Abstract:
The purpose of the PhD research was the identification of new strategies of farming and processing, with the aim to improve the nutritional and technological characteristics of poultry meat. Part of the PhD research was focused on evaluation of alternative farming systems, with the aim to increase animal welfare and to improve the meat quality and sensorial characteristics in broiler chickens. It was also assessed the use of innovative ingredients for marination of poultry meat (sodium bicarbonate and natural antioxidants) The research was developed by studying the following aspects: - Meat quality characteristics, oxidative stability and sensorial traits of chicken meat obtained from two different farming systems: free range vs conventional; - Meat quality traits of frozen chicken breast pre-salted using increasing concentrations of sodium chloride; - Use of sodium bicarbonate in comparison with sodium trypolyphosphate for marination of broiler breast meat and phase; - Marination with thyme and orange essential oils mixture to improve chicken meat quality traits, susceptibility to lipid oxidation and sensory traits. The following meat quality traits analyseswere performed: Colour, pH, water holding capacity by conventional (gravimetric methods, pressure application, centrifugation and cooking) and innovative methods (low-field NMR and DSC analysis) ability to absorb marinade soloutions, texture (shear force using different probes and texture profile analysis), proximate analysis (moisture, proteins, lipids, ash content, collagen, fatty acid), susceptibility to lipid oxidation (determinations of reactive substances with thiobarbituric acid and peroxide value), sensorial analysis (triangle test and consumer test).

Abstract:
This thesis represents a study on the application of vacuum during post-frying stage with the aim of lowering oil content in fried food. Using potato chips as an illustrative product, several protocols were initially screened, either towards the end of frying or after frying. Lowering the pressure when the product was removed from the oil resulted in a significant reduction in oil uptake. Applying vacuum triggered a continuous water vapour release from the product as a result of lower water saturation temperature, and this prevented the surface oil from penetrating into the product structure. The image generated by con focal laser scanning microscopy showed a visible boundary between the core and the crust regions, and the oil distribution was only concentrated in the crust with insignificant trace of oil in the core region. Despite the amount of oil being absorbed into fried food, the ability of oil to withstand high temperatures at the extended frying period must also be taken into consideration. Interestingly, vacuum drainage significantly inhibited the free fatty acid formation while the total oxidation value was slightly lowered, which in turn showed minimal reductions in the total colour difference value, viscosity and polar compounds. We also explored the possibility of combining moderate vacuum frying with high vacuum drainage to lower the oil uptake in fried food. As expected, high vacuum drainage significantly lowered the amount of oil taken by the potato chips, regardless to the vacuum frying conditions applied. It was also noted that the head space temperature also plays a significant role in the continuous release of water vapour from the product, aside from the product temperature. An attempt to develop fat free product was accomplished when sorbitol was used to substitute oil as frying medium. In sorbitol fried French fries, a crust was formed, similarly to oil fried product and the colour and texture developments were comparable

This research project has explored the issues surrounding East Gippsland Institute of TAFE's (EGTAFE) inability to expand the training in Hospitality and Tourism (H&T). EGTAFE is a small regional public provider of training covering a large area in regional Victoria and it has a pivotal role in the provision of knowledge and skills to assist Gippsland's transition to an innovative economy (www.egtafe.vic.edu.au/corporate/corporate). During this project discussions were held with a variety of stakeholders, namely local business owner/operators in the H&T Industry, researching issues such as industry skills requirements and also employers perception of EGTAFE. Action Research was employed as the methodology of this research project, as the cyclical process of planning, acting, observing and reflecting has facilitated the understanding and development of improvements to practices across the Hospitality and Service Industries (H&SI) team. Interviews, questionnaires and workshops were all part of the data collection method. As a result of this research project the main outcomes that have emerged are 1) a better understanding of the needs of employers and employees; and 2) the development of a strong collaborative partnership with employers. Due to the connections with industry that were made because of this research project, the H&SI team is now in a position not only to deliver industry skills requirements, but also to be a partner in the process.

