto di vista dei dati, puntando ad arrivare alla modellazione di agenti affettivi partendo da dati provenienti da persone reali ed acquisiti tramite esperimenti creati ad hoc con il preciso obiettivo di studiare reazioni e comportamenti da poter poi tradurre in modellazione. In particolare, in questo lavoro il problema viene affrontato concentrandosi in particolare sull’ambito pedonale, osservando diversi tipi di interazione coinvolgenti pedoni tramite quattro diversi esperimenti atti a raccogliere dati in grado di descrivere le interazioni operate dai soggetti per poi inserirle in un contesto di modellazione. Gli esperimenti vengono effettuati in vivo, in vitro e online, osservando le interazioni di pedoni con veicoli, ostacoli in movimento ed altri pedoni, raccogliendo dati riguardo queste diverse interazioni tramite dati fisiologici e questionari atti a profilare i partecipanti e a fornire maggiori informazioni riguardo al comportamento e alle reazioni da loro dimostrate. I dati raccolti vengono quindi utilizzati per la modellazione, prima in ambito di automi cellulari e poi, successivamente, nell’ambito dei sistemi multi-agente, mostrando come le informazioni ricavate dai dati vengano integrate all’interno dei modelli al fine di includere parametri affettivi che, in base ai valori assegnati, influenzino in un certo modo il comportamento degli agenti. Vengono poi proposte alcune simulazioni derivanti dai modelli, ai fini di osservare come i parametri affettivi introdotti influenzino il comportamento degli agenti in azione in determinate situazioni.
The modeling of agents involving emotions and affective states constitutes a relevant discussion topic in the research concerning multi-agent simulations, especially because of how the introduction of affective parameters inside the modeling process could effectively make the produced simulations more realistic. In this research area, though, the modality in which parameters regulating the affective state of agents are introduced into models, so that the agents’ behaviour and actions are influenced by them, is always based on emotional models found in literature, or on physics theories and models usually involved for the modeling of pedestrians and crowds. The approach this work presents, then, aims at tackling this problem from the point of view of data, thus wanting to get to affective agent modeling starting from data coming from real people, acquired through ad-hoc experiments with the precise goal of observing reactions and behaviour to be later translated inside a model. In particular, the focus of this work falls on the research on pedestrians and walkability, observing different types of interactions involving pedestrians through four different experiment through which gather data able to describe the participants’ interactions to then implement them in the modeling step. The proposed experiments are executed in-vivo, in-vitro and online, observing pedestrian interactions with vehicles, moving obstacles and other pedestrians, gathering data regarding these interactions through physiological data and questionnaires made for profiling purposes and in order to have more information regarding the subjects’ behaviour and reactions. The gathered data is then used for modeling, firstly from the point of view of cellular automata and then passing on to the multi-agent systems perspective, showing how the information obtained from the data is introduced inside the models to be parametrized in affective parameters that, depending on the assigned values, could influence in a certain way the behaviour of the agents. After that, some simulation instances derived from the models are presented, as to observe how the affective parameters that were introduced in the models actively influence the behaviour of agents acting and moving in certain situations.

Please see the pdf version of the abstract for an accurate reproduction.] Atherosclerosis is a common degenerative disease in which the clinical manifestations are often through stroke or myocardial infarction. Some of the established risk factors for atherosclerosis include elevated plasma low-density lipoprotein (LDL)-cholesterol levels, obesity, diabetes mellitus (DM) and cigarette smoking. Of the risk factors, an elevation in plasma LDL is one of the most established and the most researched. This is partly a consequence of the deposition of cholesterol within arterial intima being a crucial step in the progression of atherosclerosis, combined with the finding that LDL particles are a major transporter of cholesterol in circulation. Recently there is increasing evidence showing a role of the other major transporter of cholesterol in circulation, chylomicron remnants, in the progression of atherosclerosis. The notion of atherosclerosis as a postprandial phenomenon has been further substantiated by the emergence of evidence showing a direct role of chylomicron remnants in arterial cholesterol deposition. Based on evidence that chylomicron remnants are proatherogenic, the suggestion arises that accumulation of postprandial lipoproteins in plasma may add another dimension of risk to the development of coronary artery disease (CAD). This thesis tests the general hypothesis that individuals with or at high risk of CAD have postprandial dyslipidaemia and that this metabolic abnormality is correctable with a class of lipid-lowering drugs called statins. To test the hypothesis, clinical studies were conducted in normolipidaemic CAD patients, heterozygous familial hypercholesterolaemia (FH) and postmenopausal women with type 2 DM. Determination of postprandial dyslipidaemia by comparison with control populations were conducted initially in each patient group (Studies 1, 3 and 5), followed by intervention studies investigating possible modulation of the dyslipidaemia with a statin (Studies 2, 4 and 6). Six observation statements based on case-control comparisons of postprandial lipaemia in patients with or at risk of CAD and the effects of statins on postprandial dyslipidaemia in the patient groups were derived from the general hypothesis. The observation statements were examined in the individual studies described below. Postprandial lipoprotein metabolism was assessed using a number of methods. For comparison of postprandial lipaemia in Studies 1 and 2, a classic oral fat challenge was utilised. As markers of chylomicrons and chylomicron remnants, retinyl palmitate and triglyceride were measured postprandially as well as apolipoprotein (apo) B48 concentrations, a specific marker of intestinal lipoproteins. ApoB48 was also measured in the fasting state and found to predict the postprandial responses of retinyl palmitate, triglyceride and apoB48. This suggested that fasting measurement of apoB48 could be used as a simple indicator of postprandial dyslipidaemia. Consequently for Studies 3 - 6, fasting apoB48 measurements were used as primary markers of postprandial dyslipidaemia. Other markers for chylomicrons and their remnants utilised were fasting plasma concentrations of remnant-like particle-cholesterol (RLP-C) and apoC-III. As well as these static markers, chylomicron remnant catabolism was measured using a stable isotope breath test. The breath test involves the intravenous injection of a chylomicron remnant-like emulsion labelled with ¹³C-oleate and measurement of enriched ¹³CO2 in expired breath by isotope ratio mass spectrometry. The fractional catabolic rate (FCR) of the injected emulsion was subsequently calculated using multi-compartmental modeling (SAAM II). The studies are presented in this thesis as published and unpublished works. In Study 1, postprandial lipoprotein metabolism was compared between 18 normolipidaemic CAD patients (cholesterol 4.54 ± 0.12 mmol/L, triglyceride 1.09 ± 0.16) with 13 asymptomatic healthy controls using an oral fat challenge. Normolipidaemic CAD patients had higher postprandial area-under-curve (AUC) for triglyceride (+34%, p=0.019), retinyl palmitate (+74%, p=0.032) and apoB48 (+36%, p<0.001). Fasting apoB48 was also higher (+41%, p=0.001) and found to correlate significantly with AUC of triglyceride (p=0.017), retinyl palmitate (p=0.001) and apoB48 (p<0.001). The data suggest that normolipidaemic CAD patients have increased concentrations of intestinal lipoproteins in the fasting and postprandial state. In addition to these findings, significant correlations of fasting apoB48 with postprandial markers (p<0.02) suggests the fasting marker to be a simpler surrogate marker for the degree of total postprandial lipaemia. Study 2 investigated the effect of atorvastatin treatment on postprandial dyslipidaemia found in the 18 near-normolipidaemic CAD patients from Study 1. The trial was conducted in a randomised, placebo-controlled design, using oral fat challenges before and after 12-weeks atorvastatin/placebo treatment. Compared with the placebo group, atorvastatin decreased the total postprandial AUC for iii triglyceride (-22%, p=0.05) and apoB48 (-34%, p=0.013). Fasting markers of apoB48 (-35%, p=0.019) and RLP-C (-36%, p=0.032) also decreased significantly. Atorvastatin was also found to increase LDL-receptor activity by +218% (p<0.001) as reflected in binding studies. The data suggest atorvastatin reduces the fasting levels of intestinal lipoproteins as well as total postprandial lipaemia, but without acute dynamic changes in postprandial lipaemia. The reduction in fasting and total postprandial lipoprotein levels could be partly attributed to an increase in LDL-receptor mediated removal from circulation. In Study 3, postprandial lipaemia was compared in 15 heterozygous FH patients with 15 healthy controls. FH patients had higher fasting concentrations of apoB48 (+56%, p<0.001) and RLP-C (+48%, p=0.003). The elevation in these fasting markers of chylomicrons and their remnants suggests FH patients have postprandial dyslipidaemia due to an accumulation of these particles in plasma. Study 4 examined the effects of long- (> 6 months) and short-term (4 weeks) simvastatin treatment on modulating postprandial dyslipidaemia found in the 15 FH patients from Study 3. Short- and long-term simvastatin treatment decreased the fasting concentrations of apoB48 (-29% and 15% respectively, p<0.05) and RLP-C (both -38%, p<0.001), but did not significantly alter the FCR of the injected chylomicron remnant-like emulsion. The data suggest that in heterozygous FH both long- and short-term simvastatin treatments decrease the fasting markers of postprandial lipoproteins by mechanisms that may not be mediated via processes differentiated by the 13CO2 breath test. This implies that the effect on postprandial lipaemia may be from a decrease in production and/or a possible increase in catabolism of triglyceride-rich lipoproteins (TRLs). In Study 5, postprandial lipaemia was compared in 24 postmenopausal women age and body mass index matched with 14 postmenopausal women with type 2 DM. Postmenopausal diabetic women were found to have higher fasting concentrations of apoB48 (+21%, p=0.021) and apoC-III (+16%, p=0.042) as well as lower FCR of the chylomicron remnant-like emulsion (-50%, p<0.001). The data suggest that postmenopausal diabetic women have postprandial dyslipidaemia, and that this is due to delayed catabolism of chylomicron remnants. Study 6 was an hypothesis-generating exercise examining the effects of 4-weeks pravastatin treatment on postprandial dyslipidaemia found in 7 postmenopausal women with type 2 DM from Study 5. Although plasma LDL-cholesterol was reduced (-19%, p=0.028), there were no significant effects found on fasting apoB48 concentrations (-12%, p=0.116) or the FCR of the chylomicron remnant-like emulsion (+38%, p=0.345). A larger sample size of patients and/or treatment with a more potent statin at a dosage known to affect chylomicron remnant metabolism would be required to demonstrate a significant reduction in postprandial dyslipidaemia in postmenopausal women with type 2 DM. The results of the above mentioned studies combined support the general hypothesis that postprandial dyslipidaemia is a feature of patients with or at risk of CAD. This defect may be demonstrated using fasting apoB48 as an indicator of the degree of postprandial lipaemia. Postprandial dyslipidaemia may reflect a reduction in catabolism, as suggested with the breath test in type 2 DM, and/or an over overproduction of chylomicrons. Both these mechanisms would also increase competition for lipolysis and clearance pathways between hepatically and intestinally-derived lipoproteins. The exact mechanisms by which postprandial dyslipidaemia occurs are yet to be determined. Statins appear to improve defective postprandial lipaemia in patients with or at risk of CAD, which is in agreement with the general hypothesis. The effectiveness of a statin is dependant on their potency in inhibiting cholesterol biosynthesis and increasing receptor mediated clearance of LDL and chylomicron remnants. The studies conducted in this thesis show that postprandial dyslipidaemia can be reduced by statins but not to the extent demonstrated in controls. However, the demonstrated reduction in fasting and total postprandial lipaemia translates to a lowering in overall arterial exposure to circulating proatherogenic particles. The elevation in fasting and postprandial levels of proatherogenic chylomicron remnants found in the patient groups described in this thesis indicates another dimension to their risk of coronary disease. The reductions in the overall levels of proatherogenic particles in patients with or at high CAD risk, infers a possible reduction in the risk of coronary disease in these patients.