The advent of globalisation and the re-admittance of South Africa into the international market place have presented both opportunities and threats to local fast food outlets and franchises. These food outlets are faced with foreign competition and the need to adapt their strategies in order to cope with the continued onslaught of these highly efficient, foreign, service orientated organisations if they intend to survive. This research explored the strategies these fast food outlets should utilise to deal with their competition and to facilitate their market growth and organisational development which could lead to them being successfully franchised at a later stage. The study revealed that if any significant growth and organisational development was to be expected, the fast food outlet had to concentrate on customer satisfaction and retention through the innovative use of the 7P’s of the marketing mix in its strategy formulation process. Attention also needed to be given to the competitive forces in the external environment so that competitive strategies were in line with market developments. The triangulation method was applied in this study. In terms of this method, qualitative and quantitative research techniques are combined in order to improve the reliability and validity of the research findings. This method proved to be an effective research tool as the findings in the quantitative research were verified by the findings in the qualitative research. Franchising in South Africa, especially the Eastern Cape, where unemployment is highest, is important as it has the potential for job creation and economic empowerment of the disadvantaged communities. It is therefore essential that every endeavour be made to ensure the success of the emerging entrepreneurs through assisting them to develop strategies that will ensure sustainable development and growth.

The most affected cell types in cerebellar ataxias are the cerebellar neurons, which are not readily accessible for cellular and molecular investigation. Pluripotent stem cell (PSC) technology has emerged as an important tool for generating diverse types of neurons, which are used in order to better understand the human nervous system development and pathologies. In this chapter, the strategies for the differentiation of human PSCs toward cerebellar neurons are overviewed, followed by an outlook of their further optimization and diversification by implementing the knowledge from cerebellar development and new cell culture approaches. The optimization stategies are based on the recent progress made in defining the cell populations in mature and developing mouse and human cerebellum. The cellular phenotypes and organization in mouse and human cerebellum are briefly presented, followed by an overview of our current knowledge about their development, which includes pattering, proliferation, neurogenesis, gliogenesis, migration, connectivity and maturation. To date, however, relatively few studies have used induced PSCs (iPSCs) to model cerebellar ataxias and even fewer have looked directly to cerebellar neurons. The reported iPSC-derived in vitro models for cerebellar ataxias are reviewed, followed by an outlook of how to improve these models by generating and exporing the cerebellar neurons.

Abstract The challenge in the Oil&Gas industry to remain competitive in a low oil price whilst dealing with minimization of operational risk and uptime asset maximization is leading offshore Companies to evolve thought proactive and predictive maintenance approaches. In the event of unplanned intervention due to anomalies or warning messages at the dispatching center, the decision on the size of the support vessel and its utilization for straordinary maintenance could be time consuming with potential high cost impact, also due to loss of production. Even the new generation of remote condition and monitoring systems, which allow to improve the capability of operators for early warnings and surveillance, provide a reliable solution for emergencies. In this context, resident subsea drones enable on-demand inspection whilst eliminating the need for support vessel and allow operator to manage the risk in continued operations also for dangerous areas restricted to human access. A case study relevant to a new subsea field development have been conducted. Distinctive Reliability, Availability and Maintainability (RAM) analyses have been performed with the aim to get insight on the subsea production system availability considering a resident drone and to demonstrate how the so called "stategic maintainability" can be applied successfully in the decision-making process while reducing the OPEX. The former related to conventional IMR (Inspection, Maintenance & Repair) based on Condition Based and RBI, Risk Based Inspection approach, the latter related to strategic maintenability with resident drones. The application of such analysis required a multi-disciplinary approach together with the possibility of processing historical data in operating conditions. Historical data sources (e.g. OREDA dataset) were collected to obtain failure rates and active repair times typical of subsea equipment. Direct experience gained in over forty years of inspection and maintenance activities together with recent developments on subsea resident robotics allow the understanding of real internvention timing. Results show that resident subsea drones applied for early inspection and light intervention are confirmed timely and costless solution respect to conventional IMR services. They represent the first aid for environmental surveillance and subsea inspection in case of emergency and provide a relevant saving of subsea production un-availability. The economic value emerged from the presented case study represents a step change for OPEX optimization and motivates Best-in-Class Operators to get an insight case-by-case for both green and aging fields.