In recent years Major League Baseball (MLB) has seen a trend of long term multimillion dollar contracts. The purpose of this study was to examine Major League Baseball player’s performance before and after signing a free-agent contract. This research helps to explain the relationship between reward and performance in professional baseball players. Subjects include 65 major league baseball players that were free agents in the years of 2005 and 2006. The group of 65 players consisted of 34 position players and 31 pitchers. The individual statistical analysis of position players’ batting average, homeruns, and runs batted in were used. The statistics that were analyzed for the pitchers were earned run average, innings pitched, strikeouts, and wins and losses. The results that showed significance was batting average, homeruns, runs batted in, and earned run average. The results of this study can be a useful tool for the front office of Major League Baseball teams.

Russia has always been a primary factor in the development of the Baltic States. It is impossible to analyse Baltic security without looking into the processes inside Russia and the prevailing trends vis-a-vis the Baltic States. However, the changes in the Baltic security landscape in the 21st century lack a comprehensive analysis. This thesis seeks to bridge the gap. Two key aims are being pursued in this thesis. The first is to present an analysis of Russia's European agenda under President Vladimir Putin and to examine the place of the Baltic States in this agenda. The second aim is to define Russia-related threats and challengers to the Baltic States, as well as prospects in Russo-Baltic relations. To attain these aims, inter-active approach to international relations, comprising three levels of analysis - the international system, the nation state (domestic level) and the individual (personality) level - has been applied. The neo-realist paradigm of international relations theory, comparative analysis and the Knudsen model, which addresses the peculiarities of relations between great powers and small states, are the methodological framework of the thesis. When analysing the development of Russo-Baltic relations in 1990-2006, this thesis focuses on the evolution of the Baltic States from factors to actors and their chance of shaping Russo-Baltic relations from within the enlarged EU and NATO. It also examines possibilities for more active engagement of Russia in the Baltic region. The thesis concludes with an analysis of perspectives for the Baltic States in countering Russia-related threats and building cooperativer elations with Russia. The author maintains that 'high politics' in Russo-Baltic relations has ended, yet, the tensions do remain in 'low politics'. Russia seeks to retain her political and economic influence in the Baltics by exploiting various tools, primarily economic levers and Baltic dependence upon Russian energy.

Abstract:
Surgical myocardial revascularization remains the standard of care for patients with multi-vessel coronary artery disease. A growing body of evidence indicates that systemic inflammation and myocardial oxidative stress are associated with the development of postoperative atrial fibrillation (POAF) and low cardiac output syndrome in patients undergoing cardiac surgery. Statins have been shown to exert rapid anti-inflammatory and antioxidant effects by inhibiting myocardial NOX2 oxidases and by increasing the bioavailability of nitric oxide (NO). However, whether these so-called pleiotropic effects of statins result in improved patient outcomes remains to be established. To provide further insights into the mechanisms of action and impact on clinical outcomes of peri-operative statin treatment in patients undergoing cardiac surgery, I studied the molecular mechanisms underlying the myocardial nitroso-redox balance in samples of the right atrial appendages (RAA) obtained before (PRE) and after cardiopulmonary bypass (CPB) and reperfusion (POST) and setup two double-blind randomised placebo-controlled trials: 1) STARR (Statin Treatment on Atrial Refractoriness and Reperfusion injury), which tested the effect of Atorvastatin (80 mg once daily for up to 6 days before surgery and 5 days after) on the atrial effective refractory period (AERP, over 4 post-operative days) and superoxide production in paired PRE- and POST- RAA samples from 60 patients 2) STICS (Statin Treatment In Cardiac Surgery), which assessed the effects of peri-operative treatment with Rosuvastatin (20mg od) on POAF (assessed by continuous holter ECG monitoring for 5 days postoperatively) and myocardial injury (assessed by serial troponin I measurements) in 1922 patients undergoing elective cardiac surgery. I observed that atrial superoxide production increased significantly after reperfusion due to increased mitochondrial and NOX2 oxidase activity and to uncoupling of NOS activity. NOS activity in RAA samples decreased significantly after reperfusion (by 60%), but this reduction was not prevented by BH4 supplementation (10 μM) or NOX2 inhibition. Instead, I identified increased endothelial NOS S-glutathionylation as the main mechanism responsible for NOS uncoupling after reperfusion. In STARR, atorvastatin prevented increase in RAA superoxide production, maintained the functionally coupled status of NOS and NO bioavailability after reperfusion but had no measurable effect on postoperative AERP. In STICS, treatment with rosuvastatin significantly reduced LDL-C concentration by 48 hours after surgery but had no effect on the incidence of POAF (203 (21%) of the Rosuvastatinallocated patients vs. 197 (20%) of the placebo-allocated patients) or on perioperative myocardial damage (P = 0.80). Pre-defined subgroup analyses (age, sex, prior statin use, baseline troponin concentration, duration of randomized treatment before surgery, type of cardiac surgery, and postoperative use of anti-inflammatory drugs) did not identify any category of patient who benefited from perioperative rosuvastatin treatment. Nor were there beneficial effects on any of the other in-hospital clinical outcomes that were assessed. In conclusion, cardiac surgery on CPB is associated with myocardial nitroso redox imbalance that is reversed by perioperative intensive therapy with statins. However, these effects have no beneficial effects on common in-hospital complications after elective cardiac surgery. Although the benefits of long-term statin therapy in patients requiring myocardial revascularization are well established, the work presented in this thesis does not support routine use of perioperative intensive therapy with statins for the prevention of postoperative complications in patients undergoing elective cardiac surgery.

This project examines the scientific developments, strategic considerations, and political circumstances that led to the rise and fall of herbicidal warfare in Vietnam. The historical narrative draws on a wide range of primary and secondary source literature on the Vietnam War and the Cold War, the history of science, and American and international history of the 1960s and 1970s. The author conducted archival research in the United States in a variety government and non-government research facilities and toured formerly sprayed areas in Vietnam. This project utilizes oral history interviews of American and Vietnamese scientists who were involved in some aspect of the Agent Orange controversy. The thesis explains why American scientists were able to force an end to the herbicide program in 1971 and ensure that the United States would not engage in herbicidal warfare in the future. This political success can be understood only in the context of two major political transformations in the Vietnam Era: the collapse of Cold War containment as a salient model of American foreign policy, and the development of globally-oriented environmental politics and security regimes. The movement to end herbicidal warfare helped shift the meaning of security away from the Cold War toward transnational efforts to combat environmental problems that threaten all of the world's people.

This thesis develops a candidate-centred conception of American presidential nominations. Candidates' choices in nomination politics remain under-theorised. The literature on nominations has tended either to downplay the role of candidates' independent influence or to suggest that the impact of their choices is too idiosyncratic to theorize about. I reject both of these positions; and instead develop the basic elements of a theory in which candidates are the principal agents of change in nomination contests. I argue that candidates make distinct identity, tactical, and management choices, and I show that this simple frame can be used to connect aspirants' varying goals to their choices and actions. In my theory, candidates' prospects remain relatively stable unless a shift occurs in their competitive setting in response to an unexpected event - for instance, a surprising election result. These shifts, or critical junctures, define a candidate's path to his party's presidential nomination. I argue that the rival candidates' choices dominate the development of these critical junctures and, therefore, that candidates' choices are crucial to nomination outcomes. Structural factors, the actions of non-candidates and the effects of exogenous events, account for a minority of critical junctures. In the empirical chapters of this study, I examine the Democratic and Republican nomination contests in selected years before the McGovern-Fraser reforms (1912, 1924, 1932) and in post-reform cases (1972, 1976, 1980) to demonstrate the pervasive influence of candidates' choices in contrasting institutional settings. These cases confirm my basic claim about the centrality of candidates' choices and also suggest significant ways in which candidates' choices have changed between 1912 and 1980.

This thesis examines the motivations of bureaucrats in two government agencies: the Environmental Protection Agency in the US, and the Environment Agency for England and Wales. The model employed in this work is a Trifocal Model which utilises Rational Choice, Institutional and Cultural approaches in answering the thesis question. The aim of this work is two-fold: one aim is to explain motivations in two agencies; the second aim is to suggest why the existing literature in the field of bureaucracy often fails to capture the diversity of bureaucratic motivations. The claim is that the adherence to one particular paradigmatic approach prevents scholars from attaining a comprehensive understanding of motivations. This work focuses on two elements of the Trifocal Approach, namely institutional and cultural explanations. Rational Choice explanations are given a limited explanatory role in this work, in large part because of the restricted usefulness of an approach which takes the preferences of agents as given. This thesis uses a scientific approach to the analysis of qualitative data, allowing other researchers to make use of, and indeed to question, the findings presented below. The argument in this thesis suggests why scholars must pay more attention to what those people within bureaucracies tell us about themselves and their motivations. To take the preferences of agents as givens is to ignore much of what is most important about the study of politics that is, where preferences come from, and how they shape the political behaviour we observe in bureaucracies. This thesis will show that public sector reforms are often flawed, often failing to consider the interplay of cultural and institutional effects, and how these effects have a bearing on the motivations of staff in organisations undergoing reform. Furthermore, cultural and institutional factors must be considered whenever one considers the question what is it that motivates bureaucrats.

Abstract:
The purpose of this thesis was to examine two works of science fiction by the Polish writer and satirist, Stanislaw Lem, which exemplify two broad philosophical themes central to his writings; viz., cosmology and personhood. In "The New Cosmogony," Lem focuses on the central problem of the philosophy of scientific cosmology, namely, what kind of explanation is-applicable to that science. Lem synthesizes religious and scientific views of the creation of the universe in his model of the cosmos as a Game of Civilizations with advanced technologies. Central to this work is the philosophical notion of intentionality. In "The Mask," Lem raises questions about personhood, consciousness, freedom, determinism, and responsibility by embedding them in the autobiographical narrative of a conscious, female machine. Lem's views on cosmology and his views on personhood both involve intentionality - the intentions of civilizations, on the one hand, and the intentions of a King and of the machine he created, on the other. Both works challenge all preconceptions about philosophy through the use of paradox, irony, and sometimes ridicule. Lem provides no answers, however. He is interested in setting up futuristic technological situations that are capable of undermining our basic assumptions about the world.

Abstract:
Postal survey research was conducted between September and November, 1986, to gather information concerning the evolution, existence and extent of mental health services available to police personnel. Questionnaires were mailed to all 366 municipal, county, and state police agencies in the United States that employed 200 or more workers. Usable data were obtained from 76.8% of the agencies surveyed. Of the 281 respondents who returned usable data, 65.1% reported the existence of mental health services available to their police personnel. The majority of respondents (58.6%) perceived their mental health programs as being equally reactive and preventive in orientation. The most frequently reported existing components were outside agency counseling, stress management seminars, and testing of potential police recruits. Over half (54.8%) of the responding police agencies reported having between 10 and 19 components in their respective mental health programs. The implementation dates and evolution of twenty-five (25) components were examined, and specific components of various police agencies were also revealed. The majority of respondents (70.7%) reported their mental health programs were available to sworn and nonsworn personnel and their families. Almost all respondents (98.3%) viewed their programs as being cost effective. Also, most agencies were satisfied with the four treatment resources listed, which included in-house counseling, outside agency counseling, hospital in-patient programs, and alcohol/drug rehabilitation centers. Slightly over half (50.8%) of the respondents stated their service programs were entitled "Employee Assistance Program." Of the 300 staff workers holding mental health degrees, 101 were reported to have doctoral degrees in psychology. The most frequently reported personality theory utilized by staff members was eclecticism (48.5%). The prevailing high interest in police mental health services is discussed as well as possible reasons why some police managers may be apathetic towards the implementation of such services. Ways of educating police managers as to the benefits of police mental health programs are listed as well as topics for future research.

Historians of the American South have been diverse in their descriptions of the master-slave relationship over the last half-century, and have engaged in lengthy discussions in an attempt to answer the intricate question of what life was like between slaves and their masters. The phenomenon of slave runaways has perhaps offered the most convincing evidence of the troubles on southern plantations, which has been used in recent decades to emphasize negotiation and agency in the shaping of master-slave relations. The last twenty years have been consequently marked by a plethora of studies that accentuate non-traditional slave holding as it becomes clearer that masters had to compromise with their human chattel. Through an examination 9,975 runaway slave advertisements and 943 testimonies of former slaves, this study illustrates how black bondsmen absented themselves so to negotiate the terms of their working and living conditions. It traces the acts of individual slave runaways in place of broader generalizations that have for a long time contributed to some of the myths and legends of American slavery through examination of the many reasons that slaves chose to stay in bondage.

Abstract:
In this study, I examined the Supreme Court's agenda from 1801 to 1993 to determine the composition and dynamics of the issues that have dominated the business of the Court. Specifically, I set out to test empirically Robert G. McCloskey's (now standard) characterization of the Supreme Court's history, which sees it as dominated by nationalism/federalism issues before the Civil War, by economic issues just after the War through the 1930s, and by civil rights and liberties since the 1930s. The question that drove my investigation was "Is McCloskey's interpretation, which appears to be based on the great cases of Supreme Court history, an accurate description of the agenda represented in the Supreme Court's total body of reported decisions?" To test McCloskey's historical theses I employed concepts adapted from Richard Pacelle's (1991) important work on the agenda of post-Roosevelt Court and used the methods of classical historical analysis and of interrupted time-series analysis. Data for my research came from existing datasets and from my own collection (I coded the manifest content of thousands of Supreme Court's decisions from 1887 back to 1801). The most important finding from my analyses is that McCloskey not withstanding, the pre-Civil War Supreme Court's agenda was clearly dominated by economic issues of various sorts, not by nationalism/federalism as previously believed. Another key finding is that partisanship had a pronounced impact on the Court's attention to this category of issueseven in the periods when the Supreme Court had very little control of its docket. These results suggest that Supreme Court scholars should reassess or rethink their previous notion of the Court's pre-Civil War agendathe now well-established view that nation-state issues dominated the business of the Court in its formative yearsand the idea (often expressed implicitly) that the Court's mandatory jurisdiction suppressed attitudinal factors on the Court in the earlier eras.

Abstract:

Individual burials are always representative of both individuals and collective actors. The physical remains, material culture, and represented practices in burials can be used in concert to study identities and social personas amongst individual and collective actors. These identities and social personas are the result of the interaction between agency and structure, where both individuals and groups act to change and reproduce social structures.

The three burials upon which this study is based are currently held in the collections of the Milwaukee Public Museum. They are all indigenous burials created in Wisconsin in the 19th century. Biological sex, stature, age, and pathologies were identified from skeletal analysis and the material culture of each burial was analyzed using a Use/Origin model to attempt to understand how these individuals negotiated and constructed identities within a colonial system.
